Abstract
本发明公开了一种润肠通便的组合物,所述组合物包括益生菌复合物和益生元复合物,按照重量份数计,益生菌复合物包括:嗜酸乳杆菌1~5份、乳双歧杆菌0.5~2份、乳酸片球菌0.5~4份、长双歧杆菌0.5~4份、植物乳杆菌0.5~2份;益生元复合物包括:低聚果糖3~40份、低聚半乳糖2~30份、抗性糊精0~30份、低聚木糖0~20份、低聚异麦芽糖0~30份、菊粉1~15份、水苏糖1~8份;本发明还公开了所述组合物的应用以及一种包含所述组合物的润肠通便固体饮料。本发明的组合物将益生菌与益生元结合,可以减轻致病菌粘附,调节肠道菌群,显著降低肠易激综合症状的发病率,经过人群试食实验表明,具有促进排便、缓解腹痛、腹胀、腹泻症状的功效。

背景技术
人体肠道菌群生态与人体的健康状态息息相关,肠内正常菌群在维持人体功能和肠道内环境的稳定方面具有重要作用,包括保持人体正常的组织学及解剖学结构、促进维生素和小分子有机酸的合成、参与机体物质代谢、促进营养物质消化吸收等,同时肠道也是人体最大的免疫器官。但是,受不良饮食习惯和生活习惯的影响,人群肠道微生态容易失衡,出现便秘、肠胃不适、腹胀、腹泻等症状,亦对身心健康产生不良的影响。
中医认为,便秘是百病之祖。便秘除了给患者带来痛苦不悦之外,长期滞留在肠内的粪便还会引起肠内微生态环境恶化、肠胃功能紊乱、内分泌失调等问题。据我国流行病学调查,人群便秘的发病率约为9%-13%,其中25%-50%为老年性便秘。女性特殊的身体结构和生理特点如妊娠激素刺激、盆底肌肉薄弱等,会导致便秘发生率远高于男性。老年人因胃肠功能退化、膳食纤维摄取过少等导致排便困难、排便次数减少、粪便干结等便秘症状,对生活质量产生严重影响。特别是高血压、冠心病患者在排便过程中,易发生血压升高,诱发心肌梗死或血管破裂,危害生命。
采用西药药物治疗便秘具有起效快的优势,但易形成依赖性或抗药性,且长期服用此类药效强烈的药物容易引起结肠变黑、肠道平滑肌萎缩、肠道神经损害等不良反应。中草药类常见的泻药如大黄、番泻叶成分复杂,具有一定的毒性,大黄中的蒽醌类物质会干扰肠道内铁元素吸收,而番泻叶性寒,长期服用可能会加重体内寒气,对人体造成不利影响。
益生菌是经过分离鉴定、安全评价及功能试验后获得肯定的有益菌类,可通过定殖在人体内,改变宿主某一部位菌群组成的一类对宿主有益的活性微生物。益生菌具有维持肠道菌群结构平衡、抑制肠道炎症、保护肠道粘膜、促进营养物质吸收、提高机体免疫力等功效。由于肠道菌群与人体多项生理功能相关,因此肠道菌群失衡会产生多种生理疾病。益生菌通过定殖肠道,利用益生元自身大量繁殖,挤占肠粘膜位点和营养,获取相对有害菌的优势,从而抑制有害菌。肠内有益菌发酵低聚糖等益生元会产生大量醋酸和乳酸等短链脂肪酸,能促进肠道的蠕动、增加粪便湿润度,并保持一定的渗透压,故可以改善和防止便秘。此外,由于腹泻、腹胀、腹痛等多由有害菌引起,因此益生菌抑制有害菌可以起到预防和缓解腹泻症状的功效。
益生元是一类不被宿主消化吸收却能够选择性地促进体内有益菌代谢和增殖,从而改善宿主健康的有机物质。益生元主要作用是促进有益菌增殖,改善肠道微生态,从而间接促进脂质、蛋白质和矿物质代谢。另外,因大部分益生元都属于可溶性膳食纤维,因此具有其基本特性,包括增加粪便体积和水分、加速肠蠕动、降低粪便pH值等,大量临床实验表明,服用益生元对洁肠通便、改善和防止便秘具有显著效果。
在现有肠道健康的消费群体中,存在常年有便秘症状,希望通过功能食品等改善便秘情况的人群;或偶尔有肠胃不适,希望改善肠胃的人群;或易腹泻,希望不服用药物治疗腹泻的人群;或希望通过日常养护,预防腹泻及其他肠道疾病的人群。而益生菌和益生元制剂符合此类需求,利用益生菌或益生元调节肠道菌群已成为热点,以此为基础开发的功能食品或药品有着广阔的应用前景。
现有技术中包括以益生菌为主要原料的活菌类产品或者以益生元为主要原料的非活菌类产品。因菌株来源复杂,大部分活菌产品存在菌株功效依据尤其是临床数据支撑不足的问题。益生菌进入人体后受消化道干扰、固有菌群抵抗等因素,存活难、定殖难,并且益生菌只有在充足益生元供应的条件下才能更好地发挥促进排便等功效,而制剂规格限制了活菌产品中益生元的使用量,导致产品起效慢、体感差。而单纯的益生元产品虽然对排便有显著促进作用,但对腹泻、腹胀等症状的干预效果有限,甚至低聚果糖、菊粉在起效过程中会因大量产气而加剧腹胀反应。
此外,现有技术开发的产品起效机理主要是促进有益菌增殖,通过代谢益生元产生代谢活性产物来发挥作用。但是许多肠道不适症状是由于肠道炎症刺激引起的,益生菌和益生元无法有效干预。
发明内容
为解决上述技术问题,本发明旨在提供一种润肠通便的组合物、固体饮料及应用,能够在促进排便、改善便秘和腹泻等症状以及调节肠道菌群等方面发挥显著作用。
为达到上述目的,本发明提供如下技术方案:
一种润肠通便的益生菌组合物,包括益生菌复合物和益生元复合物。
优选的,按照重量份数计,所述益生菌复合物包括以下组分:嗜酸乳杆菌1~5份、乳双歧杆菌0.5~2份、乳酸片球菌0.5~4份、长双歧杆菌0.5~4份、植物乳杆菌0.5~2份。
优选的,所述嗜酸乳杆菌为嗜酸乳杆菌UALa-01;所述乳双歧杆菌为乳双歧杆菌UABla-12;所述乳酸片球菌为乳酸片球菌CCFM7902;所述长双歧杆菌为长双歧杆菌BL21;所述植物乳杆菌为植物乳杆菌Lp90。
嗜酸乳杆菌UALa-01和乳双歧杆菌UABla-12是经过多项人体临床实验验证功效的益生菌菌株,其中,在对100名便秘患者的临床实验中发现,患者服用UALa-01和乳双歧杆菌UABla-12后排便性状可以由便秘转为正常状态,肠道内大肠杆菌等有害菌减少,乳杆菌等有益菌增多。对IBS患者的临床实验发现,服用两种益生菌可以显著改善受试者的腹痛、腹胀、胃肠胀气、腹泻、便秘等症状。此外,通过对受试者粪便样品的检测,发现嗜酸乳杆菌UALa-01经口服进入人体后,存活率和定殖能力均达到较高水平。
优选的,所述益生菌复合物的活菌数量为10~30亿CFU/g。
优选的,按照重量份数计,所述益生元复合物为15~75份。
优选的,所述益生元复合物包括以下重量份数的组分:低聚果糖3~40份、低聚半乳糖2~30份、抗性糊精0~30份、低聚木糖0~20份、低聚异麦芽糖0~30份、菊粉1~15份、水苏糖1~8份。
更进一步优选的,按照重量份数计,所述益生元复合物包括以下组分:低聚果糖3~40份、低聚半乳糖2~30份、菊粉1~15份、水苏糖1~5份。
低聚果糖、低聚半乳糖、菊粉、水苏糖均为国际公认的益生元,它们的组合可以满足所有菌属有益菌的增殖供养需求。经过多项人体临床研究证明,低聚果糖和菊粉在增加排便次数、提高排便状况、促进双歧杆菌和乳杆菌增殖方面具有显著效果。低聚半乳糖在改善排便、促进有益菌增殖、预防和缓解腹泻、抑制有害菌吸附方面具有显著效果。水苏糖除具有益生元基本功效外,还具有抗氧化、保护肝脏等多种生理功效,且起效量低。
与单纯的益生菌或益生元相比,本发明所提供的组合物采用二者混合组成的合生元具有更明显的优势。一项针对636例IBS患者的临床研究显示,每天服用3g的低聚果糖和双歧杆菌组成的膳食补充剂,持续5周。即可增加患者排便次数,明显缓解IBS轻度症状患者的腹痛和腹胀症状。表明采用合生元可显著降低产品发挥功效时所需的起效量。
本发明第二方面在于提供如上所述的组合物在制备具有润肠通便功能的药物、普通食品、功能食品、保健食品或天然产品中的应用。
需要说明的是,上述组合物可以与药学或者食品学上可接受的载体制成粉剂、颗粒剂、胶囊剂或者片剂。
本发明第三方面在于提供一种润肠通便的固体饮料,包括以下组分:上述的组合物、野樱莓果粉、接骨木莓果粉、银耳多糖、糖和/或糖醇。
优选的,所述野樱莓果粉为野樱莓果汁粉、野樱莓提取物、野樱莓果浆粉中的一种或多种,果粉的干燥方式可采用喷雾干燥、冷冻干燥、真空干燥等形式。
优选的,所述接骨木莓果粉为接骨木莓果汁粉、接骨木莓提取物、接骨木莓果浆粉中的一种或多种,果粉的干燥方式可采用喷雾干燥、冷冻干燥、真空干燥等形式。
优选的,所述银耳多糖包括银耳多糖提取物。
优选的,所述糖包括蔗糖、果糖、葡萄糖、麦芽糖、异麦芽酮糖中的一种或多种。
优选的,所述糖醇包括赤藓糖醇、木糖醇、异麦芽酮糖醇、山梨糖醇中的一种或多种。
优选的,按照重量份数计,所述固体饮料包括以下组分:低聚果糖3~40份、低聚半乳糖2~30份、菊粉1~15份、水苏糖1~5份、赤藓糖醇2~10份、结晶果糖2~10份、野樱莓果汁粉0.5~4份、接骨木莓果汁粉0.5~4份、银耳多糖0.5~2份、嗜酸乳杆菌1~5份、乳双歧杆菌0.5~2份、乳酸片球菌0.5~4份、长双歧杆菌0.5~4份、植物乳杆菌0.5~2份。
进一步优选的,按照重量份数计,所述固体饮料包括以下组分:低聚果糖25~35份、低聚半乳糖15~30份、菊粉10~15份、水苏糖3~5份、赤藓糖醇5~10份、结晶果糖5~10份、野樱莓果汁粉1~4份、接骨木莓果汁粉1~4份、银耳多糖0.5~1.5份、嗜酸乳杆菌1~4份、乳双歧杆菌0.5~1.5份、乳酸片球菌1~4份、长双歧杆菌1~4份、植物乳杆菌0.5~1.5份。
研究表明,花青素对炎症反应引起的肠屏障通透性有一定的保护作用。本发明将低聚果糖、低聚半乳糖、菊粉和花青素组成复合物分别用于动物和细胞实验研究,发现其可以减轻肠道致病菌粘附,显著降低肠易激综合症状的严重程度,并调节了肠道菌群。而对嗜酸乳杆菌UALa-01的研究过程中也发现,花青素与嗜酸乳杆菌UALa-01结合可以显著抑制肠上皮细胞面临的大肠杆菌侵袭。因此,本发明将益生菌、益生元和花青素(野樱莓、接骨木莓富含花青素)组成复合物,对于治疗许多因肠道炎症刺激引起的不适症状具有积极效果。
野樱莓含有大量生物活性物质,如花色素苷、多酚类物质和抗氧化物等,其果实中已知含量最高的成分是花青素、黄酮、多酚。野樱莓中含有超过葡萄80倍,枸杞20倍,蔓越莓15倍,黑醋栗10倍,蓝莓5倍,阿萨伊莓果4倍以上的花青素和多酚。此外,野樱莓还具有抗氧化、保护心血管健康、抑制胃溃疡、抑制饮食肥胖等多种功效。接骨木莓是花青素含量仅次于野樱莓的一种莓果。除此之外,接骨木莓在支持免疫系统、提供抗氧化力、支持上呼吸道健康等方面均具有一定的作用。
银耳多糖对促进人体肠道健康同样具有积极作用。益生菌与肠黏膜上皮细胞紧密结合后可以提高黏膜防御屏障,阻止致病菌的定植和入侵,提高抗炎细胞因子水平,与病原竞争性黏附于上皮细胞,促进上皮细胞分泌黏液,使其在黏膜和微生物之间形成保护层,防止益生菌易位等。银耳多糖可能具有果寡糖、甘露寡糖、木糖的类似作用,能直接到达后肠作为双歧杆菌和乳酸杆菌特异性的营养物质,促进肠道有益微生物菌群的形成。
与现有技术相比,本发明具有以下优点:
(1)本发明的组合物将益生菌和益生元结合,可以减轻致病菌粘附,显著降低肠易激综合症状的发病率,并调节肠道菌群;
(2)本发明的组合物中添加了嗜酸乳杆菌和乳双歧杆菌,经过A级人体临床实验,证明其具有促进排便、调节肠道菌群、抑制致病菌、缓解腹痛、腹胀、腹泻症状的功效,并且活菌可以顺利存活进入肠道定殖;
(3)本发明的组合物中添加的乳酸片球菌、长双歧杆菌、植物乳杆菌取自中国人传统发酵食品或中国人肠道,是获得专利授权(乳酸片球菌,专利号:ZL201210054656.2;长双歧杆菌,专利号:ZL201510217639.X;植物乳杆菌,专利号:ZL201510218310.5)的国产菌株,更适合中国人肠道;
(4)本发明的组合物中添加了包括低聚半乳糖、水苏糖等在内的多种益生元构成的益生元混合粉(物),可以促进组合物中所有益生菌菌株的增殖,与益生菌形成协调增效作用,且益生元添加量可以达到通便、有益菌增殖等功效的起效量,对于改善排便状况具有显著效果;
(5)本发明的益生菌固体饮料中添加了富含花青素的野樱莓和接骨木莓果粉,2种莓果的花青素含量分别为14.8mg/g和13.75mg/g,远高于其它果蔬(如蓝莓为4.86mg/g)。花青素与益生菌组合物结合,可以发挥保护肠壁、抑制有害菌黏附的作用,协同增强治疗肠道疾病的综合症效果;
(6)本发明的益生菌固体饮料以天然莓果调节风味和色泽,自然清香,酸甜适口,未添加香精香料和色素;且原料均为普通食品,适用范围广,安全健康。

具体实施方式
下面结合实施例对本发明作进一步详细的描述,但本发明的实施方式不限于此。在未作说明的情况下,本发明所采用的试剂、设备和方法均为本技术领域常规市购的试剂、设备和常规使用的方法。
实施例1:
一种润肠通便的组合物,按照重量份数计,包括以下组分:嗜酸乳杆菌2.5份、乳双歧杆菌1份、乳酸片球菌2份、长双歧杆菌2份、植物乳杆菌1份、低聚果糖32份、低聚半乳糖25份、菊粉12份、水苏糖6份。
本实施例还提供一种润肠通便的固体饮料,将上述润肠通便的组合物以粉剂的形式添加到所述固体饮料中。按照重量份数计,所述固体饮料包括:嗜酸乳杆菌2.5份、乳双歧杆菌1份、乳酸片球菌2份、长双歧杆菌2份、植物乳杆菌1份、低聚果糖32份、低聚半乳糖25份、菊粉12份、水苏糖5份、野樱莓果汁粉2份、接骨木莓果汁粉2份、银耳多糖1份、赤藓糖醇5份、结晶果糖5份。
实施例2:
一种润肠通便的组合物,按照重量份数计,包括以下组分:嗜酸乳杆菌3份、乳双歧杆菌1份、乳酸片球菌1份、长双歧杆菌1份、植物乳杆菌1份、抗性糊精6份、低聚果糖4份、低聚半乳糖2份、菊粉2份、水苏糖1份。
上述润肠通便的组合物可以采用颗粒剂的形式应用于普通或者功能性的食品或饮料中。
实施例3:
一种润肠通便的组合物,按照重量份数计,包括以下组分:嗜酸乳杆菌1份、乳双歧杆菌2份、乳酸片球菌4份、长双歧杆菌0.5份、植物乳杆菌0.5份、低聚果糖10份、低聚半乳糖15份、抗性糊精15份、低聚木糖10份、低聚异麦芽糖15份、菊粉1份、水苏糖5份。
上述润肠通便的组合物可以采用胶囊剂的形式应用于普通或者功能性的食品或饮料中。
实施例4:
一种润肠通便的组合物,按照重量份数计,包括以下组分:嗜酸乳杆菌5份、乳双歧杆菌0.5份、乳酸片球菌0.5份、长双歧杆菌4份、植物乳杆菌2份、低聚果糖20份、低聚半乳糖30份、抗性糊精30份、低聚木糖20份、低聚异麦芽糖30份、菊粉15份、水苏糖1份。
上述润肠通便的组合物可以采用片剂的形式应用于普通或者功能性的食品或饮料中。
效果实施例
采用实施例1的润肠通便固体饮料为样品,进行人群试食实验,评价产品的润肠通便、改善排便情况的实际效果。
一、受试人群筛选
依据下表1的人群筛选标准,排除妊娠和哺乳期妇女,排除过敏体质及对保健食品过敏者,排除有心血管、肝、肾和造血系统等严重疾病者。共招募受试者72人,年龄介于20~65岁,其中男士21人,女士51人。登记受试者初始肠道健康状态、排便情况、排便次数、粪便性状,进行统计。

二、剂量及方法
采用实施例1的润肠通便固体饮料,制成规格为10g/袋的粉剂产品,受试人群每天服用1袋,连续进行10天。受试者在实验期间按照日常的饮食习惯和作息习惯生活。待实验结束后,收集受试者的肠道健康状态、排便情况、排便次数、粪便性状信息,进行统计分析。
三、实验结果
实验结果显示:
(1)如图1所示:本次受试人群反馈体感最明显的感受依次是“排便更加顺畅”、“产气增多”、“粪便更加湿润软化”、“排便次数增加”,显示本品对体验者排便状况有明显改善;
(2)如图2所示:本次受试人群中反馈“有轻微改善”以上的人群占比为73.78%,显示本品对大部分人群可以产生积极体感;
(3)如图3、图4所示:本次受试人群中粪便状况为“分离的硬团,像果核”的人群占比由受试前的9.72%下降至3.23%;粪便状况为“香肠形状,但有团块;松散的团块,边缘粗糙,像泥浆状的粪便”的人群占比由受试前的25.00%下降至12.90%;粪便状况为“像香肠或蛇,平滑而且软;像香肠,但在它的表面有裂痕;软的团块,有明显边缘”的人群占比由实验前的48.75%上升至67.74%,显示本品可以使人群粪便性状呈现明显改善。
注:图3、图4中,1代表一级(粪便等级),表示粪便性状:像香肠或蛇,平滑而且软;像香肠,但在它的表面有裂痕;软的团块,有明显边缘;2代表二级(粪便等级),表示粪便性状:香肠形状,但有团块;松散的团块,边缘粗糙,像泥浆状的粪便;3代表三级(粪便等级),表示粪便性状:分离的硬团,像果核;4代表其它。
以上所述,仅为本发明的较佳的具体实施例,但本发明的保护范围并不局限于此,任何熟悉本技术领域的技术人员在本发明揭露的技术范围内,根据本发明的技术方案及其构思加以等同替换或改变,都应涵盖在本发明的保护范围内。

1.一种润肠通便的组合物,其特征在于,包括益生菌复合物和益生元复合物。
2.根据权利要求1所述的润肠通便的组合物,其特征在于,按照重量份数计,所述益生菌复合物包括以下组分:嗜酸乳杆菌1~5份、乳双歧杆菌0.5~2份、乳酸片球菌0.5~4份、长双歧杆菌0.5~4份、植物乳杆菌0.5~2份。
3.根据权利要求2所述的润肠通便的组合物,其特征在于,所述嗜酸乳杆菌为嗜酸乳杆菌UALa-01;所述乳双歧杆菌为乳双歧杆菌UABla-12;所述乳酸片球菌为乳酸片球菌CCFM7902;所述长双歧杆菌为长双歧杆菌BL21;所述植物乳杆菌为植物乳杆菌Lp90。
4.根据权利要求1所述的润肠通便的组合物,其特征在于,所述益生菌复合物的活菌数量为10~30亿CFU/g。
5.根据权利要求1所述的润肠通便的组合物,其特征在于,按照重量份数计,所述益生元复合物为15~75份;
益生元复合物包括以下重量份数的组分:低聚果糖3~40份、低聚半乳糖2~30份、抗性糊精0~30份、低聚木糖0~20份、低聚异麦芽糖0~30份、菊粉1~15份、水苏糖1~8份。
6.根据权利要求5所述的润肠通便的组合物,其特征在于,所述益生元复合物包括以下重量份数的组分:低聚果糖3~40份、低聚半乳糖2~30份、菊粉1~15份、水苏糖1~5份。
7.如权利要求1~6任一项所述的组合物在制备具有润肠通便功能的药物、普通食品、功能食品、保健食品或天然产品中的应用。
8.一种润肠通便的固体饮料,其特征在于,包括以下组分:如权利要求1~6任一项所述的组合物、野樱莓果粉、接骨木莓果粉、银耳多糖、糖和/或糖醇。
9.根据权利要求8所述的润肠通便的固体饮料,其特征在于,所述野樱莓果粉为野樱莓果汁粉、野樱莓提取物、野樱莓果浆粉中的一种或多种;
所述接骨木莓果粉为接骨木莓果汁粉、接骨木莓提取物、接骨木莓果浆粉中的一种或多种;
所述银耳多糖包括银耳多糖提取物;
所述糖包括蔗糖、果糖、葡萄糖、麦芽糖、异麦芽酮糖中的一种或多种;
所述糖醇包括赤藓糖醇、木糖醇、异麦芽酮糖醇、山梨糖醇中的一种或多种。
10.根据权利要求9所述的润肠通便的固体饮料,其特征在于,按照重量份数计,所述固体饮料包括以下组分:低聚果糖25~35份、低聚半乳糖15~30份、菊粉10~15份、水苏糖3~5份、赤藓糖醇5~10份、结晶果糖5~10份、野樱莓果汁粉1~4份、接骨木莓果汁粉1~4份、银耳多糖0.5~1.5份、嗜酸乳杆菌1~4份、乳双歧杆菌0.5~1.5份、乳酸片球菌1~4份、长双歧杆菌1~4份、植物乳杆菌0.5~1.5份。
