Abstract

本发明公开了一种新生牛血清无菌混合工艺:在GMP车间内,将需要混合的密封包装的新生牛血清在万级条件下置于乙醇消毒池中,浸泡10~20min,以杀灭包装袋表面的细菌、支原体等微生物,然后通过无菌传递窗进入百级车间,在百级条件下晾干包装袋,然后将袋中新生牛血清倾入无菌混合罐中,搅拌混匀,然后进入常规多级过滤系统及其他常规程序。本发明的整套混合工艺具有全程封闭的特点,消除了操作过程中细菌、支原体、噬菌体等微生物的污染机会,适用于畜牧业疫苗药业。本发明具有操作简单、方便有效等优点。

Description

一种新生牛血清无菌混合工艺
技术领域
本发明涉及一种新生牛血清的无菌混合工艺。
背景技术
目前国内市场的牛血清生产均按照国标规定内容生产。国标对新生牛血清产品规定符合如下要求:1、新生牛血清根据出生后时间的不同(0.5小时内、出生2小时内、出生4小时内、出生14小时内)分为顶级、超级、特技和普通4级。2、规定蛋白总量为3.5-5.0(W/V),血红蛋白含量≤0.035(W/V),内毒素含量≤10EU/ml,无菌试验阴性、支原体阴性和大肠杆菌噬菌体阴性。3、牛腹泻病毒抗体和口蹄疫抗体阴性。4、对血清促进BHK细胞和SP2/0的细胞生长的情况做了规定(见表1)。
但在实际新生牛血清生产过程中,一般工艺难以做到无支原体污染。为了减少支原体污染,公开号为CN1961981A的发明专利公开了一种用多联血袋全封闭无菌生产原料动物血清的方法,从源头杜绝了支原体的污染。然而仅仅在原料牛血清的生产过程中杜绝支原体的污染是远远不够的,原因为:产品牛血清生产过程环节众多、繁杂,存有多个污染漏洞,比如:为了保障产品血清的均一性,以保障细胞培养过程的稳定性和疫苗病毒的产量,必须将多批次的牛血清进行混合。目前血清混合工艺多是在洁净环境开放条件下完成的,皮肤、唾液气溶胶、呼吸气溶胶、汗渍等人体脱落物以及原料血清外包装携带的包括支原体在内的微生物都有可能在混合过程进入牛血清。为了保障血清无菌状态,可以采取多级过滤程序,以除掉混合过程中污染的细菌。但是,支原体具有形态学可变特性,高压条件下支原体可以通过变型而通过微孔滤膜,因此,多级过滤程序并不能防止支原体的污染,产品血清污染支原体的几率依然非常大。
发明内容
针对上述现有技术的不足,本发明提供了一种可彻底断绝支原体污染的新生牛血清无菌混合工艺。
一种新生牛血清无菌混合工艺,其特征在于:在GMP车间内,将需要混合的密封包装的新生牛血清在万级条件下置于乙醇消毒池中,浸泡10~20min,以杀灭包装袋表面的细菌、支原体等微生物,然后通过无菌传递窗进入百级车间,在百级条件下晾干包装袋,然后将袋中新生牛血清倾入无菌混合罐中,搅拌混匀,然后进入常规多级过滤系统及其他常规程序。
所述乙醇消毒池中乙醇的体积分数为72%~78%。
本发明的整套混合工艺具有全程封闭的特点,消除了操作过程中细菌、支原体、噬菌体等微生物的污染机会,适用于畜牧业疫苗药业。本发明具有操作简单、方便有效等优点。
具体实施方式
下面结合实施例对本发明作进一步的说明:
实施例:原料新生牛血清的生产:最小批量50头新生牛血清
1.1 20080001号新生牛血清的生产
1.1.1新生牛血清档案:
a新生牛血清编号20080001,
b新生牛的隶属关系:山东佳宝乳业乳牛基地
c出生地:山东省济南市长清区佳宝乳业乳牛基地产房。
d出生时间:2008年6月17日13时.25分。
e取血时间:2008年6月17日14时10分
f体重:45kg,精神好,未进食,无腹泻,体表粘液未干;
g新生牛母亲的健康状况,第2胎
h母牛疫苗接种史:口蹄疫疫苗、牛流行热、布氏杆菌、炭疽
i母牛病史:1年来未发生明显疾病;
j周围牛群的疫情发生情况:3年来未发生重大疫情。
k周围畜牧养殖动物的疫情发生情况:隔离喂养,与周围动物无接触。也无禽流感等疫情发生。
1.1.2 血清采集:无菌封闭式采血(可以采用公开号为CN1961981A的发明专利公开的方法)。每份血清取4份留样,分别为①、②、③、④、⑤,做标签后-20冻存备做检测;
1.2 20080002~50号新生牛血清的生产
1.2.1 新生牛血清档案:
1.2.2 血清采集:同20080001号新生牛血清的采集方法。留样5份,标签后-20冻存备做检测。
2 留样检测:
包括微生物学检测(细菌、真菌、支原体、E.Coli(C300 or K-12)噬菌体)、细菌内毒素检测、蛋白含量检测、血红蛋白定量检测、血清抗体检测(牛腹泻病毒抗体和口蹄疫抗体)。
3产品新生牛血清的生产:
3.1 归类合并:
3.1.1 乙醇灭菌程序:首先将原料血清通过物流通道运输至GMP车间的乙醇池,在72~78%乙醇中浸泡15~20min,以杀灭包装袋表面的细菌、支原体等微生物,然后通过传递窗口将浸泡过的原料血清传递至下一个生产程序。
3.1.2 混合均一程序:GMP车间百级条件下混合车间进行:将传递窗口输入的原料牛血清无菌条件下打开并加入混合罐中,搅拌混匀,然后通过封闭式管道,将血清泵入“多级过滤和灌装程序”。
3.1.3 多级过滤和无菌灌装程序:将混合均一的血清通过过滤、自动灌装系统灌装、加塞。
3.1.4:赋予标签、档案:
3.1.5 抽检、赋予合格证:抽检内容包括:无菌检测、细菌毒素检测、支原体检测、抗体检测、细胞增殖试验。
实施例中除3.1.1、3.1.2外,其他均为公知技术。

1.一种新生牛血清无菌混合工艺,其特征在于:在GMP车间内,将需要混合的密封包装的新生牛血清在万级条件下置于乙醇消毒池中,浸泡10~20min,然后通过无菌传递窗进入百级车间,在百级条件下晾干包装袋,然后将袋中新生牛血清倾入无菌混合罐中,搅拌混匀,然后进入常规多级过滤系统及其他常规程序;
所述乙醇消毒池中乙醇的体积分数为72%~78%。
