IL-12含有的p35和p40亚基经优化后,在CHO细胞中可获得稳定高效的表达,3号优化基因序列(含天然信号肽)经表达后,在CHO-DG44细胞内,表达量为天然基因的2倍;9号优化基因序列(含鼠源信号肽)经表达后,表达量为天然基因的3倍。在CHO-3E7细胞中,3号优化基因序列表达量为天然基因的4倍;9号优化基因序列(含鼠源信号肽)经表达后,表达量为天然基因的10.6倍。这些数据证明,人IL-12的p35和p40亚基的基因密码经优化后均比天然的人IL-12的基因表达量高。利用优化的3#序列,我们已经获得高表达的稳定细胞株,在无血清的培养基内,可以达到200mg/L远远高于已有的报道。
