Abstract
本发明属于保健品技术领域,具体涉及一种含有花青素的组合物及其制备方法与应用。本发明提供的含有花青素的组合物包括如下组分及其重量份数:越橘提取物8~15份,红石榴提取物8~20份,针叶樱桃粉5~15份,蔓越莓粉10~25份,雨生红球藻提取物1~3份,复合酵素5~8份,益生元1~6份。本发明提供的一种含有花青素的组合物首次将几种强氧化性原料复配使用,同时加入复合酵素和益生元,具有较高的抗氧化性能,对DPPH自由基的清除率高达88.4%,对羟自由基的清除率高达99.1%,并且有助于肠道健康,使机体对营养物质能有更好的吸收,可广泛应用于保健护肤产品。

本发明属于保健品技术领域,具体涉及一种含有花青素的组合物及其制备方法与应用。
背景技术
衰老可分为两类:生理性衰老和病理性衰老。前者指成熟期后出现的生理性退化过程,后者是由于各种外来因素(包括各种疾病)所导致的老年性变化。随着人们物质生活水平的提高,抗衰老已经成为人们生活中的一个重要议题,尤其是对于女性,为了保持年轻的体态和容貌,在抗衰老的产品中也是不惜重金。20世纪80年代人们逐渐认识到清除自由基和抗氧化是营养保健的重要前提和基础,而花青素作为清除自由基能力最强的、其他抗氧化剂所无可比拟的抗氧化剂。
花青素是一种水溶性色素,可以随着细胞液的酸碱改变颜色。细胞液呈酸性则偏红,细胞液呈碱性则偏蓝,花青素是构成花瓣和果实颜色的主要色素之一。花青素为植物二级代谢产物,在生理上扮演重要的角色,花青素是纯天然的抗衰老的营养补充剂,研究证明是当今人类发现最有效的植物抗氧化剂,它的抗氧化性能比维生素E高出五十倍,比维生素C高出二十倍,它对人体的生物有效性是100%。花青素由于其卓越的抗氧化能力,它可以清除人体内致病的自由基,减缓细胞死亡和细胞膜变性,从而延缓衰老。在欧美等国家花青素享有“皮肤维生素”、“口服化妆品”的美誉,其主要功效为:(1)它可以维护皮肤胶原合成,抑制弹性蛋白酶,从而减少皱纹产生;(2)花青素可抑制络氨酸酶活性,具有防晒美白的作用;(3)花青素具有的多羟基结构使它在空气中易吸湿,且能与多糖(透明质酸)、蛋白质、脂类(磷脂)、多肽等复合,从而达到保湿收敛皮肤的效果。
中国专利申请CN108719704A公开了一种富含花青素的饮料组合物,包括:紫薯浓缩汁5~30重量份、不老梅汁5~28重量份、苹果汁3~20重量份和水10~40重量份,本发明制备的富含花青素的饮料虽然能在一定程度被人体吸收,但是其中的花青素成分不稳定,易分解,不利于人们的使用。
本发明旨利用花青素的强抗氧化能力,开发出一种以花青素为主要成分的组合物,具有抗氧化、保湿、抗皱、美白作用,已解决抗氧化性差和利用率低、稳定性较差等问题,可应用于女性护肤品及保健食品。
发明内容
为了解决现有技术中存在的问题,本发明的目的在于提供一种含有花青素的组合物及其制备方法与应用,本发明提供的一种含有花青素的组合物首次将几种强氧化性原料复配使用,同时加入复合酵素和益生元,具有较高的抗氧化性能,对DPPH自由基的清除率高达88.4%,羟自由基的清除率高达99.1%,并且有助于肠道健康,使机体对营养物质能有更好的吸收,可广泛应用于保健护肤产品。
本发明的技术方案是:
一种含有花青素的组合物,包括如下组分及其重量份数:越橘提取物8~15份,红石榴提取物8~20份,针叶樱桃粉5~15份,蔓越莓粉10~25份,雨生红球藻提取物1~3份,复合酵素5~8份,益生元1~6份。
进一步地,所述的含有花青素的组合物,由如下组分及其重量份数组成:越橘提取物12份,红石榴提取物16份,针叶樱桃粉10份,蔓越莓粉18份,雨生红球藻提取物2份,复合酵素7份,益生元4份。
进一步地,所述益生元为菊粉、低聚半乳糖、低聚果糖、水苏糖、膳食纤维的一种或几种。
进一步地,所述复合酵素包括以下组分及其重量份数:蕨麻15~26份,芦荟10~18份,银耳4~10份,干酵母0.1~0.3份。
进一步地,所述复合酵素由以下组分及其重量份数组成:蕨麻21份,芦荟15份,银耳8份,干酵母0.2份。
进一步地,所述复合酵素的制备步骤如下:
将蕨麻、芦荟、银耳洗净切碎,高温灭菌后,置入无菌发酵罐中,并添加干酵母,密封存放于室温下10~30天后,经200目过滤,即得。
另外,本发明还提供了一种含有花青素的组合物的制备方法,所述制备步骤如下:
S1、将越橘提取物,红石榴提取物,雨生红球藻提取物搅拌5~10min,混合均匀,得混合液;
S2、将针叶樱桃粉,蔓越莓粉,复合酵素和益生元加入到步骤S1所得的混合液中,120~200r/搅拌15~20min,即得。
另外,本发明还提供了一种含有花青素的组合物在制备抗氧化保健品和化妆品中的应用。
进一步地,所述的含有花青素的组合物在抗氧化保健品和化妆品中的添加量为10~60%。
本发明所用越橘提取物主要成分为花青素,含量高、稳定性好。越橘提取物中发现超过15种不同的花青素。越橘花青素是高级医药中间体,有助于维持毛细血管完整性和稳定胶原质。花青素也是有力的抗氧化剂。众多的临床研究显示越橘在治疗循环系统不适、静脉曲张和其它静脉和动脉不适方面十分有效。同时,花青素可稳定内皮细胞中磷脂,通过增加胶原质及黏多糖维持动脉壁结构的完整性,以此通过合成物来保护静脉和动脉。花青素同样防止血小板在内皮细胞表面的聚集和粘着。研究还显示越橘可通过刺激视紫质产生来起到在亚铁血红素所致和糖尿病导致视网膜病变中协作治疗的作用。
红石榴性味甘、酸涩、温,果实中含有丰富的维生素C、石榴多酚和花青素等物质,有很好的排毒、抗氧化功效。在药用、美容、食疗方面都有重要作用。本发明采用的红石榴提取物中含有大量的石榴多酚和花青素,抗氧化性高出绿茶3倍,更是维生素C的20倍,能有效中和自由基,促进新陈代谢、排出毒素。红石榴提取物具有抗氧化、消炎抗菌等功效,有效成分为花青素、石榴多酚、鞣花酸,这些成分都是极强的抗氧化物质,能有效延缓肌肤衰老、保湿、抗皱。
针叶樱桃富含维生素C,是世界上已知含维生素C最丰富的水果,同时针叶樱桃还含有维生素A、B1、B2、E、P、烟碱酸、抗衰老因子(SOD)、钙、铁、锌、钾和蛋白质等多种营养物质,营养价值极高,有“生命之果”的美誉。其中维生素P能防止维生素C被氧化而受到破坏,增强VC的吸收和利用效果。
蔓越莓粉含有丰富的维他命A、维他命C、维他命E、花色素、马尿酸、儿茶素、疫苗素等,具有非常好之抗氧化、抗菌及净化效益。特别是蔓越莓含有超级热门的抗氧化物——原花青素,藉由其特殊的抗氧化能力以及自由肌清除者的条件,可避免细胞受破坏并维持细胞的健康与活力。在预防和治疗尿路感染,降低皮肤癌发病率,消化系统,松弛和清理血管,龋齿、胃溃疡、肾结石,心脏病发作,动脉阻塞和中风等有着重要的作用。
雨生红球藻是一种虾青素富集天然海洋微藻,能避免UV造成的皮肤损伤,能调理皮肤色素沉着,能清除自由基,具有很好的抗氧化,防衰老的功能。从雨生红球藻中提取的天然产物,含有丰富的营养成分,以虾青素为主,具有强效的抗氧化作用,延缓肌肤衰老。
本发明提供的含有花青素的组合物,首次将几种强氧化性原料复配使用,各组分配比合理,协同作用,有效的增加了本发明提供的含有花青素的组合物的抗氧化能力。本发明人意外的发现,当在组分中添加量一定量的复合酵素后,能有利于人体的吸收,并能进一步提升抗氧化能力,同时加入益生元,有助于肠道健康,使机体对营养物质能有更好的吸收,从里到外进行全面提升。
与现有技术相比,本发明提供的含有花青素的组合物具有以下优势:
(1)本发明提供的含有花青素的组合物,本发明首次将几种强氧化性原料复配使用,同时加入复合酵素和益生元,进一步提升抗氧化能力,有助于肠道健康,使机体对营养物质能有更好的吸收,从里到外进行全面提升。
(2)本发明提供的含有花青素的组合物,抗氧化能力显著,具有较强的自由基清除能力和抗氧化能力,不仅能够促进人体新陈代谢、改善皮肤,而且还具有一定的抗氧化和抗衰老的功效,对DPPH自由基的清除率高达88.4%,羟自由基的清除率高达99.1%,可广泛用于护肤产品。
(3)本发明提供的含有花青素的组合物,对增强视力、消除眼睛疲劳、延缓脑神经衰老有显著的功效,同时对由糖尿病引起的毛细血管病也有较好治疗作用,还能增强心肺功能、预防老年痴呆等多种疾病,人们长期服用能够起到提神醒脑以及提高免疫力等优点,更能够明显改善不同程度视疲劳引起的精神症状,如注意力不集中、记忆力下降等,可广泛用于保健产品。
具体实施方式
以下通过具体实施方式的描述对本发明作进一步说明,但这并非是对本发明的限制,本领域技术人员根据本发明的基本思想,可以做出各种修改或改进,但是只要不脱离本发明的基本思想,均在本发明的保护范围之内。
越橘提取物购自湖北信康医药化工有限公司,红石榴提取物、蔓越莓粉、雨生红球藻提取物购自陕西斯诺特生物技术有限公司,针叶樱桃粉购自兰州沃特莱斯生物科技有限公司。
本发明所用的其余试剂均为常用试剂,均可在常规试剂生产销售公司购买。
实施例1一种含有花青素的组合物
所述的含有花青素的组合物,由如下组分及其重量份数组成:越橘提取物8份,红石榴提取物8份,针叶樱桃粉5份,蔓越莓粉10份,雨生红球藻提取物1份,复合酵素5份,低聚半乳糖1份。
所述复合酵素包括以下组分及其重量份数:蕨麻15份,芦荟10份,银耳4份,干酵母0.1份。
所述复合酵素的制备步骤如下:
将蕨麻、芦荟、银耳洗净切碎,高温灭菌后,置入无菌发酵罐中,并添加干酵母,密封存放于室温下10天后,经200目过滤,即得。
所述的含有花青素的组合物的制备方法,所述制备步骤如下:
S1、将越橘提取物,红石榴提取物,雨生红球藻提取物搅拌5min,混合均匀,得混合液;
S2、将针叶樱桃粉,蔓越莓粉,复合酵素和低聚半乳糖加入到步骤S1所得的混合液中,120r/搅拌15min,即得。
实施例2一种含有花青素的组合物
所述的含有花青素的组合物,由如下组分及其重量份数组成:越橘提取物12份,红石榴提取物16份,针叶樱桃粉10份,蔓越莓粉18份,雨生红球藻提取物2份,复合酵素7份,益生元4份。
所述益生元为菊粉、低聚果糖、水苏糖按重量比1:5:3组成。
所述复合酵素包括以下组分及其重量份数:蕨麻21份,芦荟15份,银耳8份,干酵母0.2份。
所述复合酵素的制备步骤如下:
将蕨麻、芦荟、银耳洗净切碎,高温灭菌后,置入无菌发酵罐中,并添加干酵母,密封存放于室温下20天后,经200目过滤,即得。
所述的含有花青素的组合物的制备方法,所述制备步骤如下:
S1、将越橘提取物,红石榴提取物,雨生红球藻提取物搅拌8min,混合均匀,得混合液;
S2、将针叶樱桃粉,蔓越莓粉,复合酵素和益生元加入到步骤S1所得的混合液中160r/搅拌18min,即得。
实施例3一种含有花青素的组合物
所述的含有花青素的组合物,由如下组分及其重量份数组成:越橘提取物15份,红石榴提取物20份,针叶樱桃粉15份,蔓越莓粉25份,雨生红球藻提取物3份,复合酵素8份,益生元6份。
所述益生元为菊粉、苏糖按重量比1:1组成。
所述复合酵素包括以下组分及其重量份数:蕨麻26份,芦荟18份,银耳10份,干酵母0.3份。
所述复合酵素的制备步骤如下:
将蕨麻、芦荟、银耳洗净切碎,高温灭菌后,置入无菌发酵罐中,并添加干酵母,密封存放于室温下30天后,经200目过滤,即得。
所述的含有花青素的组合物的制备方法,所述制备步骤如下:
S1、将越橘提取物,红石榴提取物,雨生红球藻提取物搅拌10min,混合均匀,得混合液;
S2、将针叶樱桃粉,蔓越莓粉,复合酵素和益生元加入到步骤S1所得的混合液中,200r/搅拌20min,即得。
对比例1一种含有花青素的组合物
所述的含有花青素的组合物,由如下组分及其重量份数组成:越橘提取物12份,红石榴提取物16份,针叶樱桃粉10份,蔓越莓粉18份,雨生红球藻提取物2份,益生元4份。
所述益生元为菊粉、低聚果糖、水苏糖按重量比1:5:3组成。
所述的含有花青素的组合物的制备方法与实施例2类似。
与实施例2的区别在于,所述的含有花青素的组合物中未添加复合酵素。
对比例2一种含有花青素的组合物
所述的含有花青素的组合物,由如下组分及其重量份数组成:红石榴提取物16份,针叶樱桃粉10份,蔓越莓粉18份,雨生红球藻提取物2份,复合酵素7份,益生元4份。
所述益生元为菊粉、低聚果糖、水苏糖按重量比1:5:3组成。
所述复合酵素包括以下组分及其重量份数:蕨麻21份,芦荟15份,银耳8份,干酵母0.2份。
所述复合酵素的制备步骤与实施例2类似。
所述的含有花青素的组合物的制备方法与实施例2类似。
与实施例2的区别在于,所述的含有花青素的组合物中未添加越橘提取物。
对比例3一种含有花青素的组合物
所述的含有花青素的组合物,由如下组分及其重量份数组成:越橘提取物12份,红石榴提取物16份,针叶樱桃粉10份,蔓越莓粉18份,雨生红球藻提取物2份,复合酵素7份,益生元4份。
所述益生元为菊粉、低聚果糖、水苏糖按重量比1:5:3组成。
所述复合酵素包括以下组分及其重量份数:芦荟15份,银耳8份,干酵母0.2份。
所述复合酵素的制备步骤与实施例2类似。
所述的含有花青素的组合物的制备方法与实施例2类似。
与实施例2的区别在于,所述复合酵素中未添加蕨麻。
试验例一、DPPH自由基清除能力试验
1.试验材料:实施例1~3,对比例1~3制备的含有花青素的组合物。
2.试验试剂:1,1-二苯基-2-三硝基苯肼(DPPH)、无水乙醇。
3.试验仪器:紫外可见光分光光度计、恒温箱。
4.试验方法:
(1)DPPH溶液的配制:称取8mg DPPH,用50%乙醇溶液溶解后转移至100ml容量瓶中,定容,摇匀,得到浓度为2.0×10-4mol/L的DPPH乙醇溶液,于4℃下保存备用。
(2)分别取实施例1~3、对比例1~3制备的含有花青素的组合物2mL放入试管中,并加入浓度为2.0×10-4mol/L的DPPH乙醇溶液,得到经配制的实施例1~3、对比例1~3待测试样,摇匀后置于暗处反应30min,然后测定其在517nm处的吸光度值(A1);用2.0mlDPPH·乙醇溶液加等体积的去离子水为空白对照样,测定其在517nm处的吸光度值(A0);分别用2.0ml各样品溶液加等体积的去离子水作为样品对照,测定其在517nm处的吸光度值(A2),并按下式计算自由基清除率:
自由基清除率=[A0-(A1-A2)/A0]×100%
每个待测试样测三组平行样,求平均值,结果如表1所示

由表1可知,实施例1~3制备的含有花青素的组合物的DPPH自由基清除率较高,均在80%以上,其中实施例2的DPPH自由基清除率最高,为本发明的最佳实施例,说明本发明制得的含有花青素的组合物具有较强的抗氧化、抗衰老的功效。而对比例1~3的制得的含有花青素的组合物对DPPH自由基的清除率较实施例1~3的明显降低,其中,对比例1(未添加复合酵素)和对比例2(未添加越橘提取物)制备的含有花青素的组合物对DPPH自由基的清除率分别为52.3%和46.8%,而对比例3中,当未复合酵素中未添加蕨麻时,制备的护肤基质的DPPH自由基清除率也有了明显的下降,对比例1~3中DPPH自由基清除率的下降说明了本发明提供的含有花青素的组合物中添加的复合酵素与其他组分相互协同作用,较好的维持了本发明的抗氧化性。
试验例二、羟自由基清除活性测定
1.试验材料:实施例1~3,对比例1~3制备的含有花青素的组合物。
2.试验方法:取0.75mmol/L的邻二氮菲溶液1ml于10ml具塞试管中,依次加入2ml0.2mmol/L,pH为7.5的PBS缓冲液、1ml0.75mmol/L的硫酸亚铁混合均匀,加入1ml样品溶液,混匀后加入1ml0.01%的过氧化氢,用去离子水定容,37℃水浴加热60min,在510nm出测定其吸光度值。
对羟自由基的清除率按下式计算:清除率=[(A1-A0)/(A2-A0)]×100%;
其中,A1为加入过氧化氢和样品时的吸光度;A0为加入过氧化氢但不加样品时的吸光度;A2为过氧化氢和样品均不加时的吸光度。此外,样品溶液的配置方法为:分别取100ml的实施例1~3、对比例1~3制备的含有花青素的组合物于6个烧杯中,于45℃微波加热60min,冷却至室温后得到本试验例所需的样品溶液。
3.试验结果
试验结果如表2所示。

由表2可知,实施例1~3制备的含有花青素的组合物,均在98%以上,其中实施例2的羟自由基清除率最高,达到了99.1%为本发明的最佳实施例,说明本发明制得的含有花青素的组合物具有较强的抗氧化功效。而对比例1~3的制得的含有花青素的组合物对羟自由基的清除率较实施例1~3的明显降低,对比例1~3中羟自由基清除率的下降说明了本发明提供的含有花青素的组合物中添加的复合酵素与其他组分相互协同作用,抗氧化性能优异,可广泛应用于护肤品和保健品中。
上述实施例仅例示性说明本发明的原理及其功效,而非用于限制本发明。任何熟悉此技术的人士皆可在不违背本发明的精神及范畴下,对上述实施例进行修饰或改变。因此,举凡所属技术领域中具有通常知识者在未脱离本发明所揭示的精神与技术思想下所完成的一切等效修饰或改变,仍应由本发明的权利要求所涵盖。
Claims (9)
1.一种含有花青素的组合物,其特征在于,包括如下组分及其重量份数:越橘提取物8~15份,红石榴提取物8~20份,针叶樱桃粉5~15份,蔓越莓粉10~25份,雨生红球藻提取物1~3份,复合酵素5~8份,益生元1~6份。
2.如权利要求1所述的含有花青素的组合物,其特征在于,由如下组分及其重量份数组成:越橘提取物12份,红石榴提取物16份,针叶樱桃粉10份,蔓越莓粉18份,雨生红球藻提取物2份,复合酵素7份,益生元4份。
3.如权利要求1或2所述的含有花青素的组合物,其特征在于,所述益生元为菊粉、低聚半乳糖、低聚果糖、水苏糖、膳食纤维的一种或几种。
4.如权利要求1或2所述的含有花青素的组合物,其特征在于,所述复合酵素包括以下组分及其重量份数:蕨麻15~26份,芦荟10~18份,银耳4~10份,干酵母0.1~0.3份。
5.如权利要求4所述的含有花青素的组合物,其特征在于,所述复合酵素由以下组分及其重量份数组成:蕨麻21份,芦荟15份,银耳8份,干酵母0.2份。
6.如权利要求4所述的含有花青素的组合物,其特征在于,所述复合酵素的制备步骤如下:
将蕨麻、芦荟、银耳洗净切碎,高温灭菌后,置入无菌发酵罐中,并添加干酵母,密封存放于室温下10~30天后,经200目过滤,即得。
7.一种如权利要求1-6任一项所述的含有花青素的组合物的制备方法,其特征在于,所述制备步骤如下:
S1、将越橘提取物,红石榴提取物,雨生红球藻提取物搅拌5~10min,混合均匀,得混合液;
S2、将针叶樱桃粉,蔓越莓粉,复合酵素和益生元加入到步骤S1所得的混合液中,120~200r/搅拌15~20min,即得。
8.一种如权利要求1-7任一项所述的含有花青素的组合物在制备抗氧化保健品和化妆品中的应用。
9.如权利要求8所述的应用,其特征在于,所述的含有花青素的组合物在抗氧化保健品和化妆品中的添加量为10~60%。
