具体实施方式
本发明提供了一种AAV定点整合构建表达FVIII的人293细胞株及其构建方法,以及利用该人细胞株表达有活性的重组FVIII的方法。
进一步的讲,本发明是利用rAAV整合质粒与表达整合必需的AAVRep蛋白的质粒共转染HEK293细胞,经过筛选获得稳定表达FVIII的293细胞株,一方面避免了重组病毒构建及纯化过程,更简单易于操作;另一方面无重组病毒基因组的分子量限制,具有更广泛的应用性。
以下结合一较佳实施例及相应附图对本发明的技术方案作进一步的说明。
本实施例可包括如下步骤:
一、扩增BDD基因,构建rAAV整合质粒pTRP5BDDGFP。此质粒是在张春构建的rAAV质粒pTRP5GDNFGFP基础上改造而成,将pTRP5GDNFGFP质粒中GDNF基因替换为BDD基因。
其中,所述BDD基因系B区缺失的FVIII基因(BDD-FVIII),其全长4316bp,编码1438个氨基酸,其相应序列请参见SEQIDNO:1和SEQIDNO:2,而其结构请参考图4。
一般来说,成熟FVIII蛋白包含3个区,其结构为A1-A2-B-A3-C1-C2。虽然B区的功能尚未确定,但很多研究表明B区缺失对FVIII(BDD-FVIII)的促凝血和辅因子活性没有影响;另外,研究表明,B区缺失能够将mRNA水平提高17倍之多,也因此伴有翻译产物的增加。在本发明中,通过选择BDD-FVIII,可以获得高表达量的FVIII蛋白。
又,前述pTRP5BDDGFP的构建过程包括:
首先,以BamHI和PstI从pTRUF11(请参阅图1b以及CharacterizationofaBipartiterAAVVectorforSiteSpecificIntegration,HumGeneTher.,2007,18:787-797.)中切出GFP序列,以及,用BamHI和PstI将neoR从pTRP5GDNF(参阅图1c)中切除;
而后,将GFP和pTRP5GDNF连接得到pTRP5GDNFGFP(参阅图1d);
最后,用NheI和NsiI将GDNF序列从pTRP5GDNFGFP中切除,回收骨架部分与BDD基因连接,得到pTRP5BDDGFP,其中含有GFP作为筛选标记。
显然,本领域人员能够依据本领域之常识而在前述构建过程中选择采用合适的反应条件及产物纯化、收集等操作程序,继而获得最终产物。
二、将整合质粒pTRP5BDDGFP与表达AAVRep蛋白的pSVAV2质粒(张春博士构建,请参阅CharacterizationofaBipartiterAAVVectorforSiteSpecificIntegration,HumGeneTher.,2007,18:787-797.)共转染HEK293细胞(购自中国科学院典型培养物保藏委员会细胞库),连续传代3次;将细胞克隆至96孔板,共克隆3次,筛选出稳定表达BDD-FVIII的293细胞株。利用流式细胞仪检测荧光细胞的比率。
当然,本领域人员依据本发明的内容,亦可很容易想到采用其它具有AAVRep蛋白表达功能的质粒替代前述pSVAV2质粒。
三、对所得细胞株分别进行PCR、RT-PCR和Westernblot鉴定,以确定基因整合及BDD蛋白的表达。
四、检测阳性细胞培养上清中的BDD蛋白活性。
本发明利用rAAV整合质粒与表达Rep蛋白的质粒共转染HEK293细胞,经过三轮筛选获得稳定表达FVIII的293细胞株。此方法避免了重组病毒构建及纯化过程,更简单易于操作,而且整合率高。以人293细胞作为宿主,表达的FVIII蛋白的结构与功能更接近于天然FVIII蛋白。
更具体的讲,本实施例中具体可以通过如下步骤实现:
一、利用primerpremier5.0软件设计引物,上游引物:5’-CATCGCTAGC GCCACCATGCAAATAGAGCTC,含NheI酶切位点和kozak序列;下游引物:5’-GCAGAACCAATGCATTCAGTAGAGGTCCTG,含NsiI酶切位点。PCR反应体系为50μL,包含50ng模板质粒pZp9FVIII△BS(戚正武院士惠赠,参阅TheGeneExpressionofCoagulationFactorVIIIinMammalianCellLines.ThrombosisResearch.,1999,95:105–115)、上、下游引物各0.3μM、dNTP0.2mM、MgSO41.5mM、1×PCR缓冲液和1UKOD-Plus-Neo(东洋纺生物科技有限公司,上海)。PCR扩增过程采用两步法:94°C预变性2min,98°C变性10sec,68°C延伸3min,共35个循环。用NheI和NsiI双酶切质粒rAAV质粒pTRP5GDNFGFP(该质粒可参照前述构建过程及图1b-图1d构建),将GDNF基因酶切下来(37°C,2h),回收骨架部分;将BDD基因PCR产物也用NheI和NsiI双酶切回收,连接至rAAV质粒骨架(T4DNA连接酶22°C,30min),使其位于AAV两个ITR之间,并有GFP作为筛选标记。
前述的各种酶均可由市场获得,例如,可购自NEB公司。
该步骤一最终所获rAAV重组质粒pTRP5BDDGFP的图谱和鉴定结果请参阅图1a和图2。
二、将HEK293细胞接种于24孔板中(相应条件可以为:购自hyclone公司的10%胎牛血清DMEM培养基,37°C,5%CO2,培养12h左右),待细胞长到60%左右,利用PEI将整合质粒pTRP5BDDGFP与表达AAVRep蛋白的pSVAV2质粒(张春博士构建,请参阅CharacterizationofaBipartiterAAVVectorforSiteSpecificIntegration,HumGeneTher.,2007,18:787-797.)以50:1(1μg:20ng)共转染HEK293细胞,继续培养,长满后将细胞传代,连续传3代后仍然可观察到荧光细胞;细胞计数,以每孔10个细胞接种于96孔板中,培养7天左右,挑出有荧光的细胞继续培养于24孔板中;待细胞长满后,单克隆至96板中,如此连续克隆3次,筛选出稳定表达荧光的293细胞株。利用流式细胞仪测得荧光细胞的比率约为98%,具体请参阅图3a-图3b。
三、按常规操作对所得细胞株分别进行PCR、RT-PCR和Westernblot鉴定。Rep蛋白介导的AAV定点整合位于人19号染色体的AAVS1区,用AAVD-序列上游引物(5’-AGGAACCCCTAGTGATGGAG)和AAVS1下游引物(5’-TCAGAGGACATCACGTGGTG)扩增,以确定基因的整合。具体方法,提取整合细胞的总DNA为模板200ng/50μL,上、下游引物各0.3μM、dNTP0.2mM、MgSO41.5mM、1×PCR缓冲液和1UKOD-Plus-Neo。采用降落PCR法扩增:94°C预变性2min;98°C变性10sec,74°C延伸1min,5个循环;98°C变性10sec,72°C延伸1min,5个循环;98°C变性10sec,70°C延伸1min,5个循环;98°C变性10sec,68°C延伸1min,30个循环。
Trizol法提取细胞的总RNA,用FVIII基因下游引物逆转录成cDNA,然后进行PCR扩增,上游引物:5-CCAGGGTGCCCGTCAGAAGT,下游引物:5-GCAGAACCAATGCATTCAGTAGAGGTCCTG,方法同上。
重组细胞株293-BDD培养于25cm2培养瓶中,待细胞长至80~90%,改用无血清培养48h,收集培养上清,用超滤离心法浓缩,进行SDS-PAGE电泳;Bioworld公司FactorVIII(S2194)pAb作为一抗(1:800倍稀释);ABGENTHRPGoatanti-rabbitIgGantibody作为二抗(1:8000倍稀释)。
鉴定结果表明,BDD基因已被整合至293细胞染色体中,并能在细胞中稳定表达,所获得的细胞株命名为293-BDD。
四、重组细胞株293-BDD培养于25cm2培养瓶中,待细胞长至80~90%,改用无血清培养24h,收集培养上清,用COAMATICFactorVIII试剂盒检测阳性细胞表达的BDD蛋白活性,结果表明,其凝血活性为14mU/105cellsmL-124hrs-1。
需要指出的是,以上所述仅为本发明较佳的具体实施方式,但本发明的保护范围并不局限于此,任何熟悉该技术的人在本发明所揭露的技术范围内,可轻易想到的变化或替换,都应涵盖在本发明的保护范围之内。因此,本发明的保护范围应该以权利要求的保护范围为准。
