Abstract
本发明公开了一种食用菌全活性物质的制备与应用,其以食用菌为原料,将其粉碎后采用极性从小到大的乙醇溶液及水进行分步提取,再经复合蛋白酶酶解,分别得到食用菌85%‑95%醇提物、食用菌55%‑65%醇提物、食用菌水提取物及酶解物,即获得食用菌全活性物质。经药效学实验验证,所得全活性物质能够显著的降低血清中TC)、甘油三脂(TG)以及低密度脂蛋白胆固醇(LDL‑C)的水平。根据中医“君臣佐使”理论以及现代营养学理论,可将所得食用菌全活性物质中的一种或几种与其他具有相同功效的药食同源活性成分复配,以制备具有辅助增强免疫力、调节血糖、血脂、血压及保肝等功效的营养功能性食品。

技术领域
本发明属于天然活性成分分离技术领域,具体涉及一种食用菌全活性物质的制备与应用。
背景技术
随着生活水平的提高和生活节奏的改变,被称为“富贵病”的“三高症”发病率越来越高,尤其高发于50岁以上的中老年人。高血脂可以引起血管栓塞;高血压可以引起脑出血和脑血管破裂;高血糖可以引起糖尿病。全世界每年死于心脑血管疾病的人数高达1500万人,居各种死因首位。但目前临床上常用的口服降血糖血脂药均有副作用,可引起消化系统的不良反应,有些还会引起麻疹、贫血、白细胞和血小板减少、头晕和乏力等。因此寻找开发具有增强机体免疫力作用的保健食品,以辅助药物治疗,在有效的控制血糖和血脂的同时降低副作用已引起人们的关注。
食用菌是指子实体硕大、可供食用的蕈菌(大型真菌),通称为蘑菇。中国已知的食用菌有350多种,其中多属担子菌亚门,常见的有香菇、草菇、蘑菇、木耳、银耳、猴头、竹荪、松口蘑(松茸)、口蘑、红菇、灵芝、虫草、松露、百灵和牛肝菌等,是一类有机、营养、保健的绿色食品。食用菌产业是一项集食用菌经济效益、生态效益和社会效益于一体的短平快农村经济发展项目。发展食用菌产业符合人们消费增长和农业可持续发展的需要,是农民快速致富的有效途径,市场潜力巨大。
食用菌药用价值在国内外近年来不断地更新,其含有的生物活性物质如高分于多糖、β-葡萄糖、RNA复合体、天然有机锗、核酸降解物、cAMP和三萜类化合物等对维护人体健康有重要的利用价值。已知食用菌的药用保健价值有:①抗癌作用:食用菌的多糖体能刺激抗体的形成,提高并调整机体内部的防御能力,降低某些物质诱发肿瘤的发生率,并对多种化疗药物有增效作用,此外,栗蘑中富含有机硒,可作补硒食品,若长期食用,几乎可以防止一切癌变;②抗菌、抗病毒作用;③降血压、降血脂、抗血栓、抗心律失常、强心等;④健胃、助消化作用;⑤止咳平喘、祛痰作用;⑥利胆、保肝、解毒;⑦降血糖;⑧通便利尿;⑨免疫调节。
虽然食用菌有显著并且明确的降血脂降血压,以及抗癌、保肝健胃等功效,但目前在高效制备食用菌活性成分的技术上,往往存在食用菌活性物质制备不完全、制作工艺繁琐、利用率低等问题。针对这些问题,本发明提供了一种采用顺序溶剂(极性从小到大)提取制备食用菌活性物质的方法,而目前尚未见此类报道,且此方法对促进食用菌大规模工业化生产具有良好的开发前景。
发明内容
针对现有技术的不足,本发明提供了提供一种食用菌全活性物质的制备与应用,其可解决目前存在的活性物质制备不完全、制作工艺繁琐、利用率低等问题,对促进食用菌产品大规模工业化生产具有良好的应用前景。
为实现上述目的,本发明采用如下技术方案:
一种食用菌全活性物质的制备方法,其是以食用菌为原料,将其粉碎后采用极性从小到大的乙醇溶液及水进行分步提取,再经复合蛋白酶酶解,分别得到食用菌85%-95%醇提物、食用菌55%-65%醇提物、食用菌水提取物及酶解物,即获得食用菌全活性物质;所述食用菌为猴头菇、香菇、竹荪中一种或几种。
所述食用菌全活性物质的制备方法包括如下步骤:
1)将食用菌子实体用粉碎机磨成细粉备用;
2)将制得的食用菌细粉用85wt%-95wt%的乙醇溶液为溶剂,在45-70℃条件下超声-微波协同萃取或超声辅助提取0.5-3h,过滤,获得食用菌85%-95%醇提液;
3)将步骤2)在剩余的滤渣用55wt%-65wt%的乙醇溶液为溶剂,在45-70℃条件下超声-微波协同萃取或超声辅助提取0.5-3h,过滤,获得食用菌55%-65%醇提液;
4)再将步骤3)剩余的滤渣用去离子水为溶剂,在70-100℃条件下超声-微波协同萃取或超声辅助提取0.5-3h,过滤,获得食用菌水提液;
5)将步骤4)水提剩余物在37-65℃条件下,用食用菌细粉重量0.2-2%的复合蛋白酶酶解0.5-3h,然后过滤,获得食用菌酶解物;
6)将步骤2)~5)所得产物经减压浓缩、干燥后,即分别制得食用菌85%-95%醇提物、食用菌55%-65%醇提物、食用菌水提取物以及酶解物。
步骤2)~4)的提取过程中溶剂与物料的比例为1:10-1:30 kg/L。
根据中医的“君臣佐使”理论以及现代营养学理论,可将本发明所得食用菌全活性物质中的一种或几种与其他具有相同功效的药食同源活性成分复配,以制备具有辅助增强免疫力、调节血糖、血脂、血压及保肝等功效的口服液、胶囊、片剂、冲剂、固体饮料等不同剂型的营养功能性食品。
本发明的有益之处在于:
(1)本发明用于从小到大不同极性的溶剂对食用菌细粉进行分步提取,再经过滤、减压浓缩、干燥分别制备食用菌85%-95%醇提物、食用菌55%-65%醇提物、食用菌水提物以及酶解物,可实现食用菌活性物质的全面提取,提高资源利用率。
(2)根据中医“君臣佐使”以及现代营养学理论,将本发明所得食用菌全活性物质中的一种或几种与其他具有相同功效的药食同源活性成分复配,制备的功能性食品长期食用可起到具有辅助增强免疫力、调节血糖、血脂、血压、保肝等功效。
具体实施方式
为了使本发明所述的内容更加便于理解,下面结合具体实施方式对本发明所述的技术方案做进一步的说明,但是本发明不仅限于此。
实施例1 一种猴头菇全活性物质胶囊
1)将猴头菇子实体用粉碎机磨成细粉备用;
2)按料液比1:30 kg/L将制得的猴头菇细粉用95wt%的乙醇溶液为溶剂,在70℃条件下超声-微波协同萃取0.5h,过滤,获得猴头菇95%醇提液;
3)按料液比1:30 kg/L将步骤2)剩余的滤渣用55wt%的乙醇溶液为溶剂,在70℃条件下超声-微波协同萃取0.5h,过滤,获得猴头菇55%醇提液;
4)按料液比1:30 kg/L将步骤3)剩余的滤渣用去离子水为溶剂,在100℃条件下超声-微波协同萃取0.5h,过滤,获得猴头菇水提液;
5)将步骤4)水提剩余物在37℃条件下,用猴头菇细粉重量2%的复合蛋白酶酶解0.5h,然后过滤,获得猴头菇酶解物;
6)将步骤2)~5)所得产物经减压浓缩、干燥后,即分别制得猴头菇95%醇提物、猴头菇55%醇提物、猴头菇水提取物以及酶解物,混合均匀,在70℃干燥4h,得到胶囊填充物;
7)将所得胶囊填充物经软胶囊压丸机进行制备,制得重量为2mg/粒的胶囊,塑铝包装即可。
实施例2 一种竹荪全活性物质胶囊
1)将竹荪子实体用粉碎机磨成细粉备用;
2)按料液比1:10 kg/L将制得的竹荪细粉用85wt%的乙醇溶液为溶剂,在45℃条件下超声辅助提取3h,过滤,获得竹荪85%醇提液;
3)按料液比1:10 kg/L将步骤2)在剩余的滤渣用65wt%的乙醇溶液为溶剂,在45℃条件下超声辅助提取3h,过滤,获得竹荪65%醇提液;
4)按料液比1:10 kg/L将步骤3)剩余的滤渣用去离子水为溶剂,在70℃条件下超声辅助提取3h,过滤,获得竹荪水提液;
5)将步骤4)水提剩余物在65℃条件下,用竹荪细粉重量0.2%的复合蛋白酶酶解3h,然后过滤,获得竹荪酶解物;
6)将步骤2)~5)所得产物经减压浓缩、干燥后,即分别制得竹荪85%醇提物、竹荪65%醇提物、竹荪水提取物以及酶解物,混合均匀,在70℃干燥4h,得到胶囊填充物;。
7)将所得胶囊填充物经软胶囊压丸机进行制备,制得重量为2mg/粒的胶囊,塑铝包装即可。

1.一种食用菌全活性物质的制备,其特征在于:以食用菌为原料,将其粉碎后采用极性从小到大的乙醇溶液及水进行分步提取,再经复合蛋白酶酶解,分别得到食用菌85%-95%醇提物、食用菌55%-65%醇提物、食用菌水提取物及酶解物,即获得食用菌全活性物质;
其中所述食用菌为猴头菇、香菇、竹荪中一种或几种。
2.根据权利要求书1所述食用菌全活性物质的制备,其特征在于:包括如下步骤:
1)将食用菌子实体用粉碎机磨成细粉备用;
2)将制得的食用菌细粉用85wt%-95wt%的乙醇溶液为溶剂,在45-70℃条件下超声-微波协同萃取或超声辅助提取0.5-3h,过滤,获得食用菌85%-95%醇提液;
3)将步骤2)剩余的滤渣用55wt%-65wt%的乙醇溶液为溶剂,在45-70℃条件下超声-微波协同萃取或超声辅助提取0.5-3h,过滤,获得食用菌55%-65%醇提液;
4)再将步骤3)剩余的滤渣用去离子水为溶剂,在70-100℃条件下超声-微波协同萃取或超声辅助提取0.5-3h,过滤,获得食用菌水提液;
5)将步骤4)水提剩余物在37-65℃条件下用复合蛋白酶酶解0.5-3h,然后过滤,获得食用菌酶解物;
6)将步骤2)~5)所得产物经减压浓缩、干燥后,即分别制得食用菌85%-95%醇提物、食用菌55%-65%醇提物、食用菌水提取物以及酶解物。
3.根据权利要求书2所述食用菌全活性物质的制备,其特征在于:提取过程中溶剂与物料的比例为1:10-1:30 kg/L。
4.根据权利要求书2所述食用菌全活性物质的制备,其特征在于:步骤5)中复合蛋白酶的添加量为食用菌细粉重量的0.2-2%。
5.一种如权利要求书1所述方法制备的食用菌全活性物质的应用,其特征在于:将所得食用菌全活性物质中的一种或几种与具有相同功效的药食同源活性成分复配,以制备包括口服液、胶囊、片剂、冲剂及固体饮料中任意一种形式的营养、功能性食品。
