有鉴于此,我们不可能单单靠饮食摄取足够的微量营养素。微量营养素能促进免疫系统的正常运行,并使解毒和细胞修复机制能够正常运作,以保护我们免受慢性病的侵害。充分吸收微量营养素,如维他命、矿物质和许多其他植物素加上适当的食物进取是维持健康身体最重要的一环。因此,人们从植物中提取无机的微量营养素以弥补饮食中缺失的营养,但是目前的提取工艺得到的微量元素往往并不能很好的被人体吸收和利用。
发明内容
为了解决现有技术中存在的上述问题,本发明提供一种富含白藜芦醇、花青素以及多种微量元素的提取物及其制备方法,本发明所述提取物的吸收好,生物利用度高,具有显著抗氧化延缓衰老、养护心脑血管的作用。
本发明的技术方案为:
一种抗氧化延缓衰老、养护心脑血管的提取物的制备方法,包括如下步骤:
(1)制备葡萄浸膏
从葡萄中取葡萄皮和葡萄籽,得葡萄皮和葡萄籽混合物;向所述葡萄皮和葡萄籽混合物中加入第一复合酶进行酶解,得到酶解液;向所述酶解液中加入乙醇,超声波粉碎后,依次进行萃取、过滤、浓缩,得到葡萄浸膏;
(2)制备混合果子粉
将黑加仑、覆盆子、蓝莓清洗后打碎,得到混合果肉;向所述混合果肉中加入第二复合酶进行酶解,得到酶解液;将所述酶解液依次进行灭菌、过滤、浓缩、干燥,得到混合果子粉;
(3)制备抗氧化延缓衰老、养护心脑血管的提取物
酵母菌活化后,经培养,得到种子液;向所述种子液中加入步骤(1)所述葡萄浸膏和步骤(2)所述混合果子粉,经发酵培养得到发酵液;将所述发酵液进行固液分离,得到上清液和酵母菌体;将所述上清液依次进行干燥、灭菌,即得所述抗氧化延缓衰老、养护心脑血管的提取物。
进一步地,步骤(1)中,所述第一复合酶为糖苷酶和纤维素酶的混合物,所述糖苷酶、纤维素酶、葡萄皮和葡萄籽混合物的质量之比为(0.5-1.5):(3-7):(1-3);所述酶解反应的温度为35-40℃,所述酶解反应的时间为7-9h,所述酶解反应的pH值为7.5-8。
进一步地,步骤(1)中,所述乙醇与酶解液的质量之比为1:4-1:6;所述超声波粉碎的时间为35-55min;所述萃取为微波萃取,所述微波萃取的温度为30-40℃,所述微波萃取的功率为400-600w,所述微波萃取的时间为3-7min;所述浓缩为减压浓缩,所述减压浓缩的温度为50-60℃,所述减压浓缩的真空度为-80cmHg以下。
进一步地,步骤(2)中,所述黑加仑、覆盆子、蓝莓的质量之比为(1-3):(1-3):(1-3);所述第二复合酶为糖苷酶、木瓜蛋白酶、果胶酶和纤维素酶的混合物,所述糖苷酶、木瓜蛋白酶、果胶酶、纤维素酶以及混合果肉的质量之比为(0.5-1.5):(0.5-1.5):(0.5-1.5):(3-7):(8-12);所述酶解反应的温度为35-40℃,所述酶解反应的时间为7-9h,所述酶解反应的pH值为7.5-8。
进一步地,步骤(2)中,所述灭菌的温度为90℃,所述灭菌的时间为8-12min,进行所述灭菌2次以上;所述过滤后的滤液中固形物的粒径为20-40目;所述浓缩为在温度为35-45℃的条件下加热0.5-1.5h。
进一步地,步骤(2)中,所述干燥为喷雾干燥,所述喷雾干燥的进料流量 为15-20mL/min,进风温度为120-160℃,热空气流量为30-40m 3/h,压缩空气流量为460-520L/h,出风温度为120-140℃。
进一步地,步骤(3)中,所述种子液的体积、葡萄浸膏的体积和混合果子粉的质量之比为(800-1200mL):(80-120mL):(200-400g)。
进一步地,步骤(3)中,所述发酵培养的温度为32-42℃,所述发酵培养的搅拌转速为160-200rpm,所述发酵培养的时间为22-26h。

所述方法制备得到的抗氧化延缓衰老、养护心脑血管的提取物。
葡萄皮和葡萄籽,富含白藜芦醇,含量分别为3.44mg/g和12.4mg/g,并以反式白藜芦醇的形式存在。白藜芦醇至少在21科、31属的72种植物中以糖苷的形式存在,它是许多植物受到真菌感染、紫外线照射或病理情况下产生的一种植物抗毒素,可以提高植物抵抗病原体的能力。白藜芦醇耐高温性差、容易失去活性、提取出来的成品不稳定,必须要在比较温和的条件下提取。白藜芦醇因结构不稳定,在血液中滞留时间较短,生物活性低。已有研究证实,白藜芦醇作为化学预防剂在肿瘤的启动、促进及发展3个阶段都具有抗肿瘤的功效,目前认为,其作用机制包括抑制细胞色素酶、诱导解毒酶、抑制环氧化酶、抑制蛋白激酶、拮抗雌激素受体、促进肿瘤细胞分化和凋亡。此外,白藜芦醇可增强颈动脉血中一氧化碳的含量,降低乳酸脱氢酶的含量,提高血管的张发力和增加动脉流量,减少心肌梗死的面积,透过抑制血小板凝聚和调节血脂水平两种机制来发挥对心血管疾病的预防作用。透过抑制人体低密度蛋白(LDL)的氧化,还能抑制膜脂的过氧化,减少H 2O 2的产生,起到抗氧化、抗自由基的作用。在大约500种不同的抗氧化剂中,最近的研究表明,白藜芦醇和原花青素是葡萄酒中两种最重要的多酚类抗氧化剂,它能减轻各种健康问题。营养与中间代谢(Nutrition&Intermediary Metabolism)杂志不久前发布的一项研究发现,白藜芦醇有益于II型糖尿病患者扩张脑补血管,促进脑部血液循环。多项 研究显示,饮用含有白藜芦醇的红酒有助于延缓增龄性认知衰减。这种保护效果可能与其抗氧化和抗炎活性相关。此外,白藜芦醇能够保护受损的脑细胞。
黑加仑,维生素C含量突出,达到464mg/100g,烟酸、泛酸的含量也相对较高。黑加仑中的色素含量很丰富,达4.53%,黑加仑色素是一种水溶性天然色素,属花色苷类,着色力强,主要成份是花青素-3-葡糖苷、花青素-3-芸香苷、翠雀素-3-葡糖苷、翠雀素-3-芸香苷四大类。花青素可以抑制、阻碍由胶原、花生烯酸、肾上腺素、三磷酸腺苷等引起的血小板凝固,从而能预防血管内血小板凝固引起的血栓及动脉硬化。黑加仑果汁的总酚类含量达到2863mg/100g,抗氧化力达到27.86TEACmmol/100g,赋予其清除体内自由基、延缓机体衰老的功能。黑加仑浓缩汁中含有常量元素6种、人体必需微量元素7种,人体可能必需微量元素4种,另外还含有其它种类元素17种。常量元素Ca、Mg、P、K含量很丰富,特别是Ca的含量为水果之王,含量达到83mg/100g,另外对人体直接或间接有益的Fe、Zn、Cu、Mn、Ni、B、Ba、Rb、Sr等微量元素含量也较丰富。
覆盆子,富含花青素,花青素是一种有着特殊分子结构的生物类黄酮,是目前国际上公认的清除人体内自由基最有效的天然抗氧化剂,具有非常强的体内活性。实验证明,其抗氧化能力是VE的50倍,VC的20倍。具有预防癌症、增进视力、清除体内有害的自由基、改善睡眠、调节内分泌紊乱、加固血管、抗过敏、抗突变、抗辐射、改善循环、预防心脑血管疾病,保护肝脏的作用,被称为“口服的皮肤化妆品”。不仅可防止皮肤皱纹的提早生成,更能补充营养及消除体内有害的自由基,是天然的阳光遮盖物,能够阻止紫外线侵害皮肤。在实验中发现,太阳可以杀死人类50%的皮肤细胞。但是如果用花青素加以保护,则大约有85%的皮肤细胞可以幸免于死。
蓝莓,主要有效成分包括花青素、花色素、花色甙,具有良好的营养保健作用,具有抗自由基,延缓衰老,防止脑神经老化、保护视力、强心、抗癌、软化血管、增强人机体免疫等辅助功能,营养价值高。
本发明的有益效果为:
1、本发明所述的抗氧化延缓衰老、养护心脑血管的提取物的制备方法,先分别制备葡萄浸膏和混合果子粉,然后与酵母混合发酵,通过酵母的新陈代谢,将葡萄浸膏中的白藜芦醇和混合果子粉中的花青素以及多种微量元素进行有机转化,形成更易于被人体吸收的氨基酸螯合物的形式,从而增强了营养物质的吸收,提高了生物效价。
2、动物试验证明,本发明可以显著延长小鼠寿命以及大鼠负重游泳时间,降低血尿素氮含量,减少血乳酸堆积,提高肝糖原含量,表现出显著的抗疲劳活性;同时能够提高大鼠血清中超氧化物歧化酶活力,加快丙二醛的清除速率,具有显著的抗氧化活性。
3、本发明采用纯天然原料制作,不添加任何化学原料,长期食用无毒副作用。
4、本发明可以为易疲劳人群提供能量,改善其精神状况,还具有保护身体机能不被破坏,延缓衰老的作用,此外,本发明还适用于胆固醇高、长期饮酒以及吸烟人士,促进其新陈代谢、保护心脑血管免受有害物质伤害。
具体实施方式
下面通过具体实施例,对本发明作进一步说明。
实施例1
本实施例提供一种抗氧化延缓衰老、养护心脑血管的提取物的制备方法,包括如下步骤:
(1)制备葡萄浸膏
从葡萄中取葡萄皮和葡萄籽,得葡萄皮和葡萄籽混合物;向所述葡萄皮和葡萄籽混合物中加入糖苷酶和纤维素酶进行酶解,所述糖苷酶、纤维素酶、葡萄皮和葡萄籽混合物的质量之比为0.5:3:1,所述酶解反应的温度为35℃,所述酶解反应的时间为7h,所述酶解反应的pH值为7.5,得到酶解液;向所述酶解液中加入乙醇,所述乙醇与酶解液的质量之比为1:4,超声波粉碎35min后, 进行微波萃取,所述微波萃取的温度为30℃,所述微波萃取的功率为400w,所述微波萃取的时间为3min,过滤后进行减压浓缩,所述减压浓缩的温度为50℃,所述减压浓缩的真空度为-80cmHg,得到葡萄浸膏;
(2)制备混合果子粉
将黑加仑、覆盆子、蓝莓以1:3:3的质量比混合后清洗,然后打碎,得到混合果肉;向所述混合果肉中加入糖苷酶、木瓜蛋白酶、果胶酶和纤维素酶进行酶解,所述糖苷酶、木瓜蛋白酶、果胶酶、纤维素酶以及混合果肉的质量之比为0.5:0.5:0.5:3:8;所述酶解反应的温度为35℃,所述酶解反应的时间为7h,所述酶解反应的pH值为7.5,得到酶解液;将所述酶解液在温度为90℃条件下灭菌8min,进行所述灭菌2次,经20目滤膜过滤,在温度为35℃的条件下加热0.5h进行浓缩,然后喷雾干燥,所述喷雾干燥的进料流量为15mL/min,进风温度为120℃,热空气流量为30m 3/h,压缩空气流量为460L/h,出风温度为120℃,得到混合果子粉;
(3)制备抗氧化延缓衰老、养护心脑血管的提取物
酵母菌活化后,经培养,得到种子液;向所述种子液中加入步骤(1)所述葡萄浸膏和步骤(2)所述混合果子粉,所述种子液的体积、葡萄浸膏的体积和混合果子粉的质量之比为800mL:80mL:200g,经发酵培养得到发酵液,所述发酵培养的温度为32℃,所述发酵培养的搅拌转速为160rpm,所述发酵培养的时间为22h;将所述发酵液进行固液分离,得到上清液和酵母菌体;将所述上清液进行喷雾干燥,所述喷雾干燥的进口温度为65℃,所述喷雾干燥的水滴速度为40mL/min,所述喷雾干燥的出水温度为85℃,杀菌,即得所述抗氧化延缓衰老、养护心脑血管的提取物。
实施例2
本实施例提供一种抗氧化延缓衰老、养护心脑血管的提取物的制备方法,包括如下步骤:
(1)制备葡萄浸膏
从葡萄中取葡萄皮和葡萄籽,得葡萄皮和葡萄籽混合物;向所述葡萄皮和葡萄籽混合物中加入糖苷酶和纤维素酶进行酶解,所述糖苷酶、纤维素酶、葡萄皮和葡萄籽混合物的质量之比为1.5:7:3,所述酶解反应的温度为40℃,所述酶解反应的时间为9h,所述酶解反应的pH值为8,得到酶解液;向所述酶解液中加入乙醇,所述乙醇与酶解液的质量之比为1:6,超声波粉碎55min后,进行微波萃取,所述微波萃取的温度为40℃,所述微波萃取的功率为600w,所述微波萃取的时间为7min,过滤后进行减压浓缩,所述减压浓缩的温度为60℃,所述减压浓缩的真空度为-80cmHg,得到葡萄浸膏;
(2)制备混合果子粉
将黑加仑、覆盆子、蓝莓以3:1:3的质量比混合后清洗,然后打碎,得到混合果肉;向所述混合果肉中加入糖苷酶、木瓜蛋白酶、果胶酶和纤维素酶进行酶解,所述糖苷酶、木瓜蛋白酶、果胶酶、纤维素酶以及混合果肉的质量之比为1.5:1.5:1.5:7:12;所述酶解反应的温度为40℃,所述酶解反应的时间为9h,所述酶解反应的pH值为8,得到酶解液;将所述酶解液在温度为90℃条件下灭菌12min,进行所述灭菌2次,经40目滤膜过滤,在温度为45℃的条件下加热1.5h进行浓缩,然后喷雾干燥,所述喷雾干燥的进料流量为20mL/min,进风温度为160℃,热空气流量为40m 3/h,压缩空气流量为520L/h,出风温度为140℃,得到混合果子粉;
(3)制备抗氧化延缓衰老、养护心脑血管的提取物
酵母菌活化后,经培养,得到种子液;向所述种子液中加入步骤(1)所述葡萄浸膏和步骤(2)所述混合果子粉,所述种子液的体积、葡萄浸膏的体积和混合果子粉的质量之比为1200mL:120mL:400g,经发酵培养得到发酵液,所述发酵培养的温度为42℃,所述发酵培养的搅拌转速为200rpm,所述发酵培养的时间为26h;将所述发酵液进行固液分离,得到上清液和酵母菌体;将所述上清液进行喷雾干燥,所述喷雾干燥的进口温度为85℃,所述喷雾干燥的水滴速度为60mL/min,所述喷雾干燥的出水温度为90℃,杀菌,即得所述抗氧化延缓衰老、养护心脑血管的提取物。
实施例3
本实施例提供一种抗氧化延缓衰老、养护心脑血管的提取物的制备方法,包括如下步骤:
(1)制备葡萄浸膏
从葡萄中取葡萄皮和葡萄籽,得葡萄皮和葡萄籽混合物;向所述葡萄皮和葡萄籽混合物中加入糖苷酶和纤维素酶进行酶解,所述糖苷酶、纤维素酶、葡萄皮和葡萄籽混合物的质量之比为1:5:2,所述酶解反应的温度为37℃,所述酶解反应的时间为8h,所述酶解反应的pH值为7.6,得到酶解液;向所述酶解液中加入乙醇,所述乙醇与酶解液的质量之比为1:5,超声波粉碎45min后,进行微波萃取,所述微波萃取的温度为35℃,所述微波萃取的功率为500w,所述微波萃取的时间为5min,过滤后进行减压浓缩,所述减压浓缩的温度为55℃,所述减压浓缩的真空度为-80cmHg,得到葡萄浸膏;
(2)制备混合果子粉
将黑加仑、覆盆子、蓝莓以1:1:1的质量比混合后清洗,然后打碎,得到混合果肉;向所述混合果肉中加入糖苷酶、木瓜蛋白酶、果胶酶和纤维素酶进行酶解,所述糖苷酶、木瓜蛋白酶、果胶酶、纤维素酶以及混合果肉的质量之比为1:1:1:5:10;所述酶解反应的温度为37℃,所述酶解反应的时间为8h,所述酶解反应的pH值为7.7,得到酶解液;将所述酶解液在温度为90℃条件下灭菌10min,进行所述灭菌2次,经30目滤膜过滤,在温度为40℃的条件下加热1h进行浓缩,然后喷雾干燥,所述喷雾干燥的进料流量为18mL/min,进风温度为140℃,热空气流量为35m 3/h,压缩空气流量为490L/h,出风温度为130℃,得到混合果子粉;
(3)制备抗氧化延缓衰老、养护心脑血管的提取物
酵母菌活化后,经培养,得到种子液;向所述种子液中加入步骤(1)所述葡萄浸膏和步骤(2)所述混合果子粉,所述种子液的体积、葡萄浸膏的体积和混合果子粉的质量之比为1000mL:100mL:300g,经发酵培养得到发酵液,所 述发酵培养的温度为37℃,所述发酵培养的搅拌转速为180rpm,所述发酵培养的时间为24h;将所述发酵液进行固液分离,得到上清液和酵母菌体;将所述上清液进行喷雾干燥,所述喷雾干燥的进口温度为75℃,所述喷雾干燥的水滴速度为50mL/min,所述喷雾干燥的出水温度为90℃,杀菌,即得所述抗氧化延缓衰老、养护心脑血管的提取物。

与空白组比较,本发明能显著延长大鼠负重游泳时间。与空白组比较,本发明高剂量组能显著降低大鼠血清中BUN;中剂量组和高剂量组均能显著降低LD量。与空白组比较,本发明高、中、低剂量组能显著提高大鼠肝脏中肝糖元含量。大鼠负重游泳时间的长短可反映出抗疲劳能力。BUN为蛋白质的代谢物,强运动时,蛋白质的代谢加快,BUN的清除率可反映出机体抗疲劳能力。人体在强烈运动的过程中主要通过无氧氧化供能,其代谢产物LD是引发疲劳的原因之一。LD的清除有利于疲劳的恢复。糖原是运动过程中的主要能源物质,肝糖原含量越高,抗疲劳活性越强。本发明可延长大鼠负重游泳的时间,降低BUN含量,减少LD堆积,提高肝糖原含量,表现出显著的抗疲劳活性。

与空白组比较,本发明中剂量组和高剂量组能极显著提高大鼠血清中SOD的活力;三个剂量组能显著降低血清中MDA的含量,且抗氧化作用随本发明的剂量增加而增强。SOD是生物体内清除自由基的一种重要酶,能使机体有效抵御自由基的毒性;MDA是脂质过氧化最重要的产物之一,其含量高低可反映出抗氧化能力强弱。本发明能提高大鼠血清中SOD活力,加快MDA的清除速率,具有显著的抗氧化活性。

  1. 一种抗氧化延缓衰老、养护心脑血管的提取物的制备方法,其特征在于,包括如下步骤:
    (1)制备葡萄浸膏
    从葡萄中取葡萄皮和葡萄籽,得葡萄皮和葡萄籽混合物;向所述葡萄皮和葡萄籽混合物中加入第一复合酶进行酶解,得到酶解液;向所述酶解液中加入乙醇,超声波粉碎后,依次进行萃取、过滤、浓缩,得到葡萄浸膏;
    (2)制备混合果子粉
    将黑加仑、覆盆子、蓝莓清洗后打碎,得到混合果肉;向所述混合果肉中加入第二复合酶进行酶解,得到酶解液;将所述酶解液依次进行灭菌、过滤、浓缩、干燥,得到混合果子粉;
    (3)制备抗氧化延缓衰老、养护心脑血管的提取物
    酵母菌活化后,经培养,得到种子液;向所述种子液中加入步骤(1)所述葡萄浸膏和步骤(2)所述混合果子粉,经发酵培养得到发酵液;将所述发酵液进行固液分离,得到上清液和酵母菌体;将所述上清液依次进行干燥、灭菌,即得所述抗氧化延缓衰老、养护心脑血管的提取物。
  2. 根据权利要求1所述的抗氧化延缓衰老、养护心脑血管的提取物的制备方法,其特征在于,步骤(1)中,所述第一复合酶为糖苷酶和纤维素酶的混合物,所述糖苷酶、纤维素酶、葡萄皮和葡萄籽混合物的质量之比为(0.5-1.5):(3-7):(1-3);所述酶解反应的温度为35-40℃,所述酶解反应的时间为7-9h,所述酶解反应的pH值为7.5-8。
  3. 根据权利要求1所述的抗氧化延缓衰老、养护心脑血管的提取物的制备方法,其特征在于,步骤(1)中,所述乙醇与酶解液的质量之比为1:4-1:6;所述超声波粉碎的时间为35-55min;所述萃取为微波萃取,所述微波萃取的温 度为30-40℃,所述微波萃取的功率为400-600w,所述微波萃取的时间为3-7min;所述浓缩为减压浓缩,所述减压浓缩的温度为50-60℃,所述减压浓缩的真空度为-80cmHg以下。
  4. 根据权利要求1所述的制备抗氧化延缓衰老、养护心脑血管的提取物的制备方法,其特征在于,步骤(2)中,所述黑加仑、覆盆子、蓝莓的质量之比为(1-3):(1-3):(1-3);所述第二复合酶为糖苷酶、木瓜蛋白酶、果胶酶和纤维素酶的混合物,所述糖苷酶、木瓜蛋白酶、果胶酶、纤维素酶以及混合果肉的质量之比为(0.5-1.5):(0.5-1.5):(0.5-1.5):(3-7):(8-12);所述酶解反应的温度为35-40℃,所述酶解反应的时间为7-9h,所述酶解反应的pH值为7.5-8。
  5. 根据权利要求1所述的制备抗氧化延缓衰老、养护心脑血管的提取物的制备方法,其特征在于,步骤(2)中,所述灭菌的温度为90℃,所述灭菌的时间为8-12min,进行所述灭菌2次以上;所述过滤后的滤液中固形物的粒径为20-40目;所述浓缩为在温度为35-45℃的条件下加热0.5-1.5h。
  6. 根据权利要求1所述的制备抗氧化延缓衰老、养护心脑血管的提取物的制备方法,其特征在于,步骤(2)中,所述干燥为喷雾干燥,所述喷雾干燥的进料流量为15-20mL/min,进风温度为120-160℃,热空气流量为30-40m 3/h,压缩空气流量为460-520L/h,出风温度为120-140℃。
  7. 根据权利要求1所述的制备抗氧化延缓衰老、养护心脑血管的提取物的制备方法,其特征在于,步骤(3)中,所述种子液的体积、葡萄浸膏的体积和混合果子粉的质量之比为(800-1200mL):(80-120mL):(200-400g)。
  8. 根据权利要求1所述的制备抗氧化延缓衰老、养护心脑血管的提取物的制备方法,其特征在于,步骤(3)中,所述发酵培养的温度为32-42℃,所述发酵培养的搅拌转速为160-200rpm,所述发酵培养的时间为22-26h。
  9. 根据权利要求1所述的制备抗氧化延缓衰老、养护心脑血管的提取物的制备方法,其特征在于,步骤(3)中,所述干燥为喷雾干燥,所述喷雾干燥的进口温度为65-85℃,所述喷雾干燥的水滴速度为40-60mL/min,所述喷雾干燥 的出水温度为85-90℃。
  10. 权利要求1-9任一项所述方法制备得到的抗氧化延缓衰老、养护心脑血管的提取物。
