技术领域
本发明涉及医疗材料领域,特别是涉及一种具有抗菌消炎功效的医用无纺布及制备方法。
背景技术
无纺布是一种不需要纺纱织布而形成的织物,其是将纺织短纤维或者长丝进行定向或随机撑列,形成纤网结构,然后采用机械、热粘或化学等方法加固而成。与传统的工艺相比,具有工艺流程短、生产速度快、产量高、成本低、用途广等特点。目前,无纺布广泛应用于医疗卫生用布、家庭装饰用布、农业用布等。
申请公布号CN103882690A(申请号201210561852.9)的中国专利文献公开了一种医用壳聚糖纤维无纺布的生产方法,将壳聚糖纤维无纺布放入整理液浸泡处理,之后再放入浓度为95%以上的乙醇中浸泡40—60min消毒处理,浸泡之后再使用浓度为95%以上的乙醇冲洗2—3次。申请公布号CN102605612A(申请号201210059384.5)的中国专利文献公开了一种无纺布,该无纺布为经天然复合液处理的无纺布,所述的天然复合液由桉叶油/青蒿液、薄荷油、丹参液和医用酒精组成。申请公布号CN104264371A(申请号201410454909.4)的中国专利文献公开了一种医用无纺布的加工方法,依次包括以下步骤:步骤一、喷涂,步骤二、浸泡、压干等步骤。
目前,无纺布广泛应用于医疗卫生用布、家庭装饰用布、农业用布等。但目前现有无纺布,功能较单一,无法同时满足医用的多种需要。
发明内容
针对现有技术的不足,本发明的目的是提供一种具有良好的止血、抗菌消炎、消肿止痛等功效的医用无纺布及其制备方法。本发明所得产品无毒副作用,具有良好的生物可降解性和良好的生物相容性,止血速度快,能够缩短治疗时间,避免伤口感染,防止粘连,容易与伤口贴合,促进伤口愈合,具有良好的透气性。
本发明的技术方案是:
具有抗菌消炎功效的医用无纺布,取下述重量配比的主要原料制备而成:壳聚糖50-70,蒲公英3-7,红孩儿4-8,葛根2-5,丝瓜络6-10,丁葵草3-6,两面针6-10,地皮消4-8,太白菊2-5,母猪藤根3-7,甘油适量,醋酸钠适量。
具有抗菌消炎功效的医用无纺布,优选的方案是,取下述重量配比的主要原料制备而成:壳聚糖55-65,蒲公英4-6,红孩儿5-7,葛根3-4,丝瓜络7-9,丁葵草4-5,两面针7-9,地皮消5-6,太白菊3-4,母猪藤根5-6,甘油适量,醋酸钠适量。
具有抗菌消炎功效的医用无纺布,更加优选的方案是,取下述重量配比的主要原料制备而成:壳聚糖62,蒲公英5,红孩儿6,葛根3.5,丝瓜络8,丁葵草4.5,两面针8,地皮消5.5,太白菊3.5,母猪藤根5.5,甘油适量,醋酸钠适量。
上述具有抗菌消炎功效的医用无纺布的制备方法是:
(1)将红孩儿研成粗粉,过50目筛,加醋拌匀,醋的用量为红孩儿重量的1/4-2/3(优选的,醋的用量为红孩儿重量的1/3-3/5;更加优选的,醋的用量为红孩儿重量的2/5),闷2-4小时(优选的,闷2.5-3.5小时;更加优选的,闷3小时),晒干,粉碎,放入锅中文火炒10-18分钟(优选的,放入锅中文火炒12-16分钟;更加优选的,放入锅中文火炒14分钟),加入红孩儿重量6.7-8.9倍的水(优选的,加入红孩儿重量7-8倍的水;更加优选的,加入红孩儿重量7.5倍的水),煎煮1小时,滤出药液;
(2)将丝瓜络洗净,晒干,切成段,先将锅用中火烧热,撒入麦麸,待冒烟时,投入切成段的丝瓜络,用麦麸拌炒至黄色为度,取出,筛去麸皮;
(3)将步骤(2)所得与除壳聚糖、红孩儿和丝瓜络外的其余7味原料药粉碎,过40目筛,加入7味原料药5-8倍的水煎煮1小时(优选的,加入7味原料药5.5-7倍的水煎煮1小时;更加优选的,加入7味原料药6.2倍的水煎煮1小时),滤出药液,继续加入7味原料药4.5-6倍的水煎煮1小时(优选的,继续加入7味原料药5-5.6倍的水煎煮;更加优选的,继续加入7味原料药5.2倍的水煎煮1小时),滤出药液,合并两次药液;
(4)将步骤(1)和步骤(3)所得混合,以4200r/min离心分离18分钟,取上层液,浓缩成相对密度为0.45-0.65的药液(优选的,浓缩成相对密度为0.5-0.6的药液;更加优选的,浓缩成相对密度为0.55的药液),备用;
(5)将壳聚糖溶于体积分数为2%的醋酸溶液中,壳聚糖的脱乙酰度为75%-95%(优选的,壳聚糖的脱乙酰度为80%-90%;更加优选的,壳聚糖的脱乙酰度为88%),配制成质量分数为2%的壳聚糖溶液,过滤,除去不溶物;
(6)将步骤(4)和步骤(5)所得混合,充分搅拌,加入甘油和醋酸钠,甘油的体积为步骤(4)和步骤(5)所得总体积的1/5,醋酸钠的质量为壳聚糖质量的1/20,混合均匀,得纺丝原液,将纺丝原液挤出喷丝孔进入凝固浴,纺丝原液凝固析出而形成纤维,将所得纤维切成长度为5cm丝束,于盖尔梳理机开纤梳理,使之成棉絮状纤维,重新置于盖尔梳理机上辊压打片,使其厚度为45-80g/m2,(优选的,其厚度为50-70g/m2;更加优选的,使其厚度为60g/m2),即得。
本发明提供的具有抗菌消炎功效的医用无纺布制备方法简单。除此之外,本发明的优良效果还表现在:
1、可生物降解,具有良好的透气性;
2、具有良好的止血、抗菌消炎、消肿止痛等功效,止血速度快,能够缩短治疗时间,避免伤口感染,防止粘连,所得无纺布容易与伤口贴合,促进伤口愈合;
3、安全有效,效果显著且无任何毒副作用。
具体实施方式
下面结合实施例和实验例详细说明本发明的技术方案,但保护范围不限于此。
实施例1具有抗菌消炎功效的医用无纺布,取下述重量份的主要原料制备而成(每份为500g):壳聚糖50份,蒲公英3份,红孩儿4份,葛根2份,丝瓜络6份,丁葵草3份,两面针6份,地皮消4份,太白菊2份,母猪藤根3份,甘油适量,醋酸钠适量。
上述具有抗菌消炎功效的医用无纺布的制备方法是:
(1)将红孩儿研成粗粉,过50目筛,加醋拌匀,醋的用量为红孩儿重量的1/4,闷2小时,晒干,粉碎,放入锅中文火炒10分钟,加入红孩儿重量6.7倍的水,煎煮1小时,滤出药液;
(2)将丝瓜络洗净,晒干,切成段,先将锅用中火烧热,撒入麦麸,待冒烟时,投入切成段的丝瓜络,用麦麸拌炒至黄色为度,取出,筛去麸皮;
(3)将步骤(2)所得与除壳聚糖、红孩儿和丝瓜络外的其余7味原料药粉碎,过40目筛,加入7味原料药5倍的水煎煮1小时,滤出药液,继续加入7味原料药4.5倍的水煎煮1小时,滤出药液,合并两次药液;
(4)将步骤(1)和步骤(3)所得混合,以4200r/min离心分离18分钟,取上层液,浓缩成相对密度为0.45的药液,备用;
(5)将壳聚糖溶于体积分数为2%的醋酸溶液中,壳聚糖的脱乙酰度为75%,配制成质量分数为2%的壳聚糖溶液,过滤,除去不溶物;
(6)将步骤(4)和步骤(5)所得混合,充分搅拌,加入甘油和醋酸钠,甘油的体积为步骤(4)和步骤(5)所得总体积的1/5,醋酸钠的质量为壳聚糖质量的1/20,混合均匀,得纺丝原液,将纺丝原液挤出喷丝孔进入凝固浴,纺丝原液凝固析出而形成纤维,将所得纤维切成长度为5cm丝束,于盖尔梳理机开纤梳理,使之成棉絮状纤维,重新置于盖尔梳理机上辊压打片,使其厚度为45g/m2,即得。
本发明所得具有抗菌消炎功效的医用无纺布的使用方法是:用本发明所得产品包扎创口或吸附创口组织渗出物。
实施例2具有抗菌消炎功效的医用无纺布,取下述重量份的主要原料制备而成(每份为500g):壳聚糖70份,蒲公英7份,红孩儿8份,葛根5份,丝瓜络10份,丁葵草6份,两面针10份,地皮消8份,太白菊5份,母猪藤根7份,甘油适量,醋酸钠适量。
上述具有抗菌消炎功效的医用无纺布的制备方法是:
(1)将红孩儿研成粗粉,过50目筛,加醋拌匀,醋的用量为红孩儿重量的2/3,闷4小时,晒干,粉碎,放入锅中文火炒18分钟,加入红孩儿重量8.9倍的水,煎煮1小时,滤出药液;
(2)将丝瓜络洗净,晒干,切成段,先将锅用中火烧热,撒入麦麸,待冒烟时,投入切成段的丝瓜络,用麦麸拌炒至黄色为度,取出,筛去麸皮;
(3)将步骤(2)所得与除壳聚糖、红孩儿和丝瓜络外的其余7味原料药粉碎,过40目筛,加入7味原料药8倍的水煎煮1小时,滤出药液,继续加入7味原料药6倍的水煎煮1小时,滤出药液,合并两次药液;
(4)将步骤(1)和步骤(3)所得混合,以4200r/min离心分离18分钟,取上层液,浓缩成相对密度为0.65的药液,备用;
(5)将壳聚糖溶于体积分数为2%的醋酸溶液中,壳聚糖的脱乙酰度为95%,配制成质量分数为2%的壳聚糖溶液,过滤,除去不溶物;
(6)将步骤(4)和步骤(5)所得混合,充分搅拌,加入甘油和醋酸钠,甘油的体积为步骤(4)和步骤(5)所得总体积的1/5,醋酸钠的质量为壳聚糖质量的1/20,混合均匀,得纺丝原液,将纺丝原液挤出喷丝孔进入凝固浴,纺丝原液凝固析出而形成纤维,将所得纤维切成长度为5cm丝束,于盖尔梳理机开纤梳理,使之成棉絮状纤维,重新置于盖尔梳理机上辊压打片,使其厚度为80g/m2,即得。
典型病例一:吴XX,男,32岁。吴先生在骑车时踩空,让脚踏板拉在腿上两道长伤口,伴有疼痛。到附近诊所给予消炎以及涂抹药膏后,包扎了普通纱布,血流仍不止。使用本实施例所得具有抗菌消炎功效的医用无纺布包扎伤口,25秒后血流止,疼痛很快消失。伤口愈合后,未留伤疤,未出现感染以及粘连现象。
实施例3具有抗菌消炎功效的医用无纺布,取下述重量份的主要原料制备而成(每份为500g):壳聚糖55份,蒲公英4份,红孩儿5份,葛根3份,丝瓜络7份,丁葵草4份,两面针7份,地皮消5份,太白菊3份,母猪藤根5份,甘油适量,醋酸钠适量。
上述具有抗菌消炎功效的医用无纺布的制备方法是:
(1)将红孩儿研成粗粉,过50目筛,加醋拌匀,醋的用量为红孩儿重量的1/3,闷2.5小时,晒干,粉碎,放入锅中文火炒12分钟,加入红孩儿重量7倍的水,煎煮1小时,滤出药液;
(2)将丝瓜络洗净,晒干,切成段,先将锅用中火烧热,撒入麦麸,待冒烟时,投入切成段的丝瓜络,用麦麸拌炒至黄色为度,取出,筛去麸皮;
(3)将步骤(2)所得与除壳聚糖、红孩儿和丝瓜络外的其余7味原料药粉碎,过40目筛,加入7味原料药5.5倍的水煎煮1小时,滤出药液,继续加入7味原料药5倍的水煎煮1小时,滤出药液,合并两次药液;
(4)将步骤(1)和步骤(3)所得混合,以4200r/min离心分离18分钟,取上层液,浓缩成相对密度为0.5的药液,备用;
(5)将壳聚糖溶于体积分数为2%的醋酸溶液中,壳聚糖的脱乙酰度为80%,配制成质量分数为2%的壳聚糖溶液,过滤,除去不溶物;
(6)将步骤(4)和步骤(5)所得混合,充分搅拌,加入甘油和醋酸钠,甘油的体积为步骤(4)和步骤(5)所得总体积的1/5,醋酸钠的质量为壳聚糖质量的1/20,混合均匀,得纺丝原液,将纺丝原液挤出喷丝孔进入凝固浴,纺丝原液凝固析出而形成纤维,将所得纤维切成长度为5cm丝束,于盖尔梳理机开纤梳理,使之成棉絮状纤维,重新置于盖尔梳理机上辊压打片,使其厚度为50g/m2,即得。
典型病例二:陈XX,女,46岁。陈女士在做菜时,左手食指不慎被菜刀割伤,出现5cm左右的伤口,到附近医院进行处理后,最后包上了纱布。在自己换纱布时,纱布跟伤口粘合在一起,把伤口撕裂了,血流不止。遂到医院就诊。使用本实施例所得具有抗菌消炎功效的医用无纺布包扎伤口,20s后血止住。再次更换时,未出现伤口和无纺布之间的粘合。伤口愈合后,未留伤疤,未出现感染以及粘连现象。
实施例4具有抗菌消炎功效的医用无纺布,取下述重量份的主要原料制备而成(每份为500g):壳聚糖65份,蒲公英6份,红孩儿7份,葛根4份,丝瓜络9份,丁葵草5份,两面针9份,地皮消6份,太白菊4份,母猪藤根6份,甘油适量,醋酸钠适量。
上述具有抗菌消炎功效的医用无纺布的制备方法是:
(1)将红孩儿研成粗粉,过50目筛,加醋拌匀,醋的用量为红孩儿重量的3/5,闷3.5小时,晒干,粉碎,放入锅中文火炒16分钟,加入红孩儿重量8倍的水,煎煮1小时,滤出药液;
(2)将丝瓜络洗净,晒干,切成段,先将锅用中火烧热,撒入麦麸,待冒烟时,投入切成段的丝瓜络,用麦麸拌炒至黄色为度,取出,筛去麸皮;
(3)将步骤(2)所得与除壳聚糖、红孩儿和丝瓜络外的其余7味原料药粉碎,过40目筛,加入7味原料药7倍的水煎煮1小时,滤出药液,继续加入7味原料药5.6倍的水煎煮1小时,滤出药液,合并两次药液;
(4)将步骤(1)和步骤(3)所得混合,以4200r/min离心分离18分钟,取上层液,浓缩成相对密度为0.6的药液,备用;
(5)将壳聚糖溶于体积分数为2%的醋酸溶液中,壳聚糖的脱乙酰度为90%,配制成质量分数为2%的壳聚糖溶液,过滤,除去不溶物;
(6)将步骤(4)和步骤(5)所得混合,充分搅拌,加入甘油和醋酸钠,甘油的体积为步骤(4)和步骤(5)所得总体积的1/5,醋酸钠的质量为壳聚糖质量的1/20,混合均匀,得纺丝原液,将纺丝原液挤出喷丝孔进入凝固浴,纺丝原液凝固析出而形成纤维,将所得纤维切成长度为5cm丝束,于盖尔梳理机开纤梳理,使之成棉絮状纤维,重新置于盖尔梳理机上辊压打片,使其厚度为70g/m2,即得。
典型病例三:王XX,女,27岁。王女士进行双眼皮手术后,用本实施例所得具有抗菌消炎功效的医用无纺布包扎伤口,伤口未出现感染、粘连现象,伤口愈合良好。
实施例5具有抗菌消炎功效的医用无纺布,取下述重量份的主要原料制备而成(每份为500g):壳聚糖62份,蒲公英5份,红孩儿6份,葛根3.5份,丝瓜络8份,丁葵草4.5份,两面针8份,地皮消5.5份,太白菊3.5份,母猪藤根5.5份,甘油适量,醋酸钠适量。
上述具有抗菌消炎功效的医用无纺布的制备方法是:
(1)将红孩儿研成粗粉,过50目筛,加醋拌匀,醋的用量为红孩儿重量的2/5,闷3小时,晒干,粉碎,放入锅中文火炒14分钟,加入红孩儿重量7.5倍的水,煎煮1小时,滤出药液;
(2)将丝瓜络洗净,晒干,切成段,先将锅用中火烧热,撒入麦麸,待冒烟时,投入切成段的丝瓜络,用麦麸拌炒至黄色为度,取出,筛去麸皮;
(3)将步骤(2)所得与除壳聚糖、红孩儿和丝瓜络外的其余7味原料药粉碎,过40目筛,加入7味原料药6.2倍的水煎煮1小时,滤出药液,继续加入7味原料药5.2倍的水煎煮1小时,滤出药液,合并两次药液;

(5)将壳聚糖溶于体积分数为2%的醋酸溶液中,壳聚糖的脱乙酰度为88%,配制成质量分数为2%的壳聚糖溶液,过滤,除去不溶物;
(6)将步骤(4)和步骤(5)所得混合,充分搅拌,加入甘油和醋酸钠,甘油的体积为步骤(4)和步骤(5)所得总体积的1/5,醋酸钠的质量为壳聚糖质量的1/20,混合均匀,得纺丝原液,将纺丝原液挤出喷丝孔进入凝固浴,纺丝原液凝固析出而形成纤维,将所得纤维切成长度为5cm丝束,于盖尔梳理机开纤梳理,使之成棉絮状纤维,重新置于盖尔梳理机上辊压打片,使其厚度为60g/m2,即得。
本发明所用主要原料的作用如下:
壳聚糖:壳聚糖是一种天然高分子聚合物,属于氨基多糖,学名为[(1.4)-2-乙酰氨基-2-脱氧-β-D-葡萄糖]。是至今为止发现的唯一带阳离子电荷的碱性多糖,壳聚糖在自然界中广泛存在于低等生物菌类,藻类的细胞,节支动物虾、蟹、昆虫等的外壳中。生物相容性好、毒性低、可生物降解,广泛应用于食品、医药、保健、生物工程等领域。1、抗菌作用。壳聚糖对多种细菌的生长具有抑制作用,表现出类似抗生素的特征。研究表明,壳聚糖对表皮葡萄球菌、金黄色葡萄球菌、大肠埃希杆菌、铜绿假单胞菌、白色念珠菌等实验菌株均具有抑制作用,甚至对一般抗生素难以抑制的白色念珠菌也有抑制作用。壳聚糖的抑菌抗菌机制,一般认为大分子壳聚糖通过自身的所带的正电荷与微生物细胞膜所携带的负电荷的相互作用,破坏细菌细胞壁原有结构,造成细胞成分的泄漏而起到抗菌作用。而小分子的壳聚糖,通过渗透进入细胞内与带有阴离子的生物大分子发生类似“絮凝”作用,扰乱细胞的正常生理功能,阻断DNA的生物合成,从而抑制细菌的繁殖和生长。2、促进皮肤组织愈合和修复。壳聚糖可促进上皮细胞的再生,通过介导细胞增殖而促进伤口愈合,通过温和的急性炎性反应吸引大量的多形核细胞和巨噬细胞以清除组织碎片和血凝块。壳聚糖可促进纤维细胞的迁移,对基质细胞有趋化、迁移、激活作用,并加速细胞增殖和组织重塑过程,促进皮肤组织修复。壳聚糖的降解产物N-乙酰葡萄糖胺(NAG)对组织的疤痕修复非常重要。3、止血作用。壳聚糖本身具有止血作用。同时,壳聚糖通过其与红细胞膜之间的相互作用,主要是对红细胞的凝集作用而实现止血。其止血机理是:带正电荷的壳聚糖与细胞表面带负电荷的神经氨酸残基的受体发生相互作用,促进血小板的聚集,激活凝血系统,交联红细胞形成血块。而且壳聚糖具有明显的膜形成作用,可以加速伤口愈合和防止大出血。4、成膜性。壳聚糖以其氢键相互交联成网状结构,是一种理想的成膜物。对壳聚糖成膜特性的研究表明,成膜性与其凝胶的粘度相关。粘度随浓度的增加而增大,呈典型亲水性胶质特性。壳聚糖的止血、抑菌、抗菌性、生物相容性、促进伤口愈合以及易于形成凝胶的性质赋予它用作止血材料的良好性能。
蒲公英:基原:为菊科植物蒲公英、碱地蒲公英、东北蒲公英、异苞蒲公英、亚洲蒲公英、红梗蒲公英等同属多种植物的全草。药性:味苦,甘,性寒。归肝、胃经。功用主治:清热解毒,消痈散结。主乳痈,肺痈,肠痈,痄腮,疔毒疮肿,目赤肿痛,感冒发热,咳嗽,咽喉肿痛,胃火,肠炎,痢疾,肝炎,胆囊炎,尿路感染,蛇虫咬伤。
红孩儿:基原:为秋海棠科植物裂叶秋海棠的全草。药性:甘、酸,寒。功用主治:清热解毒,散瘀消肿。主肺热咳嗽,疔疮痈肿,痛经,闭经,风湿热痹,跌打肿痛,蛇蛟伤。
葛根:基原:为豆科植物野葛的干燥根。药性:甘、辛,凉。归脾、胃经。功用主治:解肌退热,生津,透疹,升阳止泻。用于外感发热头痛、项背强痛,口渴,消渴,麻疹不透,热痢,泄泻,高血压颈项强痛。
丝瓜络:基原:为葫芦科植物丝瓜或奥丝瓜成熟果实的维管束。药性:味苦,性凉。归肺、肝、胃经。功用主治:通经活络,解毒消肿。主胸胁疼痛,风湿痹痛,经脉拘挛,乳汁不通,肺势咳嗽,痈肿疮毒,乳痈。
丁葵草:基原:豆科丁葵草属植物丁葵草的全草。药性:苦,甘,凉。功用主治:清热解毒,凉血散瘀,除湿利湿。主治风热感冒,咽痛,疮疡肿毒,毒蛇咬伤,黄疸,泄泻,疳积。
两面针:基原:为芸香科植物两面针的干燥根。药性:苦、辛,平;有小毒。归肝、胃经。功用主治:行气止痛,活血化瘀,祛风通络。用于气滞血瘀引起的跌打损伤、风湿痹痛、胃痛、牙痛,毒蛇咬伤;外治汤火烫伤。
地皮消:基原:为爵床科植物地皮消的全草。药性:味微苦,辛,性凉。功用主治:清热解毒,散瘀消肿。主肺热咳嗽,咽喉肿痛,痄腮,瘰疬,痈肿疮毒,跌打骨折,外伤感染。
太白菊:基原:为菊科植物萎软紫菀的全草。药性:味苦;微辛;性凉。功用主治:清热止咳。主肺热咳嗽;肺痈;百日咳。
母猪藤根:基原:为葡萄科植物母猪藤的根。药性:辛,凉,有毒。归肺、脾经。功用主治:清热解毒。主肺痈,疮疖。
甘油:甘油是一种无色或微黄色透明粘稠液体,无毒、无臭、无腐蚀性,具有较强的吸湿性,可与无机酸和有机酸反应生成多种酯类,在一定条件下可形成聚合甘油。在自然界中甘油主要以甘油酯的形式广泛存在于动植物体内,在棕榈油和其他极少数油脂中含有少量甘油。甘油是肥皂工业的副产物,也可用特种酵母发酵糖蜜制得。也可以丙烯为原料合成甘油。以油脂为原料制取肥皂时可得到甘油。也可用发酵或人工合成法制取。甘油可用于制造硝化甘油,醇酸树脂等。也可用作飞机和汽车液体燃料的抗冻剂,玻璃,纸的增塑剂以及化妆品、皮革、烟草、纺织品等的吸湿剂。在实验室中可用以保存标本。甘油大量用作化工原料,用于制造合成树脂、塑料、油漆、硝酸甘油、油脂和蜂蜡等,还用于制药、香料、化妆品、卫生用品等工业中。甘油(丙三醇)是重要的轻化工原料。在化学工业中用于产生环氧氯丙烷、改性醇酸树脂和酚醛树脂等。在医药工业中用于制取润滑剂。
食品工业中作甜味剂、保湿剂。烟草的吸湿剂和溶剂。在国防工业中是硝化炸药甘油的原料。飞机和汽车燃料的抗冻剂。在日化工业中用于牙膏和香精的生产。此外,也广泛用在造纸、皮革、玻璃纸、纺织工业中。
醋酸钠:无色无味的结晶体,在空气中可被风化,可燃。易溶于水,微溶于乙醇,不溶于乙醚。用途:用作照相、印染、化学试剂及肉类防腐,制作暖手袋等。
本发明所得具有抗菌消炎功效的医用无纺布(实施例5)体外降解试验
分别将本发明所得具有抗菌消炎功效的医用无纺布剪2cm×2cm的小块,分为4组,每组4块,干至恒重称其重量,置于试管中,加入3mL1%溶菌酶溶液,置37℃恒温振荡水浴。溶菌酶溶液每48h置换,分别在第1、2、4、6周取出1组,经蒸馏水洗净,80℃烘干4h后称其干重,计算其降解率。
降解率=(实验前重量一实验后重量)/实验前重量×100%。
试验表明,各组无纺布均有不同程度的重量减轻,其降解率随时间延长而增加,但其外形无明显变化。体外在1%溶菌酶溶液中1周平均降解6.74%,2周降解11.35%,4周降解15.67%,6周降解20.15%。
本发明所得具有抗菌消炎功效的医用无纺布体内降解及生物相容性实验
成年兔16只,随机分为4组,每组4只,将本发明所得具有抗菌消炎功效的医用无纺布切成2cm×2cm的小块,经环氧乙烷消毒后,在每只兔皮肤与深筋膜之间植入无纺布块。分别于2、4、8、12周各处死1组动物,取无纺布及其周围组织观察其大体变化。
试验表明,各组无纺布大致保持了植入前形状。随时间推移,无纺布逐渐变薄,表面变粗糙,4周,无纺布缩小至约为植入前的3/4;无纺布的周围有纤维组织包绕,并有纤维组织长入其内,难以将纤维组织与无纺布分离。
该无纺布在体外降解速度缓慢(4周降解15.67%,而在体内4周其体积约为植入前的3/4),说明该无纺布在体内的降解速度快于体外,具有良好的生物可降解性。植入体内后,纤维结缔组织长入无纺布内,证明该无纺布具有良好的生物相容性。
本发明所得医用无纺布是以壳聚糖、蒲公英、红孩儿和葛根等主要原料制备而成。其中,壳聚糖的止血、抑菌、抗菌性、生物相容性、促进伤口愈合以及易于形成凝胶的性质赋予它用作止血材料的良好性能。蒲公英清热解毒,消痈散结。红孩儿清热解毒,散瘀消肿。葛根解肌退热,生津,透疹,升阳止泻。丝瓜络通经活络,解毒消肿。丁葵草清热解毒,凉血散瘀,除湿利湿。两面针行气止痛,活血化瘀,祛风通络。地皮消清热解毒,散瘀消肿。太白菊清热止咳。母猪藤根清热解毒。本发明所得医用无纺布具有良好的止血、抗菌消炎、消肿止痛等功效,具有良好的生物可降解性和良好的生物相容性,止血速度快,能够缩短治疗时间,避免伤口感染,促进伤口愈合,具有良好的透气性。本发明所得医用无纺布安全有效,效果显著且无任何毒副作用。
最后应说明的是,实施例只是本发明最优的具体实施方式而已,并不用于限制本发明,尽管参照前述实施例对本发明进行了详细的说明,对于本领域的技术人员来说,其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分技术特征进行等同替换。凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (10)
1.具有抗菌消炎功效的医用无纺布,其特征在于,取下述重量配比的主要原料制备而成:壳聚糖50-70,蒲公英3-7,红孩儿4-8,葛根2-5,丝瓜络6-10,丁葵草3-6,两面针6-10,地皮消4-8,太白菊2-5,母猪藤根3-7,甘油适量,醋酸钠适量。
2.如权利要求1所述的具有抗菌消炎功效的医用无纺布,其特征在于,取下述重量配比的主要原料制备而成:壳聚糖55-65,蒲公英4-6,红孩儿5-7,葛根3-4,丝瓜络7-9,丁葵草4-5,两面针7-9,地皮消5-6,太白菊3-4,母猪藤根5-6,甘油适量,醋酸钠适量。
3.如权利要求1所述的具有抗菌消炎功效的医用无纺布,其特征在于,取下述重量配比的主要原料制备而成:壳聚糖62,蒲公英5,红孩儿6,葛根3.5,丝瓜络8,丁葵草4.5,两面针8,地皮消5.5,太白菊3.5,母猪藤根5.5,甘油适量,醋酸钠适量。
4.如权利要求1~3任一所述的具有抗菌消炎功效的医用无纺布的制备方法,其特征在于,步骤如下:
(1)将红孩儿研成粗粉,过50目筛,加醋拌匀,醋的用量为红孩儿重量的1/4-2/3,闷2-4小时,晒干,粉碎,放入锅中文火炒10-18分钟,加入红孩儿重量6.7-8.9倍的水(优选的,加入红孩儿重量7-8倍的水;更加优选的,加入红孩儿重量7.5倍的水),煎煮1小时,滤出药液;
(2)将丝瓜络洗净,晒干,切成段,先将锅用中火烧热,撒入麦麸,待冒烟时,投入切成段的丝瓜络,用麦麸拌炒至黄色为度,取出,筛去麸皮;
(3)将步骤(2)所得与除壳聚糖、红孩儿和丝瓜络外的其余7味原料药粉碎,过40目筛,加入7味原料药5-8倍的水煎煮1小时(优选的,加入7味原料药5.5-7倍的水煎煮1小时;更加优选的,加入7味原料药6.2倍的水煎煮1小时),滤出药液,继续加入7味原料药4.5-6倍的水煎煮1小时(优选的,继续加入7味原料药5-5.6倍的水煎煮;更加优选的,继续加入7味原料药5.2倍的水煎煮1小时),滤出药液,合并两次药液;
(4)将步骤(1)和步骤(3)所得混合,以4200r/min离心分离18分钟,取上层液,浓缩成相对密度为0.45-0.65的药液,备用;
(5)将壳聚糖溶于体积分数为2%的醋酸溶液中,壳聚糖的脱乙酰度为75%-95%,配制成质量分数为2%的壳聚糖溶液,过滤,除去不溶物;
(6)将步骤(4)和步骤(5)所得混合,充分搅拌,加入甘油和醋酸钠,甘油的体积为步骤(4)和步骤(5)所得总体积的1/5,醋酸钠的质量为壳聚糖质量的1/20,混合均匀,得纺丝原液,将纺丝原液挤出喷丝孔进入凝固浴,纺丝原液凝固析出而形成纤维,将所得纤维切成长度为5cm丝束,于盖尔梳理机开纤梳理,使之成棉絮状纤维,重新置于盖尔梳理机上辊压打片,使其厚度为45-80g/m2,即得。
5.如权利要求4所述的具有抗菌消炎功效的医用无纺布的制备方法,其特征在于,步骤(1)中醋的用量为红孩儿重量的1/3-3/5(优选醋的用量为红孩儿重量的2/5)。
6.如权利要求4所述的具有抗菌消炎功效的医用无纺布的制备方法,其特征在于,步骤(1)中闷2.5-3.5小时(优选闷3小时)。
7.如权利要求4所述的具有抗菌消炎功效的医用无纺布的制备方法,其特征在于,步骤(1)中放入锅中文火炒12-16分钟(优选放入锅中文火炒14分钟)。
8.如权利要求4所述的具有抗菌消炎功效的医用无纺布的制备方法,其特征在于,步骤(4)中浓缩成相对密度为0.5-0.6的药液(优选浓缩成相对密度为0.55的药液)。
9.如权利要求4所述的具有抗菌消炎功效的医用无纺布的制备方法,其特征在于,步骤(5)中壳聚糖的脱乙酰度为80%-90%(优选壳聚糖的脱乙酰度为88%)。
10.如权利要求4所述的具有抗菌消炎功效的医用无纺布的制备方法,其特征在于,步骤(6)中使其厚度为50-70g/m2(优选使其厚度为60g/m2)。
