(1) Preparing the American cockroach extract: adding 1.8 times of water into the periplaneta americana coarse powder, performing flash extraction for 2 times under the condition of 100V voltage for 2min each time to obtain an extracting solution, and performing centrifugal separation on the extracting solution at 3600r/min for 30min to obtain periplaneta americana extract;
(2) Preparation of the Pearl dew grass extract: adding deionized water with the weight 8 times of that of the pearl dew grass into the pearl dew grass, boiling with big fire, turning into small fire, continuously boiling for 35min, and filtering to obtain first filtrate and filter residue; adding deionized water with the weight 6 times of that of the filter residue, boiling with strong fire, and continuing boiling with small fire for 25min; filtering to obtain a second filtrate; combining the first filtrate and the second filtrate to obtain a pearl dew grass extract;

The comparative example provides a preparation method of a pearl water weed extract paste, which comprises the following steps: preparation of the Pearl dew grass extract: adding deionized water with the weight 8 times of that of the pearl dew grass into the pearl dew grass, boiling with big fire, turning into small fire, continuously boiling for 35min, and filtering to obtain first filtrate and filter residue; adding deionized water with the weight 6 times of that of the filter residue, boiling with strong fire, and continuing boiling with small fire for 25min; filtering to obtain a second filtrate; combining the first filtrate and the second filtrate to obtain a pearl dew grass extract; adding gelatin 10% of its own weight into the herba Phyllanthi Urinariae extract, stirring at 700r/min for 6min, standing to remove oil and precipitate, filtering, and concentrating to two thirds of its volume to obtain herba Phyllanthi Urinariae extract spray.
Experimental example 1 in vitro bacteriostasis test for bovine mastitis
The experimental method comprises the following steps:
(1) Main reagent and culture medium: beef extract, peptone, yeast powder, agar powder and common agar medium;
(2) Test strain: coli, staphylococcus aureus and streptococcus agalactiae;
(3) Test strain preparation: activating and inoculating the test strain into LB liquid medium, culturing for 18-24 hr to obtain 1×10 strain 6 CFU/ml was ready for use.
(4) The test method comprises the following steps: preparing a sterilizing common agar culture medium plate according to a conventional method, and respectively inoculating different test strains on different plates; punching (3 mm) the inoculated strain plate with a puncher, injecting 25 μl of medicinal liquid (spray provided in examples 1-13 and comparative examples 1-10, blank group without medication) into each hole, culturing at 37deg.C for 24 hr, observing and recording the diameter (mm) of each bacteriostasis zone of each medicine, and the result is shown in Table 1;

The data in Table 1 show that the composition prepared by the preparation method of the embodiment of the application has excellent antibacterial capability on escherichia coli, staphylococcus aureus and streptococcus agalactiae.
Experimental example 2 animal experiment
230 cows identified as having recessive mastitis were selected, wherein the condition of the cow udder is: the breast is slightly red and swollen, the infected milk area is hardened, and curd blocks or purulent secretion are arranged in the curd juice.
The cows are randomly divided into 23 experimental groups of 10 cows each; experiment groups 1-12 the compositions of examples 1-12 were sprayed on the surface of the dairy cow breast, respectively; the compositions of comparative examples 1-9 were sprayed on the udder surface of cows, respectively, in experimental groups 13-21; experimental group 22 was a blank control group, which was not subjected to any treatment; the treatment of experimental group 23 was performed by using the traditional method of injecting penicillin and streptomycin into the dairy room in combination with oral dandelion powder and magnesium sulfate solution to compress the affected part of the breast, and the cure rate (calculated by taking the disappearance of the symptoms as a cure standard and the cure rate as the cure standard) after 3 days of administration (administration twice a day is administration after the milk area of the affected cow is cleaned, and the dosage of the experimental group is the same) is counted, and the result is shown in table 2:

As can be seen from the data in table 2, the composition provided in examples 1 to 12 of the present application has an excellent effect of treating cow papilloma, and the cure rate in 3 days can be up to 97% or more, which indicates that the composition prepared by the preparation method provided in the present application can greatly shorten the treatment duration and has a remarkable treatment effect.
The lower cure rates of experimental groups 13 and 14 (corresponding to comparative examples 1 and 2) compared to experimental groups 2-12 demonstrate that the various process parameters in the preparation of the pearl dew grass extract of the present application can be synergistically enhanced to provide higher levels of active ingredients in the resulting pearl dew grass extract. The lower cure rates of experimental groups 15 and 16 (corresponding to comparative examples 3 and 4) compared to experimental groups 1-12 demonstrate that the various process parameters in the preparation of the American cockroach extract of the present application can synergistically cooperate to increase the active ingredient content of the resulting American cockroach extract. The lower cure rates of experimental groups 17 and 18 (corresponding to comparative examples 5 and 6) compared with experimental groups 1-12 indicate that the synergistic interaction can be achieved in the ratio range of the pearl water weed extract and the American cockroach extract provided by the application, so that the effect of the obtained composition for treating cow papillitis is better. The lower cure rates of experimental groups 19 and 20 (corresponding to comparative examples 7 and 8) compared to experimental groups 1-12 demonstrate the synergistic interaction within the process parameters of the preparation of the compositions provided herein, so that the resulting compositions have a better effect in treating cow papilloma. The lower cure rate of experimental group 21 (corresponding to comparative example 9) compared with experimental groups 1-12 indicates that the application adopts the pearl dew grass extract and the American cockroach extract to have a synergistic effect, and the prepared composition has a significantly better effect for treating cow mammitis than the effect of singly adopting the pearl dew grass extract.
In conclusion, the composition for treating the dairy cow mastitis provided by the invention has few raw material medicines, is short in time and remarkable in effect when being used for treating the dairy cow mastitis, has no toxic or side effect and is not easy to generate drug resistance. The preparation method of the composition for treating cow mastitis provided by the invention is simple and efficient, and is suitable for large-scale industrial production.

Claims (6)
1. A composition for treating cow mastitis, comprising: the raw materials comprise, by weight, 10-20 parts of American cockroach extract and 8-13 parts of pearl water weed extract;
the preparation method of the composition comprises the following steps:
(1) The preparation of the periplaneta americana extract comprises the following steps: adding 1.5-2 times of water into the periplaneta americana coarse powder, performing flash extraction for 2-3 times under the condition of 92-108V for 2min each time to obtain an extracting solution, and performing centrifugal separation on the extracting solution to obtain the periplaneta americana extract;
(2) The preparation of the pearl water weed extract comprises the following steps: adding deionized water with the weight 7-9 times of that of the pearl dew grass into the pearl dew grass, boiling with strong fire, turning to low fire, continuously boiling for 30-40min, and filtering to obtain first filtrate and filter residue; adding deionized water with the weight 5-7 times of the weight of the filter residue into the filter residue, boiling with strong fire, and continuing boiling with small fire for 22-28min; filtering to obtain a second filtrate; combining the first filtrate and the second filtrate to obtain the pearl dew grass extract;
(3) Preparation of the composition: mixing the pearl dew grass extract and the American cockroach extract according to the weight parts to obtain a mixed solution, adding a natural clarifying agent into the mixed solution, stirring for 5-8min at the rotating speed of 600-800r/min, standing to remove grease and sediment, filtering and concentrating to obtain the composition.
2. The composition for treating cow mastitis according to claim 1, wherein: 13 parts of the periplaneta americana extract and 10 parts of the pearl water weed extract.
3. The composition for treating cow mastitis according to claim 2, wherein: in the step (2), the rotational speed of centrifugation is 3500-3800r/min, and the time is 30min.
4. A composition for treating bovine mastitis according to claim 3, wherein: the natural clarifying agent comprises one of chitosan, 101 fruit juice clarifying agent and gelatin.
5. The composition for treating bovine mastitis according to claim 4, wherein: the amount of the natural clarifying agent is 8-12% of the weight of the mixed solution.
6. The composition for treating cow mastitis according to claim 1, wherein: the formulation of the composition comprises one of ointment, spray, film coating agent and transdermal plaster.
