Description
技术领域
本发明涉及一种含牛蒡多糖具有调节肠道菌群功能的多肽组合物及应用,属于保健食品技术领域。
背景技术
肠道是人体重要的消化器官,负责营养物质消化和吸收,同时保持机体内环境稳定的屏障,肠道屏障包括机械屏障、生物屏障、化学屏障以及免疫屏障。不同的屏障具有相对的结构基础和不同的调控机制和生物学功能,它们之间通过信号通路有机的结合在一起,共同抵御外来抗原性物质对机体的侵袭,防止肠内微生物群失调。
人体作为一个复杂的生态系统寄生了数以百万亿计的微生物,除皮肤表面外主要的菌群集中在胃肠道系统。肠道菌群是一个与宿主存在共生关系的复杂生态系统。人体肠道内的微生物超过99%都是细菌,约100万亿细菌,经16 S测序鉴定出1000-1150个不同的种类,足以证明肠道菌群种类的多样性。
菌群平衡与肠道健康,甚至人体机体健康息息相关。肠道菌群中既包含了对身体有益的微生物群,如乳酸杆菌和占主导地位的双歧杆菌,又包含一些潜在的致病菌,如铜绿假单胞菌、变形杆菌、大肠埃希菌等,它们处于一种共生的状态。健康人体中有益菌的比例达到70%,并与有害菌相互作用,共同维持着肠内微生态的平衡,维持人体健康。一旦机体由于各种因素影响体内外环境变化而使肠道中生态平衡破坏,如使用抗生素使肠道有益菌被抑制,有害菌大量繁殖, 从而是肠道菌群失调,引起腹泻等多种临床症状,导致疾病。
目前使用的调节肠道菌群功能的产品包括有益生菌类产品,消化酶类产品,低聚糖等益生元类产品,中药类产品等。这些产品各有其缺陷,益生菌类产品和消化酶类产品成本较高,保存条件要求严格,活性影响条件较大,保质期短;低聚糖等益生元类产品虽然贮存使用方便,无毒副作用,但单独使用调整肠道菌群的效果也比较有限;中药类产品以中医理论为基础,可以对肠道和机体进行整体性调整,但是副作用较大,易引起腹泻,口感较差。
发明内容
本发明的目的在于提供一种含牛蒡多糖具有调节肠道菌群功能的多肽组合物,该组合物将肠道微生态学理论和营养学、功能医学以及药食同源的中医药理论结合在一起,通过合适的原料选择和配伍,达到调节肠道菌群有益菌和有害菌的生长,缓解肠道菌群失调,维持肠道正常功能的目的。
为实现本发明的目的,本发明技术方案如下:
一种含牛蒡多糖的调节肠道菌群的多肽组合物,由下述组分按所述重量份数组成:多肽粉20份~50份、低聚肽粉20份~40份、谷氨酰胺5份~20份、浓缩水果粉5份~20份、牛蒡多糖1份~10份、低聚木糖1份~10份、水苏糖1份~10份、酒石酸0份~10份、三氯蔗糖0份~10份。
优选地,本发明含牛蒡多糖的调节肠道菌群的多肽组合物由下述组分按所述重量份数组成:多肽粉25份~45份、低聚肽粉21份~35份、谷氨酰胺8份~15份、浓缩水果粉10份~15份、牛蒡多糖2份~6份、低聚木糖1份~6份、水苏糖2份~8份、酒石酸1份~5份、三氯蔗糖1份~5份。
本发明提供了含牛蒡多糖的调节肠道菌群的多肽组合物在制备抗肿瘤和改善胃肠功能的药品、特殊医学用途配方食品、保健食品或食品中的应用。
本发明含牛蒡多糖具有调节肠道菌群功能的多肽组合物中,牛蒡多糖可以降低内源性自由基的产生,促进肠道有益菌乳杆菌和双歧杆菌的增殖,而且随着多糖剂量增加,增殖效果也较为明显。食源性多肽(包括大豆肽粉、乳清蛋白肽粉,骨胶原蛋白肽粉、小麦低聚肽粉、玉米低聚肽粉等)可以改善小肠粘膜组织形态结构,增加小肠绒毛的高度和表面,加深隐窝的深度,改善和维持肠壁粘膜细胞结构和功能的完整性。食源性多肽还可以增加巨噬细胞的吞噬率和吞噬指数,增强淋巴细胞的增殖能力,促进抗体形成细胞数量增加,提高机体的免疫水平。谷氨酰胺(glutamine,GLN)是快速分裂型细胞(如肠道上皮细胞、淋巴细胞和其他免疫细胞)的重要能源,是危重症病人的条件必需氨基酸。在肠屏障受损、细菌内毒素易位时,补充谷氨酰胺可减轻肠黏膜萎缩,修复肠屏障,降低肠壁通透性.并能促进淋巴细胞、单核一巨噬细胞增殖,增强免疫功能,减少肠道细菌内毒素易位。
本发明是在现有肠道微生态学理论的基础上,以营养学和功能医学为基础进行配方组配,筛选出能有效调节肠道菌群的多肽类成分和低聚糖类成分,辅以提供营养底物且具有保护肠粘膜屏障作用的谷氨酰胺,再配以药食同源的牛蒡多糖配伍,从而获得一种具有调节肠道菌群功能的复方多肽类组合物。经试验发现:本发明提供的含牛蒡多糖具有调节肠道菌群功能的多肽组合物可以显著调节肠道菌群的数量,提高肠道益生菌的数量,维持肠道正常功能。

实施例1
一种含牛蒡多糖的调节肠道菌群的多肽组合物,由下述组分按所述重量份数组成:大豆肽粉20份、乳清蛋白多肽粉25份、小麦低聚肽粉10份、玉米低聚肽粉12份、谷氨酰胺8份、西番莲水果粉10份、牛蒡多糖3份、低聚木糖5份、水苏糖5份、酒石酸1份、三氯蔗糖1份。
上述含牛蒡多糖的调节肠道菌群的多肽组合物的制备方法包括如下步骤:
1)按照上述配比称取大豆肽粉粉、乳清蛋白多肽粉、小麦低聚肽粉、玉米低聚肽粉、谷氨酰胺、西番莲水果粉、牛蒡多糖、低聚木糖和水苏糖;
2)将步骤1)的各原料与按照上述配比称取的酒石酸和三氯蔗糖混合,过65目筛,置于多维混合机中,混合均匀;
3)将步骤2)所得产品按国家标准进行抽检,检验合格后,分装,质量部门出具合格证明后,外包,入库。
实施例2

上述含牛蒡多糖的调节肠道菌群的多肽组合物的制备方法包括如下步骤:
1)按照上述配比称取大豆肽粉、小麦低聚肽粉、玉米低聚肽粉、谷氨酰胺、菠萝水果粉、牛蒡多糖、低聚木糖和水苏糖;

上述含牛蒡多糖的调节肠道菌群的多肽组合物的制备方法包括如下步骤:
1)按照上述配比称取大豆肽粉粉、乳清蛋白多肽粉、骨胶原蛋白肽粉、玉米低聚肽粉、谷氨酰胺、甜橙水果粉、牛蒡多糖、低聚木糖和水苏糖;

本发明保健食品组合物药效学研究:
本发明以体外结肠发酵模型模拟人结肠及其微生物菌群来测试不同样品
1、实验仪器
BS2202S型电子天平,德国赛多利斯公司;
HFsafe-1200LC型生物安全柜,上海力申科学仪器有限公司;
LRH-250生化培养箱,上海一恒科学仪器有限公司;
EpicS XL流式细胞仪,美国Beckmen公司;
2、实验试剂
NaCl,NaH2PO4,Na2HPO4,多聚甲醛,NaOH,HCl,EDTA,SDS,甲酰胺等均购自国药试剂;
3、实验方法
(1)肠道菌定植
采取健康男性捐赠者粪便以结冷胶和黄原胶制备胶粒,准备5支无菌厌氧管,每支加入新鲜制备的胶粒 1.5g,并加入 17ml新鲜配置的培养基加盖.旋紧,37℃恒温厌氧培养箱培养.在此阶段,每 12h 更换一次新鲜培养基,共需 24h。剩余培养基包扎严密后置于冰箱冷藏。
(2)发酵实验
将最后一次的培养液除掉之后,将5支厌氧管按如下发酵体系:
①空白、②阳性对照样品(菊粉)、③实施例1、④实施例2、⑤实施例3
所有分组于37℃恒温厌氧培养箱中培养,并做好标记。在发酵至24h时,分别取 2ml 菌液用于荧光原位杂交。
(3)荧光原位杂交及流式细胞术分析
1) 配制 PBS 缓冲液:145mM NaCl,1.4mM NaH2PO4,8mM Na2HPO4,调 pH 至7.4。
2) 配制 4%多聚甲醛(PFA):每次配制 10mL,现配先用。
取 0.4g 多聚甲醛加入 10mL 容量瓶中,加 8 毫升 PBS,加 1 毫升2M 的 NaOH溶液,放到 60℃的水浴里,10 分钟,拿出来用手颠倒几次,充分溶解,再用 HCl 调 pH 到7.2,定容。
3) 取 2mL 发酵液,4000g 离心分离 10min,去除上清,用 750uLPBS 缓冲液洗涤细胞. 4000r离心 10min,弃上清.添加 750uL 的 PBS,重新悬浮,加入 250μL 的 4%的PFA 进行固定(PFA 终浓度 1%)。
4)4℃孵育 24h,4000r 离心 10min,弃上清,用 500μL 的 PBS 完全固定。再次4000r 离心 10min,弃上清,用 500μl 的 PBS 完全固定,加入 500μL 的乙醇重新彻底悬浮菌体,放置于-20℃可保存数月。
5)配制TE 缓冲液:称取 1 mol/L Tris,用 HCl 调节 pH 至 8.0. 配制 TE 缓冲液:100 mmol/L Tris-HCl,50 mmol/L EDTA,调 pH 至 8.0。
6)将菌悬液调至合适浓度,取 600uL 菌悬液于 1.5mL离心管中,离心,沉淀用1mLPBS 洗涤,10000g 离心 3min,加入1ml PBS 洗涤,再次离心,加入 1 mL TE 缓冲液,离心,再加入 1mL TE 缓冲液洗涤,离心,弃上清。
7)取 1 mL 新鲜配制的 TE 缓冲液,加入 1 mg 溶菌酶混匀,制备成 1 mg/mL 的溶菌酶溶液,加入 1.5mL离心管的沉淀中,37℃下放置 25~50 min,超声 25min,进行透化.离心后沉淀用 1mL PBS 缓冲液冲洗,再离心后沉淀用 1 mL 杂交缓冲液(900 mmol/LNaCl,20 mmol/L Tris-HCl, 0.01% SDS,去离子甲酰胺浓度取决于探针)冲洗,8000r/min离心 3min,收集沉淀,加入 600ul的0%杂交液悬浮菌体。取 50 μL 菌悬液加入杂交小管中,并加入 4 μL 合适浓度探针(杂交液浓度不同的探针,需先离心再加入相应浓度的杂交液),在每种探针的合适杂交温度下暗处反应一定时间。杂交完毕后,每管中加入杂交缓冲液 150 μL,4℃,10000 r/min 离心 3 min.沉淀物在 500μL 洗脱缓冲液(0.9 mmol/LNaCl,20mmol/L Tris-HCl,5 mmol/L EDTA,0.01% SDS)中重新悬浮,37℃放置 20min,室温下离心收集沉淀,加入 500 μL 无菌 PBS 缓冲液上机,结果以细菌与特异性探针杂交的数目与总菌EUB338 探针杂交的数目比例来计。
(4)结果分析

由表1所示结果可以看出,实施例1、2、3与菊粉和空白对照相比,总菌数量增多,实施例1、2、3促进有益菌(双歧杆菌和乳杆菌)增殖的效果都优于菊粉,且促进双歧杆菌和乳杆菌增殖的效果各有不同。实施例1、2、3对有害菌(粪肠球菌和大肠杆菌)增殖的效果均低于空白对照,显示其均有抑制有害菌繁殖的作用。
2)菌群构成丰富性

在健康的人体肠道菌群中优势菌群主要包括:1)硬壁门的Clostridiumcoccoides- Eubacterium rectale group和Clostridium leptum group,2)拟杆菌门的Bacteroides- Prevotella species,3)放线菌门的Bifidobacterium和Atopobium genera。这些菌群约占总菌的90%,是肠膜菌群的主要构成者,具有营养及免疫调节作用,肠道的非优势菌群如肠球菌、肠杆菌,在肠道微生态平衡时是无害的,在特定的条件下具有侵袭性,对动物宿主有害。
由表2所示结果可以看出,实施例1优势菌群中梭球菌含量最高,促进梭球菌增殖效果最强,实施例2优势菌群中拟杆菌含量最高,促进拟杆菌增殖效果最强,实施例3优势菌群中乳杆菌含量最高,促进乳杆菌增殖效果最强。非优势菌群中阳性对照菊粉的肠杆菌、大肠杆菌和粪肠球菌含量最低,低于空白对照,说明菊粉肠杆菌、大肠杆菌和粪肠球菌等机会致病菌繁殖的抑制作用最强。实施例1、2、3的肠杆菌、大肠杆菌和粪肠球菌含量低于空白对照,说明实施例1、2、3均对肠杆菌、大肠杆菌和粪肠球菌等机会致病菌繁殖具有抑制作用。

1.一种含牛蒡多糖的调节肠道菌群的多肽组合物,其特征在于,包括下述组分按所述重量份数组成:多肽粉20份~50份、低聚肽粉20份~40份、谷氨酰胺5份~20份、浓缩水果粉5份~20份、牛蒡多糖1份~10份、低聚木糖1份~10份、水苏糖1份~10份、酒石酸0份~10份、三氯蔗糖0份~10份。
2.根据权利要求1所述的含牛蒡多糖的调节肠道菌群的多肽组合物,其特征在于,包括下述组分按所述重量份数组成:多肽粉25份~45份、低聚肽粉21份~35份、谷氨酰胺8份~15份、浓缩水果粉10份~15份、牛蒡多糖2份~6份、低聚木糖1份~6份、水苏糖2份~8份、酒石酸1份~5份、三氯蔗糖1份~5份。
3.根据权利要求1所述的含牛蒡多糖的调节肠道菌群的多肽组合物,其特征在于,所述含牛蒡多糖的调节肠道菌群功能的多肽组合物制成胶囊剂、片剂、散剂、颗粒剂或口服液。
4.根据权利要求1所述的含牛蒡多糖的调节肠道菌群的多肽组合物,其特征在于,所述的多肽粉可以为大豆肽粉、乳清蛋白多肽粉、卵白蛋白肽、骨胶原蛋白肽粉、豌豆肽粉、核桃肽粉其中的一种、几种或者全部混合。
5.根据权利要求1所述的含牛蒡多糖的调节肠道菌群的多肽组合物,其特征在于,所述的低聚肽粉为小麦低聚肽粉、玉米低聚肽粉、鱼胶原低聚肽粉其中的一种、几种或者全部混合。
6.根据权利要求1所述的含牛蒡多糖的调节肠道菌群的多肽组合物,其特征在于,所述的浓缩水果粉为甜橙果汁粉、西番莲果汁粉粉、菠萝果汁粉、百香果果汁粉、苹果果汁粉其中的一种、几种或者全部混合。
7.根据权利要求1所述的含牛蒡多糖的调节肠道菌群的多肽组合物,其特征在于,所述含牛蒡多糖按以下步骤制备得到:先将牛蒡洗净,切片或打碎,用水浸泡后煎煮,收集滤液,浓缩得到牛蒡浸提液;然后在获得的牛蒡浸提液中加入乙醇或乙醇溶液,至乙醇终体积百分比浓度为60%-78%,放置于4℃静置4小时及以上,离心收集沉淀,将沉淀用水溶解,然后在水中透析去除蛋白质、低聚糖、无机盐、乙醇等;浓缩透析液,喷雾干燥,得到牛蒡多糖。
8.根据权利要求1至7任一项所述的含牛蒡多糖的调节肠道菌群的多肽组合物在制备抗肿瘤和改善胃肠功能的药品、特殊医学用途配方食品、保健食品或食品中的应用。
