Skin moisture test
The moisture content of the skin is determined by two factors, namely internal and external factors, the moisture retention capacity of the horny layer of the skin is greatly changed, the moisture content can be changed from 10-60%, and the moisture content mainly comprises the breathing process of skin sweating and the water mixture in the skin. External factors including ambient temperature, humidity, drugs, cosmetics, etc., can determine and change the moisture content of the skin.
The moisture test adopts a worldwide recognized CORNEOMETER-capacitance method, the moisture test principle is based on that the dielectric constant of water and other substances is greatly changed, according to the difference of moisture content, a measuring capacitor with a proper shape can change along with the change of the capacitance of the skin, and the capacitance of the skin is in a measuring range, so that the moisture content of the skin can be measured. The result is expressed by a set humidity Measurement Value (MMV). The MMV is a value of 0-150, the capacitance measuring method is superior to other methods, and because the tested skin is not in unnatural contact with the testing probe and almost no current passes through the tested skin, the testing result is not influenced by polarization effect and ionic conductivity. The instrument probe and the water in the skin have no inertia in the process of establishing balance, and can realize quick measurement, thereby eliminating the influence of active skin on the measurement result.
The measuring process of the skin hydration rate MMV is greatly influenced by the ambient temperature and humidity. The best effect is generally selected when the measurement is carried out in an environment with 20 ℃ and 50% relative humidity.
When measuring, the moisture test probe is vertically pressed on the surface of the skin to be measured, the top of the probe is pressed back for a certain distance, a spring is arranged in the probe to keep the top of the probe to be pressed on the surface of the skin under the pressure of 0.16N, the result is displayed on the host within one second, and prompt sound is given.
The test substance and the tested part of each group are randomly numbered, and the comparison sequence of the test substance and the tested part is determined. Before the test, the subjects need to uniformly wash the inner sides of the forearms of both hands with clean water. Measurement marks were made on the inner forearm of the subject after washing. In this experiment, the left and right forearms were marked with two test areas. Each area is coated with a test substance at intervals of 1cm, and each test area is 5 × 5cm2The amount of the test sample was 0.2 g. After the subject was rested in a constant environment for 30min, the test site blank value was measured using a Corneometer, and 5 points were fixedly measured for each zone in a certain order to obtain an average value thereof. The specialist then takes care of applying the sample and starts timing. According to experimentsThe design measures the change in MMV value at each time separately. And subtracting the blank value from the average value detected each time to obtain the change of the MMV value in the time period, and obtaining the increase rate of the MMV value after removing the blank value. The results are shown in Table 1.
The skin moisture content increase rate calculation formula is as follows:
skin moisture content increase rate%t-MMV0)/MMV0×100%,
In the formula: MMV0-MMV of the skin before application; MMVtRefers to MMV of the skin t time after application.

And (3) whitening test: tyrosinase inhibition assay
The existing evaluation methods of the efficacy of the whitening agent mainly comprise a tyrosinase inhibition method, a cell biology method, an animal experiment method and a human body experiment method. Tyrosinase is a rate-limiting enzyme in a melanin synthesis pathway, and mainly influences the generation of melanin by influencing the conversion of tyrosine into dopa and the oxidation of dopa into dopaquinone, and a tyrosinase inhibition method is usually used for determining the inhibition rate of a whitening agent on the activity of tyrosinase, has short determination time, is simple and convenient to operate, requires low cost, and is suitable for large-flux screening of the whitening agent. Therefore, the product whitening test in the patent adopts a tyrosinase inhibition method for determination.

Hydroxyl radical scavenging assay
Skin aging is an extremely complex process, namely, is part of endogenous body aging and is influenced by exogenous photothermal radiation, life style, diet, emotion, hormone level and other multiple factors. In the process of skin aging, damage caused by active oxygen plays an important role in skin aging. When the skin is directly exposed to sunlight, cosmic rays, air and physicochemical polluting species, it can generate free radicals and accelerate skin aging. This example tested the effect of scavenging hydroxyl radicals of 7 masks prepared with the formulations of examples 1-5 and comparative examples 1 and 2.
Phenanthroline and Fe2+A complex is formed, which has a maximum absorption peak at 510nm and is a commonly used redox indicator, and the color change of the complex can sensitively reflect the change of the redox state of the solution. H2O2/Fe2+The system can generate hydroxyl free radical, namely o-phenanthroline-Fe through Fenton reaction2+The water solution is oxidized into phenanthroline-Fe by hydroxyl radical3+Thereafter, its maximum absorption peak at 510nm disappeared. If a hydroxyl radical scavenger exists in the reaction system, the hydroxyl radical generated by the Fenton reaction is completely or partially removed by the scavenger, and the phenanthrene-Fe is o-diazepine2+The damage to the complex will be reduced. Based on this principle, a colorimetric assay can be established that reflects the hydroxyl radical scavenging effect of radical scavengers as a510 changes.
1.5mL of 5mmol/L phenanthroline, 3.0mL of 0.5mol/L pH7.4 phosphate buffer solution and 7.5mmol/L FeSO are sequentially added into a 10mL reaction system41.0mL, mix immediately. Then 3.0mL of sample to be tested and 0.1% H are added2O21.0mL, and the volume was adjusted to 10mL with distilled water. After mixing, the mixture was subjected to a constant temperature water bath at 37 ℃ for 60min, and then the absorbance value was measured at a wavelength of 510 nm. Each sample was run in triplicate and averaged.

Clearance rate 95.5% 88.7% 91.2% 96.3% 97.7% 75.5% 80.5%
From the results of table 1, it can be seen that the effect is most excellent when the content of the humectant polyglutamic acid is 0.1 part. When the humectant also contains sodium hyaluronate and glycerin, the clearance rate of hydroxyl free radical is improved due to the compounding effect of the sodium hyaluronate and the glycerin. The absence of both rose hip fermentation filtrate and polyglutamic acid decreases the clearance of hydroxyl radicals.
All the subjects do not have any adverse reactions such as skin allergy in the test process, and the adverse reactions indicate that the skin is soft and has excellent skin feeling after the mask disclosed by the embodiment of the invention is used.
In addition, the DPPH radical clearance of the hot water rose hip leach solutions and the different fermentation filtrates were also tested and the results are shown in table 4.
TABLE 4 DPPH radical scavenging rates of Hot Water Leaching solutions and fermentation broths of Rose hips
Treatment method DPPH radical scavenging Rate%
Hot water extraction 53.1
Lactobacillus fermentation liquor 81.7
Yeast fermentation liquor 62.2
Fermentation of lactic acid bacteria yeast 92.4
As can be seen from table 4, the lactic acid bacteria fermentation enhanced the antioxidant capacity of the rose hip extract.
