Abstract
本发明公开了一种具有防治儿童青少年自闭症的益生菌矿物粉及其应用,益生菌矿物粉包括矿物类和萜类组份、药食两用组份、益生菌组份和维生素组份。经临床对比试验发现,采用本发明的益生菌矿物粉组合物具有很好的降低儿童孤独症评定量表(CARS)和儿童孤独症家长评定量表(ABC)的数值,并且能够提高Gesell发育量表(社会适应DQ、个人社交DQ、语言DQ)的数值。并且本发明通过药理试验表明,本发明提高的益生菌矿物粉可提高脑内神经递质5‑羟色胺和脑内神经营养因子的含量,可用于自闭症的新选择,且安全性高。

技术领域
本发明涉及一种有防治儿童青少年自闭症的益生菌矿物粉及其应用。
背景技术
自闭症是神经系统失调导致的发育障碍,自闭症又称孤独症,中国公益研究院2012年4月发布的《中国自闭症儿童现状分析报告》显示,目前中国自闭症患儿数约164万人,并且仍有很多潜在患者尚未被识别。早在2010年,全球自闭症患者就已达到6700万人,超过全世界癌症、艾滋病患者的总和。这种“无药可医”的疾病,已经成为世界上公认的重大公共卫生问题和沉重的社会负担。自闭症的临床表现为社会交往障碍、交流障碍、兴趣狭窄和刻板重复的行为方式。该病的发病机理目前尚未明确,可能与遗传因素、免疫系统异常、神经内分泌和神经递质功能失调等。现有治疗药物主要有中枢兴奋药、抗精神病药物等。目前的治疗药物治疗效果不理想,尤其是长期服用后带来较大的副作用,给儿童青少年身体发育造成较大的不良影响。
随着人类对肠道微生物菌群的研究,逐渐发现肠道是一个非常复杂的微环境,肠道菌群对人体的免疫力,神经发育、内分泌等等都具有重要的影响,有人提出肠-脑轴学说,说明肠道微生物对人体的健康,包括情志具有重要的影响,而儿童、青少年精细化饮食和脾胃差挑食,肠道菌群构成不合理,出现优势菌群少,对儿童、青少年的身体发育和神经系统发育都具有不良影响。
另外天然药物具有成分多、作用环节多、靶点多的特点,可通过多靶点起作用,产生协同效益,在抗抑郁方面具有较好的作用。
因此,很有必要在现有技术的基础上研究开发出适用于儿童青少年自闭症,疗效可靠,安全毒副作用小的新防治方法。
发明内容
发明目的:本发明的目的是为了解决现有技术的不足,提供一种配比科学合理,具有很好的治疗自闭症作用,并且安全可靠,不良反应低的益生菌矿物粉。
本发明提供的益生菌矿物粉不但可以改善肠道优势菌群,改善肠道微环境,达到改善情志认知障碍的作用。并且通过矿物粉起到补充微量元素和加强肠道益生菌繁殖的目的,二者配伍可起到协同增效的目的。另外本发明配伍药食两用的药材,可有效改善自闭症儿童症状,促进康复;再复配儿童青少年身体骨骼、神经发育的多种维生素,加强治疗效果;本发明各组份按优势比例配伍后,相辅相成,起到协同增效,达到临床治疗儿童青少年自闭症的功效。

(1)矿物类和萜类组份:方解石、石膏、珍珠、牡蛎,冰片;
(2)药食两用组份:栀子、莲子、益智仁、山药、核桃、沙棘、山楂、大枣、小麦;
(3)益生菌组份:Lactobacillus.rhamnosus GG菌、Lactobacillus.rhamnosusHN001菌、Lactobacillus.reuteri 1E1菌、Bifidobacterium.breve M-16V菌、Bifidobacterium.infantis M-63菌、Bifidobacterium adolescentis IM38菌、Bifidobacterium.animalis.ssp.lactis HN019菌、Bifidobacterium.longum BB536菌、Barnesiella intestinihominis、Romboutsia lituseburensis、Butyricicoccuspullicaecorum;
(4)维生素组份:维生素B1、维生素B6、维生素B12、维生素C、维生素D、维生素E、叶酸。

(1)矿物类和萜类组份:方解石1~5克、石膏5~15克、珍珠2~8克、牡蛎5~15克,冰片1~3克;
(2)药食两用组份:栀子2~6克、莲子5~15克、益智仁1~5克、山药5~25克、沙棘5~25克、核桃5~25克、山楂3~9克、大枣5~25克、小麦5~15克;
(3)益生菌组份:Lactobacillus.rhamnosus GG菌1~2Billion、Lactobacillus.rhamnosus HN001菌400~600Million、Lactobacillus.reuteri 1E1菌300~400Million、Bifidobacterium.breve M-16V菌100~200Million、Bifidobacterium.infantis M-63菌100~200Million、Bifidobacterium adolescentisIM38菌200~400Million、Bifidobacterium.animalis.ssp.lactis HN019菌300~500Million、Bifidobacterium.longum BB536菌100~300Million、Barnesiellaintestinihominis100~200Million、Romboutsia lituseburensis100~200Million、Butyricicoccus pullicaecorum100~200Million;

最佳优选方案,它由重量比为10:5:0.01的矿物类和萜类组份、药食两用组份和维生素组份混合物与益生菌组份制成。
作为优选方案,以上所述的具有防治儿童青少年自闭症的益生菌矿物粉,所述的方解石、石膏、珍珠和牡蛎通过超微粉碎机粉碎得到;
栀子、莲子、益智仁、山药、核桃、沙棘、山楂、大枣和小麦通过粉碎机粉碎过200目。
作为优选方案,以上所述的具有防治儿童青少年自闭症的益生菌矿物粉,将益生菌矿物粉与食品载体制备成胶囊、片剂、颗粒剂等便于儿童青少年服用或与葡萄糖制成糖果、果冻等。
本发明所述的具有防治儿童青少年自闭症的益生菌矿物粉在制备防治儿童青少年自闭症营养餐、保健品或功能饮料中的应用。
有益效果:本发明提供的具有防治儿童青少年自闭症的益生菌矿物粉和现有技术相比具有以下优点:
1、本发明提供的具有防治儿童青少年自闭症的益生菌矿物粉,采用矿物和益生菌组合,同时复配改善情志药食两用药材和添加人体生长发育的维生素。其中方解石、石膏、珍珠、牡蛎能够提供儿童青少年身体发育所需的必须Ca、Mn、Fe、Zn、Mg、Pb、Sr、Ba、Co、Si、Al、Na、Ba、Cu、Mn、Ti、Sr等等微量元素,并且矿物粉为益生菌繁殖提供必须的元素需求,从而在儿童、青少年肠道内提供优势菌群。另外冰片又称龙脑,本发明在矿物粉中特意添加冰片可以起到开窍促精神恢复的作用,与矿物元素合用相辅相成,可取得预料不到的技术效果。现代医学研究表明,身体肠道菌群是一个非常复杂的微环境,肠道内菌种多达300多种,与多种疾病密切相关,本发明通过大量实验筛选,优选多种特定的益生菌作为外源性补充,使其在自闭症儿童或青少年肠道内形成有利于自闭症治愈的微生物菌群,实验结果表明可大幅促进自闭症儿童精神恢复和活动能力,取得了非常好的预料不到的技术效果。
并且本发明经过大量实验筛选,根据儿童青少年自闭症发病特征,筛选安全有效的药食两用的原料,特别是采用改善情志的栀子、莲子和益智仁配伍,对儿童自闭症情志低落,不爱表达和对外界的恐惧等症状具有很好的改善作用,另外选择性添加健脾山药、益智核桃和大枣、促胃肠道消化提供膳食纤维的沙棘、山楂和小麦。配伍矿物和益生菌后可起到协同增效的作用。并且补充儿童青少年身体发育需要的维生素,改善内环境和新陈代谢,增强体质,促进自闭症儿童更乐于参与运动,改善症状。
2、经临床对比试验发现,采用本发明的益生菌矿物粉组合物具有很好的降低儿童孤独症评定量表(CARS)和儿童孤独症家长评定量表(ABC)的数值,并且能够提高Gesell发育量表(社会适应DQ、个人社交DQ、语言DQ)的数值。并且本发明通过药理试验表明,本发明提高的益生菌矿物粉可提高脑内神经递质5-羟色胺等的含量,同时具有较好的神经营养因子的功效。
3、并且通过对比试验结果表明,本发明提供的益生菌矿物粉组合物显著优于单个的益生菌组份或矿物组份及其药食两用组份。表明各组份相辅相成,协同增效,多靶点发挥治疗功效。且安全有效,无毒副作用。

一种具有防治儿童青少年自闭症的益生菌矿物粉,它由重量比为10:5:0.05的矿物类和萜类组份、药食两用组份和维生素组份制成的混合物和益生菌组份制成,密封袋装,每袋5克;
(1)矿物类和萜类组份为:方解石5克、石膏10克、珍珠2克、牡蛎5克,冰片1克;
(2)药食两用组份为:栀子5克、莲子10克、益智仁5克、山药20克、核桃25克、沙棘10克、山楂6克、大枣20克、小麦10克;
(3)维生素组份:维生素B11.5毫克、维生素B6 1.5毫克、维生素B12 10微克、维生素C 20毫克、维生素D 0.15毫克、维生素E10毫克、叶酸0.5毫克。

实施例3临床治疗自闭症疗效观察
(1)受试益生菌矿物粉:本发明实施例1,实施例3和对比实施例1~3配伍得到的益生菌矿物粉。
(2)孤独症研究对象:招募孤独症患儿60例作为研究被试,患者通过美国精神障碍诊断和统计手册(DSM-Ⅳ)儿童孤独症的诊断标准来确诊,平均年龄6岁左右,其中男孩30例,女孩30例。在这些被试中。实验对象随机分为实施例1治疗组、实施例3治疗组、对比实施例1~3对照组和空白对照组,每组10人。
(3)治疗方法:实施例1治疗组、实施例3治疗组、对比实施例1~3对照组均口服制备得到的益生菌矿物粉(开袋即食),每天2次,每次10克,可与食物混合一起服用。用药3个月后进行治疗效果评测。同时采用高通量测序仪检测肠道微生物治疗前后的变化。
(4)治疗效果评价:治疗前后采用CARS量表、ABC量表、Gesell发育量表(社会适应DQ、个人社交DQ、语言DQ),采用问卷形式对孤独症儿童的各项进行评测。采集的数据采用SPSS 18.0软件进行统计分析。
(5)结果:结果显示治疗组治疗前后CARS量表、ABC量表数值差异显著(P﹤0.05),而空白对照组治疗前后评分无明显变化。治疗后实施例1和实施例3治疗组CARS、ABC量表评分均明显降低,差异显著(P﹤0.05),而对比实施例1~3对照组CARS、ABC量表评分有所降低但是差异不显著,结果见表1。治疗后实施例1和3治疗组患儿社会适应DQ、语言DQ、个人社交DQ值与对照组比较均明显升高,差异显著(P﹤0.05),且实施例1的疗效优于实施例3。而对比实施例1~3对照组社会适应DQ、语言DQ、个人社交DQ值升高不明显。结果见表1和表2。
(6)肠道微生物变化:采用高通量测序仪测序分析16s rRNA,结果显示实施例1和3治疗组肠道微生物的多样性显著增加(P﹤0.01),随机森林(Random Forests)分析显示,双歧杆菌属数量和乳酸菌等有益菌显著增加(P﹤0.01)。

实施例4对神经递质和神经营养因子的影响
一、实验材料与药物
1.实验仪器
动物多重行为测试仪(上海吉量仪器有限公司);BT125型电子天平(赛多利斯科学仪器有限公司);KQ-250E型超声波清洗器(昆山禾创超声仪器有限公司)。Agilent1290Series高效液相色谱仪;Agilent QQQ 6410A三重四级杆质谱仪;分析色谱柱:ACEC18-AR,1.8μm,2.1x100 mm;BT125型电子天平(赛多利斯科学仪器有限公司);超纯水制备仪(南京易普易达公司)。多功能酶标仪(Enspire,Perkin-Elmer公司),高速离心机(Allegra X-12R Centrifuge和Microfuge 22R Centrifuge,Backman公司),恒温水浴锅(Bluepard公司)。
2.药物
受试药物:本发明实施例1和对比实施例1~3配伍得到的益生菌矿物粉;5-羟色胺均购自Sigma-Aldrich公司。
4.实验动物
SPF级雄性ICR种小鼠,体重18~22g,购自南京青龙山动物养殖场。
5.慢性压力应激抑郁小鼠糖水偏好性测试实验
将小鼠安置于培养室,静养7d后,腹腔注射丙戊酸,剂量500mg/kg,隔周再注射一次丙戊酸,剂量500mg/kg,出现觅食能力差,平板翻正能力、抓握发射明显低,视为造模成功。造模成功小鼠分为空白对照组、实施例1和对比实施例1~3组,空白对照组给于生理盐水,实施例1和对比实施例1~3组,按3mg/kg每天给于配伍得到的益生菌矿物粉,连续灌胃7d。
7.神经递质测定
(1)样品处理
灌胃给药后7d后解剖取全脑组织,按5g/mL的浓度值加入一定体积的甲酸(0.5M),高速分散制成匀浆液,15000转/分离心5分钟后取上清液。
(2)液相色谱分离
层析条件:流动相:0.1%甲酸水溶液(A),乙腈(B)。流速:0.2ml/min。梯度洗脱:0.0min:95%A;2.0min:90%A;8.0min:85%A;10min:80%A。
(3)质谱检测
干燥气温度(Drying gas temperature):340℃;干燥气流速(Drying gas flow):10L/min;毛细管电压(Vcap):4000V;雾化电压(Nebulizer):35psig;检测极性(Polarity):Positive;扫描模式(Scan mode):多离子检测模式(MRM);检测离子对(Ionpairsdetection):177.1>160.1,177.1>115.1(5-羟色胺);
8.海马组织神经营养因子含量测定
解剖获得海马组织,液氮迅速冷冻,粉碎。称取相应重量组织,加入10倍体积的组织裂解液(10mM HEPES,pH 7.5,1M NaCl,1mM EDTA,1mM EGTA,0.5%Triton X-100,5mMbenzamidine HCl,10μM aprotinin,10μM leupeptin),利用NGF与BDNF ELISA试剂盒,按试剂盒所列操作步骤,测定NGF与BDNF的含量。

二、实验结果
1.对脑内神经递质影响
本发明提供的实施例1的益生菌矿物粉相比空白对照组,可显著提高小鼠脑内5-羟色胺的含量,有显著性差异(P<0.05);并且与对比实施例1~3相比具有更加显著的上调作用,如表3所示。
2.对脑内神经营养因子影响
小鼠经灌胃7天后,本发明提供的实施例1的益生菌矿物粉相比空白对照组,可显著提升海马组织内的脑源性神经营养因子(BDNF)和神经生长因子(NGF)的含量,具有著性差异(P<0.05)。且与对比实施例1~3相比,具有更好的保护脑内神经营养因子的作用,取得了预料不到的技术效果。如表3所示:

Claims (3)
1.一种具有防治儿童青少年自闭症的益生菌矿物粉,其特征在于,它由重量比为10:5:0.05的矿物类和萜类组份、药食两用组份和维生素组份制成的混合物和益生菌组份制成,密封袋装,每袋5克;
(1)矿物类和萜类组份为:方解石5克、石膏10克、珍珠2克、牡蛎5克,冰片1克;
(2)药食两用组份为:栀子5克、莲子10克、益智仁5克、山药20克、核桃25克、沙棘10克、山楂6克、大枣20克、小麦10克;
(3)维生素组份:维生素B1 1.5毫克、维生素B6 1.5毫克、维生素B12 10微克、维生素C20毫克、维生素D 0.15毫克、维生素E 10毫克、叶酸0.5毫克;
其中方解石、石膏、珍珠和牡蛎通过超微粉碎机粉碎过200目得到;所述的栀子、莲子、益智仁、山药、核桃、山楂、大枣和小麦通过粉碎机粉碎过200目得到;
(4)益生菌组份的组成为:Lactobacillus.rhamnosus GG菌1Billion、Lactobacillus.rhamnosus HN001菌500Million、Lactobacillus.reuteri 1E1菌300Million、Bifidobacterium.breve M-16V菌100Million、Bifidobacterium.infantisM-63菌100Million、Bifidobacterium adolescentis IM38菌200Million、Bifidobacterium.animalis.ssp.lactis HN019菌300Million、Bifidobacterium.longum BB536菌100Million、Barnesiella intestinihominis 100Million、Romboutsia lituseburensis100Million、Butyricicoccus pullicaecorum 100Million。
2.根据权利要求1所述的具有防治儿童青少年自闭症的益生菌矿物粉,其特征在于,将益生菌矿物粉与食品载体制备成胶囊、片剂或与葡萄糖制成糖果。
3.权利要求1所述的具有防治儿童青少年自闭症的益生菌矿物粉在制备防治儿童青少年自闭症营养餐、保健品或功能饮料中的应用。
