(1) the preparation of the periplaneta americana extract: adding water 1.5 times the weight of Periplaneta americana coarse powder, performing flash extraction for 3 times under the voltage of 108V for 2min each time to obtain an extract, and performing centrifugal separation on the extract at 3500r/min for 30min to obtain a Periplaneta americana extract;
(2) preparing the herba rhodiolae extract: adding deionized water 8 times of the weight of the pearl dew grass into the pearl dew grass, boiling the pearl dew grass with strong fire, then continuously boiling the pearl dew grass with soft fire for 35min, and filtering the pearl dew grass to obtain first filtrate and filter residue; adding deionized water 6 times of the weight of the filter residue, boiling with strong fire, and further boiling with slow fire for 25 min; filtering to obtain a second filtrate; mixing the first filtrate and the second filtrate to obtain herba Hydrocotylis Parvifolii extract;
(3) preparation of the composition: mixing 13 parts of the pearl dew grass extract and 10 parts of the periplaneta americana extract according to the parts by weight to obtain a mixed solution, adding 10% of gelatin of the weight of the mixed solution into the mixed solution, stirring and mixing the mixture for 6min at the rotating speed of 700r/min, standing the mixed solution to remove grease and sediment, filtering the mixed solution, and concentrating the filtered mixed solution to two thirds of the volume of the mixed solution to obtain the spray composition.
Example 10
The present embodiment provides a method for preparing a composition for treating bovine mastitis, comprising:
(1) the preparation of the periplaneta americana extract: adding water 2 times the weight of Periplaneta americana coarse powder, performing flash extraction for 2 times under the voltage of 92V, 2min each time to obtain an extract, and performing centrifugal separation on the extract at 3800r/min for 30min to obtain a Periplaneta americana extract;
(2) preparing the herba rhodiolae extract: adding deionized water 8 times of the weight of the pearl dew grass into the pearl dew grass, boiling the pearl dew grass with strong fire, then continuously boiling the pearl dew grass with soft fire for 35min, and filtering the pearl dew grass to obtain first filtrate and filter residue; adding deionized water 6 times of the weight of the filter residue, boiling with strong fire, and further boiling with slow fire for 25 min; filtering to obtain a second filtrate; mixing the first filtrate and the second filtrate to obtain herba Cymbopogonis Citrari extract;
(3) preparation of the composition: mixing 13 parts of the pearl dew grass extract and 10 parts of the periplaneta americana extract according to the parts by weight to obtain a mixed solution, adding 10% of gelatin of the weight of the mixed solution into the mixed solution, stirring and mixing the mixture for 6min at the rotating speed of 700r/min, standing the mixed solution to remove grease and sediment, filtering the mixed solution, and concentrating the filtered mixed solution to two thirds of the volume of the mixed solution to obtain the spray composition.

(2) preparing the herba rhodiolae extract: adding deionized water 8 times of the weight of the pearl dew grass into the pearl dew grass, boiling the pearl dew grass with strong fire, then continuously boiling the pearl dew grass with soft fire for 35min, and filtering the pearl dew grass to obtain first filtrate and filter residue; adding deionized water 6 times of the weight of the filter residue, boiling with strong fire, and further boiling with slow fire for 25 min; filtering to obtain a second filtrate; mixing the first filtrate and the second filtrate to obtain herba Cymbopogonis Citrari extract;

The comparative example provides a preparation method of a margarita and aquatic weed extract paste, comprising the following steps: preparing the herba rhodiolae extract: adding deionized water 8 times of the weight of the pearl dew grass into the pearl dew grass, boiling the pearl dew grass with strong fire, then continuously boiling the pearl dew grass with soft fire for 35min, and filtering the pearl dew grass to obtain first filtrate and filter residue; adding deionized water 6 times of the weight of the filter residue, boiling with strong fire, and further boiling with slow fire for 25 min; filtering to obtain a second filtrate; mixing the first filtrate and the second filtrate to obtain herba Cymbopogonis Citrari extract; adding gelatin 10% of its weight into herba Cymbopogonis Citrari extract, stirring and mixing at 700r/min for 6min, standing to remove oil and precipitate, filtering, and concentrating to two-thirds of its volume to obtain herba Cymbopogonis Citrari extract spray.
Experimental example 1 in vitro bacteriostatic test for mastitis in milk cow
The experimental method comprises the following steps:
(1) main reagents and culture medium: beef extract, peptone, yeast powder, agar powder and common agar culture medium;
(2) test strains: escherichia coli, staphylococcus aureus, and bovine streptococcus agalactiae;
(3) preparation of test strains: activating and inoculating the test strain in LB liquid culture medium, culturing for 18-24h, and preparing into 1 × 10 6 CFU/ml for use.
(4) The test method comprises the following steps: preparing a sterilized common agar culture medium plate according to a conventional method, and respectively inoculating different test strains on different plates; punching holes (3mm) on the plate inoculated with the strain by using a puncher, injecting 25 μ l of liquid medicine (the spray provided by examples 1-13 and comparative examples 1-10, the blank group is not used with medicine) into each hole, culturing at 37 ℃ for 24h, observing and recording the size (mm) of the inhibition zone diameter of each medicine to each bacterium, and the result is shown in table 1;

The data in table 1 show that the composition prepared by the preparation method of the embodiment of the application has excellent bacteriostatic ability on escherichia coli, staphylococcus aureus and bovine streptococcus agalactiae.
Experimental example 2 animal experiments
Selecting 230 cows identified as having subclinical mastitis, wherein the condition of the cow breasts is as follows: the breast is slightly red and swollen, the affected breast area becomes hard, and there are clotted milk or purulent secretion in the coagulated milk.
Randomly dividing the cows into 23 experimental groups, and 10 cows in each group; experimental groups 1-12 the compositions of examples 1-12 were sprayed onto the surface of a cow's udder, respectively; experimental groups 13-21 the compositions of comparative examples 1-9 were sprayed on the surface of cow udder, respectively; experimental group 22 was a blank control group, which was not subjected to any treatment; the experimental group 23 was treated by the traditional method of injecting penicillin and streptomycin into the breast chamber in combination with orally administered dandelion powder and magnesium sulfate solution for fomentation of the affected part of the breast, and the cure rate after 3 days of administration (twice daily administration, both administration after the affected cow breast areas were cleaned and the administration amount of the experimental group was the same) was counted (the cure rate was calculated as the cure standard, the cure rate was calculated as the number of cured breast areas of 10 cows/the number of affected breast areas of 10 cows: 100%), and the results are shown in table 2:

As can be seen from the data in table 2, the compositions provided in examples 1 to 12 of the present application have excellent effect of treating bovine papillitis, and the cure rate of 3 days can be as high as 97% or more, which indicates that the compositions prepared by the preparation method provided in the present application can greatly shorten the treatment period and have significant treatment effect.
The cure rates of the experimental groups 13 and 14 (corresponding to the comparative examples 1 and 2) were lower than those of the experimental groups 2-12, which shows that the synergistic effect can be achieved within the process parameter ranges in the preparation of the extract of the herba rhodiolae so as to obtain the extract of the herba rhodiolae with higher active ingredient content. The cure rates of experimental groups 15 and 16 (corresponding to comparative examples 3 and 4) were lower than those of experimental groups 1-12, indicating that synergy can be achieved within the process parameter ranges in the preparation of the periplaneta americana extract of the present application, so that the contents of active ingredients in the obtained periplaneta americana extract are higher. The cure rate of the experimental groups 17 and 18 (corresponding to the comparative examples 5 and 6) is lower than that of the experimental groups 1 to 12, which shows that the pearl dew grass extract and the periplaneta americana extract provided by the application can be synergistically synergistic within the range of the mixture ratio, so that the obtained composition has better effect of treating the bovine papillitis. The lower cure rates of experimental groups 19 and 20 (corresponding to comparative examples 7 and 8) compared to experimental groups 1-12 indicate that synergistic synergy can be achieved within the process parameters in the preparation of the compositions provided herein, such that the resulting compositions are more effective in treating bovine papillitis. The cure rate of the experimental group 21 (corresponding to the comparative example 9) is lower than that of the experimental groups 1-12, which shows that the application adopts the herba rhodiolae extract and the periplaneta americana extract to have the synergistic effect, and the effect of the prepared composition for treating the cow papillitis is obviously better than that of the composition adopting the herba rhodiolae extract alone.
In conclusion, the composition for treating the cow mastitis provided by the invention has the advantages of few raw material medicine types, short time, obvious effect, no toxic or side effect and difficulty in generating drug resistance when being used for treating the cow mastitis. The preparation method of the composition for treating cow mastitis provided by the invention is simple and efficient, and is suitable for large-scale industrial production.

2. The composition for use in the treatment of bovine mastitis according to claim 1, wherein: the raw materials also comprise a herba rhodiolae extract.
3. The composition for use in the treatment of bovine mastitis according to claim 2, wherein: according to the parts by weight, the American cockroach extract is 10-20 parts, and the margarita aquatic plant extract is 8-13 parts.
4. The composition for use in the treatment of bovine mastitis according to claim 3, wherein: according to the weight parts, the American cockroach extract is 13 parts, and the pearl dew grass extract is 10 parts.

A method of preparing a composition for treating bovine mastitis according to claims 1 to 4, wherein: the method comprises the following steps: (1) the preparation of the periplaneta americana extract: adding water with the weight 1.5-2 times of the weight of the periplaneta Americana coarse powder into the periplaneta Americana coarse powder, performing flash extraction for 2-3 times under the voltage of 92-108V for 2min each time to obtain an extracting solution, and performing centrifugal separation on the extracting solution to obtain the periplaneta Americana extract.
6. The method of preparing a composition for treating bovine mastitis according to claim 5, wherein: further comprising:
(2) preparing the herba rhodiolae extract: adding deionized water 7-9 times of the weight of herba Cymbopogonis Citrari, boiling with strong fire, decocting with slow fire for 30-40min, and filtering to obtain first filtrate and residue; adding deionized water 5-7 times of the weight of the filter residue, boiling with strong fire, and decocting with slow fire for 22-28 min; filtering to obtain a second filtrate; combining the first filtrate and the second filtrate to obtain the herba Hydrocotyles extract;
