Disclosure of Invention
The invention aims to provide a high-efficiency, natural and safe traditional Chinese medicine composition which can be used for preventing and treating broiler ascites syndrome. The active ingredients of the traditional Chinese medicine composition are prepared from ligusticum wallichii, poria cocos, ginseng leaves, panax notoginseng and plantain seeds, and the traditional Chinese medicine composition is good in curative effect, low in toxicity, free of residue, green and environment-friendly.
The invention also aims to provide a preparation method and application of the traditional Chinese medicine composition.
The purpose of the invention is realized by the following technical scheme:
the invention provides a traditional Chinese medicine composition for preventing and treating ascites syndrome of poultry, which contains active ingredients, wherein the active ingredients are prepared from ligusticum wallichii, poria cocos, ginseng leaves, panax notoginseng and plantain seeds.
The active ingredients are prepared from the following components in parts by weight: 35-90 parts of ligusticum wallichii, 15-60 parts of poria cocos, 10-50 parts of ginseng leaves, 10-45 parts of panax notoginseng and 20-70 parts of plantain seeds.
Further, the active ingredients are prepared from the following components in parts by weight: 40-80 parts of ligusticum wallichii, 20-50 parts of poria cocos, 10-40 parts of ginseng leaves, 15-30 parts of panax notoginseng and 20-60 parts of plantain seeds.
Furthermore, the active ingredients are prepared from the following components in parts by weight: 50-70 parts of ligusticum wallichii, 25-40 parts of poria cocos, 20-40 parts of ginseng leaves, 20-30 parts of panax notoginseng and 30-50 parts of plantain seeds.
Wherein, the traditional Chinese medicine composition can be prepared into powder, pulvis, granules, oral liquid or injection.
The invention also provides a preparation method of the traditional Chinese medicine composition, which comprises the following steps:
1) weighing the raw materials in parts by weight, crushing, sieving and mixing;
2) optionally, adding 6-12 times of distilled water for decocting for three times, each time for 1.5 hours, combining decoction liquids, filtering, concentrating the filtrate to a proper amount, adding 1-5 times of absolute ethyl alcohol, fully and uniformly stirring, standing for 1-5 hours for precipitation, filtering, concentrating the filtrate to an extract, drying, adding a corresponding pharmaceutically acceptable carrier or auxiliary material, and preparing into a corresponding dosage form.
The traditional Chinese medicine composition can be used as a feed additive, is added into basic ration of poultry, and can prevent and treat ascites syndrome of the poultry after being fed, so the invention also provides a feed additive and a feed containing the traditional Chinese medicine composition, and the feed comprises premix, concentrated feed and batch feed.
The traditional Chinese medicine composition can be used for preparing the medicines for treating the ascites syndrome of the poultry. The traditional Chinese medicine composition has the following formula principle: the rhizoma ligustici wallichii is capable of activating blood and qi, dispelling wind and relieving pain; tuckahoe has the effects of promoting diuresis and eliminating dampness, strengthening spleen and calming heart; the ginseng leaf has the effects of tonifying qi and promoting the production of body fluid; the panax notoginseng can dissipate blood stasis and relieve swelling, diminish inflammation and relieve pain; che Qian Zi can excrete dampness and induce diuresis. The formula has the beneficial effects that the ligusticum wallichii and the poria cocos in the formula are used together, so that the formula has the wonderful effects of promoting qi circulation, removing blood stasis and removing dampness, and has the effect of activating spleen. Semen plantaginis and rhizoma Schizocapsae Plantagineae have effects of relieving swelling, removing blood stasis, inducing diuresis, eliminating body edema, invigorating qi, promoting fluid production, and improving body resistance. Modern pharmacological studies show that the ligusticum wallichii is a common traditional Chinese medicine for treating cardiovascular and cerebrovascular diseases, can dilate blood vessels, improve microcirculation, relax pulmonary artery, reduce pulmonary arterial pressure and resist myocardial ischemia, and the pachyman in the poria cocos can obviously enhance the immune function of an organism. The effect of treating both principal and secondary aspect of disease is achieved by fundamentally acting on the focus.
The traditional Chinese medicine composition has the advantages and beneficial effects that:
1) the traditional Chinese medicine composition is prepared according to the syndrome differentiation theory of traditional Chinese veterinary medicine formulas, and has the advantages of quick response and obvious curative effect.
2) The traditional Chinese medicine composition is used for preventing and treating broiler ascites syndrome, and overcomes the problems of residue and drug resistance caused by western medicines.
3) The traditional Chinese medicine composition is convenient to use, has a simple preparation process, and can be prepared into powder, granules, oral liquid, injection and the like.

The following examples are intended to illustrate the invention but are not intended to limit the scope of the invention.
Example 1
(1) Weighing the following raw materials in parts by weight:
45 parts of ligusticum wallichii, 30 parts of poria cocos, 20 parts of ginseng leaves, 45 parts of panax notoginseng and 35 parts of plantain seeds.
(2) Pulverizing the raw medicinal materials weighed in the step (1), sieving, and fully mixing to prepare powder.
Example 2
(1) Weighing the following raw materials by weight:
35 parts of ligusticum wallichii, 20 parts of poria cocos, 10 parts of ginseng leaves, 15 parts of panax notoginseng and 30 parts of semen plantaginis.
(2) Pulverizing the raw materials, sieving, adding 6 times of distilled water, decocting for three times, each time for 1.5h, mixing decoctions, filtering, concentrating the filtrate to appropriate amount, adding 2 times of anhydrous ethanol, stirring, standing for 2 h, precipitating, filtering, concentrating the filtrate to extract, adding appropriate amount of soluble starch and dextrin, mixing, and drying to obtain powder (1 g per 1g is equivalent to 1g of crude drug).
Example 3
(1) Weighing the following raw materials in parts by weight:
40 parts of ligusticum wallichii, 50 parts of poria cocos, 30 parts of ginseng leaves, 20 parts of panax notoginseng and 45 parts of semen plantaginis.
(2) Pulverizing the raw materials, sieving, adding 12 times of distilled water, decocting for three times, each time for 1.5h, mixing decoctions, filtering, concentrating the filtrate to appropriate amount, adding 3 times of anhydrous ethanol, stirring, standing for 3 h, precipitating, filtering, concentrating the filtrate to extract, adding appropriate amount of soluble starch and dextrin, mixing, drying, and making into granule (each 1g is equivalent to 1g of crude drug).
Example 4
(1) Weighing the following raw materials in parts by weight:
60 parts of ligusticum wallichii, 30 parts of poria cocos, 25 parts of ginseng leaves, 20 parts of panax notoginseng and 40 parts of semen plantaginis.
(2) Pulverizing the raw materials, sieving, adding 8 times of distilled water, decocting for three times, each for 1.5 hr, mixing decoctions, filtering, concentrating the filtrate to appropriate amount, adding 5 times of anhydrous ethanol, stirring, standing for 4 hr, filtering, concentrating the filtrate to 1.0 g per ml crude drug, adding appropriate amount of sucrose, benzoic acid, and ethylparaben, and making into oral liquid.
Example 5
(1) Weighing the following raw materials in parts by weight:
50 parts of ligusticum wallichii, 25 parts of poria cocos, 20 parts of ginseng leaves, 30 parts of panax notoginseng and 30 parts of semen plantaginis.
(2) Pulverizing the raw materials, sieving, adding 10 times of distilled water, decocting for three times, each for 1.5 hr, mixing decoctions, filtering, concentrating the filtrate to appropriate amount, adding 5 times of anhydrous ethanol, stirring, standing for 5 hr, filtering, concentrating to 1.0 g crude drug per ml, adding appropriate amount of benzoic acid and ethylparaben, and making into injection.
Test example 1 clinical prevention effect of the Chinese medicinal composition of the present invention on ascites syndrome in broiler chicken
The experiment is carried out in an experimental animal house of the research center of the great north agricultural and animal husbandry technology, and the broiler ascites syndrome is induced by manufacturing a low-temperature cold environment. 360 healthy AA broilers with the age of 4 weeks are selected and randomly divided into 3 groups of 120 broilers. Group 1 is the traditional Chinese medicine composition of the invention, prepared by the embodiment 1, 1 kg of the mixture is mixed every 1g, and the mixture is eaten freely; the group 2 is a control drug group, 1 kg of vitamin C is added for every 1g, and the group is fed freely; the group 3 is a low-temperature control group, no medicine is added, and other feeding conditions are completely consistent. The temperature of the henhouse is reduced by 1-2 ℃ every day, the temperature is gradually reduced to 8-12 ℃ from the normal temperature of 25 ℃, the symptoms of the broiler chickens are observed, and the morbidity and the mortality of the ascites syndrome of the broiler chickens are recorded. The main symptoms of the sick broiler chickens are as follows: lassitude, disorder of feather, anorexia, slow movement, abdominal swelling, and fluctuation of contact pressure; the chickens which are killed by the caesarean section have a large amount of ascites in the abdominal cavity, are brownish red, have fibrin clot, have swollen liver, hydropericardium and right heart dilatation. According to results, the morbidity of the drug group is 1.70%, and no broiler death exists; the morbidity of the control drug group is 2.50 percent, and the mortality is 0.83 percent; the morbidity of the low-temperature control group is 12.50 percent, and the mortality is 11.67 percent. The morbidity and mortality of the drug group and the control drug group are obviously lower than those of the low-temperature control group (P is less than 0.05), which indicates that the traditional Chinese medicine composition can prevent the broiler ascites syndrome. The results are shown in Table 1.

Test example 2 clinical treatment effect of the Chinese medicinal composition on ascites syndrome in broiler chicken
The test is carried out in an experimental animal house of the research center of the great north agricultural and animal protection technology. 490 AA commercial broilers selected for the test were randomly divided into 7 groups of 70 broilers each. Breeding the test chickens 21 days old; after 21 days of age, the broiler chickens in the 1 st group are still normally fed as a normal-temperature control group, and the temperature of the henhouses of other groups is gradually reduced from 25 ℃ to 12 ℃ according to the temperature of 1-2 ℃ per day; adding T into the feed per kg for groups 4, 5 and 73(Triiodothyronine) 1.5 mg to induce broiler ascites syndrome. Observing symptoms of no semen collection, lying, standing difficulty, abdominal distension and the like of the broiler chickens, using the traditional Chinese medicine composition in groups 3 and 4, preparing the traditional Chinese medicine composition by adopting the embodiment 2, adding 1 kg of water for every 1g, continuously drinking for 6 days, and freely drinking; the group 6 and group 7 are used as control drug group, and each 5 g vitamin C is mixed with 1 kg water for 6 days. Group 2 as low temperature control group, group 5 as low temperature + T3The control group was normally maintained without drug administration. According to the results, the morbidity and mortality of the normal temperature control group are 2.86% and 1.40%, the morbidity and mortality of the low temperature + drug group are 1.40% and 0.00%, and the morbidity and mortality of the low temperature + vitamin c group are 2.86% and 1.40% which are significantly lower than the morbidity and mortality of the low temperature control group are 12.86% and 11.43% (P is the same as P)<0.05); low temperature + T3Control group with T3The morbidity of the broiler chickens is up to 30.0 percent, and the mortality is up to 28 percent57%; the morbidity and mortality of the low-temperature + T3+ drug group of the invention are 5.71% and 2.86%, and the morbidity and mortality of the low-temperature + T3+ vitamin c group of the invention are 7.14% and 4.29% which are obviously lower than those of the low-temperature + T3+ drug group3The control group had 30.00% morbidity and 28.57% mortality (P)<0.05), the sum of the temperature of the broiler chicken at low temperature and low temperature plus T is greatly reduced3The morbidity under the condition shows that the traditional Chinese medicine composition can treat the broiler ascites syndrome. Test example 3 clinical treatment effect of the Chinese medicinal composition on ascites syndrome in broiler chicken
500 broilers of 28 days old are fed in a certain chicken farm in the Hebei corridor, and the phenomena of poor spirit, walking delay, standing difficulty, abdomen landing, penguin-shaped appearance, abdomen expansion, feather disharmony, tachypnea, diarrhea and the like appear in part of the broilers. When the cesarean chicken has a large amount of brownish red or khaki ascites in the abdominal cavity, the chicken is clear or mixed with cellulose protein clots; the symptoms of liver swelling and blood stasis, hydropericardium, obvious right ventricle expansion, heart wall thickening and the like are diagnosed as the broiler ascites syndrome. From these, 135 chickens with similar body weight and typical symptoms were picked and divided into 3 groups of 45 chickens. The group 1 uses the traditional Chinese medicine composition prepared by the embodiment 3, 1 kg of the traditional Chinese medicine composition is mixed with 1g of the vitamin C, the traditional Chinese medicine composition is eaten freely and continuously for 6 days, the group 2 is a control medicine group, 1 kg of the vitamin C is mixed with 5 g of the vitamin C, the traditional Chinese medicine composition is eaten freely and continuously for 6 days, the group 3 is a blank control group, no medicine is given, no treatment is carried out, and other feeding conditions are kept consistent. Meanwhile, the sanitation of the internal and external environments of the house is well protected, the disinfection is performed at regular time, and the ventilation is increased. According to the results, the cure rate of the drug group is 95.56 percent, and the cure rate of the drug control group is 93.33 percent, which are obviously higher than the cure rate of the blank control group by 11.11 percent (P is less than 0.05). The traditional Chinese medicine composition can treat the broiler ascites syndrome, and the treatment effect is better than that of a control medicine. The results are shown in Table 3.

Test example 4 clinical treatment effect of the Chinese medicinal composition on ascites syndrome in broiler chicken
850 broiler chickens are newly introduced into a certain chicken farm in Hebei stone village, and most of the broiler chickens have the defects of fluffy feather, listlessness, difficult breathing and no desire to move. The abdomen is enlarged and is shaped like a water bag, and palpation has the symptoms of wave motion, thinning and cooling of the skin of the abdomen and the like. Abdominal congestion and edema of the dead chickens after the caesarean section; pericardial effusion, enlarged heart, peritoneal effusion, etc. Isolating the sick broilers in the chicken flocks and dividing the broilers into 3 groups, wherein the 1 st group is prepared from the Chinese medicinal composition of the invention by adopting the embodiment 4, 1 ml of the Chinese medicinal composition is added with 1 kg of water, and the broilers are freely drunk and normally raised; the group 2 is used as drug control group, and 5 g vitamin C is mixed with 1 kg water, and is administered freely and fed normally; group 3 was used as a blank control group, without any drug, and was normally kept under otherwise identical conditions. And recording the cure rate and the death rate of the broilers in each group every day, and counting the cure rate and the death rate. According to the results, the cure rate of the drug group of the invention is 93.85 percent, and the cure rate of the drug control group is 92.31 percent, which are obviously higher than the cure rate of the blank control group by 10.77 percent (P < 0.05). The traditional Chinese medicine composition can treat the broiler ascites syndrome. The results are shown in Table 4.
