Abstract
本发明涉及结合到人类白细胞抗原(HLA)I类或II类分子上在免疫治疗方法中用作疫苗的肿瘤相关肽新制剂。特别地,本发明涉及用于癌症免疫治疗,特别是用于肾癌、脑癌、神经胶质瘤,尤其是胶质母细胞瘤的制剂。本发明还涉及用于引起抗肿瘤免疫反应的疫苗组合物。
Description
技术领域
本发明涉及结合到人类白细胞抗原(HLA)Ⅰ类或Ⅱ类分子上在免疫治疗方法中用作疫苗的肿瘤相关肽的新制剂。特别是,本发明涉及用于癌症免疫治疗,特别是用于肾癌的制剂。本发明还涉及用于引起抗肿瘤免疫反应的疫苗组合物。
为了本发明的目的,本文引用的所有参考资料均通过引用作为整体纳入本发明。
背景技术
WO2007/028573专利中公开了一种注射用组合物,其由粉末状的肽和一种由碳酸氢钠组成的稀释剂构成,用作肾细胞癌疫苗。然而,该制剂具有多个缺点。最主要的缺点是其溶解性差,导致其在任何应用期间很难使用,比如临床使用。
为了溶解该组合物中的粉末,需要将装有待溶解肽和稀释剂的小瓶用力摇动3分钟,然后超声匀浆1分钟,再摇动1分钟。但并非所有的内科医生都能在其诊所中使用该治疗药物,因为他们通常缺少所需的设备,因而压缩的混合物可能不够均匀,不能进行有效使用。
该组合物的另一个问题在于,考虑到有效成分溶解于给定浓度的碳酸氢钠中的速度,所得到的溶液只能使用约30分钟。
此外,上述制剂的性质随时间而变化,使得安全使用几乎不可能。所公开的该制剂在-20℃到+25℃的不同温度下储存12个月后,即使超声匀浆几分钟,也得不到澄清溶液。
因此,需要溶解性和冻干粉润湿性均增强的新型肽制剂,以便提供一种安全有效的制剂,尤其是抗癌疫苗这种情况。本发明可满足这一需求。
发明内容
在本发明的一个优选实施例,本发明提供药物组合物,其包含的肿瘤相关肽数目在2至18个之间,优选为小于15个,更优选为小于13个,进而更优选为2至12个,还要更优选为3至12个;其中每个肽的长度为8至22个氨基酸,优选为9至16个氨基酸;其中所述肽在90%乙酸中的溶解度至少为2.7mg/mL;该药物组合物还包括甘露醇和泊洛沙姆188,其中所述肽:甘露醇:泊洛沙姆188的重量比范围在1:5:1.5到1:8:2.2之间(含);或该药物组合物还包括甘露醇和吐温其中肽:甘露醇:吐温的重量比范围在1:2:1.5到1:8:2.2之间(含)。
在本发明的另一个优选实施例,本发明提供一种如权利要求1所述的药物组合物,其含有至少两个肽,其中所述肽包括一个氨基酸序列,其选自由SEQIDNO:1到SEQIDNO:10,或SEQIDNO:12到SEQIDNO:23构成的组;但要求该组合物包括至少一个含有SEQIDNO:1、SEQIDNO:2、SEQIDNO:12或SEQIDNO:13或其变体或盐的肽;并还包括甘露醇和泊洛沙姆188,其中肽与甘露醇和泊洛沙姆188的重量比范围在1:5:1.5到1:8:2.2之间(含)。
在本发明的又一个优选实施例,本发明提供一种如权利要求1所述的药物组合物,其含有至少两个肽,其中所述肽包括一个氨基酸序列,其选自由SEQIDNO:1到SEQIDNO:10,或SEQIDNO:12到SEQIDNO:23构成的组;但要求该组合物包括至少一个含有SEQIDNO:1、SEQIDNO:2、SEQIDNO:12或SEQIDNO:13或其变体或盐的肽;并还包括甘露醇和吐温其中肽与甘露醇和吐温的重量比范围在1:2:1.5到1:8:2.2之间(含)。
有利地,根据本发明的制剂适合于含有疏水肽(例如IMA-CHI-001;SEQIDNO:23)的稳定制剂。例如,该肽极难溶于水,仅微溶于90%乙酸(溶解度约2.9mg/mL)。惊奇的是,现在已经能够将根据SEQIDNO:23的肽与另一种可用于生物的制剂中的任何一种肽结合制成制剂。
因此本发明申请公开了一种制剂,配制的肽数目为2至18个之间,优选为小于15个,更优选为小于13个,进而更优选为2至12个,还要更优选为3至12个,肽长度为8至22个氨基酸,优选为9至16个氨基酸;但要求肽在90%乙酸中的溶解度至少为2.7mg/mL,优选为2.9mg/mL,并进一步包括甘露醇和泊洛沙姆188,其中肽与甘露醇和泊洛沙姆188的重量比范围在1:5:1.5到1:8:2.2之间(含)。
在本发明的药物组合物的一个优选实施例中,肽与甘露醇和泊洛沙姆188的重量比范围在1:0:2到1:0:2.2之间(含)。
本发明的另一个优选实施例提供一种如上的药物组合物,其含有至少两个肽,其中所述肽包括一个氨基酸序列,其选自由SEQIDNO:12到SEQIDNO:23组成的组;但要求该组合物包括至少一个含有SEQIDNO:12或SEQIDNO:13或其变体或盐的肽;并进一步包括甘露醇和吐温其中肽与甘露醇和吐温的重量比范围在1:5:0.5到1:5:2之间(含)。
优选地,本发明肽的总长度为9至16个氨基酸。肽可能含有非肽键。
在其它优选实施例中,该药物组合物包括由SEQIDNO:1和/或SEQIDNO:2的氨基酸序列组成的肽,以及至少一个由SEQIDNO:3至SEQIDNO:10中的任意一个氨基酸序列组成的肽。
在一些优选实施例中,该药物组合物可进一步包括一个含有SEQIDNO:11氨基酸序列的肽,条件是所述肽不是相对全长肿瘤相关多肽。在其它实施例中,该药物组合物可能还包括由SEQIDNO:11氨基酸序列组成的肽。
在其它优选实施例中,该药物组合物包括由SEQIDNO:12和/或SEQIDNO:13的氨基酸序列组成的肽,以及至少一个由SEQIDNO:14至SEQIDNO:23中的任意一个氨基酸序列组成的肽。在一些优选实施例中,该药物组合物可进一步包括含有SEQIDNO:11氨基酸序列的肽,但要求所述肽不是相对全长肿瘤相关多肽。在其它实施例中,该药物组合物可进一步包括由SEQIDNO:11氨基酸序列组成的肽。
在某些实施例中,该组合物中的肽选自对肿瘤、癌症和/或患者具有特异性的肽。
在又一些优选实施例中,该药物组合物可进一步包括至少一种合适的佐剂。
佐剂能非特异性地提高或增强免疫反应(例如,由CTLs和辅助细胞-T(TH)介导的对于抗原的免疫反应),因此认为其在本发明药剂中是有用的。合适的佐剂包括,但不限于,1018ISS、铝盐、Amplivax、AS15、BCG、CP-870,893、CpG7909、CyaA、dSLIM、鞭毛蛋白或鞭毛蛋白TLR5配体、FLT3配体、GM-CSF、IC30、IC31、咪喹莫特Imiquimod(ALDARA)、瑞喹莫特resimiquimod、ImuFactIMP321、IL-2、IL-13、IL-21的白细胞介素、干扰素-α或干扰素-β或其聚乙二醇化衍生物、ISPatch、ISS、ISCOMATRIX、ISCOMs、JuvImmune、LipoVac、MALP2、MF59、单磷酰基脂A、MontanideIMS1312、MontanideISA206、MontanideISA50V、MontanideISA-51、油包水和水包油乳化剂、OK-432、OM-174、OM-197-MP-EC、ONTAK、OspA、载体系统、PLG和葡聚糖微粒、雷西莫特、SRL172、病毒体和其它病毒样颗粒、YF-17D、VEGF阱、R848、β-葡聚糖、Pam3Cys、来源于皂苷、分歧杆菌提取物和合成细菌细胞壁模仿物的Aquila'sQS21刺激子、以及其它特许专卖佐剂,如Ribi'sDetox、Quil或Superfos。优选的佐剂如Freund's或GM-CSF。一些对树突状细胞及其制剂具有特异性的免疫佐剂之前已有报道(DupuisM等人,1998年;Allison,1998年)。也可使用细胞因子。一些细胞因子直接与下列行为相关:影响树突状细胞至淋巴组织(如TNF-)的迁移,加速树突状细胞的成熟使其成为T-淋巴细胞(如GM-CSF,IL-1和IL-4)的有效抗原呈递细胞(美国专利No.5,849,589,此处通过引用特别将其整体纳入本文),发挥免疫佐剂的作用(如IL-12、IL-15、IL-23、IL-7、IFN-alpha、IFN-beta)(Gabrilovich等人,1996年)。
有报道CpG免疫刺激性寡核苷酸可加强疫苗中佐剂的作用。不受理论的限制,CpG寡核苷酸通过Toll样受体(TLR)(主要是TLR9)来活化先天(非适应性)免疫系统。CpG触发的TLR9活化增强了针对各种抗原的抗原特异性体液和细胞反应,包括肽或蛋白抗原、活病毒或死病毒、树突状细胞疫苗、自身细胞疫苗以及预防和治疗疫苗中的多糖缀合物。更重要的是,它增强了树突状细胞的成熟和分化,从而在缺少CD4T细胞的辅助下,也能增强TH1细胞的活化和产生强细胞毒性的T-淋巴细胞(CTL)。甚至当通常促进TH2偏向的疫苗佐剂(如明矾或弗氏不完全佐剂(IFA))存在时,也保持由TLR9刺激诱导的TH1偏向。当与其它佐剂一起配制或给药时,或用在微粒、纳米粒子、脂肪乳剂或类似制剂中时,CpG寡核苷酸显示更强的佐剂活性,当抗原相对较弱时,上述制剂对诱导强反应尤其必要。它们也可加速免疫反应,并使得抗原剂量降低约两个数量级,在一些实验中,抗体反应可与无CpG的全剂量疫苗相比(Krieg等人,2006年)。美国专利No.6,406,705B1描述了联合使用CpG寡核苷酸、非核酸佐剂和一种抗原来诱导抗原特异性的免疫反应。Mologen(德国柏林)生产的dSLIM(双干环免疫调节剂)是一种CpGTLR9拮抗剂,它是本发明的药物组合物的优选组分。也可使用其它TLR结合分子,如结合RNA的TLR7、TLR8和/或TLR9。
其它有用的佐剂包括,但不限于,化学改性的CpGs(如CpR、Idera)、dsRNA类似物如Poly(I:C)和AmpliGen、非CpG细菌DNA或RNA、以及免疫活性小分子和抗体,如环磷酰胺、舒尼替尼、贝伐单抗、西乐葆、NCX-4016、西地那非、他达那非、伐地那非、索拉非尼、替莫唑胺、西罗莫司脂化物、XL-999、CP-547632、帕唑帕尼、VEGFTrap、ZD2171、AZD2171、抗-CTLA4和SC58175,其可发挥治疗作用和/或用作佐剂。本发明中佐剂和添加剂的有用量和浓度可由本领域技术人员不需要太多的实验即可确定。优选的佐剂为dSLIM、干扰素-α、干扰素-β、CpG7909、IC31、咪喹莫特、瑞喹莫特、PeviTer、RNA、他达那非、替莫唑胺和JuvImmune。
优选的佐剂为dSLIM、BCG、OK432、咪喹莫特、瑞喹莫特、GMCSF、干扰素-α、PeviTer和JuvImmune或以上的组合。
在本发明的药物组合物的一个优选实施例中,佐剂选自由集落刺激因子构成的组,如粒细胞-巨噬细胞集落刺激因子(GM-CSF、沙格司亭)、咪喹莫特、瑞喹莫特和干扰素-α。
在本发明的药物组合物的一个优选实施例中,佐剂选自由集落刺激因子构成的组,如粒细胞-巨噬细胞集落刺激因子(GM-CSF、沙格司亭)、咪喹莫特和瑞喹莫特。
在本发明药物组合物的一个优选实施例中,佐剂为咪喹莫特或瑞喹莫特。
该组合物可用于肠胃外给药,如皮下、皮内、肌肉或口服给药。该肽也可与细胞因子等免疫刺激物质一起服用。
本发明的药物组合物可用作抗癌疫苗。
本发明也包括一个药剂盒,其包括至少一种上述肽和/或上述药物组合物,预先制备或装入单独容器或小瓶中进行现场混合。
优选的药剂盒包括:(a)一个装有本发明药物组合物溶液或冻干粉的容器;(b)可选地,一个装有用于所述冻干制剂的稀释剂或重溶溶液和/或至少一种佐剂的第二容器;和(c)可选地,关于(1)使用该溶液或(2)重溶复原和/或使用所述冻干制剂的说明书。
本发明的优选药剂盒还包括一个或多个(1)缓冲液、(2)稀释剂、(3)过滤器、(4)针头以及(5)注射器。

发明详述
本发明提供可用作疫苗的药物组合物,其中所述药物组合物包括各种肿瘤相关肽(TUMAPs)外加甘露醇与吐温的混合物,或甘露醇与泊洛沙姆188的混合物,该混合物为所述组合物提供稳定性和溶解性。
此处所用的“药物组合物”优选为一种包含肽、甘露醇和吐温或包含肽、甘露醇和泊洛沙姆188的冻干制剂,或优选为一种重溶复原的液体组合物。因此,此处使用的“药物组合物”一词还可能指冻干的制剂,以指明该组合物以冻干的方式存在。
优选地,该药物组合物中的肽包括至少一个SEQIDNO:1至10的肽,其位于并已确认在原发性肾癌细胞上,或至少一个SEQIDNO:11至22及11至23的肽,其位于并已确认在原发性神经胶质瘤癌细胞上。
在其它优选实施例中,该药物组合物包括由SEQIDNO:12和/或SEQIDNO:13氨基酸序列组成的肽,以及至少一个由SEQIDNO:14至SEQIDNO:23的任意一个氨基酸序列组成的肽。

在一个特别优选的实施例中,该药物组合物包括作为乙酸盐或盐酸盐的肽。
在一个更优选的实施例中,本发明的药物组合物包括作为盐酸盐的SEQIDNO:5和作为乙酸盐的其它肽。
本发明的药物组合物可能还包括至少一个作为阳性对照肽的肽,其用作免疫标记物以测试皮内给药的效率。这种肽的一个例子为衍生自HBV核心抗原(SEQIDNO:11)。
在一个特定实施例中,该药物组合物包括11个不同的肽,每一个都由SEQIDNO:1至11的氨基酸序列组成(见表1)。优选地,该药物组合物中的每个肽的量均为约1500μg至75μg,优选为约1000μg至750μg,更优选为约500μg至600μg,最优选为约578μg。优选地,每个肽均由HPLC和离子交换色谱纯化,呈白色至灰白色粉末。
在另一个特定实施例中,该药物组合物包括12个不同的肽,每一个都由SEQIDNO:11至22和SEQIDNO:11至23的氨基酸序列组成(见表1)。优选地,该药物组合物中的每个肽的量均为约1500μg至75μg,优选为约1000μg至750μg,更优选为约500μg至600μg,最优选为约578μg。优选地,每个肽均由HPLC和离子交换色谱纯化,呈白色至灰白色粉末。

本文使用的“肽”这一术语指一系列通过肽键连接的氨基酸残基,alpha-氨基与相邻氨基酸的羰基连接。肽的典型长度为MHCI类,9个氨基酸,MHCII类,更长(15或16个氨基酸),但也可短至8个氨基酸和长至16个氨基酸。
本文使用的“寡肽”这一术语指一系列通过肽键连接的氨基酸残基,α-氨基与相邻氨基酸的羰基连接。只要保持正确的表位,寡肽的长度对本发明不关键。寡肽的典型长度小于约30个氨基酸,大于约14个氨基酸。
“多肽”这一术语指一系列通过肽键连接的氨基酸残基,α-氨基与相邻氨基酸的羰基连接。只要保持正确的表位,多肽的长度对本发明不关键。与术语“肽”或“寡肽”相对,“多肽”这一术语指长度大于30个残基的蛋白质分子。
如果能诱导免疫反应,肽、寡肽、蛋白质或多肽的编码是“免疫原性”的(因此有本发明中的“免疫原”)。在本发明中,免疫原性更明确地限定为诱导CTL-介导的反应。因此,“免疫原”是可诱导免疫反应的分子,在本发明中是可诱导CTL反应的分子。
T细胞“表位”是结合于I类或II类MHC分子并随后被T细胞识别的短肽分子。结合于I类MHC分子的T细胞表位代表性的长度为8-14个氨基酸,最典型为9个氨基酸。结合于II类MHC分子的T细胞表位的典型长度为12-30个氨基酸。对于结合于II类MHC分子的表位,相同的T细胞表位可能共有一个核心片段,但羧基端和氨基端的侧翼序列长度不同,因为该肽分子的末端并非像在I类MHC分子肽结合裂缝中一样包埋于II类MHC分子肽结合裂缝的结构中。
编码I类MHC分子的有三个不同的遗传位点:HLA-A,HLA-B和HLA-C。HLA-Al、HLA-A2和HLA-All是可由这些位点表达的不同I类MHC分子的例子。
当涉及到多肽时,本文使用的术语“部分”、“片段”和“碎片”指连续的残基如氨基酸残基序列,该序列组成一个更大序列的亚群。例如,如果一个多肽同常用的肽链内切酶(如胰岛素或糜蛋白酶)一起用于治疗的话,形成的寡肽将代表该起始多肽的部分、片段或碎片。这意味着任何这样的碎片都必定包含有大体上相同的片段、碎片或部分作为其氨基酸序列的一部分,如果它们不是完全相同,也和SEQIDNO:1至23中的一个序列相同,该序列对应于SEQIDNO:1至23的自然发生的“母体”蛋白。当涉及到多核苷酸时,这些术语指将所述多核苷酸和任何常用的核酸内切酶一起用于治疗时得到的产物。
依照本发明,当涉及到一个序列时,术语“相同度”或“相同性”是指将待比较的序列(“比较序列”)与所述的或请求保护的序列(“参考序列”)对准后,二者相比较。然后根据下列公式来判定“相同度”:

其中,C是参考序列与被比较序列之间在二者对准长度上的差异数量,其中
(i)在被比较序列中无对应的对准碱基或氨基酸的参考序列中的每一个碱基或氨基酸,以及
(ii)参考序列中的每个裂隙,以及
(iii)参考序列中每个对准的碱基或氨基酸与被比较序列中一个对准的碱基或氨基酸不同时构成一个差异;
并且,R是参考序列和被比较序列在对准长度上的碱基或氨基酸数目,其中参考序列中出现的任何裂隙也计为一个碱基或氨基酸。
如果被比较序列与参考序列之间存在一个对准,其相同度按上述计算约等于或大于一指定的最小相同度,则该被比较序列与该参考序列相比具有该指定最小相同度,即使可能出现按上述计算的相同度小于该指定相同度的对准。
本发明药物组合物中的肽可通过在肽链的不同(可能是选择性地)位点取代一个或多个残基而进行改性。这些取代可能是保守性的,例如,一个氨基酸被另一个具有相似结构和性质的氨基酸取代,如一个疏水性氨基酸被另一个疏水性氨基酸所取代。更保守的取代是具有相同或相似尺寸和化学性质的氨基酸的取代,例如亮氨酸被异亮氨酸取代。在研究自然发生的同源蛋白族的序列变异时,一些氨基酸取代较之另外一些更具有耐受性,而这些取代反应中的原始氨基酸和其取代物通常在尺寸、电荷、极性和疏水性方面相似,这就是定义“保守性取代”的基础。
此处定义的保守性取代是以下五组中组内的交换:第1组--小的脂肪族非极性或略带极性的残基(Ala、Ser、Thr、Pro、Gly);第2组——极性带负电荷的残基及其氨基化合物(Asp、Asn、Glu、Gln);第3组——极性带正电荷的残基(His、Arg、Lys);第4组——大型的脂肪族非极性残基(Met、Leu、Ile、Val、Cys);和第5组——大型芳基残基(Phe、Tyr、Trp)。
次保守取代可能包括一个氨基酸被另一个具有相似性质但尺寸稍微不同的氨基酸取代,例如丙氨酸被异亮氨酸残基取代。高度非保守取代可能包括将氨基酸取代为一个极性或甚至碱性的氨基酸。然而,不能因为这些“激进”的取代可能无效就不予考虑,因为化学效果并非总是可预测的,激进的取代也可能会引起由简单的化学原理无法预测的意外效果。
当然,这些取代可能包括不同于通常的L-氨基酸的结构。因此,D-氨基酸可能被取代为L-氨基酸,后者常常出现在本发明的抗原肽中但应仍属于本发明范围内。此外,具有非标准R基团的氨基酸(如不同于天然蛋白质中常见的20种氨基酸中的R基团)也可能用于取代,以生产本发明中的免疫原和免疫原性多肽。
如果发现超过一处位置上的取代导致肽产生如下定义的基本相同或更大的抗原活性,将测试那些取代的组合,以确定是否该组合的取代导致肽抗原性的累加作用或协同作用。肽上至多不超过四个位置会同时被取代。
优选地,当与取代肽对比,测试对SEQIDNO:l至23中的肽特异的CTLs时,取代肽相对于背景达到溶解最大增加值的一半时,肽浓度不高于1mM,优选为不高于1μM,更优选为不高于1nM,进而更优选为不高于100pM,最优选为不高于10pM。优选的是,取代肽被超过一个CTLs识别,至少为两个,更优选为三个。
黏蛋白(MUC1)是高度糖基化的I型跨膜蛋白,大量过度表达在许多人类腺癌如乳腺癌和卵巢癌的细胞表面。恶性肿瘤的异常去糖基化很普遍,且揭露出肿瘤细胞的表位可能与正常细胞不同。而且,MUC1的表达已被证明出现在多发性骨髓瘤和一些B细胞非霍奇金淋巴瘤中。一些最近的报道(ApostolopoulosV和McKenzieIF。Cellularmucins:targetsforimmunotherapy。CritRev.Immunol.14:293-309(1994);FinnOJ、JeromeKR、HendersonRA、PecherG、DomenechN、Magarian-BlanderJ和Barratt-BoyesSM。MUC-1epithelialtumormucin-basedimmunityandcancervaccines。Immunol.Rev.145:61-89(1995);Barnd,1989年;Takahashi,1994年;Noto,1997年)显示,源自卵巢、乳房、胰脏和多发性骨髓瘤肿瘤的MHC-不受限制的T细胞可辨认MUC1的表位,其蛋白质核心位于串联重复。源自MUC1蛋白的两个HLA-A2-限制性的T细胞表位已被识别(Brossart,1999年,EP1484397)。其中一个肽源自MUC1蛋白的串联重复区域。第二个肽位于MUC1的信号序列内。在晚期乳腺癌或卵巢癌的患者身上,以肽脉冲的树突状细胞作疫苗接种,使用那些肽去活体诱导细胞毒性T淋巴反应已经成功(Brossart,2000年)(Wierecky,2005年)。至于肾细胞癌,MUC1表达在常规肿瘤中常见,有报道其与肿瘤等级和分期有关。对于MUC1,蛋白质的过度表达与mRNA的过度表达无关。
脂肪分化相关蛋白是包含脂肪滴的特别分化细胞和脂质聚集细胞相关疾病的标志物(Heid,1998年)。脂肪分化相关蛋白在大范围的培养细胞株产生,包括纤维母细胞、内皮细胞和上皮细胞。然而,在组织中,脂肪分化相关蛋白的表达被局限于某些细胞类型,例如,泌乳乳腺上皮细胞、肾上腺皮质细胞、雄性生殖系统的支持细胞和睾丸间质细胞,以及酒精肝硬化的皮脂腺病或肝细胞脂肪变性(Heid,1998年)。有报告指出脂肪分化相关蛋白在结直肠癌(Saha,2001年)、肝细胞癌(Kurokawa,2004年)和肾细胞癌(Young,2001年)中会过度表达。
c-Met编码一种有着酪氨酸激酶活性的异质二聚合跨膜受体,是由一个与β-亚单位以二硫键连接的α-链组成(Bottaro,1991年;Rubin,1993年)。两个亚单位均在表面表达,重型β-亚单位负责连接配体和肝细胞生长因子(HGF),α-亚单位包含一个介导不同信号传递途径的活化的细胞内区域。c-Met信号参与器官再生,这显示在肝脏、肾脏、胚胎形成、造血作用、肌肉发育,而且参与正常活化的B细胞和单核细胞在迁移和吸附作用的调节(Zarnegar,1995年;Naldini,1991年;Montesano,1998年;Schmidt,1995年;Uehara,1995年;Bladt,1995年;Takayama,1996年;Mizuno,1993年;van,V,1997年;Beilmann,2000年)。此外,大量研究显示c-Met在恶性转化和恶性肿瘤的侵袭时会过度表达。
c-Met介导肝细胞生长因子/离散因子的多功能和潜在致癌活性,包括促进细胞生长、活力、生存力、胞外基质溶解和血管新生(Bottaro1991;Rubin1993;Zarnegar1995)。HGF结合至受体会诱导c-Met的自磷酸化作用并活化下游信号事件,包括ras、磷脂酰肌醇3激酶、磷脂酶Cγ和丝裂素活化蛋白激酶相关路径。c-Met基因主要表达在上皮细胞中,并过度表达在几种恶性组织和细胞株中。有日益增加的报告显示,非上皮细胞(例如造血、神经和骨骼细胞)反应至HGF和血液学的恶性肿瘤(如多发性骨髓瘤、霍奇金病、白血病和淋巴瘤)均表达c-Met蛋白。由c-Met活化突变、c-Met放大/过度表达及HGF/c-Met自分泌环路的获得,引起致癌基因c-Met的活化,而解除对侵袭型生长表达型的控制,从而给予恶性细胞侵袭与转变的性质。值得注意的是,在HGF过渡表达的基因转殖小鼠上基础性活化c-Met可促进大范围的肿瘤发生。
G-蛋白信号调节子5(RGS5)是异质三聚体G-蛋白信号路径的负向调节子,即使它在体内的功能仍然是难了解的。RGS蛋白包括一个有着统一催化功能但组织分布多变的分子族群。它们激起具有活化的Gα亚单位的鸟苷三磷酸酶(GTPase)的内在活性,从而加速G-蛋白的失活作用。因而,RGS分子抑制G-蛋白偶合受体的信号下游(De,2000年)。最近显示在肿瘤新生血管化期间,G-蛋白信号调节子5在周细胞的诱导与活性血管重建相符。在胰脏小岛细胞致癌作用的小鼠模型和高度血管生成的星形细胞瘤中,显示RGS5在血管生成的开关期间伴随活性血管重建会在周细胞过度表达。与正常朗格汉斯小岛相比,过度表达仅限于肿瘤脉管系统。但是在创伤愈合和排卵期间,RGS5的表达是上调的(Berger,2005年)。
RGS5在RCC中的表达增加(Rae,2000年)。在另一项研究中,RT-PCR显示RGS5在所有被检查的RCC里有强表达,且其在正常肾脏中的表达是非常微弱或检测不到的(实时PCR中的反应是6.6:1)。在RCC中,肿瘤内皮细胞是RGS5的主要部位(Furuya,2004年)。此外,有报道RGS5在肝细胞癌中是窦状内皮细胞的标志物(Chen,2004年)。
载脂蛋白L1(APOL1)是结合载脂蛋白A-I的分泌高密度脂蛋白。载脂蛋白A-I是相对丰富的血浆蛋白且是HDL的主要载脂蛋白。它参与血浆中多数胆固醇脂的形成,并且促进细胞中胆固醇脂的外排。载脂蛋白L1也许在贯穿整个身体的脂质交换和运输上起一定的作用,以及从外周细胞到肝脏的反向胆固醇运输。血浆蛋白是一个有着明显约40kDa分子质量的单链多肽(Duchateau,1997年;Duchateau,2001年)。APOL1cDNA从一个活化的内皮细胞的cDNA基因库被分离,显示是被TNF-α上调,TNF-α是有效的促炎细胞因子。(Monajemi,2002年)。
KIAA0367在KazusacDNA项目中被识别,该项目的目的是为推定蛋白识别未知的人类转录长编码(Ohara1997)。尽管KIAA0367基因推定的820氨基酸长度的蛋白质产物的功能是未知的,它包含一个位于C端的CRAL-TRIO脂质键结区段剖面,可结合小疏水分子,且存在于BCL2/腺病毒E1B19-kDa蛋白交互作用蛋白2(BNIP-2)。BNIP-2涉及控制不同细胞的功能,包括细胞形态学、迁移、内嗜作用和细胞周期进展(Zhou,2005年)。KIAA0367位于染色体区域9q21。在许多肿瘤里,该区域被描述为纯合性缺失的一个共同标靶(Gursky,2001年;Weber,年2001)或异合子性的丧失(Louhelainen,2000年;Tripathi,2003年)。
可溶性鸟苷酸环化酶(sGC)是由一个alpha和一个beta亚单位组成的异质二聚体蛋白(1个血红素群组),其可催化GTP转化为第二信使cGMP,并发挥一氧化氮和硝基血管扩张剂药物的主要受体的作用(Zabel,1998年)。GUCYa3和b3在人神经胶质瘤中被过度表达。转染反义的GUCY1A3或GUCY1B3减少了裸鼠的血管再生和肿瘤生长。这也许归结为VEGF被cGMP诱导的事实(Saino,2004年)。GUCY1A3促进小鼠乳房肿瘤细胞株的肿瘤细胞迁移(Jadeski,2003年)。
周期蛋白D1属于高度恒定的周期蛋白族,更确切地属于周期蛋白D亚族(Xiong,1991年;Lew,1991年)。周期蛋白起到CDKs(周期蛋白依赖激酶)调节子的作用。不同的周期蛋白具有不同的表达和降解模式,有助于每次有丝分裂事件的时态协同(Deshpande,2005年)。周期蛋白D1与CDK4或CDK6形成一复合体且作为其调节亚单位,其活性对细胞周期G1/S的转变是必须的。CCND1与CDK4和CDK6形成一种丝氨酸/苏氨酸激酶全酶的复合体,其使得底物对此复合体具有专一性(Bates,1994年)。该蛋白显示与肿瘤抑制蛋白Rb交互作用(Loden,2002年),且该基因的表达由Rb正向调节(Halaban,1999年)。该基因的突变、放大和过度表达可改变细胞周期进展,其在各种各样的肿瘤中被频繁观察到,也可能是引起肿瘤发生的原因(Hedberg,1999年;Vasef,1999年;Troussard,2000年)。
CSPG4(硫酸软骨素蛋白多糖)代表了一种膜内在硫酸软骨素蛋白多糖。其被看作是早期细胞表面黑色素瘤进展的标志物,涉及到刺激肿瘤细胞增殖、迁移和侵袭。CSPG4在大于90%的人体黑色素瘤病变中都有很强的表达。尽管CSPG4并非严格意义上的肿瘤特异,在没有自身免疫的情况下,黑色素瘤患者和健康个体的肿瘤反应性的CD4+T-cell反应能在HLA-DR11-表达黑色素瘤细胞中识别CSPG4693-709(Erfurt等人,2007年)。
有描述CSPG4在一些正常组织中表达,此外也在活化周细胞中表达,例如内皮细胞、软骨细胞、平滑肌细胞、表皮内的一些基底角质形成细胞,以及毛囊中的细胞(Campoli等人,2004年)。
在血管生成和CNS病理反应时,高度游动的CSPG4细胞会发生快速的形态改变,并被补充到血管生长和修复的位置。CSPG4在恶性脑肿瘤的血管上被肿瘤细胞和周细胞过度表达。通过将CSPG4-阳性的人神经胶质瘤细胞株中的细胞移植到免疫缺陷的裸鼠脑中,显示这些肿瘤相对于对照组具有更高的微血管密度,说明CSPG4表达能调节宿主源肿瘤脉管系统的功能和结构(Brekkeetal.,2006)。在将GBM活检材料异种移植到裸鼠的实验中,确定CSPG4主要与肿瘤脉管系统的周细胞和基底膜上的血管有关,其表达也与高细胞增生区域有关(Chekenya等人,2002a)。此外,CSPG4表达与一个神经胶质瘤移植模型中的肿瘤进展平行(Wiranowska等人,2006年)。
CSPG4在人神经胶质瘤中差异表达,在相对高级的神经胶质瘤中表达更高(Chekenya等人,1999年)。CSPG4的高表达与多药耐药性相关,该多药耐药性由α3β1整合素/PI3K信号调节子及其下游靶标增加的活化介导,以促进细胞的存活(Chekenya等人,2008年)。
FABP7:脂肪酸结合蛋白(FABPs))是14-15kDa的胞浆蛋白,认为其参与脂肪酸(FA)的摄入、运输和导向。当在膜区室间运输FAs及将FAs带到核靶时,认为它们能增加FAs在细胞质中的溶解性。FABPs可能会调节FA浓度,并以该种方式影响各种细胞功能,例如酶活性、基因表达、细胞生长和分化(Glatz和Storch,2001年)。
FABP7mRNA在神经上皮源组织和恶性神经胶质瘤中表达(WHOIII和IV级)。基因被定位到染色体带6q22-23,该区域也包括原癌基因c-myc,并经常在恶性神经胶质瘤中缺失杂合子。对恶性神经胶质瘤细胞株的分析显示,FABP7通常与胶质纤维酸性蛋白(GFAP)共同表达,暗示恶性神经胶质瘤源的细胞可能是一种具有能正常表达蛋白和形成肿瘤能力的星形前体细胞(Godbout等人,1998)。FABP7蛋白在GBM中显示中等到强的细胞核和细胞质表达。FABP7转染的神经胶质瘤细胞显示了较之对照细胞5倍强的迁移能力。因此,与FABP7在GBM中过度表达相关的总体存活期较短,可能是因为肿瘤细胞对周围的脑薄壁组织的迁移和侵袭有所增加(Liang等人,2005年)。对FABP7在星形细胞瘤分布的进一步分析显示,在肿瘤浸润区FABP7水平升高,这说明FABP7在驱动恶性细胞浸润到相邻脑组织中起着重要的作用(Mita等人,2007年)。在神经胶质组织和所有级别的星形细胞瘤中,FABP7具有不同的表达水平和亚细胞定位。然而,似乎FABP7的细胞核定位尤其与神经胶质瘤细胞浸润表型和EGFR通路有关,因为其核转位是在EGFR活化后检测到的,且与EGFR-阳性的GBM中的预后不良有关。此外,在I级星形细胞瘤中观察不到核FABP7免疫反应性(Liang等人,2006年;Kaloshi等人,2007年)。
X连接的神经连接素4是细胞粘附蛋白族的成员之一,它似乎在神经元突触的成熟和功能上发挥作用。神经连接素家族的成员都具有相关的结构组,如N端信号肽、具有两处选择性剪接的酯酶样结构域、跨膜域前的具有低序列等同度的小连接区,以及具有高度保守的C端的短胞质部分。已发现的相关神经连接素4mRNA的最高水平在心脏中。肝脏、骨骼肌和胰腺中检测到神经连接素4mRNA的较低表达,但脑、胎盘、肺和肾脏中几乎检测不到神经连接素4mRNA(Bolliger等人,2001年)。
X连接的NLGN4基因突变是自闭症谱系障碍的一个潜在原因,有报道突变发生在患有孤独症、亚斯伯格综合症和智力低下的患者身上(Jamain等人,2003年;Laumonnier等人,2004年;Lawson-Yuen等人,2008年)。
较少描述NLGN4X与癌症的关系:在胃肠基质肿瘤中,相对于成年人,在儿童与年轻人中发现了NLGN4X的过度表达(Prakash等人,2005年)。
肌腱蛋白C:肿瘤细胞周围的细胞外基质与正常组织中的细胞外基质不同。肌腱蛋白C(TNC)是一种在与升高的迁移活性紧密相关的过程中高度上调的细胞外基质蛋白,这些升高的迁移活性如胚胎发育(Bartsch等人,1992年),伤口愈合(Mackie等人,1988年)和瘤形成过程(Chiquet-Ehrismann,1993年;Chiquet-Ehrismann和Chiquet,2003年)。此外,TNC在具有高增殖指数的肿瘤血管中过度表达,暗示TNC参与肿瘤血管生成(Kimetal.,2000)。在正常人脑中很少检测到TNC的表达,但其在恶性神经胶质瘤中高度表达(Bourdonetal.,1983)。TNC表达可由缺氧(Lal等人,2001年)或TGF-beta1诱导,提供了高级神经胶质瘤侵入健康薄壁组织的机理(Hau等人,2006年),也或者被胃泌素诱导,其显著调节人GBM细胞的迁移(Kucharczak等人,2001年)。TNC下调原肌球蛋白-1,从而使得肌动蛋白应力纤维不稳定。此外它还造成了Wnt抑制剂Dickkopf1的下调。因为肌球蛋白-1表达的降低和Wnt信号子的增加同肿瘤发生和转化紧密相关,TNC特定地调节这些信号通路以增加神经胶质瘤细胞的增殖(Ruiz等人,2004年)。
在GBM组织中观察到围绕肿瘤供给血管的TNC的血管周围染色,但其在WHOII级和III级神经胶质瘤中较为少见,暗示TNC染色的密度与肿瘤级别相关,最强的染色表明最差的预后。TNC也有助于产生室下区(SVZ)内的干细胞巢,该室下区统筹生长因子信号子以加速神经干细胞的发育。SVZ中TNC对细胞的主要影响是调节发育进展(Garcion等人,2004年)。TNC是定向人神经干细胞(NSC)迁移的最强诱导剂。肿瘤生成的ECM从而为到播散的肿瘤细胞的NSC取向提供了许可环境(Ziu等人,2006年)。
NRCAM(神经细胞粘附分子)是具有多个免疫球蛋白样C2型和纤维粘连蛋白III型结构域的一种神经跨膜细胞粘附分子。它通过与其它IgCAMs形成亲同种抗原和异嗜性抗原相互作用(Volkmer等人,1996年;Sakurai等人,1997年;Zacharias等人,1999年),参与神经细胞的导向、增生和成束(Grumet等人,1991年;Morales等人,1993年;Stoeckli和Landmesser,1995年;Perrin等人,2001年;Sakurai等人,2001年)。锚蛋白结合的NRCAM(DavisandBennett,1994)在管腔形成内皮细胞中被上调,暗示其可能在管腔形成和血管生成中起作用(Aitkenhead等人,2002年)。
NRCAM是有助于肿瘤发生的β-连环蛋白和斑珠蛋白-LEF/TCF复合物的靶基因(Conacci-Sorrell等人,2002年)。NRCAM胞外域可通过金属蛋白酶样活动从细胞表面脱落。该脱落域能激活各种信号通路,提高细胞的游动性,及导致小鼠肿瘤发生(Conacci-Sorrell等人,2005年)。
相对于正常脑,NRCAM在间变性星形细胞瘤和GBM肿瘤组织中被上调,且水平上升与侵入行为有关(Sehgal等人,1998年)。抗NRCAM的反义RNA降低了人GBM细胞的致瘤能力(Sehgal等人,1999年)。
IGF2BP3是胰岛素样生长因子-IImRNA结合蛋白家族的成员,与RNA的定位、转化和翻译调控有关。该蛋白包括一些KH(K同源)结构域,其在RNA结合中很重要,已知其参与RNA合成和代谢。表达主要发生在胚胎发育过程中,已有报道其会产生肿瘤。因此认为IGF2BP3是一种癌胚胎蛋白(Liao等人,2005年)。相对于对照组织,IGF2BP3在各种癌组织中的高转录水平暗示,IGF2BP3蛋白可能在增生的转化细胞中起着功能性作用。支持该假说的发现是:表达IGF2BP3转录的唯一一个非恶性人体组织是人胎盘,其以细胞生长和增殖为特征(Mueller-Pillasch等人,1997年)。
例如,IGF2BP3在透明细胞RCC样本中表达,其表达与原发肿瘤的高级阶段和级别相关。此外,与IGF2BP3的阳性表达相关的是远处转移的风险会增加5-10倍,及RCC死亡的风险会增加42%-50%(Hoffmann等人,2008年;Jiang等人,2006年;Jiang等人,2008年)。
IGF2BP3在胰腺癌中也高度表达。在两项研究中,大于90%的胰腺肿瘤组织样本在免疫染色后显示IGF2BP3表达,而非肿瘤的胰腺组织对IGF2BP3呈阴性。此外,该表达随肿瘤的分期而逐渐增加(Yantiss等人2005;Yantiss等人,2008年)。
发现IGF2BP3表达在高级尿道上皮肿瘤中显著增加,而其在良性尿道上皮和低级尿道上皮肿瘤中通常无表达。此外,具有IGF2BP3阳性肿瘤的患者较之IGF2BP3阴性肿瘤的患者的无肿瘤进展和无疾病的存活率要低得多(Li等人,2008年;Sitnikova等人,2008年;Zheng等人,2008年)。
短缩素(BCAN)是一种脑特异性硫酸软骨素蛋白多糖,属于凝集素蛋白聚糖家族。已报道两种BCAN亚型:一种是分泌入细胞外基质的全长亚型,一种是具有可预测糖磷脂酰肌醇(GPI)定位的序列的较短的亚型。该分泌的亚型自出生起8年内均高度表达,之后20年下调到低水平,然后在正常成年人的大脑皮层中保持该水平。GPI亚型在发育期间均以低水平表达(Gary等人,2000年)。BCAN属于蛋白多糖家族,通常被描述为在成人神经系统中阻止细胞和神经突起游动性的屏障分子(Viapiano和Matthews,2006年)。活体内,BCAN沿喙侧迁移流边界表达(Jaworski和Fager,2000年)且是神经受伤后胶质瘢痕的主要上调组分(Jaworski等人,1999年)。
BCAN在神经胶质瘤中显示了引人注目的上调,在该神经胶质瘤中可检测到较正常水平约七倍的表达增加。在非死于神经系统并发症的成人个体的大脑皮层样本中未检测到BCANmRNA。形成鲜明对比的是,在27例人神经胶质瘤的手术样品中均检测到BCANmRNA,因而提议BCAN可能是神经胶质瘤中一种独特的和选择性的标记物(Jaworski等人,1996年)。
受体型蛋白酪氨酸磷酸酶Zeta1(PTPRZ1,PTP-ξ)-PTPRZ1属于受体型蛋白酪氨酸磷酸酶家族,其编码一个单通道I型膜蛋白,该膜蛋白具有两个细胞质酪氨酸蛋白磷酸酶结构域、一个alpha-碳酸酐酶结构域和一个纤维连接蛋白III型结构域。该基因的表达可在胃癌细胞(Wu等人,2006年)、乳腺癌(Perez-Pinera等人,2007年)、各种硬化病变的髓鞘再生少突胶质细胞(Harroch等人,2002年)和缺氧条件下人胚胎肾脏细胞(Wang等人,2005年)中被诱导。
蛋白质和转录都在胶质母细胞瘤细胞中过度表达,促进其趋触性迁移(Lu等人,2005年)和在胶质母细胞瘤中的遗传DNA扩增(Mulholland等人,2006年)。
几丁质酶3-样2(CHI3L2)-CHI3L2最初从软骨细胞中鉴定出来,在如骨关节炎中上调(Stecketal.,2002)。尽管该蛋白尚未被表征,其很有可能分泌到细胞外空间。其通常被描述为类风湿性关节炎的靶标抗原。人神经胶质瘤细胞株siRNA转染(VEGF-A)诱导的抗血管生成实验造成CHI3L2的上调。
存活素(BIRC5)和BIRC5(存活素)的表达属于凋亡蛋白(IAP)家族抑制剂,其在胚胎组织和各种人类癌症中有所升高。存活素似乎可以调节细胞增殖和凋亡细胞的死亡。尤其是在胶质母细胞瘤中,可检测到极高水平的存活素表达(Angileri等人,2008)。暗示在脑神经胶质瘤中的存活素的过度表达可能在恶性增生、抗凋亡和血管生成中具有重要作用(Zhen等人,2005;Liu等人,2006年)。尤其对于胶质母细胞瘤和其它肿瘤体,存活素表达与恶性肿瘤等级(胶质母细胞瘤中存活素表达最高)和比具有存活素阴性肿瘤的患者更短的总生存期显著相关(Kajiwara等人,2003年;Saito等人,2007;Uematsu等人,2005年;Mellai等人,2008年;Grunda等人,2006年;Xie等人,2006年;Sasaki等人,2002年;Chakravarti等人,2002年)。
基质金属蛋白酶(MMP)族的蛋白质涉及在正常生理过程中细胞外基质的破坏,这些生理过程如胚胎发育、生殖、组织重建,以及关节炎和瘤转移等疾病过程(Mott,2004年)。分泌的基质金属蛋白酶7(MMP7)是一种不具有活性的29.6kDa的原蛋白,其由细胞外蛋白质酶切割而被活化。该活化酶的分子量为19.1kDa,每个亚单位可结合两个锌离子和两个钙离子(Miyazaki,1990年;Browner,1995年)。MMP7降解明胶、纤维连接蛋白和酪蛋白(Miyazaki,1990年;Quantin,1989年)且与MMP家族的多数成员不同,因为它缺乏一个恒定的C端蛋白质区段(Gaire,1994年)。经常发现MMP7在恶性组织中过度表达(Lin,2004年;Bramhall,1997年;Denys,2004年),显示其可促进活体肿瘤细胞的侵袭(Wang,2005年)。
在多种类型的癌症里,这些蛋白可作为肿瘤专一免疫反应的标靶。
B型肝炎病毒核心抗原肽HBV-001不是内源的人类肿瘤相关抗原,而是源自B型肝炎病毒核心抗原。首先,允许定量比较由本发明的药物组合物中的肿瘤相关肽(TUMAPs)诱导的T细胞反应强度,因此允许研究其刺激抗肿瘤反应的能力。第二,在患者缺乏任何T细胞反应的病例下,可作为重要阳性对照。第三,允许监控患者的免疫活性状态。
B型肝炎病毒(HBV)感染是肝脏疾病的主要起因,影响全世界约三亿五千万人(Rehermann,2005年)。由于很容易通过水平和垂直传播,且具有引起能导致肝硬化和肝细胞癌的慢性疾病的潜在能力,HBV在全世界许多国家代表对公共卫生系统的一种主要冲击。HBV基因体(Previsani,2002年)包括部分双链环状DNA。在HBV的病毒粒子中,其与核心蛋白HBc和其它蛋白包在一起形成核壳体,该核壳体由含脂质和表面蛋白质族HBs(也叫包膜蛋白质)的一个外包膜所包围。与HBc和HBs连接的抗原决定簇分别作为HBcAg和HBsAg。这些抗原与血清学(即在患者血液中发现的抗体反应)相关,是在临床上诊断HBV感染的最有用的抗原抗体系统。对于之前无HBV感染史的个体,HBc代表一种新的外来抗原。由于该抗原为有名的致免疫肽,在IMA内源自HBcAg的一个十氨基酸肽被选作阳性对照抗原。HBc肽专一的CTLs诱导将被作为患者免疫活性和成功接种的标记。
本发明的药物组合物可用于肠胃外给药,如皮下、皮内、腹腔内、静脉内、肌肉内或口服给药。为此,肽溶解于或悬浮于一药学上可接受的,优选为水性的载体中。此外,该组合物也可包含赋形剂,如抗氧化剂、防腐剂、缓冲剂、粘合剂、爆炸剂、稀释剂和调味剂。该肽也可与细胞因子等免疫刺激物质一起服用。可用于这一组合物的许多赋形剂清单可取自例如A.Kibbe所著的AmericanPharmaceuticalAssociationandpharmaceuticalpress出版的《药学赋形剂手册》(HandbookofPharmaceuticalExcipients)2000年第三版。本发明的组合物可用于腺瘤性或癌性疾病的防止、预防和/或治疗。
本发明的药物组合物可用于患有与SEQIDNO:1-10相关肽或抗体有关的腺瘤性或癌性疾病的患者,或用于患有与SEQIDNO:11至22或11至23相关肽或抗体有关的脑癌,特别是神经胶质瘤,尤其是胶质母细胞瘤癌性疾病的患者。该药物组合物中的肽可触发患者体内的T细胞介导的免疫反应。如上所述,在肾细胞癌的情况下,本发明的药物组合物优选包括含有SEQIDNO:1和/或SEQIDNO:2氨基酸序列的肽,并还包括至少额外一个含有SEQIDNO:3至SEQIDNO:10的氨基酸序列的肿瘤相关肽。在脑癌,特别是神经胶质瘤,尤其是胶质母细胞瘤癌的情况下,该药物组合物优选包括SEQIDNO:12和/或SEQIDNO:13的氨基酸序列,并还包括至少额外一个含有SEQIDNO:11以及14至22和/或23的氨基酸序列的肿瘤相关肽。
优选地,本发明药物组合物中的肽总长度为9至100个氨基酸,优选9至30个氨基酸,最优选为9至16个氨基酸。此外,至少有一个根据SEQIDNO:1至SEQIDNO:23中任何一个的肽可以包括非肽键。
本发明的一个优选药物组合物(优选为肾细胞癌疫苗)包括一个由SEQIDNO:1和/或SEQIDNO:2的氨基酸序列组成的肽,在其它实施例中,还包括至少一个由SEQIDNO:3至SEQIDNO:11的氨基酸序列组成的肽。
本发明的又一个优选药物组合物(脑癌,特别是神经胶质瘤,尤其是胶质母细胞瘤癌疫苗)包括一个由SEQIDNO:12和/或SEQIDNO:13的氨基酸序列组成的肽,在其它实施例中,还包括至少一个由SEQIDNO:11至SEQIDNO:22和/或23的氨基酸序列组成的肽。
该肽也可能是标记的,或可能是融合蛋白,或可能是杂交分子。本发明所述序列的肽预计可刺激CD8+CTL。然而,在CD4+T-细胞帮助下的刺激更为有效。因而,杂交分子的融合伴或区段合适地提供刺激CD4+T-细胞的表位。CD4+刺激表位在本领域已为人熟知并包括那些已在破伤风类毒素中确认的表位。在又一个优选实施例中,该肽是一个融合蛋白,特别包括HLA-DR抗原相关的恒定链(Ii)的N端氨基酸。在一个实施例中,本发明的肽是一个截短人蛋白或一个蛋白碎片和另一个多肽部分中的融合蛋白,假若人的那部分包括一个或多个有创造性的氨基酸序列。
可用于该药物组合物的肽可能基本上是纯的,或与免疫刺激佐剂结合,或与免疫刺激细胞因子联合使用,或通过一合适的给药系统给药,例如脂质体、微纳颗粒、胶束、乳剂、凝胶。通常肽疫苗接种需要佐剂,如此,优选为GM-CSF(人GM-CSF市售商品名为可购自Berlex,现在是BayerHealthCarePharmaceuticals)其它合适的佐剂包括Aquila'sQS21刺激子(AquilaBiotech,Worcester,MA,USA),其来自于皂苷、分歧杆菌提取物和合成细菌细胞壁模拟,以及专卖的佐剂如Ribi'sDetox。另一个由皂苷衍生的佐剂QuilA也可能使用(Superfos,Denmark)。其它佐剂如Freund's也可能有用。使得该肽与匙孔血蓝蛋白(KLH)或甘露聚糖共轭也可能是有用的(见WO95/18145以及Longenecker等人(1993)Ann.NYAcad.Sci.690,276-291)。既然佐剂被定义为能提高对抗原免疫反应的物质(MedicalDictionary,NIH),具有该功能的其它物质也可能被使用,包括但不限于toll样受体拮抗剂(TLR拮抗剂),优选为可与TLR3、7、8和9互相抗拮的物质,如精蛋白固定的RNA、CpG寡核苷酸、CpR寡核苷酸、细菌DNA、咪唑并喹啉等。
本领域已知的可增强免疫反应的其它物质包括但不限于下列物质的抑制剂:可诱导的一氧化氮合酶(iNOS)、精氨酸酶(ARG1)、吲哚胺-2,3-双加氧酶(IDO)、血管内皮生长因子受体1(VEGFR-1)、血管内皮生长因子(VEGF)、环氧合酶(COX-2)、TGF-beta受体I(TGF-beta-RI)。这些抑制剂可能是抗分子或小分子的单克隆抗体。本领域已知的对上述因子具有抑制功能和免疫反应增强效应的小分子和单克隆抗体例如:1-MT、NCX-4016、罗非西布、西乐葆、BEC、ABH、nor-NOHA、SB-505124、SD-208、LY580276、AMD3100、阿西替尼、贝伐单抗、JSI-124、CPA-7、XL-999、ZD2171、帕唑帕尼、CP-547632以及VEGF阱。
此外,能减少调节T细胞(CD4+,CD25+,FoxP3+)数量的物质也可用作佐剂。这些包括(但不限于):环磷酰胺(Cytoxan)、ONTAK(denileukindiftitox)、舒尼替尼、anti-CTLA-4(MDX-010,CP-675206)、anti-CD25、anti-CCL22以及anti-GITR。
本发明的药物组合物中肽的优选量可在约0.1至100mg之间变动,优选为约0.1至1mg之间,最优选为每500μl溶液含约300μg至800μg。如果未明确说明,术语“约”指给定值的+/-10%。本领域技术人员可根据各种因素调整肽的实际用量,例如,患者个体的免疫状态和/或特定类型癌症中存在的TUMAP量。
较之以前已知的组合物,本发明的药物组合物提供了具有极高的溶解性和冻干粉润湿性的制剂。这可以通过使用特别的赋形剂组合物实现。因此,可开发含有SEQIDNO:1至10或SEQIDNO:1至11及其变体的本发明药物组合物,其在(-20℃、+5℃、+25℃)温度下具有优异的储存稳定性,而且容易再溶解。
术语“储存稳定性”指两年内副产物的百分比上升不超过5%。此外,术语“稳定性”指特定性质(如溶解性、溶液的光学澄清度、溶液中微粒的数目)在该期限内无可觉察的改变。
术语“容易再溶解”指通过使用缓冲液或其它赋形剂,冻干粉可以在不使用超声匀浆的情况下在数秒到两分钟内完全溶解。此外,通过皮内给药和较不优选的皮下给药,该组合物可很容易地给予需要治疗的患者。所得溶液的pH值应在pH2.7至pH9之间。
在另一个实施例中,本发明的药物组合物可能包括构成骨架的糖、糖醇、氨基酸如甘氨酸、精氨酸、谷氨酸等。糖可能是单糖、二糖或三糖。这些糖可以单独使用,也可以与糖醇联合使用。糖包括作为单糖的葡萄糖、甘露糖、半乳糖、果糖或山梨糖,作为二糖的蔗糖、乳糖、麦芽糖或海藻糖,以及作为三糖的棉子糖。糖醇例如甘露糖。优选的配料为蔗糖、乳糖、麦芽糖、海藻糖、甘露醇和/或山梨糖醇,更优选甘露醇。
此外,本发明的药物组合物还可包括生理耐受性好的赋形剂(见RaymondRowe、PaulSheskey和SianOwen编辑的《药学赋形剂手册》(HandbookofPharmaceuticalExcipients)第5版,PharmaceuticalPress出版(2006年)),例如:抗氧化剂如抗坏血酸或谷胱甘肽,防腐剂如苯酚、m-甲酚、对羟基苯甲酸甲酯或对羟基苯甲酸丙酯,氯丁醇、硫柳汞或苯扎氯铵,稳定剂和骨架形成剂如蔗糖、乳糖、麦芽糖、海藻糖、甘露糖、甘露醇和/或山梨糖醇、甘露醇和/或乳糖,增溶剂如聚乙二醇(PEG),即PEG3000、3350、4000或6000,或环糊精,即羟丙基-β-环糊精、磺丁乙基-β-环糊精,或γ环糊精,或右旋糖苷,或泊洛沙姆,即泊洛沙姆泊洛沙姆188、或吐温吐温在一个优选实施例中,本发明的药物组合物包括一或多种选自抗氧化剂、骨架形成剂和稳定剂的耐受性好的赋形剂。
本发明涉及一个发现,即制剂如果包括至少两组根据SEQIDNO:1至10或SEQIDNO:1至11的肽、及成一定比的甘露醇和泊洛沙姆188,则该组合物的稳定性将大为提高,不使用超声处理的情况下即可溶解。且溶解只需要几秒钟。按照该冻干剂的说明书,在用4.2%的碳酸氢钠溶液溶解后,视觉可观察到溶液由澄清变为乳白色。本发明的该药物组合物的重溶解特征在-20℃、+5℃和+25℃下储存2年后仍可重现。
此外,本发明涉及的制剂包括至少四组根据SEQIDNO:11至SEQIDNO:22、SEQIDNO:11至SEQIDNO:23、SEQIDNO:12至SEQIDNO:22或SEQIDNO:12至SEQIDNO:23的肽,其中成一定比的甘露醇和泊洛沙姆188使得组合物易溶解。按照该冻干剂的说明书,在用4.2%的碳酸氢钠溶液溶解后,视觉可观察到溶液由澄清变为乳白色。所述制剂中的肽在-20℃、+5℃和+25°下储存至少3个月后仍稳定。
在一个优选实施例中,本发明的药物组合物包括成一个特定比的肽:甘露醇:吐温(重量比),比例范围是1:2:1.5至1:8:2.2之间(含)。本发明包括的其它优选比见下面的图表。一个尤其优选的比例是1:5:2。另一个尤其优选的比例是1:8:2。
在另一个优选实施例中,本发明的药物组合物包括成一个特定比的肽:甘露醇:泊洛沙姆188(重量比),比例范围是1:5:1.5至1:8:2.2之间(含)。本发明包括的其它优选比例见下面的图表。一个优选的比例是1:5:1。

静脉内和肌肉内给药可接受的pH范围是pH2–12,但皮下给药的pH范围则降至2.7–9.0,因为体内稀释的速率降低,导致其在注射部位放射的可能性增大。StrickleyRobertG.,Pharm.Res.,21,NO:2,201-230(2004)。
本发明含水组合物优选的pH值为7-9,更优选的pH值为8-9,更加优选的pH值为8.3-8.7。
本发明的药物组合物生理耐受性好,容易生产,剂量精确,其浓度、分解产物和集合物在储存时间内具有优良的稳定性。在-20℃的冷冻机中,在+2-8℃的冰箱中,甚至在室温(+25℃)和60%的相对湿度下,该制剂都可在2年内稳定储存。
供体内使用的本发明药物组合物优选为无菌制剂。
本发明也提供一种提高用药对象免疫反应的方法,所述方法包括向用药对象给予本发明药物组合物,其中所述肽及其变体或盐以有效量给药,优选能引起所述免疫反应的有效量。本发明还提供了该药物组合物的用途,将该药物给予用药对象以引起抗癌免疫反应。还包括使用本发明药物组合物治疗癌症,优选为肾癌,以及用其制造药物,给予用药对象以引起抗癌免疫反应,从而治疗癌症。
该药物组合物所用的肽优选为纯的或基本为纯,且希望是基本均匀的(即不含带杂质的肽或蛋白质等)。“基本为纯”指HPLC检测的纯度至少为90%,优选为至少95%的肽制剂。“基本均匀”的制剂指该制剂总重量的至少99%是肽的肽制剂。
本发明还包括一个药剂盒,其包括:
(a)一个装有上述的药物组合物溶剂或冻干剂的容器;

容纳该制剂的容器可为一多用小瓶,其允许重复(例如2-6次给药)给予该重溶复原的制剂。该药剂盒还可包括一含有合适稀释剂(如碳酸氢钠溶液)的第二容器。

本制剂的肽可通过任何可接受的途径给药,例如口服(肠衣)、鼻内、眼内、皮下、皮内、肌肉内、静脉内或透皮。优选地,给药为皮下,最优选为皮内。也可通过输液泵给药。
分析测试步骤:
用RP-HPLC分析色谱法来进行定性分析,测定纯度和肽含量。检测波长是220nm。
稳定性测试(试验制剂的稳定性):
a)肾细胞癌疫苗:
用HPLC法来测试各种冻干剂中各肽在+25℃和+40℃下的稳定性。
压力测试在+25℃和+40℃下进行,以测试制剂在环境温度和更高温度下更稳定的趋势。
根据SEQIDNOs:1至10的肽在无任何赋形剂,并加入甘露醇和泊洛沙姆188(Lutrol)或吐温的情况下冻干。
生产下列试验制剂并用HPLC法来评价其在+25℃和+40℃下的稳定性:
制剂1:不含任何赋形剂的肽;
制剂2:肽:甘露醇:(泊洛沙姆188)(重量比1:5:2)
制剂3:肽:甘露醇:吐温(重量比1:5:2);和
制剂4:肽:甘露醇:吐温(重量比1:5:1).
在储存于人工气候室之前,已经对每种制剂进行初始HPLC测量。为此,将两个小瓶中的物质完全溶解于30%乙酸,并由RP-HPLC进行重复测定。为确保单次测量的相似性,使用β-萘基丙氨酸作为内部标准并与肽一起冻干。
在7、14、21和28天后,将另外的两个小瓶从人工气候室中取出评价其稳定性。
通过对一个小瓶的重复测定来评价数据。对于一个时间点和温度,使用两个独立的小瓶来得到共计四个数据点。用这些数值来计算标准偏差,并在相关图表中以误差棒显示。
实验结果表明,含有甘露醇和的制剂中的每种肽的稳定性与不含任何赋形剂的制剂所得的结果具有可比性。含有吐温的制剂尤其在+40℃时IMA-RGS-001的降解增强,尤其是肽IMA-ADF-001的信号增加。该增加可能是由肽降解带来的杂质一起洗脱造成的。
b)胶质母细胞瘤细胞癌疫苗:
测试各种冻干剂混合物中肽制剂的溶解度和稳定性。
根据SEQIDNOs:11至23的肽在无任何赋形剂,并加入甘露醇/泊洛沙姆188(Lutrol)和甘露醇/吐温的情况下冻干。
生产下列试验制剂,并在+25℃和+40℃下,测试其在不同溶液中的溶解性和稳定性。
制剂1:不含任何赋形剂的肽;
制剂2:肽:甘露醇:(泊洛沙姆188)(重量比1:5:2)
制剂3:肽:甘露醇:吐温(重量比1:5:2)
制剂的稳定性都很好。制剂2的溶解性最好,呈无色澄清混合物,但制剂2在更高浓度情况下有轻微沉淀。制剂1完全不能溶解。
实验结果表明,含有甘露醇/Lutrol和甘露醇/吐温制剂中的每种肽的稳定性与不含任何赋形剂的制剂所得的结果具有可比性。不含任何赋形剂的制剂在合适的溶液,如碳酸氢钠(4.2%)溶液中也不溶解。

测试赋形剂泊洛沙姆188和甘露醇对T细胞的引发效率。IMA901制剂中稳定的赋形剂是泊洛沙姆188和甘露醇。这两种赋形剂在第1阶段小试中未包含在IMA901制剂中,但包含在第2阶段小试制剂中,以提高IMA901的溶解性和用中稳定性。在该项研究中,在鼠类模型肽免疫之后,通过一个小鼠模型测试这些物质对T细胞引发效率的影响。未观察到泊洛沙姆188和甘露醇有毒性效应。免疫后九天,通过四聚体染色和流式细胞仪间接体内分析T细胞引发。免疫溶液中加入泊洛/甘露醇不会改变CD8+T细胞引发效率。
测试原理
免疫:引发CD8+T细胞,免疫原性的H2-Kb结合肽Ova257-264(SIINFEKL)与通常用于小鼠免疫的佐剂(CpG脱氧寡核苷酸)一起随4.2%的重碳酸缓冲液皮内注射入8-12周大的雌性小鼠中(C57BL/6品系,每组3只)。比较含和不含泊洛沙姆188(Lutrol(16.5mg/ml))和甘露醇(41.3mg/ml)的注射液。赋形剂浓度与IMA901注射液中的相同。阳性对照组用含有经classic(Sigma-Aldrich)乳化的CpG的肽溶液进行皮下免疫。
四聚体染色:9天后,杀死小鼠,用四聚体染色和流式细胞仪间接体内分析脾脏中的Ova257-264-特异T细胞。四聚体技术使得能够特异灵敏地检测带有相容T细胞受体的T细胞。
结果:在注射部位和全身均未观察到泊洛沙姆188(LutrolBASF,Ludwigshafen,Germany)/甘露醇的毒性。相对于阴性对照组,阳性对照组、CpG组和CpG/泊洛沙姆188/甘露醇组显示出明显更高的肽特异性T细胞频率(p<0.05)。CpG组和CpG/泊洛沙姆188/甘露醇组无显著差异。
五天体外肽刺激后,通过对增殖肽-特异性细胞的四聚体染色,排除人工观察到阳性人群的可能性(未显示数据)。
总之,泊洛沙姆188/甘露醇在免疫鸡尾酒中的存在并未改变小鼠中肽引发的免疫反应,因此,预期泊洛沙姆188和甘露醇对基于肽的免疫疗法的功效和安全性通常无反作用或不利影响。
材料和方法
用于生产不同制剂的肽由瑞士BachemAG合成和供应。
实施例
实施例1:
每种肽的储备液是通过将肽根据其溶解特性溶解在合适的溶剂中制得的(见表2)。将1.47mL高浓缩的肽溶液(肽5至10)与7.34mL肽溶液1至4合并,然后加入1.47mL10%乙酸。该混合物涡旋2分钟,水浴超声1分钟。将得到的澄清溶液转移约1mL于玻璃小瓶,再加入19.2mg甘露醇和50μL吐温80储备液(将77mg吐温溶解于30%的乙酸溶液中)。几分钟后,将该溶液在-40℃下冷冻,0.06mbar下冻干14小时,再在0.003mbar下干燥6小时(见表3)。

实施例2:
生产了一个GMP批,包括2.000个小瓶,每瓶含有的每种肽为578μg,还含有甘露醇和泊洛沙姆188。该制剂包括重量比为1:5:2的肽、甘露醇和泊洛沙姆188。
根据表4中列出的量,称取每种冻干肽,置于单独的玻璃容器中。称取后,将所有的肽溶解于一指定溶剂中。

由于肽的溶解性不同,不得不按照表4中提供的顺序从肽1开始将其溶解。考虑到肽溶解性的差异和本体溶液的最后填充体积,采用不同量和浓度的乙酸溶液和注射用水(WFI)来溶解肽。为提高溶解性,每个小瓶均剧烈振荡最多5分钟,如果必要再超声最多5分钟。采用的量和浓度列于表4。
一旦肽已容易溶解,应将溶液按照表4中给出的顺序从1号肽开始混合。将溶液收集到一个无菌的玻璃容器中。用105ml的乙酸溶液(30%)润洗各肽小瓶。最后,将该溶液加入肽溶液的混合物中并搅拌五分钟。
将23.1g泊洛沙姆188和57.8g甘露醇加入到肽混合物中,将整个溶液搅拌五分钟。
含有全部10种肽和赋形剂的本体溶液经孔径尺寸为0.22μm的过滤器进行无菌过滤。将1.485ml溶液在无菌条件和惰性氮气气氛中注入已灭菌的2R玻璃小瓶中,预密封并转移入冷冻干燥机进行冻干操作。该冻干过程(见表5)包括在-45℃下冷冻小瓶,逐步升高温度至+20℃(主要干燥阶段),以及在+25℃下的最后干燥步骤。干燥完成后,用已经无菌过滤的干燥氮气将冷冻干燥机降到大气压。

再溶解步骤:上述制剂溶解于700μl碳酸氢钠(4.2%)中用于临床试验。取下小瓶上的塑料帽。再溶解前至少30分钟,打开包装,将碳酸氢钠(4.2%)从冰箱中取出,使其在注射前达到室温。准备好用于重溶复原的注射器和针头。无菌操作取出700μL稀释剂用于冻干剂的重溶复原。为溶解该冻干剂,应剧烈摇动该小瓶和稀释剂1分钟,检查溶液是否澄清,否则继续摇动直到溶液澄清。GM-CSF注射后10至30分钟,使用一新注射器在同一位置皮内给予500μL重溶复原的制剂。给药应在重溶复原后1小时之内进行。重溶复原后,溶解的冻干粉可在室温无菌条件下储存达1小时。
重溶复原后的稳定性测试显示大多数肽在重溶复原后仍然足够稳定。但有两种肽有所降低,即IMA-CCN-001和IMA-RGS-001。(见表6)。因为这些肽含有半胱氨酸残基,二聚反应会随着时间发生。

在小鼠模型中评价不同pH值(pH8.5左右)和高容量碳酸盐缓冲液对肽免疫成功性的影响。在不同注射液中使用标准免疫原性模型肽,用标准51Cr释放法测试引发效率。四聚体染色后用流式细胞仪分析来验证结果。总结而言,在pH7.5、8.5(IMA901稀释剂的pH)和pH9.5下的皮内免疫的引发效率未检测到显著差异。
此外,相对于等渗注射缓冲液,高离子强度未降低观察到的免疫反应。在以pH7.5、8.5注射液、及以IMA901稀释剂进行免疫时未观察到毒性副作用,但是在注射液为pH9.5时毒性副作用变得明显(在注射部位皮肤发生局部坏死性病变)。这些结果支持所选缓冲液为IMA901的合适稀释剂。
实施例3:
每种肽的储备液是通过将肽根据其溶解特性溶解在合适的溶剂中制得的(见表6)。

由于肽的溶解性不同,必须按照表4中提供的顺序从肽1开始将其溶解。考虑到肽溶解性的差异和本体溶液的最后填充体积,采用不同量和浓度的乙酸溶液和注射用水(WFI)来溶解肽。为提高溶解性,每个小瓶均剧烈振荡最多5分钟,如果必要再超声最多5分钟。采用的量和浓度列于表6。
一旦肽已经容易,应将溶液按照表6中给出的顺序从1号肽开始混合。将溶液收集到一个带有搅拌棒的玻璃容器中。最后,将该溶液加入肽溶液的混合物中并搅拌至少五分钟。
将6011g泊洛沙姆188和15028mg甘露醇加入到肽混合物中,将整个溶液搅拌五分钟。
含有全部13种肽和赋形剂的本体溶液经孔径尺寸为0.22μm的过滤器进行无菌过滤。将1,500ml溶液装入2R玻璃小瓶中,预密封,转移入冷冻干燥机中冻干。该冻干过程(见表7)包括在-45℃下冷冻小瓶,逐步升高温度至+20℃(主要干燥阶段),以及在+25℃下的最后干燥步骤。干燥完成后,用氮气将冷冻干燥机降到大气压。

再溶解步骤:

将624.2mg泊洛沙姆188和15606mg甘露醇加入到肽混合物中,将整个溶液搅拌五分钟。
含有全部12种肽和赋形剂的本体溶液经孔径尺寸为0.22μm的过滤器进行无菌过滤。将1.500ml溶液装入2R玻璃小瓶中,预密封,转移入冷冻干燥机中冻干。该冻干过程(见表10)包括在-45℃下冷冻小瓶,逐步升高温度至+20℃(主要干燥阶段),以及在+25℃下的最后干燥步骤。干燥完成后,用干燥氮气将冷冻干燥机降回到大气压。

不同温度下的稳定性测试显示,即使在压力(+25℃)条件下,所有的肽都充分稳定(见表11)。

Claims (15)
1.一种药物制剂,包含:
2至18个、2至15个、2至13个、2至12个或3至12个肿瘤相关肽;
其中每一个肽的长度为8至22个氨基酸或9至16个氨基酸;
其中所述肽在90%乙酸中的溶解性为至少2.7mg/mL;
还包含甘露醇和泊洛沙姆188,其中所述肽与甘露醇和泊洛沙姆188的重量比范围在1:5:1.5至1:8:2.2之间且包含两端点;
或包含甘露醇和聚山梨醇酯80,其中肽与甘露醇和聚山梨醇酯80的重量比范围在1:2:1.5至1:8:2.2之间且包含两端点。
2.根据权利要求1所述的药物组合物,其中肽与甘露醇和泊洛沙姆188的所述重量比范围在1:0:2至1:0:2.2之间且包含两端点。
3.根据权利要求1所述的药物组合物,其含有至少两个肽,其中所述肽包括一个选自由SEQIDNO:1至SEQIDNO:10,或SEQIDNO:12至SEQIDNO:23组成的氨基酸序列;但要求该组合物包括至少一个含有SEQIDNO:1,SEQIDNO:2,SEQIDNO:12或SEQIDNO:13或其变体或盐的肽;还包括甘露醇和泊洛沙姆188,其中肽与甘露醇和泊洛沙姆188的重量比范围在1:5:1.5至1:8:2.2之间且包含两端点。
4.根据权利要求3所述的药物组合物,其中肽与甘露醇和泊洛沙姆188的重量比范围在1:0:2至1:0:2.2之间且包含两端点。
5.根据权利要求1所述的药物组合物,其含有至少两个肽,其中所述肽包括选自由SEQIDNO:1至SEQIDNO:10,或SEQIDNO:12至SEQIDNO:23组成的一个氨基酸序列;但要求该组合物包括至少一个含有SEQIDNO:1、SEQIDNO:2、SEQIDNO:12或SEQIDNO:13或其变体或盐的肽;还包括甘露醇和聚山梨醇酯80,其中肽与甘露醇和聚山梨醇酯80的重量比范围在1:2:1.5至1:8:2.2之间且包含两端点。
6.根据权利要求1至5中任意一项所述的药物组合物,其中所述肽的总长度为9至16个氨基酸。
7.根据权利要求1至6中任意一项所述的药物组合物,其中至少一个所述肽包含非肽键。
8.根据权利要求1至7中任意一项所述的药物组合物,其中所述组合物包括由SEQIDNO:1和/或SEQIDNO:2的氨基酸序列组成的肽,以及至少一个由SEQIDNO:3至SEQIDNO:10中的任意一个氨基酸序列组成的肽。
9.根据权利要求1至8中任意一项所述的药物组合物,其中所述组合物包括由SEQIDNO:12和/或SEQIDNO:13的氨基酸序列组成的肽,以及至少一个由SEQIDNO:14至SEQIDNO:23中的任意一个氨基酸序列组成的肽。
10.根据权利要求1至9中任意一项所述的药物组合物,还包括一个含有SEQIDNO:11氨基酸序列的肽,但要求所述肽不是相应的全长肿瘤相关多肽,或还包括一个由SEQIDNO:11氨基酸序列组成的肽。
11.根据权利要求1至10中任意一项所述的药物组合物,其中该组合物中肽的含量具有组织、癌症和/或患者特异性。
12.根据权利要求1至11中任意一项所述的药物组合物,还包含至少一种合适的佐剂。
13.根据权利要求1至12中任意一项所述的药物组合物作为抗癌疫苗的用途。
14.一种药剂盒,包括:
(a)一个装有根据权利要求1至13中任意一项所述的药物组合物溶液或冻干粉的容器;
(b)可选地,一个装有用于所述冻干制剂的稀释剂或重溶复原溶液和/或至少一种佐剂的第二容器;和
(c)可选地,关于(1)使用该溶液,或(2)重溶复原和/或使用所述冻干制剂的说明书。
15.根据权利要求14所述的药剂盒,还包括一个或多个(1)缓冲液、(2)稀释剂、(3)过滤器、(4)针头和(5)注射器。
