Abstract
本发明涉及保健食品技术领域,具体为一种蛋白核小球藻多肽缓解增肥组合物,以及该种组合物的制备方法。本发明的组合物以蛋白核小球藻多肽为主要功效成分之一,并与大豆多肽、玉米多肽、香菇多糖、猴头菇多糖、小麦低聚多肽粉、绿茶多酚提取物及枸杞浓缩汁配伍,可充分发挥各组分的功效,各组分相互协同增益,从而使本发明的组合物能促进脂肪代谢,减少皮下脂肪的贮存,具有显著的缓解增肥的效果,增强免疫力。本发明的组合物由一定配比的所述组分组成,不仅营养丰富均衡,且可口美味、健康安全,适宜任何年龄的人群食用,此外,本发明组合物还具有健脾和健胃的功效。

技术领域
本发明涉及保健食品技术领域,尤其涉及一种蛋白核小球藻多肽缓解增肥组合物,以及该种组合物的制备方法。
背景技术
蛋白核小球藻粉主要营养成分的分析表明,其蛋白质含量为63.60%,18种氨基酸总量为55.95%,其中的8种必需氨基酸含量为23.35%,氨基酸评分(AAS)为64.3,其营养成分全面,是优良的单细胞蛋白源。蛋白核小球藻多肽是蛋白核小球蛋白的水解产物,由多种多肽分子和游离氨基酸组成,所含人体必需的氨基酸的比例平衡,含量丰富。此外,蛋白核小球藻多肽不仅具有良好的营养特性,还是极易消化吸收的多肽化合物,能促进脂肪代谢,控制体内脂肪贮存,具有良好缓解增肥作用。相较于蛋白质而言,小分子肽优越性主要体现在:更易被人体吸收利用;活性高,在较小浓度下即可发挥其特有的生理作用;分子量小,易于修饰和改造,能够通过人工化学合成等。而相较于单一的氨基酸而言,小分子肽除了具有特殊的生理活性外,在吸收通道和吸收速度上也具有氨基酸无可比拟的优越性。已有研究证实,人体小肠存在专门的低聚肽吸收通道,人体摄入的蛋白质经过多种消化酶的水解,主要以低肽的形式被吸收。许多研究表明,各种来源的生物活性肽具有抗氧化、抗肿瘤、抑菌、降压、降血糖等多种作用,成为生物医药和保健品开发的热点。目前,具有缓解增肥及降脂作用的多肽包括大豆肽、玉米活性肽、蚕蛹肽等。蛋白核小球藻多肽还具有降血糖、降血压和抑制胆固醇的作用,市场上的缓解增肥食品中尚未发现含有蛋白核小球藻多肽的产品,很值得我们去开发这一功能性保健缓解增肥食品。
另外,现今食品的流行趋势为天然、安全、方便、口味多样化和国际化。蛋白粉富含优质蛋白,除了常见的提高免疫力功能外,在维持饮食均衡等的健康状态下,摄入高蛋白还可以减脂。但目前还没有食用和携带方便的蛋白肽冲剂,尤其是具有缓解增肥降脂功能的冲剂。
发明内容
本发明针对现有技术尚未有以蛋白核小球藻多肽为主要功效成分之一的缓解增肥保健食品的问题,提供一种主要功效成分之一为蛋白核小球藻多肽,且与其它多种功效成分配伍从而具有显著缓解增肥效果的蛋白核小球藻多肽缓解增肥组合物,以及该种组合物的制备方法。
为实现上述目的,本发明采用以下技术方案。
一种蛋白核小球藻多肽缓解增肥组合物,由包括以下重量份的各组分制成:蛋白核小球藻多肽粉20-40份,大豆多肽15-20份,玉米多肽0-5份,香菇多糖0-5份,猴头菇多糖0-5份,小麦低聚多肽粉0-5份,绿茶多酚提取物0-5份,枸杞浓缩汁0-5份。
优选的,所述的蛋白核小球藻多肽缓解增肥组合物,由以下重量份的各组分制成:蛋白核小球藻多肽粉30份,大豆多肽17份,玉米多肽3份,香菇多糖3份,猴头菇多糖3份,小麦低聚多肽粉3份,绿茶多酚提取物3份,枸杞浓缩汁4份。
以上所述的蛋白核小球藻多肽缓解增肥组合物的制备方法,用于制备所述蛋白核小球藻多肽缓解增肥组合物的组分中包括枸杞浓缩汁,所述制备方法包括以下步骤:将除枸杞浓缩汁外组成蛋白核小球藻多肽缓解增肥组合物的其它各组分混合均匀,然后将枸杞浓缩汁加入混合物中并搅拌均匀,接着将混合物置于50-60℃下烘15-20min,制得蛋白核小球藻多肽缓解增肥组合物。
优选的,以上所述的蛋白核小球藻多肽缓解增肥组合物的制备方法,将混合物置于50-60℃下烘15-20min后,混合物过筛并整粒,制得颗粒状的蛋白核小球藻多肽缓解增肥组合物。
优选的,所述蛋白核小球藻多肽粉的制备方法:以蛋白核小球藻为原料,依次经破碎、酶解、超滤、真空浓缩、喷雾干燥而得。
优选的,所述大豆多肽的制备方法:以大豆为原料,依次经破碎、酶解、超滤、真空浓缩、喷雾干燥而得。

与现有技术相比,本发明的有益效果是:
蛋白核小球藻多肽是一种高品质蛋白质原料,含有生长因子和小分子肽等多种功能性蛋白质,具有适口性好、蛋白质含量高、氨基酸平衡、吸收利用率高、附加值高等特点,这种蛋白肽冲剂对肥胖、免疫调节作用显著,可广泛应用于食品、保健食品和医药领域。
玉米多肽营养丰富,含有人体必须的各种氨基酸,提供氨基酸及肽能营养,玉米肽还具有抑制血管紧张素转换酶的作用,从而具有降血压等其它作用;除了含降血压小肽外,其它的小肽具有抗疲劳、提高机体免疫力等功能,具有适口性好、蛋白质含量高、氨基酸平衡、吸收利用率高、附加值高等特点,这种蛋白肽已经广泛应用于食品、保健食品和医药领域。
香菇多糖是从香菇子实体中提取的有效活性成分,香菇多糖中的活性成分是具有分支的β-(1-3)-D-葡聚糖,主链由β-(1-3)-连接的葡萄糖基组成,沿主链随机分布着由β-(1-6)连接的葡萄糖基,呈梳状结构。香菇多糖可提高病人的免疫功能,纠正微量元素失调。香菇多糖具有抗病毒、抗肿瘤、调节免疫功能和刺激干扰素形成等作用,已经广泛应用于食品、保健食品和医药领域。
枸杞浓缩汁是用枸杞鲜果为原料经过物理方法如压榨、离心等得到的汁液,枸杞汁的主要营养成分为枸杞多糖。枸杞多糖是从枸杞中提取而得的一种水溶性多糖,已明确该多糖系蛋白多糖,分子量为22-25kD,由阿拉伯糖、葡萄糖、半乳糖、甘露糖、木糖、鼠李糖6种单糖成分组成,呈淡黄色粉末状,易吸湿。经研究表明,枸杞多糖是枸杞调节免疫、延缓衰老的主要活性成分,其可改善老年人易疲劳、食欲不振和视力模糊等症状,对恶性肿瘤、艾滋病的预防与治疗也能起到积极作用。同时枸杞多糖在改善糖尿病上同样具有明显的效果。现代研究证实,枸杞多糖对血清胰岛素水平有提高的趋势,并有修复受损胰岛细胞和促进细胞再生的功能,使糖耐量增强。因此,从以上角度来看,枸杞多糖不但不会引起血糖升高,还能改善自身正常糖代谢功能,平稳血糖。
猴头菇多糖是从猴头菇子实体中提取的有效活性成分,猴头菇的主要成分是多糖类和猴头菌素,经热水提取的猴头菇多糖对小白鼠肉瘤180和艾氏腹水癌有明显的抑制作用,同时具有助消化、利五脏的功能,对慢性胃炎、十二指肠溃疡等多种消化道疾病均有较好的疗效,能提高人体免疫能力。猴头菌素是猴头菇另一类活性成份。它可促使神经生长因子(NGF:可延长N轴突,维持N细胞生存,调节神经细胞的产生并对衰老动物神经细胞的再生发挥作用)的合成,其活性大大强于肾上腺素,其主要成分为猴头菇菌素
A.B.C.E.F,化学成份为萜类物质。猴头菌素可用于治疗智力衰退,神经衰弱及植物性神经衰退,尤其对ALZheimev型和痴呆症有良好效果。可做各类保健品、功能性食品的添加原料。
小麦低聚多肽粉以小麦为原料,经调浆、蛋白酶酶解、分离、过滤、喷雾干燥等工艺制成。2012年9月5日,卫生部发布《关于批准中长链脂肪酸食用油和小麦低聚肽作为新资源食品等的公告》批准小麦低聚肽作为新资源食品。小麦低聚多肽有抑制胆固醇上升的作用。小麦低聚肽能促进胰岛素分泌,其功能物质是低聚蛋氨酸,可用于调节人的血糖,改善糖尿病症状。小麦肽能阻碍血管紧张素酶的作用,因而有降血压作用。小麦低聚肽的特点之一是含高谷酰胺,能够有效调节神经,也可作肠功能障碍时的特殊营养物质。小麦低聚肽具有ACE抑制作用、免疫调节、抗氧化等多种生物活性,能够刺激机体淋巴细胞增殖,增强巨噬细胞吞噬功能,提高机体抵御外界病原体感染的能力,降低机体发病率等,小麦低聚肽是小麦蛋白酶解产物,能抑制血管紧张素转化酶的活性,使血管紧张素原不能转变成能使血压升高的血管紧张素Ⅱ,从而生理地降低血压,而对正常血压不起用。

蛋白核小球藻多肽缓解增肥组合物的制备方法,包括以下步骤S1-S3:
S1备料:
蛋白核小球藻多肽粉的制备:以蛋白核小球藻为原料,采用蛋白核小球藻多肽粉的现有制备技术,依次经破碎、酶解、超滤、真空浓缩、喷雾干燥而得;备用。
大豆多肽的制备:以大豆为原料,采用大豆多肽的现有制备技术,依次经破碎、酶解、超滤、真空浓缩、喷雾干燥而得;备用。
玉米多肽的制备:以玉米为原料,采用玉米多肽的现有制备技术,依次经破碎、酶解、超滤、真空浓缩、喷雾干燥而得;备用。
小麦低聚多肽粉的制备:以小麦为原料,采用小麦低聚多肽粉的现有制备技术,依次经破碎、酶解、超滤、真空浓缩、喷雾干燥而得;备用。
香菇多糖的制备:将香菇破碎,加入香菇5倍重量的纯净水并于70-90℃的温度下提取2h,共提取两次,合并两次的提取液并过滤,提取液经真空浓缩和喷雾干燥后,得到香菇多糖;备用。

枸杞浓缩汁的制备:以枸杞鲜果为原料,经压榨和离心过滤,得到枸杞浓缩汁;备用。
S2混料:按重量配比分别称取蛋白核小球藻多肽粉、大豆多肽、玉米多肽、香菇多糖、猴头菇多糖、小麦低聚多肽粉和绿茶多酚提取物,并将这些组分混合物均匀。然后按重量配比称取枸杞浓缩汁,将枸杞浓缩汁加至混合物中并搅拌均匀。
S3干燥:将步骤S2的混合物置于50-60℃下烘15-20min至干燥,制得蛋白核小球藻多肽缓解增肥组合物。
S4制粒:经步骤S3干燥后的混合物过筛,并采用现有的整粒技术将其制备成颗粒状的蛋白核小球藻多肽缓解增肥组合物。
缓解增肥效果的测定
实验测试方法:动物实验参照《保健食品功能学评价程序与检验方法规范》,小鼠于动物实验房适应性喂养1周(23±2℃,12/12h光暗循环)后,将小鼠分为基础饲料喂养组(ND-fed)和高脂饲料喂养组(HFD-fed),分别饲养40天建立肥胖小鼠模型(基础饲料;60%的高脂饲料)。经40天不同饲料喂养后,将诱导成功的肥胖小鼠(肥胖小鼠定义为比基础饲料喂养组的小鼠平均体重增加20%的小鼠)随机分组为3组:模型对照组、配方低剂量组、配方高剂量组。各组小鼠喂饲60%的高脂饲料,同时,配方低剂量组(200mg/只/天)、配方高剂量组(400mg/只/天)分别取不同剂量的实施例制备的组合物以蒸馏水溶解后进行自由进食,模型对照组以相同体积蒸馏水替代,每周称重1次,连续饲养77天,试验小鼠在末次给药后,停食停水1天,称体质量。
实施例1-6制备的蛋白核小球藻多肽缓解增肥组合物均具有缓解体重增长的效果,对于各实施例制备的组合物进行实验测试,配方低剂量组和配方高剂量组的平均体重均明显低于模型对照组的评价体重。其中,实施例3制备的蛋白核小球藻多肽缓解增肥组合物的缓解增肥效果如图1所示,用60%的高脂饲料喂养的小鼠(配方低剂量组和配方高剂量组),同时进食实施例3制备的组合物,可有效减缓体重的增长,且进食组合物的剂量越大,缓解体重增长的效果越好,配方高剂量组的平均体重低于配方低剂量组的平均体重。用60%的高脂饲料喂养但未进食实施例3制备的组合物的小鼠,平均体重明显高于配方低剂量组和配方高剂量组的平均体重。

蛋白核小球藻多肽粉20-40份,大豆多肽15-20份,玉米多肽0-5份,香菇多糖0-5份,猴头菇多糖0-5份,小麦低聚多肽粉0-5份,绿茶多酚提取物0-5份,枸杞浓缩汁0-5份。
2.根据权利要求1所述的蛋白核小球藻多肽缓解增肥组合物,其特征在于,由以下重量份的各组分制成:
蛋白核小球藻多肽粉30份,大豆多肽17份,玉米多肽3份,香菇多糖3份,猴头菇多糖3份,小麦低聚多肽粉3份,绿茶多酚提取物3份,枸杞浓缩汁4份。
3.一种如权利要求1所述的蛋白核小球藻多肽缓解增肥组合物的制备方法,其特征在于,用于制备所述蛋白核小球藻多肽缓解增肥组合物的组分中包括枸杞浓缩汁,所述制备方法包括以下步骤:将除枸杞浓缩汁外组成蛋白核小球藻多肽缓解增肥组合物的其它各组分混合均匀,然后将枸杞浓缩汁加入混合物中并搅拌均匀,接着将混合物置于50-60℃下烘15-20min,制得蛋白核小球藻多肽缓解增肥组合物。
4.根据权利要求3所述的蛋白核小球藻多肽缓解增肥组合物的制备方法,其特征在于,将混合物置于50-60℃下烘15-20min后,混合物过筛并整粒,制得颗粒状的蛋白核小球藻多肽缓解增肥组合物。
5.根据权利要求3所述的蛋白核小球藻多肽缓解增肥组合物的制备方法,其特征在于,所述蛋白核小球藻多肽粉的制备方法:以蛋白核小球藻为原料,依次经破碎、酶解、超滤、真空浓缩、喷雾干燥而得。

8.根据权利要求3所述的蛋白核小球藻多肽缓解增肥组合物的制备方法,其特征在于,所述香菇多糖的制备方法:将香菇破碎,加入香菇5倍重量的纯净水并于70-90℃的温度下提取2h,共提取两次,合并两次的提取液并过滤,提取液经真空浓缩和喷雾干燥后,得到香菇多糖。
9.根据权利要求3所述的蛋白核小球藻多肽缓解增肥组合物的制备方法,其特征在于,所述猴头菇多糖的制备方法:将猴头菇破碎,加入猴头菇20倍重量的纯净水并于70-90℃的温度下提取2h,共提取两次,合并两次的提取液并过滤,提取液经真空浓缩和喷雾干燥后,得到猴头菇多糖。
10.根据权利要求3所述的蛋白核小球藻多肽缓解增肥组合物的制备方法,其特征在于,所述绿茶多酚提取物的制备方法:将绿茶破碎,加入绿茶20倍重量的纯净水并于70-90℃的温度下提取2h,共提取两次,合并两次的提取液并过滤,提取液经真空浓缩和喷雾干燥后,得到绿茶多酚提取物。
