Abstract
一种桔远止咳微囊的制备方法,包括以下步骤:取明胶置配料锅中,加水并加热至70℃使明胶溶解,得明胶溶液,加入桔远止咳干膏粉,冷却至50‑55℃,再加入盐酸麻黄碱、薄荷脑、桔子油,最后用均质机充分混匀,制成混合溶液;将所述混合溶液中加入醋酸溶液调pH至3.6‑3.8,加入Na2SO4溶液,搅匀,得混合物;再加适量水,搅匀,得混悬液,启动冷水循环迅速降温至10℃以下,加入异丙醇搅拌,再加入氢氧化钠溶液调pH至8‑9,继续搅拌,静置20‑24小时,分离固液相,固相即为微囊。本发明可明显降低制剂的引湿性,药物生产及贮存过程不易受潮变质,能防止挥发油渗漏,降低贮存过程挥发性成份的损失,提高药物的稳定性。

技术领域
本发明涉及中成药制剂中间体的制备,具体是一种桔远止咳微囊的制备方法。
背景技术
桔远止咳片是由桔梗、远志流浸膏、百步流侵膏、盐酸麻黄碱、薄荷脑、桔子油、甘草七味中药组成。桔远止咳片收藏于国家中成药标准汇编.内科肺系(二)分册370 页,标准代号为WS-11153(ZD-1153)-2002,具有祛痰止咳功效,用于治疗咳嗽痰多,咳痰不爽。标准中桔远止咳片的处方为桔梗225g、远志流浸膏75mL、百步流侵膏50mL、盐酸麻黄碱5g、薄荷脑0.67g、桔子油1.3g和甘草113g。其制备方法为:将桔梗、甘草粉碎成细粉、过筛,加入适量蔗糖、糊精,混匀;另取盐酸麻黄碱加水使溶解后,加入远志流浸膏、百步流侵膏,混匀,再加入上述细粉及10%淀粉浆适量,制成颗粒,干燥,取桔子油及薄荷脑加乙醇适量使溶解,与上述颗粒混匀,压片,制成1000片,即得。
目前市售的桔远止咳药物基本为素片及薄膜衣片,根据市场销售、患者反馈意见及成品留样检验情况,因本产品含薄荷脑、桔子油易挥发性成份,贮存过程挥发油会慢慢渗出,导致成品外观改变,甚至出现霉点,使许多患者不敢放心使用。同时桔梗和甘草是以药材原粉直接入药的,虽然经过高温灭菌处理,但处理过程难免对药材有效成分有影响,从而影响最终疗效,再加上药材原粉不易完全清除细菌,使成品有微生物超标的风险。并且桔远止咳片剂在贮存过程极易受潮变质,从而影响药效的发挥,也直接影响着治疗的效果。因此,有必要制备能防止挥发油渗漏、不易受潮的中间体微囊,以满足临床患者的多种需求。
发明内容
本发明针对上述存在的问题,提供一种桔远止咳微囊的制备方法。本发明将药物微囊化后,可明显降低制剂吸湿受潮、减少挥发油渗漏,提高药物的稳定性。同时微囊的释放可控,峰谷血药浓度波动小、胃肠道刺激轻、疗效长、毒性降低、安全,可提高药物生物利用度和临床疗效。该制备方法工艺简单,生产成本低,耗能低,易实现工业化生产。

1.称取桔梗225份、甘草113份药材,置提取罐中,加水煎煮2-3次,每次2-3 小时,过滤,合并滤液,浓缩至药材与浸膏质量比为1:1-1.1,得浸膏;
2.将所述浸膏置夹层锅中,分别加入加远志流浸膏75份、百部流浸膏50份,煮沸,搅拌20-30分钟,加入糊精,混匀,得混合浸膏,置喷雾干燥器的沸腾制粒锅内,预热至80-90℃,启动风机,间歇喷入所述混合浸膏制粒,粉碎,得桔远止咳干膏粉;
3.取明胶10-20份,置配料锅中,加水150份,加热至70℃使明胶溶解,得明胶溶液,加入所述桔远止咳干膏粉,边加边搅拌,冷却至50-55℃,再加入盐酸麻黄碱 5份、薄荷脑0.67份、桔子油1.3份,最后用均质机充分混匀,制成混合溶液;
4.在50-55℃条件下,在所述混合溶液中加入醋酸溶液调pH至3.6-3.8,加入重量百分比为10%的Na2 SO4 溶液1000份,搅匀,得混合物;
5.在混合物中加入体积为所述混合物2-4倍量的水,搅匀,得混悬液,启动冷水循环迅速降温至10℃以下,加入占混悬液总体积6-8%的异丙醇,搅拌10-15分钟,再加入氢氧化钠溶液调pH至8-9,继续搅拌50-60分钟,静置20-24小时,分离固液相,固相即为微囊,用异丙醇清洗微囊表面,得到固体物,将固体物在40-50℃下烘 24-28h,即得桔远止咳微囊。
优选地,以上所述步骤1的加水量为药材总重量的6-8倍量。
优选地,以上所述步骤2糊精的加入量为40-60份。
优选地,以上所述步骤3中桔远止咳干膏粉的水分≤6%。
优选地,以上所述步骤3中均质机均质时间为20-30分钟,转速为 1300-1500r/min。
优选地,以上所述步骤4中醋酸溶液为体积百分比为10%的醋酸溶液。
优选地,以上所述步骤5中氢氧化钠溶液为重量百分比为10%的氢氧化钠溶液。
以上所述桔远止咳微囊的制备方法制成的微囊,所述微囊可用于任何剂型的生产,优选用于胶囊剂、片剂的生产。
以上所述桔远止咳微囊的制备方法,所述远志流浸膏参照《中国药典》2015年版一部“远志流浸膏”项下的制备方法。所述百部流浸膏参照广东省药品标准1985年上册“百部流浸膏”,其制备方法为:取百部1000g,用55%乙醇浸渍48小时,以每分钟1-3ml的速度缓缓渗漉,最初收集的漉液850ml,另器保存,继续渗漉至流出液几乎无色和无苦味时,停止渗漉。将续漉液减压,低温蒸发至稠膏状,加最初收集的渗漉液850ml,混合后,加乙醇适量使成1000ml,静置,滤过,即得。所述桔子油,也称橘子油,是从桔皮中提取得到油。
与现有技术相比,本发明的有益效果:
1、本发明方法将桔远止咳微囊化后可明显降低制剂的引湿性,药物生产及贮存过程不易受潮变质,能提高制剂的稳定性,微囊流动性好,利于制剂生产。
2、本发明方法将桔远止咳微囊后具有一定的缓控释性能,释放度与常规的桔远止咳片相比明显降低,药物释放缓慢且平稳,既可防止药物在胃内失活或减少对胃的刺激性,又能提高药物的生物利用度。
3、将薄荷脑、桔子油易挥发成份制成微囊后,能防止挥发油渗漏,可降低贮存过程挥发性成份的损失,提高药物稳定性。
4、将桔梗、甘草的原粉直接入药改为提取浸膏入药,避免了微生物极易超标的风险。
5、本发明方法得到的桔远止咳微囊的包封率达到92%以上,载药量大于42%,微囊收率大于90%,说明本发明制备工艺可行、可靠。
6、本发明方法的工艺简单、容易操作、耗能低、省时、生产成本低,容易实现工业化生产。
具体实施方式
下面将结合具体实施方式对本发明进一步说明,但不限于本发明的保护范围。
一、桔远止咳微囊的制备
实施例1
一种桔远止咳微囊的制备方法,包括以下步骤:
1.称取桔梗225份、甘草113份药材,置提取罐中,加入药材总重量8倍量的水,煎煮2次,每次3小时,过滤,合并滤液,浓缩至药材与浸膏质量比为1:1.1,得浸膏;
2.将所述浸膏置夹层锅中,分别加入远志流浸膏75份、百部流浸膏50份,煮沸,搅拌20分钟,加入40份糊精,混匀,得混合浸膏,置喷雾干燥器的沸腾制粒锅内,预热至80℃,启动风机,间歇喷入所述混合浸膏制粒,粉碎,得桔远止咳干膏粉,所述桔远止咳干膏粉的水分为5.0%;
3.取明胶10份,置配料锅中,加水150份,加热至70℃使明胶溶解,得明胶溶液,加入所述桔远止咳干膏粉,边加边搅拌,冷却至50℃,再加入盐酸麻黄碱5份、薄荷脑0.67份、桔子油1.3份,最后用均质机在转速为1300r/min下转动30min,充分混匀,制成混合溶液;
4.在50℃条件下,在所述混合溶液中加入体积百分比为10%的醋酸溶液调pH至3.6,加入重量百分比为10%的Na2 SO4 溶液1000份,搅匀,得混合物;
5.在混合物中加入体积为所述混合物2倍量的水,搅匀,得混悬液,启动冷水循环迅速降温至10℃,加入占混悬液总体积6%的异丙醇,搅拌15分钟,再加入重量百分比为10%的氢氧化钠溶液调pH至8,继续搅拌50分钟,静置24小时,分离固液相,固相即为微囊,用异丙醇清洗微囊表面,得到固体物,将固体物在40℃下烘28h,即得桔远止咳微囊。
实施例2
一种桔远止咳微囊的制备方法,包括以下步骤:
1.称取桔梗225份、甘草113份药材,置提取罐中,加入药材总重量6倍量的水煎煮3次,每次2小时,过滤,合并滤液,浓缩至药材与浸膏质量比为1:1,得浸膏;
2.将所述浸膏置夹层锅中,分别加入远志流浸膏75份、百部流浸膏50份,煮沸,搅拌25分钟,加入50份糊精,混匀,得混合浸膏,置喷雾干燥器的沸腾制粒锅内,预热至85℃,启动风机,间歇喷入所述混合浸膏制粒,粉碎,得桔远止咳干膏粉,所述桔远止咳干膏粉的水分为5.5%;
3.取明胶20份,置配料锅中,加水150份,加热至70℃使明胶溶解,得明胶溶液,加入所述桔远止咳干膏粉,边加边搅拌,冷却至50℃,再加入盐酸麻黄碱5份、薄荷脑0.67份、桔子油1.3份,最后用均质机在转速为1500r/min下转动25min,充分混匀,制成混合溶液;
4.在50℃条件下,在所述混合溶液中加入体积百分比为10%的醋酸溶液调pH至3.7,加入重量百分比为10%的Na2 SO4 溶液1000份,搅匀,得混合物;
5.在混合物中加入体积为所述混合物3倍量的水,搅匀,得混悬液,启动冷水循环迅速降温至8℃,加入占混悬液总体积7%的异丙醇,搅拌10分钟,再加入重量百分比为10%的氢氧化钠溶液调pH至8.5,继续搅拌60分钟,静置22小时,分离固液相,固相即为微囊,用异丙醇清洗微囊表面,得到固体物,将固体物在45℃下烘26h,即得桔远止咳微囊。
实施例3
一种桔远止咳微囊的制备方法,包括以下步骤:
1.称取桔梗225份、甘草113份药材,置提取罐中,加入药材总重量7倍的水煎煮3次,每次2小时,过滤,合并滤液,浓缩至药材与浸膏质量比为1:1,得浸膏;
2.将所述浸膏置夹层锅中,分别加入远志流浸膏75份、百部流浸膏50份,煮沸,搅拌30分钟,加入60份糊精,混匀,得混合浸膏,置喷雾干燥器的沸腾制粒锅内,预热至90℃,启动风机,间歇喷入所述混合浸膏制粒,粉碎,得桔远止咳干膏粉,所述桔远止咳干膏粉的水分为5.8%;
3.取明胶15份,置配料锅中,加水150份,加热至70℃使明胶溶解,得明胶溶液,加入所述桔远止咳干膏粉,边加边搅拌,冷却至55℃,再加入盐酸麻黄碱5份、薄荷脑0.67份、桔子油1.3份,最后用均质机在转速为1500r/min下转动20min,充分混匀,制成混合溶液;
4.在55℃条件下,在所述混合溶液中加入体积百分比为10%的醋酸溶液调pH至3.8,加入重量百分比为10%的Na2 SO4 溶液1000份,搅匀,得混合物;
5.在混合物中加入体积为所述混合物4倍量的水,搅匀,得混悬液,启动冷水循环迅速降温至6℃,加入占混悬液总体积8%的异丙醇,搅拌15分钟,再加入重量百分比为10%的氢氧化钠溶液调pH至9,继续搅拌60分钟,静置20小时,分离固液相,固相即为微囊,用异丙醇清洗微囊表面,得到固体物,将固体物在50℃下烘24h,即得桔远止咳微囊。
二、桔远止咳微囊的质量评价
1.测定桔远止咳微囊的包封率
采用高效液相色谱法检测。包封率=微囊中包封的盐酸麻黄碱的质量/微囊中包封与未包封的盐酸麻黄碱的含量×100%。以盐酸麻黄碱为对照品,加0.01mol/L盐酸溶液制成每1ml含50μg的溶液。分别取实施例1-3的桔远止咳微囊各0.1g,精密称定,加0.01mol/L盐酸溶液20ml,超声处理15分钟,摇匀,滤过,取滤液。检测波长为 210nm。

从表中可以看出,本发明桔远止咳微囊的包封率均达到92%以上。
2.测定桔远止咳微囊的载药量
分别称取实施例1-3的桔远止咳微囊各5g,加适量水超声20min,加入乙酸乙酯进行萃取2次,每次50ml,滤过,合并乙酸乙酯层,放在单口瓶旋蒸,然后放在105 ℃的烘干箱烘干,把烘干的单口瓶放在真空干燥器中,冷却至室温,称重,计算单口瓶中微囊里面桔远止咳干膏粉的重量,根据公式:载药量(%)=微囊里面桔远止咳重量/桔远止咳微囊重量×100%,计算载药量,计算结果见表2。

3.测定微囊的释放度
分别取本发明实施例1-3样品及市售桔远止咳片,根据中国药典2015版释放度测定法(附录XD第一法),按照2.4所述方法操作测定桔远止咳微囊的释放度。溶出条件:37±0.5℃,转速80rpm,分别在10min、20min、40min、80min取样。按照包封率测定方法检测盐酸麻黄碱含量,试剂加入完毕开始计时,10min开始测量,连续测定80min,测定结果如表3所示。

从表中可以看出,桔远止咳微囊释放比采用普通制法制成的桔远止咳片缓慢,其缓控释效果明显。
三、桔远止咳片的制备
实施例4
取实施例1的桔远止咳微囊3.2kg,加淀粉1.8kg,再加糊精适量,混匀,用70%乙醇制成颗粒,压片,包糖衣,即得桔远止咳片。
实施例5
取实施例2的桔远止咳微囊3.2kg,加淀粉1.8kg,再加糊精适量,混匀,用70%乙醇制成颗粒,压片,包糖衣,即得桔远止咳片。
四、桔远止咳胶囊的制备
实施例6
取实施例3的桔远止咳微囊2.8kg,加淀粉2.2kg,再加入适量滑石粉,混合均匀,装入胶囊,即得桔远止咳胶囊。
五、水分测定
取本发明实施例4-6及市售桔远止咳片,置稳定性试验箱,于温度40℃±2℃、相对湿度75%±5%条件下进行6个月加速试验,按照《中国药典》2015年版一部附录水分项下检验,详见表4。

试验结果表明:本发明制备的桔远止咳片剂和胶囊,与市售的桔远止咳片相比,药物的引湿性明显降低,解决了药物在生产及贮存过程极易受潮变质的问题。完成加速6月试验后检查药品的性状,结果为:市售桔远止咳片表面有油斑浸润或变色现象;实施例4、5的表面光滑,无有油斑或变色现象;实施例6胶囊表面光滑,无油斑及胶囊粘连现象。

Claims (5)
1.一种桔远止咳微囊的制备方法,其特征在于,包括以下步骤:
(1) 称取桔梗225 份、甘草113 份药材,置提取罐中,加水煎煮2-3 次,每次2-3 小时,过滤,合并滤液,浓缩至药材与浸膏质量比为1:1-1.1,得浸膏;
(2) 将所述浸膏置夹层锅中,分别加入远志流浸膏75 份、百部流浸膏50 份,煮沸,搅拌20-30 分钟,加入糊精,混匀,得混合浸膏,置喷雾干燥器的沸腾制粒锅内,预热至80-90℃,启动风机,间歇喷入所述混合浸膏制粒,粉碎,得桔远止咳干膏粉;
(3) 取明胶10-20 份,置配料锅中,加水150 份,加热至70℃使明胶溶解,得明胶溶液,加入所述桔远止咳干膏粉,边加边搅拌,冷却至50-55℃,再加入盐酸麻黄碱5 份、薄荷脑0.67 份、桔子油1.3 份,最后用均质机充分混匀,制成混合溶液;
(4) 在50-55℃条件下,在所述混合溶液中加入醋酸溶液调pH 至3.6-3.8,加入重量百分比为10%的Na2 SO4 溶液1000 份,搅匀,得混合物;
(5) 在混合物中加入体积为所述混合物2-4 倍量的水,搅匀,得混悬液,启动冷水循环迅速降温至10℃以下,加入占混悬液总体积6-8%的异丙醇,搅拌10-15 分钟,再加入氢氧化钠溶液调pH 至8-9,继续搅拌50-60 分钟,静置20-24 小时,弃去上清液,余下滤过,分离固液相,固相即为微囊,用异丙醇清洗微囊表面,得到固体物,将固体物在40-50℃下烘24-28h,即得桔远止咳微囊;
所述步骤(2) 糊精的加入量为40-60 份;
所述步骤(3) 中桔远止咳干膏粉的水分≤ 6%;
所述步骤(3) 中均质机均质时间为20-30 分钟,转速为1300-1500r/min。
2.根据权利要求1 所述桔远止咳微囊的制备方法,其特征在于:所述步骤(1) 的加水量为药材总重量的6-8 倍量。
3.根据权利要求1 所述桔远止咳微囊的制备方法,其特征在于:所述步骤(4) 中醋酸溶液为体积百分比为10%的醋酸溶液。
4.根据权利要求1 所述桔远止咳微囊的制备方法,其特征在于:所述步骤(5) 中氢氧化钠溶液为重量百分比为10%的氢氧化钠溶液。
5.一种如权利要求1 所述桔远止咳微囊的制备方法制成的微囊,其特征在于:所述微囊用于胶囊剂、片剂的生产。
