目前具有减肥作用的大多是药品或者保健食品,会添加各种化学物质来达到控制食欲、腹泻等消耗能量的方式减轻体重,久服会造成胃肠道菌群紊乱、功能失调、对身体有很大损害。例如CN201710050144.1一种具有减脂减肥功能的代餐粉,包括如下组分:精制燕麦麸粉10-20份;小麦麸粉20-30份;棕榈油粉3-6份;乳清蛋白粉10-20份;水溶性膳食纤维5-10份;甜味剂0.05-0.5份;香料0.1-0.5份,将所有原料倒入混料机混合10-20分钟后喷雾干燥,即得具有减脂减肥功能的代餐粉。本发明纯物理加工的精制燕麦麸粉不仅大幅降低了原料淀粉含量,使其具有低热量的特点,而且增加了维生素、矿物质和蛋白质的营养密度,并且富含天然结构的燕麦β葡聚糖。与生物酶解技术制备的燕麦β葡聚糖不同,精制燕麦麸粉中燕麦β葡聚糖保留了天然大分子结构。在人体每餐食用的天然结构燕麦β葡聚糖达到3克以上时,人体胃肠道中将形成极高粘度的食物糜,这不仅增加了人体饱腹感,更为重要的是大幅减缓和减少了糖分和脂肪的吸收,使人体餐后血糖得到有效控制,起到低升糖饮食的重要作用。生理学理论表明高血糖水平刺激胰岛素的分泌,胰岛素具有促进脂肪合成的作用,对减脂、减肥不利。因此,减肥人群需要通过低升糖饮食,降低餐后胰岛素水平,从而促进了脂肪的分解,实现减脂和减肥的目的。

发明内容
本发明的目的是针对现有技术中存在的技术缺陷,而提供一种蛋白谷物果蔬粉固体饮料及制备方法,该固体饮料用于减肥、改善便秘、补充人体必需的营养素和矿物质、增强体质少生病。
为实现本发明的目的所采用的技术方案是:
一种蛋白谷物果蔬粉固体饮料,按质量份数,包括:

本发明还提供一种所述蛋白谷物果蔬粉固体饮料的制备方法,将80目筛后称量好的香芋粉、阿萨伊果粉、牛磺酸、瓜拉那提取物、绿咖啡粉、马铃薯提取物、白芸豆粉、共轭亚油酸甘油酯、中链甘油三酯微囊粉、松籽油微囊粉、荷叶粉在搅拌混合设备中混合60分钟,再与其余80目筛后称量好的原料混合120分钟,即得成品。
本组方中,共轭亚油酸甘油酯、中链甘油三酯微囊粉、松籽油微囊粉、荷叶粉、卵磷脂粉具有减脂减肥的功能,藜麦粉、黑芝麻粉、大米粉、大豆分离蛋白粉、黑豆粉、荞麦粉、黑米粉、糙米粉、红豆粉、大豆粉、玉米粉、白芝麻粉、豌豆蛋白、薏米粉、大豆肽粉、胶原蛋白粉、南瓜粉、山药粉、胡萝卜粉为天然植物粉或提取的精华粉,含有丰富的膳食纤维,对消化系统起着重要作用,摄取足够的纤维不仅能增强消化功能、稀释和加速食物中的致癌物质和有毒物质的移除,还可以预防心血管疾病、癌症、糖尿病以及其它疾病。且膳食纤维可以给人以饱腹感,让人体摄取更少的食物而不会感觉到饥饿,达到减肥的目的。

制备方法:
将称量好的香芋粉、阿萨伊果粉、牛磺酸、瓜拉那提取物、绿咖啡粉、马铃薯提取物、白芸豆粉、共轭亚油酸甘油酯、中链甘油三酯微囊粉、松籽油微囊粉、荷叶粉在搅拌混合设备中混合60分钟,再与其余原料混合120分钟,即得成品。
其中,各原辅料分别过80目筛后,称量。

药学试验:
动物试验
1.1动物:
清洁级雄性SD大鼠60只,6~8周龄,体重160~200g,由浙江省实验动物中心提供,动物许可证号SCXK(浙)2014-0001。饲养环境温度20~25℃,湿度40%~60%,12h/12h光亮/黑暗条件。
1.2药物与试剂:
试验用样品:实施例1的蛋白谷物果蔬粉固体饮料;
奥利司他(浙江海正药业股份有限公司,15051901);
高糖高脂饲料(浙江省实验动物中心);
ELISA检测试剂盒(均购自上海源叶生物科技有限公司,E20160301A);
伊红染液、苏木素染液(南京建成科技有限公司,20151216)。
健康成年雌性BALB/C小鼠,体重18~22g,华西医科大学试验动物中心提供(合格证号:川实动管质第64号)。
1.3试验方法
1.3.1肥胖模型的制备与分组:
60只雄性SD大鼠适应性喂养1周后,其中50只喂食高糖高脂饲料制备DIO模型,另10只作为正常组,给予正常饲料。造模期间,各组大鼠自由饮水、摄食。造模8周后、以体重超过正常组大鼠体重的10%作为肥胖标准,选取40只造模成功的DIO大鼠,继续给予高糖高脂饲料。
根据体重随机分成模型组,奥利司他组(32.4mg/kg),蛋白谷物果蔬粉固体饮料高剂量组(200mg/kg),蛋白谷物果蔬粉固体饮料低剂量组(100mg/kg),每组10只。分别灌胃给予相应药物,连续13周,给药体积10mL/kg,模型组及正常组灌胃等量的0.4%羧甲基纤维素钠溶液。
1.3.2体重、摄食量、饮水量和Lee's指数:实验结束前1d,测定各组大鼠24h饮水量、摄食量。末次给药后,测定体重、体长(鼻尖至肛门的长度),计算Lee's指数。计算公式如下:Lee's指数=(体重×1000)^(1/3)/体长(cm)。
1.3.3肝体比及脂体比:大鼠处死后,分离肝脏、肾周及睾周脂肪并称重,计算肝体比及脂体比。计算公式如下:肝体比=肝脏质量(mg)/体重(g)×100%;脂体比=(肾周或睾周)脂肪质量(mg)/体重(g)×100%;总脂体比=(肾周+睾周)脂肪质量(mg)/体重(g)×100%。
1.3.4脂肪病理切片的比较:取睾周脂肪同一部位,10%福尔马林固定、乙醇脱水、二甲苯透明、石蜡包埋、切片、制成5μm石蜡切片,HE染色后显微镜下观察。
1.3.5统计学处理:实验数据采用x±s表示,采用SPSS 16.0软件进行统计学分析,统计学方法为单因素方差分析(One-wayANOVA)。P<0.05或P<0.01表示有统计学意义。
2结果
2.1对大鼠体重、摄食量、饮水量及Lee's指数的影响:与正常组相比,模型大鼠体重,Lee's指数升高(P<0.01);与模型组相比,雨生红球藻柑橘纤维复合固体饮料2个剂量组大鼠体重、Lee's指数均明显下降(P<0.05);而摄食量,饮水量则无明显变化(表1)。

2.2对大鼠脂体比及肝体比的影响:与正常组相比,模型大鼠肾脂体比、睾脂体比、总脂体比及肝体比升高(P<0.01);与模型组相比,蛋白谷物果蔬粉固体饮料2个剂量组大鼠肾脂体比、睾脂体比均明显下降(P<0.05);高剂量组总脂体比下降;而2个剂量组肝体比均无明显变化(表2)

2.3对大鼠睾周脂肪组织病理变化的影响:HE染色显示,给药13周后,正常大鼠脂肪细胞排列紧密,细胞边缘清晰;模型大鼠脂肪细胞体积相比于正常组明显增大(P<0.01),且细胞边缘模糊,细胞形态不规则、不完整。与模型组相比,2个剂量组均能不同程度的减小脂肪细胞的体积(P<0.01),显著改善细胞形态。
3分析
本实验采用高糖高脂饲料连续喂养8周造模,接近人类日常饮食不节的不良生活习惯,同时符合中医理论对肥胖的认识。肥胖症主要表现在体内脂肪堆积过多,而其中睾周脂肪和肾周脂肪的含量占总脂肪量的一定比例,所以常用这2个指标及它们的和与体重的比值(脂体比)来反映肥胖状况;同时,高糖高脂饲料会导致肝脏脂肪的堆积,从而引起肝脏系数的增加。而Lee's指数相当于人的身体质量指数(BMI),是评价成年大鼠肥胖程度的有效指标。本实验将大鼠体重、Lee's指数、脂肪系数、肝脏系数等指标同步比较,更全面的评价减肥药效。
实验结果表明,与正常组比较,模型组大鼠的体重、Lee's指数、肾脂体比、睾脂体比及总脂体比显著上升,提示DIO肥胖模型造模成功;给予蛋白谷物果蔬粉固体饮料组13周后,蛋白谷物果蔬粉固体饮料2个剂量组大鼠的体重、Lee's指数、肾脂体比、睾脂体比均显著下降;高剂量组大鼠总脂体比显著降低,蛋白谷物果蔬粉固体饮料对DIO大鼠具有减肥的作用。与模型组比较,蛋白谷物果蔬粉固体饮料2个剂量组大鼠的摄食量、饮水量均无明显差异,提示蛋白谷物果蔬粉固体饮料对DIO大鼠的减肥作用不是通过食欲抑制产生的。
综上所述,蛋白谷物果蔬粉固体饮料对DIO肥胖大鼠有一定的减肥作用,其作用机制可能与抑制成脂分化、减少脂肪细胞的大小相关。
临床试验:
1)一般资料:本发明人对150例病例临床观察,其中男76例,女74例;年龄最小18岁,最大60岁;单纯性肥胖57例,肥胖合并血脂血压高59例,高血脂、免疫力低下28,其他6例(包括肥胖易疲劳者、肥胖同时易患各种炎症等)。
2)治疗方法:每人每天一次,每次16g,实施例1的固定饮料。
3)疗效评定标准:
治愈:临床症状完全消失,恢复正常体重。
有效:体重减轻一部分,其余症状消失
无效:症状、体征无明显变化。
4)治疗结果:治疗结果如表3,

5)典型临床病例:
1、刘某,男,19岁,2017年单纯性肥胖,服用本品30天后,体重恢复至正常水平。
2、郭某,女,39岁,2018年血脂偏高,肥胖,服用本品30天后,血脂血压正常,体重减轻至正常水平。
3、邓某,男,46岁,2018年血脂血压高、肥胖、免疫力下降,服用本品28天后,痊愈。
4、周某、女,58岁,2017年血脂偏高,肥胖,服用本品280天后,血脂血压正常,体重减轻至正常水平。
对于实施例2、3的固体饮料,采用与实施例1同样的实验及临床试验,均取得了与实施例1相同的实验及临床试验数据结果,其中,临床试验的有效率分别97%,98%。其中,实验数据分别见下表4、5、6、7所示。
