1) Preparation of skin elastic liposome a: weighing 750mg of Fucus vesiculosus extract, 562mg of palmitoyl tripeptide-5/dunaliella salina/sodium hyaluronate/provitamin B5 complex, and 375mg of nidus Collocaliae acid in 20mL of phosphate buffer solution for ultrasonic dissolution; 2500mg of phospholipid is weighed, 500mg of cholesterol is dissolved in 20mL of diethyl ether, the solution in the phosphoric acid buffer solution with the dissolved functional components is poured into the diethyl ether solution of the phospholipid, the mixture is homogenized at a high speed for 5 minutes, and the diethyl ether is removed at 30 ℃; adding 2% mannitol, ultrasonic emulsifying, filtering with 0.22 μm filter membrane, and lyophilizing at 3 deg.C/min to-20deg.C for 40 hr to obtain liposome with particle diameter of 0.13+ -0.03 μm.
2) Wetting and dispersing the thickener in water completely to obtain a mixed solution;
3) Adding an emollient, a filler, a preservative and EDTA-2NA into the mixed solution in the step 2), heating to 65 ℃, homogenizing for 6 minutes at 3200r/min to ensure that the mixed solution is completely dispersed;
4) Adding emulsifier, daily essence and pH regulator into the step 3), homogenizing for 6 min at 3200r/min to completely emulsify, stirring and cooling, and adding liposome a to obtain cream composition containing skin elasticity improving liposome.
The preparation process of the cream containing the liposome for improving skin elasticity of the above embodiment 2 comprises the following steps:
1) Preparation of skin elastic liposome b: weighing 150mg of Fucus vesiculosus extract, 112mg of palmitoyl tripeptide-5/dunaliella salina/sodium hyaluronate/provitamin B5 complex, and 75mg of nidus Collocaliae acid in 10mL of phosphate buffer solution for ultrasonic dissolution; weighing 500mg of phospholipid, dissolving 250mg of cholesterol in 10mL of diethyl ether, pouring the solution in the phosphoric acid buffer solution with the dissolved functional components into the diethyl ether solution of the phospholipid, homogenizing at high speed for 3 minutes, and removing diethyl ether at 30 ℃; adding 1% mannitol, ultrasonic emulsifying, filtering with 0.22 μm filter membrane, and lyophilizing at 1 deg.C/min to-20deg.C for 24 hr to obtain liposome with particle diameter of 0.15+ -0.04 μm.
2) Wetting and dispersing the thickener in water completely to obtain a mixed solution;
3) Adding an emollient, a filler, a preservative and EDTA-2NA into the mixed solution in the step 2), heating to 65 ℃, homogenizing for 3 minutes at 3000r/min to ensure that the mixed solution is completely dispersed;
4) Adding emulsifying agent, daily essence and pH regulator into the step 3), homogenizing for 3 min at 3000r/min to completely emulsify, stirring and cooling, and adding liposome b to obtain cream composition containing skin elasticity improving liposome.
The preparation process of the cream containing the liposome for improving skin elasticity of the above embodiment 3 comprises the following steps:
1) Preparation of skin elastic liposome c: weighing 730mg of Fucus vesiculosus extract, 548mg of palmitoyl tripeptide-5/dunaliella salina/sodium hyaluronate/provitamin B5 complex, and 365mg of nidus Collocaliae acid in 30mL of phosphate buffer solution for ultrasonic dissolution; weighing 3000mg of phospholipid, dissolving 1000mg of cholesterol in 30mL of diethyl ether, pouring the solution in the phosphoric acid buffer solution with the dissolved functional components into the diethyl ether solution of the phospholipid, homogenizing at high speed for 8 minutes, and removing diethyl ether at 30 ℃; adding 3% mannitol, ultrasonic emulsifying, filtering with 0.22 μm filter membrane, cooling filtrate to-20deg.C at 2deg.C/min, and lyophilizing for 48 hr to obtain liposome with particle diameter of 0.11+ -0.03 μm.
2) Wetting and dispersing the thickener in water completely to obtain a mixed solution;
3) Adding an emollient, a filler, a preservative and EDTA-2NA into the mixed solution in the step 2), heating to 65 ℃, homogenizing for 8 minutes at 3500r/min to ensure that the mixed solution is completely dispersed;
4) Adding emulsifying agent, daily essence and pH regulator into the step 3), homogenizing at 3500r/min for 8 min to completely emulsify, stirring and cooling, and adding liposome c to obtain cream composition containing skin elasticity improving liposome.
Effect verification test example
1. Safety test
Test sample: example 1;
the testing method comprises the following steps: and testing the security by adopting a third party detection mechanism. The selected area is not more than 50mm 2 A suitable patch device having a depth of about 1mm is provided by adding about 0.02 to about 0.025ml (g) of a subject to the patch device by a closed patch test method. The patch tester is attached to the forearm of a subject, the test object is removed after 24 hours, skin reactions are observed after 0.5 hour, 24 hours and 48 hours respectively, and the results are recorded according to the skin reaction grading standard in cosmetic safety technical regulations 2015.

4 stages: extremely strong reactions (fusogenic herpetic reactions); obvious erythema, severe infiltration, edema, blepharospermia, and response beyond the test area.
From table 2 above, the prepared cream showed no irritation and was safe.
2. Stability test
Test sample: example 1;
the testing method comprises the following steps: internal control standard, the appearance color, viscosity, pH for three months was observed.

As can be seen from the above Table 3, the prepared face cream has no significant changes in color, viscosity and pH within three months under high and low temperature or illumination conditions, which indicates that the face cream has better stability.
3. Efficacy test of liposomes to improve skin elasticity
Test sample: liposomes of example 1 which enhance skin elasticity;
the testing method comprises the following steps: and (3) measuring the inhibition rate of the sample on the non-enzymatic glycosylation by using a fluorescence spectrophotometer by adopting an in-vitro non-enzymatic glycosylation reaction experiment.
Test results: as shown in FIG. 1, the inhibition rate of non-enzymatic glycosylation gradually increased with increasing concentration of liposome, and at 5% of the use amount, the inhibition rate of non-enzymatic glycosylation reached 74%. The liposome can obviously reduce non-enzymatic glycosylation reaction, reduce damage of collagen and elastin on local skin, and has the functions of maintaining skin supporting effect and protecting skin elasticity.
4. Evaluation of Effect of face cream on improving skin elasticity
Test sample: example 1 (experimental group) and base cream (control group: cream composition without liposome encapsulation, other ingredients content same as experimental group);
the testing method comprises the following steps: the product can be used daily after cleaning face, and can be applied to whole face in the morning. The test is performed in a room at 20-22 c with a relative humidity of 40-60% and the subject must now be conditioned for 15 minutes in the same environment.
The results were tested in the following 4 time periods, respectively: before the sample (D0), after 14 days of the sample (D14), after 28 days of the sample (D28), and after 56 days of the sample (D56).
The detection items are as follows:
usingMPA580 measures skin elasticity, the closer the measurement is to 1, indicating that skin elasticity is better. (R2 is the ratio of the rebound amount of skin without negative pressure to the maximum stretching amount when negative pressure is applied; R5 is the ratio of the elastic portion of skin recovery during the first cycle of skin test to the elastic portion of skin applying negative pressure; and R7 is the ratio of the elastic portion of skin recovery during the first cycle of skin test to the maximum stretching amount of skin during this cycle).
Test results:
as shown in fig. 2-4 and tables 4-6, the values of R2, R5 and R7 of the experimental group are significantly greater than those of the control group, which indicates that the prepared cream can effectively improve skin elasticity. The three components are coated by liposome and used simultaneously, so that the synthesis of collagen and elastin of skin can be promoted, the supporting structure in dermis of the skin can be protected, and meanwhile, moisture can be provided for the skin, so that the skin is full and elastic.

5. Synergistic experiments of face cream composition
A cream composition comprises the following raw materials in proportion as shown in Table 7.

The preparation process of the face cream comprises the following steps:
1) Wetting and dispersing the thickener in water completely to obtain a mixed solution;
2) Adding main functional components, an emollient, a filler, a preservative and EDTA-2NA into the mixed solution in the step 1), heating to 65 ℃, homogenizing for 6 minutes at 3200r/min to ensure that the mixed solution is completely dispersed;
3) Adding emulsifier, daily essence and pH regulator into the step 2), homogenizing for 6 min at 3200r/min to completely emulsify, stirring, and cooling to obtain the cream composition.
The effects of example 1 and comparative examples 1 to 7 were verified (skin elasticity improving effect evaluation), and the experimental procedures and results were as follows:
1. the tester: 80 healthy females (25-45 years old) are divided into 8 groups of 10 individuals each; the same formulation was used for each group.
2. The method comprises the following steps: the product can be used daily after cleaning face, and can be applied to whole face in the morning. The test is performed in a room at 20-22 c with a relative humidity of 40-60% and the subject must now be conditioned for 15 minutes in the same environment. As seen from the results of table 8 and fig. 5, the skin elasticity is improved: example 1> comparative examples 1-7.
The cream composition containing the skin elastic liposome can effectively improve skin elasticity, mainly has the following two reasons that firstly, three functional components of the fucus serrulata extract, the palmitoyl tripeptide-5/the salted Dunaliella salina/the sodium hyaluronate/the provitamin B5 compound and the nidus Collocaliae acid are selected by compounding, so that the cream composition can promote the synthesis of skin collagen and elastin, protect a supporting structure in the dermis layer of the skin, and simultaneously provide moisture for the skin to enable the skin to be full and elastic, wherein the fucus serrulata extract can provide certain nutrition for the skin and promote the synthesis of collagen; the palmitoyl tripeptide-5/dunaliella salina/sodium hyaluronate/provitamin B5 complex can supplement skin moisture and hyaluronic acid to increase skin plumpness; the bird's nest acid can effectively remove excessive free radicals in the skin so as to protect collagen and elastin in the dermis layer of the skin.
In addition, the liposome is used as a delivery carrier of the functional components, so that the stability of the functional components can be maintained, the transdermal efficiency of the functional components can be enhanced, and the capability of the liposome for improving skin elasticity is improved.
The present invention is not limited to the preferred embodiments, but can be modified, equivalent, and modified in any way without departing from the technical scope of the present invention.
Claims (8)
1. A cream composition comprising liposome for improving skin elasticity, characterized in that,
the cream composition comprises the following components in parts by weight:
0.1-16 parts of liposome for improving skin elasticity;
84-99.9 parts of cosmetic auxiliary material matrix;
the skin elasticity improving liposome comprises the following main components in parts by weight: 0.1-6.3 parts of a tooth edge Fucus vesiculosus extract, 0.1-4.4 parts of palmitoyl tripeptide-5/dunaliella salina/sodium hyaluronate/provitamin B5 compound and 0.1-5.5 parts of nidus Collocaliae acid;
the skin elastic liposome comprises the following main components in percentage by mass:
extract of Fucus vesiculosus: palmitoyl tripeptide-5/dunaliella salina/sodium hyaluronate/provitamin B5 complex: cubilose acid = 1: (0.5-2.6): (0.1 to 0.5); wherein palmitoyl tripeptide-5: dunaliella salina: sodium hyaluronate: provitamin B5 complex = 1: (1-4): (1-4): (1-2.5).
2. A cream composition comprising enhanced skin elasticity liposomes according to claim 1,
the cosmetic auxiliary material matrix comprises the following components in parts by weight:
0.1-28 parts of an emollient;
0.12-14.5 parts of emulsifying agent;
0.15-10.5 parts of filler;
0.24-22.4 parts of thickening agent;
0.01-5 parts of preservative;
0.02-3 parts of EDTA-2NA;
0.015-0.65 part of pH regulator;
0.02-0.5 part of daily essence;
the balance of deionized water.
3. A cream composition comprising enhanced skin elasticity liposomes according to claim 1,
the cream composition comprises the following components in parts by weight:
0.5-8 parts of liposome for improving skin elasticity, which comprises the following main components: 0.1-2 parts of a fucus extract of the tooth edge, 0.2-4 parts of palmitoyl tripeptide-5/dunaliella salina/sodium hyaluronate/provitamin B5 complex and 0.1-2.4 parts of cubilose acid;
1.5 to 20.5 parts of an emollient;
2.2-10.2 parts of emulsifying agent;
0.55-8 parts of filler;
4-20 parts of a thickener;
0.01-1 part of preservative;
0.02-1.5 parts of EDTA-2NA;
0.05-0.5 part of pH regulator;
0.04-0.42 part of daily essence;
the balance of deionized water.
4. A cream composition comprising skin-elasticity-enhancing liposomes according to any one of claim 1 to 3,
the preparation method of the liposome for improving skin elasticity comprises the following steps:
1) Weighing a certain amount of Fucus vesiculosus extract, palmitoyl tripeptide-5/dunaliella salina/sodium hyaluronate/provitamin B5 complex, and dissolving nidus Collocaliae acid in phosphate buffer solution by ultrasonic;
2) Weighing a certain amount of phospholipid and cholesterol, dissolving the phospholipid and the cholesterol in a proper amount of diethyl ether, pouring the solution in the step 1) into the diethyl ether solution of the phospholipid and the cholesterol, homogenizing at a high speed for 3-10 minutes, and removing the diethyl ether at a temperature of 30 ℃;
3) Adding a certain amount of mannitol, performing ultrasonic emulsification, filtering with a 0.22 mu m filter membrane, cooling the filtrate to-20 ℃ at 1-3 ℃/min, and performing freeze drying for 24-48 hours to prepare the liposome for improving skin elasticity.
5. The cream composition containing skin elasticity enhancing liposome according to claim 4, wherein:
the phospholipid comprises: natural phospholipids such as soybean phospholipids, lecithin, and cephalin; synthetic phospholipids, such as dipalmitoyl phosphatidylcholine, dipalmitoyl phosphatidylethanolamine, distearoyl phosphatidylcholine; the mass ratio of the phospholipid to the cholesterol is (1-6): 1.
6. the cream composition containing skin elasticity enhancing liposome according to claim 4, wherein:
the particle size of the liposome is 0.025-0.2 μm.
7. A cream composition containing skin elasticity enhancing liposomes according to claim 2, wherein:
the emollient is selected from one or more of squalane, ethylhexyl isononanoate, isohexadecane, cetyl alcohol ethyl hexanoate, tocopheryl acetate, polydimethylsiloxane, hexyldecanol, trehalose, octyldodecanol lauroyl glutamate, phytosterol, xylitol, hydrogenated polyisobutene, jojoba oil, polydecene, cetostearyl glucoside, behenyl alcohol;
the emulsifier is selected from one or more of behenpolyether-25, PEG-100 stearate, sorbitan olive oleate, polysorbate 20, polysorbate-80, isocetyl stearyl polyether-20, cetyl stearyl polyether-30, arachidyl alcohol, oleyl polyether-2 and glyceryl stearate;
the filler is one or more selected from titanium dioxide, mica powder, talcum powder and silica;
the thickener is one or more selected from acrylic acid (esters) and C10-30 alkanol acrylate crosslinked polymer, carbomer, agar, pectin, hyaluronic acid, xanthan gum and alginic acid;
the preservative is one or more selected from phenoxyethanol, parahydroxybenzoate, 1, 2-pentanediol, sodium benzoate, methylparaben and sorbic acid;
the pH regulator is one or two selected from lactic acid and arginine.
8. A process for the preparation of a cream composition containing enhanced skin elasticity liposomes as claimed in any one of claims 1 to 7, characterized in that:
the method comprises the following steps:
1) Wetting and dispersing the thickener in water completely to obtain a mixed solution;
2) Adding an emollient, a filler, a preservative and EDTA-2NA into the mixed solution in the step 1), heating to 65 ℃, homogenizing for 3-8 minutes at 2500-3500r/min to ensure that the mixed solution is completely dispersed;
3) Adding emulsifying agent, daily essence and pH regulator into the step 2), homogenizing for 3-8 min at 2500-3500r/min to completely emulsify, stirring and cooling, and adding liposome for improving skin elasticity to obtain cream composition containing liposome for improving skin elasticity.
