A blank control group of medium and cells was set up simultaneously.
After the plates were incubated in an incubator for 72 hours, the supernatant was discarded, washed twice with PBS, and then 0.5mL of trypsinized cells were added to each well for 3min, and 2mL of maintenance solution was added to each well to stop digestion. After mixing, 0.5mL of each concentration was taken out and counted. Centrifuging the rest cell suspension at 2500r/min for 5min, discarding supernatant, adding NaOH solution into the precipitate, heating to dissolve melanin, and measuring absorbance at 490nm under enzyme-linked immunosorbent assay. The melanin synthesis inhibition ratio (I') of the sample was calculated according to the formula (2).

5. Results of the experiment
The whitening anti-aging effect evaluation experiment is shown in table 4.
Figure BDA0001867504130000071
The experimental results in table 4 show that the essential oil composition of the present invention can significantly inhibit tyrosinase activity, inhibit melanin synthesis, and scavenge free radicals, and under the condition of no change of the total dosage, the effect of the essential oil composition of the present invention is significantly improved compared with the effect of a single essential oil and base oil composition and pure base oil, which indicates that the synergistic effect is exerted among different essential oils.
Test example 2 evaluation of moisturizing efficacy
Experimental materials: the products of example 1 and comparative examples 1-6 were diluted to 5 times with olive oil and used as samples to compare the change in moisture content of the skin of the test subjects after the samples were used.
Subject: a total of 30 volunteers, of which women: 15 men and 15 men, the age being 20-40 years old.
The Corneometer CM825 is adopted to measure the change of the moisture content of the stratum corneum by measuring the change of the capacitance of the stratum corneum of the skin before and after the product is used, so that the moisturizing effect of the cosmetics is evaluated.
The experimental method comprises the following steps: the testee should wash the forearm, sit at a set humidity for 30min, and then mark 3 × 3cm on the left and right forearms2The method comprises the steps of taking a left arm as a testing area of a moisturizing product, taking a symmetrical area corresponding to a right arm as a blank control, testing and detecting the moisture content of each experimental part by using a Corneometer CM825, repeating the testing for 5 times respectively to obtain an average value of each testing, recording data, calculating the average hydration rate of 20 volunteers at different time intervals, and calculating the hydration rate by using a formula (1).
Figure BDA0001867504130000081
The results are shown in Table 5 below.
TABLE 5 average skin hydration Rate for volunteers tested at different time intervals
Figure BDA0001867504130000082
As can be seen from Table 5, the skin of the sample of example 1 of the present invention still has a high moisture content 8 hours after application, and the average hydration rate still reaches above 40%, which indicates that the essential oil composition with moisturizing and repairing effects prepared by the present invention has a good moisturizing effect. The samples of comparative examples 1 to 6, however, had lower hydration rates per time period than the sample of example 1 of the present invention. Therefore, after the essential oils are scientifically combined, the moisturizing effect is obviously improved under the condition of consistent dosage, and the essential oils have good synergistic effect.
Test example 3 skin irritation test
Experimental materials: the samples of example 1 and comparative examples 1 to 6 were selected, and the change in the moisture content of the skin of the test subjects after the samples were used was compared.
Subject: a total of 30 volunteers, of which women: 15 men and 15 men, the age being 20-40 years old.
The test method comprises the following steps: 0.03mL of the sample was taken by a syringe and placed in a chamber of a spot test tape, and the spot test tape with the sample was immediately stuck to the normal skin of the left forearm from the lower part in the longitudinal direction while gently pressing the chamber one by one to expel air and uniformly distribute the test substance. The test site is marked for easy observation. Every 24 hours, and half an hour after the patch was removed, the examination was performed according to the terms of the CTFA guidelines (1981). The patch test was performed 6 times in total, and on day 6 after completion of the skin application, additional examination was performed to observe delayed reactions. The data is compared using the last examination. The identification was carried out using the following table 6 as a standard:
TABLE 6 skin irritation assay standards
Grade Symbol Identification criteria
0 - Negative reaction: has no irritation and erythema
1 ± And (3) suspicious reaction: mild erythema
2 + Weak positive reaction: erythema
3 ++ Strong positive reaction: erythema, papules, blisters
4 +++ Extremely strong positive reaction: severe edema and bulla
And (3) test results: as shown in table 7.

The results in Table 7 show that the product of example 1 of the invention has better mildness than the products of comparative examples 1-6. Therefore, the essential oil is milder than that used alone under the condition of consistent dosage after being scientifically combined.
