Abstract
本发明公开了食品饮料生产领域的一种具有提高免疫作用的益生菌固体饮料及其制备方法,具有提高免疫作用的益生菌固体饮料由益生菌粉和辅料组成;益生菌粉包括:乳双歧杆菌、嗜酸乳杆菌、副干酪乳杆菌、植物乳杆菌、鼠李糖乳杆菌、发酵乳杆菌;辅料包括:低聚果糖、银耳多糖、低聚半乳糖、酵母β‑葡聚糖、赤藓糖醇、乳糖醇、果味粉;采用低温、低湿条件下的无菌干混工艺,按配方比例混合均匀,经称重包装成固体饮料包;本发明的益生菌固体饮料具有提高免疫、改善肠道菌群、方便即食、口感清甜等优点。多种益生元的加入既能保证益生菌摄入后的定植与增殖,保护肠道健康,又能满足提高免疫的需求,安全温和、无依赖性。
Description
技术领域
本发明涉及食品饮料生产领域的一种具有提高免疫作用的益生菌固体饮料及其制备方法。
背景技术
免疫是人体自身的防御机制,是人体识别和消灭外来侵入机体的病毒、细菌等异物,识别和处理衰老、损伤、死亡、变性的自身细胞以及突变和病毒感染细胞的能力,免疫低下状态即当身体在受到外来细菌、病毒入侵时,身体抵抗能力下降的状态。机体免疫低下易导致细菌、病毒、真菌等入侵,因此免疫低下最直接的表现就是容易生病。
乳酸菌可以通过影响机体特异性和非特异性免疫应答增强免疫,一方面可以促进T、B淋巴细胞增殖分化成浆细胞产生细胞因子IL-2、IL-10及抗体IgA、IgG等参与机体的体液免疫和细胞免疫特异性调节,一方面还可以提高单核巨噬细胞的吞噬活性,诱导活性氧、溶酶体酶及单核因子等的分泌,防止病原微生物等有害物质的入侵,参与非特异性免疫调节。益生菌除了提高免疫外,还具有调节和维持肠道菌群结构平衡、降胆固醇、抗氧化、降血糖等作用。
益生元是一类膳食补充剂,能够选择性地促进体内有益菌的代谢和增殖,而不刺激有潜在致病性或腐败活性的有害细菌,进而改善宿主健康的不可被人体消化的食品成分。益生元进入人体,主要发挥以下作用:促进肠道内乳杆菌和双歧杆菌的增殖和生长;具有较强的免疫刺激作用,可以增强巨噬细胞活性使其产生抗菌素,进而刺激淋巴细胞不断分裂繁殖。
酵母β-葡聚糖是一种优质益生元,能有效促进双歧杆菌和乳杆菌在体内的增殖和定植。食用后经过口腔、食管、胃、到达小肠,通过胞呑(或胞饮)进入淋巴系统,进入免疫系统,刺激增强NK细胞、巨噬细胞等免疫细胞的吞噬活性,提高巨噬细胞的吞噬能力达到平时的10倍以上,同时刺激释放白介素以增强人体抵抗有害毒素的能力,有效增强人体清除毒素的能力;还促进IgM抗体的产生,提高人体免疫。低聚果糖、银耳多糖、低聚半乳糖等同时即是优良的益生元,自己本身也具有很好的提高免疫的作用。
固体饮料是用食品原辅料、食品添加剂等加工制成的粉末状、颗粒状或块状等,因其品种多样,冲调方便、易于贮藏和携带而广受消费者青睐。
近年来,益生菌产品和提高免疫产品都是热点,提高免疫的益生菌固体饮料既能满足人们对增强免疫的需求,又能满足人们对肠道健康、消化改善的需求。因此,开发一款安全温和,不生产依赖性的方便携带的提高免疫产品很有必要。
发明内容
本发明的目的在于提供一种具有提高免疫作用的益生菌固体饮料及其制备方法,由益生菌粉和辅料组成,既能保证货架期内的活菌数,又能保证益生菌摄入后的定植与增殖,且产品直接食用具有良好的口感,食用方便。
为了达到上述目的,本发明是通过以下技术方案实现的:
一种具有提高免疫作用的益生菌固体饮料,其特征在于:所述固体饮料由益生菌粉和辅料组成,益生菌粉由乳双歧杆菌、嗜酸乳杆菌、副干酪乳杆菌、植物乳杆菌、鼠李糖乳杆菌、发酵乳杆菌组成,辅料由低聚果糖、银耳多糖、低聚半乳糖、酵母β-葡聚糖、赤藓糖醇、乳糖醇、果味粉组成;
对上述方案作进一步说明:所述提高免疫作用的益生菌固体饮料重量百分比为:低聚果糖30%-50%、银耳多糖1%-2%、低聚半乳糖1%-2%、酵母β-葡聚糖5%-10%、赤藓糖醇8%-15%、乳糖醇15%-25%、果味粉5%-10%,乳双歧杆菌1%-3%、嗜酸乳杆菌0.5%-2%、副干酪乳杆菌1%-3%、植物乳杆菌1%-3%、鼠李糖乳杆菌1%-3%、发酵乳杆菌0.5%-2%;
对上述方案作进一步说明:所述益生菌活菌数为:乳双歧杆菌1×1011-2×1011CFU/g,嗜酸乳杆菌1×1011-2×1011CFU/g,副干酪乳杆菌1×1011-3×1011CFU/g,植物乳杆菌1×1011-4×1011CFU/g,鼠李糖乳杆菌1×1011-4×1011CFU/g,发酵乳杆菌1×1011-5×1011CFU/g;
对上述方案作进一步说明:所述的果味粉为甜橙粉、柳橙粉、草莓粉、蓝莓粉、蔓越莓粉、不老莓粉、桃子粉、抹茶粉和咖啡粉中的一种或几种;
对上述方案作进一步说明:所述固体饮料的水分含量不大于5%,水分活度在0.1-0.2之间;
一种具有提高免疫作用的益生菌固体饮料的制备方法,包括以下工艺步骤:
1)选取甘油管保藏单菌株,37℃解冻后划线接种于MRS琼脂培养基,挑取平板上的单菌落接种于MRS液体培养基培养至对数生长期作为种子发酵剂,采用高密度培养得到发酵液,使用离心机离心获得菌泥,菌泥与脱脂乳保护剂混合乳化后进行冻干、粉碎,获得乳双歧杆菌、嗜酸乳杆菌、副干酪乳杆菌、植物乳杆菌、鼠李糖乳杆菌、发酵乳杆菌冻干粉;
2)原料验收,原料水分含量需小于5%,水分活度小于0.25;
3)在GMP车间内,无菌、低温、低水分活度的条件下,使用混料机采用等量递增原则进行混料,首先取与益生菌等量的辅料与益生菌一起混匀,再逐步加辅料直至加完为止,混料机转速为60r/min;混料时间为20-30min;
4)将混合料过60-80目筛,检测水分含量和水分活度,备用;
5)将混合料称重,使用粉末灌装机分装成小包固体饮料,无菌封装;
6)检验产品活菌数、水分含量和水分活度指标;
7)装箱入库,可室温保藏,冷藏或冻藏效果更佳。
本发明的益生菌固体饮料所用乳双歧杆菌、鼠李糖乳杆菌、嗜酸乳杆菌等乳酸菌是人体肠道内的生理常驻菌,具有提高免疫的,改善肠道内环境,起到益生作用。使用益生菌与益生元结合,不仅能促进益生菌的增殖和定植,而且能发挥自身的功效。本发明的固体饮料除了提高免疫外,还具有改善肠道、降低胆固醇、助消化等功效。
本发明具有以下有益效果:本发明所用益生菌具有较好的耐酸耐胆盐及肠道定植能力,确保其有效发挥益生特性。本发明配方科学,口感良好,具有提高免疫的功效;本发明的益生菌固体饮料无其它副作用,可作为食品或保健食品长期食用。

下面将结合具体实施例对本发明的技术方案作进一步地说明。
实施例1
一种具有提高免疫作用的益生菌固体饮料,是由以下成分组成,各组份重量如下:
低聚果糖1.175g、银耳多糖0.05g、低聚半乳糖0.05g、酵母β-葡聚糖0.25g、赤藓糖醇0.4g、乳糖醇0.6g、甜橙粉0.25g,乳双歧杆菌0.05g、嗜酸乳杆菌0.02g、副干酪乳杆菌0.045g、植物乳杆菌0.03g、鼠李糖乳杆菌0.05g、发酵乳杆菌0.03g。
实施例2
一种具有提高免疫作用的益生菌固体饮料,是由以下成分组成,各组份重量如下:
低聚果糖1.4g、银耳多糖0.06g、低聚半乳糖0.06g、酵母β-葡聚糖0.2g、赤藓糖醇0.3g、乳糖醇0.555g、柳橙粉0.2g,乳双歧杆菌0.07g、嗜酸乳杆菌0.04g、副干酪乳杆菌0.03g、植物乳杆菌0.03g、鼠李糖乳杆菌0.035g、发酵乳杆菌0.02g。
实施例3
一种具有缓解腹泻作用的益生菌固体饮料,是由以下成分组成,各组份重量如下:
低聚果糖0.6g、银耳多糖0.025g、低聚半乳糖0.025g、酵母β-葡聚糖0.2g、赤藓糖醇0.3g、乳糖醇0.5g、蓝莓粉0.2g,乳双歧杆菌0.04g、嗜酸乳杆菌0.02g、副干酪乳杆菌0.02g、植物乳杆菌0.03g、鼠李糖乳杆菌0.02g、发酵乳杆菌0.02g。
本发明实施例1-3所公开的一种具有提高免疫作用的益生菌固体饮料,是经过以下工艺制备而成的:

以下为动物实验对益生菌固体饮料提高免疫的功效数据:
以市售益生菌固体饮料作为阳性对照,使用BALB/c小鼠小鼠模拟实验,利用皮下注射环磷酰胺方法建立免疫低下小鼠模型,分别使用实施例1、2、3制备的样品干预,灌胃量为0.1g/100g(样品体重比),28d后,分别观察各组小鼠的状态,测定各组小鼠的相关指标,方法及结果如下:

实验开始前,对除空白组外其余组通过采取连续3天皮下注射40mg/kg的环磷酰胺方法建立免疫低下小鼠模型。
1.3小鼠状态观察
每天定期对小鼠称重,记录小鼠体重变化,并根据体重调整灌胃量。过程中观察体毛光泽顺滑程度、精神状态、行为、进食状况等基本情况。
1.4血清指标检测
每只小鼠取眼球血0.5~1.0mL置于1.5mL离心管中,静置1h后,3000rpm离心15min使得血清充分析出,转移血清至另一离心管,-20℃冻存即为待测样。检测指标:TNFα、IFNγ、IL-1b、IL-4、IL-6、IL-10。
2实验结果
2.1小鼠状态观察
模型组小鼠与正常组比,毛色灰暗且粗糙,小鼠体重偏低且反应迟钝,腹腔注射的创口不易恢复,经样品干预后小鼠的毛色有所恢复,反应也较模型组敏捷,说明经样品干预可以改善免疫低下小鼠的状态。
2.2益生菌固体饮料对免疫力低下小鼠血清免疫指标的影响
图1为益生菌固体饮料对免疫力低下小鼠血清免疫指标的影响图与对照组小鼠相比,模型组小鼠血清中的TNFα、IFNγ、IL-1b、IL-4、IL-6、IL-10含量显著降低,说明环磷酰胺显著的影响了大鼠血清中TNFα、IFNγ、IL-1b、IL-4、IL-6、IL-10的含量。细胞因子是机体免疫系统的重要组成,既是免疫应答反应的产物,又可以增强免疫应答反应,其中TNF-α具有广泛的生物学活性,是反映机体免疫反应程度的重要指标,在免疫的发生和发展中具有重要作用。TNF-α是机体免疫反应中的重要介质,可促进淋巴细胞增殖,激活NK细胞,也具有选择性杀灭肿瘤细胞的作用;IFN-γ是免疫细胞分泌的细胞因子,具有抗肿瘤及免疫调节作用。益生菌固体饮料能明显提高免疫抑制小鼠血清IFN-γ和TNF-α水平,表明益生菌固体饮料能够通过促进细胞因子的分泌调节免疫抑制小鼠的免疫功能。
以上所述仅是本发明的一些实施方式及验证效果,对于本领域的技术人员来说,在不脱离本发明的前提下,所做出各种形式的修改、替换和改进,均属于本发明的保护范围。
Claims (6)
1.一种具有提高免疫作用的益生菌固体饮料,其特征在于:所述固体饮料由益生菌粉和辅料组成,益生菌粉由乳双歧杆菌、嗜酸乳杆菌、副干酪乳杆菌、植物乳杆菌、鼠李糖乳杆菌、发酵乳杆菌组成,辅料由低聚果糖、银耳多糖、低聚半乳糖、酵母β-葡聚糖、赤藓糖醇、乳糖醇、果味粉组成。
2.根据权利要求1所述的具有提高免疫作用的益生菌固体饮料,其特征在于:所述提高免疫作用的益生菌固体饮料重量百分比为:低聚果糖30%-50%、银耳多糖1%-2%、低聚半乳糖1%-2%、酵母β-葡聚糖5%-10%、赤藓糖醇8%-15%、乳糖醇15%-25%、果味粉5%-10%,乳双歧杆菌1%-3%、嗜酸乳杆菌0.5%-2%、副干酪乳杆菌1%-3%、植物乳杆菌1%-3%、鼠李糖乳杆菌1%-3%、发酵乳杆菌0.5%-2%。
3.根据权利要求1所述的具有提高免疫作用的益生菌固体饮料,其特征在于:所述益生菌粉活菌数为:乳双歧杆菌1×1011-2×1011CFU/g,嗜酸乳杆菌1×1011-2×1011CFU/g,副干酪乳杆菌1×1011-3×1011CFU/g,植物乳杆菌1×1011-4×1011CFU/g,鼠李糖乳杆菌1×1011-4×1011CFU/g,发酵乳杆菌1×1011-5×1011CFU/g。
4.根据权利要求1所述的具有提高免疫作用的益生菌固体饮料,其特征在于:所述的果味粉为甜橙粉、柳橙粉、草莓粉、蓝莓粉、蔓越莓粉、不老莓粉、桃子粉、抹茶粉和咖啡粉中的一种或几种。
5.一种具有提高免疫作用的益生菌固体饮料的制备方法,其特征在于:包括以下工艺步骤:
