Abstract
本发明公开了一种水基型医用超声耦合剂及其制备方法与使用方法,涉及医用超声耦合剂技术领域,包括以下重量份数的原料:枫香树叶40‑80份、牡丹皮30‑50份、黄芩35‑60份、苦参35‑60份、仙人掌40‑80份、松针50‑80份、燕麦60‑90份、竹茹50‑90份、香菇45‑65份、新鲜嫩丝瓜60‑90份、玫瑰花20‑50份、紫苏20‑40份、紫草20‑40份、天竺葵35‑50份、佛手柑30‑50份、小麦淀粉20‑40份、藻蓝蛋白5‑20份、聚丙烯酸钠5‑10份。本发明制备的超声耦合剂采用水作为溶剂,以中药植物提取物作为主要成分,其水溶性强,涂抹效果好,涂抹后水洗难度小,对人体无毒无害,对环境无污染。

技术领域
本发明涉及医用超声耦合剂技术领域,尤其涉及一种水基型医用超声耦合剂及其制备方法与使用方法。
背景技术
当超声波入射到两种不同媒质的分界面时,两种媒质阻抗相差越大,反射系数越大,穿过界面进入另一媒质的声能就越少。对于医用超声,如果让探头与皮肤“干接触”,由于二者之间空气层的强烈反射作用,探头所发射的超声波根本无法进入人体。为此,必须在探头表面和皮肤之间充填某种物质,以驱除空气,形成使超声波顺畅和不失真传播的通道,这种物质就称为医用超声耦合剂。
使用超声耦合剂的目的首先是充填探头与皮肤接触面之间的微小空隙,排除这些空隙间的微量空气对超声穿透的影响;其次是通过耦合剂的过渡作用,减小探头与皮肤之间的声阻抗差,从而减少超声能量在此界面的反射损失。另外,还起润滑作用,减小探头面与皮肤之间的摩擦,使探头能灵活地滑动探查。近年来,为防止临床交叉感染,以及满足使用超声引导穿刺所需的凝胶条件要求,国内外都已开始生产消毒、灭菌型耦合凝胶,使其具有活组织生物相容性并可生物降解,且润滑特性优异,可涂布于探头或护套内外,从而达到透声、润滑、与活组织生物相容、防止医源性交叉感染的综合效果。但是,现有的超声耦合剂大都采用水相和油相混合制备而成,耦合剂水溶性不强,不易溶解于水,检查结束后不易清洗,尤其是涂抹过程中被弄脏衣物,清洗难度更大。
发明内容
有鉴于此,本发明的目的是提供一种水基型医用超声耦合剂,使耦合剂水溶性强,清洗过程中易溶解于水,便于检查结束后耦合剂的清洗。
本发明通过以下技术手段解决上述技术问题:

进一步,小麦淀粉为小麦多孔淀粉,其制备方法如下:
取小麦淀粉,加入3-5倍体积的磷酸氢二钠和柠檬酸组成的缓冲液,调节pH至4-5,得到淀粉乳液,将得到淀粉乳液在50℃的恒温水浴锅中预热15min,然后取与小麦淀粉等量的α-淀粉酶与糖化酶,混合后用去离子水稀释,并将稀释酶液加入淀粉乳液中酶解反应5h,反应结束后加入4%的氢氧化钠溶液终止反应,然后离心,将沉淀进行抽滤,水洗3-4次后,放入60℃的烘箱中干燥,粉碎,即得到小麦多孔淀粉。
进一步,小麦多孔淀粉的制备过程中,α-淀粉酶与糖化酶的混合比例为1.5:1,稀释比例为料液比1:3-1:5。
此外,本发明还提供了一种水基型医用超声耦合剂的制备方法,包括以下工艺步骤:
1)取枫香树叶份、牡丹皮份、黄芩份、苦参份、仙人掌、松针混合,用粉碎机粉碎并过20目筛,然后加入2倍质量的去离子水,于90℃下搅拌提取2h,再用80目滤网过滤,滤液在5000r/min的转速下离心10min,取上清液过0.8μm孔径纸板精滤,将滤液用旋转蒸发仪在70℃条件下真空浓缩至浆状,得到杀菌组份;
2)取燕麦、竹茹、香菇,与去离子水按1:7的料液比混合,于90℃提取6h后,用旋转蒸发仪在70℃条件下真空浓缩至浆状,得到保湿组份;
3)取新鲜嫩丝瓜,洗净后绞碎绞汁,加入3倍量的去离子水,搅拌均匀后,过滤除去滤渣,得到润肤组份;
4)取玫瑰花、紫苏、紫草、天竺葵、佛手柑,加入2倍质量的去离子水,于90℃下搅拌提取4h,过滤除去滤渣,得到防腐组份;
5)将步骤1)-4)中得到的杀菌组份、保湿组份、润肤组份和防腐组份混合后,于60℃下搅拌加热30min,然后加入小麦淀粉、藻蓝蛋白、聚丙烯酸钠,搅拌均匀后,冷却至室温,即得到超声耦合剂。
本发明一种水基型医用超声耦合剂的使用方法如下:使用前,先将超声耦合剂于12-18℃下进行冷冻处理,然后直接涂抹于皮肤上即可。冷冻处理后的超声耦合剂能够收缩毛孔,消炎止血效果更好,能够防止伤口流出组织液,有利于超声诊断和伤口的愈合。
本发明所用的中药如下:
枫香树叶:质脆,易破碎,揉之有清香气,味辛、微苦涩,用于胃脘疼痛,伤暑腹痛,痢疾,泄泻,痈肿疮疡,湿疹,吐血,咳血,创伤出血。
牡丹皮:味苦、辛,微寒,归心、肝、肾经,具有清热凉血、活血化淤、退虚热等功效。
黄芩:味苦、性寒,具有抗炎、抗菌、抗氧化、神经保护和抗癌作用;还有清热燥湿、止血、泻火解毒等功效。
苦参:味苦,性寒,具有抗菌、抗炎、抗风湿、抗肿瘤、抗过敏、抗病毒、免疫及生物反应调节作用等多方面的药理作用和功效。
仙人掌:味苦涩,性寒,无毒,具有清热解毒、止痛消肿等功效;此外,还可以治疗烧伤、冻伤、烫伤、手癣、腮腺炎和粉刺等病症。
松针:味苦、涩,性温,用于祛风活血,明目,安神,解毒,止痒,除口臭,去口干舌燥,治便秘;并且松针中富含植物杀菌素,抗菌性好,而且不产生抗药性。
燕麦:燕麦中富含β-葡聚糖网、麦冬多糖阿、枸杞多糖和芦荟多糖同等都具有良好的保湿效果,β-葡聚糖是一种线性无分支粘多糖,通过β-(1-3)和β-(1-4)糖苷键把β-D-吡喃葡萄糖单位连接起来,形成一种高分子的聚合物,因其含有大量的亲水基团,可以吸收水分或锁住皮肤角质层水分,燕麦β-葡聚糖具有非常好的保湿功效,可以促进成纤维细胞合成胶原蛋白,促进伤口愈合,具有良好的皮肤修复功能。
竹茹:味甘、微寒,归肺、胃经,具有清热化痰、除烦止呕的作用,可用于治疗痰热咳嗽、胆火挟痰、烦热呕吐、惊悸失眠及中风痰迷等症状。浙江大学龚金炎从竹子外表皮中提取竹子精华素EZR2005,并对其保湿效果进行了研究,通过皮肤含水量测试仪以及皮肤水分流失测试仪测试,得出EZR2005具有显著的保湿性能,效果略逊于透明质酸。
香菇:香菇又叫香簟、香菌,是世界上最著名的食用菌之一,除食用外,香菇也可入药,是食药两用菌;香菇中富含香菇多糖,香菇多糖可改善肌体代谢,增强机体对多种细菌、寄生虫、病毒性感染(包括艾滋病)的免疫能力。
新鲜嫩丝瓜:丝瓜含蛋白质、脂肪、碳水化合物、钙、磷、铁及维生素B1、维生素C,还有皂甙、植物粘液、木糖胶、丝瓜苦味质、瓜氨酸等;其中B族维生素具有防止皮肤老化的作用,维生素C具有皮肤增白的作用,能保护皮肤、消除斑块,使皮肤洁白、细嫩,是不可多得的美容佳品,故丝瓜汁“美人水”之称。
玫瑰花:味辛、甘,性微温。理气解郁,化湿和中,活血散瘀,含挥发油(玫瑰油),主要为香茅醇、橙花醇、丁香油酚、苯乙醇、壬醇、苯甲醇、芳樟醇、乙酸苯乙酯,以及槲皮甙、苦味质、鞣质、没食子酸、胡萝卜素、红色素等成分,添加至化妆品中不仅能调节化妆品的气味。
紫苏:紫苏中含有紫苏黄酮,紫苏黄酮对透明质酸酶的活性具有抑制效果,过抑制透明质酸酶活性,减少透明质酸的降解,保证了真皮层透明质酸的含量,从而提高皮肤的保湿效果。
紫草:味苦,性寒,具有凉血,活血,清热,解毒,抗菌、抗炎等作用,且其物提取物也含有良好的透明质酸酶抑制剂。
天竺葵:气味甜而略重,具有止血,收缩血管,气喘,肝排毒,胆结石,肾排毒,肾结石,利尿,肌肉酸痛,油性皮肤,老化皮肤活化,疱疹,皮肤苍白,减肥,促进结疤,湿疹,灼伤晒伤,癣,月经不顺,乳房充血发炎,减轻忧郁不安,驱赶恶魔,补身,除臭,抗菌,杀菌等功效。
佛手柑:味辛苦酸,性温,具有支气管炎,喉咙痛,袪痰,呼吸困难,肺结核,胆结石,增进食欲,消化不良,胀气,尿道抗菌,膀胱炎,美白,油性皮肤,面疱,粉刺,疱疹,促进伤口愈合,湿疹,癣,减轻忧郁不安,驱赶恶魔,释放心灵,补身,抗菌,杀菌,除臭等功效。
小麦多孔淀粉:多孔淀粉是指在一定的温度下对淀粉颗粒进行水解得到的一种具有蜂窝状多孔结构的改性淀粉。多孔淀粉的表面有很多直径大约为1μm左右的小孔,颗粒内部形成中空结构,孔隙率可达到50%甚至以上,并具有一定的颗粒强度。由于多孔淀粉的中空孔结构,使得多孔淀粉具有较大的比表面积,较高的孔隙率,较低的堆积密度,具有优良的吸附性能,能够使各种中药成分的混合更加均匀稳定,同时超声耦合剂内部之间,以及超声耦合剂与皮肤表面接触更加紧密,两种媒质阻抗相差更小,使超声能量损失小,分辨力高,图像清晰。
本发明的有益效果:
1.本发明制备的超声耦合剂采用水作为溶剂,以中药植物提取物作为主要成分,其水溶性强,涂抹效果好,涂抹后水洗难度小,对人体无毒无害,对环境无污染;
2.本发明制备的超声耦合剂消毒杀菌、保湿润肤效果好,能够在较长时间内持续改善探头与患者皮肤/粘膜之间的超声耦合效果,进而减小探头面与皮肤之间的摩擦,使探头能灵活地滑动探查,并且对皮肤刺激性小,消炎止血效果好,遇到伤口后不会因刺激而流出组织液,有利于超声诊断、促进伤口愈合。
具体实施方式
以下将结合实施例对本发明进行详细说明:
实施例一
一种水基型医用超声耦合剂,包括以下重量份数的原料:枫香树叶40份、牡丹皮30份、黄芩35份、苦参35份、仙人掌40份、松针50份、燕麦60份、竹茹50份、香菇45份、新鲜嫩丝瓜60份、玫瑰花20份、紫苏20份、紫草20份、天竺葵35份、佛手柑30份、小麦多孔淀粉20份、藻蓝蛋白5份、聚丙烯酸钠5份。其制备方法如下:
1)取小麦淀粉,加入3-5倍体积的磷酸氢二钠和柠檬酸组成的缓冲液,调节pH至4-5,得到淀粉乳液,将得到淀粉乳液在50℃的恒温水浴锅中预热15min,然后按比例为1.5:1的比例取与小麦淀粉等量的α-淀粉酶与糖化酶,混合后用去离子水按1:3-1:5的料液比稀释,并将稀释酶液加入淀粉乳液中酶解反应5h,反应结束后加入4%的氢氧化钠溶液终止反应,然后离心,将沉淀进行抽滤,水洗3-4次后,放入60℃的烘箱中干燥,粉碎,即得到小麦多孔淀粉;
2)取枫香树叶份、牡丹皮份、黄芩份、苦参份、仙人掌、松针混合,用粉碎机粉碎并过20目筛,然后加入2倍质量的去离子水,于90℃下搅拌提取2h,再用80目滤网过滤,滤液在5000r/min的转速下离心10min,取上清液过0.8μm孔径纸板精滤,将滤液用旋转蒸发仪在70℃条件下真空浓缩至浆状,得到杀菌组份;
3)取燕麦、竹茹、香菇,与去离子水按1:7的料液比混合,于90℃提取6h后,用旋转蒸发仪在70℃条件下真空浓缩至浆状,得到保湿组份;
4)取新鲜嫩丝瓜,洗净后绞碎绞汁,加入3倍量的去离子水,搅拌均匀后,过滤除去滤渣,得到润肤组份;
5)取玫瑰花、紫苏、紫草、天竺葵、佛手柑,加入2倍质量的去离子水,于90℃下搅拌提取4h,过滤除去滤渣,得到防腐组份;
6)将步骤2)-5)中得到的杀菌组份、保湿组份、润肤组份和防腐组份混合后,于60℃下搅拌加热30min,然后加入藻蓝蛋白、聚丙烯酸钠和步骤1)中得到的小麦多孔淀粉,搅拌均匀后,冷却至室温,即得到超声耦合剂。
实施例二
一种水基型医用超声耦合剂,包括以下重量份数的原料:枫香树叶80份、牡丹皮50份、黄芩60份、苦参60份、仙人掌80份、松针80份、燕麦90份、竹茹90份、香菇65份、新鲜嫩丝瓜90份、玫瑰花50份、紫苏40份、紫草40份、天竺葵50份、佛手柑50份、小麦多孔淀粉40份、藻蓝蛋白20份、聚丙烯酸钠10份。其制备方法与实施例一相同。
实施例三
一种水基型医用超声耦合剂,包括以下重量份数的原料:枫香树叶72份、牡丹皮40份、黄芩45份、苦参54份、仙人掌45份、松针62份、燕麦80份、竹茹56份、香菇55份、新鲜嫩丝瓜70份、玫瑰花28份、紫苏30份、紫草32份、天竺葵40份、佛手柑42份、小麦淀粉23份、藻蓝蛋白8份、聚丙烯酸钠6份。其制备方法与实施例一相同。
实施例四
一种水基型医用超声耦合剂,包括以下重量份数的原料:枫香树叶50份、牡丹皮35份、黄芩50份、苦参38份、仙人掌70份、松针54份、燕麦65份、竹茹75份、香菇50份、新鲜嫩丝瓜73份、玫瑰花42份、紫苏34份、紫草30份、天竺葵45份、佛手柑40份、小麦淀粉36份、藻蓝蛋白14份、聚丙烯酸钠8份。其制备方法与实施例一相同。
实施例五
一种水基型医用超声耦合剂,包括以下重量份数的原料:枫香树叶65份、牡丹皮46份、黄芩40份、苦参40份、仙人掌60份、松针70份、燕麦60份、竹茹55份、香菇60份、新鲜嫩丝瓜64份、玫瑰花30份、紫苏26份、紫草25份、天竺葵38份、佛手柑30份、小麦淀粉22份、藻蓝蛋白12份、聚丙烯酸钠5份。其制备方法与实施例一相同。
取60只小白鼠,每只鼠背部剪去4×4cm被毛,用直尺各量出直径10mm的圆形区并做标记;用酒棉将皮肤消毒,后用镊子将皮肤提起,用手术剪沿标线剪去皮肤,造成一个直径10mm的圆形皮肤全层创口,然后平均分成6组并用记号笔在尾根编号,分别为3个试验组(试验组1、试验组2、试验组3、试验组4、试验组5)和1个空白组,试验组1、试验组2、试验组3、试验组4和试验组5分别均匀涂以等量的实施例一至实施例五制备得到的超声耦合剂,超声耦合剂在涂抹前先于12-18℃下进行冷冻,空白组不做任处理,以做切口的当天记为第0天,次日(第l天)涂抹耦合剂,并统计耦合剂涂抹涂,涂抹后,每日观察小白鼠的精神状态情况、涂抹耦合剂后创口情况,并统计耦合剂保湿时间(h),结果如下表所示:
由上表试验结果分析可知,使用实施例一至实施例五制备得到的超声耦合剂涂抹创口的小白鼠与创口不做任处理的小白鼠相比,使用实施例一至实施例五制备得到的医用超声耦合剂涂抹创口的小白鼠精神活跃,无不良反应,创口无组织液流出,无红肿现象,而创口不做任处理的小白鼠虽无无不良反应,但精神较沉郁,创口有少量组织液流出,并出现轻度红肿;且实施例一至实施例五制备得到的医用超声耦合剂易于涂抹,涂展性好,涂抹后保湿时间均在3.5h以上。因此,实施例一至实施例五制备得到的超声耦合剂易于涂抹,涂抹效果均匀,对皮肤刺激性小、保湿时间,遇到伤口后不会因刺激而流出组织液,且保湿时间长,有利于诊断、促进伤口愈合。
另取实施例一至实施例五制备得到的超声耦合剂,分别于手背、棉布和涤纶布上涂抹一块5×5cm的超声耦合剂层,涂抹3h后分别水洗超声耦合剂层,观测清洗难易程度。经试验得到,涂抹手背、棉布和涤纶布上的超声耦合剂层均非常容易清洗掉。

1.一种水基型医用超声耦合剂,其特征在于,所述超声耦合剂包括以下重量份数的原料:枫香树叶40-80份、牡丹皮30-50份、黄芩35-60份、苦参35-60份、仙人掌40-80份、松针50-80份、燕麦60-90份、竹茹50-90份、香菇45-65份、新鲜嫩丝瓜60-90份、玫瑰花20-50份、紫苏20-40份、紫草20-40份、天竺葵35-50份、佛手柑30-50份、小麦淀粉20-40份、藻蓝蛋白5-20份、聚丙烯酸钠5-10份,所述小麦淀粉为小麦多孔淀粉。
2.根据权利要求1所述的一种水基型医用超声耦合剂,其特征在于,所述超声耦合剂包括以下重量份数的原料:枫香树叶72份、牡丹皮40份、黄芩45份、苦参54份、仙人掌45份、松针62份、燕麦80份、竹茹56份、香菇55份、新鲜嫩丝瓜70份、玫瑰花28份、紫苏30份、紫草32份、天竺葵40份、佛手柑42份、小麦淀粉23份、藻蓝蛋白8份、聚丙烯酸钠6份。
3.根据权利要求1所述的一种水基型医用超声耦合剂,其特征在于,所述超声耦合剂包括以下重量份数的原料:枫香树叶50份、牡丹皮35份、黄芩50份、苦参38份、仙人掌70份、松针54份、燕麦65份、竹茹75份、香菇50份、新鲜嫩丝瓜73份、玫瑰花42份、紫苏34份、紫草30份、天竺葵45份、佛手柑40份、小麦淀粉36份、藻蓝蛋白14份、聚丙烯酸钠8份。
4.根据权利要求1所述的一种水基型医用超声耦合剂,其特征在于,所述超声耦合剂包括以下重量份数的原料:枫香树叶65份、牡丹皮46份、黄芩40份、苦参40份、仙人掌60份、松针70份、燕麦60份、竹茹55份、香菇60份、新鲜嫩丝瓜64份、玫瑰花30份、紫苏26份、紫草25份、天竺葵38份、佛手柑30份、小麦淀粉22份、藻蓝蛋白12份、聚丙烯酸钠5份。
5.根据权利要求1所述的一种水基型医用超声耦合剂,其特征在于,所述小麦淀粉为小麦多孔淀粉,其制备方法如下:

7.一种如要求1-4任意一项所述的水基型医用超声耦合剂的制备方法,其特征在于,包括以下工艺步骤:
1)取枫香树叶份、牡丹皮份、黄芩份、苦参份、仙人掌、松针混合,用粉碎机粉碎并过20目筛,然后加入2倍质量的去离子水,于90℃下搅拌提取2h,再用80目滤网过滤,滤液在5000r/min的转速下离心10min,取上清液过0.8μm孔径纸板精滤,将滤液用旋转蒸发仪在70℃条件下真空浓缩至浆状,得到杀菌组份;
