Abstract
本发明公开了一种富含茶藨子叶状层菌的保健口服液,按照重量百分比计包括如下组分:茶藨子叶状层菌发酵菌丝体50‑90%、香菇多糖0‑40%、葡萄糖0‑3%、食品级磷酸二氢钾0‑0.1%、麦芽汁0‑5%、其余为水。本发明的茶藨子叶状层菌发酵菌丝体中富含甾醇,具体在菌种活化以及发酵过程中均通过添加金银花根叶提取液的方式以促进菌丝在生产过程中甾醇成分的合成和集聚,同时在培养基中加入的硝酸铵、苯甲酸钠、L‑苯氨基酸以及乙酸钠等成分可以进一步有效地促进菌体的生长以及麦角甾醇的生物合成。由此配置成的口服液中富含甾醇,不仅口感好,且具有清热、消炎、抗癌、提高免疫力、调节胃肠道等功效。

技术领域
本发明涉及菌种培养领域,具体涉及一种提高茶藨子叶状层菌发酵菌丝体中甾醇含量的方法,以及用茶藨子叶状层菌发酵菌丝体配制的保健口服液。
背景技术
茶藨子叶状层菌发酵菌丝体--来源于茶藨子叶状层菌﹝Phylloporia ribis(Schumach:Fr.)Ryvarden﹞,也称金银花菌。是一种新资源食品,是茶藨子叶状层菌(从金银花植株上分离)经接种培养、发酵、干燥、粉碎等步骤制得。
茶藨子叶状层菌中含有的甾醇具有很高的药用价值,尤其是麦角甾醇具有降胆固醇、消炎、抗溃疡、抗肿瘤等作用。但是茶藨子叶状层菌种的甾醇含量较低(一般在20-30ug/l),如果能够提高茶藨子叶状层菌中的甾醇含量将有利于甾醇的提取和药用范围。因此,本发明提高一种能够提高茶藨子叶状层菌中甾醇(尤其是麦角甾醇)含量的方法。
2013-02-05,卫生部发布2013年第1号公告中称,根据《中华人民共和国食品安全法》和《新资源食品管理办法》有关规定,现批准茶藨子叶状层菌发酵菌丝体为新资源食品。但现有茶藨子叶状层菌食品大多的功能是防治咽炎(如中国专利CN201810571684.9),且食品中茶藨子叶状层菌的含量较少,成分多而复杂,食品的药用性能比较单一,保健功效一般。
发明内容
为解决上述技术问题,本发明提供了一种提高茶藨子叶状层菌发酵菌丝体中甾醇含量的方法;同时还提高一种将富含甾醇的茶藨子叶状层菌发酵菌丝体作为食品与其他辅助成分配制成具有保健功能的口服液,达到药食同源的目的。
本发明采用的技术方案是:
一种提高茶藨子叶状层菌发酵菌丝体中甾醇含量的方法,包括如下步骤:首先将从金银花植株上分离出茶藨子叶状层菌种,然后在固体培养基中培养,将培养较好的茶藨子叶状层菌菌丝接种到液体培养基中在25℃下进行培养8-10天,在液体培养基中培养2天后对液体培养基进行震摇培养,震摇培养过程中往液体培养基中补充金银花根叶提取液,金银花根叶提取液的加入量与液体培养基的体积比为0.0001-0.0005:1。
所述金银花根叶提取液的制备方法为:首先将金银花根茎和叶进行干燥和灭菌处理,然后将根茎与叶按照重量比为2-3:1的比例混合,经粉碎后加入浓度为75-80%的乙醇溶液中浸泡2-3h,然后在50-70℃下加热回流提取2-5次,每次得到的回流液合并后即得到提取液。
所述每升乙醇溶液中的金银花根叶加入量为0.3-0.5kg。
所述固体培养基的成分按照重量份计为:去皮马铃薯煮液200份、葡萄糖20份、琼脂20份、硝酸铵0.5份、纯净水1000份,在固体培养基培养过程中,每天向菌丝上喷洒金银花提取液一次。
所述液体培养基的成分按照重量份计为:去皮马铃薯煮液200份、葡萄糖20份、苯甲酸钠0.3份、L-苯氨基酸0.4份、乙酸钠0.1份、纯净水1000份,在液体培养基培养过程中,在液体培养的第二天一次性加入金银花根叶提取液。
所述固体培养基为斜面培养基。
利用上述方法制得的茶藨子叶状层菌发酵菌丝体作为原料、辅助添加香菇多糖、葡萄糖、食品级磷酸二氢钾、麦芽汁和水配置成具有保健、清热、消炎、抗癌和提高免疫力的口服液。按照重量百分比计包括如下组分:茶藨子叶状层菌发酵菌丝体50-90%、香菇多糖0-40%、葡萄糖0-3%、食品级磷酸二氢钾0-0.1%、麦芽汁0-5%、其余为水。
上述组分中还包括嗜酸乳杆菌和/或酪酸梭菌,所述嗜酸乳杆菌和/或酪酸梭菌的含量为0-2%。
本发明的有益效果是:本发明在菌种活化以及发酵过程中均通过添加金银花根叶提取液的方式以促进菌丝在生产过程中甾醇成分的合成和集聚,同时在培养基中加入的硝酸铵、苯甲酸钠、L-苯氨基酸以及乙酸钠等成分可以进一步有效地促进菌体的生长以及麦角甾醇的生物合成,制得的茶藨子叶状层菌发酵菌丝体中的甾醇含量是普通茶藨子叶状层菌发酵菌丝体的10倍以上。利用该种富含甾醇的茶藨子叶状层菌发酵菌丝体配制成保健口服液,不仅符合新资源食品的要求,而且还具有清热、消炎、抗癌、提高免疫力、调节胃肠道等功效。

菌种选择:取金银花的根部,在无菌环境下获得纯化的茶藨子叶状层菌-子实体组织两块,经过消毒后,将两子实体组织分别在无菌环境下织接种到带有斜面固体培养基的试管Ⅰ、试管Ⅱ和试管Ⅲ中,试管Ⅰ、试管Ⅱ和试管Ⅲ均置于相同的环境下(25℃,紫外线光照),待斜面培养基上活化出菌株。事先准备好10个编有标号(1、2、3……)的装液量50ml/300ml三角瓶,10个三角瓶中均装有液体培养液,其中编号1-2中分别接种试管Ⅰ中的菌丝,接种量均为1个菌丝团;编号3-6中分别接种试管Ⅱ中的菌丝,接种量均为1个菌丝团;编号7-10中分别接种试管Ⅲ中的菌丝,接种量均为1个菌丝团。接种后,10个三角瓶均置于相同的环境下培养8天。
试管Ⅰ的固体培养基的成分为:去皮马铃薯煮液200g、葡萄糖20g、琼脂20g、纯净水1000份;试管Ⅱ和试管Ⅲ的固体培养基的成分均为:去皮马铃薯煮液200g、葡萄糖20g、琼脂20g、硝酸铵0.5g、纯净水1000份。
编号1的三角瓶中的液体培养基成分为:去皮马铃薯煮液200g、葡萄糖20g、苯甲酸钠0.3g、纯净水1000份;编号2的三角瓶中的液体培养基成分为:去皮马铃薯煮液200g、葡萄糖20g、L-苯氨基酸0.4g、纯净水1000份;编号3的三角瓶中的液体培养基成分为:去皮马铃薯煮液200g、葡萄糖20g、乙酸钠0.1g、纯净水1000份;编号4-10的三角瓶中的液体培养基成分均为:去皮马铃薯煮液200g、葡萄糖20g、苯甲酸钠0.3g、L-苯氨基酸0.4g、乙酸钠0.1g、纯净水1000g;
其中,在固体培养基培养过程中,每天向试管Ⅱ和试管Ⅲ的菌丝上喷洒等量的金银花提取液一次。在液体培养基培养过程中,在液体培养的第二天分别向编号6-10的三角瓶内一次性加入等量的金银花根叶提取液,金银花根叶提取液的加入量与液体培养基的体积比为0.0003:1。
金银花根叶提取液的制备方法为:首先将金银花根茎和叶进行干燥和灭菌处理,然后将根茎与叶按照重量比为2-3:1的比例混合,经粉碎后加入浓度为75-80%的乙醇溶液中浸泡2-3h,然后在50-70℃下加热回流提取2-5次,每次得到的回流液合并后即得到提取液。
编号1-3的三角瓶的培养方式为静止培养,编号4-6的三角瓶的培养方式为180r/min震摇培养,编号7-10的三角瓶的培养方式为先静止培养2天,然后以180r/min的震摇速度震摇培养。
待培养后,将培养好的菌丝体以5000r/min,离心20min,收集各三角瓶中的菌丝体。然后利用石油醚浸提、层析的分离提取方法分别对10个三角瓶中的菌丝体中的甾醇进行提取测定,所得甾醇含量如下表:
从上表对比可以得出,本发明在菌种活化以及发酵过程中均通过添加金银花根叶提取液的方式以促进菌丝在生产过程中甾醇成分的合成和集聚,同时在培养基中加入的硝酸铵、苯甲酸钠、L-苯氨基酸以及乙酸钠等成分可以进一步有效地促进菌体的生长以及麦角甾醇的生物合成。
利用上述方法所得的茶藨子叶状层菌发酵菌丝体,将茶藨子叶状层菌发酵菌丝体粉碎研磨成微细粉,按照如下重量配比:茶藨子叶状层菌发酵菌丝体50%、香菇多糖30%、葡萄糖3%、食品级磷酸二氢钾0.1%、麦芽汁5%、嗜酸乳杆菌2%、其余为水,配制成口服液,经过密封包装即得产品。由此配置成的口服液中富含甾醇,不仅口感好,且具有清热、消炎、抗癌、提高免疫力、调节胃肠道等功效。
Claims (8)
1.一种富含茶藨子叶状层菌的保健口服液,其特征在于:按照重量百分比计包括如下组分:
茶藨子叶状层菌发酵菌丝体 50-90%
香菇多糖 0-40%
葡萄糖 0-3%
食品级磷酸二氢钾 0-0.1%
麦芽汁 0-5%
其余为水。
2.根据权利要求1所述的一种富含茶藨子叶状层菌的保健口服液,其特征在于:组分中还包括嗜酸乳杆菌和/或酪酸梭菌,所述嗜酸乳杆菌和/或酪酸梭菌的含量为0-2%。
3.根据权利要求1或2所述的一种富含茶藨子叶状层菌的保健口服液,其特征在于:所述茶藨子叶状层菌发酵菌丝体的培养方法包括如下步骤:首先将从金银花植株上分离出茶藨子叶状层菌种,然后在固体培养基中培养,将培养较好的茶藨子叶状层菌菌丝接种到液体培养基中在25℃下进行培养8-10天,其特征在于:在液体培养基中培养2天后对液体培养基进行震摇培养,震摇培养过程中往液体培养基中补充金银花根叶提取液,金银花根叶提取液的加入量与液体培养基的体积比为0.0001-0.0005:1。
4.根据权利要求3所述的一种富含茶藨子叶状层菌的保健口服液,其特征在于:所述金银花根叶提取液的制备方法为:首先将金银花根茎和叶进行干燥和灭菌处理,然后将根茎与叶按照重量比为2-3:1的比例混合,经粉碎后加入浓度为75-80%的乙醇溶液中浸泡2-3h,然后在50-70℃下加热回流提取2-5次,每次得到的回流液合并后即得到提取液。
5.根据权利要求4所述的一种富含茶藨子叶状层菌的保健口服液,其特征在于:所述每升乙醇溶液中的金银花根叶加入量为0.3-0.5kg。
6.根据权利要求3所述的一种富含茶藨子叶状层菌的保健口服液,其特征在于:所述固体培养基的成分按照重量份计为:去皮马铃薯煮液200份、葡萄糖20份、琼脂20份、硝酸铵0.5份、纯净水1000份,在固体培养基培养过程中,每天向菌丝上喷洒金银花提取液一次。
7.根据权利要求3所述的一种富含茶藨子叶状层菌的保健口服液,其特征在于:所述液体培养基的成分按照重量份计为:去皮马铃薯煮液200份、葡萄糖20份、苯甲酸钠0.3份、L-苯氨基酸0.4份、乙酸钠0.1份、纯净水1000份,在液体培养基培养过程中,在液体培养的第二天一次性加入金银花根叶提取液。
8.根据权利要求3所述的一种富含茶藨子叶状层菌的保健口服液,其特征在于:所述固体培养基为斜面培养基。
