本发明涉及新的替诺福韦盐。
背景技术
替诺福韦富马酸盐(TDF,tenofovir disoproxil fumarate,或称为富马酸替诺福韦酯)是化学名为9-[2-(R)-[[双[[(异丙氧羰基)氧基]甲氧基]磷酰基]甲氧基]丙基]腺嘌呤富马酸盐(1:1)的化合物,是在吸收时待水解的前药,并且是可用于治疗HIV-1感染和慢性乙型肝炎的膦酰基甲氧基核苷酸类似物。
TDF被批准作为用于治疗AIDS的治疗剂,然后被美国食品和药物管理局批准作为用于治疗乙型肝炎的治疗剂。替诺福韦是TDF的活性代谢物,在对拉夫米定(Zeffix)有耐受性的患者中具有高的抑制乙型肝炎病毒DNA的能力,并且是属于如由美国食品和药物管理局(FDA)分类的妊娠B类(这是在动物研究中被发现没有胎儿风险的药物)的抗病毒剂,用于评估由药物造成的胎儿损伤的风险。
然而,已知确保替诺福韦的物理化学稳定性是困难的。如在已公开的文献(Pharmaceutical Research,2001,18,234-237;Pharmaceutical Research,2000,17,1098-1103)中所报导的,替诺福韦在水的存在下水解得到甲醛,然后经过与替诺福韦的N6-胺基进行缩合反应,从而产生作为杂质的替诺福韦二聚体。
通常,为了预期药物的一致效应,不仅在药物制造后必须防止活性成分的量的分解,而且在其储存期间要防止活性成分的量的分解,并且此外,杂质或在相同时期内作为活性成分降解产物的相关物质的量的增加必须被抑制。因此,防止杂质掺入药物被认为在药物的质量控制方面非常重要。
对于各国关于纯度测试的官方汇编规定,韩国药典在纯度试验部分对相关物质有单独的规定以及美国药典对“普通杂质”有规定,其中将相关物质的总量设定为2.0%以下,或者其中在药品的各文章中根据各药物的相关化合物和色谱纯度调节相关物质的量。此外,欧洲和英国药典规定了相关物质,并且日本药典在纯度测试中规定了相关物质及其含量。
因此,本发明人已经研究了新的盐,其即使在长期储存时能够使相关物质的产生最小化并使溶解度最大化,同时表现出等于或优于常规替诺福韦富马酸盐的物理化学性质,引领了使用乙二磺酸盐的新的替诺福韦乙二磺酸盐的开发。

因此,本发明旨在提供一种新的替诺福韦盐化合物,其具有与替诺福韦富马酸盐相同或更好的物理化学性质,可以即使在长期储存时也能使相关物质的产生最小化,并且可以使溶解度最大化。

有益效果
根据本发明,与替诺福韦富马酸盐相比,替诺福韦乙二磺酸盐可以使相关物质随时间的产生最小化,由此在含有本发明的盐化合物的产品的储存期间可显著降低杂质的产生,从而提高药物的稳定性并避免了对毒性作用进行另外研究的需要。尽管pH、耐湿性和溶解性发生变化,本发明的盐化合物的稳定性可极大地改善,并且可以表现出优异的物理化学性质,并且因此可以用作用于治疗HIV-1感染和慢性乙型肝炎的药物组合物的活性成分,与药学上可接受的载体一起构成药物组合物。此外,与其他替诺福韦盐(例如乳清酸盐、天冬氨酸盐、马尿酸盐等)相比,本发明的盐化合物甚至在pH变化时也具有非常高的溶解度。
此外,即使当使用简单的方法制备本发明的替诺福韦乙二磺酸盐时,在酸加成盐的合成期间可以获得更纯的替诺福韦。

通常,本发明的药物组合物可以口服、直肠内、阴道内、鼻内、局部(例如眼内、口内和舌下)或肠胃外(例如皮下、肌内、静脉内、皮内、脊柱内或硬膜外)施用。口服给药是优选的。
本发明的药物组合物可优选以片剂、胶囊剂、粉剂、颗粒剂、滴丸剂、散剂、丸剂、酊剂或糊剂的剂型提供。片剂优选以典型片、包衣片、分散片、泡腾片、缓释片、控释片或肠溶衣片的形式提供。胶囊剂优选以典型胶囊、持续释放胶囊、控释胶囊或肠溶衣胶囊的形式提供。
当本发明的药物组合物是片剂或胶囊剂时,单位剂型中作为活性成分的替诺福韦乙二磺酸盐的量可以根据多种因素而变化,如症状和年龄。通常,在口服给药时,单剂量的活性成分的剂量范围为5mg至300mg,并且优选5mg至150mg。此外,片剂或胶囊还可包含填充剂(如淀粉、蔗糖、乳糖等),粘合剂(如水、醇、聚乙烯吡咯烷酮、预胶化淀粉等),崩解剂(如微晶纤维素、交联羧甲基纤维素钠、交联聚乙烯吡咯烷酮等),和润滑剂(如硬脂酸镁、滑石粉、二氧化硅等)。
当本发明的药物组合物是片剂或胶囊剂时,其优选包含碱性药物载体,包括碱式碳酸盐和碱式氢氧化物。碱式碳酸盐的优选实例是碳酸钙、碳酸镁、碳酸锌、碳酸亚铁或碳酸铝。碱式氢氧化物的优选实例是氢氧化镁、氢氧化钙、氢氧化铝或氢氧化亚铁。
当本发明的药物组合物是分散片时,崩解剂选择性地以约0.5至60%的量存在,以实现快速崩解。
本发明的药物组合物的剂型可以是用于注射的无菌粉末或注射溶液。剂型可以通过注射施用。
本发明的另一方面涉及预防或治疗哺乳动物中的病毒感染的方法,包括向需要其的哺乳动物施用治疗有效量的替诺福韦乙二磺酸盐。
本发明的又一方面涉及替诺福韦乙二磺酸盐在制备用于预防或治疗与病毒感染相关的疾病的药物中的用途。
在本发明中,病毒包括HIV、HBV、CMV、HSV-1、HSV-2和人类疱疹病毒,并且优选包括HIV或HBV。本发明的替诺福韦乙二磺酸盐特别适用于治疗HIV-1感染和慢性乙型肝炎。

此外,本发明涉及一种制备替诺福韦乙二磺酸盐的方法,包括使替诺福韦与乙二磺酸盐二水合物反应。替诺福韦和乙二磺酸盐二水合物优选以1:0.5-1.5的摩尔比反应。更优选地,制备替诺福韦乙二磺酸盐的方法包括使替诺福韦与乙二磺酸盐二水合物以1:0.5-0.6的摩尔比反应。
发明方式
通过以下实施例可以获得对本发明的更好理解,所述实施例用于说明,但不应解释为限制本发明的范围,并且可以由本领域技术人员在本发明的范围内适当地修改和改变。
<实施例1>替诺福韦半乙二磺酸盐的制备
将200mL无水乙醇、10.0g替诺福韦和1.98g 1,1-乙二砜二水合物(1,1-ethanedisulfone dihydrate)置于500mL反应器中,在20至30℃下搅拌1小时,冷却至0至5℃,搅拌0.5小时,过滤,用20mL无水乙醇洗涤,并在40℃下减压干燥4小时,从而得到11.2g白色替诺福韦半乙二磺酸盐(纯度为99.68%)。

<对比例1>替诺福韦富马酸盐的制备
替诺福韦富马酸盐是从中国购买的制备好的API。
<对比例2>替诺福韦单乙二磺酸盐的制备
按照如下制备替诺福韦单乙二磺酸盐。
在100mL反应器中,放入30mL的去离子水、3.0g的替诺福韦和1.2g(1.1当量)的1,1-乙二砜二水合物,在20至30℃下搅拌1小时。反应终止后,浓缩反应产物以除去去离子水,加入30mL甲苯,再次浓缩,从而除去剩余的去离子水。
将浓缩的残留物在室温下溶解在6mL甲醇中,与6mL异丙醚一起搅拌30分钟,结晶,过滤并在40℃下减压干燥4小时,从而得到3.4g白色替诺福韦单乙二磺酸盐(纯度97.51%)。
图1b显示了根据本发明的替诺福韦半乙二磺酸盐和替诺福韦单乙二磺酸盐的结构。
<试验例1>依赖于相关物质的产生的稳定性(压力条件)的评价
(1)与替诺福韦富马酸盐的比较
为了评价稳定性(压力条件),根据ICH指南,使用本发明的替诺福韦乙二磺酸盐(实施例1:TDE)和作为对照组的替诺福韦富马酸盐(对比例1:TDF),并如美国药典(USP)中所述使用高效液相色谱(HPLC)进行分析。结果示于下表1中(压力条件:60±2℃)。

从表1可以看出,与替诺福韦富马酸盐(disoproxil fumarate)相比,本发明的替诺福韦半乙二磺酸盐在压力条件下非常稳定。特别地,由替诺福韦富马酸盐产生的相关物质极少在本发明的替诺福韦半乙二磺酸盐中产生,从中可以推断本发明的替诺福韦半乙二磺酸盐保持高纯度。在初始阶段出现的相关物质中,仅替诺福韦单酯的量增加,而其它相关物质很少增加。此外,替诺福韦半乙二磺酸盐含有主要降解产物,即替诺福韦单酯,含量为0.37%,因此显著提高了稳定性。
(2)替诺福韦单乙二磺酸盐
以与上述(1)相同的方式,评价作为测试组的本发明的替诺福韦半乙二磺酸盐(实施例1:TDE(半盐)),和作为对照组的替诺福韦单乙二磺酸盐(对比例2:TDE(单盐))的稳定性3周。结果如图1c所示。
如图1c所示,与替诺福韦单乙二磺酸盐相比,本发明的替诺福韦半乙二磺酸盐在压力条件下非常稳定。特别地,本发明的替诺福韦半乙二磺酸盐随时间的变化几乎不产生相关物质,因此保持了其高纯度,并且在第3周产生非常少量的0.37的相关物质,从而表现出比替诺福韦单乙二磺酸盐高至少15倍的稳定性,其中在相同时期观察到5.62的相关物质。
因此,可以确认本发明的替诺福韦半乙二磺酸盐是能够使相关物质的产生最小化从而能够在有利于相关物质的量增加相关的条件下进行储存的一种药物,并且还使得能够长期储存。
<试验例2>溶解性的评价
溶解度在制造药物方面被认为是重要的。在具有高功效的药物具有低溶解度的情况下,产品的研发可能变得困难。如果药物具有低溶解度,其可能沉淀并且因此口服吸收可能显著降低。
评价本发明的替诺福韦半乙二磺酸盐(实施例1)和作为对照组的替诺福韦富马酸盐(对比例1)的饱和溶解度。结果示于下表2中。

<试验例3>依赖于pH变化的稳定性的评价
当药物口服施用时,其通过胃、十二指肠、小肠等在体内吸收。因此,依赖于pH变化的稳定性对药物的释放速率具有很大影响。在本发明中,使用去离子水和三种不同的pH缓冲溶液(pH 1.2、4.0和6.8)测量稳定性用于释放测试。以如下方式进行试验方法,将本发明的替诺福韦半乙二磺酸盐和作为对照组的替诺福韦富马酸盐各5mg精确称量,完全溶解在1mL乙腈和水(1:19)的混合物中,然后分别加入9mL的上述pH缓冲溶液。将如此制备的各个样品置于30℃的小室中,并使用HPLC对其随时间的量的变化进行定量。
图2和图3示出了依赖于pH变化的替诺福韦半乙二磺酸盐(实施例1:TDE)和替诺福韦富马酸盐(对比例1:TDF)的稳定性的结果。如图2和3所示,本发明的替诺福韦半乙二磺酸盐显示出与常规替诺福韦富马酸盐产物相同的稳定性。
<试验例4>PXRD(粉末X射线衍射,X射线衍射分析)
对于X射线粉末衍射,当材料的晶体结构和化合物形式不同时,衍射图案变化。将其结果与标准材料的结果进行比较,从而确认材料的晶体结构。使用Rigaku MiniFlex 600分析本发明的替诺福韦半乙二磺酸盐和对照组,即替诺福韦富马酸盐和替诺福韦单乙二磺酸盐。
本发明的替诺福韦半乙二磺酸盐A具有图5所示的X射线粉末衍射峰值,具有在4.14°、10.40°、11.67°、12.64°、12.90°、13.24°、15.64°、16.30°、16.86°、18.69°、18.74°、19.19°、19.82°、20.74°、21.36°、21.89°、22.46°、23.19°、23.72°、24.88°、25.56°、26.21°、和27.04°的那些。
本发明的替诺福韦半乙二磺酸盐B具有图6所示的X射线粉末衍射峰值,具有在3.32°、4.18°、5.13°、7.31°、8.59°、10.01°、10.42°、10.97°、11.56°、12.64°、12.95°、13.25°、14.67°、15.53°、16.39°、17.33°、18.15°、18.75°、19.28°、19.93°、20.44°、20.81°、21.37°、22.03°、22.49°、22.84°、23.28°、23.75°、24.66°、25.00°、25.65°、26.36°、27.1°、27.76°、28.16°、28.79°、30.31°、和31.17°的那些。发现其具有不同于替诺福韦富马酸盐(US 5935946)和替诺福韦单乙二磺酸盐的结晶形式。

<试验例5>DSC(差示扫描量热法)
DSC广泛用于测量热流相对于温度的变化,与材料的热传递相关。使用SCINCODSCN650,在氮气条件下以20℃/min的加热速率测量本发明的替诺福韦半乙二磺酸盐和对照组,即替诺福韦富马酸盐和替诺福韦单乙二磺酸盐。图7a示出了替诺福韦富马酸盐的DSC测量值,图7b示出了替诺福韦单乙二磺酸盐的DSC测量值,并且图8示出了替诺福韦半乙二磺酸盐的DSC测量值。
工业实用性
根据本发明,与替诺福韦富马酸盐相比,替诺福韦乙二磺酸盐可以使相关物质随时间的产生最小化,由此在含有本发明盐化合物的产品储存期间杂质的产生可以显著降低,因此提高了药物的稳定性并且避免了对毒性作用进行另外的研究的需要。
Claims (11)
1.一种替诺福韦乙二磺酸盐,其由以下化学式1表示:

2.根据权利要求1所述的替诺福韦乙二磺酸盐,其中所述化学式1的化合物是半乙二磺酸盐,其中替诺福韦和乙二磺酸盐以1:0.5的摩尔比结合。
3.根据权利要求1所述的替诺福韦乙二磺酸盐,其中所述化学式1的化合物具有以下1H核磁共振(NMR)峰:
1H NMR(500MHz,DMSO)8.44(s,1H),8.33(s,1H),5.54-5.45(m,4H),4.82-4.77(m,2H),4.38-4.35(dd,1H),4.25-4.4.21(dd,1H),4.01-3.90(m,3H),2.67(s,2H),1.23-1.22(d,12H),和1.09-1.08(d,3H)ppm。
4.根据权利要求1所述的替诺福韦乙二磺酸盐,其中所述化学式1的化合物具有以下(i)或(ii)的X射线粉末衍射峰值:
(i)替诺福韦乙二磺酸盐A:

5.一种替诺福韦乙二磺酸盐的结晶形式,其具有如图5所示的X-射线粉末衍射峰值。
6.一种替诺福韦乙二磺酸盐的结晶形式,其具有如图6所示的X-射线粉末衍射峰值。
7.一种药物组合物,其包含治疗有效量的权利要求1至4中任一项所述的替诺福韦乙二磺酸盐。
8.根据权利要求7所述的药物组合物,其中所述药物组合物以片剂、胶囊剂、粉剂、颗粒剂、滴丸剂、散剂、丸剂、酊剂或糊剂的剂型提供。
9.一种制备替诺福韦乙二磺酸盐的方法,包括使替诺福韦与乙二磺酸盐二水合物反应。
10.根据权利要求9所述的方法,其特征在于,所述替诺福韦和所述乙二磺酸盐二水合物以1:0.5-1.5的摩尔比反应。
11.根据权利要求10所述的方法,其特征在于,所述替诺福韦和所述乙二磺酸盐二水合物以1:0.5-0.6的摩尔比反应。
