Abstract
本发明公开了一种稳定性好的叶黄素纳米乳液及其制备方法,该叶黄素纳米乳液包括以下按质量份计算的组分制成:第一水相包括水相乳化剂9~40份、山梨糖醇5~15份和甘油90~146份;第二水相包括水相抗氧化剂0.1~3份、稳定剂0.1~3份和水10~61份;油相包括叶黄素4~10份、植物油30~50份、油相乳化剂0.1~1份和油相抗氧化剂4~8份。本发明通过调节配方以及高温瞬时油溶、高速剪切、高压均质的方法,制备出稳定性能好、粒径在200nm左右、乳化产率在85%以上的叶黄素纳米乳液。

技术领域
本发明涉及食品加工技术领域,更具体地,涉及一种稳定性好的叶黄素纳米乳液及其制备方法。
背景技术
叶黄素是优异的天然抗氧化剂和天然黄色素,普遍存在于蔬菜、水果和花卉等植物中,其中万寿菊是叶黄素含量最高的一种。叶黄素具有很高的营养价值和多种生理活性功能,被广泛应用于食品、保健品、化妆品、医药、烟草和动禽类饲料等多个领域,能够清除自由基起抗氧化作用,并在预防视黄斑退化、白内障、肿瘤、心血管疾病、动脉硬化、增强机体免疫力、视力保健等方面有着广泛的生物活性。我国在2007年将叶黄素作为了着色剂,可以在饮料、焙烤食品、冷冻食品、果冻和果酱类等食品中应用,同时也批准了作为营养强化剂可以添加到婴幼儿或儿童配方食品中。
叶黄素(lutein),属于类胡萝卜素的一种,分子式为C40H56O2,其基本结构是由两个不同的紫罗酮环与一个含十八碳原子的共轭双键的长链相联接而成,晶体结构呈菱格状,为鲜艳黄色,有金属光泽。叶黄素不溶于水,微溶于植物油,在脂肪族和芳香族的烃中有中等的溶解性,易溶于氯仿等。因多双键结构的存在,使得它的化学性质不稳定,在光、热、氧及酸性等条件下易发生氧化分解。
纳米乳液一般指平均粒径为50~500nm的乳液体系,是由水相、油相、表面活性剂及助表面活性剂按照合适的比例所形成的热力学稳定的胶体分散体系,由于其特有的物化特性,解决了油溶性功能成分水溶性差、易氧化、难吸收等问题,在食品药品等领域已引起广泛关注。中国专利CN108813616公开了一种制备叶黄素水溶性微乳液的方法,将水包油型乳化剂和油包水型乳化剂配合使用,但是由于加入乳化剂种类多,形成水包油、油包水乳剂的混合物,乳液的粒径未达到纳米级,叶黄素纳米乳液存在稳定性能差的问题。
发明内容
本发明要解决的技术问题是克服现有叶黄素纳米乳液稳定性差的缺陷和不足,提供一种稳定性好的叶黄素纳米乳液。
本发明的另一目的是提供一种稳定性好的叶黄素纳米乳液的制备方法。
本发明上述目的通过以下技术方案实现:
一种稳定性好的叶黄素纳米乳液,其特征在于,包括以下按质量份计算的组分制成:第一水相包括水相乳化剂9~40份、山梨糖醇5~15份和甘油90~146份;第二水相包括水相抗氧化剂0.1~3份、稳定剂0.1~3份和水10~61份;油相包括叶黄素4~10份、植物油30~50份、油相乳化剂0.1~1份和油相抗氧化剂4~8份。
本发明选用叶黄素为原料,油相抗氧化剂及乳化剂有利于减少高温对叶黄素的破坏,并且有助于形成稳定均匀的油相,并在油相和水相均加有乳化剂,油相选用亲油性的乳化剂,有助于形成均匀的油相,帮助叶黄素晶体在植物油中的溶解,水相选用亲水性的乳化剂,两种乳化剂均为小分子乳化剂,将非离子型乳化剂混合使用(如蔗糖脂肪酸酯和司盘80)可以发挥增效作用,能够有效降低界面张力而形成粒径小的颗粒,有效增加了乳液的稳定性。水相体系采用甘油和水结合的体系,以甘油为主,得益于其多羟基的结构,有利于乳化剂的溶解以便对油相进行包埋,有利于形成稳定的O/W乳化体系,但是甘油在常温下状态较稠,尤其在加入乳化剂之后,成品状态较稠使用不方便,为此进一步地在体系中加入水,以改变产品的状态,并加入山梨糖醇和稳定剂以起到保持水分的作用。
优选地,所述油相与第一水相和第二水相之和的总量为1:3~1:7。
优选地,所述水相乳化剂为聚甘油脂肪酸酯、蔗糖脂肪酸酯、改性大豆磷脂、酪蛋白酸钠、聚氧乙烯山梨醇酐单油酸酯(吐温80)中的一种或两种以上。
优选地,所述水相乳化剂的亲水亲油平衡值(HLB值)为10~18。
优选地,所述水相抗氧化剂为D-异抗坏血酸钠、抗坏血酸、抗坏血酸钠、和茶多酚、乙二胺四乙酸二钠、竹叶抗氧化物中的一种或两种以上。
优选地,所述稳定剂为焦磷酸钠、三聚磷酸钠、六偏磷酸钠、磷酸二氢钠中的一种或两种以上。
优选地,所述叶黄素和植物油的质量比为1:4~1:9。
优选地,所述叶黄素为纯度80%~100%的叶黄素晶体(HPLC方法测定)。
优选地,所述植物油为葵花籽油、玉米油、花生油、大豆油、米糠油、棕榈油、杏仁油、牛油果油、椰子油的一种或者两种以上。
优选地,所述油相乳化剂为单,双甘油脂肪酸酯(油酸、亚油酸、棕榈酸、山嵛酸、硬脂酸、月桂酸、亚麻酸)、氢化松香甘油酯、卵磷脂、山梨醇酐单油酸酯(司盘80)中的一种或两种以上。
优选地,所述油相乳化剂的HLB值为3~6。
优选地,所述油相抗氧化剂为维生素E、抗坏血酸棕榈酸酯、丁基羟基茴香醚、二丁基羟基甲苯、叔丁基对苯二酚、茶多酚棕榈酸酯、没食子酸丙酯中的一种或两种以上。
每种乳化剂都有特定的HLB值,单一乳化剂往往很难满足由多组分组成的体系的乳化要求,本发明将上述多种具有不同HLB值的水相和油相乳化剂混合使用,构成混合乳化剂,既可以满足本体系的要求,又可以增进乳化效果。
本发明保护上述叶黄素纳米乳液的制备方法,包括如下步骤:
S1.分别制得第一水相、第二水相和油相;
S2.将油相和第一水相混匀后进行剪切乳化,乳化时间2~6min,乳化转数为6000~10000转/分钟;再加入第二水相继续乳化,乳化时间为2~6min,乳化转数为6000~10000转/分钟,得到初乳液;
S3.将步骤S2制得的初乳液进行2次均质,第一次均质压力为30~50Mpa,时间为3~6min,第二次均质压力为50~100Mpa,时间为3~6min,制得叶黄素纳米乳液。
优选地,步骤S1所述第一水相的制备方法为:
将水相乳化剂、山梨糖醇、甘油混合,于65~80℃加热溶解至无固体留存,制得第一水相。
优选地,步骤S1所述第二水相的制备方法为:
将水相抗氧化剂、稳定剂、水混合并搅拌溶解至无固体留存,制得第二水相。
优选地,步骤S1所述油相的具体制备方法为:
将叶黄素、植物油搅匀,搅拌速度为4000~8000转/分钟,搅拌时间为1~3小时,得到叶黄素油悬液;将叶黄素油悬液、油相抗氧化剂、油相乳化剂混合,在惰性气体的保护下,于160~185℃条件下加热溶解,加热时间为2~5min,制得油相。
优选地,所述加热溶解的操作步骤为:
按叶黄素油悬液、油相抗氧化剂、油相乳化剂的顺序加入反应釜中,升温至160~185℃,并维持2~5min,加热反应结束后冷却至70~90℃,制得油相。
油相原料通过一个反应釜来实现高温瞬时油溶,反应釜带有外夹层,夹层通导热油,导热油升温至200~250℃后,将油相原料按叶黄素油悬液、油相抗氧化剂、油相乳化剂顺序加入反应釜中,升温至160~185℃,并维持2~5min,利用反应釜导热油外循环冷却系统将所述油相的温度进行控制,加热反应结束后冷却至70℃~90℃,保温,制得油相。为减少加热过程中氧气对叶黄素的氧化作用,设备装有充气装置,整个加热过程在惰性气体的保护下进行,有效地减少了叶黄素的损耗,惰性气体优选为氮气。
所述步骤S2分为第一水相和第二水相先后和油相进行高速剪切乳化,进一步地,为减少高速剪切导致的高温对叶黄素的破坏,在搅拌罐外加装夹层,通入冷却介质进行降温,优选地,温度控制在70~90℃,冷却介质优选为冷凝水。
所述步骤S3中,采用2次高压均质机进行均质,进一步地,为减少均质产生的高温对叶黄素的损耗,均质机加装有水冷式冷却装置,冷却器介质由进水口经分水盖进入一半冷却器管之后,再从回水盖流入另一半冷却器管进入另一侧分水盖及出水管。冷介质在双管程流过程中,吸收放出的余热由出水口排出,保持较低温度,控制在70℃~90℃。经过高压均质之后,乳液的颗粒大小达到纳米级,制备得到稳定性良好的叶黄素纳米乳液。

本发明公开了一种稳定性好的叶黄素纳米乳液,包括按一定配比的组分:第一水相为水相乳化剂、山梨糖醇、甘油;第二水相为水相抗氧化剂、稳定剂和水;油相为水相抗氧化剂、稳定剂和水,本发明通过调节配方,采用高温瞬时油溶、高速剪切、高压均质的方法制备叶黄素纳米乳液,缩短了反应时间,提高了原料利用率,并且制备出稳定性能好、粒径在200nm左右、乳化产率在85%以上的叶黄素纳米乳液,可以实现工业化。

具体实施方式
下面结合具体实施方式对本发明作进一步的说明,但实施例并不对本发明做任何形式的限定。除非另有说明,本发明实施例采用的原料试剂为常规购买的原料试剂。
本发明提供了一种稳定性好的叶黄素纳米乳液及其制备方法,以下实施例具体举例说明。
实施例1
一种稳定性好的叶黄素纳米乳液,包括以下按质量份计算的组分:第一水相为水相乳化剂(蔗糖脂肪酸酯14份、聚甘油脂肪酸酯5份)、山梨糖醇10份、甘油124.2份;
第二水相为水相抗氧化剂(D-异抗坏血酸钠0.3份)、稳定剂(六偏磷酸钠0.3份)、水40份;
油相为叶黄素晶体5份、葵花籽油35份、油相乳化剂(单,双甘油脂肪酸酯0.2份)、油相抗氧化剂(二丁基羟基甲苯3份、叔丁基对苯二酚3份)。
上述叶黄素纳米乳液的制备方法,包括如下步骤:
S1.将水相乳化剂、山梨糖醇、甘油混合置于70℃水浴条件下加热溶解至无固体留存,得到第一水相,并于70℃条件下保温;
将水相抗氧化剂、稳定剂、水混合并搅拌溶解至无固体留存,得到第二水相;
将叶黄素晶体、葵花籽油分别加入到搅拌罐中,搅拌2小时,搅拌速度为6000转/分钟,得到叶黄素油悬液;将叶黄素油悬液、油相抗氧化剂、油相乳化剂混合,在惰性气体的保护下,于180℃条件下加热溶解,加热时间为3.5min,得到油相;
S2.将油相和第一水相混合进行高速剪切乳化,乳化时间为5min,乳化转数为6000转/分钟;接着加入第二水相继续乳化,乳化时间为5min,乳化转数为6000转/分钟,得到初乳液;
S3.将上述初乳液进行2次高压均质,第一次均质压力为45Mpa,时间为5min,第二次均质压力为75Mpa,时间为5min,制备得到叶黄素纳米乳液。
实施例2
一种稳定性好的叶黄素纳米乳液,包括以下按质量份计算的组分:第一水相为水相乳化剂(蔗糖脂肪酸酯14份、酪蛋白酸钠10份)、山梨糖醇10份甘油108.9份;
第二水相为水相抗氧化剂(抗坏血酸0.5份)、稳定剂(焦磷酸钠0.5份)水50份;
油相为叶黄素5份、植物油(玉米油35份)、油相乳化剂(氢化松香甘油酯0.1份)、油相抗氧化剂(抗坏血酸棕榈酸酯6份)。
本实施例的制备方法与实施例1相同。
实施例3
一种稳定性好的叶黄素纳米乳液,包括以下按质量份计算的组分:第一水相为水相乳化剂(蔗糖脂肪酸酯20份、改性大豆磷酯5份)、山梨糖醇10份、甘油113.8份;
第二水相为水相抗氧化剂(抗坏血酸钠0.5份)、稳定剂(三聚磷酸钠0.5份)、水45份;
油相为叶黄素5份、植物油(花生油35份)、油相乳化剂(山梨醇酐单油酸酯0.2份)、油相抗氧化剂(丁基羟基茴香醚2.5份、二丁基羟基甲苯2.5份)。
本实施例的制备方法与实施例1相同。
实施例4
一种稳定性好的叶黄素纳米乳液,包括以下按质量份计算的组分:第一水相为水相乳化剂(聚氧乙烯山梨醇酐单油酸酯15份、聚甘油脂肪酸酯6份)、山梨糖醇10份、甘油119.2份;
第二水相为水相抗氧化剂(乙二胺四乙酸二钠0.8份)、稳定剂(磷酸二氢钠0.8份)、水40份;
油相为叶黄素5份、植物油(大豆油35份)、油相乳化剂(卵磷脂0.2份)、油相抗氧化剂(茶多酚棕榈酸酯4份、没食子酸丙酯4份)。
本实施例的制备方法与实施例1相同。
实施例5
一种稳定性好的叶黄素纳米乳液,包括以下按质量份计算的组分:第一水相为水相乳化剂(蔗糖脂肪酸酯12份、聚氧乙烯山梨醇酐单油酸酯12份)、山梨糖醇10份、甘油105.4份;
第二水相为水相抗氧化剂(茶多酚0.5份、竹叶抗氧化剂0.5份)、稳定剂(焦磷酸钠0.5份)、水53份;
油相为叶黄素5份、植物油(棕榈油35份)、油相乳化剂(氢化松香甘油酯0.1份)、油相抗氧化剂(抗坏血酸棕榈酸酯3份、叔丁基对苯二酚3份)。
本实施例的制备方法与实施例1相同。
实施例6
一种稳定性好的叶黄素纳米乳液,包括以下按质量份计算的组分:第一水相为水相乳化剂(酪蛋白酸钠12份、改性大豆磷酯12份)、山梨糖醇10份、甘油108.8份;
第二水相为水相抗氧化剂(抗坏血酸0.25份、抗坏血酸钠0.25份)、稳定剂(六偏磷酸钠0.5份)、水50份;
油相为叶黄素5份、植物油(大豆油35份)、油相乳化剂(单,双甘油脂肪酸酯0.1份、氢化松香甘油酯0.1份)、油相抗氧化剂(抗坏血酸棕榈酸酯3份、二丁基羟基甲苯3份)。
本实施例的制备方法与实施例1相同。
实施例7
一种稳定性好的叶黄素纳米乳液,包括与实施例1相同的组分。
上述叶黄素纳米乳液的制备方法,包括如下步骤:
S1.将水相乳化剂(蔗糖脂肪酸酯、聚甘油脂肪酸酯)、山梨糖醇、甘油混合置于75℃水浴条件下加热溶解至无固体留存,得到第一水相,并于75℃条件下保温;
将水相抗氧化剂(D-异抗坏血酸钠)、稳定剂(六偏磷酸钠)、水混合并搅拌溶解至无固体留存,得到第二水相;
将叶黄素晶体、葵花籽油分别加入到搅拌罐中,搅拌2小时,搅拌速度为8000转/分钟,得到叶黄素油悬液;将叶黄素油悬液、油相抗氧化剂(二丁基羟基甲苯、叔丁基对苯二酚)、油相乳化剂(单,双甘油脂肪酸酯)混合,在惰性气体的保护下,于183℃条件下加热溶解,优选地加热时间为3min,得到油相;
S2.将油相和第一水相混合进行高速剪切乳化,乳化时间为4min,乳化转数为8000转/分钟;接着加入第二水相继续乳化,乳化时间为4min,乳化转数为8000转/分钟,得到初乳液;
S3.将上述初乳液进行2次高压均质,第一次均质压力为45Mpa,时间为4min,第二次均质压力为80Mpa,时间为4min,制备得到叶黄素纳米乳液。
对比例1
本对比例包括以下按质量份计算的组分:
油相:叶黄素晶体5份、植物油(椰子油35份)、油相乳化剂(卵磷脂0.2份)、油相抗氧化剂(维生素E 8份);
水相:甘油113.8份、山梨糖醇10份、水相乳化剂(酪蛋白酸钠20份)、水45份、水相抗氧化剂(竹叶抗氧化物1.5份)、稳定剂(磷酸二氢钠1.5份)。
本对比例的制备方法为:
S1.将水相乳化剂、山梨糖醇、甘油混合置于70℃水浴条件下加热溶解至无固体留存,另外将水相抗氧化剂、稳定剂加入水中混合并搅拌溶解至无固体留存,然后将两者混合搅拌均匀,制得水相;
S2.将叶黄素晶体、椰子油分别加入到搅拌罐中,搅拌2小时,搅拌速度为6000转/分钟,得到叶黄素油悬液;将叶黄素油悬液、油相抗氧化剂、油相乳化剂混合,在惰性气体的保护下,于180℃条件下加热溶解,加热时间为3.5min,制得油相;
S3.将油相和水相混合进行高速剪切乳化,乳化时间为10min,乳化转数为6000转/分钟,得到初乳液;
S4.将上述初乳液进行2次高压均质,第一次均质压力为45Mpa,时间为5min,第二次均质压力为75Mpa,时间为5min,制备得到叶黄素纳米乳液。
对比例2
本对比例包括以下按质量份计算的组分:
油相:叶黄素晶体5份、植物油(米糠油35份)、油相乳化剂(山梨醇酐单油酸酯0.3份)、油相抗氧化剂(茶多酚棕榈酸酯8份);
水相:甘油105.7份、山梨糖醇10份、水相乳化剂(改性大豆磷脂22份)、水51份、水相抗氧化剂(茶多酚2份)、稳定剂(三聚磷酸钠1份)。
本对比例的制备方法与对比例1相同。
性能测试
为更好地验证本发明制备的叶黄素纳米乳液的乳化产率和粒径,对实施例及对比例制得的叶黄素纳米乳液进行了检测,检测结果如下:
(1)乳化产率的测定
叶黄素乳液乳化产率是指被乳化剂包埋的叶黄素含量占配方中加入的叶黄素总含量的比例。乳液中叶黄素总含量的测定:取稀释一定倍数的叶黄素乳液1mL,加入正己烷,超声震荡,静置取上层萃取液,重复3次,合并萃取液。取萃取液定容,在452nm下测定吸光值,根据标准曲线计算总含量A。乳液中未被包埋的叶黄素含量的测定:取1mL乳液加入3mL正己烷,振荡30s。混合均匀离心5min,取上清液用正己烷定容,在452nm下测定吸光值,根据标准曲线计算含量B。
乳化产率(%)=(叶黄素总含量A-未被包埋的叶黄素含量B)/叶黄素总含量A。检测结果见表1。
(2)粒径的测定
测定方法:采用激光粒度仪(英国马尔文仪器公司生产,型号ZetasizerNano ZS)测量乳液的粒径大小,测定前应将不同条件下制备的乳液分别用水稀释一定的倍数,考虑到PH和离子强度对乳液稳定性的影响,稀释用水的PH和离子强应和乳液样品一致。激光粒度仪的测定波长为658nm,散射角度为133°,测定时温度维持在25±1℃,平衡时间2min。
实施例1~7和对比例1~2叶黄素纳米乳液的平均粒度及粒径分布的测定结果见表1。

通过上述实验结果可以发现,选择的乳化剂、抗氧化剂、稳定剂以及乳化均质条件对乳液的稳定性有影响。本发明制备的叶黄素纳米乳液的乳化产率均在85%以上,颗粒的大小在200nm左右,达到纳米级的要求。
在光照稳定性方面,在露天光照情况下,放置90天,叶黄素的损耗率在20%左右,而对比例的叶黄素的损耗率明显较高(见表2),实施例的叶黄素纳米乳液抗光照性能优良,说明得到叶黄素纳米乳液均匀、稳定,可见图1,能够有效地将叶黄素应用到食品工业各领域。

Claims (10)
1.一种稳定性好的叶黄素纳米乳液,其特征在于,包括以下按质量份计算的组分制成:第一水相包括水相乳化剂9~40份、山梨糖醇5~15份和甘油90~146份;第二水相包括水相抗氧化剂0.1~3份、稳定剂0.1~3份和水10~61份;油相包括叶黄素4~10份、植物油30~50份、油相乳化剂0.1~1份和油相抗氧化剂4~8份。
2.根据权利要求1所述的叶黄素纳米乳液,其特征在于,所述油相与第一水相和第二水相之和的质量比为1:3~1:7。
3.根据权利要求1所述的叶黄素纳米乳液,其特征在于,所述稳定剂为焦磷酸钠、三聚磷酸钠、六偏磷酸钠、磷酸二氢钠中的一种或两种以上。
4.根据权利要求1所述的叶黄素纳米乳液,其特征在于,所述叶黄素和植物油的质量比为1:4~1:9。
5.根据权利要求1所述的叶黄素纳米乳液,其特征在于,所述植物油为葵花籽油、玉米油、花生油、大豆油、米糠油、棕榈油、杏仁油、牛油果油、椰子油的一种或者两种以上。
6.权利要求1~5任一项所述叶黄素纳米乳液的制备方法,其特征在于,包括如下步骤:
S1.分别制得第一水相、第二水相和油相;
S2.将油相和第一水相混匀后进行剪切乳化,乳化时间2~6min,乳化转数为6000~10000转/分钟;再加入第二水相继续乳化,乳化时间为2~6min,乳化转数为6000~10000转/分钟,得到初乳液;
S3.将步骤S2制得的初乳液进行2次均质,第一次均质压力为30~50Mpa,时间为3~6min,第二次均质压力为50~100Mpa,时间为3~6min,制得叶黄素纳米乳液。
7.根据权利要求6所述的制备方法,其特征在于,步骤S1所述第一水相的制备方法为:
将水相乳化剂、山梨糖醇、甘油混合,于65~80℃加热溶解至无固体留存,制得第一水相。
8.根据权利要求6所述的制备方法,其特征在于,步骤S1所述第二水相的制备方法为:
将水相抗氧化剂、稳定剂、水混匀并溶解至无固体留存,制得第二水相。
9.根据权利要求6所述的制备方法,其特征在于,步骤S1所述油相的制备方法为:
将叶黄素、植物油搅匀,搅拌速度为4000~8000转/分钟,搅拌时间为1~3小时,得到叶黄素油悬液;将叶黄素油悬液、油相抗氧化剂、油相乳化剂混合,在惰性气体的保护下,于160~185℃条件下加热溶解,加热时间为2~5min,制得油相。
10.根据权利要求9所述的制备方法,其特征在于,所述加热溶解的操作步骤为:
按叶黄素油悬液、油相抗氧化剂、油相乳化剂的顺序加入反应釜中,升温至160~185℃,并维持2~5min,加热反应结束后冷却至70~90℃,制得油相。
