本发明抗衰老美白祛痘霜,由以下重量份数的原料制成:石榴籽油6~8份、椰油酰肌氨酸钠3~4份、山嵛醇4~5份、氢化聚异丁烯6~8份、纯净水90~100份、抗坏血酸磷酸酯钠1.5~2份、十八碳烯二酸1~1.5份、季戊四醇松脂酸酯3~3.5份、熊果苷2~2.5份、龙头竹提取物2~2.5份、桃花提取物2~2.5份、罗汉柏枝提取物3~3.5份、黑喇叭菌提取物2.5~3份、核桃青皮凝结芽孢杆菌发酵提取物2.5~3份。

下面给出的实施例拟对本发明作进一步说明,但不能理解为是对本发明保护范围的限制,本领域技术人员根据本发明内容对本发明的一些非本质的改进和调整,仍属于本发明的保护范围。
本发明组方中,黑喇叭菌(Craterellus cornucopioides)提取物为抗炎祛痘组分,抑制THP-1细胞分泌炎症因子IL-1β。可以采用以下方法提取:将黑喇叭菌粉碎后,按料液比1:20,加入体积浓度为75%的乙醇,70~75℃超声处理浸提6~8小时后,过滤,离心收集上清液,然后采用活性炭柱过滤脱色,减压浓缩,真空冷冻干燥,即得。
黑喇叭菌提取物对THP-1细胞分泌炎症因子IL-1β的影响:取指数生长期的THP-1细胞,用培养液调节细胞密度约为1×106个/mL,接种于96 孔培养板中,与终质量浓度为20mg/mL的黑喇叭菌提取物共孵育4 h。取厌氧培养72h的痤疮丙酸杆菌,2500r/min 离心,用PBS 溶液清洗3~4 次,制成活菌悬液,按痤疮丙酸杆菌与THP-1 细胞数量100:1的比例加入96 孔板中,以不加药物孵育为阴性对照,加维甲酸( 20μmol /L)孵育为阳性对照,只加磷酸盐缓冲液( PBS) 为空白对照。置于37℃恒温,5% CO2及饱和湿度的培养箱培养24h。根据IL-1β ELISA 试剂盒说明书检测IL-1β的含量。IL-1β抑制率=分泌量降低值/分泌量初始值×100%。
炎症因子的分泌量是痤疮炎症发生的直观指标,由于人急性单核细胞THP-1发生炎症反应时特异性产生IL-1β,故通常将IL-1β作为痤疮炎症相关筛选的指标。通过ELISA法测定黑喇叭菌提取物对痤疮丙酸杆菌刺激的THP-1细胞IL-1β分泌的影响,结果见(图1)。痤疮丙酸杆菌诱导THP-1细胞大量分泌IL-1β。阳性对照药物维甲酸能有效地抑制IL-1β分泌,IL-1β分泌降低至47.33ng /L,对比阴性对照组,抑制率为83.2%。质量浓度为20mg/mL的黑喇叭菌提取物能明显抑制炎症细胞因子IL-1β的分泌,IL-1β分泌降低至12.56ng /L,抑制率为95.5%。黑喇叭菌提取物可作为抗炎祛痘组分。
核桃青皮凝结芽孢杆菌发酵提取物能够促进HDF-a细胞分泌Col-Ⅰ,从而达到抗衰老的功效,可以采用以下方法提取:将核桃青皮干燥后粉碎加入10~12倍量纯净水混合,100~120℃灭菌后,室温接种活化的凝结芽孢杆菌菌液,接种量为1.5%~2.5%;放入47℃恒温箱中,培养3~4天;然后将培养物在35~45℃低温烘干,再加入6~9倍量的70~80%的乙醇,超声提取3次,超声频率60kHz, 功率330W,每次90~120分钟,过滤,活性炭脱色,离心得上清液,减压浓缩,真空冷冻干燥,即得。
核桃青皮凝结芽孢杆菌发酵提取物对HDF-a细胞分泌Col-Ⅰ的影响:取指数生长期HDF-a细胞1×105个/mL,接种于96孔培养板,每孔100μL,于37 ℃,CO2培养箱中培养。24h后取上清液,3000r/min 离心10min。离心后按照试剂盒说明书,取上清液进行实验,依次与生物素标记抗体、HRP标记链霉亲和素、显色底物和终止液作用。以空白孔调零,在终止后15min内,在450nm波长处测量各孔的OD值。根据标准品的浓度及对应的OD值,计算出标准曲线的回归方程,再根据样本的OD 值计算对应的Col-Ⅰ浓度,最终浓度为实际测定浓度乘以稀释倍数。
在皮肤老化的过程中,细胞内MAPK信号通路被激活。该通路抑制转化生长因子β(TGF-β) 的表达同时上调基质金属蛋白酶( MMP)的水平,最终导致细胞中Col-Ⅰ分泌量降低,引起皮肤老化。所以Col-Ⅰ的分泌量是反应皮肤衰老的最直观指标。通过ELISA 法检测核桃青皮凝结芽孢杆菌发酵提取物对自然状态下HDF-a细胞分泌Col-Ⅰ的影响,核桃青皮凝结芽孢杆菌发酵提取物质量浓度为10-20mg/mL下进行抗衰老功效的考察,结果见(图2)。核桃青皮凝结芽孢杆菌发酵提取物能有效增加HDF-a 细胞分泌Col-Ⅰ的含量,可作为抗衰老化妆品添加剂应用。20mg/mL组分泌量为129.89mg /L,Col-Ⅰ分泌量提高了40.3%。
罗汉柏(THUJOPSIS DOLABRATA)枝提取物,具有抗菌和美白的功效;对黑色素的生成有抑制作用,对金黄色葡萄球菌、大肠杆菌、糠皮孢子菌、变异链球菌有良好的抑制作用。可选择西安全奥生物科技有限公司10:1罗汉柏枝提取物。龙头竹提取物能有效软化和舒缓肌肤并有效控制油脂分泌。可选择西安文竹生物科技有限公司10:1龙头竹提取物。桃花提取物为蔷薇科植物桃或山桃的花用纯净水提取加工,减压浓缩喷雾干燥而成的精细粉末,具有美白肌肤,杀菌保湿的作用。可选择陕西森弗天然制品有限公司10:1桃花提取物。
抗坏血酸磷酸酯钠作为美白祛斑剂,可以抑制酪氨酸酶活性,还原黑色素,具祛斑美白作用。十八碳烯二酸作为美白祛斑剂,可以减少酪氨酸酶的MRNA表达从而起到抑黑作用。熊果苷作为美白剂,能有效地抑制皮肤中的酪氨酸酶的活性,阻断黑色素的形成,通过自身与酪氨酶直接结合,加速黑色素的分解与排泄,从而减少皮肤色素沉积,祛除色斑和雀斑。
季戊四醇松脂酸酯为皮肤调理剂、柔润剂、成膜剂。椰油酰肌氨酸钠是天然的温和的表面活性剂。山嵛醇做乳化稳定剂和增稠剂使用,对皮肤无刺激作用。氢化聚异丁烯作成膜剂、柔润剂和黏度控制剂使用,有滋润、保湿、润滑的作用。石榴籽油能够营养肌肤、保持皮肤湿润、提高皮肤弹力;平衡皮肤pH值,从而保护皮肤免受自由基侵害;能够使龟裂、受刺激的皮肤快速恢复。
实施例1:抗衰老美白祛痘霜,通过以下步骤制成,按重量:
1)将石榴籽油7.5份、椰油酰肌氨酸钠3.5份、山嵛醇4.5份、氢化聚异丁烯6.5份,加入搅拌釜,加热至80~85℃,搅拌均匀;
2)将纯净水95份、抗坏血酸磷酸酯钠1.6份、十八碳烯二酸1.3份、季戊四醇松脂酸酯3.2份、熊果苷2.4份,加入搅拌釜,加热至60~65℃,搅拌均匀;
3)将步骤1)中的混合物和步骤2)中的混合物混合,抽真空均质8~10分钟,60~65℃条件下,搅拌15~20分钟;
4)当步骤3)中混合物温度降到40~45℃时,加入龙头竹提取物2.4份、桃花提取物2.2份、罗汉柏枝提取物3.3份、黑喇叭菌提取物2.8份、核桃青皮凝结芽孢杆菌发酵提取物2.6份,混合搅拌均匀;冷却出料。
实施例2:抗衰老美白祛痘霜,通过以下步骤制成,按重量:
1)将石榴籽油6份、椰油酰肌氨酸钠3份、山嵛醇4份、氢化聚异丁烯6份,加入搅拌釜,加热至80~85℃,搅拌均匀;
2)将纯净水90份、抗坏血酸磷酸酯钠1.5份、十八碳烯二酸1份、季戊四醇松脂酸酯3份、熊果苷2份,加入搅拌釜,加热至60~65℃,搅拌均匀;
3)将步骤1)中的混合物和步骤2)中的混合物混合,抽真空均质8~10分钟,60~65℃条件下,搅拌15~20分钟;
4)当步骤3)中混合物温度降到40~45℃时,加入龙头竹提取物2份、桃花提取物2份、罗汉柏枝提取物3份、黑喇叭菌提取物2.5份、核桃青皮凝结芽孢杆菌发酵提取物2.5份,混合搅拌均匀;冷却出料。
实施例3:抗衰老美白祛痘霜,通过以下步骤制成,按重量:
1)将石榴籽油8份、椰油酰肌氨酸钠4份、山嵛醇5份、氢化聚异丁烯8份,加入搅拌釜,加热至80~85℃,搅拌均匀;
2)将纯净水100份、抗坏血酸磷酸酯钠2份、十八碳烯二酸1.5份、季戊四醇松脂酸酯3.5份、熊果苷2.5份,加入搅拌釜,加热至60~65℃,搅拌均匀;
3)将步骤1)中的混合物和步骤2)中的混合物混合,抽真空均质8~10分钟,60~65℃条件下,搅拌15~20分钟;
4)当步骤3)中混合物温度降到40~45℃时,加入龙头竹提取物2.5份、桃花提取物2.5份、罗汉柏枝提取物3.5份、黑喇叭菌提取物3份、核桃青皮凝结芽孢杆菌发酵提取物3份,混合搅拌均匀;冷却出料。
对比例1:美容霜,通过以下步骤制成,按重量:
1)将石榴籽油7.5份、椰油酰肌氨酸钠3.5份、山嵛醇4.5份、氢化聚异丁烯6.5份,加入搅拌釜,加热至80~85℃,搅拌均匀;
2)将纯净水95份、抗坏血酸磷酸酯钠1.6份、十八碳烯二酸1.3份、季戊四醇松脂酸酯3.2份、熊果苷2.4份,加入搅拌釜,加热至60~65℃,搅拌均匀;
3)将步骤1)中的混合物和步骤2)中的混合物混合,抽真空均质8~10分钟,60~65℃条件下,搅拌15~20分钟;
4)当步骤3)中混合物温度降到40~45℃时,加入龙头竹提取物2.4份、桃花提取物2.2份、黑喇叭菌提取物2.8份、核桃青皮凝结芽孢杆菌发酵提取物2.6份,混合搅拌均匀;冷却出料。
对比例2:美容霜,通过以下步骤制成,按重量:
1)将石榴籽油7.5份、椰油酰肌氨酸钠3.5份、山嵛醇4.5份、氢化聚异丁烯6.5份,加入搅拌釜,加热至80~85℃,搅拌均匀;
2)将纯净水95份、抗坏血酸磷酸酯钠1.6份、十八碳烯二酸1.3份、季戊四醇松脂酸酯3.2份、熊果苷2.4份,加入搅拌釜,加热至60~65℃,搅拌均匀;
3)将步骤1)中的混合物和步骤2)中的混合物混合,抽真空均质8~10分钟,60~65℃条件下,搅拌15~20分钟;
4)当步骤3)中混合物温度降到40~45℃时,加入龙头竹提取物2.4份、桃花提取物2.2份、罗汉柏枝提取物3.3份、核桃青皮凝结芽孢杆菌发酵提取物2.6份,混合搅拌均匀;冷却出料。
对比例3:美容霜,通过以下步骤制成,按重量:
1)将石榴籽油7.5份、椰油酰肌氨酸钠3.5份、山嵛醇4.5份、氢化聚异丁烯6.5份,加入搅拌釜,加热至80~85℃,搅拌均匀;
2)将纯净水95份、抗坏血酸磷酸酯钠1.6份、十八碳烯二酸1.3份、季戊四醇松脂酸酯3.2份、熊果苷2.4份,加入搅拌釜,加热至60~65℃,搅拌均匀;
3)将步骤1)中的混合物和步骤2)中的混合物混合,抽真空均质8~10分钟,60~65℃条件下,搅拌15~20分钟;
4)当步骤3)中混合物温度降到40~45℃时,加入龙头竹提取物2.4份、桃花提取物2.2份、核桃青皮凝结芽孢杆菌发酵提取物2.6份,混合搅拌均匀;冷却出料。
祛痘抗炎测试,入选标准:临床症状和体征符合寻常型痤疮诊断标准,病变程度符合中国痤疮治疗指南分类法。判定疗效,减少率=(治疗前炎性皮损数-治疗后炎性皮损数)/治疗前炎性皮损数×100%。基本痊愈:炎性皮损数减少≥90%;显效:炎性皮损数减少60%~89%;好转:炎性皮损数减少20%~60%;无效:炎性皮损数减少<20%或加重。总有效率 =(显效+好转+痊愈例数)/总例数×100%。共有168个患有痤疮的试用者,分实施例1/对比例1/对比例2/对比例3四组,每天以1.25μl/cm2涂敷密度均匀涂敷两次,三天后测定结果见表1。通过测试可以看出本发明产品实施例1协同祛痘抗炎效果明显优于对比例1-3。
实施例4:美容霜,通过以下步骤制成,按重量:
1)将石榴籽油7.5份、椰油酰肌氨酸钠3.5份、山嵛醇4.5份、氢化聚异丁烯6.5份,加入搅拌釜,加热至80~85℃,搅拌均匀;
2)将纯净水95份、抗坏血酸磷酸酯钠1.6份、十八碳烯二酸1.3份、季戊四醇松脂酸酯3.2份、熊果苷2.4份,加入搅拌釜,加热至60~65℃,搅拌均匀;
3)将步骤1)中的混合物和步骤2)中的混合物混合,抽真空均质8~10分钟,60~65℃条件下,搅拌15~20分钟;
4)当步骤3)中混合物温度降到40~45℃时,加入龙头竹提取物2.4份、桃花提取物2.2份、罗汉柏枝提取物3.3份、黑喇叭菌提取物2.8份,混合搅拌均匀;冷却出料。
实施例5:美容霜,通过以下步骤制成,按重量:
1)将石榴籽油7.5份、椰油酰肌氨酸钠3.5份、山嵛醇4.5份、氢化聚异丁烯6.5份,加入搅拌釜,加热至80~85℃,搅拌均匀;
2)将纯净水95份、抗坏血酸磷酸酯钠1.6份、十八碳烯二酸1.3份、季戊四醇松脂酸酯3.2份、熊果苷2.4份,加入搅拌釜,加热至60~65℃,搅拌均匀;
3)将步骤1)中的混合物和步骤2)中的混合物混合,抽真空均质8~10分钟,60~65℃条件下,搅拌15~20分钟;
4)当步骤3)中混合物温度降到40~45℃时,加入桃花提取物2.2份、罗汉柏枝提取物3.3份、黑喇叭菌提取物2.8份、核桃青皮凝结芽孢杆菌发酵提取物2.6份,混合搅拌均匀;冷却出料。
实施例6:美容霜,通过以下步骤制成,按重量:
1)将石榴籽油7.5份、椰油酰肌氨酸钠3.5份、山嵛醇4.5份、氢化聚异丁烯6.5份,加入搅拌釜,加热至80~85℃,搅拌均匀;
2)将纯净水95份、抗坏血酸磷酸酯钠1.6份、十八碳烯二酸1.3份、季戊四醇松脂酸酯3.2份、熊果苷2.4份,加入搅拌釜,加热至60~65℃,搅拌均匀;
3)将步骤1)中的混合物和步骤2)中的混合物混合,抽真空均质8~10分钟,60~65℃条件下,搅拌15~20分钟;
4)当步骤3)中混合物温度降到40~45℃时,加入龙头竹提取物2.4份、罗汉柏枝提取物3.3份、黑喇叭菌提取物2.8份、核桃青皮凝结芽孢杆菌发酵提取物2.6份,混合搅拌均匀;冷却出料。
皮肤弹性测试:使用Cutometer(MPA580,C+K公司,德国),对受试者面部皮肤进行皮肤弹性的测定。Q1、R2、R5和R7值越接近1,说明皮肤弹性越好。共选取164名试用者,在湿度相同的环境中,分为4组,分别采用实施例1,对比例4-6。每天以1.8μl/cm2涂敷密度均匀涂敷两次,使用前测试初始值,一个月后测试结果,见表2。通过测试可以看到,采用本发明抗衰老美白祛痘霜的实施例1在使用一个月后,在防止皮肤衰老提高皮肤弹性方面,协同效果突出,显著高于对比例4-6。

Claims (3)

1.一种抗衰老美白祛痘霜,其特征在于由以下重量份数的原料制成:石榴籽油6~8份、椰油酰肌氨酸钠3~4份、山嵛醇4~5份、氢化聚异丁烯6~8份、纯净水90~100份、抗坏血酸磷酸酯钠1.5~2份、十八碳烯二酸1~1.5份、季戊四醇松脂酸酯3~3.5份、熊果苷2~2.5份、龙头竹提取物2~2.5份、桃花提取物2~2.5份、罗汉柏枝提取物3~3.5份、黑喇叭菌提取物2.5~3份、核桃青皮凝结芽孢杆菌发酵提取物2.5~3份;
所述核桃青皮凝结芽孢杆菌发酵提取物采用以下方法提取:将核桃青皮干燥后粉碎加入10~12倍量纯净水混合,100~120℃灭菌后,室温接种活化的凝结芽孢杆菌菌液,接种量为1.5%~2.5%;放入47℃恒温箱中,培养3~4天;然后将培养物在35~45℃低温烘干,再加入6~9倍量的70~80%的乙醇,超声提取3次,超声频率60kHz,功率330W,每次90~120分钟,过滤,活性炭脱色,离心得上清液,减压浓缩,真空冷冻干燥,即得;
所述黑喇叭菌提取物通过以下步骤提取:将黑喇叭菌粉碎后,按料液比1:20,加入体积浓度为75%的乙醇,70~75℃超声处理浸提6~8小时后,过滤,离心收集上清液,然后采用活性炭柱过滤脱色,减压浓缩,真空冷冻干燥,即得。
2.根据权利要求1所述的抗衰老美白祛痘霜,其特征在于由以下重量份数的原料制成:石榴籽油7.5份、椰油酰肌氨酸钠3.5份、山嵛醇4.5份、氢化聚异丁烯6.5份、纯净水95份、抗坏血酸磷酸酯钠1.6份、十八碳烯二酸1.3份、季戊四醇松脂酸酯3.2份、熊果苷2.4份、龙头竹提取物2.4份、桃花提取物2.2份、罗汉柏枝提取物3.3份、黑喇叭菌提取物2.8份、核桃青皮凝结芽孢杆菌发酵提取物2.6份。
3.一种制备以上任一项权利要求所述抗衰老美白祛痘霜的方法,其特征在于包括以下步骤:
1)将配方量的石榴籽油、椰油酰肌氨酸钠、山嵛醇、氢化聚异丁烯,加入搅拌釜,加热至80~85℃,搅拌均匀;
2)将配方量的纯净水、抗坏血酸磷酸酯钠、十八碳烯二酸、季戊四醇松脂酸酯、熊果苷,加入搅拌釜,加热至60~65℃,搅拌均匀;
3)将步骤1)中的混合物和步骤2)中的混合物混合,抽真空均质8~10分钟,60~65℃条件下,搅拌15~20分钟;
4)当步骤3)中混合物温度降到40~45℃时,加入龙头竹提取物、桃花提取物、罗汉柏枝提取物、黑喇叭菌提取物、核桃青皮凝结芽孢杆菌发酵提取物,混合搅拌均匀;冷却出料。
