Verfahren zur Herstellung von neuen Derivaten des Sulfanilamids
Die vorliegende Erfindung betrifft ein Verfahren zur Hestellung von neuen Derivaten des Sulfanilamids.

R1 eine Alkyl- oder Alkenylgruppe von höchstens 5 Kohlenstoffatomen oder eine Cycloalkyl- oder Cycloalkenylgruppe von höchstens 7 Kohlenstoffatomen und R. die Methyl- oder Äthylgruppe bedeutet, und ihre Additionssalze mit anorganischen oder organischen Säuren sind bisher nicht bekannt geworden.
Wie nun gefunden wurde, besitzen diese Verbindungen wertvolle pharmakologische Eigenschaften, insbesondere das l-Sulfanilyl-2-imino-3-butyl-5-äthyl-, das 1-Sul frnilyl-imino-3-tert.butyl-4-methyl- und das l-Sulfanilyl -2-imino-3-tert.butyl-5-methyl-imidazolidin weisen bei peroraler oder parenteraler Verabreichung hypoglykämische Wirkung auf, die sie als geeignet zur Behandlung der Zuckerkrankheit charakterisieren. Die hypoglykämische Wirkung wird an Standardversuchen an Warmblütern, z.B. an Kaninchen und Ratten, nachgewiesen.
In den Verbindungen der allgemeinen Formel I kann Ri als niedere Alkylgruppen z.B. die Methyl- Äthyl-, Propyl-, Isopropyl-, Butyl-, sek.Butyl-, tert.Butyl-, Isobutyl-, Pentyl-, Isopentyl-, 2,2-Dimethyl-propyl-, 1 -Me- thyl-butyl-, 1 -Äthyl-propyl-, 1 ,2-Dimethyl-propylgrappe und als Alkenylgruppe z.B. die Allyl- die 1-, 2-oder 3- -Buten-l-yl-, die 2-Methyl-propenyl-, Methallyl- oder die 3- oder 4-Penten-l-ylgruppe sein.
Ferner kann R1 beispielsweise sein: als Cycloalkylgruppe die Cyclopropyl-, Cyclopropylmethyl-, 2-Cyclopropyl-äthyl-, 3-Cyclopropyl-propyl-, Cyclobutyl-, Cyclobutylmethyl-, 2-Cyclobutyl-äthyl-, 3-Cyclobutyl-propyl-, Cyclopentyl-, 1 -Methylcyclopentyl-, 2-Methyl-cyclopentyl-, 3-Methyl-cyclopentyl-, l-Äthyl-cyclopentyl-, 2-Äthyl-cyclopentyl-, 3-Äthyl -cyclopentyl-, Cyclopentylmethyl-, 2-Methyl-cyclopentylmethyl-, 3 -Methyl-cyclopentylmethyl-, Cyclohexyl-, 1 -Methyl-cyclohexyl-, 2-Methyl-cyclohexyl-, 3-Methyl-cyclohekyl-, 4-Methyl-cyclohexyl-, Cyclohexylmethyl- oder die Cycloheptylgruppe und als Cycloalkenylgruppe die 2 -Cyclopenten-lyl-,

Die Umsetzung erfolgt vorzugsweise in Gegenwart eines mit Wasser mischbaren oder nichtmischbaren inerten organischen Lösungsmittels in An- oder Abwesenheit von Wasser. Geeignete inerte organische Lösungsmittel sind beispielsweise Kohlenwasserstoff, wie Benzol, Toluol oder Xylol. ätherartige Flüssigkeiten, wie Äther, Dioxan oder Tetrahydrofuran, chlorierte Kohlenwasserstoffe, wie Me thylenchlorid, und niedere Ketone, wie Aceton oder Me thyläthylketon. Es ist vorteilhaft, der Reaktionslösung ein säurebindendes Mittel zuzufügen. Als solche eignen sich z.B. anorganische Basen oder Salze, wie z.B. Alkalimetallhydroxide und Carbonate. Ferner lassen sich auch or organische Basen wie z.B.
Pyridin, Trimethyl- oder Tri äthylamin, N,N-Diisopropyläthylamin oder Collidin, verwenden, die im Überschuss zugefügt, auch als Lösungsmit tel eingesetzt werden können.
Die Umwandlung der Gruppe X des Reaktionspro duktes in die freie Aminogruppe, welche dieses in eine
Verbindung der allgemeinen Formel I überführt, wird je nach der Art der Gruppe X durch eine Hydrolyse, Re duktion oder reduktive Spaltung vorgenommen.
Durch Hydrolyse in die freie Aminogruppe überführbare Reste X sind beispielsweise Acylaminoreste, wie z.B. die Acetamidogruppe. Ferner sind solche Reste nie dere Alkoxycarbonylaminoreste, wie z.B. die Äthoxycar bonylaminogruppe, Aryloxycarbonylaininoreste, wie der Benzyloxycarbonylaminorest, oder Reste von entsprechenden Thiokohlensäurederivaten. Weitere Beispiele sind substituierte Methylenaminoreste, wie z.B. die Benzyliden amino- oder die p-Dimethylamino-benzylidenaminogruppe.
Die Hydrolyse zur Freisetzung der Aminogruppe kann in saurem Medium, z.B. in methanolischer Salzsäure, oder in verdünnter wässriger Salzsäure oder Schwefelsäure erfolgen oder, falls X durch einen Alkoxycarbonylamino rest verkörpert ist, auch unter milden alkalischen Bedingungen, z.B. mittels 1 an. oder 2-n. Natronlauge vorgenommen werden.
Ein Beispiel für einen durch Reduktion in die Aminogruppe überführbaren Rest X ist die Nitrogruppe und Beispiele für solche Reste, die durch reduktive Spaltung zur Aminogruppe führen, sind die Phenylazo- oder p-Dimethylamino-phenylazogruppen. Die Reduktion dieser Reste kann allgemein katalytisch, z.B. mittels Wasserstoff in Gegenwart von Raney-Nickel, Palladium- oder Platin -Kohle, in einem inerten Lösungsmittel, wie z.B. Äthanol, erfolgen. Neben diesen kommen auch andere übliche Reduktionsverfahren in Betracht, beispielsweise die Reduktion von Nitrogruppen oder die reduktive Spaltung von Azogruppen mittels Eisen in Essigsäure oder Salzsäure.
Als Ausgangsstoffe des Verfahrens werden Verbindungen der allgemeinen Formel III verwendet, deren Symbole R1 und R. mit den Gruppen übereinstimmen, die anschliessend an Formel I aufgezählt sind.
Eine Gruppe von Ausgangsstoffen der allgemeinen Formel III sind beispielsweise 2-Amino-4-methyl-sowie 2-Amino-4-äthyl-2-imidazoline, welche in 1-Stellung durch den Rest R1 substituiert sind. Zu den genannten l-R1-2- -amino-4-methyl-2-imidazolinen gelangt man z.B. wie folgt:
Man geht von 2-Chlor-propionylchlorid aus und setzt dieses Chlorid mit primären Aminen von der Formel H.N-R1 zu N-R1 -2-chlor-propionaminden um; diese Amide liefern mit Benzylamin N-R1-(2-benzylamino)-propionamide, welche mit Lithium-aluminiumhydrid zu N1-R1 - N-"-benzyl-l 2-propandiaminen reduziert werden: die Reduktionsprodukte lassen sich mit Palladiumkohle und Wasserstoff zu den entsprechenden N-R1-1,2-propandi- aminen entbenzylieren. welche sich mit Bromcyan unter Ringschluss kondensieren.
Homologe 1 -R1-2-amino-4- äthyl-2-imidazoline können analog hergestellt werden ausgehend von 2-Chlorbutyrylchlorid über die Zwischenprodukte N-R1-9-chlor-butyramide, N-R,-Zbenzylamino- -butyramide, N1-R1-N-benzvl-l .2-butandiamine und N1- - 1 -butandiamine.
Eine weitere Gruppe von Ausgangsstoffen der allgemeinen Formel III sind beispielsweise 2-Amino-5-methyl- -sowie '-Amino-5-äthyl-2-imidazoline, welche in l-Stel ]ung durch den Rest R1 substituiert sind. Die genannten l-R-2-amino-5-methyl-2-imidazoline können zum Beispiel ausgehend von 2-Chlor-propionylchlorid hergestellt werden.
Man setzt beisnielsweise dieses Carbonsäurechlorid mit Benzylamin zum N-Benzyl-2-chlor-propionamid um, welches mit primären Aminen von der Formel H2N-R1 N - Benzyl-2-(R-amino)-propionamide liefert; diese Amide werden mit Lithiumaluminiumhydrid zu N'- -Benzyl-Ni-R,-l ,-propan. diaminen reduziert, die mit Wasserstoff in Gegenwart eines Katalysators zu N'-R1- -1,2-propandiaminen enthenzyliert werden und schliess- lich werden die erhaltenen Diamine mit Bromcyan kondensiert und cyclisiert.
Die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der allgemeinen Formel I werden anschliessend gewünschtenfalls in ihre Salze mit anorganischen sowie organischen Säuren übergeführt. Die Herstellung dieser Salze erfolgt z.B. durch Umsetzung der Verbindungen der allgemeinen Formel I mit der äquivalenten Menge einer Säure in einem geeigneten wässrig-or organischen oder organischen Lösungsmittel. wie z.B. Methanol, Äthanol, Diäthyläther. Chloroform oder Methylenchlorid.
Zur Verwendung als Arzneistoffe können anstelle der freien Verbindungen der allgemeinen Formel I deren pharmazeutisch annehmbare Salze mit Säuren eingesetzt werden. Geeignete Additionssalze sind z.B. Salze mit Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfon säure, B-Hydroxyäthansulfonsäure. Essigsäure, Milchsäure, Oxalsäure, Bemsteinsäure, Fumarsäure, Maleinsäure, Äpfelsäure, Weinsäure, Citronensäure, Benzoesäure, Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, stellen jedoch keineswegs die einzigen Ausführungsformen derselben dar. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiels a) 31,0 g rohes N'-tert.Buteyl-1,2-propandiamin.dihy- drobromid werden mit einer Lösung von 13,2 Bromcyan in 220 ml Äther versetzt. Man fügt zu diesem Gemisch innerhalb 5 Minuten portionsweise eine Lösung von
11,4 g Natriumcarbonat in 88 ml Wasser. Die erhaltene Suspension wird 15 Stunden bei Raumtemperatur gerührt und 2 Tage stehengelassen. Man trennt die wässrige Schicht, welche das 1 -tert. Butyl-2-amino-4-methyl-2 -imidazolin enthält, von der Ätherlösung und verdünnt die wässrige Phase mit Waser, um das ausgefallene Natrium bromid zu lösen. Man versetzt die wässrige Lösung mit einer solchen von 23,8 g N-Acetyl-sulfanilylchlorid in
220 ml Aceton und ferner mit einer Lösung von 8,8 g Na triumhydroxid in 44 ml Wasser.
Anschliessend wird das
Gemisch 30 Minuten unter Rückfluss gekocht, abgekühlt, das ausgefallene Rohprodukt abfiltriert und aus Chloroform-Äthanol umkristallisiert. Man erhält das 4'-(2-Imino -3-tert.butyl-5-methyl- imidazolidin-l -alsulfonyl) - acetanilid vom Smp. 238-2400.
b) 35,2 g des nach a) erhaltenen Acetanilids werden in
180 ml 8-n. äthanolischer Salzsäure gelöst und diese Lösung 2 Tage bei Raumtemperatur stehengelassen. Man dampft das Reaktionsgemisch ein und löst den Rückstand in Wasser. Die Lösung wird abfiltriert und mit Natronlauge alkalisch gestellt. Die ausgefallenen Kristalle werden abfiltriert, mit Wasser gewaschen u. aus Aceton-Methanol umkristallisiert, wonach das 1 -Sulfanilyl-2-imino- -3-tert.butyl-5-methyl-imidazolidin bei 145-1460 schmilzt.
Das Ausgangsprodukt von a) lässt sich wie folgt herstellen: c) 16,35 g N-tert.Butyl-2-chlor-propionamid [vgl. J.
Am.Chem. Soc. 78, 6124 (1956)] und 25,0 g Benzylamin werden 3,5 Stunden auf eine Badtemperatur von 150-170 erhitzt. Aus der Lösung, die zunächst homogen ist, fallen Knstalle aus. Man kühlt das Reaktionsgemisch ab und fügt 150 ml 2-n. Natronlauge und Chloroform zu. Die organische Phase wird abgetrennt, mit Wasser gewaschen, getrocknet und eingedampft. Man destilliert den Rückstand. Nach einem Vorlauf von Benzylamin erhält man das N-tert.Butyl 2-benzylamino propionamid vom Kp.
123-195 /0,03 Torr. Das Hydrochlorid schmilzt bei 158 l(,2.
d) Zu einer Suspension von 9,0 g Lithiumaluminiumhydrid in 200 ml abs. Tetrahydrofuran tropft man innerhalb 15 Minuten eine Lösung von 23,4 g des nach c) erhaltenen Amids in 80 ml abs. Tetrahydrofuran. Dann wird die Suspension 38 Stunden unter Rückfluss gekocht.
Sie wird zunächst himbeerrot und im Verlauf von 30 Minuten wieder grau. Man kühlt das Reaktionsgemisch ab, zerstört das überschüssige Lithiumaluminiumhydrid unter Kühlung mit Essigsäureäthylester, anschliessend mit Wasser und mit Methanol und dampft die Suspension ein.
Der graue Rückstand wird zweimal mit kochendem Chloroform während 30 Minuten extrahiert und filtriert. Man trocknet die vereinigten Chloroformlösungen über Natriumsulfat und dampft sie ein. Das zurückbleibende öl wird destilliert. Das N1-tert.Butyl-N2-benzyl- 1 ,2-propandi- amin destilliert bei 72-860/0,005 Torr.
e) 22,0 g des nach d) erhaltenen Amins in 440 ml destilliertem Äthanol werden mit 34,0 g 48'70igem reinen Bromwasserstoff und 8,8 g 5%iger Palladiumkohle versetzt und bei Raumtemperatur und Normaldruck hydriert, Nachdem 95% der berechneten Menge Wasserstoff aufgenommen sind, fügt man noch 8,8 g 5%ige Palladiumkohle zu und hydriert weiter. Nach 17 Stunden hat das Reaktionsgemisch 98% der berechneten Menge Wasserstoff aufgenommen. Man filtriert den Katalysator ab, wäscht mit Äthanol nach und dampft das Filtrat ein.
Der Rückstand, das N1 -tert.Butyl- 1 ,2-propandiamin.dihy.
drobromid wird als Rohprodukt eingesetzt.
Beispiel 2 a) Analog Beispiel 1 a-b) erhält man folgendes Endprodukt:
Aus N1-Butyl- 1 2-propandiamin und Bromcyan das 1 -Butyl-2-amino- 4-methyl- 2- imidazolidin - hydrobromid (Rohprodukt), welches mit N-Acetyl-sulfanylchlorid das 4'-(2-Iminio-3-butyl-5-methylimidazolidin - 1 - ylsulfonyl)acetanilid vom Smp. 243-2440 liefert, dessen Hydrolyse das 1 -Sulfanilyl-2-imino-3-butyl-5 - methyl - imidazolidin vom Smp. 125-1260 ergibt.
Das Ausgangsprodukt wird wie folgt erhalten: b) Man tropft unter guter Eiskühlung innerhalb 30 Minuten 12,7 g 2-Chlor-propionylchlorid in 20 ml Chloroform zu einer Lösung von 16,1 g Butylamin in 50 ml Chloroform. Die erhaltene Lösung wird 15 Stunden bei Raumtemperatur stehengelassen, dann mehrmals mit Wasser gewaschen und die organische Phase abgetrennt.
Man trocknet die organische Phase über Natriumsulfat und dampft sie ein. Zurück bleibt das rohe N-Butyl-2 -chlor-propionamid.
c) Analog Beispiel 1 c-e) erhält man ausgehend vom rohen Amid von b) über die Zwischenprodukte N-Butyl -2-benzyl-aminopropionamid vom Kp. 151 - 1540/0,005 Torr und NI -Butyl-NB-benzyl-l 2-propandiamin vom Kp.
98-104/0,04 Torr das Nl-Butyl-1,2-propandiamin.
Beispiel 3 a) Analog Beispiel 1 a-b) erhält man folgendes Endprodukt:
Aus N' -Butyl- 1,2-butandiamin (Rohprodukt) mit Bromcan das l-Butyl-2-amino-4-äthyl-2-imidazolin-dihy- drobromid (Rohprodukt), welches mit N-Acetyl-sulfanilchlorid das 4'-(2-Imino-3 - butyl-5-äthyl-imidazolidin- 1 -alsulfonyl)-acetanilid (Rohprodukt) vom Smp. 107-1110 liefert, welches zum l-Sulfanilyl-2-imino-3-butyl-5-äthyl- imidazolidin vom Smp. 161-1630 hydrolisiert wird.
Die Ausgangsverbindung von a) wird wie folgt hergestellt: b) Analog Beispiele 2 b) und 1 c-e) erhält man ausgehend von 2-Chlor-butyrylchlorid über die Zwischenprodukte N-Butyl-2-chlor-butyramid (Rohprodukt), N-Butyl-2-benzylamino-butyramid (Rohprodukt) und Nl-Bu- tyl-N--benzyl-l ,2-butandiamin (Rohprodukt) das Nl-Bu- tyl- 1 ,2-butnadiamrn.
Beispiel 4 a) 23,6 g (0,1 Mol) l-tert.Butyl-2-amino-5-methyl-2 -imidazolinhydrobromid werden in 100 ml Wasser gelöst und mit 21,8 g (0,1 Mol) N-Acetyl-sulfanilychlorid in 200 ml Aceton versetzt. Dann tropft man innerhalb 10 Minuten eine Lösung von 10,0 g Natriumhydroxid in 40 ml Wasser zu und kocht die erhaltene Suspension 45 Minuten unter Rückfluss. Dann kühlt man das Reaktionsgemisch und dampft das Aceton im Vakuum ab. Man versetzt den Rückstand mit Wasser und nimmt das ausgefallene öl in Methylenchlorid auf. Die Methylenchloridlösung wird über Natriumsulfat getrocknet und eingedampft. Den Rückstand kristallisiert man aus Methanol. Das erhaltene 4'-(2-Imino-3-tert.butyl-4-methyl-imidazolidin-1-yl)- acetanilid schmilzt bei 178-1800.
b) Das nach a) erhaltene Reaktionsprodukt wird analog Beispiel 1 b) zum 1 -Sulfanil-2-imino-3-tert-butyl-4- -methyl-imidazolidin vom Smp. 178-1800 (aus Isopropanol) hydrolysiert.
Die Ausgangsverbindung, das l-tert.Butyl-2-imino- 5 -methyl-2-imidazolidin, kann wie folgt hergestellt werden: c) 12,7 g 2-Chlor-propionylchlorid werden in 50 ml Chloroform gelöst. Man tropft die Lösung innerhalb 45 Minuten zu einer Lösung von 22 g Benzylamin in 70 ml Chloroform, rührt das Reaktionsgemisch eine Stunde bei Raumtemperatur und fügt dann Wasser zu. Das Bezylamin löst sich. Man trennt die Chloroformlösung ab und wäscht sie mit Wasser. Die wässrige Phase wird einmal mit Wasser gewaschen und diese wässrige Phase mit Chloroform extrahiert. Die vereinigten Chloroformlösungen werden über Natriumsulfat getrocknet und eingedampft.
Man kristallisiert den Rückstand aus Methylenchlorid Cyclohexan, wonach das N-Benzyl-2-chlor-propionamid bei 74-760 schmilzt.
d) 19.75 g des nach c) erhaltenen Amids werden in 20 g tert. Butylamin gelöst und die erhaltene Lösung im Autoklav 3,5 Stunden auf 1500 erhitzt. Dann wird das Reaktionsgemisch mit Äther und Wasser verdünnt. Man trennt die organische Phase ab und extrahiert sie mehrmals mit 6-n. Salzsäure. Die wässrigen Phasen werden vereinigt und unter Kühlung mit konz. Natronlauge alkan lisch gestellt. Das öl, welches ausfällt, wird mit Äther extrahiert, die Ätherlösung über Natriumsulfat getrocknet und eingedampft. Das zurückbleibende N-Benzyl-2 -tert.butylamino.propionamid wird im Hochvakuum destilliert, Kp. 119-1200/0,01 Torr.
e) 23,4 g des nach d) erhaltenen Amids werden in 50 ml abs. Tetrahydrofuran gelöst. Man tropft die Lösung innerhalb 30 Minuten unter Kühlung zu einer Suspension von 9 g Lithiumaluminiumhydrid in 170 ml abs.
Tetrahydrofuran und kocht die Suspension 39 Stunden unter Rückfluss. Das Reaktionsgemisch wird gekühlt, mit 9 ml Wasser, 18 ml wässriger zeiger Natriumhydroxidlösung und 27 ml Wasser tropfenweise versetzt. Man filtriert vom entstandenen Niederschlag ab, dampft das Filtrat im Vakuum ein und destilliert den Rückstand unter Hochvakuum. Das entstandene N1-Benzyl-N2-tert. butyl-1,2-propandiamin siedet bei 74-800!0,01 Torr.
f) 22 g des nach e) erhaltenen Diamins werden in 220 ml Äthanol gelöst. Zur Lösung fügt man 34 g reine konzentrierte Bromwasserstoffsäure und 5 g 5%ige Palladiumkohle. Man hydriert das Gemisch mit Wasserstoff bei Atmosphärendruck und Raumtemperatur. Während der Hydrierung fügt man zweimal je 3 g Palladiumkohle zu.
Nachdem die Wasserstoffaufnahme beendet ist, filtriert man vom Katalysator ab und dampft das Filtrat im Vakuum ein. As has now been found, these compounds have valuable pharmacological properties, in particular l-sulfanilyl-2-imino-3-butyl-5-ethyl-, 1-sulfanilyl-imino-3-tert-butyl-4-methyl and l-sulfanilyl -2-imino-3-tert-butyl-5-methyl-imidazolidine have a hypoglycemic effect when administered orally or parenterally, which characterizes them as being suitable for the treatment of diabetes. The hypoglycemic effect is demonstrated in standard tests on warm-blooded animals, e.g. in rabbits and rats.

The mixture is hydrogenated with hydrogen at atmospheric pressure and room temperature. During the hydrogenation, 3 g of palladium-carbon are added twice each time.
