朝鲜蓟(CynarascolymusL.),又名法国百合、荷花百合、菜蓟、“洋蓟”、“球蓟”,是一种多年生草本块根菊科菜蓟属植物,原产于地中海沿岸,19世纪由法国引入我国。其在西班牙、法国、意大利及美国加利福尼亚栽培较多。国内,朝鲜蓟主要在上海、北京、浙江、湖南、云南等地栽培。朝鲜蓟食用部分为花托的肥嫩苞片及肉质花苞,营养丰富,富含钙、磷、铁、碘、钾等矿物质,及维生素A、B、C。同时,植株含有丰富的洋蓟素、多酚类物质(包括绿原酸)、菊糖(块根中)等活性物质,经常食用具有促进消化、保护肝脏、调节脂质代谢、降血糖、抗氧化等功效,具有极高的保健功能。朝鲜蓟也是一个低热量蔬菜。其高钾低钠的特点,尤其适宜高血压、心脑血管疾病患者食用。朝鲜蓟可口又营养,但脂肪含量只有0.2%,所以是一种很好蔬菜,同时也是一种很好的保健食材。
由于朝鲜蓟本身是后期由法国传入我国,且仅在我国南方地区有一定规模的种植,在我国的辨识率较低,对于我国消费者也较为陌生。目前,我国朝鲜蓟加工产品品种单一,除了80%的花蕾用于鲜食,其余20%主要为用于生产出口的浸渍类罐头食品,另有极少部分朝鲜蓟提取物用于功能饮料的生产,但总体规模相对较小,市场规模极小。而国外更多的是利用朝鲜蓟叶的功能性成分进行保健功能产品和草药的研发。鉴于以上朝鲜蓟较强的功能性、产品的单一化、对于我国消费群体的陌生化,朝鲜蓟新产品的开发与综合利用,将具有极好的市场潜力和广阔的前景。
现代人们工作压力大、生活节奏快、生活方式不合理甚至不健康,使得人体机能出现紊乱,随之而来的各种疾病,包括高血脂、糖尿病、心血管疾病等等。以上许多症状都与肝脏的代谢、病变有关,而肝病在一定人群中成为一种高发病,也是一种高危病。而目前市场上还没有效果明显的护肝食品,而朝鲜蓟产品具有明显的养肝护肝功效,其开发潜力和空间巨大。基于朝鲜蓟调节脂质代谢、保肝护肝的功能性,主要在于含有多酚类结构的功能性成分。而此类成分大都性质活泼,受条件影响敏感,因此功能性成分含量及功能活性的保持,以及提高产品的稳定性将是产品开发中的重点,也是难点之所在。由此可见,提供一种工艺步骤简单、适合工业化生产的朝鲜蓟功能成分的提取成为本领域技术人员亟待解决的技术难题。
发明内容
本申请实施例提供了一种朝鲜蓟提取物的制备方法,提高提取物中咖啡酸衍生物的含量,并且适合工业化生产。
本申请实施例示出一种朝鲜蓟提取物的制备方法,包括以下步骤:
蒸制:将阴干的朝鲜蓟原材料于100-120℃蒸制4-8小时;
前处理:将蒸制后朝鲜蓟原材料晾干,后经切药机切碎得到净料;
提取:称取所述净料投入反应釜中,加热水回流提取,提取液经过滤后收集;
浓缩,醇沉:将收集的提取液进行浓缩,加入乙醇溶液进行醇沉,将沉降后的上清液过滤后收集;
吸附,洗脱:将收集的滤液通过树脂柱吸附,并用乙醇水溶液通过吸附柱进行洗脱,将醇洗液过滤后收集;
浓缩,喷雾干燥:将收集的醇洗液进行真空浓缩,得到浓浆后喷雾干燥、过筛,得到朝鲜蓟提取物。
可选地,所述提取步骤具体为:向不锈钢反应釜中投入所述净料,然后加入4倍热水,加热至90-95℃开启热回流,提取3次后,后两次加水量为2~3倍,,通过0.45μm滤膜滤除杂质,收集提取液。
可选地,所述浓缩,醇沉步骤具体为:将提取液浓缩至液料比为1:1~1:2或相对密度为1.08~1.15,加入乙醇溶液,配置成乙醇浓度为50%的溶液,冷却后进行醇沉,时间为12h~24h,将沉降后的上清液通过0.45μm的滤膜滤掉杂质,收集滤液。
可选地,所述吸附,洗脱步骤具体为:将滤液通过树脂柱吸附,配置浓度为15%、45%、95%的乙醇水溶液分别通过吸附后的树脂柱进行洗脱,将醇洗液经过0.22μm微孔过滤器滤除杂质,收集醇洗液。
可选地,所述浓缩,喷雾干燥步骤具体为:将收集的醇洗在45~65℃的条件下进行真空浓缩,回收乙醇,得到浓浆后喷雾干燥、过筛,得到朝鲜蓟提取物。
可选地,所述吸附,洗脱步骤,所述树脂柱的型号为D101型,AB-8型或聚酰氨型中的一种或几种。
可选地,所述前处理步骤,切药机切碎得到净料的粒径大小为40-60目。
本申请实施例示出一种朝鲜蓟提取物的制备方法,包括以下步骤:蒸制:将阴干的朝鲜蓟原材料于100-120℃蒸制4-8小时;前处理:将蒸制后朝鲜蓟原材料晾干,后经切药机切碎得到净料;提取:称取所述净料投入反应釜中,加热水回流提取,提取液经过滤后收集;浓缩,醇沉:将收集的提取液进行浓缩,加入乙醇溶液进行醇沉,将沉降后的上清液过滤后收集;吸附,洗脱:将收集的滤液通过树脂柱吸附,并用乙醇水溶液通过吸附柱进行洗脱,将醇洗液过滤后收集;浓缩,喷雾干燥:将收集的醇洗液进行真空浓缩,得到浓浆后喷雾干燥、过筛,得到朝鲜蓟提取物。本发明的朝鲜蓟提取物具有抗氧化、保护肝脏、降低胆固醇、改善胃肠功能、增强肝脏排毒、促进消化、改善血液循环、防止动脉硬化、保护心血管、促进氨基酸代谢等生理活性,另外,朝鲜蓟中含有的洋蓟酸有治疗慢性肝炎的药理活性,具有很高的应用价值。通过本发明提取工艺所得到的含有朝鲜蓟提取物的产品可进一步开发成朝鲜蓟养生茶、朝鲜蓟休闲食品或朝鲜蓟保健茶饮料等,具有广阔的应用前景。本发明的提取工艺步骤简单,成本低,可实现工业化生产,提取的朝鲜蓟提取物咖啡酸衍生物含量高,提高朝鲜蓟的精深加工与综合开发利用水平,带来良好的经济和社会效益。

本申请实施例示出一种朝鲜蓟提取物的制备方法,包括以下步骤:
S1蒸制:将阴干的朝鲜蓟原材料于100-120℃蒸制4-8小时;
S2前处理:将蒸制后朝鲜蓟原材料晾干,后经切药机切碎得到净料;
S3提取:称取所述净料投入反应釜中,加热水回流提取,提取液经过滤后收集;
S4浓缩,醇沉:将收集的提取液进行浓缩,加入乙醇溶液进行醇沉,将沉降后的上清液过滤后收集;
S5吸附,洗脱:将收集的滤液通过树脂柱吸附,并用乙醇水溶液通过吸附柱进行洗脱,将醇洗液过滤后收集;
S6浓缩,喷雾干燥:将收集的醇洗液进行真空浓缩,得到浓浆后喷雾干燥、过筛,得到朝鲜蓟提取物。
可选地,所述S3提取步骤具体为:向不锈钢反应釜中投入所述净料,然后加入4倍热水,加热至90-95℃开启热回流,提取3次后,后两次加水量为2~3倍,通过0.45μm滤膜滤除杂质,收集提取液。

实施例1:
将阴干的朝鲜蓟原材料于120℃蒸制4小时;将蒸制后朝鲜蓟原材料晾干,后经切药机切碎得到40目净料;称取所述净料500g投入反应釜中,然后加入4倍热水,加热至90℃开启热回流,提取3次后,后两次加水量为2~3倍,通过0.45μm滤膜滤除杂质,收集提取液。将提取液浓缩至液料比为1:2,加入乙醇溶液,配置成乙醇浓度为50%的溶液,冷却后进行醇沉,时间为24h,将沉降后的上清液通过0.45μm的滤膜滤掉杂质,收集滤液。将滤液通过聚酰氨树脂柱吸附,配置浓度为15%、45%、95%的乙醇水溶液分别通过吸附后的聚酰氨树脂柱进行洗脱,将醇洗液经过0.22μm微孔过滤器滤除杂质,收集醇洗液。将收集的醇洗在65℃的条件下进行真空浓缩,回收乙醇,得到浓浆后喷雾干燥、过筛,得到朝鲜蓟提取物。
采用紫外分光光度法对上述提取物中咖啡酸衍生物含量进行测定,测得咖啡酸衍生物的含量为9.11%。
实施例2:
将阴干的朝鲜蓟原材料于120℃蒸制6小时;将蒸制后朝鲜蓟原材料晾干,后经切药机切碎得到50目净料;称取所述净料500g投入反应釜中,然后加入4倍热水,加热至95℃开启热回流,提取3次后,后两次加水量为2~3倍,通过0.45μm滤膜滤除杂质,收集提取液。将提取液浓缩至液料比为1:1,加入乙醇溶液,配置成乙醇浓度为50%的溶液,冷却后进行醇沉,时间为24h,将沉降后的上清液通过0.45μm的滤膜滤掉杂质,收集滤液。将滤液通过聚酰氨树脂柱吸附,配置浓度为15%、45%、95%的乙醇水溶液分别通过吸附后的聚酰氨树脂柱进行洗脱,将醇洗液经过0.22μm微孔过滤器滤除杂质,收集醇洗液。将收集的醇洗在65℃的条件下进行真空浓缩,回收乙醇,得到浓浆后喷雾干燥、过筛,得到朝鲜蓟提取物。
采用紫外分光光度法对上述提取物中咖啡酸衍生物含量进行测定,测得咖啡酸衍生物的含量为9.82%。
实施例3:
将阴干的朝鲜蓟原材料于120℃蒸制8小时;将蒸制后朝鲜蓟原材料晾干,后经切药机切碎得到60目净料;称取所述净料500g投入反应釜中,然后加入4倍热水,加热至95℃开启热回流,提取3次后,后两次加水量为2~3倍,通过0.45μm滤膜滤除杂质,收集提取液。将提取液浓缩至液料比为1:2,加入乙醇溶液,配置成乙醇浓度为50%的溶液,冷却后进行醇沉,时间为20h,将沉降后的上清液通过0.45μm的滤膜滤掉杂质,收集滤液。将滤液通过聚酰氨树脂柱吸附,配置浓度为15%、45%、95%的乙醇水溶液分别通过吸附后的聚酰氨树脂柱进行洗脱,将醇洗液经过0.22μm微孔过滤器滤除杂质,收集醇洗液。将收集的醇洗在55℃的条件下进行真空浓缩,回收乙醇,得到浓浆后喷雾干燥、过筛,得到朝鲜蓟提取物。
采用紫外分光光度法对上述提取物中咖啡酸衍生物含量进行测定,测得咖啡酸衍生物的含量为8.86%。

Claims (7)
1.一种朝鲜蓟提取物的制备方法,其特征在于,包括以下步骤:
蒸制:将阴干的朝鲜蓟原材料于100-120℃蒸制4-8小时;
前处理:将蒸制后朝鲜蓟原材料晾干,后经切药机切碎得到净料;
提取:称取所述净料投入反应釜中,加热水回流提取,提取液经过滤后收集;
浓缩,醇沉:将收集的提取液进行浓缩,加入乙醇溶液进行醇沉,将沉降后的上清液过滤后收集;
吸附,洗脱:将收集的滤液通过树脂柱吸附,并用乙醇水溶液通过吸附柱进行洗脱,将醇洗液过滤后收集;
浓缩,喷雾干燥:将收集的醇洗液进行真空浓缩,得到浓浆后喷雾干燥、过筛,得到朝鲜蓟提取物。

7.根据权利要求1所述的制备方法,其特征在于,所述前处理步骤,切药机切碎得到净料的粒径大小为40-60目。
