人的全身由皮肤覆盖。因此,皮肤被认为是人体最大的器官,且通常由2层,即表皮与真皮构成。表皮是位于皮肤最外的最薄的层,起到了担当皮肤的保湿及保护的重要的功能。详细地说,表皮阻止皮肤干燥,而且,防止紫外线、病毒、细菌等有害物质的侵入。进一步地,其使皮脂腺的脂与汗腺的水自然的乳化形成弱酸性的皮脂膜,防止有害物质的侵入,杀死细菌。表皮主要由角质形成细胞(keratinocytes)构成,且由角质形成细胞的增殖及分化的高度调节平衡而得以维持。表皮的角质形成细胞分化而形成基底层、棘层、颗粒层及角质层4层。特别地,角质形成细胞,主要通过物理障壁,起到保护个体不受外部环境伤害的作用,以及生成和/或分泌多种调节生物学反应的物质,包括调解免疫应答的细胞因子,从而调节皮肤的免疫反应与炎症反应。角质形成细胞起到正常的生理功能时,能够维持健康的美丽的皮肤。
真皮,向表皮供给营养成分,支撑表皮,保护身体免受外部损伤,且有贮藏水分的能力与调节体温的能力。真皮由成纤维细胞与其细胞外基质构成,其中包括神经、血管、淋巴、肌肉与皮脂腺、大汗腺、小汗腺等皮肤附属器官。作为构建真皮结构的主要构成要素成纤维细胞,其合成胶原蛋白、蛋白多糖及其他结构的糖蛋白质等细胞外基质。其中,胶原蛋白,作为占真皮的70%的纤维蛋白质,在支撑皮肤的机械强度(弹力)、结缔组织的粘合、细胞粘连、以及诱导细胞分裂与分化等起到作用(Van der M.Rest et al.,Biometerials.,11:28-31,1990;Shimokomaki M.et al.,Ann.N.Y.Acad.Sci.,580:1-7,1990;Van der Rest M.et al.,Biochimie.,72(6-7):473-484,1990)。胶原蛋白随着年龄的增加及紫外线照射的光老化而减少,因而造成胶原蛋白含量减少,皮肤的厚度变薄,进而导致皮肤的皱纹的形成(Artheu K.Balin et al.,″Aging and the skin″,1998)。相反,如果通过刺激皮肤上的胶原蛋白合成,使胶原蛋白代谢活跃,则会显现出真皮基质的成分增加,从而导致例如皱纹形成减少、皮肤强度增加及皮肤强化等效果。
人类的皮肤,随着年龄的增加,由于各种内外原因,其功能降低。由皮肤细胞的代谢过程或紫外线而生成的活性氧等氧化细胞组分等(例:细胞膜中的脂质等),从而阻碍了细胞组分的活性及生物合成。其结果是,产生皮肤老化。在皮肤老化进行的同时,皮肤上的表皮的厚度通常变薄,细胞与组织发生脱水现象,从而细胞和组织变得干燥,细小皱纹增加,且皱纹逐渐加深。进一步地,占真皮的大部分的胶原蛋白的量减少,合成胶原蛋白的成纤维细胞的活性也减少,新的胶原蛋白的合成也减少。随着真皮内胶原蛋白的量的减少,真皮的厚度也减少,其结果是,皮肤上产生皱纹,且降低了皮肤的平滑和强度。因此,为了有效地抑制如上述的皮肤老化,必须使表皮细胞及真皮细胞两者都活化和/或再生。
另一方面,AIMP1(ARS-交互多功能蛋白质,ARS-interactingmulti-functional protein 1)蛋白质,是以往的作为p43蛋白质已知的,由本发明人等进行重新命名的蛋白质(Kim S.H.et al.,Trends in BiochemicalSciences.,30:569-574,2005)。AIMP1,是由312个氨基酸构成的蛋白质,其结合多重-tRNA合成酶复合体(multi-tRNA synthetase complex),增进上述多重-tRNA合成酶的催化活性(catalytic activity)。在体外,上述AIMP1包括在脑脊髓炎、神经炎及葡萄膜炎的自身免疫疾病部位上的微神经元(microneuron)中被高水平地表达。AIMP1在特定发育阶段及组织中高水平地表达的现象,表明AIMP1与炎症反应及细胞凋亡有关系(Berger,A.C.et al.,J.Immunother.23:519-527,2000)。本发明人等,探明了AIMP1及其N-末端片段可有效地作为细胞因子、抗癌剂及血管发生抑制剂来利用(Park H.et al.,J.Leukoc.Biol.,71:223-230,2002;Park S.G.et al.,J.Biol.Chem.,277:45243-45248,2002;Park H.et al.,Cytokine,21:148-53,2002)。但是,AIMP1与胶原蛋白合成及KGF表达之间的关系,至今还没有查明。
发明内容
因此,本发明的目的,是提供刺激在体外或体内刺激胶原蛋白合成和/或KGF(角质形成细胞生长因子)表达的方法、治疗皮肤老化的方法、治疗皮肤松弛和/或皱纹的方法、增进皮肤平滑和/或强度的方法、治疗绝经对皮肤的不良效果的方法、及治疗绝经对胶原蛋白的不良效果的方法,上述方法均包括将有效量的具有序列号1的氨基酸序列的多肽或与其具有相同的生理活性的片段施用至所需受试者。
进一步地,本发明的其它的目的是提供用于刺激胶原蛋白合成和/或KGF表达的组合物、及用于防止皮肤老化的化妆品组合物,上述组合物均含有作为有效成分的、且具有序列号1的氨基酸序列的多肽或与其具有相同的生理活性的片段。
同时,本发明的另外的目的是提供了具有序列号1表示的氨基酸序列的多肽或与其具有相同生理活性的片段在用于刺激胶原蛋白合成和/或KGF表达的组合物、及用于制备防止皮肤老化的化妆品组合物方面的用途,上述组合物均以所述的多肽和其片段作为活性成分。
为了达到上述目的,本发明提供在体外或体内刺激胶原蛋白合成和/或KGF(角质形成细胞生长因子)表达的方法、治疗皮肤老化的方法、治疗皮肤松弛和/或皱纹的方法、增进皮肤平滑和/或强度的方法、治疗绝经对皮肤的不良效果的方法、及治疗绝经对胶原蛋白的不良效果的方法,上述方法均包括将有效量的选自由下述(a)、(b)及(c)所组成的组的任意一种施用于所需受试者,
(a)具有序列号1表示的氨基酸序列的分离的多肽;
(b)具有与上述(a)多肽有至少70%同源性的氨基酸序列的分离的多肽;及
(c)上述(a)或(b)多肽的片段。
进一步地,本发明还提供用于刺激胶原蛋白合成和/或KGF表达的组合物、及用于防止皮肤老化的化妆品组合物,上述组合物均以具有序列号1表示的氨基酸序列的多肽或与其具有相同的生理活性的片段作为活性成分。
进一步地,本发明还提供具有序列号1表示的氨基酸序列的多肽或与其具有相同生理活性的片段在用于制备刺激胶原蛋白合成和/或KGF表达的组合物中的用途、及用于防止皮肤老化的化妆品组合物中的用途,该组合物以所述多肽或其片段作为活性成分。
以下详细地说明本发明。
本发明具有以下特征,提供与胶原蛋白合成及KGF(角质形成细胞生长因子)表达有关的AIMP1的新用途。
AIMP1蛋白质是由312个氨基酸构成的蛋白质,是由包括前列腺癌细胞、免疫细胞及转染细胞的多种类型细胞分泌的,已知该分泌的AIMP1作用于诸如单核细胞,巨噬细胞、内皮细胞及成纤维细胞等的多种靶细胞。已知上述AIMP1的下述3个SNP(参照NCBI的SNP数据库):全长AIMP1蛋白质的氨基酸序列(序列号1)中的第79号丙氨酸(Ala)被脯氨酸(Pro)取代(序列号24、SNP注册号:rs3133166)、第104号苏氨酸(Thr)被丙氨酸(Ala)取代(序列号25、SNP注册号:rs17036670)、第117号苏氨酸被丙氨酸(序列号26、SNP注册号:rs2230255)取代。
因为上述AIMP1蛋白质分别对于多种不同的靶细胞,具有多种复杂的活性,本发明人等作出了如下假设,即AIMP1将使用不同结构的基序或结构域来实现其不同活性。为了确认上述的可能性,本发明的发明人通过下述途径确定了用于刺激胶原蛋白合成及KGF(角质形成细胞生长因子)表达的AIMP1蛋白质的功能结构域,即通过构建AIMP1的N末端片段及C末端片段,并确定各片段等对于胶原蛋白合成及KGF(角质形成细胞生长因子)表达的活性(参见实施例5)。其结果是,AIMP1蛋白质的C末端片段对于刺激胶原蛋白合成及KGF表达方面没有活性,其N末端片段则显示出了活性(参见图5)。
接下来,为了确认在AIMP1蛋白质的N末端片段上的哪个域具有上述的活性,本发明人等构建了缺失片段,并利用该缺失片段进行试验。结果是,确认AIMP1的6-46位氨基酸区域,具有刺激胶原蛋白合成及KGF表达的活性(参照图6及图7)。根据上述的试验结果,含有AIMP1蛋白质的6-46位氨基酸区域和至少由192个氨基酸构成的片段,即选自序列号2、序列号12、序列号13及序列号14的多肽具有刺激胶原蛋白合成及KGF表达的活性。进一步地,AIMP1蛋白质的1-192位氨基酸构成的片段(序列号27)及AIMP1蛋白质的6-192位氨基酸构成的片段(序列号18)也具有刺激胶原蛋白合成及KGF表达的活性(结果未图示)。
胶原蛋白,是在真皮的成纤维细胞中产生的并形成70%的真皮部分的纤维状蛋白质,对皮肤的平滑与强度起作用。因此,当胶原蛋白的合成减少,皮肤则会发生老化,所以皮肤的平滑度与强度急剧减少,其结果是皮肤产生松弛或皱纹。反过来,如果通过刺激皮肤内的胶原蛋白合成,使胶原蛋白代谢活跃,则真皮基质中的该成分增加,从而导致诸如改善皱纹、强度增进、皮肤强化等的效果。如上所述,本发明的AIMP1蛋白质及其片段,具有刺激胶原蛋白合成的活性(参见图1至图3及图5至图7)。
KGF(角质形成细胞生长因子),是构成皮肤表皮的主成分的角质形成细胞的成长因子,是由真皮成纤维细胞产生的旁分泌因子。KGF,是与上皮细胞的再生与重建有关的因子,其诱导上皮细胞生长,使皮肤的表皮层再生(Moore T.et al,Lab.Invest.60:237-244,1989;Han DS et al.,Am.J.Physiol.Gastrointest.Liver Physiol.279:G1011-1022,2000)。进一步地,也已知KGF为成纤维细胞生长因子家族的一员,即成纤维细胞生长因子7(fibroblastgrowth factor7),并且已知导入编码KGF的基因的转基因大鼠具有增厚的表皮(Guo L.et al.,EMBO J.,12(3):973-986,1993)。现在,KGF已在国际化妆品注册协会(Cosmetic Toiletry and Fragrance Association)作为化妆品原料物质进行了注册。进一步地,已知KGF与胶原蛋白互相结合,刺激角质形成细胞的细胞分裂(Ruehl M.et al.,J Biol Chem.,277:26872-8,2002)。如上所述,本发明的AIMP1蛋白质及其片段,具有刺激KGF的表达的活性(参见图4、图5及图6)。
具有如上所述活性的AIMP1蛋白质及其片段,可用于刺激皮肤的胶原蛋白合成和/或KGF表达。而且,通过刺激胶原蛋白合成和/或KGF表达,AIMP1蛋白质及其片段能够用于抑制、治疗、改善和/或预防皮肤老化或由其引起的皮肤症状(cutaneous signs)。
因此,本发明提供刺激胶原蛋白合成和/或KGF(角质形成细胞生长因子)表达的方法、治疗皮肤老化的方法、治疗皮肤松弛和/或皱纹的方法、增进皮肤平滑和/或强度的方法,其包括将有效量AIMP1蛋白质或其片段施用于所需受试者。
另一方面,绝经期中,真皮的重要变化是胶原蛋白水平减少,真皮厚度减少。与绝经以后胶原蛋白代谢的变化有关的研究,已报道观察到骨密度减少以及皮肤厚度的减少,进一步地,观察到毎年胶原蛋白水平在平行地减少(C.Castelo-Braance et al.,Maturitas.,18(3):199-206,1994)。即,绝经后皮肤胶原蛋白的减少,在绝经最初年度中减少程度最急剧,从绝经开始至绝经后20年,皮肤胶原蛋白毎年减少2.1%,绝经后5年内皮肤胶原蛋白减少30%左右。因此,据报道,随着皮肤厚度的急剧减少以及皮脂的减少,加速出现了诸如皮肤变粗,皱纹增加,皮肤的弹力急剧地减少等多种皮肤老化的现象。进一步地,已表明,绝经后女性的尿失禁频率增加的主要的原因是膀胱壁的胶原蛋白减少引起的强度的降低(Bergman A et al.,Gynecol Obstet Invest.,37(1):48-51,1994)。因此,可利用有刺激胶原蛋白合成及KGF表达活性的本发明的AIMP1蛋白质及其片段抑制、治疗、改善和/或预防绝经后的皮肤老化、绝经后的胶原蛋白代谢异常引起的异常病症或症状等。因此,本发明,提供治疗绝经对皮肤的不良效果及绝经对胶原蛋白的不良效果的方法,所述方法均包括将有效量的AIMP1蛋白质及其片段施用于所需受试者。
本发明的方法使用的AIMP1蛋白质,可具有序列号1表示的氨基酸序列。进一步地,本发明的AIMP1蛋白质也包括其功能等同物。本发明中的“功能等同物”,是指与具有序列号1表示的氨基酸序列的AIMP1具有基本上相同的生理活性的多肽。
上述“基本上相同的生理活性”,是指刺激胶原蛋白的合成和/或KGF表达的活性。上述功能等同物也可以是与序列号1表示的氨基酸序列具有至少70%、优选至少80%、更优选至少90%的序列同源性的多肽。优选地,上述功能等同物可以是序列号24、序列号25及序列号26的多肽,已知它们是AIMP1的SNP。更具体地,上述“功能等同物”,是指包含由氨基酸添加、取代或缺失而产生的与序列号1的多肽具有至少70%、优选为至少90%、更优选为至少95%的序列同源性(即,同一性)的序列、且与序列号1的多肽具有基本上相同生理活性的多肽,例如,与序列号1具有70%、71%、72%、73%、74%、75%、76%、77%、78%、79%、80%、81%、82%、83%、84%、85%、86%、87%、88%、89%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、100%的序列同源性。本文中的序列同源性或同一性被定义为,在为达到最大的同一性百分比而对序列进行比对和引入空位(gap)(如有必要)后,以及在不将保守性取代作为序列同一性的一部分的情况下,候选序列中与序列号1的氨基酸序列完全相同的氨基酸残基的百分比。对序列号1的氨基酸序列的N末端伸长、C末端伸长或内部伸长、缺失或插入等构建均不能影响同一性或同源性。进一步地,可使用通常使用的用于比较2条多肽的氨基酸位置的相似性的标准方法来确定上述序列的同一性。使用BLAST或FASTA等的计算机程序,将2条多肽氨基酸进行比对,以最优化配对(沿着1条或2条序列的全长序列,或沿着1条或2条序列的预定部分)。上述程序,提供默认的开放罚分(default opening penalty)及缺省空位罚分(default gap penalty),提供可与计算机程序结合在一起使用的PAM250(标准记分矩阵;Dayhoff et al.,in Atlas of Protein Sequence and Structure,vol 5,supp.3,1978)等的记分矩阵。例如,百分比的同一性可通过如下所述进行计算:将完全一致的序列(indentical matches)的总数乘上100,然后除以匹配范围(machted span)内的较长的序列的长度数目以及为了排列两条序列而引入到较长序列内的空位数目的总和。本发明的功能等同物的范围中,也包括维持本发明的多肽的基本骨架及生理活性的同时,通过修改本发明多肽的一部化学结构的衍生物。例如,包括为了改变本发明的肽的稳定性、贮藏性、挥发性或溶解度等的结构修饰。
进一步地,本发明也可利用AIMP1的片段。上述AIMP1的片段,可以是具有序列号1表示的氨基酸序列的多肽的部分片段或与序列号1表示的氨基酸序列具有至少70%的序列同源性的多肽的部分片段。进一步地,上述片段与AIMP1氨基酸序列中的一部分具有100%的同一性,同时显示出与本发明的AIMP1基本相同的生理活性。上述片段,可由选自序列号1的氨基酸序列或与序列号1的氨基酸序列具有至少70%的同源性的氨基酸序列中的至少10个以上,优选为41个,更优选为100个以上连续氨基酸构成。本发明的片段,优选也可以为包括选自AIMP1的氨基酸序列(序列号1)中的41个邻接氨基酸序列(序列号12),且至少由192个氨基酸构成的多肽。更优选地,本发明的片段是由选自由序列号2、序列号12、序列号13、序列号14及序列号27至序列号34构成的组中的任何一个氨基酸序列构成的多肽。上述序列号29至31的氨基酸序列,是具有序列号27氨基酸序列的多肽的SNP,而且上述序列号32至34的氨基酸序列是由序列号28的氨基酸序列组成的多肽的SNP。
本发明的多肽可通过基因工程的方法构建。首先,通过通常的方法制作编码上述AIMP1蛋白质或其片段的DNA序列。DNA序列,可通过利用适宜的引物进行PCR扩增而制作。也可通过其它的本领域公知的标准方法,例如,利用自动DNA合成仪(Biosearch或Applied Biosystems公司销售的产品)合成DNA序列。构建好的DNA序列,插入至含有1个或1个以上的表达控制序列(expression control sequence)的载体上,该表达控制序列可操作地连接在该DNA序列上,用由此在形成的重组表达载体转化宿主细胞。在适宜的培养基及条件下培养生成的转化体,从上述培养物回收DNA序列编码的基本上纯的多肽。上述回收,利用本领域上公知的方法(例如,色谱法)而进行。上述的“基本上纯的多肽”,是指本发明的多肽,基本上不包含从宿主细胞由来的任何其它蛋白质。本发明的多肽合成的基因工程的方法,可参考下述文献(Maniatis et al.,Molecular Cloning;A laboratory Manual,ColdSpring Harbor laboratory,1982;Sambrook et al.,supra;Gene ExpressionTechnology,Method in Enzymology,Genetics and Molecular Biology,Methodin Enzymology,Guthrie & Fink(eds.),Academic Press,San Diego,Calif,1991;Hitzeman et al.,J. Biol.Chem.,255:12073-12080,1990.)。
进一步地,本发明的多肽,可根据本领域公知的技术进行化学合成(Creighton,Proteins:Structures and Molecular Principles,W.H.Freeman andCo.,NY(1983))。即,本发明的多肽,可利用通常的分步液相或固相合成、片段缩合、F-MOC或T-BOC化学方法来制造(Chemical Approaches to theSynthesis of Peptides and Proteins,Williams et al.,Eds.,CRC Press,Boca RatonFlorida,(1997);A Practical Approach,Atherton & Sheppard,Eds.,IRL Press,Oxford,England,(1989))。
特别地,优选的制造方法,是利用固相合成法。本发明的多肽,可通过采用通常的固相法,使受保护的氨基酸之间进行缩合反应,其中,根据已经确定的序列从C末端开始,并依次连接第一个氨基酸、第二个氨基酸和第三个氨基酸等等。缩合反应后,可通过酸解或氨解等公知方法除去保护基及C末端氨基酸连结的载体。上述提及的多肽合成法,在相关书籍中有详细的描述(Gross and Meienhofer′s,The Peptides,vol 2.,Academic Press,1980)。
本发明的多肽合成中使用的固相载体,可以是该领域通常使用的载体,例如,可使用取代的苄型聚苯乙烯树脂(polystyrene resins of substituted benzyltype)、羟基甲基苯乙酰胺型(hydroxymethylphenylacetic amide form)的聚苯乙烯树脂、取代的苯基聚苯乙烯树脂和聚丙烯酰胺树脂,它们具有可与多肽结合的功能基团。氨基酸缩合可利用通常的方法,例如,二环己基碳二亚胺(DDC)方法、酸酐方法及活化酯方法。
本发明的多肽合成中使用的保护基等,可用通常的多肽合成中使用的保护基等,包括那些通过与酸解、还原或氨解等通常方法容易除去的基团。作为氨保护基的具体例子有甲酰基;三氟乙酰基;苄氧羰基、(邻或对)氯苯氧基羰基及(邻或对)溴苄氧基羰基等的取代的苄氧基羰基;t-丁氧基羰基及t-氨氧基羰基等的脂肪族氧基羰基等。氨基酸的羰基等,可转化成酯基来保护。酯基中,有苄基酯;甲氧基苄基酯等的取代的苄基酯;环己基酯、环庚基酯或t-丁基酯等的烷基酯等。胍基部分(尽管不需要对其进行保护)可通过硝基,或通过甲苯磺酰基、甲氧基苯磺酰基或均三甲基苯磺酰基等的芳基磺酰基保护。作为咪唑的保护基,有甲苯磺酰、苄基及二硝基苯基等。色氨酸的吲哚基,可用甲酰基等保护,或不用保护。
脱保护及从载体分离保护基团,可在各种净化剂(scavenger)的存在下,通过无水氢氟化物来进行。作为净化剂的例子,可举出苯甲醚、 (邻、间、对)甲酚、二甲基硫化物、甲苯硫酚、ethanendiol及巯基吡啶等的多肽合成中通常使用的物质。
对通过基因工程的方法制造的重组多肽或化学合成的多肽可通过下述方法进行分离及精制,例如,可通过提取法、重结晶法、多种色谱(凝胶过滤法、离子交换、沉淀、吸附、反相)、电泳、逆流分配法等本领域公知的方法,。
本发明的“有效量”,是指能够表现出选自下述效果的量,即刺激体外或体内胶原蛋白合成和/或KGF表达、治疗皮肤老化、治疗皮肤松弛和/或皱纹、增进皮肤的平滑性和/或强度、治疗绝经对皮肤的不良效果、及治疗绝经对胶原蛋白的不良效果。
本发明中,“受试者(subject)”,是指哺乳动物,特别是指包括人类在内的动物或上述动物的皮肤细胞、皮肤组织。上述受试者也可以是需要治疗的患者(patient)。进一步地,上述皮肤细胞,优选也可以是成纤维细胞。
可施用本发明的AIMP1蛋白质及其片段直至达到上述记载的效果中希望的效果。本发明的AIMP1蛋白质及其片段,可通过本领域公知的方法经由多种途径施用。即,口服或胃肠外途径,例如,可在口腔、肌肉内、静脉内、皮内、动脉内、骨髓内、鞘内、腹腔内、鼻内、阴道内、直肠内、舌下或皮下施用或施用至胃肠道、粘膜或呼吸器管。例如,可通过将本发明的多肽直接涂布在皮肤上的方法,或通过将上述多肽制造成注射用剂型,用30gauge的细注射针,在皮肤下层注入一定量或用注射针刺皮肤的方法施用。优选直接涂布在皮肤上的方法。进一步地,本发明的AIMP1蛋白质及其片段以与对靶细胞或组织(例:皮肤细胞或皮肤组织)诱发高亲和性结合的分子结合的形式,或以被包囊在某一分子内的形式施用。本发明的AIMP1蛋白质及其片段,可利用本领域公知的技术与固醇(例:胆固醇)、脂质(例:阳离子脂质、病毒体或脂质体)或靶细胞特异性结合剂(例:靶细胞特异性受体识别的配合基)结合。作为适宜的偶联剂或交联剂,例如,可包含蛋白质A、碳二亚胺、N-琥珀酰亚胺-3-(2-嘧啶二硫)丙酸酯(SPDP)等。
为了达成上述记载的用途等,将本发明的AIMP1蛋白质或其片段用于药学的、皮肤病学的或化妆品类组合物的活性成分。
上述组合物,可制造成本领域中制造的任何的剂型。优选可制成皮肤涂布用剂型。例如,可制造成溶液、混悬剂、乳剂、糊剂、凝胶、乳膏、洗剂、粉剂、肥皂、含有表面活性剂的清洁剂、油、粉末粉底、乳剂粉底、蜡粉底及喷雾剂等,但并不限于此。更优选的是,可制成软化洗剂、营养洗剂、营养乳膏、按摩乳膏、精华素、眼霜、清洁剂霜、清洁剂泡沫、清洁水、pack、喷雾剂或粉剂的形式。
本发明的组合物的剂型是糊剂、乳膏或凝胶时,载体成分可使用动物油、植物油、蜡、石蜡、淀粉、黄芪胶、纤维素衍生物、聚乙二醇、硅酮、膨润土、二氧化硅、滑石或氧化锌等。进一步地,本发明的组合物的剂型是粉剂或喷雾剂时,载体成分可使用乳糖、滑石、二氧化硅、氢氧化铝、硅酸钙或聚酰胺粉末,特别是,喷雾剂时,也可追加含有氟氯烃、丙烷/丁烷或二甲基醚等的推进剂。本发明的组合物的剂型是混悬剂时,载体成分可使用水、乙醇或丙二醇等的液体稀释剂,乙氧基化异十八烷醇、聚氧乙烯山梨醇酯及聚氧乙烯山梨醇酐酯等的悬浊剂,微晶型纤维素,偏氢氧化铝,膨润土,琼脂或黄芪胶等。进一步地,本发明的组合物的剂型是含有表面活性剂的清洁剂时,载体成分可使用脂肪醇磺酸盐/酯、脂肪醇醚磺酸盐/酯、硫代琥珀酸单酯、羟乙基磺酸盐/酯、咪唑啉衍生物、脂肪酸酰胺醚硫酸盐/酯(fatty acidamide ether sulfate)、烷基氨基甜菜碱、脂肪醇、脂肪酸甘油酯、脂肪酸二乙醇胺、植物油、羊毛脂衍生物或乙氧基化甘油脂肪酸酯等。
进一步地,本发明的组合物,可制造为美速疗法(mesotherapy)的注射用剂型。上述注射用剂型,可使用适宜的分散剂或湿润剂及悬浊化剂,通过本领域公知的技术而进行制造。例如,可在盐水或缓冲液中溶解本发明的多肽,作为用于注射的剂型。
进一步地,本发明的组合物,也可制造为口服施用的剂型。口服施用时,本发明的多肽,可与赋形剂混合以剂型化为口服片剂、含服片剂、含片(troche)、胶囊、酏剂、混悬剂、糖浆及薄片等形式。这些剂型,在活性成分以外还含有稀释剂(例:乳糖、葡萄糖、蔗糖、甘露醇、山梨醇、纤维素和/或甘氨酸)与滑润剂(例:二氧化硅、滑石、硬脂酸及其镁盐或钙盐和/或聚乙二醇)。上述片剂,可含有硅酸铝镁、淀粉膏、明胶、黄芪胶、甲基纤维素、羧甲基纤维素钠和/或聚乙烯基吡咯烷酮等的粘合剂,根据情形,可另外含有淀粉、琼脂、海藻酸或其钠盐等的崩解剂、吸收剂、着色剂、香味剂和/或甜味剂。上述剂型,可通过通常的混合、制粒或包衣的方法制造。
为了进一步强化本发明的效果,本发明的组合物可另外含有具有刺激胶原蛋白合成、刺激成纤维细胞增殖、抑制/改善皮肤老化、皮肤保湿、增进皮肤的强度/柔软、改善皱纹、强化皮肤功能等的效果的公知物质。上述物质,可使用类视黄酸(retinoic acid)、TGF(trans-forming growth factor)、白桦脂酸(betulinic acid)、桂皮酸(cinnamic acids)、氢芪(hydrostilbene)、维生素A、维生素E、维生素C、红葡萄提取粉末等,但并不限于此。此外,也可含有药学上、皮肤病学上和/或化妆品学上可接受的介质或基材,例如促进蛋白质进入皮肤的物质、防腐剂、保湿剂、乳化剂、缓冲剂等。
本发明的AIMP1蛋白质或其片段的施用量,相对于组合物总重量可以为0.001%至20%、优选为0.005%至10%。
本发明的组合物中,本发明的多肽的总有效量,可以通过单剂量施用于受试者,也可通过多次治疗方法(fractionated treatment protocol)来施用,其中在多次治疗方法中,是在较长时间内施用多剂量。含有本发明的多肽的组合物的施用量可随着该组合物的用途而不同,然而活性成分的通常有效施用量可为0.1μg至10mg,且可每日施用数次。但是,上述多肽的浓度的变化可依据多种因素,例如不仅考虑施用的途径及施用次数,还要考虑所需受试者的年龄、体重、健康状态、性别、疾病的程度、饮食状况及排泄等多种因素。因此,考虑上述方面时,本领域技术人员可确定本发明的多肽在用于上述记载的特定的用途时的有效量。本发明的组合物只要能呈现本发明的效果,其剂型、施用途径及施用方式没有特别的限制。

但是,下述实施例仅用于示例本发明,而不是将本发明的内容限制于此。
参考例1
AIMP1及片段的构建
通过本领域公知的Park等的方法(Park S.G.et al.,J. Biol.Chem.,277:45243-45248,2002),制备由312个氨基酸构成的AIMP1蛋白质(序列号1)、其N-末端片段(1-147;序列号2)及C-末端片段(148-312;序列号3)。
参考例2
AIMP1的N-末端缺失片段及C-末端缺失片段的构建
分别制备AIMP1的N-末端缺失片段和C-末端缺失片段,即,AIMP1-(6-46;序列号12)、AIMP1-(1-46;序列号13)、AIMP1-(1-53;序列号14)、AIMP1-(193-312;序列号15)、AIMP1-(1-192;序列号27)及AIMP1-(6-192;序列号28)片段。上述片段均以AIMP1的cDNA作为模板,利用各片段的特异性引物组(表1)对上述各片段进行PCR扩增合成。PCR反应条件,如下所述:在95℃下加热2分钟,使模板DNA预变性;然后进行25个如下循环:95℃下30秒;56℃下30秒;及72℃下1分钟;最后在72℃下进行5分钟延伸反应。用EcoRI及XhoI酶切上述PCR产物,并连接到用相同的酶酶切的pGEX4T3载体(Amersham Biosciences)上。用上述载体转化E.coli BL21(DE3),进行培养,诱导多肽的表达。各多肽被表达成GST-tag融合蛋白,并通过GSH琼脂糖凝胶进行提纯。为了除去脂多糖,用无热原的缓冲液(10mM磷酸钾缓冲液、pH6.0,100mM氯化钠)透析蛋白质溶液。透析后,将蛋白质溶液装载到预先用相同的缓冲液平衡的多粘菌素树脂(polymyxin resin)(Bio-Red)上,培养20分钟后进行洗脱,由此制备AIMP1的各缺失片段。

实施例1
不同AIMP1处理浓度对胶原蛋白的合成的刺激
<1-1>包皮成纤维细胞的培养及AIMP1处理
为了测定AIMP1引起的胶原蛋白转录RNA,将包皮成纤维细胞(5×104细胞/孔,公司名称:MTT、保藏编号:MC1232)在6孔板上含有10%血清的DMEM培养基上培养12小时后,在无血清培养基中培养约3小时。然后,用不同浓度的AIMP1蛋白质(序列号1)(0、20、50、100及200nM)分别处理该培养的细胞达6小时(RT-PCR)或12小时(免疫印迹法)。
<1-2>RT-PCR分析
回收上述实施例<1-1>中的由不同浓度的AIMP1处理的细胞后,用TRIzol(invitrogen)溶解。往细胞溶解液中添加10wt%的氯仿进行充分混合。将混合液在12,000g下离心分离15分钟,取上清液。往取得的上清液中添加乙醇,使最终浓度为70%。接下来,对上清液在26,000g下进行离心分离5分钟,取得沉淀物。干燥上述沉淀物,用灭菌的蒸馏水进行溶解,提取总RNA。利用3μg的总RNA制造cDNA。接下来,利用胶原蛋白特异性引物(序列号4及序列号5)进行PCR。此时,利用GADPH特异性引物(序列号6及序列号7)对cDNA进行PCR,以用作对照。PCR反应条件如下所述:在95℃下加热5分钟,使模板DNA预变性;然后进行25个如下循环:95℃下1分钟;52℃下1分钟;及72℃下1分钟;最后在72℃下延伸反应5分钟。其结果是,如图1的上部分(upper)所示,能够确认,AIMP1剂量依赖性地刺激胶原蛋白在成纤维细胞中的合成。
<1-3>免疫印迹分析
收集上述实施例<1-1>中的由不同浓度的AIMP1处理的细胞,在细胞溶解缓冲液(lysis buffer)(50mM HEPES,pH7.5,150mM NaCl,10%丙三醇,5mM EGTA,1mM原钒酸钠,10mM NaF,12mM b-甘油磷酸酯,1mM DTT,1mM PMSF,5mg/ml抑肽酶,and 1%NP40)中溶解细胞。将溶解的细胞在26,000g下离心分离15分钟,得到细胞提取物。用8%SDS-PAGE分离细胞提取物30μg,并利用抗-胶原蛋白I抗体(Abcam),通过本领域公知的通常的方法进行免疫印迹。利用抗微管蛋白抗体(Sigma)作为对照。
其结果是,图1的下部分(bottom)所示,能够从蛋白质水平确认,AIMP1剂量依赖存性地刺激胶原蛋白在成纤维细胞中的合成。
实施例2
不同时间间隔的AIMP1处理对胶原蛋白合成的刺激
<2-1>包皮成纤维细胞的培养及AIMP1处理
将包皮成纤维细胞(5×104细胞/孔)在6孔板上含有10%血清的DMEM培养基上培养12小时后,在不含血清培养基上培养3小时左右。然后,在不同时间间隔(0、2、4、6、12及24h),用100nM的AIMP1处理该培养的细胞。
<2-2>RT-PCR分析
在指定的时间回收上述实施例<2-1>的经AIMP1处理的细胞,通过与上述实施例<1-2>相同的方式进行RT-PCR。其结果是,如图2的上部分(upper)所示,能够确认,AIMP1时间依赖性地刺激成纤维细胞中的胶原蛋白的合成。
<2-3>免疫印迹分析
在指定的时间回收上述实施例<2-1>的AIMP1处理的细胞,通过与上述实施例<1-3>相同的方法进行免疫印迹分析。其结果是,如图2的下部分(bottom)所示,能够在蛋白质水平上确认,AIMP1刺激成纤维细胞中胶原蛋白的合成。
实施例3
体内AIMP1的胶原蛋白合成的刺激
通过本领域公知的基因陷阱方法(Zambrowicz,B.P.,et al.,Nature392:608-611,1998;Kim JY et al.,Proc.Natl. Acad.Sci.USA 99:7912-7916,2002),诱使雄性C57BL6小鼠中的AIMP1结构基因突变,制造AIMP1缺失的纯合小鼠(AIMP1-/-小鼠、Ho)(Park S.G.et al.,Am.J. Pathol.,in press:,2005)。然后,利用剪刀在存活8周的纯合小鼠与野生型小鼠(AIMP1+/+小鼠、WT)的背上,设立直径为0.5cm的圆形创口。伤口部位没有被敷裹。将4μg上述参考例1中制备的AIMP1蛋白质,溶解到5μl含有20%丙三醇的PBS中,并将该溶液涂布在伤口部位。每日进行两次涂布,直至受伤后3天。对照(没有进行AIMP1蛋白质处理)用含有20%丙三醇PBS进行处理。接下来,利用剪刀切除受伤部位的组织,用4%多聚甲醛固定过夜。用PBS清洗固定的组织,在30%的蔗糖液中培养4小时后,最后与OCT(最佳剪切温度(optimal cutting temperature))化合物混合,冷冻至-70℃。
将6μm的冻结切片附着在涂敷硅烷的载片上,用PBS清洗。接下来,用包含0.1%tween20与1%脱脂乳的PBS封闭载片,在37℃下与抗-胶原蛋白I抗体(Abcam)反应2小时。用包含0.1%tween20的PBS清洗3次上述载片后,添加连结FITC的二次抗体,并在37℃下温育1小时。在萤光显微镜(confocal immunofluorescence microscopy,μ-Radiance,BioRad)下观察上述切片。
如图3所示,其结果是,与AIMP1缺失的纯合小鼠(Ho)相比,能够观察到野生型小鼠(WT)中有更多量的胶原蛋白,而且通过进行AIMP1的外部处理,胶原蛋白能够进一步增加。由此,能够确认AIMP1在体内也能刺激胶原蛋白的合成。
实施例4
AIMP1对KGF表达的刺激
从经100nM的AIMP1处理2小时的包皮成纤维细胞中,通过与上述实施例<1-2>中记载的相同方法,提取总RNA。接下来,利用KGF特异性引物(序列号8及9),对提取物进行RT-PCR。分别利用细胞周期蛋白(cyclin)D特异性引物(序列号10及11)和GADPH特异性引物(序列号6及7)对提取物分别进行扩增以作为对照。PCR反应条件,如下所述:以95℃下5分钟,从而使模板DNA预变性;进行25个如下循环:95℃下1分钟,52℃下1分钟,72℃下1分钟;以及在72℃下最终延伸5分钟。
其结果是,如图4所示,由于AIMP1处理,包皮成纤维细胞中KGF的表达得到增加。由此表明,AIMP1是通过增加KGF的表达而参与皮肤的表皮层的再生。
实施例5
AIMP1片段对胶原蛋白合成及KGF表达的活性将包皮成纤维细胞(5×104细胞/孔)在6孔板含有10%血清的DMEM培养基上培养12小时,接下来在不含血清的培养基上培养3小时左右。然后,用100nM上述参考例1中制备的AIMP1的N-末端及C-末端片段在不同时间间隔下(0、1、2、4及6h)处理上述培养的细胞。接下来,利用胶原蛋白特异性引物(序列号4及5)、KGF特异性引物(序列号8及9)及GADPH特异性引物(序列号6及7),通过与上述实施例<1-2>相同的方法,进行RT-PCR。
其结果是,如图5所示,显现出了AIMP1的N-末端片段刺激胶原蛋白的合成及KGF的表达。但是,C-末端片段没有出现有意义的效果。
实施例6
AIMP1的N-末端缺失片段及C-末端缺失片段对胶原蛋白合成及KGF
表达的刺激
将包皮成纤维细胞(5×104细胞/孔)在6孔板含有10%血清的DMEM培养基上培养12小时,进一步在不含血清的培养基上培养3小时左右。接下来,分别用100nM浓度的上述参考例2中制备的AIMP1的N-末端片段(序列号12、序列号13及序列号14)及AIMP1的C-末端片段(序列号15)分别进行2小时及6小时的处理。利用胶原蛋白特异性引物(序列号4及5)、KGF特异性引物(序列号8及9)、及GADPH特异性引物(序列号6及7),通过与上述实施例<1-2>相同的方法,对上述处理的细胞进行RT-PCR。
其结果是,如图6所示,能够确认,用AIMP1的N-末端缺失片段6-46(序列号12)及含有该片段的多肽(序列号13、序列号14)处理比用AIMP1的C-末端片段193-312(序列号15)处理更能刺激胶原蛋白的合成及KGF的表达。
实施例7
AIMP1及其N-末端片段对胶原蛋白合成的刺激
将包皮成纤维细胞(5×105细胞/孔)在6孔板含有10%血清的DMEM培养基上培养48小时,进一步在不含血清的培养基上培养3小时左右。接下来,用200nM浓度的AIMP1蛋白质(序列号1)与AIMP1N-末端片段等(序列号2及序列号12)处理该培养的细胞达12小时。使用没有进行上述处理的细胞作为对照。
在指定的时间回收上述经AIMP1及其N-末端片段处理的细胞与对照细胞,并通过与上述实施例<1-3>相同的方法进行免疫印迹分析。
其结果是,如图7所示,能够在蛋白质水平上确认,用AIMP1(序列号1)及AIMP1N-末端切片(序列号2及序列号12)处理的包皮成纤维细胞中的胶原蛋白的合成比没有通过AIMP1处理的包皮成纤维细胞中的胶原蛋白的合成得到更多的刺激。

如上所述,本发明的AIMP1蛋白质及其片段,能够刺激在皮肤中的胶原蛋白合成及KGF表达。因此,本发明的AIMP1蛋白质或其片段,可有效地用于所需受试者,以刺激胶原蛋白合成和/或KGF表达、治疗皮肤老化、治疗松弛和/或有皱纹的皮肤、增进皮肤的平滑度和/或强度、治疗绝经的不良皮肤效果、及治疗绝经对胶原蛋白的不良影响。

1.一种在体外或体内刺激胶原蛋白合成和/或KGF表达的方法,其包括将有效量的选自由下述(a)、(b)及(c)所组成的组中的任意一种施用于受试者:
(a)具有序列号1表示的氨基酸序列的分离的多肽;
(b)具有与上述(a)多肽有至少70%同源性的氨基酸序列的分离的多肽;及
(c)上述(a)或(b)多肽的片段。
2.一种治疗皮肤老化的方法,其包括将有效量的选自由下述(a)、(b)及(c)所组成的组中的任意一种施用于受试者:
(a)具有序列号1表示的氨基酸序列的分离的多肽;
(b)具有与上述(a)多肽有至少70%同源性的氨基酸序列的分离的多肽;及

(a)具有序列号1表示的氨基酸序列的分离的多肽;
(b)具有与上述(a)多肽有至少70%同源性的氨基酸序列的分离的多肽;及
(c)上述(a)或(b)多肽的片段。
7.根据权利要求1~6中任一项所述的方法,其特征在于,所述(b)的多肽选自由序列号24、序列号25和序列号26组成的组。
8.根据权利要求1~6中任一项所述的方法,其特征在于,所述(c)的片段包括序列号12表示的氨基酸序列,且是由至少192个氨基酸构成的肽。
9.根据权利要求8所述的方法,其特征在于,所述(c)的片段是由选自序列号2、序列号12、序列号13、序列号14及序列号27至序列号34的氨基酸序列构成的肽。
10.一种用于刺激胶原蛋白合成和/或KGF表达的组合物,该组合物包括选自由下述(a)、(b)及(c)组成的组中的一种作为活性成分:
(a)具有序列号1表示的氨基酸序列的分离的多肽;
(b)具有与上述(a)多肽有至少70%同源性的氨基酸序列的分离的多肽;及
(c)上述(a)或(b)多肽的片段。
11.一种防止皮肤老化的化妆品组合物,该组合物包括选自由下述(a)、(b)及(c)组成的组中的一种作为活性成分:
(a)具有序列号1表示的氨基酸序列的分离的多肽;
(b)具有与上述(a)多肽有至少70%同源性的氨基酸序列的分离的多肽;及
(c)上述(a)或(b)多肽的片段。
12.根据权利要求10或11所述的组合物,其特征在于,所述(b)的多肽选自由序列号24、序列号25和序列号26组成的组。
13.根据权利要求10或11所述的组合物,其特征在于,所述(c)片段包括序列号12表示的氨基酸序列,且是由至少192个氨基酸构成的多肽。
14.根据权利要求13所述的组合物,其特征在于,所述(c)的片段是由选自序列号2、序列号12、序列号13、序列号14及序列号27至序列号34的氨基酸序列构成的肽。
15.选自由下述(a)、(b)及(c)所组成的组中的任意一种在制备用于刺激胶原蛋自合成和/或KGF表达的组合物中的用途,该组合物包括下述(a)、(b)及(c)中的一种作为活性成分:
(a)具有序列号1表示的氨基酸序列的分离的多肽;
(b)具有与上述(a)多肽有至少70%同源性的氨基酸序列的分离的多肽;及
(c)上述(a)或(b)多肽的片段。
16.选自由下述(a)、(b)及(c)所组成的组中的任意一种在制备用于防止皮肤老化的化妆品组合物中的用途,该组合物包括下述(a)、(b)及(c)中的一种作为活性成分:
(a)具有序列号1表示的氨基酸序列的分离的多肽;
(b)具有与上述(a)多肽有至少70%同源性的氨基酸序列的分离的多肽;及
(c)上述(a)或(b)多肽的片段。
17.根据权利要求15或16所述的用途,其特征在于,(b)的多肽选自由序列号24、序列号25和序列号26组成的组。
18.根据权利要求15或16所述的用途,其特征在于,所述(c)的片段包括序列号12表示的氨基酸序列,且是由至少192个氨基酸构成的多肽。
19.根据权利要求18所述的用途,其特征在于,所述(c)的片段是由选自序列号2、序列号12、序列号13、序列号14及序列号27至序列号34的氨基酸序列构成的肽。
