The invention has the beneficial effects that: (1) according to the maca wine, the formula of the health wine is optimized, multiple Chinese herbal medicines are selected, the Chinese medicinal herbs with homology of medicine and food for dispelling effects of alcohol and protecting liver such as kudzuvine flower and hovenia dulcis are compounded on the basis of tonifying kidney and strengthening yang, when the maca wine is drunk, a protective film can be formed on the liver and the stomach, the effects of protecting the liver and nourishing the stomach are achieved, the malaise symptom of drinking is relieved by accelerating urination, perspiration and excretion and decomposition, and the drunk degree is relieved, so that the maca wine has the effects of tonifying kidney and strengthening yang, meanwhile, the damage of alcohol to the liver is reduced; (2) the traditional Chinese medicine composition disclosed by the invention is prepared by selecting reasonable and comprehensive Chinese herbal medicines, wherein the maca, the cistanche and the morinda officinalis can play a role in tonifying kidney yang, benefiting essence and blood and strengthening muscles and bones; the medlar, the sealwort and the eucommia bark can play the roles of nourishing liver and kidney, nourishing yin and moistening lung; the raspberry, the cherokee rose fruit and the leek seed have the effects of securing essence and reducing urination, warming and nourishing liver and kidney, and tonifying yang and securing essence; flos puerariae lobatae and semen hoveniae can protect liver, relieve alcoholism and relieve liver injury caused by drinking; (3) the maca health-care wine is prepared by mixing the Chinese herbal medicines and the white spirit, can exert the comprehensive effects of the medicinal materials to the maximum extent, has the effects of obviously tonifying kidney and yang, promoting the kidney to produce sperm automatically, enabling the essence and qi to be full, keeping primordial qi constant and smooth blood flow, and can assist yang to play, supplement essence and blood, benefit liver and kidney and strengthen tendons and bones, so that the effects of balancing yin and yang and tonifying at the same time are achieved, and the curative effect is obvious; (4) the maca health-care wine prepared by the invention is soft in body, pure in color, sweet and tasty, harmonious in fragrance, complementary in yin and yang, and has the effect of tonifying but not drying, thus being an ideal health-care product integrating nutrition and efficacy. Detailed Description
The invention relates to a preparation method of maca health-care wine for tonifying kidney and strengthening yang, which comprises the following steps:
A. cutting maca, cistanche, morinda officinalis, polygonatum rhizome, eucommia ulmoides and liquorice into pieces with cross-sectional areas of 0.2-0.5 cm2The fragments are dried at the temperature of 30-60 ℃ after being cleaned; cleaning fructus Lycii, Rubi fructus, fructus Rosae Laevigatae, semen Allii Tuberosi, flos Puerariae Lobatae, and semen Hoveniae, and air drying;
B. putting the components obtained in the step A into a wine jar, and mixing the components according to the proportion of 1: 5-1: 10, adding 42-70 degrees of white spirit, sealing the wine jar, soaking for 30-60 days, and inverting the jar for 1-3 times every day in the soaking process;
C. and after the soaking time is over, opening the wine jar, and filtering to obtain the 35-62-degree health wine.
The preparation method takes natural medicinal and edible materials as raw materials, and prepares the maca health wine by selecting proper alcohol content and proper material-liquid ratio to blend and filter with white spirit, thereby not only fully playing the efficacies of enhancing immunity, resisting fatigue, tonifying kidney and strengthening yang, securing essence and reducing urination, protecting liver and warming stomach, strengthening tendons and bones and the like of the raw materials, but also ensuring the efficacy, aroma and taste of the product and improving the stability of the product. The preparation method of the maca health-care wine for tonifying kidney and strengthening yang comprises the following steps:

The raw materials of the maca health-care wine for tonifying kidney and strengthening yang comprise, by weight, 35 parts of maca, 15 parts of cistanche, 10 parts of morinda officinalis, 18 parts of wolfberry fruit, 10 parts of polygonatum rhizome, 15 parts of eucommia ulmoides, 15 parts of raspberry, 15 parts of cherokee rose fruit, 15 parts of leek seeds, 15 parts of pueraria flower, 15 parts of hovenia dulcis thumb and 10 parts of liquorice. The preparation method of the maca health wine is the same as that of the maca health wine in example 1.
Example 4
The maca health wine for tonifying kidney and strengthening yang comprises, by weight, 30 parts of maca, 20 parts of cistanche, 15 parts of morinda officinalis, 15 parts of wolfberry fruit, 12 parts of rhizoma polygonati, 15 parts of eucommia ulmoides, 10 parts of raspberry, 15 parts of cherokee rose fruit, 15 parts of leek seeds, 8 parts of pueraria flower, 8 parts of hovenia dulcis thunb and 10 parts of liquorice. The preparation method of the maca health wine is the same as that of the maca health wine in example 1.
Comparative example 1
A maca health wine comprises, by weight, 46 parts of maca, 20 parts of cistanche, 15 parts of morinda officinalis, 15 parts of wolfberry fruit, 12 parts of rhizoma polygonati, 15 parts of eucommia ulmoides, 10 parts of raspberry, 15 parts of cherokee rose fruit, 15 parts of leek seeds and 10 parts of liquorice. The preparation method of the maca health wine is the same as that of the example 1. The difference between the comparative example and the example 4 is only that the pueraria flower and the hovenia dulcis thunb are deleted from the ingredients, the content of the maca is improved, and the preparation method of the health wine is unchanged.

The invention relates to a maca health wine for tonifying kidney and strengthening yang, which comprises, by weight, 15 parts of maca, 35 parts of cistanche, 15 parts of morinda officinalis, 15 parts of wolfberry fruit, 12 parts of rhizoma polygonati, 15 parts of eucommia ulmoides, 10 parts of raspberry, 15 parts of cherokee rose fruit, 15 parts of leek seeds, 8 parts of pueraria flower, 8 parts of hovenia dulcis thumb and 10 parts of liquorice. The comparative example is different from the example 4 only in that the content of maca in the ingredients is reduced, the content of cistanche is increased, and the preparation method of the health care wine is unchanged.
Comparative example 3
The maca health-care wine for tonifying kidney and strengthening yang comprises, by weight, 30 parts of maca, 20 parts of cistanche, 15 parts of morinda officinalis, 15 parts of wolfberry fruit, 12 parts of rhizoma polygonati, 15 parts of eucommia ulmoides, 10 parts of raspberry, 15 parts of cherokee rose fruit, 15 parts of leek seeds, 16 parts of pueraria flower and 10 parts of liquorice. First, sensory evaluation
The same amount of maca health care wine in the above examples and comparative examples was taken for sensory evaluation. The evaluation mode adopts 10 points, 3 points of fragrance, 3 points of color and 4 points of taste, 10 experienced sensory panel judges the taste of the wine, and according to the total points, the results are shown in the following table 2:

1.1 test sample: the maca health-care wine prepared by the embodiments and the comparative examples listed in the invention is used as a test sample to perform evaluation tests on the effects of tonifying kidney and strengthening yang, promoting impotence and protecting liver.

2. Method and results
2.1 Effect on castration and Kidney deficiency in the Extra organs of rats
2.1.1 Effect on the sexual organs of castrated Male rats
(1) Taking 70 male rats with the weight of 102-126 g, removing bilateral testicles, injecting penicillin into muscles after operation, dividing three days after castration into 7 groups at random, wherein each group comprises 10 castration model groups, examples 1-4 and comparative examples 1 and 2, continuously irrigating the stomach for 20 days, and each day comprises 20ml of physiological saline water for irrigating the stomach of the castration model groups and corresponding maca health-care wine for irrigating the stomach of the examples and comparative examples; another 10 non-castrated rats were kept and observed in the same manner as normal control group. At 20d, the rat penis was placed with a stimulation electrode of a WQ-10B multi-use electronic acupoint apparatus, and a stimulation intensity of 3mA was applied to the electrode, and the penis erection latency (the interval from the start of stimulation to the erection of penis) was recorded, and the results are shown in Table 3.

As can be seen from the table 1, the maca health care wine prepared by the formula can improve the excitability of penis of castrated male rats to external stimulation, has the effect of shortening the erection latent period of the penis of the male rats, has significant difference (P is less than 0.05) compared with a castration model group, and has significant difference (P is less than 0.01) compared with a normal control group.
(2) Taking 70 male rats with the weight of 100-110 g, castrating into 7 groups, 10 male rats in each group, namely a castration model group, 1-4 male rats and comparative examples 1 and 2 male rats, and continuously performing intragastric administration for 20 days, wherein the daily volume is 20 ml; the castration model group comprises normal saline for gastric lavage, and the examples and comparative example groups comprise corresponding maca health care wine for gastric lavage; another 10 non-castrated rats were kept and observed in the same manner as normal control group. Animals were sacrificed after weighing on day 20, the seminal vesicle, prostate and glandulae preputiales were weighed rapidly and the amount per 100g of weight was calculated and the results are shown in table 4.

The results show that: the weight of animals (castrated rats) with kidney deficiency and the weight of each reproductive organ are obviously lower than those of normal control animals, which indicates that the molding of the animals with kidney deficiency is successful. The weight of the seminal fluid sac, the prostate and the glandula preputialis of the male rat of the maca health-care wine for gastric perfusion is obviously higher than that of a castration model group, which shows that the maca health-care wine has the effect of promoting the development of male organs after being taken for a long time.
2.1.2 Effect on the adnexal organs of castrated female rats
Taking 70 female rats with the weight of 100-110 g, cutting off bilateral ovaries under ether anesthesia, performing conventional anti-inflammatory treatment, randomly dividing the rats into 7 groups after 3 days, wherein each group comprises 10 female rats, namely a castration model group, examples 1-4 and comparative examples 1 and 2, and performing continuous intragastric administration for 20 days, wherein the daily volume is 20 ml; the castration model group comprises normal saline for gastric lavage, and the examples and comparative example groups comprise corresponding maca health care wine for gastric lavage; another 10 non-castrated rats were kept and observed in the same manner as normal control group. After day 20 the animals were sacrificed and the weight of the preputial glands and bilateral uteri was quickly weighed and calculated per 100g of body weight, the results are shown in table 5.

The results show that: the weight of periglandular glands and uterus of female animals of the maca health-care wine after gastric perfusion is obviously higher than that of a castration model group, so that the maca health-care wine has the effect of promoting development of female organs after long-term administration.
2.2 Effect on animals with Yang deficiency
Taking 70 mice, each half of the mice is male and female, randomly dividing the mice into 7 groups, wherein each group comprises 10 mice, namely a yang deficiency disease model group, examples 1-4 groups and comparative examples 1 and 2 groups, injecting 25mg/kg/d of hydrocortisone, continuously irrigating the stomach for 20d, and 20ml of the mice per day, and irrigating corresponding maca health care wine for the stomach of the yang deficiency disease model group, the examples and the comparative examples; another 10 animals were kept and observed in the same manner as the normal control group. Each group of mice was dosed once a day for 7 days, and the survival time (water temperature 10 ℃, water depth 50cm, width 50cm, swimming in swimming pool 120cm long) and the number of autonomic activities (within 10 min) of each group of mice were recorded after 1 hour of the last dose, and the test results are shown in Table 6.

The results show that: the large dose of hydrocortisone can cause the cold resistance and fatigue resistance of mice to be reduced, and the maca health care wine prolongs the low-temperature swimming survival time of the mice with yang deficiency, and has significant difference compared with a yang deficiency group and a normal control group. The maca health-care wine can obviously enhance the autonomous activity of mice, and has significant difference compared with a yang deficiency group and a normal control group.
2.3 liver protection efficacy verification
The reduction of liver damage according to the present invention is achieved by improving liver protection activity provided by the synergistic combination of pueraria lobata flower and hovenia dulcis thunb included in the health wine. Therefore, comparative examples 1, 3 and 4 are selected as control groups of the embodiment of the invention to study the liver protection effect of the maca health-care wine.
40 rats are taken, half of the rats are male and female, the rats are randomly divided into 4 groups, 10 rats are taken in each group, the rats are respectively the example 4 and the comparative examples 1, 3 and 4, the medicine is continuously drenched for 30 days, 20ml of the medicine is taken every day, and the maca health care wine is corresponding to the drenching of the rats in the examples and the comparative examples. On day 30, taking 0.5ml of blood from the tail of each rat every 30min after drug filling, and determining the concentration of ethanol in the blood, wherein the determination result shows that the concentration of ethanol in the blood is highest 60min after drug filling; the Malondialdehyde (MDA), Alcohol Dehydrogenase (ADH) and superoxide dismutase (SOD) activities in the blood were measured 30 days after the end of the test. The test results are shown in Table 7.

Test results show that the anti-alcoholic substances contained in the flower of kudzuvine and the raisin tree seed in the maca health-care wine can improve the activity of ADH active Alcohol Dehydrogenase (ADH) and the activity of superoxide dismutase (SOD) in blood of a white rat after drinking for a long time, accelerate the catabolism speed of ethanol, reduce the content of ethanol in blood and the accumulation time of acetaldehyde as an intermediate metabolite in vivo, thereby reducing the accumulation of Malondialdehyde (MDA) and playing a role in reducing alcoholism and protecting liver function. The liver tissue HE staining result shows that the anti-alcoholism substances contained in the flower of kudzuvine and the hovenia dulcis thunb in the maca health-care wine can reduce the liver function damage caused by hepatic cell steatosis, hepatic cell necrosis, fatty liver and the like after the rats drink the wine for a long time, and the liver-protecting effect is achieved.
The pharmacodynamic test result shows that: the maca health-care wine has the effects of improving the excitability of penis of a castrated rat to external stimulation and shortening the latent period of penile erection. The maca health wine has the effects of increasing the weight of seminal fluid sacs, prostates, preputial glands and uteri for animals with kidney deficiency (castrated rats), and is identical to the effects of tonifying kidney and strengthening yang and promoting yang to wither. Can be used for improving cold tolerance, prolonging low temperature swimming time, and enhancing activity of animals (hydrocortisone mice) with yang deficiency. The sobering-up substances contained in the flower of kudzuvine and the raisin tree seed in the maca health-care wine can accelerate the catabolic speed of ethanol, reduce the ethanol content in blood and the accumulation time of an intermediate metabolite acetaldehyde in vivo, reduce the liver function damage phenomenon caused by hepatic cell steatosis, hepatic cell necrosis, fatty liver and the like after a rat drinks for a long time, and play a role in reducing alcoholism and protecting the liver function. Meanwhile, the effects are superior to the effects of the health care wine after the formula is changed according to the comparative example, which shows that the formula and the proportion of the medicinal wine are optimal, the medicinal wine can complement yin and yang, regulate the balance of yin and yang in vivo, strengthen and protect the kidney and the liver, and has obvious curative effects on the sub-health state caused by kidney-yang deficiency.
Third, clinical research
1. Trial drinking test
The maca health-care wine for tonifying kidney and strengthening yang disclosed by the invention has an obvious curative effect after being taken by patients with kidney-yang deficiency in clinic, and after being taken by 200 patients with kidney-yang deficiency in clinic (140 men and 60 women), the maca health-care wine is continuously taken for 2-3 days, and 100% of patients feel more peaceful intestines and stomach and have an enhanced appetite; 98% of patients feel refreshed and energetic after continuously taking the medicine for 7-15 days; after continuously taking the medicine for 3 months, 95 percent of patients have the renal function reaching the level of normal people, the symptoms such as soreness and weakness of waist and knees, intolerance of cold and cold limbs, impotence, premature ejaculation, spermatorrhea and the like disappear, the sexual function returns to normal, and the medicine is stopped without rebounding. After being taken by 20 cases of impotence patients clinically, the number of sperms of 9 cases of patients is increased by 35 to 45 percent after the patients continuously take the medicine for 3 months; after being taken by 18 patients with cold sperm clinically, 98 percent of the patients feel the cold sperm phenomenon relieved within 7 to 15 days, and 97 percent of the patients do not have the cold sperm phenomenon within 2 to 3 months. Because the ingredients of the maca health-care wine for tonifying kidney and strengthening yang comprise the pueraria flower and the raisin tree seed, when the health-care wine is drunk, a protective film can be formed in the liver and the stomach in advance, the effects of protecting the liver and nourishing the stomach are achieved, urination is accelerated, the uncomfortable symptoms of drinking can be relieved, and the maca health-care wine can wake up or recover the mental state quickly even if drunk, so that the daily dose of a patient can be higher than that of common medicinal wine in the market.
2. Typical case
(1) Liza, male, 50 years old, Xianning. The sexual intercourse is restrained because workers live separately in two places for a long time, and the sexual life is frequent because the sexual intercourse is not restrained in the early development stage of going back to the countryside, and the penis is gradually erected and is not hard, and the sexual intercourse is not easy to move for more than 2 years. The traditional Chinese medicine diagnosis is as follows: kidney yang deficiency. Various kidney-tonifying and yang-reinforcing medicines, health-care foods and the like have not been taken before to achieve the curative effect. Dizziness, fatigue, soreness of the waist and legs, insomnia, dreaminess, palpitation, shortness of breath, sallow complexion and poor appetite in case of fatigue.
