Abstract
本发明涉及将由slit1、slit2、slit3、robo1、robo2及vilse所组成的群中选择的基因或其表达蛋白质作为有效成分含有的预防或治疗骨折及骨质疏松症的药物组合物;含有所述蛋白质以预测骨折及骨质疏松症发生的标记组合物;含有对所述蛋白质特异性结合的抗体以预测骨折及骨质疏松症发生危险的试剂盒;及通过利用对所述蛋白质特异性结合抗体的抗原‑抗体结合反应以测定slit蛋白质表达水平的步骤,用于预测骨折及骨质疏松症发生危险的信息提供方法。本发明的slit是在细胞及动物模型中增加骨形成,减小骨吸收,并与骨质疏松症的发病率成负相关关系,由此可有效利用为预防或治疗骨折及骨质疏松症的组合物及用于预测骨折及骨质疏松症发生危险的生物标记。

技术领域
本发明涉及包含slit,robo或其表达蛋白质作为有效成分用于预防或治疗骨折或骨质疏松症的药物组合物,利用所述基因或表达蛋白质用于预测骨折及骨质疏松症发生危险的试剂盒,以及用于预测骨折及骨质疏松症发生危险的信息提供方法
背景技术
骨质疏松症是指由于骨形成的无机质和基质的含量过度减少,骨密度减少的状态,因此有骨质疏松症时易于骨折。骨质疏松症是骨密度(bone mineral density;BMD)低以及骨折危险高的最常见代谢性骨病(metabolic bone disease;MBD)(Peacock,M.,etal.,Endocr.Rev.23:303-326,2002;Akhter,M.P.,et al.,Bone.41(1):111-6,2007)。最近,由臀部、脊椎及手腕等的一般性骨质疏松症骨折住院的患者明显地增加(Fogarty,P.,et al.,Maturitas.52Suppl 1:S3-6,2005;Palacios,S.,et al.,Maturitas.15;52Suppl1:S53-60.Review,2005)。尤其是40岁以后的女性在更年期绝经后常见骨质疏松症,70岁以后,不论男女都发生老年人骨质疏松症。
目前,骨质疏松症的诊断是以X射线等的物理方法进行,但这种方法需要大规模的诊断设备,不仅存在使用X射线的安全性等的问题,而且也无法事先预测向后骨密度的减少,即,单靠骨密度的数值具有很难正确地预测有多大的骨质疏松性骨折危险的缺点。
因此,需要建立一种不仅早期发现并诊断绝经后骨质疏松症或老年人骨质疏松症,还包括其它各种骨质疏松症,并预测骨折危险,能有效治疗的迅速简便正确的方法。
对此,本发明人要开发骨折及骨质疏松症治疗机、以及用于预测骨折及骨质疏松症发生危险的标记,不断研究的结果,确认slit3在细胞及动物模型中增加骨形成,减小骨吸收,并与骨质疏松症的发病率成负相关关系,由此完成了本发明。

本发明的目的是提供,将由slit1、slit2、slit3、robo1、robo2及vilse所组成的群中选择的基因或其表达蛋白质作为有效成分含有的预防或治疗骨折及骨质疏松症的药物组合物。
本发明的另一目的是提供,将由slit1、slit2、slit3、robo1、robo2及vilse所组成的群中选择的基因或其表达蛋白质作为有效成分含有的预防或改善骨折及骨质疏松症的食品组合物。

本发明的slit1、slit2、slit3、robo1、robo2或vilse蛋白质不仅包含具有其自然型氨基酸序列的蛋白质,其氨基酸序列变异体也包含于本发明的范围。slit1、slit2、slit3、robo1、robo2或vilse蛋白质的变异体是指slit1、slit2、slit3、robo1、robo2或vilse自然氨基酸序列和一个以上的氨基酸残基通过缺失、插入、非保守性或保守性替换、或这些的组合具有相异序列的蛋白质。整体上不改变分子活性的蛋白质及肽中的氨基酸替换在该领域中早已公布(H.Neurath,R.L.Hill,The Proteins,Academic Press,New York,1979)。最普遍的替换是氨基酸残基Ala/Ser,Val/Ile,Asp/Glu,Thr/Ser,Ala/Gly,Ala/Thr,Ser/Asn,Ala/Val,Ser/Gly,Thy/Phe,Ala/Pro,Lys/Arg,Asp/Asn,Leu/Ile,Leu/Val,Ala/Glu,Asp/Gly之间的替换。根据情况,可由磷酸化(phosphorylation)、硫酸化(sulfation)、乙酰化(acetylation)、糖基化(glycosylation)、甲基化(methylation)、法尼基化(farnesylation)等来修饰(modification)。
所述slit1、slit2、slit3、robo1、robo2或vilse蛋白质或其的变异体可以从自然提取或合成(Merrifleld,J.Amer.chem.Soc.85:2149-2156,1963)或是可由DNA序列为基本的基因重组方法来制造(Sambrook et al,Molecular Cloning,Cold Spring HarbourLaboratory Press,New York,USA,2,1989)。

所述表达载体可以使用将所述slit1、slit2、slit3、robo1、robo2或vilse基因插入并能表达的在本领域公布的表达载体。例如,可以使用pBK-CMV(Staratagene)、pCR3.1(Invitrogen)等的表达载体。

如上所述,表达构建体可以用多种方法使用在本发明的治疗。因此,所述表达构建体可以直接投入在患者需要治疗的部位。
在本发明的其它实施例中,所述药物组合物可以包含增加slit1、slit2、slit3、robo1、robo2或vilse表达的活性因子作为有效成分。
在本发明中,所谓“增加slit1、slit2、slit3、robo1、robo2或vilse表达的活性因子”是指直接或间接地作用在slit1、slit2、slit3、robo1、robo2或vilse上,改善、诱导、刺激、增加slit1、slit2、slit3、robo1、robo2或vilse生物活性的物质。所述物质包含如有机或无机化合物一样的单一化合物,如肽、蛋白质、核酸、碳水化合物及脂质一样的生物体高分子化合物及复数化合物的复合体等。增加所述slit3表达的活性因子可以利用在由slit3表达、活性、或功能的减小所发生疾病的预防、改善及治疗。并不特别限定所述物质使slit1、slit2、slit3、robo1、robo2或vilse活性化的机制。例如,所述物质可以作用为使转录、翻译等的基因表达增加,或使非活性型转换成活性型的机制。优选地,使slit1、slit2、slit3、robo1、robo2或vilse活性化的物质是如肽、蛋白质、核酸、碳水化合物及脂质一样的生物体高分子化合物。对核酸及蛋白质序列已公布的slit3而言,本技术领域的技术人员利用本领域的技术可以制造或筛选作用为诱导剂或活性剂的如有机或无机化合物一样的单一化合物,如肽、蛋白质、核酸、碳水化合物及脂质一样的生物体高分子化合物及复数化合物的复合体等。
在本发明中,所述slit3是在分化的破骨细胞中分泌,在成骨细胞中进行迁移、生存能力、增殖、分化以及增加OPG生成的功能,且具有增加骨形成,抑制骨损失,增加VEGF的表达促进血管形成(angiogenesis)及骨形成的优异效果。另外,根据本发明的slit3功能和活性的减少可能为骨折及骨质疏松症的原因。因此,为使所述基因功能的正常化及活性化,对基因或蛋白质的处理可能成为治疗骨折及骨质疏松症的主要关键。
因此,所述药物组合物是,将含有所述基因的表达载体或含有该表达载体的宿主细胞增殖或激活后投入患者,便可以治疗或预防骨折及骨质疏松症,并且通过所述基因或其表达蛋白质,可以抑制破骨细胞的分化,且促进成骨细胞的分化、增殖、迁移,并治疗骨折及骨质疏松症。
在制造本发明的药物组合物方面,还可以包含通常使用的适量载体、赋形剂及稀释剂。而且,根据通常的方法,可以制作成粉剂、颗粒剂、片剂、胶囊剂、混悬剂、乳剂、糖浆剂、喷雾剂等的口服剂、外用剂、栓剂及无菌注射溶液形式的剂型使用。在本技术领域知晓的合适制剂使用在文献(Remington's Pharmaceutical Science,最近,Mack PublishingCompany,Easton PA)记载的较好。可以包含的载体、赋形剂及稀释剂有乳糖、葡萄糖、蔗糖、山梨糖醇、甘露醇、木糖醇、赤藓糖醇、麦芽糖醇、淀粉、阿拉伯胶、藻酸盐、明胶、磷酸钙、硅酸钙、纤维素、甲基纤维素、微晶纤维素、聚乙烯吡咯烷酮、水、羟基苯甲酸甲酯、羟基苯甲酸丙酯、滑石粉、硬脂酸镁和矿物油等。将所述组合物制剂化时,使用常用的填料、增量剂、粘合剂、湿润剂、崩解剂、表面活性剂等的稀释剂或赋形剂而制造。口服固体制剂包含片剂、丸剂、粉剂、颗粒剂、胶囊剂等,这种固体制剂是在所述组合物中混合至少一种以上的赋形剂,例如淀粉、碳酸钙(calcium carbonate)、蔗糖、乳糖、明胶等而制造。而且,除了简单的赋形剂以外,也使用如硬脂酸镁、滑石粉等润滑剂。口服液相制剂有混悬剂、液剂、乳剂、糖浆剂等,除了常用的简单稀释剂,例如水、液状石蜡以外,可以包含多种赋形剂,例如湿润剂、甜味剂、芳香剂、保存剂等。非口服制剂包含无菌水溶液、非水溶剂、悬浮剂、乳剂、冻干制剂、栓剂。非水溶剂、悬浮剂可以使用丙二醇(propylene glycol),聚乙二醇,像橄榄油的植物油,像醋酸乙酯的可注射酯等。栓剂基质可以使用witepsol、聚乙二醇、吐温(tween)61、可可脂、月桂精脂、甘油明胶等。

本发明药物组合物的优选投入量是根据患者的状态和体重、疾病的程度、药物形式、投入途径及时间而不同,但本技术领域的技术人员可以适当地选择投入量。为了获得较好的效果,本发明的组合物1天可以投入0.001mg/kg至10000mg/kg的量。所述组合物的投入可以一天一次投入,亦可以分成多次投入。
本发明的药物组合物对个体可以通过多种途径投入。可以预料投入的所有方式,例如,通过注射可以投入口腔、直肠或静脉、肌肉、皮下、子宫内、硬膜或脑血管内(intracerebroventricular)。
本发明的组合物用于预防或治疗骨折及骨质疏松症,可以单独使用,或与手术、放射线治疗、激素治疗、化学治疗及生物反应调节剂一起使用。
另外,本发明提供,将由slit1、slit2、slit3、robo1、robo2及vilse组成的群中选择的蛋白质作为有效成分含有的预防或改善骨折及骨质疏松症的食品组合物。
在本发明中,所述‘预防或改善骨折及骨质疏松症的食品组合物’是指具有疾病的预防及改善、生物体防御、免疫、病后康复、老化抑制等生物体调节功能的食品,且长期服用时必须对人体无害才可。
本发明的组合物是以预防及改善骨折及骨质疏松症为目的,可以添加在健康食品中。本发明的蛋白质或增加其表达的活性因子作为食品添加物使用时,可将所述成分直接添加,或与其他食品或食品成分一起使用,并可以根据通常的方法适当地使用。有效成分的混合量可根据使用目的(预防、健康或治疗处理)适当地决定。通常,制造食品或饮料时,本发明的蛋白质或增加其表达的活性因子相对于原料添加15重量%以下,优选地添加10重量%以下的量。然而,在以健康及卫生为目的,或以调节健康为目的长期摄取时,可以在所述范围以下的量,而且在安全性方面,没有任何问题,因此,有效成分是可以使用所述范围以上的量。
所述食品的种类并无特别的限制。可以添加所述物质的食品为例,有肉类、火腿、面包、巧克力、糖类、点心类、饼干类、比萨饼、方便面、其它面类、口香糖类、包含冰淇淋的乳制品、各种汤、饮料、茶、酒精饮料及复合维生素等,并且包含通常意义上的所有健康食品。
本发明的健康饮料组合物像通常的饮料一样可以包含各种香味剂或天然碳水化合物等作为附加成分。所述天然碳水化合物可以使用如葡萄糖、果糖的单糖,如麦芽糖、蔗糖的双糖,以及如糊精、环糊精的天然甜味剂,或如糖精、阿斯巴甜的合成甜味剂等。所述天然碳水化合物的比率是通常每100ml本发明的组合物约0.01至10g,优选地是约0.01至0.1g。
所述以外,本发明的组合物可以包含各种营养剂、维生素、电解质、香味剂、着色剂、果胶酸及其盐、褐藻酸及其盐、有机酸、保护性胶体增稠剂、pH调节剂、稳定剂、防腐剂、甘油、酒精、在碳酸饮料中使用的碳酸化剂等。除此之外,本发明的组合物可以包含天然果汁、用于制造果汁饮料及蔬菜饮料的果肉。这种成分可以单独或组合而使用。这种添加剂的比率虽然不很重要,但本发明的组合物是通常每100重量份在0.01至0.1重量份的范围内选择。
另外,本发明提供,含有由slit1、slit2、slit3、robo1、robo2及vilse所组成的群中选择的蛋白质以预测骨折及骨质疏松症发生的标记组合物。
在本发明的实施例中,以可能有骨质疏松症的患者为对象,关于骨质疏松症发病率和slit3蛋白质的相关关系,经分析从患者的血液中获得的slit3蛋白质的蛋白质组,发现骨质疏松症高发病率组的血中slit3蛋白质浓度的表达特异性减少,因此可以作为诊断或治疗骨折及骨质疏松症的蛋白质标记,选择了slit3。
本发明的slit3蛋白质是与骨质疏松症的发病率成负相关关系,可以有效利用为预测骨折及骨质疏松症发生危险的生物标记。
在本发明中使用的术语‘蛋白质组(proteome)’是指从基因组可以制造的所有蛋白质,这是在一个细胞或组织根据特异性生理状态或病理状态,其蛋白质组的现象不断地变化的动态概念。蛋白质组学(proteomics)概括地指研究这种蛋白质组的方法和技术,通过集中研究基因的表达、翻译后变形(post-translational modification)、与其他蛋白质的结合,将细胞内的变形过程及网络形成与疾病的进展过程联系,在整体上理解蛋白质特性的研究领域。如此,蛋白质组代表一个细胞或组织上的生理状态或病理状态,是最适合于寻找能直接用于诊断疾病的诊断标记的方法。而且,像骨折及骨质疏松症的情形,确认到特定基因的表达干涉疾病的进展并促进骨折及骨质疏松症时,则确认并鉴定其蛋白质的存在,可以作为进行诊断或治疗骨折及骨质疏松症的目的蛋白质。虽然利用基因组学(Genomics)的优越灵敏度和简便扩增基因的优点,也进行不少的诊断及治疗剂的开发,但在DNA或mRNA阶段的变化实际上可能不直接联系到细胞内活性蛋白质变化的观点上,具有理论上的问题,再进一步,实际没有基因物质的体液为例,蛋白质组学是唯一的研究方法。目前,以侵入性接近(non-invasive approach)方式用于诊断的有血浆、血清、尿、脑脊液、羊水、分泌液等体液,且许多研究人员为了发掘作为诊断标记的疾病特异性蛋白质,采用蛋白质组学方法。
在本说明书中,“预测骨折及骨质疏松症发生的标记”是指作为基准将骨质疏松症高发病率组和低发病率组的血液样本区分的蛋白质物质。在本发明中,这蛋白质标记在骨质疏松症高发病率组和低发病率组的特征是其存在量相对地多或少。
因为本发明是在骨质疏松症高发病率组和低发病率组的血液中分析蛋白质组,这样确认的蛋白质标记可以说是对骨质疏松症的发病具有特异性,因此,可以有效用于诊断骨折及骨质疏松症。更进一步,鉴于这样确认的蛋白质标记在骨质疏松症高发病率组的变化现象明显,此蛋白质标记的生理学功能可能对骨质疏松症的发病有直接的关联,因此,该蛋白质标记可有效地用于研究骨质疏松症发病机制或开发骨质疏松症治疗的目的蛋白质。
另外,本发明提供,含有对由slit1、slit2、slit3、robo1、robo2及vilse所组成的群中选择的蛋白质或其免疫原性片段特异性结合的抗体以预测骨折及骨质疏松症发生的试剂盒。
在本发明中,抗体是指指向抗原性部位的特异性蛋白质分子。便于本发明的目的,抗体是指对标记蛋白质特异性结合的抗体,并包含多克隆抗体、单克隆抗体及重组抗体全部。
如上所述,查明了预测骨折及骨质疏松症发生危险的标记蛋白质,因此利用本技术领域的知晓技术,可以容易制造利用该标记蛋白质的抗体生成。

另外,本发明提供骨折及骨质疏松症治疗剂的筛选方法,其包含:(a)从slit1、slit2、slit3、robo1、robo2及vilse所组成的群中选择基因导入细胞培养的步骤;(b)将被检物质与所述(a)步骤的细胞接触的步骤;以及(c)在所述(b)步骤的细胞将slit1、slit2、slit3、robo1、robo2或vilse蛋白质的表达水平与没有处理被检物质的对照组比较的步骤。
另外,本发明提供,以序列号17的氨基酸序列表示的slit3蛋白质的LRR2区域重组肽及包含其的预防或治疗骨折及骨质疏松症的药物组合物。
所述slit3蛋白质的LRR2区域是在slit蛋白质中的四个leucine-rich repeat(LRR)区域中的一个,其中第二区域LRR2与受体结合。本发明人制造作为slit3的LRR2区域的小小重组肽后,利用其确然对破骨细胞分化的效果,发现所述重组肽明显地抑制破骨细胞的分化(图13)。因此,所述重组肽可有效用于骨质疏松症的治疗。

关于所述SNP的NCBI refSNP ID表示所述SNP的序列及其位置。凡是本技术领域的技术人员利用所述号码可以容易地确认SNP的位置及序列。在分析中使用的reference序列是Genome Reference Consortium在2009年2月收集公开的序列,可以在NCBI assemblydatabase的GRCh37(hg19)(http://genome.ucsc.edu/cgi-bin/hgGateway)确认详细的内容。对NCBI登记的SNP的refSNP ID相应的具体序列根据继续研究基因的结果可能略有变更,理所当然,本技术领域的技术人员应该理解这种变更序列也属于本发明的范围。

根据下述的实施例可以确认,以骨密度(BMD)在平均以上的组和严重低BMD的组为对象,对在所述基因存在的基因多态性进行研究的结果,所述基因的SNP位置的对立基因与严重低BMD状态的危险性有显着相关(参照表6)。具体地说,在表示低BMD危险状态的SNP位置,表示的基因型以refSNP ID:rs7655084为例,是GT,GG基因型;以refSNP ID:rs1549909为例,基因型是TT基因型;以refSNP ID:rs10036727为例,基因型是TT基因型;以refSNPID:rs3821735为例,基因型是CC,CT基因型;以refSNP ID:rs78817248为例,基因型是CG,GG基因型;以refSNP ID:rs12418548为例,基因型是GG,GA基因型。
因此,本发明的所述SNP根据严重低的骨密度可有效使用于预测骨折及骨质疏松症发生。
在本发明中,包含所述SNP的多核苷酸或其互补多核苷酸可能是由10个以上连续的碱基构成的DNA片段,DNA片段的大小不是整个基因的full length,而是包含一个SNP部位碱基的任意大小片段也无碍,但优选地,其特征是,10至数百个碱基,更优选地,100-500个碱基适合。100-500个碱基时,可以利用为用于探测所述SNP的探针或引物,而且更多的碱基时,可以利用在PCR-RFLP等。
另外,本发明提供,含有所述SNP的多核苷酸,或含有其互补多核苷酸的预测骨折及骨质疏松症发生的标记组合物,或预测用试剂盒。
在本发明中,包含所述SNP的试剂盒可以通过本技术领域知晓的SNP试剂盒制造方法来体现。例如,所述试剂盒以微阵列体现时,将包含SNP的多核苷酸或其互补多核苷酸固定在基板上,通过核酸杂交及检测杂交结果,可以容易检测SNP。
通过本技术领域知晓的方法,将包含所述SNP的多核苷酸或其互补多核苷酸可容易固定在基板上。另外,在微阵列上的核酸杂交及杂交结果的检测方法也可以以本技术领域知晓的方法执行。例如,以产生核酸样本可检测信号的标记物质(例如,荧光物质)标记后,在微阵列上杂交,并检测从所述标记物质产生的信号,由此可以检测杂交结果。
以下,通过实施例对本发明进行更详细的说明。这些实施例只是本发明的例示而已,理所当然,本技术领域中具有通常知识的技术人员应该理解本发明的范围并不受限于这些实施例。
[实施例]
材料和试剂
NFATc1的抗体是从Santa Cruz Biotechnology(Santa Cruz,CA,USA)购买。slit3的抗体及重组slit1/2/3是分别从Abcam(Cambridge,MA,USA)及R&D System Inc.(Minneapolis,MN,USA)购买。Robo1和Robo2fc嵌合体是从R&D system Inc.购买。
实施方法1.细胞培养,器官培养及条件培养基的收集
原代小鼠骨髓细胞(BMCs)是将5-6周龄C57BL/6小鼠的大腿骨(femur)和胫骨(tibia)冲洗(flushing)获得后,以5%CO2加湿的条件下,在包含10%胎牛血清(FBS;Gibco,Grand Island,NY,USA),100U/mL盘尼西林,及100μg/mL链霉素的最小必需培养基(α-MEM;Wel Gene,Daegu,Korea)以37℃培养。培养24小时后,收集非贴壁细胞(nonadherent cells)在48孔板以1.0×105细胞/孔密度培养。3天以上以30ng/mL M-CSF(R&D System Inc.)培养的BMCs作为骨髓巨噬细胞(bone marrow macrophages,BMMs)使用。在这步骤的细胞当作破骨细胞前体(osteoclast precursors)。BMCs是利用每隔2-3天交换的培养基,以30ng/mL M-CSF及50ng/mL可溶性(soluble)RANKL(R&D System Inc.)培养,由此诱导分化成破骨细胞。在其它方法,小鼠(murine)巨噬细胞Raw264.7细胞(ATCC,Manassas,VA,USA)也是用相同容量的RANKL,制造了破骨细胞-类似细胞。
为了进行利用人破骨细胞的免疫共沉淀(co-immunoprecipitation)实验,从一般健康的志愿者获得了外周血。血液是以相同容量的α-MEM稀释后,利用LymphoprepTM(Axis-Shield,Oslo,Norway)密度梯度离心原理分离了外周血单核细胞(peripheral bloodmononuclear cells,PBMCs)。用autoMacs buffer 悬浮所述细胞,再利用CD14-阳性单核细胞分离器(autoMacs magnetic cell separator;Miltenyi Biotech,Auburn,CA,USA)筛选。在有M-CSF(25ng/mL)的状态下,在6孔板培养PBMCs(3.0-4.0×106/well)3天后,以M-CSF(25ng/mL)及RANKL(30ng/ml)再培养。细胞完全分化成破骨细胞需要7-9天。
以新生C57BL/6小鼠来源颅骨(calvaria)的顺序胶原酶消化(sequentialcollagenase digestion)分离原代小鼠破骨细胞,并保持在含10%FBS的α-MEM中。将小鼠MC3T3-E1前成骨细胞株(preosteoblast cell line,ATCC)以5%CO2加湿的条件下在含有10%FBS,100U/mL盘尼西林,及100μg/mL链霉素的α-MEM以37℃培养。所述培养基是每周交换2次。生长到80%汇合,利用trypsin-EDTA(Gibco)继代培养了所述细胞。为了进行利用人成骨细胞的免疫共沉淀实验,从肋骨(ribs)分离了原代人骨髓间充质干细胞(bone marrowstromal cells,BMSCs)。肋骨是从没有代谢性骨病的患者在开胸手术(thoracotomy)时被遗弃。将所述肋骨在无菌状态下切除,并清理软组织后以纵向(longitudinally)打开。将露出的骨髓以无血清α-MEM经几次的洗涤冲洗掉,在1,400rpm离心分离10分钟。将细胞团块在培养基再悬浮,用LymphoprepTM(Axis-Shield,Oslo,Norway)获得人BMSC分划。将所述细胞以3×107/75-cm2细胞密度接种在75-cm2塑料培养烧瓶,并在含有10%FBS,100U/mL盘尼西林,及100μg/mL链霉素的α-MEM培养。培养基是2周后开始每周交换2次。所述细胞生长到80-90%汇合时,用0.01%胰蛋白酶及0.05%EDTA继代培养。在实验使用了第2次继代培养细胞。
进行了6周龄C57BL/6小鼠来源大腿骨的器官培养。将所述骨髓强有力地冲洗,细胞用H2O振荡培养24小时再去除。然后,将所述骨用α-MEM仔细洗涤,与BMMs共培养。
在破骨细胞生成(osteoclastogenesis)或骨吸收(bone resorption)期间,在指定的日子花24小时收集了CM。将获得的CM用0.45-μm膜滤器过滤,在-70℃保管直到使用。
实验方法2.在CM中破骨细胞形成及吸收活性的测定
在破骨细胞生成的不同阶段收集的CM中,破骨细胞形成的程度是利用0.2U/L低检测界限值的市售ELISA试剂盒(Immunodiagnostic Systems,Boldon,UK)根据制造商的说明书测定TRAP-5b(tartrate-resistant acid phosphatase-5b)进行了评价。在CM中的吸收活性是利用2.0ng/mL低检测界限值的ELISA kit(RatLaps;Immunodiagnostic Systems)测定CTX(C-telopeptide)进行了评价。
实验方法3.趋化性及伤口愈合分析
趋化性(chemotaxis)分析是利用具有8-μm微孔聚碳酸酯膜的Transwell在Boydenchamber system中进行(Costar,Corning,NY,USA)。细胞是以含0.1%FBS的α-MEM每100μL具有1.0×105细胞的密度在上室接种6小时后,在之后的24小时期间在下室以CM或slit3进行处理。在上部膜上的细胞用棉签擦拭全部去除。将侵入在下部膜的细胞固定,用苏木精染色。最后,利用计算机视频成像系统(Olympus,Tokyo,Japan)计数所述细胞。
为了进行伤口愈合分析,层面细胞用塑料针造成伤口。在含0.1%FBS的α-MEM产生细胞迁移(cell migration)后,标记伤口并在O小时和24小时的时刻利用倒立显微镜进行测定。定量是利用Quantity One(BioRad,Hercules,CA,USA)测定各伤口愈合(woundclosure)区域而完成。数据是以伤口愈合的百分比来表示。
实验方法4.细胞生存力及分化的测定
细胞生存力是根据制造商的说明书利用细胞计数试剂盒-8(CCK-8;Dojindo,Kumamoto,Japan)测定的。简单地说,将10μL的WST-8dye(2-[2-methoxy-4-nitrophenyl]-3-[4-nitrophenyl]-5-[2,4-disulfophenyl]-2H-tetr azolium,monosodium salt)添加在96孔板的各孔中反应2小时后,在450nm利用酶标仪(SPECTRAmax 340PC;MolecularDevices,Palo Alto,CA,USA)以650nm的参考波长测定了吸光度。
细胞分化是利用Brd-U(5-bromo-2’-deoxyuridine)测定的。成骨细胞种在96孔板反应24小时后,在α-MEM继代培养了24小时。接着,将所述细胞用Brd-U反应24小时后,用BrdU标记及检测试剂盒(Roche,Mannheim,Germany)测定了细胞分化。
实验方法5.蛋白质组学实验计划
准备样本
Raw 264.7细胞是在含10%FBS的α-MEM的每100mm盘以4.0×105细胞的密度接种后,将所述细胞在有可溶性50ng/mL RANKL或没有的状态下处理一天分化成破骨细胞。第二天,将所述细胞以无血清及没有酚红的培养基洗涤3次后,将培养基替换成有RANKL或没有的6mL无血清及无酚红的α-MEM,培养一天。CM是以0.45μm膜滤器过滤后在-70℃保管。将在CM的蛋白质冻干沉淀。
通过反相C18HPLC及胰蛋白酶消化的分划
蛋白质的复合体是利用由C18-HPLC柱(214TP5125,2.1×150mm;Vydac Grace,Hesperia,CA,USA),自动取样器,及UV检测器(215nm wavelength;Peptide LibrarySupport Facility,Pohang,Korea)构成的毛细管HPLC系统,在0.3mL/min的流速经120分钟利用0-60%梯度的ACN(acetonitrile)及0.1%TFA(trifluoroacetic acid)分离成96个分划。
为了进行胰蛋白酶消化(trypsin digestion),在50mM ABC(ammoniumbicarbonate)溶解各蛋白质分划后,添加10mM DTT(dithiothreitol)还原样本,为了半胱胺酸烷基化添加了100mM碘乙酰胺(iodoacetamide)。最后,添加500ng胰蛋白酶在37℃反应了6个小时。
LC-MS/MS分析
将各个消化的样本利用具有奈米流HPLC系统的LTQ(linear-trap quadrupole)质谱仪来分析(Thermo Fisher Scientific,San Jose,CA,USA)。肽是利用安装有C18(3μm)的反相分析柱(10cm×75μm i.d.)来分析。梯度是在250nL/minute的流速以5%CAN开始5分钟,之后60分钟以40%CAN,最后10分钟增加到80%CAN。洗脱剂(eluent)是以1.8kV的电喷射电压利用奈米粒子源注入LTQ质谱仪。所述分析法是由400-1500m/z范围的全质量分析(MS)扫描构成,对5个最强烈粒子的数据依赖质量分析(MS2)是在全MS扫描上进行了分析。
MASCOT数据库检索
在TQ质谱仪获得的数据是利用国际蛋白质指数(IPI)小鼠FASTA数据库(version3.54)及MASCOT检索引擎进行了分析。在固定的变形上,检索时考虑了根据胰蛋白酶消化的不完全切断、对蛋氨酸氧化的可变变形,及半胱氨酸的尿基甲基化(carbamidomethylation)。本发明人以信任范围95%机率评价了各个离子(P<0.05)。
功能注释及分泌性蛋白质筛选
为了功能注释(functional annotation),鉴定的蛋白质是利用基于网页的程序(http://david.abcc.ncifcrf.gov)DAVID(atabase for Annotation,Visualization,andIntegrated Discovery)数据库根据其生物过程及分子功能分类。另外,鉴定的蛋白质是利用HMM评分(SignalP 3.0hidden Markov matrix scoring)评价以便识别其分泌特性。
实验方法6.蛋白质印迹法
细胞裂解液是以裂解液(20mM Tris[pH 7.5],150mM NaCl,1mM EDTA,1%TritonX-100,2.5mM sodium pyrophosphate,1mM-glycerophosphate,1mM Na3VO4,1mM NaF,及aprotease-inhibitor mixture)在4℃制造20分钟,所述裂解液的蛋白质浓度是利用BCA蛋白质分析试剂盒(Pierce Chemical Co.,Rockford,IL,USA)测定。包含10-20μg蛋白质的样本以10%凝胶SDS-PAGE分离后,转移到硝酸纤维膜(Amersham Biosciences,Buckinghamshire,UK)上。用TBST(500mM Tris-HCL[pH 7.4],1.5M NaCl,0.1%Tween-20)中的5%脱脂乳在室温阻挡1小时后,将所述膜用第1抗体反应一夜,然后用第2抗体反应。免疫反应蛋白质是利用增强化学发光试剂盒(PerkinElmer,Waltham,MA,USA)检测。
实验方法7.免疫共沉淀法
GFP-tagged Robo1及Myc-tagged Vilse的人cDNA是从Origene(Rockville,MD,USA)公司购买。将Robo1-GFP及Vilse-Myc的cDNA用lipofectamine2000(Gibco,GrandIsland,NY,USA)在人BMSC或人PBMC转染6个小时后,用Slit3处理了细胞。所述细胞是以包含蛋白酶抑制剂混合物(protease inhibitor cocktail;Sigma-Aldrich,St.Louis,MO,USA)及磷酸酶抑制剂(phosphatase inhibitors;1mM Na3VO4,1mM NaF)的TNE缓冲液(25mMTris-HCl,pH 7.4,150mM NaCl,1%Triton X-100,1mM EDTA)进行了溶解。裂解液是在4℃用GFP抗体(Anaspec,Fremont,CA,USA)及蛋白质-G-琼脂糖珠(Amersham Biosciences,Buckinghamshire,UK)免疫沉淀了18小时。免疫沉淀物及细胞裂解液是以抗-GFP或抗-Myc抗体(Novus biological,Littleton,Co,USA)提供给免疫印迹分析。
实验方法8.RT-PCR及定量实时PCR
总RNA是根据制造商的说明书用TRIzol试剂(Invitrogen,Rockville,MD,USA)分离,cDNA是利用Superscript III First-Strand Synthesis System(Invitrogen)从1μg的总RNA合成。所有PCR扩增是利用Biometra thermocycler(GmbH,Goettingen,Germany)而执行。各目的基因的mRNA表达水平利用Quantity One program标准化为管家基因GAPDH(glyceraldehydes-3-phosphate dehydrogenase)。特异性引物对如下:

实验方法9.siRNA的转染
将关于NFATc1(Mm_NFATc1_6;Qiagen),Robo1(MSS208673;Invitrogen),Robo2(MSS241005;Invitrogen)的siRNA及nonsense siRNA(Stealth RNAiTM siRNA negativecontrol;Invitrogen)根据制造商的说明书以lipofectamine试剂(Invitrogen)进行了转染。简单地说,在含10%FBS的α-MEM培养细胞后,添加OPTI-MEM(Invitrogen)中的siRNA-试剂混合物,然后再培养细胞6个小时。将所述培养基替换成新鲜的完全α-MEM后,再培养细胞2天。
实验方法10.板状伪足(lamellipodia)的确认
将MC3T3-E1细胞在24孔板接种24小时,然后在有或没有slit3的状态下饥饿了24小时。固定所述细胞用PBS(phosphate-buffered serum)洗涤2次。将所述细胞以100ng/mL鬼笔环肽(phalloidin;Molecular Probes,Leiden,Netherlands)在37℃培养30分钟。免疫荧光图像是利用荧光显微镜(Olympus)拍摄的。
实验方法11.钙化结节的形成
将原代小鼠BMCs在12孔板以6×106细胞/孔的密度接种,并在补充10%FBS(v/v),100U/mL盘尼西林及100mg/L链霉素的α-MEM在37℃以5%CO2及95%空气加湿的孵化器培养7天。7天后去除非贴壁细胞,然后将表现BMSCs(bone marrow stromal cells)的贴壁细胞在用于诱导BMSCs成骨细胞分化的分化培养基(α-MEM containing 10%FBS[v/v]supplemented with 8mMβ-glycerophosphate and 50μg/mL ascorbic acid)再生长14天。所述培养基每隔2天或3天交换一次。在第14天,将培养物用70%乙醇固定1小时后,以40mMAlizarin red S(Sigma-Aldrich,St.Louis,MO,USA)进行了染色。
实验方法12.TRAP染色及吸收分析
将原代小鼠BMCs以30ng/mL M-CSF及50ng/mL可溶性RANKL培养4天。固定贴壁细胞,根据制造商的说明利用leukocyte acid phosphatase kit(Sigma-Aldrich)对破骨细胞的酶标记以TRAP进行了染色。将包含3个核(nuclei)以上的TRAP-阳性多核细胞当中破骨细胞,在光学显微镜(Olympus)下进行了计数。
为了测定吸收区域,将BMCs(5×104cells/well)与30ng/mL M-CSF及50ng/mL可溶性RANKL一起置放在牙本质盘(dentine discs)上。10天后,将所述载片用5%次氯酸钠水溶液洗涤去除细胞后,用苏木精对吸收坑(pits)进行了染色。为了测定每BMCs的数目吸收的区域,在牙本质盘上在苏木精的染色之前进行了TRAP染色。吸收坑的区域是利用QuantityOne Software(VersaDoc Model 3000Imaging system,Bio-Rad,Berkeley,CA,USA)进行了分析。
实验方法13.生物体内颅骨骨形成模型
对4周龄C57BL/6小鼠在3周期间每周5日,一天一次,将PBS或300μg/kg的slit3皮下注射在颅骨(calvaria)。注射实施在位于人字缝(lambdoidal suture)及冠状缝(coronal suture)之间的颅骨失状(calvarial sagittal)的左侧,右侧是当作对照组使用。牺牲所述动物后在4%PFA(paraformaldehyde)固定了颅骨。将各颅骨用14%EDTA去除钙质后,包埋在冻结切片(frozen section)用OCT(optical cutting temperature)化合物。将样本切割成7μm切片后,用苏木精及伊红染色。用光学显微镜测定了颅骨的宽度。
实验方法14.生物体内颅骨骨损失模型
将6周龄C57BL/6小鼠分成4组:对照组(0.1%BSA+PBS);slit3-处理组(0.1%BSA+300μg/kg slit3);IL-1-诱导颅骨骨损失组(2μg IL-1+PBS);及IL-1骨损失及slit3-处理组(2μg IL-1+300μg/kg slit3)。将经所述溶液处理的胶原蛋白海绵(Cellmatrix Type I-A;Nitta Gelatin Inc.,Osaka,Japan)移植在位于失状缝(sagittal suture)中间的颅骨上。移植7天后牺牲所述小鼠,将颅骨固定在4%PFA上。将各颅骨用14%EDTA去除钙质,包埋在冻结切片用OCT化合物。将样本切割成7μm切片后,用苏木精及伊红染色。用光学显微镜测定了颅骨的宽度。
实验方法15.骨髓腔(bone marrow cavity)移植
本发明人使用了11周龄C57BL/6小鼠。将胫骨(tibias)用23-guage针小心地打了个孔。将用PBS(left tibia)或300μg/kg的slit3(right tibia)GFP-标记的MC3T3-E1细胞用31-guage针注入在骨髓腔。经过2天或3天后牺牲所述小鼠,胫骨固定在4%PFA上。将各胫骨用14%EDTA去除钙质,包埋在冻结切片用OCT化合物。为例计数GFP阳性细胞,在位于荧光显微镜(Olympus)下的载片上观察了样本。
实验方法16.在slit3基因敲除小鼠胚胎上的Von Kossa染色及VEGF免疫组织化学
染色
slit3突变胚胎是交配从Mutant Mouse Regional Resource Centers(stocknumber 030759-MU;Columbia,MO,USA)购买的雌雄slit3+/-C57BL/6J小鼠而制造。胚胎组织在PBS收集,在室温固定在10%福尔马林一夜。为了组织化学分析,野生型及slit3突变的17.5天(E17.5)胚胎以4%PFA固定,并包埋在OCT化合物。将7μm切片用Von Kossa反应及核固红(nuclear fast red)染色。For IHC staining,peroxidase chromogens(Invitrogen)were used.为了免疫组织化学(IHC)染色,使用了过氧化物酶色原(peroxidasechromogens,Invitrogen)。所述前片在溶解3%H2O2的甲醇中淬灭(quench)10分钟后,在PBS洗涤,与以PBS稀释成1:200或1:500的抗-VEGF抗体(Abcam)在室温反应1小时。利用DAB(diaminobenzidine)基质使关于抗体结合的信号视觉化。对所有样本用光学显微镜进行了评价。
实验方法17.骨密度测定
全身骨密度(BMD)是利用密度测定仪(Lunar PIXImus densitometer softwareversion 1.4;GE-Lunar Co.,Madison,WI,USA)以双能X线吸收法(dual-energy X-rayabsorptiometry)进行了测定。以变动系数(coefficient of variation,CV)表示的设备精密度是1.27%。将所述小鼠用40mg/kg Zoletil50(Virbac,France)及5.6mg/kg Rompun(Bayer Korea,Seoul,Korea)的混合物肌肉注射麻醉,以伸直四肢的俯卧位(proneposition)状态置放在扫描器上。
实验方法18.测序
本发明人对染色体4上的slit2基因(NM_004787)、染色体5上的slit3基因(NM_003062)、染色体3上的robo1基因(NM_001145845)、染色体3上的robo2基因(NM_NM_002942)、及染色体11上的robo4基因(NM_019055)进行了调差。关于这些基因的目的区域是整个编码外显子、外显子-内含子边界及调控区域。设计诱饵库(bait libraries)利用Agilent eArray website functionality(https://earray.chem.agilent.com/earray/)对这些目的基因组区域的覆面进行了分析。利用BLAST(~12mismatches across thebait)以90%以上一个以上的位置映射到诱饵时,从设计中排除。最终,6,507bp目的区域以Agilent SureSelect Sequence Enrichment Kit被确定。接着,用IlluminaHiSeq2000analyzer(Illumina,San Diego,CA)对982绝经后的女性(super-normal BMDgroup=501,and severe-low BMD group=481)进行了基因测序。利用Covaris System随机使用3的基因组DNA制造了150bp的插入片段(inserts)。将这片段DNA利用T4DNA聚合酶及Klenow聚合酶进行末端修复(end-repair),将修复的末端受体寡核苷酸(Illuminapaired-end adaptor oligonucleotides)贴在粘性末端(sticky ends)上。用琼脂糖凝胶电泳分析连接混合物纯化成200-250bp大小的片段。将纯化的DNA库以SureSelectTarget Enrichment probes set(Agilent,Santa Clara,CA)杂化根据制造商的说明书在标记区域捕捉。利用被捕捉的库,根据制造商的方案,制造了HiSeq2000paired-endflowcell。PCR菌落的集群放在HiSeq2000platform进行了基因测序。
实验结果1.在初期分化阶段破骨细胞分泌诱引成骨细胞前体的潜在性偶联因子。
为了建立用于调差所述偶联现象的试管内系统,准备了关于破骨细胞生成及骨吸收的可变培养条件。本发明人将BMMs在有30ng/mL M-CSF的状态下以100ng/mL RANKL处理2天和6天,分别诱导了初期及后期破骨细胞的分化。为了诱导吸收阶段,将BMMs在有30ng/mLM-CSF的状态下与100ng/mL RANKL及小鼠大腿骨(femur)一起培养了10天。破骨细胞分化及骨吸收是在各培养基分别测定TRAP-5b活性及CTX浓度进行了评价。
如图1a所示,从RANKL-处理的细胞获得的CM与每阶段的非处理组比较,TRAP-5b活性明显增加。而且,如图1b所示,CTX浓度只是在大腿骨和RANKL都有的状态下增加。
在各阶段,将从破骨细胞培养水收集的CM在原代小鼠颅骨的成骨细胞及MC3T3-E1细胞进行了处理。RANKL-处理的BMMs的任意培养基与非处理组的上层液比较对原代成骨细胞的生存能力或分化(分别是图2a和2c),或是对MC3T3-E1细胞的生存能力或分化(分别是图2b和2d)没有显着的影响。但是伤口-愈合分析显示从RANKL-处理的BMMs获得的CM与非处理组比较明显增加了成骨细胞(图2e)及MC3T3-E1细胞(图2f)的运动性(motility)。从RANKL-处理的Raw 264.7细胞获得的CM也是增加了MC3T3-E1细胞的运动性(图2g)。尤其是,在后期分化阶段,从RANKL-处理的细胞收集的CM以更小的程度刺激前成骨细胞的运动性,但初期分化阶段从RANKL-处理的细胞获得的CM最显着地增加了前成骨细胞的运动性。与此相反,从以RANKL培养10天的破骨细胞获得的CM不管有或没有小鼠大腿骨,不刺激运动性。
这种结果是指可偶联因子(possible coupling factors)不是从骨吸收过程分泌,主要是从初期分化的破骨细胞分泌。
对于成骨细胞系的方向性迁移(directional migration),为了评价破骨细胞CM的效果,本发明人使用了Boyden chamber assay。从RANKL-处理的BMM及Raw 264.7细胞两者收集的CM也是与非处理对照组比较,明显地刺激了MC3T3-E1细胞的方向性动原(mobilization)(分别是图2h和2i)。这种结果说明由从初期破骨细胞生成诱导的因子补充前成骨细胞(preosteoblastic recruitment)的刺激可能是在骨再形成部位的偶联现象的主要机制。
实验结果2.推定为趋化性因子的鉴定。
为了鉴定对前成骨细胞作用为趋化因子的特异蛋白质,本发明人分划了LC-MS/MS。简单地说,将从RANKL-处理及-非处理的Raw 264.7细胞获得的CM的96个匹配分划在MC3T3-E1细胞处理,对于这些细胞可动性的效果则利用Boyden chamber assay做了比较。本发明人筛选对于MC3T3-E1细胞的迁移表现最显着效果的匹配分划。将所述实验方法中记载的蛋白质组学分析,对筛选的分划及相邻RANKL-处理的分划实施。与相邻分划及RANKL-非处理对照组比较,本发明人在RANKL-处理的分划鉴定了明显表达的45个肽。其中,9个肽具有分泌特性(表1)。最后,选择了slit3蛋白质,之后进行了实验。

实验结果3.Slit3是从分化的破骨细胞分泌。
为了确认从分化的破骨细胞是否分泌slit3,进行了如下的实验。将BMMs(bonemarrow macrophages)在含有30ng/mL M-CSF(macrophage colony-stimulating factor)的培养基下,加以区分100ng/mL RANKL的处理与否后培养2天,在所述细胞进行了蛋白质印迹法。而且,利用知晓为参与破骨细胞形成过程(osteoclastogenesis)的主基因NFATc1的siRNA,在BMMs切断破骨细胞形成过程,以相同的方法进行了蛋白质印迹法。其结果分别显示在图3a及图3b。
如图3a所示,确认到slit3的表达是在分化的破骨细胞的细胞裂解液(lysate)及CM(conditioned media)都显着地增加。
另外,如图3b所示,确认到由RANKL刺激的slit3的表达被NFATc1-siRNA完全切断。
通过所述结果,确认到slit3是从分化的破骨细胞分泌,不是从破骨细胞的前体分泌。
实验结果4.Slit3刺激成骨细胞的迁移、生存能力、增殖、分化及OPG生成。
为了确认在成骨细胞的slit3功能,将MC3T3-E1成骨细胞用重组slit3处理后,确认了对成骨细胞的迁移(migration)、生存能力(viability)、增殖(proliferation)、分化(differentiation)及OPG(osteoprotegerin)生成的影响。其结果分别表示在图4a至4g。
如图4a所示,将MC3T3-E1成骨细胞用slit3(0,0.1,0.5及1.0μg/mL)处理24小时的结果,确认了随着slit3的浓度,前成骨细胞(preosteoblast)的迁移增加。
另外,如图4b及图4c所示,将MC3T3-E1成骨细胞用slit3(0,0.1,0.5及1.0μg/mL)处理48小时,进行CCK-8测定法及Brd-U测定法的结果,确认由slit3的处理增加了成骨细胞的生存能力及增殖。在CCK-8测定法IGF-1是利用为阳性对照组。
另外,如图4d所示,将原代(primary)小鼠BMSCs(bone marrow stromal cells)以1.0μg/mL slit3处理,诱导成骨细胞分化的结果,确认由slit3的处理显着地增加成骨细胞分化标记ALP(alkaline phosphatase)及OCN(osteocalcin)的mRNA表达。
另外,如图4e所示,将原代小鼠颅骨成骨细胞以1.0μg/mL slit3处理48小时,进行QRT-PCR的结果,确认slit3促进OPG的生成,但对RANKL的表达没有影响。
另外,如图4f所示,在细胞迁移过程中,向细胞方向移动,且实际参与运动的板状伪足(lamellipodia)形成受刺激时,则这意味着有趋化性(chemotactic action)。
通过所述结果,确认了slit3在成骨细胞具有增加迁移、生存能力、增殖、分化及OPG生成的功能,由此对骨形成过程有贡献。
实验结果5.Slit3增加颅骨骨形成并防止IL-1诱导的颅骨骨损失。
为了验证关于骨细胞的slit3的试管内结果,进行了利用动物模型的实验。将slit3直接注入颅骨的结果,与非处理对照组比较骨的宽度增加到15%(图5)。而且,在动物模型的颅骨骨损失,处理IL-1诱导骨损失时,证明了slit3抑制由IL-1诱导的骨损失(图6)。这种结果意味着slit3可以用为增加骨形成,同时减小骨吸收的治疗剂。
实验结果6.Slit3刺激MC3T3-E1细胞向骨形成表面移动。
将GFP-标记的MC3T3-E1细胞注入骨髓腔时,slit3处理与PBS处理组比较,向骨形成表面明显地增加细胞的移动(图7)。
实验结果7.slit3在基因敲除小鼠胚胎明显地弱化骨形成及血管新生。
为了评价17.5天的slit3小鼠胚胎大腿骨中积累的钙,进行了Von Kossa染色,为了评价血管的形成,进行了VEGF IHC染色。slit3-基因敲除胚胎与野生型胚胎比较,由VonKossa(图8a)及VEGF IHC(图8b)两个方法的染色几乎没有显示。这种结果是指slit3持续地贡献于骨形成,并在增加对血管新生必需因子VEGF的表达扮演重要的角色。
实验结果8.对骨细胞的slit3作用是通过Robo1及Robo2受体传递。
因为slit配体及Robo受体被公认为在各种类型细胞中的信号传递路径(Dickinson RE,et al.,Reproduction 2010;139:697-704),所以本发明人执行RT-PCR后在骨细胞调查了Robo受体的表达。
其结果,确认了MC3T3-E1细胞表达Robo1及Robo2受体(图9a)。原代破骨细胞主要表达Robo1受体,反而,Robo3及Robo4受体的表达水平增加得很弱(图9b)。在MC3T3-E1细胞,由siRNA转染及fc嵌合体切断Robo1及Robo2的表达时,slit3-刺激的细胞的迁移及增殖被抑制(分别为图9c及9d)。关于原代破骨细胞,对于Robo1受体siRNA转染使由slit3处理引起的破骨细胞形成的减少完全恢复(图9e).
实验结果9.Vilse是在成骨细胞和破骨细胞通过Robo1受体传递slit3作用的主要
信号分子。
从人BMSC和人PBMC生成的,GFP-Robo1是成骨细胞(图10a)和破骨细胞(图10b),与Myc-vils一起分别转染时,slit3处理在免疫共沉淀法明显地增加Robo1-Vilse的相互作用。用siRNA抑制vilse时,MC3T3-E1细胞的slit3-刺激的生存能力被抑制(图10c)。这种结果意味着通过Robo1受体的slit作用在骨细胞可能由vilse传递。
实验结果10.全身BMC在Robo1-基因敲除小鼠明显地减少。
在8周龄雄性Robo1野生型、杂合子性(heterozygote)及基因敲除小鼠比较全身BMC值得结果,Robo1-基因敲除小鼠显示明显低的BMD值(图11)。
实验结果11.slit1及slit2弱化破骨细胞的分化。
进行TRAP染色,由此实验了有关破骨细胞slit家族的其他蛋白质,slit1及slit2的效果(图12)。slit1及slit2是如由slit3所观察的一样,破骨细胞的分化随容量减少。
实验结果12.重组slit3的LRR2肽弱化破骨细胞的分化。
据报slit蛋白质的LRR2区域是与其的受体Robo结合的区域。因此,本发明人以LRR2区域的肽制造了slit3的小重组肽。确认到所述重组肽明显地抑制破骨细胞的分化(图13)。这种结果是指重组肽可有效利用在骨质疏松症的治疗。
实施例13.临床实验
13-1.实验对象的选择
实验对象是在首尔峨山病院(Seoul,Korea)门诊的绝经后健康女性作为对象。所述女性都是担心骨质疏松症的可能性而门诊的情形或在健康检查时检测到骨质疏松症的情形。此时绝经是持续1年以上无月经为例经测定血清内FSH(follicle stimulatinghormone)的浓度而验证。在实验对象中,40岁以前绝经的实验对象,投入对骨代谢可能有影响的药剂6个月以上或在12个月以内的实验对象及曾经患过对骨代谢可能有影响疾病的实验对象均将排除。最后选了346位女性。
13-2.骨密度(Bone mineral density,BMD)测定
对所述13-1选出的实验对象测定了年龄、体重、身高、BMI、行为因素(吸烟、饮酒、运动等)、骨密度等。
骨密度(BMD,g/cm2)是利用DXA(Lunar;Prodigy,Madison,WI,USA)测定了腰椎(lumbar spine,L1-L4)、股骨近端(proximal femur)、股骨颈(femur neck)、大腿骨(totalfemur)、转子(trochanter)、骨干(shaft)、三角(ward)。
其结果显示在下表2。

13-3.血中slit3浓度测定
将在所述13-1选择的实验对象的空腹血液样本离心分离后,分离出上清液。血中slit3浓度是利用slit3竞争性ELISA试剂盒(Echelon Biosciences Inc,Salt Lake,UT,USA)测定,反复了2次。利用所述结果值,将各骨位置的BMD或T-score作为因变量(dependent variable),slit3浓度作为自变量(independent variable),进行了多次线性回归分析(Multiple linear regression analysis)。其结果显示在表2。协变量(Covariates)包含年龄、体重、身高、吸烟、饮酒及定期运动。

如表3所示,以可能性的混杂因素(confounding factor)补正后,确认到血中slit3浓度与腰椎、股骨颈、大腿骨、大腿三角、大腿骨干的BMD值及T-score有相关性。
另外,根据slit3的浓度以5分位分组后,观察了骨质疏松症发病率。根据WHO时,腰椎、股骨颈、大腿部中任一处的T-score≤-2.5SD时,定义为骨质疏松症。其结果显示在下表4。

如表4所示,骨质疏松症odds是以协变量补正后,确认到相比于显示血中slit3浓度最低的组(Q1),在显示最高的组(Q5)显示约低58%。
通过所述结果,确认血中slit3浓度是与骨质疏松症的发病率成负相关关系,由此可以利用为预测骨折及骨质疏松症发生危险的标记。
实验结果14.在slit2,slit3,robo1,robo2及robo4的7个SNP与低骨密度危险有关
联。
根据BMD状态的基因学研究对象的基准特性如下表5。因为两组的年龄相同,在年龄方面没有什么特别差异。与这些组的定义相同的是,BMI在严重低BMD组明显地高,反而在腰骨(lumbar spine)和股骨颈(femur neck)的BMD值和T-score在平均以上的BMD组明显地高。
处于严重低BMD状态(severe-low BMD status)危险的各种多态性(polymorphism)基因效果是通过序列分析(targeted deep sequencing analysis)及logistic回归分析(logistic regression analysis)得到了分析(表6)。在slit2有1个SNP(rs7655084),在slit3有2个SNP(rs1549909及rs10036727),在robo1有1个下游SNP,在robo2有2个SNP(rs3821735及rs78817248),以及在robo4有1个SNP(rs12418548)与严重低BMD状态的危险有明显的关联。

以上,对本发明以较佳实施例为中心进行了说明。应该理解凡是本发明的技术领域中具有通常知识的技术人员在不脱离本发明基本特性的范围下可以以变形的形态实施。因此,揭示的实施例不是以限定的观点,而是以说明的观点考量才行。本发明的范围不是在上述的说明,而是在权利要求书表示,并且与其同等范围内的所有差异点应包含于本发明。
商业上利用可能性
本发明的slit或robo蛋白质是在细胞及动物模型中增加骨形成,减小骨吸收,并与骨质疏松症的发病率成负相关关系,由此可有效利用为预防或治疗骨折及骨质疏松症的组合物及用于预测骨折及骨质疏松症发生危险的生物标记。
Claims (3)
1.一种药物组合物在制备预防或治疗骨折及骨质疏松症的药物中的用途,所述药物组合物作为有效成分含有slit3蛋白质的LRR2区域。
2.一种由slit3蛋白质的LRR2区域作为有效成分含有的食品组合物在制备预防或改善骨折及骨质疏松症的食品中的用途。
3.一种包含用于检测slit3的LRR2基因的mRNA的RT-PCR用引物的试剂盒在制备预测骨折及骨质疏松症发生的药物中的用途。
