Disclosure of Invention
Aiming at the problems existing in the prior art, the invention aims to provide a traditional Chinese medicine composition for preventing and treating duck taeniasis.
In order to achieve the aim of the invention, the technical scheme adopted by the invention is as follows:
the invention provides a traditional Chinese medicine composition for preventing and treating duck taeniasis, which is prepared from the following raw materials in parts by weight: 50-70 parts of acupuncture iron seeds, 30-50 parts of kola nut, 30-50 parts of lysimachia foenum-graecum hance, 30-50 parts of sweetsop, 30-50 parts of betel nut and 20-40 parts of rhizoma dryopteris crassifolia, wherein the source of the raw material medicine is a traditional Chinese medicine or a traditional Chinese medicine extract equivalent to the crude drug of the traditional Chinese medicine.
According to the traditional Chinese medicine composition, preferably, the traditional Chinese medicine composition is prepared from the following raw materials: 60 parts of acupuncture iron seeds, 40 parts of kola nut, 40 parts of lysimachia foenum-graecum hance, 40 parts of sweetsop, 40 parts of betel nut and 30 parts of rhizoma dryopteris crassifolia.
The second aspect of the invention provides a preparation method of the traditional Chinese medicine composition in the first aspect, which comprises the steps of mixing the acupuncture iron seeds, the kola nut, the lysimachia foenum-graecum, the sweetsop, the betel nut and the cyrtomium rhizome in proportion, adding a solvent for extraction, filtering after the extraction is completed, and collecting filtrate.
According to the above preparation method, preferably, the solvent is water or ethanol. More preferably, the solvent is water.
The third aspect of the invention provides application of the traditional Chinese medicine composition in preparation of medicines for treating taeniasis of ducks.
The fourth aspect of the invention provides a medicine for preventing and treating taeniasis of ducks, which is prepared from the traditional Chinese medicine composition in the first aspect and pharmaceutically acceptable auxiliary materials.
According to the above medicament, preferably, the medicament is in an oral dosage form.
According to the above medicine, preferably, the medicine is granule, tablet, oral liquid. More preferably, the medicament is a granule.
The fifth aspect of the present invention provides a method for preparing the medicament according to the fourth aspect, wherein the method comprises the steps of mixing the acupuncture iron seeds, the kola nut, the lysimachia foenum-graecum, the sweetsop, the betel nut and the cyrtomium rhizome in proportion, adding a solvent for extraction, filtering after the extraction is completed, and collecting filtrate; concentrating the filtrate to obtain extract, and adding pharmaceutically acceptable adjuvants to obtain the final product. Preferably, the solvent is water or ethanol. More preferably, the solvent is water.
The traditional Chinese medicine composition provided by the invention is prepared by purchasing all components in a commercially available product, and the formula principle of the traditional Chinese medicine composition is as follows:
needling iron seeds: is fruit of Philippine iron seed of Philippine. Harvesting and sun drying after 10-12 months of fruit ripening. Bitter and sour; the sex is flat. Functional insect repellent. Major taeniasis.
Vine fruit: the product is dry mature fruit of Bulletia Folium Embelia ob10ngifolia Hemsl. Picking the fruits in autumn and sun drying. Pungent taste; warm and sharp nature. Has the functions of killing parasites and raising stomach temperature. Can be used for treating taeniasis and edema.
Lysimachia foenum-graecum: the product is dry aerial part of herba Lysimachiae Foenumgraeci Lysimachia foenum-Graecum Hance of Prinsepia. Harvesting for 9-10 months, cleaning, drying in the shade or drying at low temperature. Sweet and light in taste; the sex is flat. Has effects in clearing heat, promoting qi circulation, relieving pain, and expelling parasites. Can be used for treating common cold, headache, toothache, sore throat, fullness in chest and abdomen, and parasitosis.
Sweetsop is prepared from sweetsop: is the fruit of Annonaceae plant Annona squamosa Linn. Collected in summer and autumn, and fresh or sun-dried. Sweet in taste; it is cold in nature. The function is mainly used for tonifying spleen and stomach; clearing heat and detoxicating; killing insects. Sore pain in the main malignant sore; intestinal parasitic disease.
Betel nut: the product is a dry mature seed of Areca cathu L. Harvesting mature fruit from late spring to early autumn, decocting with water, drying, removing pericarp, taking out seed, and drying. Bitter and pungent taste; warm nature. Enter stomach and large intestine meridians. Has the functions of killing parasites, removing food retention, promoting qi circulation, promoting diuresis and preventing malaria. Can be used for treating taeniasis, ascariasis, schistosomiasis, abdominal pain due to accumulation of worms, diarrhea due to accumulation of food stagnation, tenesmus, edema, tinea pedis, and malaria.
Rhizoma Cyrtomii Falcati: is rhizome of Dryopteris shanxi of Dryopteridaceae. Collected in summer and autumn, removed from leaves after digging out, cleaned, used fresh or dried in the sun. Bitter and astringent taste; cooling; it is slightly toxic. Has the functions of clearing heat and detoxicating; cooling blood and stopping bleeding; expelling parasites; preventing influenza; brain b; measles; parotitis. Primary diarrhea; hematemesis; epistaxis blood; metrorrhagia and metrostaxis; pinworm; hookworm; intestinal parasitic diseases such as cestodes.
After duck is infected with tapeworm disease, common symptoms are inappetence, emaciation, anemia, loose feathers, slow movement, pale mucous membrane, pale green or grey-white thin feces, and tapeworm joint tablets are mixed in the feces. The section can be seen as congestion, bleeding, inflammation or ulceration of the intestinal mucosa. There are a large number of parasitic insects in the intestinal lumen, even blocking the intestinal tract.
The recipe is to kill parasites, stop bleeding, break accumulation and promote qi circulation. Therefore, the invention uses the acupuncture iron seed bitter and sour, has mild property as a monarch drug, inhibits and kills tapeworm, and simultaneously drives and kills insects; the stomach warming and insect expelling effects of the herba polygoni multiflori with the assistance of Xin Wenqi sharp are adopted, the herba lysimachiae is used for promoting qi circulation and relieving pain, relieving the abdominal pain caused by the insect accumulation, the sweetsop is used for expelling cold and detoxifying and killing insects, the betel nut is used as ministerial drugs for breaking qi and removing food retention, detoxifying and killing insects, the qi circulation and relieving pain are adopted for assisting the monarch drugs to kill insects and remove food retention, the rhizoma polygonati is used for cooling blood and detoxifying, the intestine and stopping bleeding are used together, and the effects of killing insects, removing food retention and stopping bleeding are achieved together.
The experiment of the curative effect of the traditional Chinese medicine composition on the taeniasis of ducks shows that the traditional Chinese medicine composition can greatly reduce the mortality rate of ducks with taeniasis, improve the cure rate of ducks with taeniasis and effectively reduce the cultivation loss; moreover, the prevention experiment shows that the traditional Chinese medicine composition can effectively reduce the incidence rate of the taeniasis of ducks. In conclusion, the traditional Chinese medicine composition disclosed by the invention has the advantages of definite curative effect on the taeniasis of ducks, environment friendliness, safety, no toxic or side effect and prevention and treatment, and has good market application value.
Detailed Description
The present invention will be described in further detail by way of the following specific examples, which are not intended to limit the scope of the present invention.
Example 1:
a medicine for preventing and treating taeniasis of duck is prepared from Chinese-medicinal composition and pharmaceutically-acceptable auxiliary materials. The traditional Chinese medicine composition is prepared from the following raw materials in parts by weight: 60 parts of acupuncture iron seeds, 40 parts of kola nut, 40 parts of lysimachia foenum-graecum hance, 40 parts of sweetsop, 40 parts of betel nut and 30 parts of rhizoma dryopteris crassifolia. The auxiliary material is maltodextrin.

The auxiliary material is maltodextrin.

The drug efficacy verification experiment for preventing and treating the taeniasis of the invention comprises the following steps:
in order to verify the efficacy of the medicines for preventing and treating the taeniasis of ducks, efficacy verification experiments are carried out by taking the medicines prepared in the examples 1, 2 and 3 of the invention as examples.
Meanwhile, in order to compare with the medicine for preventing and treating the duck tapeworm disease, the invention also provides the following comparison experiments, namely, the comparison examples 1 to 12; the medicines of comparative examples 1 to 12 are basically the same as that of example 1 of the present invention except that the raw materials used for the traditional Chinese medicine composition in the formulation are different, and the raw materials used for the traditional Chinese medicine composition of comparative examples 1 to 12 are specifically shown in table 1.

1. Treatment efficacy experiment:
(1) Test subjects
The test objects are 8000 Beijing ducks raised in Qiji county farms in Henan province, the age of 25 days, the symptoms of duck taeniasis appear in large groups in different degrees, the sick birds eat down, the drinking water is increased, the excrement is light green or light gray, the odor is mixed with mucus and white worm segments, the emaciation is realized, some ducks walk unstably, fall to one side suddenly, and the feet do the action of rowing water. The diseased duck is dissected to find punctate bleeding in small intestine, and a large number of insects exist in intestinal cavity, even the intestinal tract is stuffed. Faeces were collected and concentrated by the process of worm eggs Bian Shi to find taeniasis and taeniasis coronans eggs. The resident field practitioner determines a typical cestodiasis infection.
(2) Experimental drugs:
the experimental drugs were the drugs prepared in examples 1 to 3 of the present invention, the drugs prepared in comparative examples 1 to 12 and the drug prepared in comparative example 13.
(3) Test grouping and medication:
510 patients were randomly selected from the patients, and the patients were randomly divided into 17 groups (experiment 1 group to experiment 17 group) of 30 patients, and each group was individually bred and labeled one by one. The feed adopted in the feeding is a self-prepared complete feed of the premix of the Beijing yukou.
The medicines adopted in the experiment 1 group to the experiment 15 group are the medicines prepared in the example 1 to the example 3 and the comparative example 1 to the comparative example 12 respectively, and the medicine taking methods of the experiment 1 group to the experiment 15 group are as follows: 2g of the test drug was dissolved in 1L of water and administered with water for 5 days.
The medicine adopted in experiment 16 is praziquantel tablet (commercial name is Kedilil, specification is 0.2/tablet, production lot number is 220322, shenyang red flag pharmaceutical Co., ltd.) and the medicine taking method is as follows: praziquantel was administered at 20mg/kg body weight, mixed with feed, and administered on the first and third days of the trial only.
The experiment 17 group is a blank control group, and is not treated by medicines, and only the same drinking water and feed are fed.
After the treatment of the experiment 1 group to the experiment 17 group according to the administration method for 5 days, the death number and the cure number of the ducks in each group are observed and recorded, and the death rate and the cure rate are calculated (the cure standard is that the spirit, feeding, excrement and the like of the sick chickens are recovered to be normal). The administration methods and statistics of the experimental results of each experimental group are shown in table 2.

Comparing the experimental results of the experiment 1 group to the experiment 15 group, the treatment effect is obviously lower than that of the combination of a plurality of raw materials when the single raw material is used for treatment; moreover, when the six raw materials are combined, the death rate of the duck is lowest, the cure rate is highest, and the treatment effect is optimal. Therefore, the invention combines six raw materials of acupuncture iron seed, vine fruit, lysimachia foenum-graecum, sweetsop, betel nut and cyrtomium rhizome to play a role in synergy, can better play a role in disinsection and has good treatment effect.
Comparing the experimental results of the experiment 1 group with the experimental results of the experiment 16 group and the experimental results of the experiment 17 group, the insecticidal effect of the composition of the invention is better than that of praziquantel as a chemical drug compared with the drug of the example 1 and the drug of the comparative example 13. Therefore, the medicine 1 can reduce the death rate and improve the cure rate when being used for treating the tapeworm disease.
2. Prevention experiment
(1) Test subjects
The test subjects were Beijing ducks raised in the Kaiqi county farm in Henan province, from which 480 healthy ducks were selected (the selected ducks were confirmed to be in good health condition by veterinary examination 1d before the test).
(2) Experimental drugs:
the experimental drugs were the drugs prepared in examples 1 to 3 of the present invention and the drugs prepared in comparative examples 1 to 12.
(3) Test grouping and medication:
480 healthy ducks were randomly divided into 16 groups (experiment 1 group to experiment 16 group), 30 in each group, and each group was individually bred and individually labeled. The feed adopted in the feeding is a self-prepared complete feed of the premix of the Beijing yukou.
The medicines adopted in the experiment 1 group to the experiment 15 group are the medicines prepared in the example 1 to the example 3 and the comparative example 1 to the comparative example 12 respectively, and the medicine taking methods of the experiment 1 group to the experiment 15 group are as follows: 1.5g of the test drug was dissolved in 1L of water, administered with water, and administered continuously for 5 days.
Experiment 16 was a blank control group, and only the same drinking water and feed were given without medication.
On day 2 of administration, the experimental groups 1 to 16 were subjected to artificial tapeworm infection, and then the mental, appetite and fecal conditions of the ducks were observed and recorded day by day. Continuous observation was performed for 8 days after infection, and morbidity and mortality were recorded for each group of ducks. The experimental results are shown in table 3.

As can be seen from table 3, the prevention effect is poor when a single raw material medicine is adopted for prevention, and the six raw material medicines of acupuncture iron seed, kola nut, lysimachia foenum-graecum hance, sweetsop, betel nut and cyrtomium rhizome are combined, so that the prevention effect is best, and the six traditional Chinese medicine combinations can play a synergistic effect, so that the infections of taeniasis can be effectively prevented, and the morbidity and mortality can be reduced.
The experiment results show that when the taeniasis of the ducks is treated, the traditional Chinese medicine composition is continuously fed for 5 days according to the proportion of adding 2g in each liter of water, so that the death rate can be effectively reduced, and the cure rate can be improved. When preventing tapeworm disease, the tapeworm disease is continuously fed for 5 days according to the proportion of adding 1.5g in each liter of water, so that the morbidity and mortality of the tapeworm disease can be effectively prevented, the protection rate is improved, and the cultivation loss is effectively reduced.
(II) safety experiments of target animals of the medicine for preventing and treating the taeniasis of the invention:
1. test materials
(1) Test animals: 120 healthy Beijing ducks with about consistent body weight at 15 days of age. And adopting a fin number to carry out marking identification. The enrolled ducks were confirmed to be in good health by veterinary inspection 1d prior to the trial. The animals were randomized into 4 groups, 30 in each of the control, low, medium and high dose groups.
(2) Test drug and drug dosage: the test drug was the drug prepared in example 1; the recommended dosage of the traditional Chinese medicine composition for treating tapeworm disease is mixed drink: 2g of drug per 1L of water. To ensure the safety of the product for clinical use on target animals, the test is performed at recommended doses. The doses of the high dose group, the medium dose group and the low dose group are 5 times, 3 times and 1 time of the recommended doses in sequence, and the administration is continued for 5 days; the control group was not treated with the drug. The dosing regimen tested is shown in table 4.

2. Observing and detecting index
During the test period, the reaction of the ducks after administration was observed daily, and the general state of the animals such as mental state, diet, mobility, growth and the like was recorded. The body weight of each group of ducks before and after drug administration was recorded. After stopping the drug, the blood is subjected to hematological detection (white blood cell count, red blood cell count and hemoglobin content measurement), blood biochemical index measurement (aspartate aminotransferase, alanine aminotransferase, albumin, total cholesterol, creatinine and the like), organ pathological section examination and the like.
3. Experimental results
(1) Clinical manifestations
Throughout the trial, each group did not die and no apparent abnormal activity of the animals associated with the test agent occurred. The mental state, activity, drinking water, collecting feces and the like of the ducks in all dosage groups are normal.
(2) Weight measurement results
The average weight results of the ducks in each group before and after the test are shown in Table 5.

As can be seen from table 5, after the end of the administration (20 days of age), there was no significant difference in the average body weight of each dose group from that of the blank group.
(3) Blood index detection result
After stopping the drug, the hematological index of each group of ducks is evaluated, namely, the red blood cells, the white blood cells, the hemoglobin and other items in blood are detected, and the results are shown in table 6.

As can be seen from table 6, there was no significant difference in the number of leukocytes and hemoglobin in each dose group compared to the blank group. The traditional Chinese medicine composition has little influence on the hematological index of ducks.
(4) Blood biochemical index detection result
After stopping the drug, each blood biochemical index of each group of ducks is detected and is subjected to statistical analysis, and the results are shown in table 7.

As can be seen from table 7, there was no significant difference in the levels of aspartic aminotransferase, alanine aminotransferase, creatinine, serum albumin, and total bilirubin among the high, medium, and low dose groups of ducks as compared to the blank control group. The traditional Chinese medicine composition has little influence on biochemical indexes of duck blood.
(5) Histopathological examination
After drug withdrawal, the animals were visually examined for heart, liver, spleen, lung and kidney without abnormal changes. At the same time, the main viscera are subjected to pathological histology examination. The results show that the heart, liver, spleen, lung, kidney of each dose group also showed no histopathological changes associated with the toxic effects of the test drug as compared to the control group. The traditional Chinese medicine composition is safe in clinical application to ducks.
