Abstract
The invention discloses a micromolecule polypeptide youth essence which comprises the following components: small molecular polypeptide mixed liquor, oligopeptide mixed liquor, deep sea collagen powder, glucan, a witch hazel extract and an auxiliary agent; the mixed solution of the small molecular polypeptide consists of tetrapeptide-30, palmitoyl pentapeptide-4, tripeptide-1 copper, nonapeptide-1, dipeptide-2, tetrapeptide-7 and tetrapeptide-15; the oligopeptide mixed solution is composed of oligopeptide-1, oligopeptide-3, oligopeptide-4 and oligopeptide-5. The invention carefully compounds a plurality of small molecular polypeptide, deep-sea collagen powder, glucan and witch hazel extract to prepare the small molecular polypeptide youth essence, has an obvious promoting effect on the melanin inhibition of facial skin, and has the effects of removing freckles, whitening, moisturizing, increasing skin elasticity, resisting wrinkles and tightening skin.
Description
Technical Field
The invention relates to the technical field of cosmetics, and particularly relates to micromolecule polypeptide youth essence and a preparation method thereof.
Background
According to the skin, white skin and ruddy luster of oriental women, the facial mask is fashion advocated and pursued by women, and then functional essence products such as whitening, moisturizing or antioxidation and the like emerge in the market endlessly. In the past decades, people mostly use physical shielding technologies such as whitening powder and the like to achieve the effect of surface modification, and the pertinence is strong, but the change of one skin state is often accompanied with the appearance of other skin problems, such as skin dryness, which easily causes the problems of dull and lusterless skin, skin senilism, pigment deposition and the like.
The existing essence products in the market are mainly added with various chemical components and natural plant components, so that certain whitening and skin-care effects are achieved, but the actual using effect is not ideal. In addition, many essence products can be added with irritant harmful substances such as preservatives and the like, and the skin can be remained after long-term use, so that certain side effects can be generated on the skin, not only an ideal skin care effect cannot be obtained, but also the skin can be damaged in serious cases, the normal physiological metabolism of the skin is influenced, and the aging of the skin is accelerated. Although some essential product formulas adopt plant Chinese herbal medicine components, active ingredients are difficult to be absorbed by body skin and are easy to be covered by other substances, and the due effect is difficult to achieve.
The small molecular polypeptide is a biochemical substance between amino acid and protein, has a molecular weight smaller than that of protein and larger than that of amino acid, and is a fragment of protein. Two or more amino acids are linked by peptide bonds, and the resulting amino acid chain is called a peptide. Peptides consisting of 2 to 15 amino acids are called small peptides or small peptides. Among them, peptides consisting of 2 to 9 amino acids are called oligopeptides, and peptides consisting of 10 to 15 amino acids or more are called polypeptides.
Chinese patent CN108785186A discloses a polypeptide skin-beautifying youth essence which comprises the following components in parts by weight: 70g of water, 0.02g of EDTA disodium, 5g of glycerol, 3g of propylene glycol, 0.08g of xanthan gum, 0.2g of allantoin, 1g of hyaluronic acid, 0.1g of nipagin methyl ester, 0.5g of D-panthenol, 0.2g of oat B-glucan, 0.1g of HPS0, 0.02g of incense, 0.1g of MTI0, 1g of centella asiatica extract, 0.1g of snake venom peptide, 80.1g of hexapeptide, 120.1g of decapeptide, 30.1g of oligopeptide, 0.1g of migpeptide and 1g of purslane extract. The invention can slow down the aging process of skin, help to moisten skin, promote collagen synthesis and skin regeneration, improve skin texture and appearance, enable skin to be elastic and reduce wrinkles; meanwhile, the water retention property of the skin is increased, so that the effects of moisturizing and moistening the skin are achieved, and the skin is protected from being damaged. There is a need for further development of youth essence products containing small molecule polypeptides.
Disclosure of Invention
The invention aims to provide a micromolecular polypeptide youth essence and a preparation method thereof, so as to solve the defects in the background technology.
The invention is realized by the following technical scheme:
a micromolecule polypeptide youth essence comprises the following components: small molecular polypeptide mixed liquor, oligopeptide mixed liquor, deep sea collagen powder, glucan, a witch hazel extract and an auxiliary agent; the mixed solution of the small molecular polypeptide consists of tetrapeptide-30, palmitoyl pentapeptide-4, tripeptide-1 copper, nonapeptide-1, dipeptide-2, tetrapeptide-7 and tetrapeptide-15; the oligopeptide mixed solution is composed of oligopeptide-1, oligopeptide-3, oligopeptide-4 and oligopeptide-5.
The auxiliary agent is a common component in a small molecular polypeptide youth essence formula, and comprises but is not limited to a gel thickening agent, a sterilization preservative, a pH regulator, base oil, an emulsifier and water.
The collagen can effectively accelerate the growth of cells in the dermis, activate epidermal cells, maintain the elasticity and firmness of the skin, prevent wrinkles from appearing, and improve or even remove the wrinkles generated by skin aging. However, the collagen extracted from terrestrial animals can activate the activity of the collagen most efficiently at the temperature of more than 35 ℃ to achieve the ideal effect. The human body surface temperature is lower than 35 ℃, and the activity of collagen cannot be well excited. The inventor finds that the activity of collagen extracted from deep-sea fish is only activated at a temperature of more than 10 ℃, and the deep-sea collagen powder added into the small-molecular polypeptide youth extract can fully activate and utilize the activity of the collagen, thereby achieving the best effect.
Glucan, also known as dextran, is a polysaccharide present in the mucus secreted by some microorganisms during growth, has a relatively high molecular weight and is mainly linked by D-glucopyranose in α, 1 → 6 bonds, with branching points of 1 → 2, 1 → 3, 1 → 4, preferably, but not limited to, β -glucan, preferably β -1, 3-glucan, β -glucan enables lymphocytes of injured organisms to rapidly recover the ability to produce cytokines (IL-1), effectively regulating the immune function of the organisms, β -1, 3-glucan has been identified as a safe substance by FDA in the United states.
In the above minor molecule polypeptide youth essence component, the gel thickener is selected from carbomer 940 or 941; the sterilizing preservative is methyl p-hydroxybenzoate; the pH regulator is preferably, but not limited to, disodium ethylenediaminetetraacetate (EDTA-2Na), potassium hydroxide; the base oil is preferably, but not limited to, white oil, silicone oil, glycerin; the emulsifier is preferably but not limited to organic olive oil emulsified wax, glyceryl stearate, fatty alcohol polyoxyethylene ether, and white beeswax.
As a preferred technical scheme, the micromolecule polypeptide youth essence comprises the following components in percentage by weight:
the balance of water, and the sum of the percentage contents of all the components is 100%.
Preferably, the formula of the micromolecule polypeptide youth essence also contains a proper amount of vitamins.
Preferably, the weight ratio of each component in the oligopeptide mixed solution is as follows: oligopeptide-1: oligopeptide-3: oligopeptide-4: oligopeptide-5 is 1: 2: 1: 2.

The tetrapeptide-30, also called as skin-brightening peptide, has the effects of whitening skin, removing freckles, inhibiting excessive melanin generation and reducing melanin deposition, has the action mechanism of mainly reducing the number of tyrosinase, inhibiting MSH (melanocyte stimulating hormone), reducing melanin generation and preventing excessive melanin generation so as to brighten the skin, and is suitable for the skin with various complexion.
Palmitoyl pentapeptide-4 can penetrate dermis to increase collagen, reverse skin aging process by reconstruction from inside to outside, stimulate proliferation of collagen, elastic fiber and hyaluronic acid, improve skin water content and water locking degree, increase skin thickness, and reduce fine lines.
Tripeptide-1 copper is also known as copper trisp peptide, copper blue peptide. The copper peptide and the copper compound thereof can be used as an activator and an antioxidant for tissue remodeling, and the copper peptide is also a signal peptide, so that the degradation of a large number of collagen aggregates outside the scar, the synthesis of normal collagen of the skin, the generation of elastin, proteoglycan and glucosaminan, the growth speed and migration of different cell types, inflammation resistance and antioxidant reaction are promoted.
The first step in melanin formation is the binding of melanocyte stimulating hormone to the MC1 receptor on melanocytes, which then activates tyrosinase to further form melanin. Nonapeptide-1 is a biomimetic peptide, has good matching property with MC1 receptor, and can be used as an antagonist of melanocyte stimulating hormone to prevent tyrosinase from being further activated, thereby reducing and avoiding the formation of melanin.

1. heating the phase A to 75-85 ℃ and completely dissolving;
2. cooling to 45-50 deg.C, adding the material of phase B into phase A, and mixing under vacuum;
3. cooling to 40-45 deg.C, adding phase C, and mixing under vacuum;
4. cooling to 35-38 deg.C, adding pH regulator until pH is 6.0-7.0, and discharging after qualified inspection;
the phase A comprises: water, emulsifier, gel thickener, base oil;
the phase B comprises: sterilizing and preserving agents;
the phase C comprises: small molecular polypeptide mixed liquor, oligopeptide mixed liquor, deep sea collagen powder, glucan and witch hazel extract.
The invention carefully compounds a plurality of small molecular polypeptide, deep-sea collagen powder, glucan and witch hazel extract to prepare the small molecular polypeptide youth essence, has an obvious promoting effect on the melanin inhibition of facial skin, and has the effects of removing freckles, whitening, moisturizing, increasing skin elasticity, resisting wrinkles and tightening skin.
Detailed Description
The invention is illustrated below by means of specific examples, without being restricted thereto. All the raw materials are conventional commercial products unless otherwise specified.
Example 1

The polypeptide skin-beautifying youth essence disclosed by Chinese patent CN108785186A comprises the following components in parts by weight: 70g of water, 0.02g of EDTA disodium, 5g of glycerol, 3g of propylene glycol, 0.08g of xanthan gum, 0.2g of allantoin, 1g of hyaluronic acid, 0.1g of nipagin methyl ester, 0.5g of D-panthenol, 0.2g of oat B-glucan, 0.1g of HPS0, 0.02g of incense, 0.1g of MTI0, 1g of centella asiatica extract, 0.1g of snake venom peptide, 80.1g of hexapeptide, 120.1g of decapeptide, 30.1g of oligopeptide, 0.1g of migpeptide and 1g of purslane extract.
Test example
1. Skin melanin content test:
the skin melanin test probe was used: mexametrmx 18, for testing the content of skin melanin. Based on the principle of spectral absorption (RGB), the amount of melanin in the skin is determined by measuring the amount of reflection of light of a particular wavelength on human skin. The emitter of the instrument probe emits light with three wavelengths to the skin surface, and the receiver detects the light reflected by the skin. Since the amount of emitted light is constant, the amount of light absorbed by the skin and thus the amount of melanin in the skin can be measured.
5 testees with darker facial complexion are selected. The face of the testee can not use any product within 3 days before the test, 2cm x 2m in the middle of the cheek is selected as a test area, any 5 points in the area are selected for measurement, and the average value is obtained and recorded. The inventive example 1 sample was used for the left face and the comparative sample was used for the right face. The first measurement was made before using the sample and recorded as the initial value. After the test is started, the face pack is used once in the morning and evening every day, a proper amount of samples are uniformly smeared around the face, the samples are guaranteed to be smeared on a test area and fully absorbed, and the face pack is tested once every full week of use for four weeks. The test results are shown in table 1:

From the data, the whitening effect of the small-molecule polypeptide youth essence of the example 1 is slightly inferior to that of the comparative small-molecule polypeptide youth essence in the first two weeks, but the effect is obviously improved in the third week and is obviously improved in the fourth week. Therefore, the micromolecule polypeptide youth essence has a remarkable long-acting improvement effect on the inhibition of the melanin of the facial skin.
2. Anti-wrinkle test
The value of the skin elasticity index R7 was measured using a skin elasticity meter Cotomermapa 580 from CK, Germany (constant negative pressure of 450mbar during the test).
30 tested persons with obvious forehead wrinkles are selected and are all women in the age of 35-45 years, and 10 persons are randomly divided into 1 group and 3 groups. The forehead of the testee can not use any product within 3 days before the test, the forehead skin is selected as a test area, any 5 points in the test area are selected for measurement, and the average value is obtained and recorded. In which the group # 1 and the group # 2 used the sample of example 1 of the present invention and the group # 3 used the sample of comparative example. The first measurement was made before using the sample and recorded as the initial value. After the test is started, the liquid is used once in the morning and evening every day, a proper amount of samples are uniformly smeared around the forehead, the liquid is guaranteed to be smeared on a test area and be fully absorbed, and the liquid is tested once every week after the liquid is used, and is used for four weeks. Specification of indexes: the closer the skin elasticity index R7 is to 1, the better the skin elasticity is. Each set of data from each test was averaged after removing one maximum and one minimum, and the test results are shown in table 2:

Test results show that the skin elasticity is obviously increased after the small molecular polypeptide youth essence is used for two weeks, and the R2 index is close to 1 after the fourth week, which shows that the skin elasticity of a tested person is greatly improved, and the effect is obviously better than that of a comparative example. The micromolecule polypeptide youth essence has the effects of increasing skin elasticity, resisting wrinkles and tightening the skin.
3. Speckle removing effect test
In order to test the freckle removing effect described in each of the examples and comparative examples of the present invention, 40 subjects were selected, randomly divided into four groups, and the youth essence or the comparative sample test article was used each day, wherein the double-blind test score test was performed using the sample of example 1 for group 1, the sample of example 2 for group 2, the sample of example 3 for group 3, and the sample of comparative example for group 4; the subjects were aged 23-50 years, during the trial period, the trial site was deprived of any drug, and the subjects continued to use the test product for one month. The skin color and the number of the color spots on the cheeks of the test subjects were observed, the initial photograph and the used photograph were compared and scored, the filling score was 100%, and the average score was obtained, and the results are shown in table 3.

2. The small molecule polypeptide youth essence of claim 1, wherein the auxiliary agents comprise a gel thickener, a bactericidal preservative, a PH regulator, a base oil, an emulsifier and water.
3. The small molecule polypeptide youth essence of claim 1, wherein the glucan is β -1, 3-glucan.
4. The small molecule polypeptide serum of claim 2, wherein the gel thickening agent is selected from carbomers 940 or 941; the sterilizing preservative is methyl p-hydroxybenzoate; the pH regulator is disodium ethylene diamine tetraacetate; the base oil is selected from white oil, silicone oil, and glycerol.
5. The small molecule polypeptide youth essence according to claim 2, wherein the emulsifier is selected from organic olive oil emulsified wax, glyceryl stearate, fatty alcohol polyoxyethylene ether and white beeswax.
7. The micromolecule polypeptide youth essence according to claim 1, wherein the weight ratio of each component in the oligopeptide mixed solution is as follows: oligopeptide-1: oligopeptide-3: oligopeptide-4: oligopeptide-5 is 1: 2: 1: 2.
10. The method for preparing the minor molecule polypeptide essence of any one of claims 1 to 9, which comprises the following steps:
1. heating the phase A to 75-85 ℃ and completely dissolving;
2. cooling to 45-50 deg.C, adding the material of phase B into phase A, and mixing under vacuum;
