Abstract
The invention discloses a traditional Chinese medicine composition for preventing and treating poultry cold, which comprises the following raw materials: 8-12 parts of dandelion, 8-12 parts of red peony root, 15-25 parts of houttuynia cordata, 15-25 parts of bighead atractylodes rhizome, 10-20 parts of poria cocos, 8-12 parts of Chinese violet, 15-25 parts of isatis root, 10-20 parts of fructus forsythiae, 5-15 parts of rhizoma anemarrhenae, 10-20 parts of ash bark, 10-20 parts of kudzu root and 8-12 parts of liquorice. The invention adopts a plurality of traditional Chinese medicines, and compounds the traditional Chinese medicines into a compound preparation after a special mixing and extracting process, thereby having unique efficacy, improving the body resistance of poultry and enhancing the capability of the body to resist pathogenic diseases; meanwhile, the feed additive has the effects of diminishing inflammation, sterilizing, resisting viruses and resisting allergic reaction, and can enhance the immunity of poultry, repair damaged mucous membranes and effectively control secondary infection. The raw materials in the traditional Chinese medicine composition are coordinated with each other to exert the drug effect to the maximum extent, and clinical application proves that the traditional Chinese medicine composition has a very good treatment effect on poultry anemofrigid cold and anemofrigid cold.

The invention relates to the technical field of medicines, and particularly relates to a traditional Chinese medicine composition for preventing and treating poultry cold.
Background
The common cold of poultry is an exogenous disease caused by the repeated affection of pathogenic wind, cold, dryness, summer-heat, dampness, fire (warm, hot) or epidemic toxin under the condition of insufficient vital qi of the poultry body. The poultry feed commonly suffers from adverse conditions such as climate change, immunization, poor feed quality and poor feeding management, is a common disease of poultry respiratory tract, is easy to be infected with bacterial diseases or secondarily infected with bacterial diseases, can cause serious consequences, and is one of the main diseases affecting the production benefit of poultry.
According to the theory of Chinese veterinarian, the common cold of poultry is divided into wind-cold common cold and wind-heat common cold, wherein the wind-cold common cold is frequently generated in alternative seasons of winter and autumn and winter, and the symptoms of fever at the upper part of the poultry, cold lower limbs, aversion to cold and aversion to wind, depression, unsmooth movement and stuffy respiratory symptoms are seen, wherein the mental state is particularly not vibrated when the temperature is low in the morning and evening; as yang is injured, the early stage of the disease is not thirsty and does not greed water, the feed intake is not obviously reduced, the trachea can be seen bright by the caesarean section of the dead poultry, only a small amount of primary blood is left at the bifurcation, the blood is more bright, a small amount of transparent foam-like secretion exists in the trachea cavity, and renal type bronchiectasis and the like can be caused. Because the cold is classified into wind-cold type and wind-heat type, the treatment methods are different, and wind-cold type cold is treated with pungent and warm drugs, such as Jingfang Baidu san; cold-natured drugs for wind-heat type common cold, such as Shuanghuanglian oral liquid, etc., can reduce or discharge heat evil out of the body. For wind-cold syndrome, misuse of cold drugs can easily damage yang qi of poultry; when the heat-clearing agent is applied by mistake to the patient with wind-heat, it will consume the body fluids and even cause other diseases. Therefore, veterinary medicines used for preventing and treating poultry cold generally need to be treated by differentiation of symptoms and signs, and few medicines with treatment effects on poultry cold due to wind-cold and wind-heat cold are reported at present.
Disclosure of Invention
Aiming at the prior art, the inventor combines the clinical experience of the inventor for many years, starts from the therapeutic principle of veterinary medicines for treating wind-cold type common cold and wind-heat type common cold, and prepares the traditional Chinese medicine composition with the therapeutic effect on the wind-cold type common cold and the wind-heat type common cold of poultry by appropriately processing 12 raw medicines of dandelion, red paeony root, houttuynia cordata, bighead atractylodes rhizome, poria cocos, Chinese violet, isatis root, weeping forsythia, common anemarrhena rhizome, ash bark, kudzuvine root and liquorice.

The compatibility of the Chinese medicines is characterized by mutual reinforcement, mutual assistance, mutual incompatibility, mutual killing, mutual aversion and contradistinction, and different compatibility relations exist between different Chinese medicines, so that when more than two Chinese medicines are used, the compatibility relations between the mutual reinforcement and mutual assistance between the Chinese medicines must be selected, and the clinical effect of the product is obviously improved.
In the traditional Chinese medicine composition for preventing and treating the poultry cold, all raw material components are organic whole, and one is not essential. In the research and development process, the inventor finds that the overall action effect of the traditional Chinese medicine composition is obviously reduced by reducing any one raw material component in the composition or replacing the raw material components of the traditional Chinese medicine composition by the raw material medicines with similar medicine properties; other raw material components are added on the basis of the traditional Chinese medicine composition, so that the overall effect of the traditional Chinese medicine composition is not obviously improved, and even the overall effect of the traditional Chinese medicine composition is reduced.
The preparation method of the traditional Chinese medicine composition for preventing and treating the poultry cold can crush the raw materials and uniformly mix the raw materials for use; or decocting in water; or prepared according to other methods.
In a preferred embodiment of the present invention, the preparation method of the traditional Chinese medicine composition for preventing and treating poultry cold comprises:
(1) pulverizing herba Taraxaci and radix Paeoniae Rubra, mixing, adding 10-15 weight times of water, heating to 70-90 deg.C, keeping warm, leaching for 1-2 hr, extracting for 2-3 times, filtering, discarding residue, and mixing filtrates; after rough filtration, the filtrate is separated into effective fractions with molecular weight of 5000-;
(2) Pulverizing herba Violae, rhizoma anemarrhenae and cortex Fraxini, mixing, and subjecting to supercritical CO2Extracting under 25-30MPa at 30-40 deg.C for 2-4h with 95% (volume fraction) ethanol as entrainer in an amount of 8-12% of the total weight of the raw materials to obtain extract B;
(3) pulverizing herba Houttuyniae, Atractylodis rhizoma, Poria, radix Isatidis, fructus forsythiae, radix Puerariae and Glycyrrhrizae radix, mixing, adding 8-12 times of water, decocting for 2-4 times, and mixing decoctions to obtain extract C;
(4) mixing extract A, extract B and extract C, and drying.
Preferably, in the step (4), the drying is freeze drying.
The traditional Chinese medicine components are complex, one medicinal material contains water-soluble components and oil-soluble or alcohol-soluble components, the pharmacological activities of different extraction parts are different, and some active components are easy to inactivate at high temperature, so that the effective components of the traditional Chinese medicine can be extracted and separated by selecting a proper extraction method and extraction process, the effective components can be extracted to the maximum extent, the activity of the effective components can be ensured, and the clinical effect of the product can be ensured. In addition, the mixed extraction of various Chinese medicines by adopting a proper method can increase the solubility of the effective components of the medicines, eliminate or weaken the toxicity of certain medicines, generate new substances and play a new curative effect.
Tests prove that by adopting the optimal preparation method of the invention, the raw material medicines in the traditional Chinese medicine composition are classified and mixed and extracted by adopting different extraction methods, the active ingredients in the traditional Chinese medicine composition can be extracted to the maximum extent, the activity of the active ingredients is ensured, and ineffective and even harmful ingredients are removed, so that the clinical effect of the product is ensured.
The application of the traditional Chinese medicine composition in preparing the feed additive for preventing and treating the poultry cold is also within the protection scope of the invention.
In the application, the specific application method comprises the following steps: every 1g of the traditional Chinese medicine composition is mixed with 500g of the material for continuous use for 3-5 days.
The invention also protects the application of the traditional Chinese medicine composition in preparing a medicinal preparation for preventing and treating the poultry cold.
In a third aspect of the invention, the invention provides a pharmaceutical preparation for preventing and treating poultry cold, which is prepared from the traditional Chinese medicine composition and pharmaceutically acceptable auxiliary materials.
The medicinal preparation can be made into various dosage forms, such as powder, solution, soluble powder or injection.
The pharmaceutically acceptable auxiliary materials are conventional veterinary auxiliary materials, such as: sweetening agent, disintegrating agent, lubricant, adhesive, etc. Preferably one or more of cyclodextrin, starch, sodium carboxymethyl cellulose, and maltodextrin.
The invention has the beneficial effects that:
on the basis of carrying out a large amount of researches on clinically frequent poultry wind-cold type common cold and wind-heat type common cold, the traditional Chinese medicines and the pharmacodynamic characteristics of the effective components of the traditional Chinese medicines are combined, pathogenic factors are considered comprehensively, a plurality of traditional Chinese medicines are adopted and are compounded into a compound preparation through a special mixing and extracting process, so that the compound preparation has a unique effect, the body resistance of the poultry can be improved, and the pathogenic disease resistance of the poultry can be improved; meanwhile, the feed additive has the effects of diminishing inflammation, sterilizing, resisting viruses and resisting allergic reaction, and can enhance the immunity of poultry, repair damaged mucous membranes and effectively control secondary infection. The raw materials in the traditional Chinese medicine composition are coordinated with each other to exert the drug effect to the maximum extent, and clinical application proves that the traditional Chinese medicine composition has a very good treatment effect on poultry anemofrigid cold and anemofrigid cold.

The traditional Chinese medicine has complex ingredients, and particularly for the traditional Chinese medicine compound, the achievement of the drug effect is realized through the interaction among a plurality of effective ingredients. The effective components of the same raw material medicine are obtained by different extraction methods, and the medicine effects of the effective components are different. In order to further improve the overall curative effect of the traditional Chinese medicine composition for preventing and treating the poultry cold, the raw material medicines in the traditional Chinese medicine composition are respectively extracted by different methods, so that ineffective impurities are removed as far as possible to purify the preparation, and the synergistic effect among different extracts is exerted. Multiple tests prove that the dandelion and the red paeony root are extracted by adding water, and effective fractions with the molecular weight of 5000-; subjecting herba Violae, rhizoma anemarrhenae and cortex Fraxini to supercritical CO2Extracting to obtain effective components; decocting herba Houttuyniae, Atractylodis rhizoma, Poria, radix Isatidis, fructus forsythiae, radix Puerariae and Glycyrrhrizae radix with water to obtain effective components; the three effective components are mixed again, so that the effect of preventing and treating the cold of the poultry can be exerted to the maximum extent.
In order to make the technical solutions of the present application more clearly understood by those skilled in the art, the technical solutions of the present application will be described in detail below with reference to specific embodiments.
The test materials used in the examples and comparative examples of the present invention are conventional in the art and are commercially available.
Example 1:

(1) pulverizing herba Taraxaci and radix Paeoniae Rubra, mixing, adding 12 weight times of water (the amount of water is 12 weight times of herba Taraxaci and radix Paeoniae Rubra), heating to 80 deg.C, keeping the temperature, leaching for 2 hr, extracting for 2 times, filtering, removing residue, and mixing filtrates; after rough filtration, the filtrate is separated into effective fractions with molecular weight of 5000-;
(2) pulverizing herba Violae, rhizoma anemarrhenae and cortex Fraxini, mixing, and subjecting to supercritical CO2Extracting under 28MPa at 35 deg.C for 3h with 95% (volume fraction) ethanol as entrainer, wherein the entrainer accounts for 10% of the total weight of the raw materials (i.e. the entrainer is added in an amount of 10% of the total weight of herba Violae, rhizoma anemarrhenae and cortex Fraxini), to obtain extract B;
(3) pulverizing herba Houttuyniae, Atractylodis rhizoma, Poria, radix Isatidis, fructus forsythiae, radix Puerariae and Glycyrrhrizae radix, mixing, adding 10 weight times of water, decocting for 3 times, and mixing decoctions to obtain extract C;
(4) mixing the extract A, the extract B and the extract C, and freeze-drying to obtain the traditional Chinese medicine composition for preventing and treating the poultry wind-cold type common cold.
Example 2:
the raw material medicine comprises the following components:
8 g of dandelion, 12 g of red peony root, 15 g of houttuynia cordata, 25 g of bighead atractylodes rhizome, 10 g of poria cocos, 12 g of Chinese violet, 15 g of isatis root, 20 g of forsythia, 5 g of rhizoma anemarrhenae, 20 g of ash bark, 10 g of kudzu root and 12 g of liquorice.
The preparation method is the same as example 1.
Example 3:
the raw material medicine comprises the following components:
12 g of dandelion, 8 g of red peony root, 25 g of houttuynia cordata, 15 g of bighead atractylodes rhizome, 20 g of poria cocos, 8 g of Chinese violet, 25 g of isatis root, 10 g of forsythia, 15 g of rhizoma anemarrhenae, 10 g of ash bark, 20 g of kudzu root and 8 g of liquorice.
The preparation method is the same as example 1.
Comparative example 1:
in the research and development process, the compatibility of a plurality of raw material medicines with similar medicinal properties and the research on medicinal effects are carried out, and the result shows that even if the medicinal properties of a certain raw material medicine are similar, the difference of the medicinal effects of the raw material medicine is obvious after the raw material medicine is matched with other different raw material medicines for use. (3) pulverizing herba Houttuyniae, Atractylodis rhizoma, Poria, radix Isatidis, fructus forsythiae, radix Puerariae and Glycyrrhrizae radix, mixing, adding 10 weight times of water, decocting for 3 times, and mixing decoctions to obtain extract C;
(4) mixing the extract A, the extract B and the extract C, and freeze-drying to obtain the traditional Chinese medicine composition A.
Comparative example 2:
in the research and development process, the invention also carries out addition, subtraction, optimization and investigation on the composition types of the raw material medicines in the formula. For example: the red peony root, the rhizoma anemarrhenae and the ash bark in the formula of the medicine in the embodiment 1 are omitted, the rest is the same as the embodiment 1, and the traditional Chinese medicine composition B is prepared according to the method in the embodiment 1.
For another example, 3 kinds of bulk drugs of reed rhizome, chrysanthemum and spina date seed are added on the basis of the drug formula in the example 1, the dosage of the bulk drugs refers to the dosage in the pharmacopoeia, the added 3 kinds of bulk drugs are decocted with 7 kinds of bulk drugs of houttuynia cordata and the like in water, and the traditional Chinese medicine composition C is prepared according to the method in the example 1.
Comparative example 3:
the dosage of each raw material medicine in the traditional Chinese medicine composition is marked, even if the medicine composition of the prescription is the same, if the dosage of the medicine is different, the efficacy of the medicine is divided into different magnitudes, and the compatibility relationship is changed by monarch, minister, assistant and guide, so that the efficacy is different. In the research and development process, the content of each raw material in the traditional Chinese medicine composition is investigated and the drug effect is researched, and the result shows that the difference of the drug effect is obvious due to the improper mixture ratio of the raw materials. For example: doubling the weight parts of the dandelion, the bighead atractylodes rhizome, the Chinese violet and the forsythia in the embodiment 1, and halving the weight parts of the red paeony root, the anemarrhena and the ash bark; the other raw materials were used in the same amounts as in example 1, and the preparation method of example 1 was followed to prepare a Chinese medicinal composition D.
Comparative example 4:
the traditional Chinese medicine has very complex components, contains various active ingredients, ineffective ingredients and no toxic ingredients, and has obvious difference in drug effect of traditional Chinese medicine compound preparations prepared by different methods. In the research and development process, the preparation method of the traditional Chinese medicine composition is researched, and the result shows that the traditional Chinese medicine compositions prepared by different methods have obvious difference in efficacy and effect even if the raw materials adopted by the traditional Chinese medicine compositions are consistent. For example, the preparation method of the Chinese medicinal composition in example 1 is adjusted to: weighing the raw materials according to the parts by weight, crushing, mixing, adding 10 times of water by weight, decocting for 3 times, combining decoction, concentrating, drying, and preparing into the traditional Chinese medicine composition E.
Test example 1: safety determination of traditional Chinese medicine composition on chick embryo
The Chinese medicinal compositions prepared in examples 1 to 3 were prepared into solutions with a concentration of 1g/ml using sterile physiological saline, respectively, and measured by the method of inoculating the compositions into allantoic cavities of chicken embryos. The method comprises the following specific steps:
diluting the Chinese medicinal composition with normal saline by 1, 5, 10, 50 and 100 times, respectively injecting into allantoic cavity of SPF chick embryo of 9 days old at a dose of 0.2 ml/body, incubating in a constant temperature incubator at 37 deg.C, injecting for 1 time at an interval of 12h, continuously incubating for 10 chick embryos per group, observing the state of chick embryo every day, discarding the chick embryo dead in 24h, taking out the chick embryo dead in 25-72h, freezing in a refrigerator at 4 deg.C for 6h, and dissecting to observe the development and death status of chick embryo. The live embryos are continued to be hatched until hatching. The maximum concentration which is safe to the chick embryos and does not cause death is taken as the minimum lethal concentration, and the safety of the traditional Chinese medicine composition to the chick embryos is determined according to the minimum lethal concentration.
As a result, it was found that none of the chick embryos were killed by inoculating the Chinese medicinal composition prepared in examples 1 to 3, and thus the Chinese medicinal composition of the present invention was safe to the chick embryos.
Test example 2:
the Chinese medicinal compositions prepared in example 1 and comparative examples 1 to 4 were prepared into solutions having a concentration of 1mg/ml with sterile water, respectively.
2 x 10 to2EID50Inoculating 0.1ml of influenza virus strain to 9-day-old SPF chick embryos, inoculating sterile normal saline to a blank control group, incubating for 4 hours at 37 ℃, and then respectively inoculating the prepared solutions, inoculating the solution prepared from the traditional Chinese medicine composition in the embodiment 1 to the test group, respectively inoculating the solution prepared from the traditional Chinese medicine composition A to the traditional Chinese medicine composition E to the control group 1 to the control group 5, and inoculating 0.1ml of influenza virus strain to each embryo; equivalent sterile normal saline is inoculated in the challenge control group; incubating at 37 ℃, discarding dead chick embryos within 24h, photographing eggs every 6h for 5 days, observing and recording the dead chick embryos time each time, and counting the protection rate of each group of chick embryos. The test results are shown in Table 1.

As can be seen from table 1, the protective effect of the traditional Chinese medicine composition of example 1 of the present invention on the SPF chick embryos infected with influenza virus is significantly better than that of comparative example 1 (traditional Chinese medicine composition a). The composition conforms to the compatibility of the traditional Chinese medicines, has a synergistic effect among the components, and is not simple replacement and combination of the raw material medicines with similar medicinal properties.
As can be seen from the comparison between the traditional Chinese medicine composition prepared in example 1 and the traditional Chinese medicine compositions B to D, on the basis of the medicine formula in example 1, the protective effect of the traditional Chinese medicine composition on the SPF chick embryos infected with influenza virus can be reduced no matter whether the types of the raw material medicines are increased or decreased, or the dosage of each raw material medicine in the traditional Chinese medicine composition in example 1 is adjusted.
Compared with the traditional Chinese medicine composition E prepared in the embodiment 1, the preparation method has a very key effect on the efficacy of the prepared traditional Chinese medicine composition, and the traditional Chinese medicine composition prepared by the preparation method can remarkably improve the protection effect on the SPF chick embryos infected with influenza viruses.
Test example 3:
in order to verify the treatment effect of the traditional Chinese medicine composition on the wind-cold type common cold of the poultry, a chicken which has the wind-cold type common cold in a certain chicken farm is selected for testing.
1. The test method comprises the following steps:
400 chickens suffering from wind-cold type common cold (symptoms: upper part fever of poultry, lower limb coldness, aversion to cold and aversion to wind, mental depression, unsmooth movement and respiratory tract symptoms, wherein the mental state is particularly not vibrated when the temperature is low in the morning and evening) are randomly and averagely divided into a test 1 group, a test 2 group, a test 3 group and a control group, wherein the test 1 group to the test 3 group respectively take the traditional Chinese medicine compositions prepared in the examples 1-3, and the traditional Chinese medicine compositions are respectively taken according to 500g of mixed materials per 1 g; the control group was administered with the powder of JING FANG BAI DU, which is commercially available, according to the instruction. and (3) curing: the chicken has good spirit and good appetite, the lower limb body temperature returns to normal, and the symptoms of cough, sneezing and head swinging disappear;
the method has the following advantages: the symptoms are obviously relieved, the spirit is improved, the appetite is better, and the symptoms of cough, sneeze and head swing are occasionally caused;
and (4) invalidation: the symptoms are not obviously improved, and some symptoms even aggravate.

In the test process, the chickens in the test 1 group and the test 2 group do not die, the mortality rate of the chickens in the test 3 group is 1%, and the mortality rate of the chickens in the control group is 5%.
Test example 4:
in order to verify the treatment effect of the traditional Chinese medicine composition on the wind-heat type common cold of the poultry, a chicken which has the wind-heat type common cold in a certain chicken farm is selected for testing.
1. The test method comprises the following steps:
400 chickens suffering from wind-heat type common cold (symptoms: poultry is fever but not aversion to cold, the body is relatively stretched, the water intake is increased at the initial stage due to yin injury, the sick poultry mostly show dehydration symptoms, and the food intake is reduced after the disease is generally attacked for 1-2 d) are randomly and evenly divided into a test 1 group, a test 2 group, a test 3 group and a control group, wherein the test 1 group to the test 3 group respectively take the traditional Chinese medicine compositions prepared in the examples 1-3, and the traditional Chinese medicine compositions are respectively taken according to 500g of mixed materials per 1 g; the control group was administered with the commercially available SHUANGHUANGLIAN oral liquid according to the instruction. All the above groups were used together for 3 days.
2. The therapeutic effect judgment standard is as follows:

and (4) invalidation: the symptoms are not obviously improved, and some symptoms even aggravate.

In the test process, the death rate of the chicken in the test 1 group is not increased, the death rate of the chicken in the test 2 group and the chicken in the test 3 group is 1 percent, and the death rate of the chicken in the control group is 4 percent.

1. The traditional Chinese medicine composition for preventing and treating the poultry cold is characterized in that the poultry cold is wind-cold type cold or wind-heat type cold and comprises the following raw materials:
8-12 parts of dandelion, 8-12 parts of red peony root, 15-25 parts of houttuynia cordata, 15-25 parts of bighead atractylodes rhizome, 10-20 parts of poria cocos, 8-12 parts of Chinese violet, 15-25 parts of isatis root, 10-20 parts of fructus forsythiae, 5-15 parts of rhizoma anemarrhenae, 10-20 parts of ash bark, 10-20 parts of kudzu root and 8-12 parts of liquorice;
the traditional Chinese medicine composition is prepared by the following method:
(1) pulverizing herba Taraxaci and radix Paeoniae Rubra, mixing, adding 10-15 weight times of water, heating to 70-90 deg.C, keeping warm, leaching for 1-2 hr, extracting for 2-3 times, filtering, discarding residue, and mixing filtrates; after rough filtration, the filtrate is separated into effective fractions with molecular weight of 5000-;
(2) pulverizing herba Violae, rhizoma anemarrhenae and cortex Fraxini, mixing, and subjecting to supercritical CO2Extracting under 25-30MPa at 30-40 deg.C for 2-4h with 95 vol% ethanol as entrainer in an amount of 8-12 wt% of the raw materials to obtain extract B;
(3) pulverizing herba Houttuyniae, Atractylodis rhizoma, Poria, radix Isatidis, fructus forsythiae, radix Puerariae and Glycyrrhrizae radix, mixing, adding 8-12 times of water, decocting for 2-4 times, and mixing decoctions to obtain extract C;
(4) mixing extract A, extract B and extract C, and drying.
2. The traditional Chinese medicine composition according to claim 1, which is characterized by comprising the following raw materials:
10 parts of dandelion, 10 parts of red peony root, 20 parts of houttuynia cordata, 20 parts of bighead atractylodes rhizome, 15 parts of poria cocos, 10 parts of Chinese violet, 20 parts of isatis root, 15 parts of forsythia, 10 parts of rhizoma anemarrhenae, 15 parts of ash bark, 15 parts of kudzu root and 10 parts of liquorice.
3. Use of the Chinese medicinal composition of claim 1 or 2 in the preparation of feed additive for preventing and treating poultry cold.
4. Use of the Chinese medicinal composition of claim 1 or 2 for the preparation of a pharmaceutical preparation for preventing and treating poultry cold.
5. A pharmaceutical preparation for preventing and treating poultry cold, which is characterized by being prepared from the traditional Chinese medicine composition of claim 1 or 2 and pharmaceutically acceptable auxiliary materials.
6. The pharmaceutical formulation of claim 5, wherein the pharmaceutically acceptable excipient is a disintegrant, a lubricant, a sweetener, or a binder.
