本发明涉及一种灵芝破壁孢子粉牡丹子油复合软胶囊的制备方法,灵芝孢子粉过筛,淘洗、分离后烘干;烘干后的灵芝孢子粉低温细胞壁脆化,在破壁机中,在‑20℃下破壁,0℃~5℃干燥密封保存;灵芝破壁孢子粉与牡丹子油组成的油剂混合在一起,机械搅拌打浆后用胶体磨处理,得混合浆液,在50℃保温;制胶罐中升温真空脱气,加入明胶和甘油,适量甘油,得胶液;胶液压制成胶片;胶片与混合浆液通过自动旋转制囊机压制成软胶囊;定形及整形,洗擦丸,干燥晾丸,拣丸得灵芝破壁孢子粉牡丹子油复合软胶囊。本发明低温破壁技术和软胶囊明胶皮膜对氧气的隔绝作用,有效保护了灵芝破壁孢子粉和牡丹子油的功能成分活性。

背景技术
灵芝孢子粉是灵芝在生长成熟期,从灵芝菌褶中弹射出来的极其微小的卵形生殖细胞即灵芝的种子,每个灵芝孢子只有4~6个微米,是活体生物体,双壁结构,外被坚硬的几丁质纤维素所包围,人体很难充分吸收,破壁后更适合人体肠胃直接吸收。破壁后孢子粉的脂肪及水溶性多糖的含量分别比未破壁的提高了38.95%和21.83%。其他的研究也表明,破壁孢子粉中的还原糖和多肽较不破壁孢子粉易于提取,有效物质含量明显提高;更为重要的是,破壁孢子粉醇提液比不破壁孢子粉具有更强的体外毒杀癌细胞的活性。破壁灵芝孢子粉含有灵芝多糖、灵芝多肽、三萜类、氨基酸、蛋白质等活性物质,破壁后这些生物活性物质易被空气中的氧气氧化,所以要密封保存。
灵芝孢子油破壁方法有滚压法、高压均质法、生物酶法、高压爆破法、震荡磨法等,滚压法破壁温度低,但破壁效率低;高压均质法需要以溶剂为媒介,破壁时温度可升高至100℃,高温易使功能成分失活;生物酶法破壁效率低,破壁率一般在60%以下;高压爆破法设备复杂,破壁率低;传统震荡磨法破壁效率高,存在重金属超标问题。本专利将经低温脆化与震荡磨破壁相结合,在保证破壁率的前提下降低破壁时间,解决了震荡磨破壁存在的重金属超标问题,同时,低温破壁可保持功能成分活性。
牡丹子仁含油量丰富,含油率在24.12%~33.00%;牡丹子仁油的主要成分为亚麻酸、油酸、亚油酸、棕榈酸和硬脂酸,其中不饱和脂肪酸含量为83.05%~90.0%,亚麻酸含量达31.56%~66.85%,是中国特有的木本坚果油,营养丰富而独特,且有降血脂、降血糖等医疗保健作用,是一种极具开发潜力的油料资源。
软胶囊也称软胶丸剂,它是将油类或对明胶物无溶解作用的非水溶性的液体或混悬液等封闭于胶囊壳中而成的一种制剂,其形状有圆形,椭圆形,鱼形,管形等。其具有如下优点:液状油性物可直接封入胶囊,无需使用吸附,包埋之类的添加剂,而且软胶囊油性物含量高达60%~85%(重量),而象包接物一般的含量在50%以下。
灵芝破壁孢子粉抗氧化活性物质含量很高,牡丹子油不饱和脂肪酸含量达90%以上,常规产品中因为与氧气接触,灵芝破壁孢子粉和牡丹子油中的生物活性物质容易被氧化,从而使保健功能降低,制备成软胶囊可保持功能成分活性。
CN104586902A公开了一种超临界技术生产北虫草灵芝孢子粉软胶囊的方法,虫草孢子粉和灵芝孢子粉按比例混合后,在温度50℃一60℃,压力20Mpa~30Mpa,萃取1.5~3小时,得精油和水溶性有效成分;萃取出的精油在40℃下加入橄榄油、蜂蜡搅匀,将辅酶Q、天然维生素E;按重量比,混合精油、橄榄油放入蜂蜡中,搅匀后过胶体磨,过筛,存放,明胶加水浸泡与甘油、水混合成明胶液混合物,刮去泡沫后在40℃下保温,精油混合物与明胶混合物混合制成软胶囊,在20℃下保温获得成型软胶囊,整粒,用90%的酒精洗涤、吹干,制成成品软胶囊;在 GMP条件下经过杀菌、包装、检验、出厂。
CN1836676公开了一种以灵芝孢子粉为主要药物而制成的软胶囊,其配方组成包括一定比例的稀释剂和助悬剂,该软胶囊具有较好稳定性。同时还公开了该软胶囊的制备方法和质量控制方法。灵芝孢子粉软胶囊囊液包含灵芝孢子粉、稀释剂和助悬剂,稀释剂为植物油或聚乙二醇-400,助悬剂为蜂蜡、聚乙二醇-6000或卵磷脂中的任一种。
发明内容
本发明的目的是针对上述现状,旨在提供一种具有灵芝破壁孢子粉、牡丹子油的营养和保健功能,且加工工艺可保证功能成分不流失;产品具有提高人体免疫力、清血脂等功效,有功效加成作用的灵芝破壁孢子粉牡丹子油复合软胶囊的制备方法。
本发明目的的实现方式为,灵芝破壁孢子粉牡丹子油复合软胶囊的制备方法,具体步骤如下:
1)将灵芝孢子粉过40目筛,淘洗、分离后50℃~60℃烘干;
2)取经步骤1)烘干的灵芝孢子粉在-20℃~-25℃冷冻,同时,开启震荡磨制冷机,震荡磨温度降到-20℃~-25℃,然后加入破壁机,在-20~-25℃下破壁 30~40min,于0℃~5℃干燥环境密封保存;
3)将经步骤2)破壁灵芝孢子粉20~30份与牡丹子油70~80份组成的油剂混合在一起,机械搅拌打浆后用胶体磨处理,得到混合浆液,在50℃保温;
4)在制胶罐中先放入蒸馏水,升温至50℃,真空脱气,然后保温并加入明胶和甘油,再加入所需量的甘油搅拌均匀后抽真空,胶液应无气泡,50℃保温;
所述水与明胶的比为1:1;
5)将步骤4)得到的胶液放入80~90℃保温箱内,取出压制成胶片;将制成合格的胶片与步骤3)得到的混合浆液通过自动旋转制囊机压制成软胶囊;
6)将压制成的软胶囊在网机内20℃下吹风定形,待定形4小时后,整形:
7)用乙醇在洗擦丸机中洗去经步骤6)整形胶囊表面油层,吹干洗液;再在网机内吹干;
8)将经步骤7)干燥后的软胶囊进行人工拣丸或机械拣丸拣丸,合格的软胶囊丸放入洁净干燥的容器中,称量,得到灵芝破壁孢子粉牡丹子油复合软胶囊。
本发明利用低温脆化破壁技术和软胶囊明胶皮膜对氧气的隔绝作用,有效保护了灵芝破壁孢子粉和牡丹子油的保健成分,可保证功能成分不流失;将具有保健作用的牡丹子油与灵芝破壁孢子粉相结合,产品具有两种原料的保健作用,还有功效加成作,有提高人体免疫力、清血脂等功效;且食用方便。
具体实施方式
灵芝破壁孢子粉是公认的具有提高免疫力、抗肿瘤等作用的保健品,牡丹子油也是最近几年兴起的保健用油。灵芝孢子粉中不饱和脂肪酸等抗氧化成分含量很高,破壁后这些成分易被空气中的氧气氧化从而使保健作用降低,铝塑包装等常规的产品形态不能使物料与氧气充分隔绝,而软胶囊可以达到这个目的,使灵芝破壁孢子粉功效很好地保持。
本发明将灵芝孢子粉过筛,淘洗、分离后烘干;烘干后的灵芝孢子粉低温细胞壁脆化,在破壁机中,在-20℃下破壁,0℃~5℃干燥密封保存;灵芝破壁孢子粉与牡丹子油组成的油剂混合在一起,机械搅拌打浆后用胶体磨处理,得混合浆液,在50℃保温;制胶罐中升温真空脱气,加入明胶和甘油,适量甘油,得胶液;胶液压制成胶片;胶片与混合浆液通过自动旋转制囊机压制成软胶囊;定形及整形,洗擦丸,干燥晾丸,拣丸得灵芝破壁孢子粉牡丹子油复合软胶囊。
拣丸采用人工拣丸或机械拣丸,拣去大小丸、异形丸、明显网印丸、漏丸、瘪丸、薄壁丸、气泡丸等,
下面用具体实施例详述对本发明。
实施例1
1)将灵芝孢子粉过40目筛,淘洗、分离后50℃烘干
2)取经步骤1)烘干的灵芝孢子粉在-20℃冷冻,同时,开启震荡磨制冷机,震荡磨温度降到-20℃,然后加入破壁机,在-20℃下破壁30min,破壁率可达90%,于0℃干燥环境密封保存;
3)将经步骤2)破壁灵芝孢子粉20份与牡丹子油80份组成的油剂混合在一起,机械搅拌打浆后用胶体磨处理,得到混合浆液,在50℃保温;
4)在制胶罐中先放入蒸馏水,升温至50℃,真空脱气,然后保温并加入明胶和甘油,再加入所需量的甘油搅拌均匀后抽真空,胶液应无气泡,50℃保温;
所述水与明胶的比为1:1;
5)将步骤4)得到的胶液放入80℃保温箱内,取出压制成胶片;将制成合格的胶片与步骤3)得到的混合浆液通过自动旋转制囊机压制成软胶囊;

本实施例所得灵芝破壁孢子粉牡丹子油复合软胶囊为深棕色,椭圆形,内部为灵芝破壁孢子粉和牡丹子油悬浊液,外被明胶,使用方法为温开水冲服,有提高人体免疫力、清血脂等作用。
实施例2,同实施例1,不同的是,
1)将灵芝孢子粉过40目筛,淘洗、分离后55℃烘干
2)取经步骤1)烘干的灵芝孢子粉在-20℃冷冻,同时,开启震荡磨制冷机,震荡磨温度降到-25℃,然后加入破壁机,在-25℃下破壁40min,于0℃干燥环境密封保存;
3)将经步骤2)破壁灵芝孢子粉20份与牡丹子油80份组成的油剂混合在一起,机械搅拌打浆后用胶体磨处理,得到混合浆液,在50℃保温;
5)将步骤4)得到的胶液放入85℃保温箱内,取出压制成胶片;将制成合格的胶片与步骤3)得到的混合浆液通过自动旋转制囊机压制成软胶囊。
实施例3,同实施例1,不同的是,
1)预处理:将灵芝孢子粉过40目筛,淘洗、分离后60℃烘干备用;
2)破壁:烘干后的灵芝孢子粉在-25℃冷冻,同时,开启震荡磨制冷机,使震荡磨温度降到-22℃,然后将物料加入破壁机,在-22℃下破壁,破壁时间35min,破壁率可达95%,密封保存于5℃干燥环境;
3)胶体磨处理:将25份灵芝破壁孢子粉粉剂与75份牡丹子油组成的油剂混合在一起,机械搅拌打浆后用胶体磨处理,得到混合浆液,在50℃保温。
5)将步骤4)得到的胶液放入90℃保温箱内,取出压制成胶片;将制成合格的胶片与步骤3)得到的混合浆液通过自动旋转制囊机压制成软胶囊。
实施例4、同实施例1,不同的是,
1)预处理:将灵芝孢子粉过40目筛,淘洗、分离后50℃烘干备用;
2)破壁:烘干后的灵芝孢子粉在-25℃冷冻,同时,开启震荡磨制冷机,使震荡磨温度降到-25℃,然后将物料加入破壁机,在-25℃下破壁,破壁时间35min,破壁率可达94%,密封保存于3℃干燥环境;
3)胶体磨处理:将30份灵芝破壁孢子粉粉剂与70份牡丹子油组成的油剂混合在一起,机械搅拌打浆后用胶体磨处理,得到混合浆液,在50℃保温。
5)将步骤4)得到的胶液放入85℃保温箱内,机压制胶片,取出压制成胶片;将制成合格的胶片与步骤3)得到的混合浆液通过自动旋转制囊机压制成软胶囊。
实施例5,同实施例1,不同的是,
1)预处理:将灵芝孢子粉过40目筛,淘洗、分离后55℃烘干备用;
2)破壁:烘干后的灵芝孢子粉在-23℃冷冻,同时,开启震荡磨制冷机,使震荡磨温度降到-25℃,然后将物料加入破壁机,在-25℃下破壁,破壁时间40min,破壁率可达97%,密封保存于0℃干燥环境;
5)将步骤4)得到的胶液放入87℃保温箱内,机压制胶片,取出压制成胶片;将制成合格的胶片与步骤3)得到的混合浆液通过自动旋转制囊机压制成软胶囊。
Claims (1)
1.灵芝破壁孢子粉牡丹子油复合软胶囊制备方法,其特征在于:具体步骤如下:
1)将灵芝孢子粉过40目筛,淘洗、分离后50℃~-60℃烘干
2)取经步骤1)烘干的灵芝孢子粉在-20℃~-25℃冷冻,同时,开启震荡磨制冷机,震荡磨温度降到-20℃~-25℃,然后加入破壁机,在-20~-25℃下破壁30~40min,于0℃~5℃干燥环境密封保存;
3)将经步骤2)破壁灵芝孢子粉20~30份与牡丹子油70~80份组成的油剂混合在一起,机械搅拌打浆后用胶体磨处理,得到混合浆液,在50℃保温;
4)在制胶罐中先放入蒸馏水,升温至50℃,真空脱气,然后保温并加入明胶和甘油,再加入所需量的甘油搅拌均匀后抽真空,胶液应无气泡,50℃保温;

8)将经步骤7)干燥后的软胶囊进行人工拣丸或机械拣丸拣丸,合格的软胶囊丸放入洁净干燥的容器中,称量,得到灵芝破壁孢子粉牡丹子油复合软胶囊。
