背景技术
铁皮石斛(Dendrobium officinale Kimura et Migo)是兰科石斛属植物,是一种名贵的中药材,具有悠久的应用历史,被列为九大仙草之首,是药中黄金。现代医学研究表明:铁皮石斛中所含有的石斛多糖,生物碱,菲类化合物等具有较高的医疗保健作用,能够提高机体免疫力。
枸杞是一种传统的中药,在中国也有较悠久的药用历史,古中医用其治疗虚劳精亏,内热消渴,血虚萎黄等,现代医学研究表明枸杞具有增强非特异性免疫的作用。
沙棘是一种含有多种维生素、多种微量元素、多种氨基酸和其他生物活性物质的药用食物,在中国具有悠久的药用历史。古代中医藏学,蒙医将沙棘列为重要的药用植物,用于止咳、平喘、活血化瘀等。现代研究表明沙棘中含有丰富的营养物质,尤其是过时含有人体不能合成的,人的身心健康不可缺少的多种维生素。
水果含有丰富的维生素、矿物质及微量元素,且某些水果还含有一些特定的营养物质。
现有保健品大多采用胶囊、冲剂或口服液等传统形式,不便于食用和携带,因此,很有必要在现有保健品基础上,研发一种便于携带和食用的保健品。咀嚼片是一种便于携带与使用的加工形式,据统计目前有关石斛的咀嚼片有多种,但大多采用单一的形式制作,难以满足人们对多种营养的复合需求。
发明内容
本发明所要解决的技术问题是提供一种含丰富营养物质的石斛复合咀嚼片及其制备方法。
为解决上述技术问题,本发明提供了一种石斛复合咀嚼片,其组分包括:石斛、枸杞、沙棘,另外还包括各种不同口味的水果,其重量配比为:石斛10份,枸杞4-6份,沙棘2-3份,水果1-8份。
所述石斛复合咀嚼片的制备方法包括如下步骤:
(1)鲜品处理:按重量配比取石斛鲜条10份、枸杞4-6份、沙棘2-3份、水果1-8份洗净,沥干表面水分,切成片或段备用;
(2)干燥:采用冷冻干燥设备将处理后的石斛茎段、枸杞、沙棘和水果真空干燥,至其含水量在9%-10%;
(3)粉碎:将干燥石斛茎段放入超微粉碎机进行破壁粉碎,枸杞、沙棘、水果分别进行粉碎;
(4)过筛:将石斛粉末过200目筛网,与枸杞粉、沙棘粉、果粉混合后过80目筛网;
(5)加辅料:在混合粉中加入微晶纤维素和蜂蜜,充分混合;
(6)压片:将混合物放入压片机压成直径为8-16mm的各种片剂,压完后需检查压片是否存在没有压实的情况,并将未压实的片剂从新压片;
(7)灭菌、包装:将压好的石斛复合片放在紫外灯管下杀菌,装入包装器皿即可。

据了解,微晶纤维素(MCC)、糊精和淀粉均可作为咀嚼片的填充剂,压制片剂。本发明通过测定三种填充剂成形性和硬度以期选择最佳的填充剂。实验结果表明,选用微晶纤维素(MCC)作为填充剂所得石斛复合咀嚼片,片剂成形性最好,淀粉次之,糊精最差。因此在本发明中选用微晶纤维素(MCC)作为石斛复合咀嚼片的填充剂。

本发明的咀嚼片具有提高免疫力的作用,其实验方法及数据如下:
1、剂量:本发明的铁皮石斛营养咀嚼片的人体推荐剂量为9g/天,在实验检测中检测剂量设置低、中、高三个剂量,即0.5g/kg,1.0g/kg、3.0g/kg,分别相当于人体剂量的5、10、30倍。对各实验组小鼠经口服用本发明样品30天,另设置空白对照。

1)将200只小鼠按重量随机分为五个组,每组40只。一组用于NK细胞活性检测和淋转实验,二组用于DTH、脾指数和胸腺指数实验,三组进行溶血空斑和溶血素实验,四组进行碳廓清实验,五组进行吞噬实验。
2)喂养并称重:按剂量设计连续喂养小鼠30天,每周称重一次,计算实验过程中小鼠体重变化。
3)脏器/体重比值(胸腺指数、脾指数)测定:小鼠连续服用30天,颈椎脱臼处处死,取胸腺、脾脏称重,分别计算胸腺指数和脾指数。
4)细胞免疫功能测定
a) ConA诱导的小鼠淋巴细胞转化实验:小鼠连续给样30天后,颈椎脱臼处死,取脾脏,制成脾淋巴细胞悬液,调整细胞浓度为3*106个/ml,将细胞悬浮液分两孔加入24孔培养板中,每孔1ml,一孔加75µlConA液(相当于7.5µg/ml),另一孔作为对照,置5%CO2,37℃培养72h,培养结束前4h,每孔加入MTT(5mg/ml)50µl/孔,继续培养4h,培养结束后,每孔加入1ml酸性异丙醇,吹打混匀,使紫色结晶完全溶解后,以570nm波长进行比色,记录实验结果并进行统计。
b) DTH测定及胸腺指数、脾指数的测定:致敏:小鼠连续给养30天后,将小鼠腹部去毛,范围约3*3cm,将1%DNFB的溶液10µl均匀涂抹于小鼠右耳进行攻击。24h后称重,颈椎脱臼处死,用打孔器取下直径8mm的左右耳片,称重并计算肿胀度。再取胸腺、脾脏称重,分别计算胸腺指数、脾指数。
5)体液免疫功能测定
a)抗体产生检测试验:动物连续给养样品30天后,将脱纤维绵羊红细胞免疫5天后,动物颈椎脱臼处死,取脾脏,制成脾细胞悬浮液,调整细胞浓度为5*106个/ml。将表层培养基(1g琼脂糖加双蒸水至100ml)加热溶解后,放入45℃水浴保温,与等量pH7.2-7.4、2倍浓度的Hanks液混合,分装小试管,每管0.5ml,再向管内加50µl110%SRBC,20µl中温育1.5h,然后各排比做倍比稀释,每孔加1%SRBC100µl震荡后37℃放置3h,当血球对照出现沉落后观察结果。
6)单核—巨噬细胞功能测定实验
a)小鼠碳廓清实验:小鼠连续给予样品30天后,在尾部静脉注射1:3稀释的印度墨汁,待墨汁注入,立即计时。注入墨汁后2-10min后,分别从眼静脉取血20µl,并将其加到2ml0.1%碳酸钠溶液中,用分光光度计在600nm处测定OD值,以碳酸钠溶液作空白对照。根据动物体重、肝重和脾重计算吞噬指数。
b)小鼠腹腔巨噬细胞吞噬鸡红细胞实验:动物连续给养样品30天后,每鼠腹腔注射20%鸡红细胞悬浮液1ml,间隔30min,颈椎脱臼处死,固定于鼠板上,剪开腹部皮肤,注射生理盐水2ml,转动鼠板1min,吸出腹腔洗液1ml,分滴于2片玻片上,37℃孵箱湿孵30min,用生理盐水漂洗晾干,用油镜镜检,计算吞噬百分率(%)和吞噬指数。
7)NK细胞活性测定:小鼠连续给养样品30天后,动物颈椎脱臼处死,取脾脏,制成脾脏悬浮液(效应细胞),取传代后24Hyac-1细胞加入RPMI1640完全培养液,调整细胞浓度为4*105个/ml(靶细胞),取靶细胞和效应细胞各100µl(效靶比50:1),加入U型96孔板,靶细胞自然释放孔加靶细胞和培养液各100µl,最大释放孔加靶细胞和1%NP40各100µl,上述各项均设三个重复组,于37℃、5%CO2培养箱中培养4h,每孔吸取上清液100µl置平地96孔板中,同时加入LDH基质液100µl,反应30min,每孔加入1mol/l的HCL30µl终止,用酶标仪在490nm处测定OD值。
4、结果
实验过程中小鼠体重观察的结果如表1所示。

5、结论:在本实验条件下,细胞免疫功能、体液免疫功能、NK细胞活性和单核-巨噬细胞功能均为阳性,根据《保健食品检验与评价技术规范》中“增强免疫功能”的结果判定,认为小鼠经口服用本发明产品30天后,具有增强免疫力功能。
显然,上述实施例仅仅是为清楚地说明本发明所作的举例,而并非是对本发明的实施方式的限定。对于所属领域的普通技术人员来说,在上述说明的基础上还可以做出其它不同形式的变化或变动。这里无需也无法对所有的实施方式予以穷举。而这些属于本发明的精神所引伸出的显而易见的变化或变动仍处于本发明的保护范围之中。
Claims (1)
1.一种所述石斛复合咀嚼片的制备方法,其特征在于包括如下步骤:
(1)鲜品处理:按重量配比取石斛鲜条10份、枸杞5份、沙棘2份、水果哈密瓜6份,沥干表面水分,切成片或段备用;
(2)干燥:采用冷冻干燥设备将处理后的石斛茎段、枸杞、沙棘和水果真空干燥,至其含水量在9%-10%;
(3)粉碎:将干燥石斛茎段放入超微粉碎机进行破壁粉碎,枸杞、沙棘、水果分别进行粉碎;
(4)过筛:将石斛粉末过200目筛网,与枸杞粉、沙棘粉、果粉混合后过80目筛网;
(5)加辅料:在混合粉中加入微晶纤维素和蜂蜜,充分混合;
(6)压片:将混合物放入压片机压成直径为8-16mm的各种片剂;
(7)灭菌、包装:将压好的石斛复合片放在紫外灯管下杀菌,装入包装器皿即可。
