本発明は、QOLを改善するための組成物に関する。
近年、「生活の質」などと訳されるQOL(Quality of Life)という指標が活用されている。QOLとは、一般に、個々の人生の内容の質や、社会的にみた「生活の質」のことを指し、ある人がどれだけ人間らしい生活や自分らしい生活を送り、人生に幸福を見出しているか、ということを尺度としてとらえる概念ともいえる。そのため、QOLは、個々の日々の生活とも密接に関係しており、例えば、個人の日々の睡眠、疲労、腸機能等とも大きく関係しているといえる。
睡眠は、ヒトなどの高等動物にとって重要な生理機能のひとつである。近年では、ライフスタイルの変化により生活リズムが乱れるなどの理由から、良質な睡眠を充分に取ることができず、仕事や学業などの日中の機能障害が生じた状態、いわゆる不眠に悩む人々が多い。
これに対し、様々な不眠改善剤や睡眠改善剤などが開発されている。例えば、ラクトバチルス・ヘルベティカスLBH MIKI-020(FERM P-13678)を含む乳酸菌、前記乳酸菌による発酵物、及び前記乳酸菌の菌体破砕物を含む配合物を4週間摂取したヒトに対し、OSA睡眠調査票MA版による睡眠評価を行った結果、因子I(起床時睡眠)と因子II(入眠と睡眠維持)において、試験開始前(0週)に比べて、試験終了後(4週)に有意な改善がみられたことが報告されている(特許文献1)。
これに対し、様々な不眠改善剤や睡眠改善剤などが開発されている。例えば、ラクトバチルス・ヘルベティカスLBH MIKI-020(FERM P-13678)を含む乳酸菌、前記乳酸菌による発酵物、及び前記乳酸菌の菌体破砕物を含む配合物を4週間摂取したヒトに対し、OSA睡眠調査票MA版による睡眠評価を行った結果、因子I(起床時睡眠)と因子II(入眠と睡眠維持)において、試験開始前(0週)に比べて、試験終了後(4週)に有意な改善がみられたことが報告されている(特許文献1)。
また、近年では、仕事や活動によって生じた心身の疲労を感じている人々も多い。また、精神的(中枢性)疲労の一因として脳内セロトニン濃度の上昇が挙げられており、運動やストレスなどにより、血液中のアミノ酸バランスが崩れると、脳内のセロトニン濃度が上昇する。
一般的に、心身の疲労を改善するためには十分な休息を取ることが有効であるが、十分な休息が取れない人々のために、様々な栄養ドリンクやサプリメントなどが開発されている。例えば、大豆サポニンBグループを含有する錠剤を8週間摂取したヒトに対し、VASによる評価を行った結果、「日々の疲労感」、「疲れ易さ・疲れの取れ易さ」、「肩こり」、「朝起きた時の気分」の数値で表される疲労に大幅な改善が見られたことが報告されている(特許文献2)。
また、ラクトバチルス・ヒルガルディーK-3(FERM P-18422)を用いて所定の発酵原料を発酵して得られた発酵液を加熱殺菌した後、ろ過し、得られたろ液を噴霧乾燥して得られたγ-アミノ酪酸(GABA)を含有する清涼飲料水を、運動開始30分前のヒトが摂取したところ、運動後、POMSにより評価される項目「疲労」の数値の上昇が抑制され、かつ、脳内セロトニン濃度の上昇が抑制されたことが報告されている(特許文献3)。
一般的に、心身の疲労を改善するためには十分な休息を取ることが有効であるが、十分な休息が取れない人々のために、様々な栄養ドリンクやサプリメントなどが開発されている。例えば、大豆サポニンBグループを含有する錠剤を8週間摂取したヒトに対し、VASによる評価を行った結果、「日々の疲労感」、「疲れ易さ・疲れの取れ易さ」、「肩こり」、「朝起きた時の気分」の数値で表される疲労に大幅な改善が見られたことが報告されている(特許文献2)。
また、ラクトバチルス・ヒルガルディーK-3(FERM P-18422)を用いて所定の発酵原料を発酵して得られた発酵液を加熱殺菌した後、ろ過し、得られたろ液を噴霧乾燥して得られたγ-アミノ酪酸(GABA)を含有する清涼飲料水を、運動開始30分前のヒトが摂取したところ、運動後、POMSにより評価される項目「疲労」の数値の上昇が抑制され、かつ、脳内セロトニン濃度の上昇が抑制されたことが報告されている(特許文献3)。
また、近年では、不規則な生活や運動不足などの不摂生、ホルモンバランスの崩れ、精神的ストレスの負荷等によって腸機能が悪化している人々も多い。
これに対し、整腸作用を発揮する様々な食品や医薬品が開発されている。例えば、ラクトバチルス・ブレビスの死菌とラクトフェリンとを有効成分として含有する錠剤を1日当たり3錠、夕食後から寝る前に4週間経口摂取したヒトにおいて、1週間の排便回数、排便量が多くなり、優れた腸内環境改善効果が得られたことが報告されている(特許文献4)。
これに対し、整腸作用を発揮する様々な食品や医薬品が開発されている。例えば、ラクトバチルス・ブレビスの死菌とラクトフェリンとを有効成分として含有する錠剤を1日当たり3錠、夕食後から寝る前に4週間経口摂取したヒトにおいて、1週間の排便回数、排便量が多くなり、優れた腸内環境改善効果が得られたことが報告されている(特許文献4)。
一方で、ラクトバチルス・ヘルベティカスという細菌が知られている。ラクトバチルス・ヘルベティカスは、高効率でD-アミノ酸を分泌することが知られており、当該細菌を用いて高効率でD-アミノ酸を製造する方法が開発されている(特許文献5)。
しかし、ラクトバチルス・ヘルベティカスが、QOLを改善する効果を奏することは知られていない。
本発明は上記事情に鑑みてなされたものであって、生活の質(Quality of Life (QOL))を改善するための技術の提供を課題とする。好ましくは、不眠を改善するため、疲労を改善するため、又は整腸のための技術の提供を課題とする。
本発明者らは、ラクトバチルス・ヘルベティカスが、QOL改善作用、好ましくは、不眠改善作用、疲労改善作用、整腸作用を有することを見出し、本発明を完成するに至った。
本発明は、ラクトバチルス・ヘルベティカスを含む、QOLを改善するための組成物を提供することができる。

ラクトバチルス・ヘルベティカスは飲食品や医薬品に含有することができる細菌であるため、QOLを改善するための飲食品や医薬品として調製することができ、対象は簡便に摂取等することができる。
以下、本発明を詳細に説明する。ただし、本発明は以下の形態に限定されず、本発明の範囲内で自由に変更することができるものである。

尚、本発明に係る、ラクトバチルス・ヘルベティカスを含む、QOLを改善するための組成物、また、好ましい態様でもある、不眠を改善するための組成物、疲労を改善するための組成物、整腸のための組成物は、いずれも混合物であってよく、その成分が均一であるか不均一であるかを問わない。
また、本明細書では、本発明に係る、QOLを改善するための組成物、また、好ましい態様でもある、不眠を改善するための組成物、疲労を改善するための組成物、整腸のための組成物を、「本発明の組成物」とまとめて記載することがある。
また、本発明の組成物は、ラクトバチルス・ヘルベティカスを単独で含んでもよいし、QOL改善作用を発揮する限り、他の成分を含んでもよい。
また、本明細書では、「摂取」、「摂取する」、「摂取した」などの語は、医薬組成物に用いられる場合には、それぞれ、「投与」、「投与される」、「投与された」などの語に置き換えることができる。

また、本発明の組成物は、ラクトバチルス・ヘルベティカスを単独で含んでもよいし、QOL改善作用を発揮する限り、他の成分を含んでもよい。
また、本明細書では、「摂取」、「摂取する」、「摂取した」などの語は、医薬組成物に用いられる場合には、それぞれ、「投与」、「投与される」、「投与された」などの語に置き換えることができる。
本発明の組成物は、ラクトバチルス・ヘルベティカス(Lactobacillus helveticus)を含む。本発明で用いるラクトバチルス・ヘルベティカスとしては、対象がラクトバチルス・ヘルベティカスを含む組成物を摂取したときにQOL改善作用、また、好ましい態様においては、不眠改善作用、疲労改善作用、又は整腸作用が発揮される限り特に制限されない。例えば、ラクトバチルス・ヘルベティカスMCC1848(NITE BP-01671)、ラクトバチルス・ヘルベティカスJCM 1120(JCM 1120Tと表記されることがある。Tは標準株であることを表す。)、ラクトバチルス・ヘルベティカスDSM 20075、ラクトバチルス・ヘルベティカスSBT 2171等が挙げられる。好ましくは、ラクトバチルス・ヘルベティカスMCC1848(NITE BP-01671)である。本発明の組成物では、ラクトバチルス・ヘルベティカスとして、一を用いてもよいし、複数を用いてもよい。
ラクトバチルス・ヘルベティカスMCC1848(NITE BP-01671)は、2013年7月29日に、独立行政法人製品評価技術基盤機構特許微生物寄託センター(〒292-0818 千葉県木更津市かずさ鎌足2-5-8 122号室)に、ブダペスト条約に基づき国際寄託されている(受託番号:NITE BP-01671)。
ラクトバチルス・ヘルベティカスJCM 1120は、独立行政法人理化学研究所バイオリソースセンター微生物材料開発室(JCM)より入手することができる。
ラクトバチルス・ヘルベティカスDSM 20075は、Deutsche Sammlung von Mikroorganismen und Zellkulturen GmbH(住所:Inhoffenstraβe 7B, 38124 Braunschweig, Germany)より入手することができる。

なお、ラクトバチルス・ヘルベティカスMCC1848(NITE BP-01671)には、当該細菌名で所定の機関に寄託や登録がなされている株そのもの(以下、説明の便宜上、「寄託株」ともいう。)に限られず、それと実質的に同等な株(「派生株」または「誘導株」ともいう。)も包含される。すなわち、「ラクトバチルス・ヘルベティカスMCC1848(NITE BP-01671)」には、受託番号NITE BP-01671で上記寄託機関に寄託されている株そのものに限られず、それと実質的に同等な株も包含される。「上記寄託株と実質的に同等の株」とは、上記寄託株と同一の種に属し、本発明の効果が得られ、さらにその16SrRNA遺伝子の塩基配列が、上記寄託株の16SrRNA遺伝子の塩基配列に対して、好ましくは98%以上、より好ましくは99%以上、さらに好ましくは100%の同一性を有し、かつ、好ましくは上記寄託株と同一の菌学的性質を有する株をいう。上記寄託株と実質的に同等の株は、例えば、当該寄託株を親株とする派生株であってよい。派生株としては、寄託株から育種された株や寄託株から自然に生じた株が挙げられる。育種方法としては、遺伝子工学的手法による改変や、突然変異処理による改変が挙げられる。突然変異処理としては、X線の照射、紫外線の照射、ならびにN-メチル-N'-ニトロ-N-ニトロソグアニジン、エチルメタンスルフォネート、およびメチルメタンスルフォネート等の変異剤による処理が挙げられる。寄託株から自然に生じた株としては、寄託株の使用の際に自然に生じた株が挙げられる。そのような株としては、寄託株の培養(例えば継代培養)により自然に生じた変異株が挙げられる。派生株は、一種の改変により構築されてもよく、二種またはそれ以上の改変により構築されてもよい。
このことは、既出の他の寄託菌にも同様に適用される。

本発明の組成物に含有し得るラクトバチルス・ヘルベティカスの菌体としては、市販品を用いてもよく、適宜製造して取得したものを用いてもよい。例えば、ラクトバチルス・ヘルベティカスMCC1848(NITE BP-01671)の加熱殺菌体の市販品であれば、森永乳業株式会社から入手することができる。
本発明の組成物に含有し得るラクトバチルス・ヘルベティカスは、ラクトバチルス・ヘルベティカスを培養することにより容易に取得することができる。培養方法は、ラクトバチルス・ヘルベティカスが増殖できる限り、特に制限されない。培養方法としては、例えば、ラクトバチルス属細菌(乳酸菌)の培養に通常用いられる方法を、そのまま、あるいは適宜修正して、用いることができる。培養温度は、例えば、25~50℃であってよく、35~42℃であることが好ましい。培養は、好ましくは嫌気条件下で実施することができ、例えば、炭酸ガス等の嫌気ガスを通気しながら実施することができる。また、培養は、液体静置培養等の微好気条件下で実施することもできる。培養は、例えば、ラクトバチルス・ヘルベティカスが所望の程度に増殖するまで実施することができる。
培養に用いる培地は、ラクトバチルス・ヘルベティカスが増殖できる限り、特に制限されない。培地としては、例えば、ラクトバチルス属細菌の培養に通常用いられる培地を、そのまま、あるいは適宜修正して、用いることができる。すなわち、炭素源としては、例えば、ガラクトース、グルコース、フルクトース、マンノース、セロビオース、マルトース、ラクトース、スクロース、トレハロース、デンプン、デンプン加水分解物、廃糖蜜等の糖類を資化性に応じて用いることができる。窒素源としては、例えば、アンモニア、硫酸アンモニウム、塩化アンモニウム、硝酸アンモニウム等のアンモニウム塩類や硝酸塩類を用いることができる。また、無機塩類としては、例えば、塩化ナトリウム、塩化カリウム、リン酸カリウム、硫酸マグネシウム、塩化カルシウム、硝酸カルシウム、塩化マンガン、硫酸第一鉄等を用いることができる。また、ペプトン、大豆粉、脱脂大豆粕、肉エキス、酵母エキス等の有機成分を用いてもよい。また、ラクトバチルス属細菌の培養に通常用いられる培地として、具体的には、強化クロストリジア培地(Reinforced Clostridial medium)、MRS培地(de Man, Rogosa, and Sharpe medium)、mMRS培地(modified MRS medium)、TOSP培地(TOS propionate medium)、TOSP Mup培地(TOS propionate mupirocin medium)が挙げられる。
本発明の組成物に含有し得るラクトバチルス・ヘルベティカスとしては、ラクトバチルス・ヘルベティカスの菌体またはそれを含有する画分を、特に制限されず用いることができる。すなわち、ラクトバチルス・ヘルベティカスとしては、例えば、培養により得られた培養物をそのまま用いてもよく、培養物を希釈又は濃縮して用いてもよく、培養物から回収した菌体を用いてもよい。また、本発明の効果を損なわない限り、加熱や凍結乾燥等の種々の追加操作を培養後に行うことができる。すなわち、本発明の組成物に含有し得るラクトバチルス・ヘルベティカスとして、具体的には、ラクトバチルス・ヘルベティカスの培養物、又は前記培養物から回収した菌体、それらの処理物が挙げられ、前記処理物としては希釈物、濃縮物、又は乾燥物等が挙げられる。
なお、菌体は、生菌体でも死菌体でもよく、通常は死菌体を含有する形態で使用されるのが好ましい。生菌体であれば、プロバイオティクスとしての効果を期待することができる。また、菌体は、生菌体からなるものであってもよく、死菌体からなるものであってもよく、生菌体と死菌体の混合物であってもよい。
死菌体としては、加熱等により殺菌された死菌体(すなわち、加熱殺菌体等)が挙げられる。加熱殺菌体は、例えば、上記のようにしてラクトバチルス・ヘルベティカスを培養後、得られた培養液を遠心分離し、凍結乾燥して生菌末を得た後、滅菌水に懸濁し、90℃で30分間の加熱殺菌をして得ることができる。
その他の死菌体の調製法として、噴霧乾燥法(スプレードライ法)、レトルト殺菌法、凍結乾燥法、UHT殺菌法、加圧殺菌法、高圧蒸気滅菌法、乾熱滅菌法、流通蒸気消毒法、電磁波殺菌法、電子線滅菌法、高周波滅菌法、放射線滅菌法、紫外線殺菌法、酸化エチレンガス滅菌法、過酸化水素ガスプラズマ滅菌法、化学的殺菌法(アルコール殺菌法、ホルマリン固定法、電解水処理法)等が挙げられる。
死菌体としては、加熱等により殺菌された死菌体(すなわち、加熱殺菌体等)が挙げられる。加熱殺菌体は、例えば、上記のようにしてラクトバチルス・ヘルベティカスを培養後、得られた培養液を遠心分離し、凍結乾燥して生菌末を得た後、滅菌水に懸濁し、90℃で30分間の加熱殺菌をして得ることができる。

本発明の組成物を摂取する対象としてはヒトである。また、該ヒトは男性でも女性でもよいが、好ましくは女性である。また、該ヒトは20歳以上65歳未満のヒトが好ましい。また、該ヒトの、POMS2短縮版により評価される項目FI(疲労-無気力)の数値は、好ましくは50以上、より好ましくは55以上、さらに好ましくは60以上であり、一方で、好ましくは75以下、より好ましくは73以下、さらに好ましくは70以下である。また、2回の同一の評価で2回とも項目FI(疲労-無気力)の数値が前記数値範囲内であるヒトがより好ましい。
本発明におけるQOLとしては、例えば、睡眠面の健康の程度、身体面の健康の程度等が挙げられる。睡眠面の健康の程度を改善することとしては、例えば、対象の不眠の程度を改善すること等が挙げられる。また、身体面の健康の程度を改善することとしては、例えば、対象の疲労の程度を改善すること、対象の腸機能の発揮の程度を改善すること等が挙げられる。
本発明の組成物が含有する前記ラクトバチルス・ヘルベティカスは、対象の不眠、疲労、腸機能を改善する作用を有する。そのため、本発明の組成物は、不眠を改善するための組成物、疲労を改善するための組成物、腸機能を改善するための組成物(腸機能を改善するための組成物は、整腸のための組成物といってもよい。)としても有用である。
本発明の組成物が含有する前記ラクトバチルス・ヘルベティカスは、対象の不眠、疲労、腸機能を改善する作用を有する。そのため、本発明の組成物は、不眠を改善するための組成物、疲労を改善するための組成物、腸機能を改善するための組成物(腸機能を改善するための組成物は、整腸のための組成物といってもよい。)としても有用である。
本発明の一態様は、前記ラクトバチルス・ヘルベティカスを含む、不眠を改善するための組成物である。以下、当該組成物を「態様1の組成物」と記載することがある。
態様1の組成物が含むラクトバチルス・ヘルベティカスは、不眠改善作用を有する。すなわち、例えば、対象がラクトバチルス・ヘルベティカスを摂取したときに、摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)よりも、不眠が改善する。したがって、前記ラクトバチルス・ヘルベティカスは、不眠を改善するための組成物の有効成分として使用することができる。
また、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象と比較して、ラクトバチルス・ヘルベティカスを含む組成物を摂取した対象の方が、不眠が改善する。すなわち、前記不眠は、ラクトバチルス・ヘルベティカスを含まない組成物(例えば、プラセボ)を摂取した対象の不眠と比較して改善することが評価される不眠であってもよい。尚、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象の不眠は改善してもよいし、改善しなくてもよい。
また、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象と比較して、ラクトバチルス・ヘルベティカスを含む組成物を摂取した対象の方が、不眠が改善する。すなわち、前記不眠は、ラクトバチルス・ヘルベティカスを含まない組成物(例えば、プラセボ)を摂取した対象の不眠と比較して改善することが評価される不眠であってもよい。尚、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象の不眠は改善してもよいし、改善しなくてもよい。
本態様により改善される不眠としては、対象がラクトバチルス・ヘルベティカスを摂取したときに改善される不眠であれば特に制限されないが、「寝床についてから実際に寝るまで時間がかかる状態であること」、「夜間、睡眠の途中で目が覚める状態であること」、「希望する起床時間より早く目覚めて、それ以降、眠れない状態であること」、「夜の眠りや昼寝も合わせて、睡眠時間が足りていない状態であること」、「全体的な睡眠の質について満足していない状態であること」、「日中の気分が滅入っている状態であること」、「日中の身体的および精神的な活動の状態が低下している状態であること」、「日中の眠気がある状態であること」、「起床時に眠気や疲労を感じすっきり起きられない状態であること」、及び、「睡眠状態がよくない状態であること」からなる群から選択される一又は複数が好ましい。また、本態様の用途は、睡眠の質及び/又は目覚めの良さを改善することであってもよい。
本発明により改善される不眠は、自覚する不眠であってもよいし、当該技術分野において通常行われる検査によって客観的に判定され得る不眠であってもよいし、当該技術分野において通常行われるアンケートによって判定され得る不眠であってもよいが、好ましくは、当該技術分野において通常行われる検査によって客観的に判定され得る不眠、当該技術分野において通常行われるアンケートによって判定され得る不眠である。
本態様により改善される不眠が、当該技術分野において通常行われる検査によって客観的に判定され得る不眠である場合、例えば、アテネ不眠尺度(Athens Insomnia Scale, AIS)により評価される項目の合計得点で表される不眠等が挙げられる。本態様により改善される不眠は、一の検査によって客観的に判定され得る不眠であってもよく、複数の検査によって客観的に判定され得る不眠でもよい。好ましくは、アテネ不眠尺度により評価される項目の合計得点で表される不眠である。
アテネ不眠尺度とは、世界保健機関(WHO)が中心になって設立した「睡眠と健康に関する世界プロジェクト」が作成した検査であり、当該技術分野において広く用いられている検査である。アテネ不眠尺度を用いた検査は、周知のとおり、項目「寝床についてから実際に寝るまで時間がかかること」、項目「夜間、睡眠の途中で目が覚めること」、項目「希望する起床時間より早く目覚めて、それ以降、眠れないこと」、項目「夜の眠りや昼寝も合わせて、睡眠時間が足りていないこと」、項目「全体的な睡眠の質について満足していないこと」、項目「日中の気分が滅入っていること」、項目「日中の身体的および精神的な活動の状態が低下していること」、及び項目「日中の眠気があること」について、被験者が過去1か月間に少なくとも週3回以上経験したものを、0点、1点、2点、3点の4段階で回答し、その合計得点で判定する検査である。尚、アテネ不眠尺度を用いた検査では、合計得点が、3点以下の場合は「睡眠がとれている」と、4~5点の場合は「不眠症の疑いが少しある」と、6点以上の場合は「不眠症の可能性が高い」と判断される。
本態様により改善される不眠が、アテネ不眠尺度により評価される項目の合計得点で表される不眠である場合、対象が態様1の組成物を摂取したときの当該合計得点が、対象が態様1の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の当該合計得点を基準として、2以上減少することが好ましく、5以上減少することがより好ましく、7以上減少することがさらに好ましい。一方で、例えば、20以下減少することが好ましい。
このとき、対象が態様1の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の当該合計得点は、特に制限されないが、好ましくは4以上、より好ましくは5以上、さらに好ましくは6以上であり、一方で、好ましくは20以下、より好ましくは17以下、さらに好ましくは15以下である。
また、本態様の用途が、睡眠の質及び/又は目覚めの良さを改善することであって、その改善が、アンケートの項目「睡眠の質・目覚めの良さ」で評価される場合、対象が態様1の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の「睡眠の質・目覚めの良さ」を基準とした、対象が態様1の組成物を摂取したときの「睡眠の質・目覚めの良さ」について、「良くなった」、及び「少し良くなった」と回答した人数が、「変わらない」、「少し悪くなった」、及び「悪くなった」と回答した人数よりも多いことが好ましく、有意に多いことがより好ましい。
態様1の組成物は、不眠改善作用を有しており、不眠が改善することによって予防又は治療され得る疾患、症候、症状又は障害等の予防又は治療に使用することができる。尚、「治療」には、改善も含まれる。また、対象は、一又は複数の疾患、症候、症状又は障害等を有していてよい。
このような疾患、症候、症状又は障害等としては、例えば、睡眠障害(例えば、不眠症、過眠症(例えば、ナルコレプシー等)、睡眠時随伴症、睡眠相後退症候群(睡眠相後退障害と称されることがある。)等)等が挙げられる。
尚、不眠症とは、寝つきが悪い、何度も目がさめる、眠りが浅いといった症状が慢性化している状態である。
過眠症とは、日中に過剰な眠気のため日常生活に支障をきたす状態である。
ナルコレプシーとは、日中に耐えがたい眠気に襲われ、突然その場で眠ってしまう症状であり、過眠症の一態様である。
睡眠時随伴症とは、睡眠中に起こる行動や心身の異常の総称であり、一般的に「夢遊病」として知られる症状である。
睡眠相後退症候群とは、夜型生活が続き、朝起きられなくなる症候群である。
このような疾患、症候、症状又は障害等としては、例えば、睡眠障害(例えば、不眠症、過眠症(例えば、ナルコレプシー等)、睡眠時随伴症、睡眠相後退症候群(睡眠相後退障害と称されることがある。)等)等が挙げられる。
尚、不眠症とは、寝つきが悪い、何度も目がさめる、眠りが浅いといった症状が慢性化している状態である。
過眠症とは、日中に過剰な眠気のため日常生活に支障をきたす状態である。
ナルコレプシーとは、日中に耐えがたい眠気に襲われ、突然その場で眠ってしまう症状であり、過眠症の一態様である。

本発明の他の一態様は、前記ラクトバチルス・ヘルベティカスを含む、疲労を改善するための組成物である。以下、当該組成物を「態様2の組成物」と記載することがある。
態様2の組成物が含有するラクトバチルス・ヘルベティカスは、疲労改善作用を有する。すなわち、例えば、対象がラクトバチルス・ヘルベティカスを摂取したときに、摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)よりも、疲労が改善する。したがって、前記ラクトバチルス・ヘルベティカスは、疲労を改善するための組成物の有効成分として使用することができる。
また、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象と比較して、ラクトバチルス・ヘルベティカスを含む組成物を摂取した対象の方が、疲労が改善する。すなわち、前記疲労は、ラクトバチルス・ヘルベティカスを含まない組成物(例えば、プラセボ)を摂取した対象の疲労と比較して改善することが評価される疲労であってもよい。尚、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象の疲労は改善してもよいし、改善しなくてもよい。
また、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象と比較して、ラクトバチルス・ヘルベティカスを含む組成物を摂取した対象の方が、疲労が改善する。すなわち、前記疲労は、ラクトバチルス・ヘルベティカスを含まない組成物(例えば、プラセボ)を摂取した対象の疲労と比較して改善することが評価される疲労であってもよい。尚、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象の疲労は改善してもよいし、改善しなくてもよい。
本態様により改善される疲労としては、対象がラクトバチルス・ヘルベティカスを摂取したときに改善される疲労であれば特に制限されないが、「疲労及び/又は無気力」が好ましい。また、「ぐったりしている状態であること」、「つかれている状態であること」、「へとへとである状態であること」、「だるい状態であること」、「うんざりである状態であること」、及び「精根尽き果てた状態であること」からなる群から選択される一又は複数も好ましい。また、「疲労を感じている状態であること」及び「ぐったりしている状態であること」からなる群から選択される一又は複数も好ましい。また、「疲れに関して問題がある状態であること」、「もっと休養が必要な状態であること」、「眠気またはだるさを感じている状態であること」、「何かを始めるのが困難である状態であること」、「問題なく何かを始めても、それを続けているうちに体が弱っていく状態であること」、「行動力にかけている状態であること」、「以前より筋力が衰えている状態であること」、「身体が弱っていると感じる状態であること」、「集中するのに苦労する状態であること」、「物事を明確に考えるのが大変である状態であること」、「話しているときに言い間違いをする状態であること」、「的確な言葉を探すのに以前より苦労する状態であること」、「記憶力がいつもより悪い状態であること」、及び「以前は関心があったことに興味を持てなくなった状態であること」からなる群から選択される一又は複数も好ましい。また、本態様の用途は、疲労感及び/又は体のだるさを改善することであってもよい。
本態様により改善される疲労は、自覚する疲労であってもよいし、当該技術分野において通常行われる検査によって客観的に判定され得る疲労であってもよいし、当該技術分野において通常行われるアンケートによって判定され得る疲労であってもよいが、好ましくは、当該技術分野において通常行われる検査によって客観的に判定され得る疲労、当該技術分野において通常行われるアンケートによって判定され得る疲労である。
本態様により改善される疲労が、当該技術分野において通常行われる検査によって客観的に判定され得る疲労である場合、例えば、POMSにより評価される項目「疲労」の数値で表される疲労、POMS2により評価される項目「疲労-無気力」の数値で表される疲労、VASにより評価される項目「疲労を感じている」の数値で表される疲労、VASにより評価される項目「ぐったりしている」の数値で表される疲労、CFSにより評価される項目の合計得点で表される疲労等が挙げられる。本態様により改善される疲労は、一の検査によって客観的に判定され得る疲労であってもよく、複数の検査によって客観的に判定され得る疲労でもよい。好ましくは、POMSにより評価される項目「疲労」の数値で表される疲労、POMS2により評価される項目「疲労-無気力」の数値で表される疲労、VASにより評価される項目「疲労を感じている」の数値で表される疲労、VASにより評価される項目「ぐったりしている」の数値で表される疲労、及びCFSにより評価される項目の合計得点で表される疲労からなる群から選択される一又は複数である。
POMS(登録商標)及びPOMS2(商標)は、それぞれ、Profile of Mood States、Profile of Mood States 2nd Editionの略であって、マルチ-ヘルス システムズ インコーポレイテッド(Multi-Health Systems Inc.)が提供するものであり、当該技術分野において広く用いられている検査である。POMS2はPOMSの改訂版であり、その主要部は前記POMSから変更されていない。尚、POMS及びPOMS2には、成人用や青少年用のように対象となる母集団により検査方法が異なるものや、全項目版や短縮版のように検査時の質問項目数が異なるものがあるが、対象の状況に応じて検査方法や質問項目数を適宜選択すればよい。
「ぐったりしていること」についてのPOMS2を用いた検査は、周知のとおり、項目「ぐったりする」について、被験者が0点、1点、2点、3点の4段階で回答し、所定の方法で算出した得点をもって「ぐったりしていること」を判定する検査である。「つかれていること」、「へとへとであること」、「だるいこと」、「うんざりであること」、「精根尽き果てたこと」についても同様である。

VASは、Visual Analogue Scaleの略であって、当該技術分野において広く用いられている検査である。「疲労を感じている」についてのVASを用いた検査は、周知の通り、「0」を「疲労を全く感じない」、「100」を「疲労を非常に感じる」(これまで経験した一番大きい疲労を感じる)として、現在の疲労が10 cmの直線上のどの位置にあるかを被験者が示して判定する検査である。「ぐったりしていること」についても同様である。
CFSは、Chalder Fatigue Scaleの略であって、米国疾病予防管理センター(CDC)が疲労の検査において使用を推奨している、当該技術分野において広く用いられている検査である。CFSを用いた検査は、周知のとおり、項目「疲れに関して問題があること」、項目「もっと休養が必要であること」、項目「眠気またはだるさを感じていること」、項目「何かを始めるのが困難であること」、項目「問題なく何かを始めても、それを続けているうちに体が弱っていくこと」、項目「行動力にかけていること」、項目「以前より筋力が衰えていること」、項目「身体が弱っていると感じること」、項目「集中するのに苦労すること」、項目「物事を明確に考えるのが大変であること」、項目「話しているときに言い間違いをすること」、項目「的確な言葉を探すのに以前より苦労すること」、項目「記憶力がいつもより悪いこと」、及び項目「以前は関心があったことに興味を持てなくなったこと」について、被験者が0点、1点、2点、3点の4段階で回答し、その合計得点で判定する検査である。
本態様により改善される疲労が、POMS2により評価される項目「疲労-無気力」の数値で表される疲労である場合、対象が態様2の組成物を摂取したときの当該項目「疲労-無気力」の数値が、対象が態様2の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の当該項目「疲労-無気力」の数値を基準として、2以上減少することが好ましく、4以上減少することがより好ましく、8以上減少することがさらに好ましい。一方で、例えば、20以下減少することが好ましい。
このとき、対象が態様2の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の当該項目「疲労-無気力」の数値は、特に制限されないが、好ましくは50以上、より好ましくは55以上、さらに好ましくは60以上であり、一方で、好ましくは75以下、より好ましくは73以下、さらに好ましくは70以下である。
本態様により改善される疲労が、VASにより評価される項目「疲労を感じている」の数値で表される疲労である場合、対象が態様2の組成物を摂取したときの当該項目「疲労を感じている」の数値が、対象が態様2の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の当該項目「疲労を感じている」の数値を基準として、5以上減少することが好ましく、10以上減少することがより好ましく、15以上減少することがさらに好ましい。一方で、例えば、80以下減少することが好ましい。
このとき、対象が態様2の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の当該項目「疲労を感じている」の数値は、特に制限されないが、好ましくは50以上、より好ましくは55以上、さらに好ましくは60以上であり、一方で、好ましくは95以下、より好ましくは90以下、さらに好ましくは85以下である。
本態様により改善される疲労が、VASにより評価される項目「ぐったりしている」の数値で表される疲労である場合、対象が態様2の組成物を摂取したときの当該項目「ぐったりしている」の数値が、対象が態様2の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の当該項目「ぐったりしている」の数値を基準として、5以上減少することが好ましく、10以上減少することがより好ましく、15以上減少することがさらに好ましい。一方で、例えば、80以下減少することが好ましい。
このとき、対象が態様2の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の当該項目「ぐったりしている」の数値は、特に制限されないが、好ましくは50以上、より好ましくは55以上、さらに好ましくは60以上であり、一方で、好ましくは95以下、より好ましくは90以下、さらに好ましくは85以下である。
本態様により改善される疲労が、CFSにより評価される項目の合計得点で表される疲労である場合、対象が態様2の組成物を摂取したときの当該項目の合計得点が、対象が態様2の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の当該項目の合計得点を基準として、2以上減少することが好ましく、4以上減少することがより好ましく、6以上減少することがさらに好ましい。一方で、例えば、30以下減少することが好ましい。
このとき、対象が態様2の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の当該合計得点は、特に制限されないが、好ましくは10以上、より好ましくは15以上、さらに好ましくは20以上であり、一方で、好ましくは40以下、より好ましくは35以下、さらに好ましくは30以下である。
また、本態様の用途が、疲労感及び/又は体のだるさを改善することであって、その改善が、アンケートの項目「疲労感・体のだるさ」で評価される場合、対象が態様2の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の「疲労感・体のだるさ」を基準とした、対象が態様2の組成物を摂取したときの「疲労感・体のだるさ」について、「良くなった」、及び「少し良くなった」と回答した人数が、「変わらない」、「少し悪くなった」、及び「悪くなった」と回答した人数よりも多いことが好ましく、有意に多いことがより好ましい。
態様2の組成物は、疲労改善作用を有しており、疲労が改善することによって予防又は治療され得る疾患、症候、症状又は障害等の予防又は治療に使用することができる。尚、「治療」には、改善も含まれる。また、対象は、一又は複数の疾患、症候、症状又は障害等を有していてよい。
このような疾患、症候、症状又は障害等としては、例えば、思考能力の低下、刺激に対する反応の低下、注意力の低下、注意散漫、動作緩慢、行動量の低下、眼のかすみ、頭痛、肩こり、腰痛等が挙げられる。

本発明の他の一態様は、前記ラクトバチルス・ヘルベティカスを含む、整腸のための組成物である。以下、当該組成物を「態様3の組成物」と記載することがある。
態様3の組成物が含有するラクトバチルス・ヘルベティカスは、整腸作用を有する。本態様における整腸作用とは、腸機能を改善する作用をいう。すなわち、対象がラクトバチルス・ヘルベティカスを摂取したときに、摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)よりも、腸機能が改善する。したがって、前記ラクトバチルス・ヘルベティカスは、整腸のための組成物の有効成分として、また、腸機能を改善するための組成物の有効成分として使用することができる。
また、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象と比較して、ラクトバチルス・ヘルベティカスを含む組成物を摂取した対象の方が、腸機能が改善する。すなわち、前記腸機能は、ラクトバチルス・ヘルベティカスを含まない組成物(例えば、プラセボ)を摂取した対象の腸機能と比較して改善することが評価される腸機能であってもよい。尚、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象の腸機能は改善してもよいし、改善しなくてもよい。
また、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象と比較して、ラクトバチルス・ヘルベティカスを含む組成物を摂取した対象の方が、腸機能が改善する。すなわち、前記腸機能は、ラクトバチルス・ヘルベティカスを含まない組成物(例えば、プラセボ)を摂取した対象の腸機能と比較して改善することが評価される腸機能であってもよい。尚、ラクトバチルス・ヘルベティカスを含まない組成物を摂取した対象の腸機能は改善してもよいし、改善しなくてもよい。
本態様により改善される腸機能としては、対象がラクトバチルス・ヘルベティカスを摂取したときに改善される腸機能であれば特に制限されない。また、本態様の用途は、お腹の調子を改善することであってもよい。
本態様により改善される腸機能は、自覚する腸機能であってもよいし、当該技術分野において通常行われる検査によって客観的に判定され得る腸機能であってもよいし、当該技術分野において通常行われるアンケートによって判定され得る腸機能であってもよいが、好ましくは、アンケートによって判定され得る腸機能である。
また、本態様の用途がお腹の調子を改善することであって、その改善が、アンケートの項目「お腹の調子」で評価される場合、対象が態様3の組成物を摂取しないとき(又は摂取する前、又はプラセボを摂取したとき)の「お腹の調子」を基準とした、対象が態様3の組成物を摂取したときの「お腹の調子」について、「良くなった」、及び「少し良くなった」と回答した人数が、「変わらない」、「少し悪くなった」、及び「悪くなった」と回答した人数よりも多いことが好ましく、有意に多いことがより好ましい。
態様3の組成物は、整腸作用を有しており、整腸によって予防又は治療され得る疾患、症候、症状又は障害等の予防又は治療に使用することができる。尚、「治療」には、改善も含まれる。また、対象は、一又は複数の疾患、症候、症状又は障害等を有していてよい。
このような疾患、症候、症状又は障害等としては、例えば、排便障害が挙げられる。
排便障害としては、例えば、便秘、下痢等が挙げられる。尚、本明細書においては、下痢は便失禁を含むものとする。
便秘とは、排便が困難になっている状態を指す障害であり、便が固い、排便に苦労する、何日も排便がない場合等が挙げられ、排便回数及び/又は排便量が少ない。
下痢とは、健常時の便と比較して非常に緩いゲル(粥)状又は液体状の便が排泄される障害であり、軟便や泥状便、水様便等と称され、排便回数及び/又は排便量が多い。
便失禁とは、無意識又は意に反して排便してしまう障害であり、排便回数及び/又は排便量が多い。便失禁としては、例えば、腹圧性便失禁、切迫性便失禁、溢流性便失禁、機能性便失禁等が挙げられる。
このような疾患、症候、症状又は障害等としては、例えば、排便障害が挙げられる。

また、便秘としては、例えば、通過遅延型便秘、機能性便秘、器質性便秘又は薬剤誘発性便秘等が挙げられる。

本発明の組成物における前記ラクトバチルス・ヘルベティカスの含量は、組成物の態様によって適宜設定されるが、総量で、1×104~1×1013cfu/gまたは1×104~1×1013cfu/mlの範囲内であることが好ましく、1×105~1×1012cfu/gまたは1×105~1×1012cfu/mlの範囲内であることがより好ましく、1×106~1×1011cfu/gまたは1×106~1×1011cfu/mlの範囲内であることがさらに好ましい。
尚、本明細書では、「cfu」は、colony forming unit(コロニー形成単位)を表す。また、本明細書では、前記ラクトバチルス・ヘルベティカスが死菌の場合、cfu/gまたはcfu/mlは、個細胞/gまたは個細胞/mlと置き換えることができる。

本発明の組成物の摂取量は、組成物の形態、用法、対象、対象の齢、性別、及びその他の条件等により適宜設定されるが、それを摂取した対象において、QOL改善作用、また、好ましい態様においては、不眠改善作用、疲労改善作用、又は整腸作用が発揮される限り特に制限されない。前記ラクトバチルス・ヘルベティカスの総量として、1日当たり、1×104~1×1013cfuの範囲内であることが好ましく、1×105~1×1012cfuの範囲内であることがより好ましく、1×106~1×1011cfuの範囲内であることがさらに好ましい。
尚、本明細書では、前記ラクトバチルス・ヘルベティカスが死菌の場合、cfuは、個細胞と置き換えることができる。
また、本発明の組成物は、1日1回又は複数回に分けて摂取することができる。また、数日又は数週間に1回の摂取としてもよいが、毎日摂取することが好ましい。また、摂取期間は5週間以上が好ましい。

また、本発明の飲食品組成物は、サプリメントであってもよく、例えばタブレット状のサプリメントであってもよい。サプリメントである場合には、一日当りの食事量及び摂取カロリーについて他の食品に影響されることなく、前記ラクトバチルス・ヘルベティカスを摂取できる。

本発明の飲食品組成物は、通常の飲食品の原料に、前記ラクトバチルス・ヘルベティカスを添加することにより製造することができ、前記ラクトバチルス・ヘルベティカスを添加すること以外は、通常の飲食品と同様にして製造することができる。前記ラクトバチルス・ヘルベティカスの添加は、飲食品組成物の製造工程のいずれの段階で行ってもよい。また、添加した前記ラクトバチルス・ヘルベティカスによる発酵工程を経て、飲食品組成物が製造されてもよい。そのような飲食品組成物としては、乳酸菌飲料、及び発酵乳等が挙げられる。また、搾乳された母乳や調製乳に添加する態様も挙げられ、添加後の乳を乳幼児に摂取させることも想定される。
本発明の飲食品組成物には、飲食品組成物製造のための原料、及び食品添加物等、飲食品組成物の製造工程又は製造後に飲食品組成物に添加されるものも含まれる。例えば、前記ラクトバチルス・ヘルベティカスは、発酵乳製造用スターターとして使用することができる。また、前記ラクトバチルス・ヘルベティカスを、製造された発酵乳に後から添加することもできる。
また、本発明の飲食品組成物には、本発明の効果を損なわない限り、公知の又は将来的に見出されるプレバイオティクス効果を有する成分又はプレバイオティクス効果を補助する成分(以下「プレバイオティクス」と称する。)を使用することができる。例えば、本発明の飲食品組成物は、ホエイタンパク質、カゼインタンパク質、大豆タンパク質、若しくはエンドウ豆タンパク質(ピープロテイン)等の各種タンパク質若しくはその混合物、分解物;ロイシン、バリン、イソロイシン若しくはグルタミン等のアミノ酸;ビタミンB6若しくはビタミンC等のビタミン類;クレアチン;クエン酸;フィッシュオイル;又は、イソマルトオリゴ糖、ガラクトオリゴ糖、キシロオリゴ糖、大豆オリゴ糖、フラクトオリゴ糖、ラクチュロース等のオリゴ糖等のプレバイオティクスと、前記ラクトバチルス・ヘルベティカスとを配合して製造することができる。
本発明の飲食品組成物における前記ラクトバチルス・ヘルベティカスの含量は、飲食品組成物の態様によって適宜設定されるが、総量で、1×104~1×1013cfu/gまたは1×104~1×1013cfu/mlの範囲内であることが好ましく、1×105~1×1012cfu/gまたは1×105~1×1012cfu/mlの範囲内であることがより好ましく、1×106~1×1011cfu/gまたは1×106~1×1011cfu/mlの範囲内であることがさらに好ましい。
本発明の飲食品組成物の摂取量は、飲食品組成物の形態、用法、対象、対象の齢、性別、及びその他の条件等により適宜設定されるが、それを摂取した対象において、QOL改善作用、また、好ましい態様における、不眠改善作用、疲労改善作用、又は整腸作用が発揮される限り特に制限されない。前記ラクトバチルス・ヘルベティカスの総量として、1日当たり、1×104~1×1013cfuの範囲内であることが好ましく、1×105~1×1012cfuの範囲内であることがより好ましく、1×106~1×1011cfuの範囲内であることがさらに好ましい。
尚、本発明の飲食品組成物は、1日1回又は複数回に分けて摂取することができる。また、数日又は数週間に1回の摂取としてもよいが、毎日摂取することが好ましい。また、摂取期間は5週間以上が好ましい。

本発明の飲食品組成物は、単独で摂取してもよいし、他の飲食品組成物若しくは飲食品又は医薬組成物若しくは医薬と共に摂取してもよい。例えば、QOLを改善するため他の飲食品組成物若しくは飲食品、又はQOLを改善するための医薬組成物若しくは医薬等と共に摂取してもよい。このことは、前記QOLを改善することが、不眠を改善すること、疲労を改善すること、整腸である場合も同様である。
本発明の飲食品組成物は、QOLを改善するため、また、好ましい態様における、不眠を改善するため、疲労を改善するため、又は整腸のため等との用途が表示された飲食品組成物又は飲食品として販売することができる。また、これ以外でも、上記用途によって二次的に生じる効果を表す文言であれば、使用できることはいうまでもない。
前記「表示」とは、需要者に対して上記用途を知らしめるための全ての行為を意味し、上記用途を想起・類推させうるような表示であれば、表示の目的、表示の内容、表示する対象物及び媒体等の如何に拘わらず、すべて本発明の「表示」に該当する。しかしながら、需要者が上記用途を直接的に認識できるような表現により表示することが好ましい。
具体的には、本発明の飲食品組成物又は飲食品に係る商品又は商品の包装に上記用途を記載する行為、商品又は商品の包装に上記用途を記載したものを譲渡し、引渡し、譲渡若しくは引渡しのために展示し、輸入する行為、商品に関する広告、価格表若しくは取引書類に上記用途を記載して展示し、若しくは頒布し、又はこれらを内容とする情報に上記用途を記載して電磁気的(インターネット等)方法により提供する行為等が例示でき、特に包装、容器、カタログ、パンフレット、POP等の販売現場における宣伝材、その他の書類等への表示が好ましい。
具体的には、本発明の飲食品組成物又は飲食品に係る商品又は商品の包装に上記用途を記載する行為、商品又は商品の包装に上記用途を記載したものを譲渡し、引渡し、譲渡若しくは引渡しのために展示し、輸入する行為、商品に関する広告、価格表若しくは取引書類に上記用途を記載して展示し、若しくは頒布し、又はこれらを内容とする情報に上記用途を記載して電磁気的(インターネット等)方法により提供する行為等が例示でき、特に包装、容器、カタログ、パンフレット、POP等の販売現場における宣伝材、その他の書類等への表示が好ましい。
また、表示としては、行政等によって許可された表示(例えば、行政が定める各種制度に基づいて認可を受け、そのような認可に基づいた態様で行う表示)であることが好ましい。例えば、保健機能食品など、より具体的には保健機能食品、健康食品、機能性食品、経腸栄養食品、特別用途食品、栄養機能食品、医薬用部外品等としての表示を例示することができ、その他消費者庁によって認可される表示、例えば、特定保健用食品、栄養機能食品、機能性表示食品、これに類似する制度にて認可される表示を例示できる。後者の例としては、特定保健用食品としての表示、条件付き特定保健用食品としての表示、身体の構造や機能に影響を与える旨の表示、疾病リスク低減表示、科学的根拠に基づいた機能性の表示等を例示することができる。さらに詳細には、健康増進法に規定する特別用途表示の許可等に関する内閣府令(平成二十一年八月三十一日内閣府令第五十七号)に定められた特定保健用食品としての表示(特に保健の用途の表示)、及びこれに類する表示等を例示することができる。
前記表示としては、本発明の飲食品組成物が、ラクトバチルス・ヘルベティカスを含む、QOLを改善するための飲食品組成物である場合には、例えば、「QOLを改善したい方へ」、「生活の質を改善したい方へ」等の表示が挙げられる。
前記表示としては、本発明の飲食品組成物が、ラクトバチルス・ヘルベティカスを含む、不眠を改善するための飲食品組成物である場合には、例えば、「不眠を改善したい方へ」、「寝床についてから実際に寝るまで時間がかかる方へ」、「夜間、睡眠の途中で目が覚める方へ」、「希望する起床時間より早く目覚めて、それ以降、眠れない方へ」、「夜の眠りや昼寝も合わせて、睡眠時間が足りていない方へ」、「全体的な睡眠の質について満足していない方へ」、「日中の気分が滅入っている方へ」、「日中の身体的および精神的な活動の状態が低下している方へ」、「日中の眠気がある方へ」、「起床時に眠気や疲労を感じすっきり起きられない方へ」、「睡眠状態がよくない方へ」等の表示が挙げられる。
前記表示としては、本発明の飲食品組成物が、ラクトバチルス・ヘルベティカスを含む、疲労を改善するための飲食品組成物である場合には、例えば、「疲労を改善したい方へ」、「疲労のある方へ」、「無気力の方へ」、「ぐったりしている方へ」、「つかれている方へ」、「へとへとである方へ」、「だるい状態にある方へ」、「物事にうんざりしている方へ」、及び「精根尽き果てた方へ」、「疲労を感じている方へ」、「疲れに関して問題がある方へ」、「もっと休養が必要である方へ」、「眠気またはだるさを感じている方へ」、「何かを始めるのが困難である方へ」、「問題なく何かを始めても、それを続けているうちに体が弱っていく方へ」、「行動力にかけている方へ」、「以前より筋力が衰えている方へ」、「身体が弱っていると感じる方へ」、「集中するのに苦労する方へ」、「物事を明確に考えるのが大変である方へ」、「話しているときに言い間違いをする方へ」、「的確な言葉を探すのに以前より苦労する方へ」、「記憶力がいつもより悪い方へ」、「以前は関心があったことに興味を持てなくなった方へ」、「体がだるい方へ」等の表示が挙げられる。
前記表示としては、本発明の飲食品組成物が、ラクトバチルス・ヘルベティカスを含む、整腸のための飲食品組成物である場合には、例えば、「腸機能を改善したい方へ」、「お腹の調子を整えたい方へ」、「お通じを改善したい方へ」、「便秘がちの方へ」、「お通じの悪い方へ」、「下痢気味の方へ」、「お腹のゆるい方へ」等の表示が挙げられる。
さらに、対象がヒトであるの本発明の飲食品組成物の一例として、調製乳が挙げられる。
調製乳には、調製粉乳、調製液状乳が含まれる。
調製粉乳は、乳および乳製品の成分規格等に関する省令(乳等省令)において、「生乳、牛乳、特別牛乳、またはこれらを原料として製造した食品を加工し、または主要原料とし、乳幼児に必要な栄養分を加え粉末状にしたもの」として定義される。
調製液状乳は、前記省令において、「生乳、牛乳、特別牛乳、またはこれらを原料として製造した食品を加工し、または主要原料とし、乳幼児に必要な栄養分を加え液状にしたもの」として定義される。
また、調製乳は、各種の蛋白質、油脂、炭水化物、ミネラル類、ビタミン類等の栄養成分が配合されたものであって、粉末状又は液状に加工されたものも含まれる。
また、調製乳にはさらに、健康増進法で規定される特別用途食品における「乳児用調製粉乳」、「乳児用調製液状乳」「妊産婦、授乳婦用粉乳」が含まれ、幼児向け調製粉乳、成人用栄養粉末、高齢者用栄養粉末等の態様も含まれる。

また、調製乳にはさらに、健康増進法で規定される特別用途食品における「乳児用調製粉乳」、「乳児用調製液状乳」「妊産婦、授乳婦用粉乳」が含まれ、幼児向け調製粉乳、成人用栄養粉末、高齢者用栄養粉末等の態様も含まれる。
本発明の医薬組成物は、前記ラクトバチルス・ヘルベティカスを含有する限り特に制限されない。本発明の医薬組成物としては、前記ラクトバチルス・ヘルベティカスをそのまま使用してもよく、生理的に許容される液体又は固体の製剤担体を配合し製剤化して使用してもよい。
本発明の医薬組成物の剤形は特に制限されず、具体的には、錠剤、丸剤、散剤、液剤、懸濁剤、乳剤、顆粒剤、カプセル剤、シロップ剤、坐剤、注射剤、軟膏剤、貼付剤、点眼剤、及び点鼻剤等を例示できる。また、製剤化にあたっては、製剤担体として通常使用される賦形剤、結合剤、崩壊剤、滑沢剤、安定剤、矯味矯臭剤、希釈剤、界面活性剤、又は注射剤用溶剤等の添加剤を使用することができる。
また、前記製剤担体としては、剤形に応じて、各種有機又は無機の担体を用いることができる。固形製剤の場合の担体としては、例えば、賦形剤、結合剤、崩壊剤、滑沢剤、安定剤、矯味矯臭剤等が挙げられる。
賦形剤としては、例えば、乳糖、白糖、ブドウ糖、マンニット、ソルビット等の糖誘導体;トウモロコシデンプン、馬鈴薯デンプン、α-デンプン、デキストリン、カルボキシメチルデンプン等のデンプン誘導体;結晶セルロース、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース、カルボキシメチルセルロース、カルボキシメチルセルロースカルシウム等のセルロース誘導体;アラビアゴム;デキストラン;プルラン;軽質無水珪酸、合成珪酸アルミニウム、メタ珪酸アルミン酸マグネシウム等の珪酸塩誘導体;リン酸カルシウム等のリン酸塩誘導体;炭酸カルシウム等の炭酸塩誘導体;硫酸カルシウム等の硫酸塩誘導体等が挙げられる。
結合剤としては、例えば、上記賦形剤の他、ゼラチン;ポリビニルピロリドン;マクロゴール等が挙げられる。
崩壊剤としては、例えば、上記賦形剤の他、クロスカルメロースナトリウム、カルボキシメチルスターチナトリウム、架橋ポリビニルピロリドン等の化学修飾されたデンプン又はセルロース誘導体等が挙げられる。
滑沢剤としては、例えば、タルク;ステアリン酸;ステアリン酸カルシウム、ステアリン酸マグネシウム等のステアリン酸金属塩;コロイドシリカ;ビーガム、ゲイロウ等のワックス類;硼酸;グリコール;フマル酸、アジピン酸等のカルボン酸類;安息香酸ナトリウム等のカルボン酸ナトリウム塩;硫酸ナトリウム等の硫酸塩類;ロイシン;ラウリル硫酸ナトリウム、ラウリル硫酸マグネシウム等のラウリル硫酸塩;無水珪酸、珪酸水和物等の珪酸類;デンプン誘導体等が挙げられる。
安定剤としては、例えば、メチルパラベン、プロピルパラベン等のパラオキシ安息香酸エステル類;クロロブタノール、ベンジルアルコール、フェニルエチルアルコール等のアルコール類;塩化ベンザルコニウム;無水酢酸;ソルビン酸等が挙げられる。
矯味矯臭剤としては、例えば、甘味料、酸味料、香料等が挙げられる。
なお、経口投与用の液剤の場合に使用する担体としては、水等の溶剤、矯味矯臭剤等が挙げられる。

本発明の医薬組成物における前記ラクトバチルス・ヘルベティカスの含量は、剤形、用法、対象、対象の齢、性別、疾患等の種類、その程度、及びその他の条件等により適宜設定されるが、総量で、1×104~1×1013cfu/gまたは1×104~1×1013cfu/mlの範囲内であることが好ましく、1×105~1×1012cfu/gまたは1×105~1×1012cfu/mlの範囲内であることがより好ましく、1×106~1×1011cfu/gまたは1×106~1×1011cfu/mlの範囲内であることがさらに好ましい。
対象に対する本発明の医薬組成物の投与量は、剤形、用法、対象、対象の齢、性別、疾患等の種類、その程度、及びその他の条件等により適宜設定されるが、それを摂取した対象において、QOL改善作用、また、好ましい態様における、不眠改善作用、疲労改善作用、又は整腸作用が発揮される限り特に制限されない。前記ラクトバチルス・ヘルベティカスの総量として、1日当たり、1×104~1×1013cfuの範囲内であることが好ましく、1×105~1×1012cfuの範囲内であることがより好ましく、1×106~1×1011cfuの範囲内であることがさらに好ましい。
本発明の医薬組成物の投与時期は特に限定されず、適宜投与時期を選択することが可能である。また、予防的に投与してもよい。また、投与形態は製剤形態、対象、対象の齢、性別、疾患等の種類、その程度、その他の条件等に応じて決定されることが好ましい。なお、本発明の医薬組成物は、いずれの場合も1日1回又は複数回に分けて投与することができる。また、数日又は数週間に1回の投与としてもよいが、毎日投与することが好ましい。また、投与期間は5週間以上が好ましい。
本発明の医薬組成物は、単独で投与してもよいし、他の医薬組成物若しくは医薬又は飲食品組成物若しくは飲食品と共に投与してもよい。例えば、QOLを改善するための他の医薬組成物若しくは医薬、又はQOLを改善するための飲食品組成物若しくは飲食品等と共に投与してもよい。このことは、前記QOLを改善することが、不眠を改善すること、疲労を改善すること、整腸である場合も同様である。

以下に実施例を用いて本発明を具体的に説明するが、本発明はこれら実施例に限定されるものではない。
<被験者>
以下の選択基準に該当し、除外基準に該当しないものを対象とした。条件を満たす13人を被験者とした。

2)POMS 2短縮版 FI(疲労-無気力)スコアが2回のスクリーニングとも50以上の者
(除外基準)
1)肝、腎、心、肺、消化器、血液、内分泌系および代謝系等の治療を受けている者またはこれらの重篤な既往歴がある者
2)乳酸菌・ビフィズス菌配合食品(サプリメントなど、ヨーグルトを除く)を常用している者
3)気分や疲労に影響を与える医薬品、サプリメントを常用している者
4)薬物アレルギーまたは重篤な食物アレルギーを有する者または既往を有する者
5)妊娠している者、試験期間中妊娠の意思がある者、授乳中の者
6)過去1ヶ月以内に他の医薬品や食品の試験に参加した者
7)被験者背景、身体所見、診察、理学的検査などの結果から、試験責任者または試験責任医師により被験者として不適当と判断された者
本試験の実施に際しては、事前に被験者全員に試験の主旨を十分に説明したうえで、本人の自由意思で書面による参加の同意を得た。本試験はヘルシンキ宣言を遵守し、特定非営利活動法人 全国臨床研究協議会 倫理審査委員会の承認を得たうえで、被験者の安全を第一に考慮して実施した。
以下の選択基準に該当し、除外基準に該当しないものを対象とした。条件を満たす13人を被験者とした。

[実施例1]
<試験品の製造>
ラクトバチルス・ヘルベティカスMCC1848(NITE BP-01671)の菌体培養液を加熱殺菌し、濃縮、乾燥させた。この菌体乾燥物をマルトデキストリンと混合し、2×1010個細胞/gの菌末を得た。この菌末を1包1 gのスティックフィルム袋に小分けし、試験品とした。
<試験品の製造>
ラクトバチルス・ヘルベティカスMCC1848(NITE BP-01671)の菌体培養液を加熱殺菌し、濃縮、乾燥させた。この菌体乾燥物をマルトデキストリンと混合し、2×1010個細胞/gの菌末を得た。この菌末を1包1 gのスティックフィルム袋に小分けし、試験品とした。
<試験方法>
単群前後比較試験を実施した。被験者は、試験品を1日1回、1包(ラクトバチルス・ヘルベティカスMCC1848(NITE BP-01671)の加熱殺菌体2×1010個細胞)を水などに溶かし毎朝朝食前に5週間にわたり摂取した。摂取前、摂取後において、各種検査及びアンケートを行った。
単群前後比較試験を実施した。被験者は、試験品を1日1回、1包(ラクトバチルス・ヘルベティカスMCC1848(NITE BP-01671)の加熱殺菌体2×1010個細胞)を水などに溶かし毎朝朝食前に5週間にわたり摂取した。摂取前、摂取後において、各種検査及びアンケートを行った。
<統計解析>
被験者13名全員を解析対象とし、試験品摂取前の各種検査の数値と試験品摂取後の各種検査の数値を対応のあるt検定によって評価した。試験後のアンケートでは、「変化しない」を基準としたときの1変量t検定によって評価した。

<結果>
(POMS2短縮版の結果)
POMS2短縮版の項目「疲労-無気力(FI)」の数値を表1に示す。試験品摂取前の数値を基準として、摂取後では有意に数値が低下した。

(アンケートの結果)
アンケートの結果を表5に示す。「お腹の調子」、「疲労感・体のだるさ」、「睡眠の質・目覚めの良さ」のすべての項目において、試験品摂取前を基準として、「変わらない」、「少し悪くなった」、「悪くなった」と回答した人数よりも、「良くなった」、「少し良くなった」と回答した人数が有意に多かった。
アンケートの結果を表5に示す。「お腹の調子」、「疲労感・体のだるさ」、「睡眠の質・目覚めの良さ」のすべての項目において、試験品摂取前を基準として、「変わらない」、「少し悪くなった」、「悪くなった」と回答した人数よりも、「良くなった」、「少し良くなった」と回答した人数が有意に多かった。
Claims (8)
- ラクトバチルス・ヘルベティカスを含む、不眠を改善するための組成物。
- 前記不眠がアテネ不眠尺度により評価される項目の合計得点で表される不眠である、請求項1に記載の組成物。
- ラクトバチルス・ヘルベティカスを含む、疲労を改善するための組成物。
- 前記疲労が下記(1)~(5)からなる群から選択される一又は複数である、請求項3に記載の組成物。
(1)POMS(登録商標)により評価される項目「疲労」の数値で表される疲労
(2)POMS2(商標)により評価される項目「疲労-無気力」の数値で表される疲労
(3)VASにより評価される項目「疲労を感じている」の数値で表される疲労
(4)VASにより評価される項目「ぐったりしている」の数値で表される疲労
(5)CFSにより評価される項目の合計得点で表される疲労 - ラクトバチルス・ヘルベティカスを含む、整腸のための組成物。
- 前記ラクトバチルス・ヘルベティカスが、ラクトバチルス・ヘルベティカスMCC1848 (NITE BP-01671)である、請求項1~5のいずれか一項に記載の組成物。
- 飲食品組成物である、請求項1~6のいずれか一項に記載の組成物。
- 医薬組成物である、請求項1~6のいずれか一項に記載の組成物。
