本发明涉及中药提取纯化技术领域,具体是一种通过中药萃取脂溶性成分获得的蜡油混的物,通过如下方法制备,蜂蜡加热融化后,加入到醇提药液中,所述醇提药液温度65~75℃、相对密度1.08~1.20,充分搅拌混匀,冷却,静置12~24小时,取上部药液,过滤分离,得到蜡油混合物。同时,克服了醇提药液在后续浓缩、干燥、粉碎中,对易挥发性成分、脂溶性成分造成一定损失,脂肪油等造成不易干燥、不易粉碎等技术难点。

技术领域
本发明涉及中药提取纯化技术领域,具体是一种从中药醇提液中萃取分离挥发性成分、脂溶性成分及脂肪油的制备方法。
背景技术
中药提取纯化工艺是指根据临床用药和制剂要求,用适宜溶剂和方法从净药材(饮片)中富集有效物质,除去杂质的过程。常用溶剂有水或水与乙醇配制不同浓度的乙醇。前者称水提药液,后者称醇提药液。在提取、纯化及其后续的制剂过程中,浓缩、干燥、粉碎是必要的工艺环节。目前,醇提药液在后续浓缩、干燥、粉碎中,对易挥发性成分、脂溶性成分造成一定损失,脂肪油等造成不易干燥、不易粉碎等技术难点。
发明内容
本发明的目的是在中药醇提液中加入蜂蜡萃取分离挥发性成分、脂溶性成分及脂肪油,得到蜡油混合物,进而解决了目前醇提药液在后续浓缩、干燥、粉碎中存在的问题。
本发明是采用如下技术方案实现的:
一种通过中药萃取脂溶性成分获得的蜡油混的物,通过如下方法制备,蜂蜡加热融化后,加入到醇提药液中,所述醇提药液温度65~75℃、相对密度1.08~1.20,充分搅拌混匀,冷却,静置12~24小时,取上部药液,过滤分离,得到蜡油混合物,主要用于中药软胶囊药液配制的助悬剂。
其中,醇提药液的获得方法如下,在中药配方中加50%~95%的乙醇,回流提取2~3次得醇提液,过滤,回收乙醇,减压浓缩至相对密度1.08~1.10、温度为65~75℃的清膏。
蜂蜡系蜜蜂科昆虫中华蜜蜂Apis cerana Fabricius或意大利蜂Apis melliferaLinnaeus分泌的蜡精制而成。蜂蜡溶于氯仿、醚、非挥发油、挥发油等,几乎不溶于水,熔点为62~67℃。蜂蜡中70%~75%的成分是具有偶数碳链,长度为24~36个碳的单羟基直链一元醇与直链酸酯化所形成的各种酯的混合物。直链酸所含的偶数碳原子最多可达36个,其中一些为C18位的羟基酸。所形成的酯主要是棕榈酸蜂花酯。同时存在的还有游离酸(约14%)和烃(约12%),以及约1%的游离蜡醇和脂肪酸的硬脂醇酯。蜂蜡常用于中药软胶囊剂药液配制的助悬剂。
上述蜡油混合物的优点如下:
1、分离得到的蜡油混合物作为中药软胶囊药液配制的助悬剂,减少制剂中辅料用量。
2、利用蜂蜡提取保留醇提药液中的挥发性成分、脂溶性成分及脂肪油。
3、有利于醇提药液的后续干燥、粉碎工艺。
本发明设计合理,在中药的醇提液中萃取分离挥发性成分、脂溶性成分及脂肪油,获得蜡油混合物。同时,克服了醇提药液在后续浓缩、干燥、粉碎中,对易挥发性成分、脂溶性成分造成一定损失,脂肪油等造成不易干燥、不易粉碎等技术难点。
具体实施方式
实施例1
山金连软胶囊加蜡收脂工艺中蜡油混合物的制备方法。
山银花等八味加65%的乙醇,回流提取3次,提取时间本领域技术人员根据实际情况确定,过滤,回收乙醇,浓缩至相对密度1.08~1.10、温度65℃的清膏,加入蜂蜡,加热融化,充分搅拌,冷却至0~5℃,静置12h,过滤,分离得到蜡油混合物。滤液进行后续的浓缩、干燥、粉碎等工艺,避免了其中易挥发性成分、脂溶性成分的损失,而且脂肪油被提取后,更容易干燥和粉碎。
加蜡收脂工艺参数如下:
蜂蜡量:12g(以处方量计);
醇提液:醇提液量:1800ml(以处方量计),
药液温度:65℃以上,
相对密度:1.08~1.10(65℃);
冷却温度:0~5℃;
静置时间:12小时;
上部药液取量:180ml(以处方量计);
过滤、分离收集蜡油混合物量60g。
实施例2
正气消暑软胶囊加蜡收脂工艺中蜡油混合物的制备方法。
姜厚朴等五味加入65%乙醇,回流提取2次,提取时间本领域技术人员根据实际情况确定,过滤,回收乙醇,浓缩至相对密度1.08~1.10、温度67℃的清膏,加入适量蜂蜡,加热融化,充分搅拌,冷却至0~5℃,静置24h,过滤,分离得到蜡油混合物。滤液进行后续的浓缩、干燥、粉碎等工艺,避免了其中易挥发性成分、脂溶性成分的损失,而且脂肪油被提取后,更容易干燥和粉碎。
加蜡收脂工艺参数如下:
蜂蜡量:12g(以处方量计);
醇提液:醇提液量:1500ml(以处方量计),
药液温度:67℃,
相对密度:1.08~1.10(65℃);
冷却温度:0~5℃;
静置时间:24小时;
上部药液取量:150ml(以处方量计);
过滤、分离收集蜡油混合物量48g。

Claims (3)

1.一种通过中药萃取脂溶性成分获得的蜡油混合物,其特征在于:通过如下方法制备,蜂蜡加热融化后,加入到醇提药液中,所述醇提药液温度65~75℃、相对密度1.08~1.20,充分搅拌混匀,冷却,静置12~24小时,取上部药液,过滤分离,得到蜡油混合物。
2.根据权利要求1所述的通过中药萃取脂溶性成分获得的蜡油混合物,其特征在于:所述醇提药液的获得方法如下,在中药配方中加50%~95%的乙醇,回流提取2~3次得醇提液,过滤,回收乙醇,减压浓缩至相对密度1.08~1.10、温度为65~75℃的清膏。
3.根据权利要求1或2所述的通过中药萃取脂溶性成分获得的蜡油混合物,其特征在于:蜂蜡和醇提药液混匀后的冷却温度为0~5℃。
