Abstract

本发明提供冬虫夏草的无性阶段中华被毛孢的工厂化培养发酵,发酵产物虫草菌丝体具有与野生冬虫夏草相同的药理作用,并且人工虫草菌体的必需氨基酸含量成倍于天然冬虫夏草。

Description

本发明是冬虫夏草的无性阶段一中华被毛孢(1987年11月3日保藏于中国典型培养物保藏中心,保藏号为CCTCCNO:M87101)的深层发酵工艺技术,工厂化生产冬虫夏草菌体。
冬虫夏草是只产干中国西南西北高原的保健滋补珍品。野生的冬虫夏草,产区局限,产量稀少,资源缺乏,价格昂贵。中华被毛孢及其完成有性阶段冬虫夏草的技术,将中华被毛孢在特定条件下,侵染人工饲养的寄生—斜脉蝠蛾幻虫,完成有性阶段,周期长,要求条件严格,成本较高。本发明的优点是:深层发酵工艺条件容易控制、周期较短,成本较低,发酵所得的冬虫夏草菌体成分与天然冬虫夏草成分类同,并可诱导产生人类需要的代谢产物,可以人为地控制菌丝体产量,以此为原料生产用于保健、延缓衰老的滋补品,用于治疗性的药物,可以最大限度的满足人们日益增加的对保健、防病、治病的需要。
以下详细叙述本发明的主要内容:
1.斜面培养:斜面培养基:麸皮30克,蛋白胨10克,奶粉10克,蚕蛹粉3克,维生素10毫克,MgSO4·7H2O0.5克,磷酸二氢钾1克,琼脂20克,水1000毫升,将中华被毛孢(Hirsutella sinensis yu,Liu,Guo etFang,sp,nov.)(CCTCC No.M87101)菌株接种于斜面培养基上,在15-18℃的条件下培养30天;
2.液体培养:液体培养基:麸皮30克,奶粉10克,蚕蛹粉汁3克,牛肉汁10克,维生素10毫克,MgSO4·7H2O0.5克,磷酸二氢钾1克,水1000毫升,pH6.5,分装三角瓶,体积为三角瓶容量的1/5,灭菌后置摇床,在18±2℃的温度下,90次/分的往复式摇床上振荡培养8天后,菌丝体即进入生长旺盛期,可以进行再扩大培养。
3.发酵生产
(1)种子罐培养
培养基:麸皮30千克奶粉10千克蚕蛹粉汁3千克酵母10千克维生素10克水1000升pH6.5,以5%接种量接入液体培养菌丝,在温度18±2℃,通气量1∶1(V/V/V)的条件下,连续发酵168小时。此时菌丝体生长旺盛,经镜检无细菌污染,可转入发酵罐生产。
(2)发酵罐生产
培养基:麸皮30千克奶粉10千克蚕蛹粉汁3千克酵母粉10千克维生素10克MgSO4·7H2O0.5千克磷酸二氢钾1千克水1000千克。发酵条件:温度18±2℃,通气量1∶1(V/V/V),pH6.5,连续发酵7天,在发酵开始的第24小时后,每隔16小时取发酵液样,测定pH值、还愿糖及氨态氮,当pH下降至5.5-5.0,还原糖1%以下,氨态氮0.1%以下,镜检菌丝体已产生大量分生孢子梗及分生孢子时,就可出罐。

浙江农业大学用日立835—50氨基酸分析仪对天然冬虫夏草和虫草菌体进行对比分析结果如下:(表见文后)
*色氨酸在水解过程中全部破坏,无法测出。
从上述结果看,必需氨基酸均成倍于天然冬虫夏草。

Claims (3)

1.中华被毛孢的深层发酵工艺,其特征在于取新鲜冬虫夏草虫体(菌核(Hirsutella sinensisyu,Liu,Guo et Fang,sp,nov.)CCTCC No.M87101部分,去表皮,切成薄片,置下述培养基上培养,或挑取子囊孢子培养:
斜面培养基:麸皮30克,蛋白胨10克,奶粉10克,蚕蛹粉3克,维生素10克,硫酸镁0.5克,磷酸二氢钾1克,琼脂20克,水1000毫升,pH6.5,在15-18℃的温度下培养30天;
液体培养基:麸皮30克,奶粉10克,蚕蛹粉汁3克,牛肉汁10克,维生素10毫克,硫酸镁0.5克,磷酸二氢钾1克,水1000毫升,pH6.5,在18±22的温度下,90次/分的往复式摇床上振荡培养8天后扩大培养。
2.根据权利要求2所说的发酵工艺,其特征在于种子罐发酵培养基:麸皮30克,奶粉10克,蚕蛹粉汁3克,酵母粉10克,维生素10毫克,水1000毫升,pH6.5,通气量1∶1(V/V),在18±2℃温度的条件下发酵7天后转罐,接种量5%。
3.根据权利要求1或2所说的发酵工艺,其特征在于发酵罐培养基:麸皮30千克,奶粉10千克,蚕蛹粉汁3千克,酵母粉10千克,维生素10克,硫酸镁0.5千克,磷酸二氢钾1千克,水1000千克,温度18±2℃,通气量1∶1(V/V),连续发酵168小时出罐。
