Abstract
本发明涉及包含辛烯基琥珀酸酐改性的金合欢胶和类胡萝卜素的组合物。已经发现根据本发明的组合物允许以低浊度生产具有非常高的颜色强度以及颜色稳定性的乳液。这些组合物可被用于食物饮料、动物饲料、化妆品组合物或药物组合物的富集、强化和/或着色。本发明还涉及通过将组合物和饮料成分混合制造饮料的方法。本发明还涉及通过该方法可获得的饮料。
Description
本发明涉及包含辛烯基琥珀酸酐改性的金合欢胶(gum acacia)和类胡萝卜素的组合物。这些组合物可被用于对食物饮料、动物饲料、化妆品组合物或药物组合物进行富集、强化和/或着色。本发明还涉及通过将组合物和饮料成分混合制造饮料的方法。本发明还涉及通过该方法可获得的饮料。
用于对食物、饮料、动物饲料、化妆品组合物或药物组合物进行富集、强化、或着色的含有类胡萝卜素(例如β-胡萝卜素)的组合物是现有技术已知的(例如:WO2008110225)。β-胡萝卜素是一种优选的着色剂化合物,这归因于其浓烈的和对上述应用而言非常令人愉悦的橙色。因为β-胡萝卜素被分类为略溶于水的活性剂,并且其中使用β-胡萝卜素的最终产品通常是水性组合物如饮料,所以不得不添加额外的化合物,以避免含β-胡萝卜素的相从产品中分离,所述分离会导致相应的产品不可接受。
因此,类胡萝卜素通常与辅助化合物(诸如淀粉、树胶、或鱼明胶)组合,从而在含类胡萝卜素的水性组合物中避免相分离。然而,这些辅助化合物常对最终产品的颜色特性和营养特性具有负面影响。因此期望开发具有高颜色强度的新颖的类胡萝卜素组合物。特别存在对改进的辅助化合物的需要,所述辅助化合物在使用它们的最终产品中具有非常良好的粒度、浊度、口味、乳化、乳液稳定性和颜色等性质。
一种天然的水胶体——金合欢胶(也称作阿拉伯树胶(gumarabic))在饮料乳液中被广泛用作乳化剂/稳定剂。它是高度溶于水的(以重量计多达50%),并且其水溶液提供可乳化性、乳液稳定性、包封能力和成膜能力。对金合欢树的茎和枝条施加压力时从中获得呈粘性渗出物的金合欢胶。金合欢胶从Acacia Senegal树中收集,并且其中小部分在非洲撒哈拉区域的若干国家中从Acacia seyal树中收集。金合欢胶是带有鼠李糖和葡萄糖醛酸末端单元的阿拉伯半乳聚糖多糖,其含有2%的蛋白质(OH-脯氨酸、丝氨酸、脯氨酸)和四种糖(L-阿拉伯糖、L-鼠李糖、D-半乳糖、D-葡糖醛酸)(Idris et al,Food Hydrocolloids,Part I to III,12,1998,379-388)。
然而,作为乳化剂,金合欢胶被认为不如产品中通常使用的乳化剂明胶好,并且未被标准化。由于与物种、地理位置和个体生长季节相关的不相似的功能性,不同的批次间可能产生不均衡的树胶性能。另外,金合欢胶的性能取决于土壤、气候和树的年龄。
除了被用作乳化剂外,金合欢胶是碳水化合物例如水解淀粉的最常见的载体材料之一。在香料工业中,阿拉伯树胶在喷雾干燥的应用中被用作固定剂,其中所述树胶包封香料化合物,保护其免于氧化和挥发。少量工作显示金合欢胶(Krishnan et al.,Carbohydrates Polymers,61,2005,95-102;Krishnan et al.Carbohydrates Polymers,62,2005,309-315;Buffo et al,Perfumers&Flavorist,25,2000,45-54)、麦芽糖糊精和改性的食物淀粉的掺合物可代表一种包封基质,所述包封基质在香味保持和抗氧化保护方面具有改进的特性。
在着色产品例如饮料中,还常期望保持饮料的光学透明度(opticalclarity)。用于补充的略溶于水的着色剂例如类胡萝卜素(例如β-胡萝卜素)可以以许多形式获得,但是当添加进饮料时会趋向于提高视觉混浊度。成环(ringing),即液体顶部独立的脂溶性β-胡萝卜素层的形成,也是许多已知的β-胡萝卜素制剂的一个问题。向饮料中添加略溶性物质但不提高视觉混浊度或不成环的一种手段是将所述物质包封在脂质体中。然而,这是一种昂贵的方法,并且脂质体中物质的浓度倾向于较低。
因此,可以以滋补量(restorative amount)或营养补充量添加至饮料的、令人满意的略溶于水的着色剂(例如类胡萝卜素)的组合物应当不影响添加它的饮料的光学透明度,并且不改变所述饮料的感官性质。此外,着色剂组合物不应当引起成环。
WO2002/069981公开了基于二羧酸酐和水胶体的水溶性酯化水胶体,其适合用于生产水包油型乳液,其特征在于,乳液中至少60%的油滴的直径小于2μm。更具体地,辛烯基琥珀酸酐改性的金合欢胶被描述为比金合欢胶本身明显更有效的乳化剂。然而,WO2002/069981仅公开了水包油型乳液(通常为香精油(flavour oils))。
因此,仍然需要用于对食物、饮料、动物饲料、化妆品组合物或药物组合物进行富集、强化和/或着色的类胡萝卜素组合物,所述类胡萝卜素组合物应未表现出上述问题,即未表现出分离现象并且对得到的产物提供增强的颜色强度和颜色稳定性。
因此,提供具有改进的颜色强度和颜色稳定性的类胡萝卜素组合物是本发明的一个目的。这些组合物应当额外地具有上文指出的期望特性,例如在光学透明度和乳液稳定性方面的非常良好的特性。
已经惊奇地发现,包含辛烯基琥珀酸酐改性的金合欢胶类以及任选地还包含其他辅剂和/或赋形剂的本发明的胡萝卜素组合物可以与水混合,从而所得混合物具有高的颜色强度和颜色稳定性。通过使用导致小粒径的特定乳化技术已经实现了意想不到的高颜色值。这种红色至橙色的颜色对可以使用组合物的食物、饮料、动物饲料、化妆品组合物或药物组合物是有利的。此外,没有得到类胡萝卜素从所得混合物中分离。在不存在其他辅助化合物(例如鱼凝胶)或除β-胡萝卜素以外的着色化合物的情况下,可以实现有利的颜色。
因此,本发明还涉及下述组合物,所述组合物包含5至85重量%之间、优选25至85重量%之间、最优选55至85重量%之间的辛烯基琥珀酸酐改性的金合欢胶,和0.1至50重量%的至少一种类胡萝卜素,其中所述重量%基于总组合物以干物质计算。
在本发明的一个优选的实施方式中,基于总组合物以干物质计算,组合物包含0.1和30重量%之间、进一步优选0.2和15重量%之间、最优选0.5和10重量%之间、以及甚至最优选1和5重量%之间的类胡萝卜素。
在本发明的最优选的实施方式中,组合物包含55至85重量%之间的辛烯基琥珀酸酐改性的金合欢胶和1至6重量%之间的至少一种类胡萝卜素,其中所述重量%基于总组合物以干物质计算,并且其中类胡萝卜素选自由β-胡萝卜素和虾青素组成的组。
优选地,本发明的组合物不包含除所述至少一种类胡萝卜素化合物外的其他着色物质。优选地,本发明的组合物不包含鱼明胶。
根据WO2002/069981,本领域技术人员可以生产辛烯基琥珀酸酐改性的金合欢胶。根据本发明的最优选的辛烯基琥珀酸酐改性的金合欢胶可以以A-2010粉末的商品名从TIC Gums,Inc.,Belcamp,(MD)USA购买。
在本发明的另一个优选的实施方式中,组合物的特征在于,类胡萝卜素选自由阿朴胡萝卜醛(apocarotinal)、叶黄素、胭脂树橙、虾青素、番茄红素、β-胡萝卜素、角黄素、桔黄素、玉米黄质、隐黄质、β-阿朴-8’-胡萝卜醛和β-阿朴-12’-胡萝卜醛组成的组。更优选地,至少一种类胡萝卜素是β-胡萝卜素或虾青素,更优选地,至少一种类胡萝卜素为β-胡萝卜素。
在本发明的另一个优选的实施方式中,组合物的特征在于,组合物还包含一种或多种辅剂和/或赋形剂,其中优选这些辅剂和/或赋形剂不代表油。
优选的赋形剂和/或辅剂选自由单糖、二糖、寡糖和多糖、水溶性抗氧化剂和脂溶性抗氧化剂组成的组。
可存在于本发明组合物中的单糖和二糖的例子是蔗糖、转化糖、木糖、葡萄糖、果糖、乳糖、麦芽糖、蔗糖和糖醇。
寡糖和多糖的例子是淀粉、淀粉水解产物例如糊精和麦芽糖糊精,特别是具有5到65葡糖当量(DE)范围的那些,以及葡萄糖浆,特别是具有20到95DE范围的那些。术语“葡糖当量”(DE)表示水解程度,是以干重为基础以D-葡萄糖计的还原糖的量;该标度以DE接近0的天然淀粉和DE为100的葡萄糖为基础。
基于总组合物以干物质计算,根据本发明的组合物优选地包含少于30%、进一步优选少于20重量%、进一步优选少于10重量%、进一步优选少于5重量%的一种或多种油。最优选地,组合物不包含任何油。
本文上下文中使用的术语“油”包括适用于组合物的期望用途的任何甘油三酯或任何其他油。甘油三酯适当地为植物油或脂肪、优选地为玉米油、葵花子油、大豆油、红花油、菜籽油、花生油、棕榈油、棕榈仁油、棉籽油、橙油、柠檬烯、橄榄油或椰子油。
固体组合物可额外含有一种或多种抗结块剂,例如硅酸或磷酸三钙等等,并且至多为10重量%,优选地为0.1到5重量%。
水溶性抗氧化剂可以是例如抗坏血酸或其盐(优选地为抗坏血酸钠)、水溶性多酚例如羟基酪醇和橄榄苦苷、苷元、表没食子儿茶素没食子酸酯(EGCG)或迷迭香提取物或橄榄提取物。
脂溶性抗氧化剂可以是例如生育酚,例如dl-α-生育酚(即合成的生育酚)、d-α-生育酚(即天然生育酚)、β-生育酚或γ-生育酚,或其两种或多种的混合物;丁基化的羟基甲苯(BHT);丁基化的羟基茴香醚(BHA);乙氧基喹啉、没食子酸丙酯;叔丁基羟基喹啉;或6-乙氧基-1,2-二羟基-2,2,4-三甲基喹啉(EMQ),或脂肪酸的抗坏血酸酯,优选地为棕榈酸抗坏血酸酯或硬脂酸抗坏血酸酯。
另外,所述组合物还可以包含水或任何其他溶剂。如果组合物为液体,其可包含从几个ppm到更高浓度的溶剂(例如水)。
表1示出了基于总组合物以干重计算的,组合物各成分的优选用量(重量%)。另外,表1中规定的用量可与上文所述的各成分的优选用量组合。

可以通过现有技术中已知的方法来制造根据本发明的组合物。作为例子,组合物的制造包含以下步骤:
I)在室温下搅拌至少1小时,将辛烯基琥珀酸酐改性的金合欢胶溶解于水中;
II)向步骤I)的溶液中添加有机相,所述有机相包含至少一种类胡萝卜素并任选地包含至少一种有机溶剂
III)用本领域技术人员已知的传统乳化方法将步骤II)的混合物匀化,
IV)在减压下蒸发有机溶剂,
V)通过喷雾干燥、粉末捕获或其他方法干燥乳液。
因此,在另一个实施方式中,根据本发明的组合物的特征在于,其为乳液。乳液是两种或多种不混溶(不可共混)的液体的混合物。乳液是被称为胶体的更上位的一类物质两相体系的部分。虽然术语胶体和乳液有时可以互换使用,但乳液倾向于暗示分散相和连续相二者都是液体。在乳液中,一种液体(分散相)分散于另一种(连续相)之中。
本发明的组合物优选地为添加剂组合物,并且优选地用作添加剂组合物。
令人惊奇的是,与标准的基于金合欢胶的乳液相比,以及与WO02/069981中描述的水包油型乳液相比,具有根据本发明的组合物的乳液的粒径非常小。因此,本发明还涉及如上所述根据本发明的组合物用于食物、饮料、动物饲料、化妆品组合物或药物组合物的富集、强化和/或着色,优选地用于饮料的富集、强化和/或着色的用途,其中至少50%、优选至少70%、更优选至少80%、最优选至少90%、甚至更优选至少95%的乳液颗粒具有小于1微米的直径。可以通过现有技术已知的方法(例如:Coulter Counter)测量粒度。
在另一个实施方式中,本发明涉及根据本发明的乳液用于饮料的富集、强化和/或着色的用途,其中,以5ppm的浓度包含所述乳液的饮料的比浊法测量值(nephelometric value)低于20NTU。NTU代表比浊法测量的浊度单位,并且可以由本领域技术人员用校准过的浊度计测量。
在另一个实施方式中,本发明涉及根据本发明的乳液用于饮料的富集、强化和/或着色的用途,其中5ppm的乳液具有至少800、优选至少1000、最优选至少1200、甚至更优选1400的颜色强度值(E1/1),其中颜色强度值E1/1为1%的组合物溶液和1cm厚度的吸光度,并且如下计算:E1/1=(Amax-A650)*稀释因子/(样品重量*产品形式的含量以%计)。
通过测量组合物的吸光度来进行乳液颜色强度值(对应于乳化性能)的测定。通过使用包含5%的β-胡萝卜素的模型系统来测定根据本发明的乳液的颜色强度值。
为了测量消光系数,将足量的根据本发明的组合物分散、溶解于水中和/或用水稀释。将所得“溶液”稀释至5ppm的β-胡萝卜素的最终浓度,并且以水作为参比测量其UV/VIS谱。从所得的UV/VIS谱测定最大或肩峰(shoulder)的特定波长下的吸光度,Amax。另外,测定650nm处的吸光度A650。颜色强度E1/1为1%溶液和1cm厚度的吸光度,其计算方法如下:E1/1=(Amax-A650)*稀释因子/(样品重量*产品形式的含量以%计)。
本发明的其他方面为含有上述组合物的食物、饮料、动物饲料、化妆品组合物或药物组合物。
其中可使用本发明的产品形式用于饮料的富集、强化和/或着色的饮料可以是碳酸饮料,例如调味的塞耳特斯水(flavored seltzer water)、软饮或矿物饮品,以及非碳酸饮料,例如调味的水、果汁、水果宾治(fruit punches)和这些饮料的浓缩形式。它们可以基于天然的水果汁或蔬菜汁或基于人工香料。还包括在内的是酒精饮料和即食饮料粉(instant beverage powder)。另外,还包括含糖饮料,含有无热量人工甜味剂的减肥饮料(dietbeverage)。
另外,得自天然或合成来源的乳制品也属于食物产品的范围,其中本发明的产物形式可被用于产品的富集、强化和/或着色。这类产品的典型例子是乳饮品、冰激凌、乳酪、酸乳等等。乳替代产品如豆浆饮品和豆腐制品也包括在该应用范围内。
还包括在内的是含有用于产品的富集、强化和/或着色的本发明的产品形式的甜品(sweets),如糖果制品(confectionery product)、糖果、口香糖、甜点,例如冰激凌、果冻(jellies)、布丁、即食布丁粉等等。
还包括在内的是谷物、点心、曲奇、意大利面(pasta)、汤和沙司(sauces)、蛋黄酱、沙拉酱等等,其含有用于产品的富集、强化和/或着色的本发明的产品形式。另外,用于乳和谷物的水果制备物也包括在内。
借由本发明的组合物添加至食物产品中的β-胡萝卜素的最终浓度以食物组合物的总重为基础可以优选地从0.1到50ppm、尤其是从1到30ppm、更优选3到20ppm,例如约6ppm,并取决于待着色或强化的具体食物制品和着色或强化的预期等级。
优选地通过以本发明组合物的形式向食物产品中添加类胡萝卜素,来获得本发明的食物组合物。为了食物或药物制品的富集、强化和/或着色,可根据本身已知的、应用可分散于水中的固体产品形式的方法使用本发明的组合物。
通常,根据特定的应用,组合物可作为水性储存溶液添加,或者作为与适用于其他食物成分的干粉混合物或预混合物添加。根据最终应用的组成,可以使用例如干粉搅拌器、低剪力混合器、高压匀化器或高剪力混合器完成混合。显而易见的是,这类技术是本领域技术人员已知的。
其中使用本发明的组合物作为着色剂的药物组合物例如片剂或胶囊也在本发明的范围内。可如下完成片剂的着色:以液体或固体着色组合物的形式,向片剂涂层混合物中单独地添加组合物,或向片剂涂层混合物的一种组分中添加组合物。可通过在胶囊物质的水溶液中掺入组合物,制备有色的硬壳或软壳胶囊。
其中使用本发明的组合物作为活性成分的药物组合物例如片剂(例如咀嚼片、泡腾片或薄膜包衣片)或胶囊(例如硬壳胶囊)也在本发明的范围内。产品形式通常作为粉末被添加至制片混合物中,或以用于生产胶囊的本身已知的方式被填充进胶囊中。
其中使用本发明的组合物作为色素沉着着色剂(例如用于卵黄、肉鸡(table poultry)、仔鸡(broilers)或水生动物)或作为活性成分的动物饲料产品也在本发明的范围内,所述动物饲料制品例如为营养成分的预混物、复合饲料、乳替代物、液体膳食或饲料制剂。
其中使用本发明的组合物作为着色剂或作为添加剂或作为活性成分的化妆品(cosmetics)、美容用品(toiletries)和皮肤用品(derma product)也在本发明的范围内,所述化妆品、美容用品和皮肤用品即皮肤和头发护理制品例如乳霜、洗剂、沐浴用品、唇膏、洗发水、护发素、喷雾或凝胶。
本发明还涉及制造饮料的方法,其包含下述步骤:匀化如权利要求1至5中任意一项所述的组合物,以及将基于类胡萝卜素含量的1至50ppm、优选5ppm的乳化的组合物与饮料的其他常用成分混合。
此外,本发明涉及通过如上所述制造饮料的方法可得到的饮料。
本发明还通过以下的实施例进一步阐述,所述实施例并非意图限制本发明的范围。
实施例
实施例1
包含辛烯基琥珀酸酐改性的金合欢胶和β-胡萝卜素的组合物的制备
在反应容器中,在室温下用锚式搅拌器将辛烯基琥珀酸酐改性的金合欢胶(A-2010)溶解于去离子水中,直到全部溶解。然后将有机相加入到水相中,并匀化所得到的混合物。最终,在减压下蒸发溶剂,并将所得产品转化成干燥形式。

实施例2
液体乳液的表征
借由Zeta电势测量来表征实施例1的液体乳液。分析前,将该材料稀释于milli-Q水中,从而获得用于测量的足够的散射所需要的0.1至1%重量/体积的最小浓度。因为这种测量不依赖于尺寸,所以选择Smoluchowski理论用于目标计算。根据这种方法,该产品表现出~51.5mV的Zeta电势值。
用动态光散射进行粒度分析。用milli-Q水稀释乳液,直到浓度低于0.1%(体积),然后在两个角度下测量:173°和90°。所得粒度为173°时247nm以及90°时226nm。
另外使用激光衍射来测定粒度。在这种情况下,在测量单元中直接稀释乳液至20%的合适遮光率,并且采用Sauter直径作为目标值。所有粒子低于1μm,并且Sauter直径为270nm。
对于颜色强度测量,采用5ppm的β-胡萝卜素溶液,并使用UV/Vis光谱仪分析。根据最大吸光度(Amax)和650nm处的吸光度(A650),通过下面的公式计算颜色强度:(Amax-A650)*稀释因子/(样品重量*产品形式的含量以%计)。得到的值是1549。
使用相同溶液(5ppmβ-胡萝卜素)来测定材料的混浊度(浊度)。相应产品的比浊法测量值为13.6NTU。
实施例3
液体组合物的喷雾干燥
在氮气下将如实施例1中制备的300gβ-胡萝卜素乳液在配备有两个流体喷嘴和高性能气旋器(cyclone)的Mini Spray Dryer B-290Advanced(Büchi)中喷雾干燥。将乳液加热至40℃,工艺参数设为:T°in:190℃,T°out:85℃,流速:40m3.hr-1,抽吸率(aspirator):90%。
收集40g产品,并贮存在氮气下,以待进一步表征。
实施例4
喷雾干燥形式的表征
借由Zeta电势测量来表征实施例3中制备的喷雾干燥的产品。分析前,将该材料以0.1至1%重量/体积的最小浓度溶解于milli-Q水中,以获得足够的散射。因为这种测量不依赖于尺寸,所以选择Smoluchowski理论用于目标计算。根据这种方法,该产品表现出~60mV的Zeta电势值。
用动态光散射进行粒度分析。将喷雾干燥的产品溶解于milli-Q水中,直到浓度低于0.1%(体积),然后在两个角度下测量:173°和90°。所得粒度为173°时203nm以及90°时213nm。
另外使用激光衍射来测定粒度。在这种情况下,在测量单元中直接稀释所溶解的喷雾干燥的产品至20%的合适遮光率,并且采用Sauter直径作为目标值。所有粒子低于1μm,并且Sauter直径为270nm。
对于颜色强度测量,采用相应的5ppm的β-胡萝卜素溶液,并使用UV/Vis光谱仪分析。根据最大吸光度(Amax)和650nm处吸光度(A650),通过下面的公式计算颜色强度:
(Amax-A650)*稀释因子/(样品重量*产品形式的含量以%计)。得到的值是1400。
使用相同溶液(5ppmβ-胡萝卜素)来测定材料的混浊度(浊度)。相应产品的比浊法测量值为14NTU。
实施例5
含辛烯基琥珀酸酐改性的金合欢胶和虾青素的组合物的制备
在反应容器中,在室温下用锚式搅拌器使辛烯基琥珀酸酐改性的金合欢胶(A-2010)溶解于去离子水中,直到全部溶解。然后将溶剂中虾青素、蜂蜡和dl-α-生育酚的溶液加入到水相中,并匀化所得到的混合物。最终,在减压下蒸发溶剂,并将所得产品通过喷嘴喷雾。用玉米淀粉处理所得粉末,并干燥。

Claims (12)
1.包含5至85重量%之间的辛烯基琥珀酸酐改性的金合欢胶和0.1至50重量%之间的至少一种类胡萝卜素的组合物,其中所述重量%基于总组合物以干物质计算。
2.如权利要求1所述的组合物,其特征在于,基于总组合物以干物质计算,所述组合物包含0.5至10重量%之间的至少一种类胡萝卜素。
3.如权利要求1或2中任意一项所述的组合物,其特征在于至少一种类胡萝卜素选自由阿朴胡萝卜醛、叶黄素、虾青素、番茄红素、β-胡萝卜素、角黄素、桔黄素组成的组。
4.如权利要求1至3中任意一项所述的组合物,其特征在于至少一种类胡萝卜素为β-胡萝卜素。
5.如权利要求1至4中任意一项所述的组合物,其特征在于所述组合物还包含一种或多种辅剂和/或赋形剂。
6.如权利要求1至5中任意一项所述的组合物,其特征在于所述组合物为乳液。
7.根据权利要求6所述的组合物用于食物、饮料、化妆品组合物或药物组合物的富集、强化和/或着色的用途,其中至少50%的乳液颗粒具有小于1微米的直径。
8.如权利要求6中所述的组合物的用途,其中至少90%的乳液颗粒具有小于1微米的直径。
9.如权利要求6中所述的组合物的用途,其中相应产品的比浊法测量值低于20NTU。
10.如权利要求6中所述的组合物的用途,其中所述乳液具有至少800的颜色强度值(E1/1),其中所述颜色强度E1/1为1%的溶液和1cm厚度下的吸光度,并且如下计算:E1/1=(Amax-A650)*稀释因子/(样品重量*产品形式的含量以%计)。
11.制造饮料的方法,其包含如下步骤:
将如权利要求1至5中任意一项所述的组合物匀化,以及
将乳化的组合物与饮料的其他常用成分混合。
12.通过如权利要求11所述的方法可得到的饮料。
