Abstract
The invention discloses a food, a health-care product or a pharmaceutical composition, which is a preparation prepared from the following raw materials in parts by weight: 50-100 parts of ginkgo, 10-40 parts of ginseng, 20-50 parts of angelica, 50-120 parts of medlar, 50-100 parts of sea buckthorn, 100 parts of raw hawthorn, 110 parts of kudzuvine root, 180 parts of pine mushroom, 10-20 parts of auricularia polytricha and 10-90 parts of highland barley. Also discloses a preparation method and application of the food, the health care product or the pharmaceutical composition. The food, the health care product or the pharmaceutical composition provided by the invention has the advantages of simple preparation process and low cost, and is suitable for mass production; the food, the health-care product or the pharmaceutical composition can obviously increase the times of mouth opening and obviously prolong the gasp stopping time after the head of a mouse is cut off, can effectively improve the hypoxia tolerance of the body, has no toxic or side effect, can be taken for a long time, and has good market application prospect.

Technical Field
The invention relates to a food, a health-care product or a pharmaceutical composition for improving anoxia resistance, a preparation method and application thereof.
Background
Oxygen is very important to sustain life activities of the body. Hypoxia is a pathological process in which the metabolism, function, and even morphological structure of a tissue are abnormally changed due to insufficient oxygen supply or oxygen deficiency of the tissue. Over-strong or long-term hypoxia stress can bring serious harm to the organism, and the death of important organs of the organism, such as heart, brain, and the like, can be caused due to insufficient energy supply if the organism is not treated in time.
Hypoxia is of many types, mainly classified as environmental, physiological, pathological and motor hypoxia. People are extremely easy to be damaged by oxygen deficiency and appear as environmental oxygen deficiency when working and fighting in special environments such as plateau, high altitude, diving, tunnel and the like; the body of the old is in a chronic hypoxia state with different degrees due to physiological aging of each organ, and the physiological hypoxia is shown; patients with respiratory system diseases and cardiovascular and cerebrovascular diseases have pathological hypoxia caused by the reduction of oxygen carrying capacity and oxygen uptake capacity of blood; the oxygen consumption in the body of a high-intensity mental and nervous manual worker is not enough to compensate the oxygen consumption in the body by simply breathing through the lung due to the rapid increase of the oxygen consumption in the body, so that cells are in an anoxic environment and show exercise hypoxia. The improvement of the hypoxia tolerance of the organism is to reduce the damage of hypoxia to the organism by means of reducing the hypoxia stress intensity of the organism or promoting the establishment of hypoxia adaptation and the like, so that the organism can maintain normal physiological functions as far as possible in a long-term hypoxia environment.
At present, the main clinical and common hypoxia-resistant western medicines include calcium channel blockers (nimodipine and flunarizine), free radical scavengers (allopurinol) and some central inhibitory drugs (hibernating mixture), but the drugs have non-negligible toxic and side effects, can only be used for treating various transient hypoxic diseases, and cannot meet the requirements of long-term operation on the hypoxia-resistant medicines in special environments such as plateau, aviation, aerospace and diving.
In recent years, people have conducted extensive research on the effective components and pharmacological effects of traditional Chinese medicines, and found that many single traditional Chinese medicines and extracts thereof have anti-hypoxia effects, such as: rhodiola root, angelica, hovenia acerba, sea buckthorn, ginseng, kudzu root and the like. Compared with single medicine, the traditional Chinese medicine compound preparation prepared according to the monarch, minister, assistant and messenger formula of the traditional Chinese medicine prescriptions has better integrity and comprehensiveness, wider application and better curative effect. Chenyufang and the like, the research on the influence of a traditional Chinese medicine compound preparation on rat hypoxia tolerance and the action mechanism thereof, and the 3 compounds disclosed in the Chinese veterinary medicine journal 2003 and in the 06 th year have the effects of improving the hypoxia tolerance of the organism and relieving the damage of hypoxia to the organism. Application No.: 201510089468, title of the invention: the patent application of a composition for preventing and treating acute altitude stress and hypoxia tolerance discloses a composition for preventing and treating acute altitude stress and hypoxia tolerance, which is prepared from the following raw materials in parts by weight: 3-18 parts of platycodon grandiflorum, 2-20 parts of poria cocos, 3-12 parts of ligusticum wallichii, 3-12 parts of radix angelicae, 5-16 parts of uncaria and 5-20 parts of caulis polygoni multiflori.
At present, the report of improving the hypoxia tolerance of organisms by matching ginkgo, ginseng, angelica, medlar, sea buckthorn, raw hawthorn, kudzu root, tricholoma matsutake, auricularia polytricha and highland barley is not found.

The hypoxia tolerance of cells of a human body is directly related to qi and blood of the human body, and the functions of supplementing qi, nourishing yin and activating blood circulation are main treatment methods for improving the hypoxia tolerance of the cells, and the key point of sufficient qi and blood of the human body is the transportation and transformation of spleen qi and the dispersion and descent of lung qi during the qi and blood replenishing.
In the food, the health-care product or the pharmaceutical composition, the ginseng has the effects of greatly invigorating primordial qi, recovering pulse, relieving depletion, tonifying spleen, benefiting lung and promoting the production of body fluid; the angelica has the functions of enriching and activating blood, and the compatibility of the angelica and the angelica is the monarch drug for tonifying qi and enriching blood. Sea-buckthorn can promote digestion, eliminate stagnation, promote blood circulation and remove blood stasis; the raw hawthorn has the effects of promoting digestion and strengthening the stomach, promoting qi circulation and removing blood stasis, is compatible with the Chinese angelica to enhance the effects of promoting blood circulation and removing blood stasis, is compatible with the ginseng to enhance the effects of invigorating the spleen and supplementing qi, is a ministerial drug, is compatible with the radix puerariae to release muscles and promote the production of body fluid, and is compatible with the ginseng and the Chinese angelica to nourish yin and promote the production of body fluid to enrich the blood. (1) pulverizing radix Puerariae, and sieving with 100 mesh sieve.
(2) Parching semen Avenae Nudae, pulverizing, and sieving with 100 mesh sieve.
(3) Mixing 8 kinds of raw materials including semen Ginkgo, Ginseng radix, radix Angelicae sinensis, fructus Lycii, fructus Hippophae, fructus crataegi, Tricholoma matsutake and Auricularia, extracting with water twice (each for 30 min), filtering, mixing filtrates, concentrating into extract, adding radix Puerariae powder, and drying at 60 deg.C; mixing the dried product with 60g white sugar, and pulverizing; adding semen Avenae Nudae powder, 150g maltodextrin, and 90% ethanol, mixing, granulating, and drying at 60 deg.C to obtain granule.

1. The food, the health product or the pharmaceutical composition of the invention: 50g of ginkgo, 10g of ginseng, 20g of angelica, 50g of medlar, 50g of sea-buckthorn, 100g of raw hawthorn, 110g of kudzu root, 10g of tricholoma matsutake, 10g of auricularia polytricha and 10g of highland barley.
2. The preparation method comprises the following steps:

The food, the health product or the pharmaceutical composition of the invention: 100g of ginkgo, 40g of ginseng, 50g of angelica, 120g of medlar, 100g of sea buckthorn, 150g of raw hawthorn, 180g of kudzu root, 20g of tricholoma matsutake, 50g of auricularia polytricha and 90g of highland barley.
2. The preparation method comprises the following steps:
(1) pulverizing radix Puerariae, and sieving with 100 mesh sieve.
(2) Parching semen Avenae Nudae, pulverizing, and sieving with 100 mesh sieve.
(3) Mixing 8 raw materials of ginkgo, ginseng, angelica, medlar, sea buckthorn, raw hawthorn, matsutake mushroom and auricularia polytricha, extracting with water for 30min, filtering, extracting filter residue with water for 30min, mixing the two filtrates, concentrating into extract, adding kudzu root powder, and drying at 60 ℃; pulverizing the dried product and 70g white sugar, adding semen Avenae Nudae powder, 200g maltodextrin, and 90% ethanol, mixing, granulating, and drying at 60 deg.C to obtain granule.

1.2 healthy mice 160, half male and female, weight (20 + -2 g), were obtained from the animal laboratory center of the academy of traditional Chinese medicine, Sichuan province.
2 method of experiment
2.1 animal grouping and administration methods
The mice are randomly divided into 3 groups, namely a rhodiola rosea, ginseng and wolfberry capsule group as positive control drugs, a composition of the invention and a distilled water group, and 40 mice in each group. The mice were gavaged with 10mL/kg daily 1 time daily for 30 consecutive days using gavage. The mouse decapitation experiment is performed to observe the wheezing times and the wheezing time.

As shown in Table 1, the food, the health product or the pharmaceutical composition has the functions of obviously increasing the mouth opening times of mice and prolonging the gasp time of the mice. And the composition has short onset time (effective in 7 days), obvious aging relation (better than 7 days in 15 days), and better effect than a positive control group.
From the results, compared with the rhodiola rosea, ginseng and wolfberry capsule, the food, the health-care product or the pharmaceutical composition can obviously increase the times of mouth opening of the mouse after head breaking and obviously prolong the gasp stopping time, which shows that the food, the health-care product or the pharmaceutical composition can effectively improve the hypoxia tolerance of the body.

And crushing the reduced pressure dried sample and the white granulated sugar, adding the highland barley powder and the maltodextrin, mixing uniformly, and granulating.
2.2 study of drying Process
Considering that many nutrients are inactivated by high temperature heating, high temperature addition drying is excluded. If low-temperature heating drying is adopted, the time consumption is long; if freeze drying is adopted, the requirements on instruments are high, the energy consumption is high, and the requirements are not consistent with large-scale production, so that reduced pressure drying is selected, and the pressure drying temperature is optimized. The temperature of reduced pressure drying is respectively 50 ℃, 55 ℃ and 60 ℃, the extract of the prescription dose is concentrated into extract, and the extract is added with the radix puerariae for reduced pressure drying when the extract is hot.
2.3 taste-modifying study
The raw material extract has a large sour taste, and is added with white sugar for flavoring, and the addition amount of the white sugar is optimized. 3g, 4g, 5g, 6g, 7g and 8g of white sugar are added into the prescription for one day respectively to compare the original bitter taste with the existing sweetness, and products with moderate sour taste and sweetness are selected.

The product type is temporarily defined as thick soup and made into granules, and the addition amount of maltodextrin, the ethanol concentration and the drying time are optimized in consideration of good molding of the product. As can be seen from Table 2, the temperature of reduced pressure drying is 60 deg.C, and the extract-like concentrate can be obtained in the shortest time.

As shown in Table 3, when the amount of white sugar added was 6g, the sour taste was moderate and the sweetness was moderate.
3.3 Molding conditions of product particles with different maltodextrin addition amounts and different ethanol concentrations
The results of the experiment are shown in table 4:

As can be seen from Table 4, the amount of maltodextrin added was 15 g/square, the ethanol concentration was 90%, and the degree of product formation was the best at a drying temperature of 60 ℃.
From the above results, the preparation process of the food, health product or pharmaceutical composition of the invention (1) comprises pulverizing radix Puerariae, and sieving with 100 mesh sieve; (2) parching semen Avenae Nudae, pulverizing, and sieving with 100 mesh sieve; (3) mixing 8 raw materials of ginkgo, ginseng, angelica, medlar, sea buckthorn, raw hawthorn, matsutake mushroom and auricularia polytricha, extracting with water for 30min, filtering, extracting filter residue with water for 30min, mixing the two filtrates, concentrating into extract, adding kudzu root powder, and drying at 60 ℃; mixing the dried product with 6g of white granulated sugar, and pulverizing; adding semen Avenae Nudae powder, 15g maltodextrin, and 90% ethanol, mixing, granulating, and drying at 60 deg.C to obtain granule; is a preferred process.
In conclusion, the food, the health-care product or the pharmaceutical composition provided by the invention has the advantages of simple preparation process, low cost and suitability for mass production; the food, the health-care product or the pharmaceutical composition can obviously increase the times of mouth opening and obviously prolong the gasp stopping time after the head of a mouse is cut off, can effectively improve the hypoxia tolerance of the body, has no toxic or side effect, can be taken for a long time, and has good market application prospect.

1. A food, health product or pharmaceutical composition for improving anoxia endurance is characterized in that: the preparation is prepared from the following raw materials in parts by weight: 90 parts of ginkgo, 15 parts of ginseng, 30 parts of angelica, 100 parts of medlar, 90 parts of sea buckthorn, 120 parts of raw hawthorn, 150 parts of kudzu root, 15 parts of tricholoma matsutake, 30 parts of auricularia polytricha and 50 parts of highland barley.

The food, nutraceutical, or pharmaceutical composition of claim 1, wherein: the composition is a preparation prepared by taking raw medicinal materials of ginkgo, ginseng, angelica, medlar, sea backthern, raw hawthorn, kudzu root, tricholoma matsutake, auricularia polytricha and highland barley, water extract or organic solvent extract as effective components and adding white granulated sugar, maltodextrin and 90% ethanol as auxiliary materials.

5. A process for preparing a food, nutraceutical or pharmaceutical composition according to any of claims 1 to 4, characterized in that: it comprises the following steps:
(1) weighing raw material medicines;
(2) directly pulverizing the raw materials, or decocting the raw materials in water or extracting with organic solvent, concentrating the extractive solution, adding white sugar, maltodextrin, and 90% ethanol, and making into common preparation.
6. The method of claim 5, wherein: the step (2) comprises the following steps:
(1) parching semen Avenae Nudae; pulverizing radix Puerariae and cooked semen Avenae Nudae respectively, and sieving with 100 mesh sieve;
(2) mixing 8 kinds of raw materials including semen Ginkgo, Ginseng radix, radix Angelicae sinensis, fructus Lycii, fructus Hippophae, fructus crataegi, Tricholoma matsutake and Auricularia, extracting with water twice each for 30min, filtering, mixing filtrates, concentrating into extract, adding radix Puerariae powder, and drying at 50-60 deg.C; pulverizing the dried product and 50-70 parts of white granulated sugar, adding highland barley powder, 150-200 parts of maltodextrin and 90% ethanol, mixing, granulating, and drying at 50-60 deg.C.
