Abstract
本发明公开了一种具有通便功效的冠突散囊菌发酵大豆粉制备工艺,将大豆清洗后用pH为6.5‑7.0的弱碱水浸泡12h,然后按照豆水比1:7的比例加水磨浆,用纱布过滤后得到豆乳;本发明的目的在于提供一种具有通便功效的冠突散囊菌发酵大豆粉及其制备工艺,具备很好自然通便功效和溶解性好、分散速度快、抗氧化性强的优点,解决了大都停留在传统的大豆制品开发与研究,缺乏创新性的、精细加工的大豆产品,而传统药物存在一定的副作用,列如,会损伤肠道粘膜、影响肠道微生态环境、引起电解质紊乱、具有药物依懒性等,不适合长期使用,同时传统大豆粉生产中,制备的大豆粉溶解性较差,易结块,风扇速度慢的问题。

背景技术
便秘是肠道功能异常的一种常见病症,随着人们生活水平的提高、饮食结构改变生活节奏加快社会心理等因素的影响,其发病率呈逐年上升趋势。我国成年人便秘的发病率为3.2%~11.6%,老年人患病率为11.5%,70岁以上人群患病率为11.7%~19.5%。虽然便秘不会直接导致死亡,危及生命,但是对人体危害仍不可轻视,便秘会给人体带来痛苦、降低生活品质,已成为影响现代人生活质量的重要病症。目前,防止便秘的产品以药物、润肠通便中草药、膳食纤维等为主。然而,药物和某些润肠通便中草药普遍存在一定的副作用,例如,会损伤肠道粘膜、影响肠道微生态环境、引起电解质絮具有药物依赖性等,不适合长期食用。目前,合理调整饮食结构、摄入足量膳食纤维是缓解和预防便秘的重要措施之一,大豆粉含有丰富的蛋白质、氨基酸及脂肪酸,营养价值较高,深受消费者喜爱。
目前,湿法工艺是制备大豆粉最常用的方法,其工艺过程一般是将大豆浸泡后磨浆,再经浓缩调配及喷雾干燥制得大豆粉。然而,大豆粉溶解性不高,其氟溶解指数为70~80%,存在冲调性差等问题,影响了大豆粉的食用性。提高大豆粉溶解性的方法有很多,如酶解处理、添加还原剂和乳化剂以及一些物理、化学方法等,近年来,蛋白质糖基化修饰备受关注,能够改善蛋白质乳化性、溶解性、抗菌和抗氧化作用,同时降低蛋白质的致敏性。此外,这种反应可以自发的发生,不需要添加化学试剂,被认为是在食品应用中最有效的绿色改性方法之一,具有很好的应用前景。葡甘露聚糖和葡聚糖属于大分子中性多糖,具有减肥、修饰肠道微生物代谢和降低胆固醇等生理功能可作为食品添加剂用于食品加工,这两种多糖可以与豆乳中蛋白质发生糖基化反应,其多羟基的分子结构能够改善大豆粉的溶解性,且反应后的糖基化产物功能性也得到改善,大豆是我们日常生活中经常食用的食材,有丰富的优质蛋白、不饱和脂肪酸、钲及B族维生素,是人们日常生活中重要的蛋白来源。同时,由于大豆中淀粉含量低,因而可为糖尿病人食用。大豆异黄酮是一种结构类似雌激素的物质,通过摄入大豆可预防女性因雌激素分泌水平降低而导致的骨质疏松。因而,大豆是一种具有极高营养价值的食品,虽然目前对于大豆的研究与开发已经比较多,但大都停留在传统的大豆制品开发与研究,缺乏创新性的、精细加工的大豆产品,而传统药物存在一定的副作用,列如,会损伤肠道粘膜、影响肠道微生态环境、引起电解质紊乱、具有药物依懒性等,不适合长期使用,同时传统大豆粉生产中,制备的大豆粉溶解性较差,易结块,风扇速度慢的问题。

为实现上述目的,本发明提供如下技术方案:一种具有通便功效的冠突散囊菌发酵大豆粉,包括以下重量组成份:
富勒烯微囊粉20~75wt%,水溶性膳食纤维20~50wt%,L-阿拉伯糖0.01~lwt%,水苏糖0.5~15wt%,果蔬粉2~20wt%,大豆70~85%,葡甘露聚糖3~8%,冠突散囊菌4~6%。
一种具有通便功效的冠突散囊菌发酵大豆粉制备工艺,包括以下步骤:
步骤1:将大豆清洗后用pH为6.5-7.0的弱碱水浸泡12h,然后按照豆水比1:7的比例加水磨浆,用纱布过滤后得到豆乳;
步骤2:向豆乳中添加葡甘露聚糖,所述的葡甘露聚糖添加量为豆乳质量的1-3%,在压力为20MPa下进行高压均质处理5min,随后经过超声波处理,干燥化得到大豆粉;
步骤3:将大豆粉与葡聚糖混合后进行研磨得到混合粉,所述大豆粉与葡聚糖质量比为6-8:1,将混合粉进行微波热风联合处理,所述的微波功率为800-1000W,热风温度为65-85℃,处理时间为4-12min,将处理后的大豆粉进行超微粉碎即得速溶大豆粉;
步骤4:取速溶大豆粉,加水后进行灭菌操作,得到速溶大豆粉溶液,水的加入量为大豆粉体积的3-5倍;
步骤5:向速溶大豆粉溶液中接种冠突散囊菌与润肠通便组合物,得到的发酵基质置于恒温26-30℃条件下发酵培养5-8天;
步骤6:按1×107-2×108CFU/100g大豆粉的量接种冠突散;
步骤7:在发酵过程中,每隔48小时在无菌条件下翻动发酵基质,直至发酵结束,将发酵后得到的成品真空冷冻干燥;
步骤8:粉碎后过60目筛,所述冠突散囊菌从茯砖茶中分离得到,得到的大豆为非转基因速溶黄大豆;
步骤9:选取的润肠通便组合物中含有富勒烯微囊粉、水溶性膳食纤维、L-阿拉伯糖、水苏糖和果蔬粉。
优选的,所述在步骤2中,将均质后的豆乳进行超声处理,所述的超声功率为200-400W,超声温度为85-95℃,超声时间为10-30min,超声处理后将豆乳冷却至室温,然后进行真空浓缩、喷雾干燥得到大豆粉。
优选的,所述在步骤3中优选葡甘露聚糖添加量为豆乳质量的2%,超声处理优选参数为:超声功率300W,超声温度90℃,超声时间20min,优选大豆粉与葡聚糖质量比为7:1,微波热风联合处理优选参数为:微波功率900W,热风温度75℃,处理时间8min。
优选的,所述在步骤5中,所述富勒烯微囊粉中含有富勒烯类化合物食用植物油、食用乳化剂壁材以及任选的稳定剂;优选地,以所述富勒烯微囊粉的总重量为基准。

富勒烯微囊粉20~75wt%,水溶性膳食纤维20~50wt%,L-阿拉伯糖0.01~lwt%,水苏糖0.5~15wt%,果蔬粉2~20wt%,大豆70~85%,葡甘露聚糖3~8%,冠突散囊菌4~6%。

将大豆清洗后用pH为6.5-7.0的弱碱水浸泡12h,然后按照豆水比1:7的比例加水磨浆,用纱布过滤后得到豆乳;向豆乳中添加葡甘露聚糖,的葡甘露聚糖添加量为豆乳质量的1-3%,在压力为20MPa下进行高压均质处理5min,随后经过超声波处理,干燥化得到大豆粉;在步骤2中,将均质后的豆乳进行超声处理,的超声功率为200-400W,超声温度为85-95℃,超声时间为10-30min,超声处理后将豆乳冷却至室温,然后进行真空浓缩、喷雾干燥得到大豆粉,将大豆粉与葡聚糖混合后进行研磨得到混合粉,大豆粉与葡聚糖质量比为6-8:1,将混合粉进行微波热风联合处理,的微波功率为800-1000W,热风温度为65-85℃,处理时间为4-12min,将处理后的大豆粉进行超微粉碎即得速溶大豆粉;在步骤3中优选葡甘露聚糖添加量为豆乳质量的2%,超声处理优选参数为:超声功率300W,超声温度90℃,超声时间20min,优选大豆粉与葡聚糖质量比为7:1,微波热风联合处理优选参数为:微波功率900W,热风温度75℃,处理时间8min,取速溶大豆粉,加水后进行灭菌操作,得到速溶大豆粉溶液,水的加入量为大豆粉体积的3-5倍;在步骤5中,富勒烯微囊粉中含有富勒烯类化合物食用植物油、食用乳化剂壁材以及任选的稳定剂;优选地,以富勒烯微囊粉的总重量为基准,食用乳化剂选自蔗糖脂肪酸酯、聚甘油脂肪酸酯、甘油脂肪酸酯、聚氧乙烯山梨糖醇酐脂肪酸酯、酪蛋白酸钠、卵磷脂、硬脂酸钠、单甘油有机酸脂肪酸酯、甜菜碱、氨基酸盐琥珀酸甘油脂肪酸酯、硬脂酰乳酸钠、吐温-80司盘-80、聚甘油酯、丙二醇脂肪酸脂和聚山梨醇脂肪酸酯中的至少一种;或者,食用乳化剂为A,或A和B的混合物,A为低聚果糖、抗性糊精、脱脂奶粉中的一种或者两种,B为酪蛋白酸钠、单,双甘油脂肪酸酯、辛烯基琥珀酸淀粉钠、抗坏血酸、抗坏血酸钠、磷脂、天然维生素E、三聚磷酸钠和抗坏血酸棕榈酸酯中的一种,稳定剂选自异抗坏血酸及其盐、没食子酸及其衍生物、葡萄糖酸、三聚磷酸钠、复合磷酸盐、磷酸氢二钾、聚磷酸钠、偏磷酸钠、羧甲基纤维素钠和琼脂中的至少一种,水溶性膳食纤维选自阿拉伯胶、果胶、角叉胶、瓜尔豆胶、亚麻籽胶、卡拉胶、愈疮胶、黄原胶、琼脂、低聚果糖、燕麦B-葡聚糖、聚葡萄糖、菊粉、抗性糊精、低聚木糖、低聚半乳糖、甜菊糖、大豆低聚糖、低聚异麦芽糖、抗性淀粉、真菌多糖、壳聚糖、魔芋粉、海藻酸钠和藻酸钠中的至少一种,准向速溶大豆粉溶液中接种冠突散囊菌与润肠通便组合物,得到的发酵基质置于恒温26-30℃条件下发酵培养5-8天;按1×107-2×108CFU/100g大豆粉的量接种冠突散;在发酵过程中,每隔48小时在无菌条件下翻动发酵基质,直至发酵结束,将发酵后得到的成品真空冷冻干燥;粉碎后过60目筛,冠突散囊菌从茯砖茶中分离得到,得到的大豆为非转基因速溶黄大豆;选取的润肠通便组合物中含有富勒烯微囊粉、水溶性膳食纤维、L-阿拉伯糖、水苏糖和果蔬粉,水溶性膳食纤维选自阿拉伯胶、果胶、角叉胶、瓜尔豆胶、亚麻籽胶、卡拉胶、愈疮胶、黄原胶、琼脂、低聚果糖、燕麦B葡聚糖、聚葡葡糖、菊粉、抗性糊精、低聚木糖、低聚半乳糖、甜菊糖、大豆低聚糖、低聚异麦芽糖、抗性淀粉、真菊多糖、壳聚糖、魔芊粉海藻酸钠和藻酸钠中的至少一种,果蔬粉选自苹果粉、猕猴桃粉、桑萁粉、无花果粉、红枣粉、枸杞粉、百合粉、山药粉、胡萝卜粉、芹菜粉、柑橘粉、葡菊粉、茭白粉、柠檬粉、苦瓜粉、南瓜粉、番茄粉、草莓粉、蓝莓粉、西瓜粉、樱桃粉、蔓越莓粉、橘皮粉、金桔粉、百香果粉、芒果粉、木瓜粉、哈密瓜粉、香蕉粉、菠萝粉、姜黄粉、杨桃粉、黄瓜粉、番石榴粉、甘蓝熟粉、茼蒿粉、空心菜粉、莴苣熟粉、黑艺麻熟粉、黑莓粉、莲藕粉、肉桂粉、白萝卜熟粉、山竹粉和荔枝粉中的至少一种。

Claims (10)
1.一种具有通便功效的冠突散囊菌发酵大豆粉,其特征在于:包括以下重量组成份:
富勒烯微囊粉20~75wt%,水溶性膳食纤维20~50wt%,L-阿拉伯糖0.01~lwt%,水苏糖0.5~15wt%,果蔬粉2~20wt%,大豆70~85%,葡甘露聚糖3~8%,冠突散囊菌4~6%。
2.一种具有通便功效的冠突散囊菌发酵大豆粉制备工艺,其特征在于:包括以下步骤:
步骤1:将大豆清洗后用pH为6.5-7.0的弱碱水浸泡12h,然后按照豆水比1:7的比例加水磨浆,用纱布过滤后得到豆乳;
步骤2:向豆乳中添加葡甘露聚糖,所述的葡甘露聚糖添加量为豆乳质量的1-3%,在压力为20MPa下进行高压均质处理5min,随后经过超声波处理,干燥化得到大豆粉;
步骤3:将大豆粉与葡聚糖混合后进行研磨得到混合粉,所述大豆粉与葡聚糖质量比为6-8:1,将混合粉进行微波热风联合处理,所述的微波功率为800-1000W,热风温度为65-85℃,处理时间为4-12min,将处理后的大豆粉进行超微粉碎即得速溶大豆粉;
步骤4:取速溶大豆粉,加水后进行灭菌操作,得到速溶大豆粉溶液,水的加入量为大豆粉体积的3-5倍;
步骤5:向速溶大豆粉溶液中接种冠突散囊菌与润肠通便组合物,得到的发酵基质置于恒温26-30℃条件下发酵培养5-8天;
步骤6:按1×107-2×108CFU/100g大豆粉的量接种冠突散;
步骤7:在发酵过程中,每隔48小时在无菌条件下翻动发酵基质,直至发酵结束,将发酵后得到的成品真空冷冻干燥;
步骤8:粉碎后过60目筛,所述冠突散囊菌从茯砖茶中分离得到,得到的大豆为非转基因速溶黄大豆;
步骤9:选取的润肠通便组合物中含有富勒烯微囊粉、水溶性膳食纤维、L-阿拉伯糖、水苏糖和果蔬粉。
3.根据权利要求2所述的一种具有通便功效的冠突散囊菌发酵大豆粉制备工艺,其特征在于:所述在步骤2中,将均质后的豆乳进行超声处理,所述的超声功率为200-400W,超声温度为85-95℃,超声时间为10-30min,超声处理后将豆乳冷却至室温,然后进行真空浓缩、喷雾干燥得到大豆粉。
4.根据权利要求2所述的一种具有通便功效的冠突散囊菌发酵大豆粉制备工艺,其特征在于:所述在步骤3中优选葡甘露聚糖添加量为豆乳质量的2%,超声处理优选参数为:超声功率300W,超声温度90℃,超声时间20min,优选大豆粉与葡聚糖质量比为7:1,微波热风联合处理优选参数为:微波功率900W,热风温度75℃,处理时间8min。
5.根据权利要求2所述的一种具有通便功效的冠突散囊菌发酵大豆粉制备工艺,其特征在于:所述在步骤5中,所述富勒烯微囊粉中含有富勒烯类化合物食用植物油、食用乳化剂壁材以及任选的稳定剂;优选地,以所述富勒烯微囊粉的总重量为基准。
6.根据权利要求2所述的一种具有通便功效的冠突散囊菌发酵大豆粉制备工艺,其特征在于:所述食用乳化剂选自蔗糖脂肪酸酯、聚甘油脂肪酸酯、甘油脂肪酸酯、聚氧乙烯山梨糖醇酐脂肪酸酯、酪蛋白酸钠、卵磷脂、硬脂酸钠、单甘油有机酸脂肪酸酯、甜菜碱、氨基酸盐琥珀酸甘油脂肪酸酯、硬脂酰乳酸钠、吐温-80司盘-80、聚甘油酯、丙二醇脂肪酸脂和聚山梨醇脂肪酸酯中的至少一种;或者,所述食用乳化剂为A,或A和B的混合物,A为低聚果糖、抗性糊精、脱脂奶粉中的一种或者两种,B为酪蛋白酸钠、单,双甘油脂肪酸酯、辛烯基琥珀酸淀粉钠、抗坏血酸、抗坏血酸钠、磷脂、天然维生素E、三聚磷酸钠和抗坏血酸棕榈酸酯中的一种。
7.根据权利要求2所述的一种具有通便功效的冠突散囊菌发酵大豆粉制备工艺,其特征在于:所述稳定剂选自异抗坏血酸盐、没食子酸衍生物、葡萄糖酸、三聚磷酸钠、复合磷酸盐、磷酸氢二钾、聚磷酸钠、偏磷酸钠、羧甲基纤维素钠和琼脂中的至少一种。
8.根据权利要求2所述的一种具有通便功效的冠突散囊菌发酵大豆粉制备工艺,其特征在于:所述水溶性膳食纤维选自阿拉伯胶、果胶、角叉胶、瓜尔豆胶、亚麻籽胶、卡拉胶、愈疮胶、黄原胶、琼脂、低聚果糖、燕麦B-葡聚糖、聚葡萄糖、菊粉、抗性糊精、低聚木糖、低聚半乳糖、甜菊糖、大豆低聚糖、低聚异麦芽糖、抗性淀粉、真菌多糖、壳聚糖、魔芋粉、海藻酸钠和藻酸钠中的至少一种。
9.根据权利要求2所述的一种具有通便功效的冠突散囊菌发酵大豆粉制备工艺,其特征在于:所述水溶性膳食纤维选自阿拉伯胶、果胶、角叉胶、瓜尔豆胶、亚麻籽胶、卡拉胶、愈疮胶、黄原胶、琼脂、低聚果糖、燕麦B葡聚糖、聚葡葡糖、菊粉、抗性糊精、低聚木糖、低聚半乳糖、甜菊糖、大豆低聚糖、低聚异麦芽糖、抗性淀粉、真菊多糖、壳聚糖、魔芊粉海藻酸钠和藻酸钠中的至少一种。
10.根据权利要求2所述的一种具有通便功效的冠突散囊菌发酵大豆粉制备工艺,其特征在于:所述果蔬粉选自苹果粉、猕猴桃粉、桑萁粉、无花果粉、红枣粉、枸杞粉、百合粉、山药粉、胡萝卜粉、芹菜粉、柑橘粉、葡菊粉、茭白粉、柠檬粉、苦瓜粉、南瓜粉、番茄粉、草莓粉、蓝莓粉、西瓜粉、樱桃粉、蔓越莓粉、橘皮粉、金桔粉、百香果粉、芒果粉、木瓜粉、哈密瓜粉、香蕉粉、菠萝粉、姜黄粉、杨桃粉、黄瓜粉、番石榴粉、甘蓝熟粉、茼蒿粉、空心菜粉、莴苣熟粉、黑艺麻熟粉、黑莓粉、莲藕粉、肉桂粉、白萝卜熟粉、山竹粉和荔枝粉中的至少一种。
