本发明公开了一种沙棘叶中黄酮类成分的制备方法,步骤如下:将沙棘叶经过挑选、整理、去刺,烘干,粉碎后过筛;取沙棘叶粉末,用石油醚回流除去油脂、色素等杂质,挥干;将经过处理的样品放入与微波实验炉配套的双口烧瓶中,加入乙醇溶液,装好搅拌、冷凝装置,微波提取功率440‑460W,提取2‑4次,每次15‑25min,合并提取液,减压浓缩得浸膏;将浸膏经硅胶色谱柱分离,依次用体积比为65:1、45:1、17:1、9:1的氯仿‑甲醇洗脱,收集17:1洗脱部分,浓缩即得。该提取方法省时、高效、节能,可以富集沙棘叶中黄酮类成分,具有降低胆固醇,增强免疫力的作用,可以用于药品制备。

本发明涉及一种沙棘叶中黄酮类成分的制备方法。
背景技术
沙棘Hippophae rhamnoides L. 俗名醋柳、酸溜溜等,是胡颓子科沙棘属的灌木亚乔木,有很高的生态价值。我国的沙棘资源丰富,占世界沙棘总面积的90%。沙棘叶中富含黄酮类化合物,从沙棘果和叶中提取的总黄酮具有抗心肌缺血、提高耐氧能力、降低血清胆固醇及增强免疫力等广泛的药理作用,沙棘总黄酮在医疗、保健方面有着广阔的开发前景。
沙棘叶中总黄酮的提取研究主要是采用水浸提法、碱水浸提法以及溶剂回流提取法,这些方法提取时间长、提取率低。微波是一种非电离的电磁辐射,被辐射物质的极性分子在微波电磁场中快速转向及定向排列,从而产生撕裂和相互摩擦引起发热,同时可以保证能量的快速传递和充分利用。微波提取技术的研究表明,微波技术应用与天然产物的提取具有选择性高、操作时间短、溶剂耗量少、有效成分得率高的特点。

本发明的目的在于提供一种沙棘叶中黄酮类成分的制备方法。
本发明通过下面技术方案实现:
一种沙棘叶中黄酮类成分的制备方法,包括如下步骤:将沙棘叶经过挑选、整理、去刺,在100-105℃下烘烤至干,经粉碎后过40目筛;取10-20份沙棘叶粉末,用75-85份石油醚回流3-5h除去油脂、色素等杂质,挥干;将经过处理的样品放入与微波实验炉配套的双口烧瓶中,加入90-100份体积分数为70-80%乙醇溶液,装好搅拌、冷凝装置,微波提取功率440-460W,提取2-4次,每次15-25min,合并提取液,减压浓缩得浸膏;将浸膏经硅胶色谱柱分离,依次用体积比为65:1、45:1、17:1、9:1的氯仿-甲醇洗脱,收集17:1洗脱部分,浓缩即得;各原料均为重量份。
