Abstract
本发明涉及一种富硒营养强化剂的制备方法,步骤如下:一、提取天然有机硒母料,包括将天然原料清洗、再经粉碎、超微研磨、酶解、酶灭活、离心分离、纳滤、浓缩干燥等一系列工序制得天然有机硒提取物。二、复配:将天然有机硒提取物与抗氧化剂、维生素、氨基糖苷抗生素、蛋白质粉、矿物质按质量比(1.2‑1.5):(0.8‑1.2):(0.2‑0.5):(0.2‑0.5):(0.6‑1.2)混合均匀混合均匀,用100‑120目筛子过筛,得到得到复配后的富硒营养强化剂。本发明制备的富铬食品抗氧化剂安全无毒、稳定性高,具有抗氧化、预防胆固醇、心脑血管等多种疾病的功效。
Description
技术领域
本发明涉及一种营养强化剂的制备方法,特别是涉及一种富硒营养强化剂的制备方法。
背景技术
硒是联合国卫生组织所确定的人体所必需的微量元素,是人和动物体内谷胱甘肽过氧化物酶(glutathione per-oxidase,GSHPX)的辅助因子,GSHPX广泛存在于生命机体的肝、肾、心、肺等,硒代替半胱氨酸为该酶提供其活性中心的必需基团,GSHPX利用谷胱甘肽使有毒性的过氧化物还原为无害的羟基化合物,使过氧化物分解,清除活性自由基部位,保护细胞膜结构和功能,修复分子损伤。因此,硒元素具有抗氧化、抗癌、抗衰老、增强机体免疫力等多种生物学功能。缺硒会导致人体克山病、大骨节病和心血管等疾病,硒摄入过多则会引起中毒,因此硒的摄入量对人体具有重要的意义。世界上许多国家都规定了每人每天的硒元素摄入量,1976年国际硒学会推荐标准是60ug,美国科学院食品与营养委员会的日推荐标准是50-200ug,中国营养学会1988年推荐标准是每天50ug。
目前,普通的水果蔬菜中的硒含量较低,谷物、海洋动物、肉类(特别是肝脏)中的硒含量稍微较高,但多是大分子硒,吸收较为困难,亚硒酸钠之类的无机硒自带毒性,不能直接用于食品中,天然植物中的硒元素提取又受生长土壤的限制,其来源很不稳定,难以满足人体摄入硒元素的需要。因此开发含有硒元素的食品添加剂是很有意义的,目前,通过微生物合成法富集硒元素是一种开发硒食品安全有效的途径,其中,国内外专家对富硒酵母的研究较为广泛,国家发明专利CN 103911299 B公开了一种富硒酵母及其制备方法,通过在酵母菌株中加入富硒酵母培养物,所述富硒酵母培养物包括亚硒酸盐、氨基酸及其它营养元素,制得的富硒酵母中的硒含量在3000ppm以上,营养均衡且容易吸收。国家发明专利CN 106906206 A 公开了一种富硒酵母,将乙醇假丝酵母Candidaethanolica进行氮离子注入诱变,经多次诱变筛选,获得耐受高浓度亚硒酸钠的乙醇假死酵母突变菌种,将种子培养液接种到含有发酵培养基中,发酵至对数生长期,在发酵产物中补加亚硒酸钠,继续发酵培养,最后经离心、干燥后制得淡黄色粉末状产品。
上述的方法,虽然实现了硒元素的大批量生产,但都是利用无机硒及其它营养元素配制成营养液,对酵母进行发酵或者突变,以得到富硒酵母,其生产工艺复杂,发酵或者突变的周期长,且亚硒酸盐或者硒酸盐毒性较高,分子活性高,很容易分解或者与其它分子结合形成新的化合物,不能保证100%得到富硒酵母。因此,我们新开发一种富硒营养强化剂,直接从富硒麦芽、海藻或螺旋藻等天然物质中提取有机硒,将其化学分解为更容易吸收的小分子硒,另加入其它植物营养元素、氨基酸及其它含氮化合物等物质,得到一种无污染、硒含量高、无毒无害的富硒营养强化剂。
发明内容
为解决现有技术中发酵时间长,亚硒酸盐或者硒酸盐自带毒性,且分子活性较高,容易分解或被氧化,本发明提供一种制造安全无毒、稳定性高,具有抗氧化、防癌、抗衰老性能的富硒营养强化剂的方法。
本发明解决其技术问题所采用的技术方案是:
一、提取天然有机硒母料
(1)、清洗:将一定量的天然原料用30-40℃的水洗净2-3遍并烘干;所述天然原料为富硒麦芽、海藻或螺旋藻。
(2)、粉碎:将洗净的天然原料用粉碎机进行粉碎,过100目筛。
(3)、超微研磨:利用球磨仪对粉碎后的粒子进行超微研磨,研磨时间10-30min,功率10-30HZ,得到酶解原料,放入0-3℃下进行保藏;球磨中加入适量保护液,所述保护液由下列重量份的原料加工而成:氧化型谷胱甘肽1-3份,黄原胶3-6份,燕麦β-葡聚糖5-10份,蜂胶粉1-3份,聚赖氨酸0.5-1份,60-80份去离子水。
天然原料研磨处理最大的技术障碍是避免营养成分流失、维持研磨液新鲜、原料组织不萎缩变质,本发明采用燕麦β-葡聚糖、氧化型谷胱甘肽、蜂胶和聚赖氨酸的水溶液进行预处理,很好了解决了这一技术障碍,燕麦β-葡聚糖能在原料组织表面形成一层保护膜,且与组织细胞之间物质交联,避免营养成分流失,实践中还发现,黄原胶的添加,可以使得均质液均一稳定性好,长时间放置不会分层,可以显著改善酶解效果,该保护膜中还均匀分布有氧化型谷胱甘肽,与组织细胞中固有的还原型谷胱甘肽互补协同作用,实现高效抗氧化功能,避免原料的有益成分被氧化,避免组织细胞变质品质下降,避免制得的酶解粉褐变,另外,蜂胶和聚赖氨酸具有天然的抑菌杀菌性能,有效避免预处理和均质过程中杂菌的滋生,维持均质液的新鲜度;
(4)、酶解:将步骤(3)中的酶解原料与水1:5(重量)混合,并搅拌均匀,调节PH值到5.6-6.7左右,加入0.8%-1.2%的复合酶并不断搅拌,酶解时间2-4h,酶解温度45-55℃;所述的复合酶包括蛋白酶、纤维素酶、淀粉酶按重量比(0.8-1.2):(0.8-1.2):(0.5-1.2)组成。
(5)、酶灭活:酶解结束后,酶解液于110-120℃、加热3-5s灭酶,降温至30-50℃.
(6)、离心分离:将灭活后的有机硒提取液在离心机中进行离心分离,得到有机硒清液,离心机转速4000-6000rmp,离心时间5-10min。
(7)、纳滤:将有机硒清液用纳米膜进行透析浓缩,所述的纳米膜截留分子量为150-500Da,得到过滤后的有机硒提取液。
(8)、浓缩干燥:将过滤后的有机硒提取液在温度60-80℃、真空度-0.075-0.095mpa下进一步浓缩至固形物含量为30-40%,然后再喷雾干燥器中干燥得到含水率为4%-7%的粉末状有机硒提取物。
前述步骤(3)中氧化型谷胱甘肽的制备方法步骤如下:
1)取还原型谷胱甘肽,于40-45℃溶解,获得50-100g/L的还原型谷胱甘肽溶液,测其pH值为A1;
2)用氢氧化钡调pH至A2,7.0≤A2≤7.2;
3)加入一定量双氧水,边加边搅拌,加完后继续搅拌反应0.2-0.5h;
4)反应结束后,加入一定量二氧化锰,搅拌5-10min,然后充分滤去二氧化锰;实践中发现,多余的双氧水必需除去,否则干燥过程中被氧化的谷胱甘肽20-30%会恢复到还原型状态,影响氧化型谷胱甘肽的纯度;
5)用硫酸调节pH至A3,A1-0.1≤A3≤A1;
6)充分滤去硫酸钡沉淀,喷雾干燥,获得氧化型谷胱甘肽。
二、复配
将天然有机硒提取物与抗氧化剂、维生素、氨基糖苷抗生素、蛋白质粉、矿物质按质量比(1.2-1.5):(0.8-1.2):(0.2-0.5):(0.2-0.5):(0.6-1.2)混合均匀,用100-120目筛子过筛,得到得到复配后的富硒营养加强剂。
所述的抗氧化剂为天然VE、改性葡聚糖、木糖醇或麦芽糖醇。
所述维生素为VB1(硫胺素)、VB2(核黄素)、VB11叶酸或维生素C。
所述蛋白质粉为:酪蛋白、乳清蛋白或豌豆蛋白。
所述矿物质为:氯化钙、氯化钾或氯化钠。
本发明的有益效果是:
1、利用球磨仪对粉碎后的原材料颗粒进一步研磨,使得其能更充分的被酶催化水解,减少了酶催化的时间,使得大分子的有机硒更容易被分解成小分子有机硒,更有利于被人体吸收。
2、通过在球磨的时候加入保护液,避免原料在球磨的过程中被氧化或者在高温下被分解,同时起到了一定的抑菌作用,使得研磨的效果更好,得到纯度更高的有机硒提取液。
3、利用离心机对其离心分离,可得到纯度更纯的有机硒提取液,最后经浓缩干燥后得到的的粉末状有机硒容易保存,不易变质。
4、通过在有机硒提取物中加入抗氧化剂、维生素、氨基糖苷抗生素、蛋白质粉、矿物质制得富硒营养强化剂,兼具抗氧化、美白、补钙、防癌等多重功效,易于生产,对人体无副作用。
具体实施方式
下面通过具体实施例,对本发明的技术方案作进一步的具体说明。
实施例1:
一、提取天然有机硒母料
(1)、清洗:将富硒麦芽150g用30-40℃的水洗净2-3遍并烘干。
(2)、粉碎:将烘干后的富硒麦芽用粉碎机进行粉碎,过100目筛。
(3)、超微研磨:利用球磨仪对粉碎后的富硒麦芽粒子进行超微研磨,研磨时间20min,功率20HZ,得到酶解原料,放入3℃下进行保藏;球磨中加入适量保护液,所述保护液由下列重量份的原料加工而成:氧化型谷胱甘肽1份,黄原胶3份,燕麦β-葡聚糖5份,蜂胶粉1份,聚赖氨酸0.5份,60份去离子水。
(4)、酶解:将步骤(3)中的酶解原料与水1:5(重量)混合,并搅拌均匀,调节PH值到6,加入0.8%重量比为0.8:0.5的纤维素酶和淀粉酶并不断搅拌,酶解时间2h,酶解温度45℃。
(5)、酶灭活:酶解结束后,酶解液于110℃、加热5s灭酶,降温至30℃;
(6)、离心分离:将灭活后的有机硒提取液在离心机中进行离心分离,得到有机硒清液,离心机转速4000rmp,离心时间5min。
(7)、纳滤:将有机硒清液用纳米膜进行透析浓缩,所述的纳米膜截留分子量为150Da,的到浓缩后的有机硒提取液。
(8)、浓缩干燥:将浓缩后的有机硒提取液在温度60℃、真空度-0.075mpa下进一步浓缩至固形物含量为30%,然后再喷雾干燥器中干燥得到含水率为4%的粉末状有机硒提取物。
前述步骤(3)中氧化型谷胱甘肽的制备方法步骤如下:
1)取还原型谷胱甘肽,于40℃溶解,获得50g/L的还原型谷胱甘肽溶液,测其pH值为A1。
2)用氢氧化钡调pH至A2=7.0。
3)加入一定量双氧水,边加边搅拌,加完后继续搅拌反应0.2h。
4)反应结束后,加入一定量二氧化锰,搅拌5min,然后充分滤去二氧化锰;
5)用硫酸调节pH至A3=A1-0.1。
6)充分滤去硫酸钡沉淀,喷雾干燥,获得氧化型谷胱甘肽。
二、复配
将天然有机硒提取物与抗氧化剂(天然VE)、维生素(VB1硫胺素)、蛋白质(酪蛋白)、矿物质(氯化钙)按1.2:0.8:0.2:0.2:0.6混合均匀,用100目筛子过筛,得到得到复配后的富硒营养加强剂。
实施例二:
一、提取天然有机硒母料
(1)、清洗:将180g海藻用30-40℃的水洗净2-3遍并烘干。
(2)、粉碎:将烘干后的海藻用粉碎机进行粉碎,过100目筛。
(3)、超微研磨:利用球磨仪对粉碎后的海藻粒子进行超微研磨,研磨时间25min,功率25HZ,得到酶解原料,放入3℃下进行保藏;球磨中加入适量保护液,所述保护液由下列重量份的原料加工而成:氧化型谷胱甘肽2份,黄原胶4.5份,燕麦β-葡聚糖7.5份,蜂胶粉3份,聚赖氨酸0.75份, 80份去离子水。
(4)、酶解:将步骤(3)中的酶解原料与水1:5(重量)混合,并搅拌均匀,调节PH值到6左右,加入0.8%重量比为0.8:1.2的纤维素酶、淀粉酶并不断搅拌,酶解时间3h,酶解温度50℃。
(5)、酶灭活:酶解结束后,酶解液于115℃、加热4s灭酶,降温至40℃;
(6)、离心分离:将灭活后的有机硒提取液在离心机中进行离心分离,得到有机硒清液,离心机转速5000rmp,离心时间7min。
(7)、纳滤:将有机硒清液用纳米膜进行透析浓缩,所述的纳米膜截留分子量为250Da,的到浓缩后的有机硒提取液。
(8)、浓缩干燥:将浓缩后的有机硒提取液在温度70℃、真空度0.045mpa下进一步浓缩至固形物含量为35%,然后再喷雾干燥器中干燥得到含水率为5%的粉末状有机硒提取物。
前述步骤(3)中氧化型谷胱甘肽的制备方法步骤如下:
1)取还原型谷胱甘肽,于42℃溶解,获得75g/L的还原型谷胱甘肽溶液,测其pH值为A1。
2)用氢氧化钡调pH至A2=7.1。
3)加入一定量双氧水,边加边搅拌,加完后继续搅拌反应0.35h;
4)反应结束后,加入一定量二氧化锰,搅拌7.5min,然后充分滤去二氧化锰;
5)用硫酸调节pH至A3=A1-0.05。
6)充分滤去硫酸钡沉淀,喷雾干燥,获得氧化型谷胱甘肽。
二、复配
将天然有机硒提取物与抗氧化剂(改性葡聚糖、木糖醇)、维生素(VB2核黄素)、蛋白粉(豌豆蛋白)、矿物质(氯化钙、氯化钾)按1.35:1:0.35:0.35:0.9混合均匀,用110目筛子过筛,得到得到复配后的富硒营养加强剂。
实施例3:
一、提取天然有机硒母料
(1)、清洗:将200g的螺旋藻用30-40℃的水洗净2-3遍并烘干。
(2)、粉碎:将烘干后的螺旋藻用粉碎机进行粉碎,过100目筛。
(3)、超微研磨:利用球磨仪对粉碎后的螺旋藻粒子进行超微研磨,研磨时间30min,功率30HZ,得到酶解原料,放入3℃下进行保藏;球磨中加入适量保护液,所述保护液由下列重量份的原料加工而成:氧化型谷胱甘肽2份,黄原胶4.5份,燕麦β-葡聚糖7.5份,蜂胶粉2份,聚赖氨酸0.75份,70份去离子水。
(4)、酶解:将步骤(3)中的酶解原料与水1:5(重量)混合,并搅拌均匀,调节PH值到6.7左右,加入1.2%的蛋白酶、淀粉酶并不断搅拌,酶解时间4h,酶解温度55℃。
(5)、酶灭活:酶解结束后,酶解液于120℃、加热5s灭酶,降温至50℃;
(6)、离心分离:将灭活后的有机硒提取液在离心机中进行离心分离,得到有机硒清液,离心机转速6000rmp,离心时间10min。
(7)、纳滤:将有机硒清液用纳米膜进行透析浓缩,所述的纳米膜截留分子量为500Da,的到浓缩后的有机硒提取液。
(8)、浓缩干燥:将浓缩后的有机硒提取液在温度80℃、真空度0.095mpa下进一步浓缩至固形物含量为40%,然后再喷雾干燥器中干燥得到含水率为7%的粉末状有机硒提取物。
前述步骤(3)中氧化型谷胱甘肽的制备方法步骤如下:
1)取还原型谷胱甘肽,于45℃溶解,获得100g/L的还原型谷胱甘肽溶液,测其pH值为A1。
2)用氢氧化钡调pH至A2=7.2。
3)加入一定量双氧水,边加边搅拌,加完后继续搅拌反应0.5h。
4)反应结束后,加入一定量二氧化锰,搅拌10min,然后充分滤去二氧化锰。
5)用硫酸调节pH至A3=A1。
6)充分滤去硫酸钡沉淀,喷雾干燥,获得氧化型谷胱甘肽。
二、复配
将天然有机硒提取物与抗氧化剂(改性葡聚糖、麦芽糖醇)、维生素(VB11叶酸、维生素C)、蛋白质(乳清蛋白)、矿物质(氯化钠、氯化钙)按1.5:1.2:0.5:0.5:1.2混合均匀,用100-120目筛子过筛,得到得到复配后的富硒营养加强剂。
实施例1-3制备得到的富硒营养强化剂性能评价:
将普通营养强化剂与实施例得到的营养强化剂进行营养元素含量分 析,结论如下表所示:
综上:根据GB/T14880-2012食品营养强化剂使用标准,该营养强化剂 中各项营养元素的含量均没有超出国家规定的限制,且该营养强化剂相对 普通营养强化剂硒含量增加近一倍,能满足人体对微量元素硒的摄取。
将普通营养强化剂与实施例得到的营养强化剂用真空包装(真空度0.08MPa),放在温度4℃、20℃、30℃下贮藏6个月,每隔一个月测定其水分含量及营养元素含量,考察其变化趋势,得出实施例中的营养强化剂耐低温及耐高温的性能较好,在温度为4℃和30℃时,实施例中营养强化剂的各组分的营养元素随贮存时间的推移变化不明显,几乎没有营养元素的流失,营养保存率较高,稳定性较好。而普通营养强化剂在过高温度时容易氧化分解,且营养元素流失很快。
以上所述的实施例只是本发明的一种较佳的方案,并非对本发明作任何形式上的限制,在不超出权利要求所记载的技术方案的前提下还有其它的变体及改型。
Claims (9)
1.一种富硒营养强化剂的制备方法,其特征在于:所述方法步骤如下:
S1:清洗:将富含有机硒的天然原料用30-40℃的水洗净2-3遍并烘干;
S2:粉碎:将洗净的原料用粉碎机进行粉碎,过100目筛;
S3:超微研磨:利用球磨仪对粉碎后的原料进行超微研磨,研磨时间10-30min,得到酶解原料,放入0-3℃下进行保藏,超微研磨中加入适量保护液;
S4:酶解:将超微研磨后的酶解原料与水质量比1:5混合,并搅拌均匀,调节pH值到5.6-6.7左右,加入原料质量0.8%-1.2%的复合酶并不断搅拌,酶解时间2-4h,酶解温度45-55℃;
S5:酶灭活:酶解结束后,酶解液于110-120℃、加热3-5s灭酶,降温至30-50℃;
S6:离心分离:将灭活后的酶解液在离心机中进行离心分离,离心机转速4000-6000rmp,离心时间5-10min,得到富含有机硒的酶解清液;
S7:纳滤:将富含有机硒的酶解清液用纳米膜进行透析浓缩,得到过滤后的富含有机硒的酶解清液;
S8:浓缩干燥:将过滤后的富含有机硒的酶解清液在温度60-80℃、真空度-0.075-0.095mpa下进一步浓缩至固形物含量为30-40%,然后再喷雾干燥器中干燥得到含水率为4%-7%的粉末状有机硒提取物;
S9:复配:将有机硒提取物与抗氧化剂、维生素、蛋白质粉、矿物质按质量比(1.2-1.5):(0.8-1.2):(0.2-0.5):(0.2-0.5):(0.6-1.2)混合均匀,用100-120目筛子过筛,得到复配后的富硒营养强化剂。
2.如权利要求1所述的富硒营养强化剂的制备方法,其特征在于:所述富含有机硒的天然原料为富硒麦芽、海藻或螺旋藻。
3.如权利要求1所述的富硒营养强化剂的制备方法,其特征在于:所述保护液由下列重量份的原料加工而成:氧化型谷胱甘肽1-3份,黄原胶3-6份,燕麦β-葡聚糖5-10份,蜂胶粉1-3份,聚赖氨酸0.5-1份,60-80份去离子水。
4.如权利要求1所述的富硒营养强化剂的制备方法,其特征在于:所述的复合酶由蛋白酶、纤维素酶和淀粉酶按重量比(0.8-1.2):(0.8-1.2):(0.5-1.2)组成。
5.如权利要求1所述的富硒营养强化剂的制备方法,其特征在于:所述的抗氧化剂为天然VE、改性葡聚糖、木糖醇或麦芽糖醇的一种或几种组合。
6.如权利要求1所述的富硒营养强化剂的制备方法,其特征在于:所述的维生素为VB1、VB2、VB11或维生素C的一种或几种组合。
7.如权利要求1所述的富硒营养强化剂的制备方法,其特征在于:所述的蛋白质粉为:酪蛋白、乳清蛋白或豌豆蛋白的一种或几种组合。
8.如权利要求1所述的富硒营养强化剂的制备方法,其特征在于:所述矿物质为氯化钙、氯化钾或氯化钠的一种或几种组合。
9.如权利要求3所述的富硒营养强化剂的制备方法,其特征在于:
所述氧化型谷胱甘肽的制备方法步骤如下:
1)取还原型谷胱甘肽,于40-45℃溶解,获得50-100g/L的还原型谷胱甘肽溶液,测其pH值为A1;
2)用氢氧化钡调pH至A2,7.0≤A2≤7.2;
3)加入一定量双氧水,边加边搅拌,加完后继续搅拌反应0.2-0.5h;
4)反应结束后,加入一定量二氧化锰,搅拌5-10min,然后充分滤去二氧化锰;
5)用硫酸调节pH至A3,A1-0.1≤A3≤A1;
6)充分滤去硫酸钡沉淀,喷雾干燥,获得氧化型谷胱甘肽。
