背景技术
随着生活节奏的加快和工作压力的加重,人们常常忽略身体状况,处于亚健康状态,长期以往,就会出现气血不足、免疫力下降、加速衰老等症状。传统的养生方法周期长,效果不明显,阿胶作为传统补药,具有良好的补充气血功效,并且现代越来越多其他养生产品和阿胶共同熬制制作成阿胶糕,方便易携带。
阿胶中85%的成分为胶原蛋白,而胶原蛋白口服后需要被蛋白酶降解为低肽或氨基酸后才能吸收,而阿胶的主要食用人群通常会伴随着脾胃虚弱症状,食用阿胶后可能会导致消化系统负担加重,甚至恶心,呕吐等,因此,在生产中就对阿胶进行进一步的降解有利于人体的吸收。
公开号为CN106387898A的中国专利申请公开了一种不上火不油腻的阿胶糕的制备方法,该发明是通过对芝麻、核桃仁蒸汽加热预处理;并且对阿胶进行烊化;混合制备阿胶糕;本发明解决传统阿胶糕口感油腻易上火的问题,提高了阿胶糕的品质,但是该阿胶糕不易消化,有效成分吸收率低。
公开号为CN106480146A的中国专利申请公开了一种低苦味阿胶低聚肽及其制备方法与应用。其制备方法包括:将阿胶酶解产生的酶解液进行柱层析,收集洗脱液得到所述阿胶低聚肽。所用洗脱剂为乙醇的水溶液;所述乙醇的水溶液的体积百分浓度具体为15-30%。该发明采用未添加任何辅料的阿胶块或阿胶汁为酶解原料,显著降低了阿胶低聚肽的苦味,提高了消费者对产品的接受度,对阿胶低聚肽产品的开发具有十分重要的意义。该发明使用的复合蛋白酶进行酶解,由于酶作用条件并不相同,导致酶解效果差。
发明内容
为了解决现有技术存在的问题,本发明提供了一种红枣阿胶糕及其制备方法,本发明不仅对红枣进行了预处理,使红枣阿胶糕口感更佳,并且对阿胶进行了酶解处理,使阿胶成分更容易吸收,补血效果更佳。
本发明具体技术方案如下所示:
一种红枣阿胶糕,主要由以下成分及其重量份数组成:红枣15-30份、阿胶 15-30份、核桃仁5-8份、黑芝麻3-5份、冰糖1-2份、黄酒10-12份、枸杞子 5-8份、黄精1-3份、山药5-7份、陈皮1-3份、人参1-3份、高麦芽糖浆1-2份和蜂蜜2-4份。
优选的,所述的红枣阿胶糕,由以下成分及其重量份数组成:红枣22份、阿胶23份、核桃仁6份、黑芝麻4份、冰糖1.5份、黄酒11份、枸杞子7份、黄精2份、山药6份、陈皮2份、人参2份、高麦芽糖浆1.5份和蜂蜜3份。
进一步的,所述的红枣为脱水干枣,处理工艺为:红枣洗净后,采用60-70℃温度通风干燥至红枣水分含量为23-25%,室温下放置20-24h使红枣表里水分分布均匀,最后切成5-12mm红枣丁,即得。
本发明还提供了所述的红枣阿胶糕的制备方法,
S1:将阿胶粉碎成60-80目阿胶粉,加入20-30倍阿胶粉质量的丙醇二酸水溶液进行混合,然后80-90℃水浴25-30min,降温至37-40℃,得到阿胶混合液,加入1mol/L的盐酸调节pH值为1.2-1.8,加入胃蛋白酶,胃蛋白酶与阿胶粉用量比为0.8×105-1.2×105U:1g,37-40℃水浴1.5-2h,得到阿胶胃酶解液,使用1mol/L的氢氧化钠调节pH值为7.5-8.5,加入胰脂肪酶、辅脂酶、胰蛋白酶、肠肽酶,与阿胶粉用量比为5000-7000U:2000-3000U: 4000-6000U:2000-3000U:1g,37-40℃水浴2.5-4h,然后80-85℃灭活15-20min,得到阿胶酶解液,加入同等体积的乙醚进行萃取,取水层,沸水浴蒸去乙醚,离心去除上清液,最后冷冻干燥,得到阿胶去油低肽粉A;
S2:将人参、陈皮、枸杞子、黄精和山药干燥后粉碎至30-50目,加入水使其没过药材,放入蒸器中95-100℃蒸制20-30min,得到蒸品B;
S3:将黑芝麻放入锅中,在锅底温度为100-110℃的条件下炒制15-20min,冷却至室温,与核桃仁混合后粉粹至1000-1200目,得到混合油C;
S4:将冰糖加入黄酒中充分溶解,然后边搅拌边加入阿胶去油低肽粉A、高麦芽糖浆和蜂蜜熬制10-15min,继续加入蒸品B、混合油C和红枣,搅拌均匀后放入模具中,自然冷却,即得。
进一步的,所述的红枣阿胶糕的制备方法,步骤S1中所述的丙醇二酸水溶液质量分数为1-2%。
进一步的,所述的红枣阿胶糕的制备方法,步骤S4中使用的红枣为脱水干枣,处理工艺为:红枣洗净后,采用60-70℃温度通风干燥至红枣水分含量为 23-25%,室温下放置20-24h,最后切成5-12mm红枣丁,即得。

红枣:红枣的医疗保健价值极高,具有养胃健脾、补血益气、和解药毒、保护肝脏、增强肌力等功效,红枣中微量元素铁的吸收利用率较高,可作为缺铁性贫血的食疗补血。红枣中的黄酮类化合物有着镇静、催眠和降压功效。红枣还具有增强肌力、抗疲劳的作用。
阿胶:氨基酸和微量元素学说认为阿胶的补血作用是因其含有人体必需氨基酸和微量元素所致。阿胶中的甘氨酸通过调节血清铁,促进血红蛋白的合成;精氨酸可使机体生长激素和睾丸酮分泌增多,促进血红蛋白的合成:苏氨酸、组氨酸、赖氨酸均具有生血作用。阿胶含有丰富的铁元素和大量的动物蛋白,有效地控制缺铁性贫血,并且服用阿胶可补充机体必需的铜元素,并能加速铁的吸收和利用,使吸收的铁与锰等微量元素协同产生生血效应。
核桃仁:核桃仁中含有丰富的蛋白质、维生素、碳水化合物及微量元素,药理研究证明,核桃仁具有清除自由基、抗衰老、健脑益智等功能。
黑芝麻:黑芝麻含有丰富的油酸、亚油酸、棕榈酸等油脂成分,长期食用具有调节血脂、促肾上腺作用,保护肝脏、抗衰老和增强免疫力的作用。
枸杞子:枸杞子主要含有枸杞多糖、甜菜碱、类胡萝卜素、氨基酸和维生素等,枸杞子中的枸杞多糖具有调节细胞免疫和体液免疫的功能,并且具有降血糖作用、降血脂,抗肿瘤和抗癌的作用,并且枸杞子还能减轻辐射对骨髓的抑制作用,可促进骨髓细胞增殖,刺激造血系统功能,有抗γ射线辐射的作用。
黄精:黄精含有黄精多糖、甾体皂苷、黄铜、氨基酸和微量元素,具有延缓衰老、改善学习能力和记忆能力、抗炎、抗病毒作用、抑菌作用、调脂、抗疲劳作用。
山药:含有脂肪酸、山药多糖、氨基酸和微量元素,具有良好的抗氧化、降血糖血脂、保护肝损伤、提高免疫力、抗肿瘤抗突变的作用。
陈皮:陈皮含有萜类化合物、黄铜、生物碱、维生素和微量元素等,具有降血脂、抗血小板凝聚、抗氧化、抗衰老、抗菌、抗肿瘤等作用,并且陈皮对消化系统具有良好的调节作用,本发明与阿胶配合使用调理脾胃,有助于阿胶吸收,增强效果。
人参:本发明人参为人工种植,主要活性成分为人参皂苷、人参多糖和微量元素等,具有调节中枢神经系统的生理功能,提高机体免疫功能,强心、抗休克,保护和刺激骨髓的造血功能,增加肾上腺皮质激素分泌,调整血糖水平,显著抑制胆固醇的吸收,抗肿瘤,延缓衰老,加强机体的适应性,其提取物能明显促进大鼠器官核酸和蛋白质的合成。
本发明公开的红枣阿胶糕含有多种中药成分,配合使用具有良好的补血、抗疲劳和增强免疫力的作用。
本发明通过安全性评价试验发现,动物实验模型豚鼠食用后没有任何过敏或是急性中毒的表现,说明本发明安全性高。
并且通过动物模型实验发现,本发明公开的红枣阿胶糕对盐酸苯肼导致的家兔贫血模型具有良好的治疗效果,食用后,对血液中的血红蛋白具有显著的提升作用,并且可以提升红细胞数、血小板数目,连续食用10天后,血液指数恢复正常水平,说明本发明具有良好的补血作用。
进一步的,通过对小鼠负重游泳实验测定,发现本发明提供的红枣阿胶糕比传统的阿胶糕抗疲劳功效更好。并且通过小鼠脾重指数试验发现,本发明提供的红枣阿胶糕具有良好的增强免疫力功效。
便溏多见于脾虚,脾胃是人体纳运食物及化生气血最重要的脏腑,阿胶、红枣与其他药物配伍具有良好的补血补气,治疗脾胃虚弱的作用,但是阿胶属于大补中药,其中含有较多的油脂和高蛋白,直接食用会加重脾胃的消化负担,造成消化不良,本发明通过对阿胶中油脂和蛋白进行仿生酶解,可以直接被肠道吸收,既减轻了消化系统的负担,又增强了脾胃功能,对脾虚导致的便溏具有良好的调理功能。
本发明提供的红枣阿胶糕中红枣为干燥后产品,口感好,并且公开的成分阿胶经过酶解反应,使营养成分更易吸收。
与现有技术相比,本发明具有以下优点:(1)安全性高,本发明中中药成分均经过炮制,去除毒性,并且通过对阿胶酶解降低消化系统负担;(2)补血功能强;(3)抗疲劳,增强免疫性。
具体实施方式
通过以下具体实施例对本发明作进一步的说明,使本领域技术人员更加理解本发明技术方案,本发明使用的成分均为常规产品,其中阿胶购自东阿阿胶股份有限公司;黄酒购自浙江古越龙山绍兴酒股份有限公司。
实施例1 一种红枣阿胶糕
所述的红枣阿胶糕,由以下成分及其重量份数组成:红枣22份、阿胶23 份、核桃仁6份、黑芝麻4份、冰糖1.5份、黄酒11份、枸杞子7份、黄精2 份、山药6份、陈皮2份、人参2份、高麦芽糖浆1.5份和蜂蜜3份;
所述的红枣为脱水干枣,处理工艺为:红枣洗净后,采用65℃温度通风干燥至红枣水分含量为24%,室温下放置22h,最后切成8mm红枣丁,即得;
所述的红枣阿胶糕的制备方法,
S1:将阿胶粉碎成70目阿胶粉,加入25倍阿胶粉质量的丙醇二酸水溶液进行混合,丙醇二酸水溶液质量分数为1.5%,然后85℃水浴28min,降温至38℃,得到阿胶混合液,加入1mol/L的盐酸调节pH值为1.5,加入胃蛋白酶,胃蛋白酶与阿胶粉用量比为1.0×105U:1g,38℃水浴1.8h,得到阿胶胃酶解液,使用 1mol/L的氢氧化钠调节pH值为8.0,加入胰脂肪酶、辅脂酶、胰蛋白酶、肠肽酶,与阿胶粉用量比为6000U:2500U:5000U:2500U:1g,38℃水浴3.2h,然后82℃灭活18min,得到阿胶酶解液,加入同等体积的乙醚进行萃取,取水层,沸水浴蒸去乙醚,离心去除上清液,最后冷冻干燥,得到阿胶去油低肽粉A;
S2:将人参、陈皮、枸杞子、黄精和山药干燥后粉碎至40目,加入水使其没过药材,放入蒸器中98℃蒸制25min,得到蒸品B;
S3:将黑芝麻放入锅中,在锅底温度为105℃的条件下炒制18min,冷却至室温,与核桃仁混合后粉粹至1100目,得到混合油C;
S4:将冰糖加入黄酒中充分溶解,然后边搅拌边加入阿胶去油低肽粉A和高麦芽糖浆熬制13min,熬制温度为125℃,继续加入蒸品B、混合油C和红枣,搅拌均匀后放入模具中,自然冷却,即得。
实施例2 一种红枣阿胶糕
所述的红枣阿胶糕,由以下成分及其重量份数组成:红枣30份、阿胶15 份、核桃仁8份、黑芝麻3份、冰糖2份、黄酒12份、枸杞子8份、黄精1份、山药7份、陈皮1份、人参3份、高麦芽糖浆1份和蜂蜜4份;
所述的红枣与实施例1处理工艺相同;
所述的红枣阿胶糕的制备方法与实施例1类似。
实施例3 一种红枣阿胶糕
所述的红枣阿胶糕,由以下成分及其重量份数组成:红枣15份、阿胶30 份、核桃仁5份、黑芝麻5份、冰糖1份、黄酒10份、枸杞子5份、黄精3份、山药5份、陈皮3份、人参1份、高麦芽糖浆2份和蜂蜜2份;
所述的红枣与实施例1处理工艺相同;
所述的红枣阿胶糕的制备方法与实施例1类似。
对比例1 一种红枣阿胶糕
所述的红枣阿胶糕,由以下成分及其重量份数组成:红枣22份、阿胶25 份、核桃仁6份、黑芝麻4份、冰糖1.5份、黄酒11份、枸杞子7份、山药6 份、黄精2份、人参2份、高麦芽糖浆1.5份和蜂蜜3份;
所述的红枣与实施例1处理工艺相同;
所述的红枣阿胶糕的制备方法与实施例1类似。
与实施例1的区别是不添加陈皮,增加阿胶的含量。
对比例2 一种红枣阿胶糕
所述的红枣阿胶糕,由以下成分及其重量份数组成:红枣22份、阿胶23 份、核桃仁6份、黑芝麻4份、冰糖1.5份、黄酒11份、枸杞子7份、黄精2 份、山药6份、陈皮2份、人参2份、高麦芽糖浆1.5份和蜂蜜3份;
所述的红枣与实施例1处理工艺相同;
所述的红枣阿胶糕的制备方法为,
S1:将阿胶粉碎成70目阿胶粉,加入25倍阿胶粉质量的丙醇二酸水溶液进行混合,丙醇二酸水溶液质量分数为1.5%,然后85℃水浴28min,降温至38℃,得到阿胶混合液,加入1mol/L的盐酸调节pH值为1.5,加入胃蛋白酶,胃蛋白酶与阿胶粉用量比为1.0×105U:1g,38℃水浴1.8h,得到阿胶胃酶解液,使用 1mol/L的氢氧化钠调节pH值为8.0,加入胰蛋白酶、肠肽酶,与阿胶粉用量比为5000U:2500U:1g,38℃水浴3.2h,然后82℃灭活18min,得到阿胶酶解液,加入阿胶酶解液2倍体积的乙醚进行萃取,取水层,沸水浴蒸去乙醚,离心去除上清液,最后冷冻干燥,得到阿胶去油低肽粉A;
S2:将人参、陈皮、枸杞子、黄精和山药干燥后粉碎至40目,加入水使其没过药材,放入蒸器中98℃蒸制25min,得到蒸品B;
S3:将黑芝麻放入锅中,在锅底温度为105℃的条件下炒制18min,冷却至室温,与核桃仁混合后粉粹至1100目,得到混合油C;
S4:将冰糖加入黄酒中充分溶解,然后边搅拌边加入阿胶去油低肽粉A、高麦芽糖浆和蜂蜜熬制13min,熬制温度为125℃,继续加入蒸品B、混合油C和红枣,搅拌均匀后放入模具中,自然冷却,即得。
与实施例1的区别是红枣阿胶糕的制备方法步骤S1中不添加胰脂肪酶、辅脂酶,增加乙醚的用量。
试验例1、安全性试验
试验对象:实施例1-3与对比例1-2制备得到的红枣阿胶糕;
动物模型:15只6周龄的豚鼠,体重350±10g;
试验方法:将豚鼠禁食(不禁水)24h后,以10g/kg(红枣阿胶糕/豚鼠体重)喂食红枣阿胶糕,连续喂养7日,2次/日;结果:豚鼠无一死亡,体重平均增长 0.8g,MTD相当人用量(9g/日)的70倍,证明该红枣阿胶糕食用健康,并且未发现豚鼠有兴奋不安、呼吸困难等过敏反应,观察一周亦未见其它过敏反应。
试验例2、补血试验
试验对象:实施例1-3与对比例2制备得到的红枣阿胶糕;
动物模型:取48只家兔(雌雄各半,2±0.3kg),用2%盐酸苯肼(分析纯,注射用水配制成2%浓度后过0.22μm微孔滤膜除菌)按15mg/kg(盐酸苯肼溶液/ 家兔体重)脊背皮下注射,每2d一次,连续注射3次,造模后,选取贫血程度接近的30只家兔作为动物模型;
试验方法:将红枣阿胶糕加同样质量的水碾碎后,以10g/kg(红枣阿胶糕/豚鼠体重)对贫血家兔进行灌胃处理,设置空白对照,连续10天,抗凝检测家兔血液中RBC,Hb,PLT,每组6只家兔,取平均值,结果如表1所示。

从表1中可以看出,家兔经盐酸苯肼注射后,血红蛋白(Hb)、红细胞数 (RBC)、血小板数(PLT)显著降低,同时食用本发明实施例1-3与对比例2 制得的红枣阿胶糕,发现本发明实施例1-3制得的红枣阿胶糕食用后,家兔血液各项指标恢复正常,而对比例2指标接近正常值,效果比实施例1-3相对较差,说明本发明公开的红枣阿胶糕具有良好的补血功能。
试验例3、抗疲劳试验
试验对象:实施例1-3与对比例2制得的红枣阿胶糕;
动物模型:选体重25-30g健康雄性昆明种小鼠30只,禁食(不禁水)24h后,以10g/kg(红枣阿胶糕/小鼠体重)喂食红枣阿胶糕,连续喂养7日,2次/日,设置空白组,正常进食;
试验方法:将小鼠置于水深30cm,水温25±0.5℃的水箱中游泳,鼠尾根部负荷5%体重的石子,记录小鼠力竭时间(s),以判断抗疲劳能力,结果如表2所示(平行试验6次,取平均值)。

从表2中可以看出,红枣阿胶糕具有良好的抗疲劳作用,其中本发明公开的的实施例1-3制备得到的红枣阿胶糕抗疲劳效果更佳。
试验例4、免疫增强试验
试验对象:实施例1-3与对比例1-2制备得到的红枣阿胶糕;
动物模型:取体重25-30g健康雄性昆明种小鼠36只,分为6组,小鼠腹腔注射氢化可的松(HC)25mg/kg,1次/d,连续7d,得到动物模型;
试验方法:将红枣阿胶糕加同样质量的水碾碎后,以10g/kg(红枣阿胶糕重量/ 小鼠体重)对小鼠进行灌胃处理,设置空白对照(正常进食),连续24天,测定小鼠脾重和脾重指数,具体如表3所示(平行试验6次,取平均值)。

从表3中可以看出,红枣阿胶糕具有良好的免疫增强作用,其中本发明公开的实施例1-3制备得到的红枣阿胶糕增强免疫作用更强。
试验例5、调理便溏作用
试验对象:实施例1-3与对比例2制备得到的红枣阿胶糕;
试验方法:选100名志愿者(脾虚导致的便溏,病症在5-10天),分5组,连续使用红枣阿胶糕3天,设置对照组(直接食用阿胶),观察结果,统计不再便溏的人数,具体见表4。

从表4可以看出,本发明制得的红枣阿胶糕具有良好的调理便溏的功效。
Claims (7)
1.一种红枣阿胶糕,其特征在于,原料及其重量份数为:红枣15-30份、阿胶15-30份、核桃仁5-8份、黑芝麻3-5份、冰糖1-2份、黄酒10-12份、枸杞子5-8份、黄精1-3 份、山药5-7份、陈皮1-3份、人参1-3份、高麦芽糖浆1-2份和蜂蜜2-4份;
所述红枣阿胶糕的制备方法,包括以下步骤:
S1:将阿胶粉碎成60-80目阿胶粉,加入20-30倍阿胶粉质量的丙醇二酸水溶液进行混合,然后80-90℃水浴25-30min,降温至37-40℃,得到阿胶混合液,加入1mol/L的盐酸调节pH值为1.2-1.8,加入胃蛋白酶,胃蛋白酶与阿胶粉用量比为0.8×105-1.2×105U:1g,37-40℃水浴1.5-2h,得到阿胶胃蛋白酶解液,使用1mol/L的氢氧化钠调节pH值为7.5-8.5,加入胰脂肪酶、辅脂酶、胰蛋白酶、肠肽酶,与阿胶粉用量比为5000-7000U:2000-3000U:4000-6000U: 2000-3000U:1g,37-40℃水浴2.5-4h,然后80-85℃灭活15-20min,得到阿胶酶解液,加入同 等体积的乙醚进行萃取,取水层,沸水浴蒸去乙醚,离心去除上清液,最后冷冻干燥,得到阿胶去油低肽粉A;
S2:将人参、陈皮、枸杞子、黄精和山药干燥后粉碎至30-50目,加入水使其没过药材,放入蒸器中95-100℃蒸制20-30min,得到蒸品B;
S3:将黑芝麻放入锅中,在锅底温度为100-110℃的条件下炒制15-20min,冷却至室温,与核桃仁混合后粉粹至1000-1200目,得到混合物C;
S4:将冰糖加入黄酒中充分溶解,然后边搅拌边加入阿胶去油低肽粉A、高麦芽糖浆和蜂蜜熬制10-15min,继续加入蒸品B、混合物C和红枣,搅拌均匀后放入模具中,自然冷却,即得。
2.根据权利要求1所述的红枣阿胶糕,其特征在于,原料及其重量份数为:红枣22份、阿胶23份、核桃仁6份、黑芝麻4份、冰糖1.5份、黄酒11份、枸杞子7份、黄精2份、山药 6份、陈皮2份、人参2份、高麦芽糖浆1.5份和蜂蜜3份。
3.根据权利要求1或2所述的红枣阿胶糕,其特征在于,所述的红枣为脱水干枣,处理工艺为:红枣洗净后,采用60-70℃温度通风干燥至红枣水分含量为23-25%,室温下放置20-24h,最后切成5-12mm红枣丁,即得。
4.根据权利要求1所述的红枣阿胶糕,其特征在于,步骤S1中所述的丙醇二酸水溶液质量分数为1-2%。
5.根据权利要求1所述的红枣阿胶糕,其特征在于,步骤S4中使用的红枣为脱水干枣,处理工艺为:红枣洗净后,采用60-70℃温度通风干燥至红枣水分含量为23- 25%,室温下放置20-24h,最后切成5-12mm红枣丁,即得。
6.根据权利要求1所述的红枣阿胶糕,其特征在于,步骤S4中熬制温度为120-130℃。
7.根据权利要求1或2所述的红枣阿胶糕在制备调理便溏保健品中的应用。
