本发明涉及一种功能性饲用甜味剂的制备方法,属于饲料添加剂技术领域。本发明培育复合菌种培养液与米糠,鱼糜,玉米粉,蔗糖,豆粕粉,植物基甜味料发酵制得甜味培养料,并喂养蝇蛆,最后筛选灭活制得功能性饲用甜味剂。本发明用产胞外多糖酵母菌发酵培养料,提高培养料糖含量,并与植物乳杆菌、乳酸菌LB1通过培养料进入虫体,改善内外肠道营养吸收率,提高免疫力,并且在虫体死亡后,将臭味物质发酵产生酯类、辛酸、十六醛以及甜味等物质改善风味,增加甜度的同时增加鲜味和营养成分,可有效刺激味觉,促进食欲,以达到增加饲料采食的效果,且可提高消化吸收能力,显著增加食欲,从而保证仔猪的健康生长,食用后仔猪皮红毛亮,提高免疫力。

目前饲料甜味剂的主要的甜源是采用糖精及其盐,同时辅以其他的成分对糖精及其盐的口感进行修饰,常用的有甜蜜素、阿斯巴甜、三氯蔗糖、葡萄糖等,但糖精及其盐本身具有金属苦味,会影响饲粮的适口性,因此,对于饲料甜味剂的改善,使其具有以下特点:1、无毒性和致癌作用,无不良余味,不会对动物的生长发育产生损害作用,不能在动物体内积累,从而间接危害人类健康;2、产生甜味快,有与蔗糖一样的甜味特征,有足够的甜度;3、稳定性好。在生产、储存与使用中甜味剂本身不挥发损失,不影响饲料中其他营养成分;4、无生理活性,是水溶性的,被动物采食后,能溶于口腔的唾液中被味蕾感知;5、成本低廉,性价比优越。
发明内容
本发明所要解决的技术问题:针对目前饲料甜味剂的主要的甜源是采用糖精及其盐,同时辅以其他的成分对糖精及其盐的口感进行修饰,但糖精及其盐本身具有金属苦味,会影响饲粮的适口性的问题,本发明提供了一种功能性饲用甜味剂的制备方法。
为解决上述技术问题,本发明采用的技术方案是:
一种功能性饲用甜味剂的制备方法,其特征在于,具体制备步骤为:
(1)将罗汉果与新鲜甜菊叶水洗打浆后混合加入质量分数为70%乙醇溶液中,在70~75℃恒温水浴下,用60W超声波超声辅助提取50~60min,离心分离后收集上清液,并将沉淀重复浸提1~2次,合并上清液,并在60~65℃下,减压蒸发至干后装入研钵中研磨,得植物基甜味料;
(2)取葡萄糖、豆粕粉、硫酸镁、氯化钠加入去离子水中混合均匀,用氢氧化钠溶液调节pH至中性,再转入灭菌锅中,在110~120℃下灭菌12~20min,得液体培养基;
(3)将产胞外多糖酵母菌、植物乳杆菌、乳酸菌LB1,接种至液体培养基中进行菌种培养,在35~37℃下,以150~200r/min振荡培养30~36h,过滤得复合益生菌液;
(4)取复合益生菌液与去离子水混合,再加入米糠、鱼糜、玉米粉、蔗糖、豆粕粉、植物基甜味料,搅拌均匀后装入发酵罐中,发酵2~3天,得甜味培养料;
(5)用甜味培养料喂养家蝇幼虫1~2月后筛选12~15目幼虫,将筛选出的幼虫置于洁净的饲养盒中饥饿处理15~20h,再用清水淋洗15~20min后用质量分数2%氯化钠溶液浸泡1~2h,取出后用去离子水洗涤冷冻干燥,得功能性饲用甜味剂。
所述罗汉果浆液、甜菊叶浆液与乙醇溶液的用量为500~600重量份罗汉果浆液,200~300重量份甜菊叶浆液加入2000~3000重量份乙醇溶液。
所述葡萄糖、豆粕粉、硫酸镁、氯化钠的用量为15~20重量份葡萄糖,12~15重量份豆粕粉,0.20~0.25重量份硫酸镁,5~6重量份氯化钠。
所述产胞外多糖酵母菌、植物乳杆菌、乳酸菌LB1的接种量为3~5%。
所述复合益生菌液、米糠、鱼糜、玉米粉、蔗糖、豆粕粉、植物基甜味料的重量为0.010~0.02份复合益生菌液,70~75份米糠,3~4份鱼糜,13~15份玉米粉,3~4份蔗糖,5~8份豆粕粉,0.010~0.015份植物基甜味料。
本发明的有益技术效果是:
本发明通过产胞外多糖酵母菌、植物乳杆菌、乳酸菌LB1,培育复合菌种培养液,并将其与米糠,鱼糜,玉米粉,蔗糖,豆粕粉,植物基甜味料发酵制得甜味培养料,并喂养蝇蛆,最后筛选灭活制得功能性饲用甜味剂。本发明用产胞外多糖酵母菌发酵培养料,提高培养料糖含量,并与植物乳杆菌、乳酸菌LB1通过培养料进入虫体,改善内外肠道营养吸收率,提高免疫力,并且在虫体死亡后,将臭味物质发酵产生酯类、辛酸、十六醛以及甜味等物质改善风味,增加甜度的同时增加鲜味和营养成分,可有效刺激味觉,促进食欲,以达到增加饲料采食的效果,且可提高消化吸收能力,显著增加食欲,从而保证仔猪的健康生长,食用后仔猪皮红毛亮,提高免疫力。
具体实施方式
取罗汉果用水洗净表皮后剖开去核,再加入匀浆机中破碎打浆10~15min,得罗汉果浆液,取新鲜甜菊叶用水洗净后加入匀浆机中破碎打浆10~12min,得甜菊叶浆液,取500~600g罗汉果浆液,200~300g甜菊叶浆液加入2~3L质量分数为70%乙醇溶液中,在70~75℃恒温水浴下,并用60W超声波超声辅助提取50~60min,再装入离心机中,以4000~6000/min离心分离10~15min,收集上清液,并将沉淀重复浸提1~2次,合并上清液,将合并后的上清液装入旋转蒸发仪中,在60~65℃下,减压蒸发至干后装入研钵中研磨,得植物基甜味料,取15~20g葡萄糖,12~15g豆粕粉,0.20~0.25g硫酸镁,5~6g氯化钠,加入0.8~1.0L去离子水中,以300~400r/min搅拌混合10~15min后,用质量分数为5%氢氧化钠溶液调节pH至中性,随后转入灭菌锅中,在110~120℃下灭菌12~20min,得液体培养基,按接种量3~5%,将产胞外多糖酵母菌、植物乳杆菌、乳酸菌LB1,接种至液体培养基中进行菌种培养,在35~37℃下厌氧培养15~20h,过滤得复合益生菌液,取10~20g益生菌液与10~15L去离子水混合,再加入70~75kg米糠,3~4kg鱼糜,13~15kg玉米粉,3~4kg蔗糖,5~8kg豆粕粉,10~15g植物基甜味料,搅拌均匀后装入发酵罐中,发酵2~3天,得甜味培养料,用甜味培养料喂养家蝇幼虫1~2月,收集10~15目幼虫,将筛选出的幼虫置于洁净的饲养盒中饥饿处理15~20h,并用清水淋洗15~20min,再用质量分数2%氯化钠溶液浸泡1~2h,取出后用去离子水洗涤1~3次,转入冷冻干燥箱中,在﹣50~﹣35℃下干燥3~6h,得功能性饲用甜味剂。
实例1

实例2
取罗汉果用水洗净表皮后剖开去核,再加入匀浆机中破碎打浆12min,得罗汉果浆液,取新鲜甜菊叶用水洗净后加入匀浆机中破碎打浆11min,得甜菊叶浆液,取550g罗汉果浆液,250g甜菊叶浆液加入2L质量分数为70%乙醇溶液中,在72℃恒温水浴下,并用60W超声波超声辅助提取55min,再装入离心机中,以5000/min离心分离12min,收集上清液,并将沉淀重复浸提1次,合并上清液,将合并后的上清液装入旋转蒸发仪中,在62℃下,减压蒸发至干后装入研钵中研磨,得植物基甜味料,取18g葡萄糖,13g豆粕粉,0.24g硫酸镁,5g氯化钠,加入0.9L去离子水中,以350r/min搅拌混合12min后,用质量分数为5%氢氧化钠溶液调节pH至中性,随后转入灭菌锅中,在115℃下灭菌15min,得液体培养基,按接种量4%,将产胞外多糖酵母菌、植物乳杆菌、乳酸菌LB1,接种至液体培养基中进行菌种培养,在36℃下厌氧培养18h,过滤得复合益生菌液,取15g益生菌液与12L去离子水混合,再加入72kg米糠,3kg鱼糜,14kg玉米粉,3kg蔗糖,6kg豆粕粉,12g植物基甜味料,搅拌均匀后装入发酵罐中,发酵2天,得甜味培养料,用甜味培养料喂养家蝇幼虫1月,收集12目幼虫,将筛选出的幼虫置于洁净的饲养盒中饥饿处理18h,并用清水淋洗18min,再用质量分数2%氯化钠溶液浸泡1h,取出后用去离子水洗涤2次,转入冷冻干燥箱中,在﹣40℃下干燥4h,得功能性饲用甜味剂。
实例3
取罗汉果用水洗净表皮后剖开去核,再加入匀浆机中破碎打浆15min,得罗汉果浆液,取新鲜甜菊叶用水洗净后加入匀浆机中破碎打浆12min,得甜菊叶浆液,取600g罗汉果浆液,300g甜菊叶浆液加入3L质量分数为70%乙醇溶液中,在75℃恒温水浴下,并用60W超声波超声辅助提取60min,再装入离心机中,以6000/min离心分离15min,收集上清液,并将沉淀重复浸提2次,合并上清液,将合并后的上清液装入旋转蒸发仪中,在65℃下,减压蒸发至干后装入研钵中研磨,得植物基甜味料,取20g葡萄糖,15g豆粕粉,0.25g硫酸镁,6g氯化钠,加入1.0L去离子水中,以400r/min搅拌混合15min后,用质量分数为5%氢氧化钠溶液调节pH至中性,随后转入灭菌锅中,在120℃下灭菌20min,得液体培养基,按接种量5%,将产胞外多糖酵母菌、植物乳杆菌、乳酸菌LB1,接种至液体培养基中进行菌种培养,在37℃下厌氧培养20h,过滤得复合益生菌液,取20g益生菌液与15L去离子水混合,再加入75kg米糠,4kg鱼糜,15kg玉米粉,4kg蔗糖,8kg豆粕粉,15g植物基甜味料,搅拌均匀后装入发酵罐中,发酵3天,得甜味培养料,用甜味培养料喂养家蝇幼虫2月,收集15目幼虫,将筛选出的幼虫置于洁净的饲养盒中饥饿处理20h,并用清水淋洗20min,再用质量分数2%氯化钠溶液浸泡2h,取出后用去离子水洗涤3次,转入冷冻干燥箱中,在﹣50℃下干燥6h,得功能性饲用甜味剂。
对比例:玉米-豆粕型基础饲粮。
用添加本发明制备的功能性饲用甜味剂的玉米-豆粕型基础饲粮与对比例进行断奶仔猪饲养,猪舍温度控制在(25±3)℃,并保持良好的通风。试验管理按照 常规管理规程及正常免疫程序进行饲养。试验期间断奶仔猪自由饮水、自由采食。
将混合饲料每天分2~4次进行投喂,并且要保证每天都有1h的空槽时间即可。
饲养结果如表1所示:

由上表可知,本发明制备的功能性饲用甜味剂,可有效刺激味觉,促进食欲,以达到增加饲料采食的效果,且可促进消化吸收,提高日增重量。
Claims (5)
1.一种功能性饲用甜味剂的制备方法,其特征在于,具体制备步骤为:
(1)将罗汉果与新鲜甜菊叶水洗打浆后混合加入质量分数为70%乙醇溶液中,在70~75℃恒温水浴下,用60W超声波超声辅助提取50~60min,离心分离后收集上清液,并将沉淀重复浸提1~2次,合并上清液,并在60~65℃下,减压蒸发至干后装入研钵中研磨,得植物基甜味料;
(2)取葡萄糖、豆粕粉、硫酸镁、氯化钠加入去离子水中混合均匀,用氢氧化钠溶液调节pH至中性,再转入灭菌锅中,在110~120℃下灭菌12~20min,得液体培养基;
(3)将产胞外多糖酵母菌、植物乳杆菌、乳酸菌LB1,接种至液体培养基中进行菌种培养,在35~37℃下,以150~200r/min振荡培养30~36h,过滤得复合益生菌液;
(4)取复合益生菌液与去离子水混合,再加入米糠、鱼糜、玉米粉、蔗糖、豆粕粉、植物基甜味料,搅拌均匀后装入发酵罐中,发酵2~3天,得甜味培养料;
(5)用甜味培养料喂养家蝇幼虫1~2月后筛选12~15目幼虫,将筛选出的幼虫置于洁净的饲养盒中饥饿处理15~20h,再用清水淋洗15~20min后用质量分数2%氯化钠溶液浸泡1~2h,取出后用去离子水洗涤冷冻干燥,得功能性饲用甜味剂。
2.如权利要求1所述的一种功能性饲用甜味剂的制备方法,其特征在于,所述罗汉果浆液、甜菊叶浆液与乙醇溶液的用量为500~600重量份罗汉果浆液,200~300重量份甜菊叶浆液加入2000~3000重量份乙醇溶液。
3.如权利要求1所述的一种功能性饲用甜味剂的制备方法,其特征在于,所述葡萄糖、豆粕粉、硫酸镁、氯化钠的用量为15~20重量份葡萄糖,12~15重量份豆粕粉,0.20~0.25重量份硫酸镁,5~6重量份氯化钠。
4.如权利要求1所述的一种功能性饲用甜味剂的制备方法,其特征在于,所述产胞外多糖酵母菌、植物乳杆菌、乳酸菌LB1的接种量为3~5%。
5.如权利要求1所述的一种功能性饲用甜味剂的制备方法,其特征在于,所述复合益生菌液、米糠、鱼糜、玉米粉、蔗糖、豆粕粉、植物基甜味料的重量为0.010~0.02份复合益生菌液,70~75份米糠,3~4份鱼糜,13~15份玉米粉,3~4份蔗糖,5~8份豆粕粉,0.010~0.015份植物基甜味料。
