Abstract

Description

用于对食物、饮料、动物饲料、化妆品或药物组合物进行富集、强化、或着色的含有脂溶性活性成分(例如β-胡萝卜素)的组合物是本领域已知的。β-胡萝卜素是一种优选的着色剂化合物,这归因于其浓烈的和对上述应用而言非常令人愉悦的橙色。因为最终的组合物通常是水性组合物如饮料,所以不得不向组合物中添加额外的用于富集、强化和/或着色的化合物,以避免脂肪(油)相从产物中分离,所述分离会导致相应的产品不可接受。

茄替胶(Gum ghatti),也已知为印度胶(Indian gum)或阔叶榆绿木(Axlewood)作为来自Anogessias latifolia的渗出物获得,所述Anogessiaslatifolia是属于使君子科(Combretaceae)的树。茄替胶是与L-阿拉伯糖、D-半乳糖、D-甘露糖、D-木糖和D-葡糖酸(D-glucorinic acid)的复合多糖。茄替胶可含有多达5%或甚至更多的蛋白质。其被用于包封香料或油、用作纤维来源、成膜剂、粘附剂等等。

根据本发明的组合物与US2005/0287221A1中公开的组合物存在显著差异。
在着色制品如饮料中,也通常期望保持饮料的光学澄清度。用于补充的脂溶性着色剂如类胡萝卜素(例如β-胡萝卜素)能够以许多形式获得,但是被添加至饮料时会趋向于提高可视的混浊度。成环现象(Ringing,即液体顶部形成分离的脂溶性β-胡萝卜素层)也是许多已知的β-胡萝卜素配制物的问题。向饮料中添加脂溶性物质而不提高可视混浊度或者成环的一种手段是将所述物质包封在脂质体中。然而,这是昂贵的工艺,并且脂质体中物质的浓度往往较低。

因此,仍然存在对不显示上述问题,即不显示分离现象的脂溶性活性成分组合物的需要,所述组合物用于食物、饮料、动物饲料、化妆品或药物组合物的富集、强化和/或着色。在脂溶性活性成分是着色剂的情况下,组合物应当还为得到的产物提供提高的颜色强度和颜色稳定性。
因此,提供具有如上所述期望特性(例如关于光学透明度和乳液稳定性的非常良好的特性和/或改进的颜色强度和颜色稳定性)的脂溶性活性成分的组合物是本发明的一个目的(无论可应用在何处)。例如通过使用不同的乳化技术来改进制备脂溶性活性成分组合物的方法也是本发明的一个目的。

本发明涉及包含茄替胶和一种或多种脂溶性活性成分的组合物,其中所述组合物包含以总组合物干物质为基础少于40重量%的油,进一步优选少于35重量%的油,进一步优选少于30重量%的油。最优选根据本发明的组合物包含从0重量%到10重量%的油。
在本发明的一个优选的实施方案中,组合物包含以总组合物干物质为基础20重量%和85重量%之间,进一步优选30重量%和65重量%之间的茄替胶。
本文使用的术语“脂溶性活性成分”是指:选自由维生素A、D、E、K及其衍生物组成的组的维生素;多不饱和脂肪酸;亲脂性健康成分;类胡萝卜素;调味剂或芳香物质;以及它们的混合物。
根据本发明适合的多不饱和脂肪酸(PUFA)是单或多不饱和羧酸,其优选地具有16到24个碳原子,尤其是1到6个双键,优选地具有4或5或6个双键。
不饱和脂肪酸可属于n-6系列和n-3系列二者。n-3多不饱合酸的优选的例子是二十碳-5,8,11,14,17-五烯酸和二十二碳-4,7,10,13,16,19-六烯酸;优选的n-6多不饱合酸的例子是花生四烯酸和γ-亚麻酸。
多不饱合脂肪酸的优选的衍生物是其酯,例如甘油酯,尤其是甘油三酯;尤其优选的是乙酯。n-3和n-6多不饱和脂肪酸的甘油三酯是特别优选的。
甘油三酯可含有3个一致的不饱和脂肪酸,或2或3个不同的不饱和脂肪酸。它们也可部分含有饱和的脂肪酸。
当衍生物是甘油三酯时,通常用甘油酯化三个不同的n-3多不饱和脂肪酸。在本发明的一个优选的实施方案中使用甘油三酯,其中30%的脂肪酸部分是n-3脂肪酸,且其中25%是长链不饱和脂肪酸。在又一个优选的实施方案中,使用可商业获得的‘30’n-3Food Oil(DSMNutritional Products Ltd,Kaiseraugst,瑞士)。
在本发明的另一个优选的实施方案中,PUFA酯是’75’n-3EE。ROPUFA’75’n-3EE是乙酯形式的精制鱼油(marine oil),其具有最小72%n-3脂肪酸乙酯的含量。其用混合的生育酚、抗坏血酸棕榈酸盐、柠檬酸稳定,并含有迷迭香提取物。
根据本发明,使用含有多不饱和脂肪酸的甘油三酯的天然存在的油(一种或多种组份)可以是有利的,例如鲸鱼油(鱼油)和/或植物油,使用从经发酵的生物质或经遗传修饰的植物中提取的油也是有利的。
包含多不饱和脂肪酸甘油三酯的优选的油为橄榄油、葵花子油、月见草籽油、琉璃苣油(borage oil)、葡萄籽油、大豆油、花生油、小麦胚芽油(wheat germ oil)、南瓜子油、胡桃油、芝麻籽油、菜籽油(芸苔)、黑加仑籽油(blackcurrant seed oil)、猕猴桃籽油、来自特定真菌的油和鱼油。
多不饱和脂肪酸的优选的例子为例如亚油酸、亚麻酸、花生四烯酸、二十二碳六烯酸、二十碳五烯酸等等。
根据本发明优选的健康成分是:白藜芦醇;藁本(ligusticum);选自辅酶Q10(也称作“Q10”)、辅酶Q9和/或其还原形式(相应的泛醇)的泛醌和/或泛醇(一种或多种组分);染料木黄酮和/或α-硫辛酸。
本发明的特别优选的脂溶性活性成分为类胡萝卜素,特别是β-胡萝卜素、番茄红素、叶黄素、胭脂树橙(bixin)、虾青素、阿朴胡萝卜素醛、β-阿朴-8′-胡萝卜素醛、β-阿朴-12′-胡萝卜素醛、角黄素、隐黄素、柠檬黄质和玉米黄素。最优选的是β-胡萝卜素。
在本发明的一个优选的实施方案中,以总组合物干物质为基础,组合物包含0.1和70重量%之间、进一步优选0.1和30重量%之间、进一步优选0.5和20重量%之间、最优选0.5和15重量%之间的一种或多种脂溶性活性成分。
在本文上下文中使用的表述“油(oil)”包括适用于所述组合物的预期用途的任何甘油三酯或任何其它油(例如萜烯)。油优选地为植物油或脂肪,优选地为玉米油、向日葵油、大豆油、红花油、油菜籽油、花生油、棕榈油、棕榈仁油、棉籽油、橙油、柠檬烯(limonene)、橄榄油或椰子油。
在本发明的又一个优选的实施方案中,组合物的特征在于其还包含至少一种额外的植物胶质;特别优选所述又一种植物胶质是金合欢胶。根据本发明的组合物中使用的金合欢胶是Acacia senegal或Acacia seyal,优选地是Acacia Seyal。
如果根据本发明的组合物还包含至少一种额外的植物胶质,则优选所述组合物还包含至少一种改性淀粉。
术语“改性的食物淀粉”是指由通过已知的化学方法用疏水片段取代的淀粉制成的改性淀粉。例如,可用环状二羧酸酐如琥珀酸酐和/或戊二酸酐处理淀粉,所述酸酐用烷基或烯基烃基取代。
本发明的一种尤其优选的改性淀粉具有下式(I):
其中St是淀粉,R是亚烷基,R′是疏水基团。优选地R为低级亚烷基,例如二亚甲基或三亚甲基。R′可以是烷基或链烯基,优选地具有5到18个碳原子。式(I)的一种优选的改性淀粉为辛烯基琥珀酸淀粉钠(StarchSodium Octenyl Succinate,也称“OSA-淀粉”)。本文使用术语“OSA-淀粉”表示用辛烯基琥珀酸酐(OSA)处理过的任何淀粉(来自任何天然来源例如玉米、小麦、木薯、马铃薯或是合成的)。取代程度,即相对于羟基总数的酯化羟基数量,通常在从0.1%到10%的范围内,优选地在从0.5%到5%的范围内,更优选地在从2%到4%的范围内变化。
OSA-淀粉可含有其他水胶体,如淀粉、麦芽糖糊精、碳水化合物、胶质(gum)、玉米糖浆等,并可选地含有任何典型的乳化剂(作为共乳化剂),如脂肪酸单甘油酯和二甘油酯、脂肪酸的多甘油酯、卵磷脂、脱水山梨醇单硬脂酸酯、植物纤维和/或糖。
OSA-淀粉可以商品名HiCap 100、Capsul、Capsul HS、Purity Gum2000、UNI-PURE、HYLON VII等从例如National Starch商业获得;或从Roquette Frères商业获得;或以商品名C*EmCap从CereStar商业获得;或从Tate&Lyle商业获得。HiCap 100是特别优选的。
如果根据本发明的组合物还包含至少一种额外的植物胶质,则进一步更优选的是根据本发明的组合物额外地包含一种或多种寡糖,如糊精和麦芽糖糊精,特别是具有5到65葡糖当量(DE)的那些,和葡萄糖浆,特别是具有20到95DE的那些。术语“葡糖当量”(DE)表示水解程度,是以干重为基础以D-葡萄糖计的还原糖的量;该标度以DE接近0的天然淀粉和DE为100的葡萄糖为基础。优选地,在根据本发明的组合物中使用麦芽糖糊精。
本发明还涉及用于制造上文所述组合物的方法,所述方法包括以下步骤(所述方法可以以本文所述用量使用所述成分来进行):
I)将茄替胶和任选地一种或多种额外的植物胶质溶于水中;
II)任选地向步骤I)的溶液中添加至少一种改性淀粉;
III)向步骤II)的溶液中添加有机相,所述有机相包含一种或多种脂溶性活性成分和少于40重量%的油(以总组合物干物质为基础),并且在所述脂溶性活性成分是类胡萝卜素的情况下还包含有机溶剂;
IV)在30℃和100℃之间、进一步优选45℃和80℃之间、进一步优选在50℃和70℃之间的温度下乳化步骤III)的混合物;
V)在减压下蒸发所述有机溶剂;
VI)通过喷雾干燥、粉末捕获或其它方法干燥所述乳液。
干燥步骤可用本领域技术人员已知的任何常规干燥步骤进行,优选的是喷雾干燥和/或粉末捕获工艺,其中被喷雾的悬浮液微滴在吸附剂(如淀粉或硅酸钙或硅酸或碳酸钙或其混合物)的床上被捕获并随后干燥。
需要时还优选在所述方法的步骤II)中添加寡糖,优选地添加麦芽糖糊精。
合适的有机溶剂是卤代的脂肪族烃,脂肪族酯,脂肪族和环状碳酸酯,脂肪族酯和环状酯(内酯),脂肪族和环状酮,脂肪族醇及其混合物。
卤代脂肪族烃的例子是单卤代或多卤代的线性、支化或环状C1-到C15-烷烃。特别优选的例子是单氯化或多氯化或溴化的线性、支化或环状C1-到C15-烷烃。优选地使用CHCl3。
脂肪族酯和环状酯(内酯)的例子是乙酸乙酯、乙酸异丙酯和乙酸正丁酯;和γ-丁内酯。
脂肪族和环状酮的例子是丙酮、二乙基甲酮和异丁基甲基甲酮;和环戊酮和异佛尔酮。
环状碳酸酯的例子具体是碳酸乙二醇酯和碳酸丙二醇酯及其混合物。
脂肪族醚的例子是二烷基醚,其中所述烷基片段具有1到4个碳原子。一种优选的例子是二甲基醚。
脂肪族醇的例子是乙醇、异丙醇、丙醇和丁醇。
本发明还涉及上述组合物用于食物、饮料、动物饲料、化妆品或药物组合物的富集、强化和/或着色的用途。
本发明还涉及通过将上述组合物与饮料的其它常见成分混合来制造饮料的方法。
本发明还涉及能够通过上述方法获得的饮料。
业已惊讶地发现,本发明的组合物能够与水混合,由此得到的混合物具有高度稳定性。另外,没有出现脂溶性活性成分从得到的混合物分离。如果所述脂溶性活性成分是β-胡萝卜素,则不存在辅助化合物如鱼胶或除β-胡萝卜素之外的着色化合物时也实现有利的颜色。
还出乎意料地发现,使用小量油(例如低至1重量%)时根据本发明的组合物具有改进的特性,例如颜色强度和颜色稳定性。
如果没有相反的定义,则本发明的化合物的用量(重量%)是指所述化合物以总组合物干物质为基础(不含溶剂如有机溶剂和/或水的所有成分的总量)的重量%。根据本发明的组合物优选地是乳液。
固体组合物可额外含有抗结块剂如硅酸或磷酸三钙等等,并且至多10重量%,优选地为0.1重量%到5重量%。
水溶性抗氧化剂可例如是抗坏血酸或其盐,优选地为抗坏血酸钠,水溶性多酚如羟基酪醇和橄榄苦苷,苷元,表没食子儿茶素没食子酸酯(EGCG)或迷迭香或橄榄的提取物。
脂溶性抗氧化剂可例如是生育酚,例如dl-α-生育酚(即合成的生育酚)、d-α-生育酚(即天然生育酚)、β-或γ-生育酚或两种或更多这些生育酚的混合物;基化的羟基甲苯(BHT);丁基化的羟基茴香醚(BHA);乙氧喹,没食子酸丙酯;叔丁基羟基喹啉;或6-乙氧基-1,2-二羟基-2,2,4-三甲基喹啉(EMQ),或脂肪酸的抗坏血酸酯,优选地为棕榈酸抗坏血酸酯或硬脂酸抗坏血酸酯。
另外,所述组合物可包含水。
优选地,本发明的组合物除类胡萝卜素化合物之外不含有其它着色物质。优选地,本发明的组合物不含有鱼明胶。
表1示出了以总组合物干物质为基础组合物优选成分的优选用量(重量%)。表1所示用量可额外地与如上所述的成分优选用量组合。

本发明还涉及上述组合物用于食物、饮料、动物饲料、化妆品或药物组合物的富集、强化和/或着色,优选地用于饮料的富集、强化和/或着色的用途。不存在“成环”,即用含有本发明组合物的饮料装填的瓶表面上不存在不溶部分不想要的分离。
本发明的其它方面是含有上文所述的组合物的食物、饮料、动物饲料、化妆品和药物组合物。
可使用本发明的制品形式作为着色剂或添加剂成分的饮料可以是碳酸饮料,例如调味的塞耳特斯水(flavored seltzer water)、软饮或矿物饮品,以及非碳酸饮料,例如调味的水、果汁、水果宾治(fruit punches)和这些饮料的碳酸形式。它们可以基于天然的水果汁或蔬菜汁或基于人工香料。还包括在内的是酒精饮料和即食饮料粉(instant beverage powder)。另外,还包括含糖饮料、不含热量并含有人工甜味剂的减肥饮料(diet beverage)。
另外,得自天然或合成来源的乳制品也属于食物制品的范围,其中本发明的制品形式可被用作着色剂或添加剂成分。这类制品的典型例子是乳饮品、冰激凌、乳酪、酸乳等等。乳替代制品如豆浆饮品和豆腐制品也包括在该应用范围内。
还包括在内的是作为着色剂或作为添加剂成分含有本发明的制品形式的甜品(sweets),如糖果制品(confectionery product)、糖果、口香糖、甜点,例如冰激凌、果冻(jellies)、布丁、即食布丁粉等等。
还包括在内的是谷物、点心、曲奇、意大利面(pasta)、汤和沙司(sauces)、蛋黄酱、沙拉酱等等,其含有本发明的制品形式作为着色剂或添加剂成分。另外,用于乳和谷物的水果制备物也包括在内。
借由本发明的组合物添加至食物制品中的一种或多种脂溶性活性成分(优选类胡萝卜素,特别是β-胡萝卜素)的终浓度以食物组合物的总重为基础可优选地从0.1到50ppm,尤其是1到30ppm,更优选3到20ppm,例如约6ppm,并取决于有待着色或强化的具体食物制品和着色或强化的预期等级。
优选地通过向食物制品中添加本发明组合物形式的脂溶性活性成分,来获得本发明的食物组合物。为了食物或药物制品的着色或强化,可根据本身已知的方法使用本发明的组合物来应用可分散于水中的固体制品形式。
通常,组合物可作为水性储存溶液添加,或者作为与适用于特定应用的其他食物成分的干粉混合物或预混合物添加。根据最终应用的组成,可例如使用干粉搅拌器、低剪力混合器、高压匀化器或高剪力混合器完成混合。显而易见的是,这类技术是本领域普通技术人员已知的。
其中使用本发明的组合物作为着色剂的药物组合物如片剂或胶囊也在本发明的范围内。可以通过向片剂包衣混合物中单独添加液体或固体着色剂组合物形式的组合物,或者通过向片剂包衣混合物的组分之一中添加所述组合物,来实现片剂的着色。可以通过在胶囊物质的水性溶液中掺入所述组合物,来制备着色的硬壳或软壳胶囊。
其中本发明的组合物作为活性成分使用的药物组合物如片剂(如咀嚼片、泡腾片或包膜片)或胶囊(如硬壳胶囊)也在本发明的范围内。产物形式典型地作为粉末被添加至制片混合物中,或以本身已知的用于生产胶囊的方式被填充进胶囊中。
其中使用该组合物作为色素沉着着色剂(例如用于卵黄、肉鸡(tablepoultry)、小鸡(broilers)或水生动物)或作为活性成分的动物饲料制品也在本发明的范围内,所述动物饲料制品例如营养成分的预混物、复合饲料、乳替代物、液体膳食或饲料制剂。
其中使用本发明的组合物作为着色剂或作为添加剂或作为活性成分的化妆品(cosmetics)、美容用品(toiletries)和皮肤用品(derma product)也在本发明的范围内,所述化妆品、美容用品和皮肤用品即皮肤和头发护理制品例如乳霜、洗剂、沐浴用品、唇膏、洗发水、护发素、喷雾或凝胶。
本发明的饮料和组合物是在下文所述的测试方法中显示优良表现的那些,尤其是显示有利的颜色色度的那些。
本发明的饮料的例子是运动饮料、补充维生素的水和其中需要添加维生素的饮料。还包括用于恢复腹泻导致的电解质损失的饮料。还包括碳酸饮料例如调味的塞耳特斯水(flavored seltzer water)、软饮或矿物饮品,以及非碳酸水果汁和蔬菜汁、宾治(punches)和这些饮料的浓缩形式。
本发明还涉及通过将如上所述的组合物与其他常用成分混合制造饮料的工艺。
另外,本发明涉及可通过如上所述的制造饮料的工艺获得的饮料。
本发明的组合物优选地是添加剂组合物,并优选地以添加剂组合物形式使用。
物理稳定性
如果微滴内部相的分布与时间无关,则乳液被认为是物理稳定的。可能的不稳定是乳状液分层(creaming)、沉降、奥斯特瓦尔成熟(Ostwaldripening)、聚结、相转化和聚集。
沉降和乳状液分层是本领域公知的,并且已使用斯托克斯定律(Stoke’s Law)详细描述于文献中。沉降和乳状液分层的速率受密度、介质粘度、颗粒尺寸的影响。为此进行颗粒尺寸D(3,2)的测量来证明本发明。另外,一些类胡萝卜素配制物的颜色改变是D(3,2)值的函数,例如对β-胡萝卜素而言,微滴最小时颜色最趋向于黄色。
根据本发明制造方法步骤V)的胡萝卜素乳液显示从0.05μm到1μm的尺寸范围(用Mastersizer S进行测量)。
乳液中的絮凝程度也取决于电动力(electrokinetic force)。所有系统的稳定性取决于微滴之间吸引力和排斥力(例如范德华力、静电、立体稳定作用)之间的平衡。根据DLVO模型,电势曲线中正的初级最大值(positive primary maximum)在室温下应当>1.5kT(>37.5mV),所述正的初级最大值起到针对不可逆絮凝的能垒(energy barrier)的作用。这一静电排斥可以通过由电泳迁移率计算的ζ电势来定量。通过使用Smoluchowski等式测量ζ电势,进行了本发明乳液的稳定性平价。在我们的案例中,ζ电势总是高于-38mV(用ZetasizerNano SZ进行测量)。
对工业中颜色的应用而言,不仅测量显著的颜色,而且精确测定颜色差异。CIE L*a*b*(CIELAB)是International Commission on Illumination(Commission Internationale d′Eclairage,因此其缩写为CIE)规定的最全面的颜色空间(color space)。所述颜色空间描述了人眼可见的所有颜色,并且被创建来用作参照的与设备无关的模型。CIELAB的三个坐标表示颜色的亮度(L*,L*=0得到黑色,L*=100表示白色),其在红色/品红色(magenta)和绿色之间的位置(a*,负值表示绿色,正值表示品红色)和其在黄色与蓝色之间的位置(b*,负值表示蓝色,正值表示黄色)。L、a和b之后的星号(*)是全名的一部分,因为它们代表L*、a*和b*,与Hunter的L、a和b区分。在我们的系统内,与窄尺寸分布相关的颗粒尺寸的降低与L*的增加直接相关。另外,颗粒尺寸的降低被转化为基于β-胡萝卜素的产物形式的颜色从浅红色(reddish)到浅黄色(yellowish)迁移。在我们的案例中,L*总是高于70,b*为正。

在烧杯上将茄替胶倒入水中,并在室温下搅拌,直至其完全溶解。同时将含有β-胡萝卜素、玉米油、α-生育酚的有机相溶于溶剂中。然后向水相中添加有机相,并乳化得到的混合物(FLUID 5000RPM)。最后在减压下蒸发溶剂。通过粉末捕获(在淀粉床上)将得到的产物转化成干燥形式。测定了产物的颗粒尺寸、颜色测量和ζ电势。

Claims (13)
1.包含茄替胶和一种或多种脂溶性活性成分的组合物,其中所述组合物包含以总组合物干物质为基础少于40重量%的油。
2.根据权利要求1的组合物,其特征是所述脂溶性活性成分是类胡萝卜素,特别是β-胡萝卜素、番茄红素、叶黄素、胭脂树橙、虾青素、阿朴胡萝卜素醛、β-阿朴-8′-胡萝卜素醛、β-阿朴-12′-胡萝卜素醛、角黄素、隐黄素、柠檬黄质和/或玉米黄素。
3.根据权利要求2的组合物,其特征是所述组合物包含以总组合物干物质为基础0.1重量%和30重量%之间的类胡萝卜素。

II)任选地向步骤I)的溶液中添加至少一种改性淀粉;

11.根据权利要求11的方法,其特征是在步骤II)中还添加一种或多种寡糖。
12.权利要求1到10中任一项所述的组合物用于食物、饮料、动物饲料、化妆品或药物组合物的富集、强化和/或着色的用途。
13.含有权利要求1到10中任一项所述的组合物的饮料。
