背景技术
滨海白首乌又名耳叶牛皮消,为萝藦科鹅绒藤属植物,是滨海的特色植物资源。滨海白首乌味甘、苦,性微温,含有蛋白质、淀粉、磷脂、游离氨基酸等多种营养成分,并含有C21甾苷,多糖,苯酮等生物活性物质。药理研究证实,白首乌具有抗氧化清除自由基、抗肿瘤、免疫调节、降血脂、护肝、抗炎、抗疲劳以及延缓衰老的功效。
葛根是豆科植物野葛的干燥根,全国均产,具有解肌退热、透疹、生津止渴、升阳止泻等功效,临床常用于治疗外感发热头痛、项背强痛、麻疹不透等。现代研究表明,葛根作为药食两用中药,其含异黄酮、三萜、香豆素、生物碱等多种活性成分,具有抗氧化、保肝、调节血压、血脂、血糖、免疫等药理作用。
目前,市场上白首乌和葛根主要以原料和营养粉为主,深加工较少。而且目前深加工方法尚未成熟,加工过程中营养成分不能完全析出,同时还存在营养成分容易流失的缺陷。本发明中以白首乌葛根为原料,利用生物酶解、生物转化技术,促进了以结合态形式存在的功能活性物质的释放,二者相辅相成,提高其抗氧化、保肝护肝、降血脂和免疫的功能作用,更有助于人体健康。而且本发明利用湿法制粒技术将其制成含片,可实现对白首乌葛根原料的综合利用,提高其营养价值和经济价值,使用方便,便于储存,具有广阔的市场发展潜力。
经查文献及专利,尚未发现通过生物酶解原材料,利用湿法制粒技术制备白首乌葛根口服含片的报道。
发明内容
本发明是通过以下技术方案实现的:
步骤1,原料制粉:
(1)白首乌、葛根预处理:挑选新鲜白首乌、葛根经清洗、去皮后,切成8-10g的碎粒;
(2)打浆:将切成碎粒的白首乌(50-70重量份)和葛根(30-50重量份)混合,进行磨浆处理,磨浆时加水料为原料4-8倍,100-120目筛网过滤,所得滤渣二次磨浆,将两次所得浆液混合;
(3)酶处理:取果胶酶UTC-PE40(60-80重量份,湖南尤特尔)、纤维素酶(10-20重量份)、半纤维素酶(10-20重量份)混合,按混合浆0.1-0.5%的重量比加入到混合浆中,温度控制为45-55℃,时间为1-2h;
(4)均质:将浆液均质处理,温度为60-70℃,压力为30-40Mpa;
(5)喷雾干燥:将均质好的混合液进行喷雾干燥,得到白首乌葛根粉。
步骤2,以白首乌葛根粉为原料,加入稀释剂的预混料、部分崩解剂的预混料,以聚乙烯吡咯烷酮K30为粘合剂制粒。
(1)将稀释剂可溶性淀粉、异麦芽糖醇,崩解剂交联聚乙烯吡咯烷酮、低取代羟丙纤维素,矫味剂蛋白糖,润滑剂聚乙二醇4000,粘合剂聚乙烯吡咯烷酮K30粉碎,过80-100目筛,备用。
(2)稀释剂的预混料可为可溶性淀粉与异麦芽糖醇粉末按质量比为1∶(2~3)的混合物;崩解剂的预混料为交联聚乙烯吡咯烷酮、低取代羟丙纤维素粉末按照质量比为1∶(1~2)的混合物;粘合剂为溶有蛋白糖粉末的10%聚乙烯吡咯烷酮K30的乙醇溶液。
(3)取白首乌葛根粉末,加入稀释剂的预混料、崩解剂的预混料,于容器中混匀,加入溶有蛋白糖粉末的10%聚乙烯吡咯烷酮K30的乙醇溶液,制软材,软材用24目筛制粒,湿颗粒于50~60℃条件下烘干干燥,每隔0.5小时翻动1次,以加快干燥速度,干燥至颗粒水分含量5%以下,制备白首乌葛根含片的干颗粒(以1000片为基数,制备颗粒时稀释剂预混料的用量为165~172g;崩解剂用量为25~30g)。
步骤3,将白首乌葛根含片的干颗粒与部分崩解剂预混料及润滑剂预混料混合均匀,压片,制备白首乌葛根含片。混合过程如下:将干燥好的颗粒,用24目筛整粒备用;将聚乙二醇4000粉末与微粉硅胶粉末按质量比2∶(0.8~1.5)混合均匀,制备润滑剂的预混料,备用;再将崩解剂预混料20g及润滑剂预混料6g采用等量递加混合法混合均匀;最后再将上述粉末与整粒后的颗粒采用等量递加法混合均匀,压片,即得白首乌葛根含片。
步骤4,成品检验入库:将包装后的成品进行抽检,各项指标符合质量标准后,方可入库。
本发明的有益效果主要体现在:

2.利用湿法制粒技术,创造性地在制备白首乌葛根口服含片时选用可溶性淀粉与异麦芽糖醇的预混料为稀释剂,在粘合剂聚乙烯吡咯烷酮K30的乙醇溶液溶入了蛋白糖粉末,获得了表面光滑、色泽均一、入口细腻的白首乌葛根口服含片,
3.将白首乌粉和葛根粉制作成口服含片,可以随身携带随时口服含化,食用比口服液、胶囊方便而且易消化吸收,符合消费者对营养与健康的双重需求,具有广阔的市场发展潜力。
具体实施方式
实施例1
1.原料制粉:
(1)白首乌、葛根预处理:挑选新鲜白首乌、葛根经清洗、去皮后,切成8-10g的碎粒;
(2)打浆:将切成碎粒的白首乌(60重量份)和葛根(40重量份)混合,进行磨浆处理,磨浆时加水料为原料4倍,100目筛网过滤,所得滤渣二次磨浆,将两次所得浆液混合;
(3)酶处理:按混合浆的重量比,取果胶酶0.3%、纤维素酶0.1%、半纤维素酶0.1%混合,加入到浆液中,温度控制为45℃,时间为2h;
(4)均质:将浆液均质处理,温度为60℃,压力为40Mpa
(5)喷雾干燥:将均质好的混合液进行喷雾干燥,得到白首乌粉和葛根粉。
2.以白首乌葛根粉为原料,加入稀释剂的预混料、部分崩解剂的预混料,以聚乙烯吡咯烷酮K30为粘合剂制粒。
(1)将稀释剂可溶性淀粉、异麦芽糖醇,崩解剂交联聚乙烯吡咯烷酮、低取代羟丙纤维素,矫味剂蛋白糖,润滑剂聚乙二醇4000,粘合剂聚乙烯吡咯烷酮K30粉碎,过100目筛,备用。
(2)稀释剂的预混料为可溶性淀粉与异麦芽糖醇粉末按质量比为1∶3的混合物;崩解剂的预混料为交联聚乙烯吡咯烷酮、低取代羟丙纤维素粉末按照质量比分别为1∶1的混合物;粘合剂为溶有蛋白糖粉末的10%聚乙烯吡咯烷酮K30的乙醇溶液。
(3)取白首乌葛根粉末,加入稀释剂的预混料、崩解剂的预混料,于容器中混匀,加入溶有蛋白糖粉末的10%聚乙烯吡咯烷酮K30的乙醇溶液,制软材,软材用24目筛制粒,湿颗粒于50℃条件下烘干干燥,每隔0.5小时翻动1次,以加快干燥速度,干燥至颗粒水分含量3%,制备白首乌葛根含片的干颗粒(以1000片为基数,制备颗粒时稀释剂预混料的用量为170g;崩解剂用量为30g)。
3.将白首乌葛根含片的干颗粒与部分崩解剂预混料及润滑剂预混料混合均匀,压片,制备白首乌葛根含片。混合过程如下:将干燥好的颗粒,用24目筛整粒备用;将聚乙二醇4000粉末与微粉硅胶粉末按质量比2∶1混合均匀,制备润滑剂的预混料,备用;再将崩解剂预混料20g及润滑剂预混料6g采用等量递加混合法混合均匀;最后再将上述粉末与整粒后的颗粒采用等量递加法混合均匀,压片,即得白首乌葛根含片。
4.成品检验入库:将包装后的成品进行抽检,各项指标符合质量标准后,方可入库。
实施例2
1.原料制粉:
(1)白首乌、葛根预处理:挑选新鲜白首乌、葛根经清洗、去皮后,切成8-10 g的碎粒;
(2)打浆:将切成碎粒的白首乌(70重量份)和葛根(30重量份)混合,进行磨浆处理,磨浆时加水料为原料8倍,120目筛网过滤,所得滤渣二次磨浆,将两次所得浆液混合;
(3)酶处理:取果胶酶0.5%、纤维素酶0.2%、半纤维素酶0.2%混合,加入到浆液中,温度控制为55℃,时间为1h;
(4)均质:将浆液均质处理,温度为70℃,压力为30Mpa;
(5)喷雾干燥:将均质好的混合液进行喷雾干燥,得到白首乌粉和葛根粉。
2.以白首乌葛根粉为原料,加入稀释剂的预混料、部分崩解剂的预混料,以聚乙烯吡咯烷酮K30为粘合剂制粒。
(1)将稀释剂木糖醇、蔗糖,崩解剂交联聚乙烯吡咯烷酮、低取代羟丙纤维素,矫味剂蛋白糖,润滑剂聚乙二醇4000,粘合剂聚乙烯吡咯烷酮K30粉碎,过120目筛,备用。
(2)稀释剂的预混料为可溶性淀粉与异麦芽糖醇粉末按质量比为1∶2的混合物;崩解剂的预混料为交联聚乙烯吡咯烷酮、低取代羟丙纤维素粉末按照质量比分别为1∶2的混合物;粘合剂为溶有蛋白糖粉末的10%聚乙烯吡咯烷酮K30的乙醇溶液。
(3)取白首乌葛根粉末,加入稀释剂的预混料、崩解剂的预混料,于容器中混匀,加入溶有蛋白糖粉末的10%聚乙烯吡咯烷酮K30的乙醇溶液,制软材,软材用24目筛制粒,湿颗粒于60℃条件下烘干干燥,每隔0.5小时翻动1次,以加快干燥速度,干燥至颗粒水分含量5%,制备白首乌葛根含片的干颗粒(以1000片为基数,制备颗粒时稀释剂预混料的用量为165g;崩解剂用量为25g)。
3.将白首乌葛根含片的干颗粒与部分崩解剂预混料及润滑剂预混料混合均匀,压片,制备白首乌葛根含片。混合过程如下:将干燥好的颗粒,用24目筛整粒备用;将聚乙二醇4000粉末与微粉硅胶粉末按质量比2∶1混合均匀,制备润滑剂的预混料,备用;再将崩解剂预混料20g及润滑剂预混料6g采用等量递加混合法混合均匀;最后再将上述粉末与整粒后的颗粒采用等量递加法混合均匀,压片,即得白首乌葛根含片。
4.成品检验入库:将包装后的成品进行抽检,各项指标符合质量标准后,方可入库。
Claims (5)
1.一种白首乌葛根口服含片,其通过如下方法制得:
1).挑选新鲜的白首乌和葛根,经清洗、去皮、切粒后,打浆处理。将复合酶按一定比例混合后加入混合浆中酶解;酶解结束后,进行均质和喷雾干燥处理,得到白首乌粉和葛根粉。
2).以白首乌葛根粉为原料,加入稀释剂的预混料和崩解剂的预混料,以聚乙烯吡咯烷酮K30为粘合剂制粒。
3).将白首乌葛根含片的干颗粒与部分崩解剂预混料及润滑剂预混料混合均匀,压片,制备白首乌葛根含片。
2.根据权利要求1所述方法,其特征在于,步骤1)中:取果胶酶(60-80重量份)、纤维素酶(10-20重量份)、半纤维素酶(10-20重量份)混合,按混合浆0.1-0.5%的重量比加入到混合浆中,温度控制为45~55℃,时间为1~2h;均质温度为60-70℃。压力为30-40Mpa。
3.根据权利要求1所述的方法,其特征在于,所述步骤2)中制备颗粒时稀释剂的预混料可为可溶性淀粉与异麦芽糖醇粉末按质量比为1∶(2~3)的混合物;崩解剂的预混料为交联聚乙烯吡咯烷酮、低取代羟丙纤维素粉末按照质量比为1∶(1~2)的混合物;粘合剂为溶有蛋白糖粉末的10%聚乙烯吡咯烷酮K30的乙醇溶液。制备颗粒时稀释剂预混料的用量,以1000片为基数,其用量为165~172g;崩解剂预混料的用量,以1000片为基数,其用量为25~30g。
4.根据权利要求1所述方法,其特征在于,步骤3)中白首乌葛根口服含片的制粒方法为湿法制粒。
5.根据权利要求1所述的方法,其特征在于,所述步骤3)中混合过程如下:将干燥好的颗粒,用24目筛整粒备用;将聚乙二醇4000粉末与微粉硅胶粉末按质量比2∶(0.8~1.5)混合均匀,制备润滑剂的预混料,备用;再将崩解剂预混料20g及润滑剂预混料6g采用等量递加混合法混合均匀;最后再将上述粉末与整粒后的颗粒采用等量递加法混合均匀,压片,即得白首乌葛根含片。
