das gegebenenfalls am N-Atom eine Benzylgruppe aufweilst, reduziert oder ein entspre chendes basisch substituiertes Phenylpropanol, das gegebenenfalls am N-Atom eine Benzylgruppe aufweist, mit l-Phenyl-2-halogen-propan umsetzt und einen allfälligen an der Aminogruppe vorhandenen Benzylrest abspaltet.
Die neuen Verfahrenserzeugnisse stellen bei hervorragender Verträglichkeit wertvolle Arzneimittel mit insbesondere herz-und kreislaufwirksamen Eigenschaften dar. Darüber hinaus sind sie z. B. als Zwi schenprodukte zur Herstellung von Arzneimitteln geeignet.
Nach dem erfindungsgemässen Verfahren werden die Verfahrensprodukte der angegebenen Struktur- formel durch Reduktion von Phenytaceton in Gegenwart von Aminen der Formel

Annalen der Chemie 603, (1957), Seite 189].
Als Amine kommen beispielsweise in Frage : 1, 1-Diphenyl-1-hydroxy-3-amiho-propan, l-o-, m-oder p-Methoxyphenyl-1-phenyl
1-hydroxy-3-amino-propan, 3-Phenyl-3-hydroxy-1-amino-pentan,
3-Phenyl-3-hydroxy-1amino-butan,
3-Phenyl-3-isopropyl-3-hydroxy-1-amino-propan und 3-Phenyl-3-iso- (oder sek.)-butyl-3- hydroxy-1-amino-propan.
Die Reduktion von Phenylaceton in Gegenwart der Amine erfolgt vorzugsweise katalytisch. Als Katalysatoren verwendet man Metallba der 8. Gruppe des Periodensystems, vorzugsweise Nickelkatalysato- ren, in Gegenwart von hiorfür üblichen Lösungsmitteln, z. B. mit Wasser mischbaren aliphatischen Alkoholen. Es können auch Edelmetalle, wie Palla dium oder Platin oder Raney-Katalysatoren, verwen det werden. Ebenso kann man auch mit nasoierendem Wasserstoff, z. B. mit Aluminiumamalgam und Alto- hol, Natriumamatgam, Lithiumaluminiumhydrid oder Natriumborhydrid, reduzieren. Die Reduktion ist ferner auch elektrolytisch durchführbar.
Es ist auch möglich, die Verfahrensprodukte aus den vorstehend erwähnten Aminen durch Umsetzung mit l-Phenyl-2-halogen-propanen herzustellen, wobei 'als Halogenatome Chlor, Brom od'er Jod in Frage kommen. Die Umsetzung erfolgt zweckmässig in Gegenwart einer basischen Verbindung, um den bei der Reaktion frei werdenden Halogenwasserstoff zu binden. Zu diesem Zweck kann jedoch auch das um- ? zusetzende Amin im Uberschuss eingesetzt werden.
A1S basische Mittel eignen sich z. B. tertiÏre organische Basen, wie Trimethylamin, Triäthylamin, Pyridin oder Chinolin sowie anorganische Verbindungen, beispielsweise Natrium-, Kalium- oder Calcitumcarbonat bzw.-bicarbonat. Die Umsetzung kann mit oder ohne Verwendung eines geeigneten organischen Lösungsmittels erfolgen. Als solche Lösungsmittel eignen sich beispielsweise organische Lösungsmittel, wie Alkohole, Benzol, Toluol, ¯ther oder Chloroform. Arbeitet man in Gegenwart der genannten or ganischen Basen, so k¯nnen thèse Verbindungen gleichzeitig sowohl als L¯sungsmittel als auch als Halogenwasserstoff-bindende Mittal fungieren.
Im Falle, da¯ der Substituent R2 in der allgemeinen Formel f r die Verfahrenserzeugnisse für eine Benzylgruppe steht, wird der Benzylrest entweder im Verlauf der Reaktion oder im Anschluss an die HensteNung der Verfahrenserzeugnisse abgespalten.
Diese Abspaltung erfolgt vorteilhaft katalytisch unter Verwendung von Edelmetallkatalysatoren, wie Palla dium-Mohr. Die Abspaltung des Benzylrestes erübrigt sich in den Fäflen, wo zur Herstellung der Verfahrens- erzeugnisse von einer Hydrierungsreaktion in Gegenwart von Edelmetalkatalysatoren Gebrauch gemacht wird, da bei einer solchen Umsetzung die Benzyl- abspaltung im gleichen Arbeitsgang erfolgt.
Die erhaltenen basisch substituierten Propanole können gegebenenfalls durch Wasserabspaltung in die entsprechenden Alkenylamine bergef hrt werden. Die Umsetzung wird in Gegenwart der üblichen Dehyd'ratisierungsmittel, beispielsweise Phosphorsäure, p-Toluolsulfosäure, Phosphorpentoxyd oder Essigsäureanhydrid, entsprechend der in Liebigs Annalen der Chemie 603, S. 189, (1957) angegebenen Vorschrift durchgeiihrt.
Die Verfahrenserzeugnisse können als basische Verbindungen mit Hilfe von anorganischen oder organischen Säuren in die entsprechenden Salze iibergeführt werden. Als anorganische Säuren kommen beispielsweise in Betracht : HalogenwasserstoffsÏuren, wie Chlorwasserstoffsäure und Bromwasserstoffsäure sowie Schwefelsäure, Phosphorsäure und Amidosulfonsäure. A) ! s organische Säuren seien beispielsweise genannt : AmeisensÏure, EssigsÏure, Pro pionsäure, MilchsÏure, GlykolsÏure, GluconsÏure, Maleinsäure,. Bemsteinsäure, Weinsäure, Benzoe- sÏure, Salicylsäure, Zitronensäure, Acetursäure, Oxy ÏthansulfonsÏure und ¯thylendiamintetraessigsÏure.
Die Verfahrensprodukte stellen wertvolie ArzneiL mittel dar und zeichnen sich insbesondere durch her vorragendfe Herz-und Kreislaufwirkungen aus. So f hrt z. B. die Verabreichung von 1-Phenyl-2-[1',1'diphenyl-1'-hydroxy-propyl-(3')]-amino-propan-hy drochloritl im Versuch am isolierten Kaninchen herzen nach Langendorff bei Dauerinfusion von 1,2 γ/ Minute zu einer bedeutenden Zunahme der Coronar Durchströmung, die bei Infusion von 2 γ/Minute am grössten wird. Auch nach Beendigung der Infusion hält die Durchströmungsvermehrung noch lange an.
Ausserdem kommt es zu einer Senkung der Herzfrequenz, die sich auch am gesamten Tier bemerk- bar macht. Die gleiche Verbindung bewirkt auch im Versuch am isolierten diurchströmten Kaninchenohr nach Kraskow-Pissemsky in geringen Dosen eine ausgeprägta Gefϯerweiterung. Beispielsweise f hren 5 γ des Produktes zu einer schwachen, 150 y zu einer starken Gefässerweiterung.
Die Verfahrensprodukte sihd bereits, in so geringer Dosis coronar-und peripher-gefϯerweiternd, dassdieToxizitat(fürdieoben genannte Verbindung beträgt die Dos. let min', an der Maus 15 bis 20-mg/kg) praktisch nicht ins Gewicht fallt.
Auch im klinischen Versuch besitzt die genannte Verbindung eines gefässerweiternde Wirkung,-obei eine erweiternde Wirkung auf die Coronargefä & e deutlich ist. Sie bewirkt ausserdem ei'ne Steigerung der peripheren Durchblutung. Die coronargefässerwei- ternde Wirkung setzt sehr rasch ein und hÏlt im Vergleich zu den bekannten Nitroverbindungen, beispiels- weise dem Tetrasalpetersäureester des Brythols lange (mehrere Stunden) an. Als Einzeldosis werden 5 bis
10 mg verabreicht.
Die durch Dehydratisierung aus den basisch substituierten Propanolen erhältlichen entsprechenden Alkenylamine weisen ebenfalls eine gute Herz-und Kreislaufwirkung auf. *)
Die Verfahrenserzeugnisse können als solche oder in Form entsprechender Salze, gegebenenfalls unter Beimischung pharmazeutisch üblicher Trägerstoffe parenteral oder orali appliziert werden. Im Falle der oralen Applikation kommen als Darreichungsform vorzugsweise Tabletten oder Dragées in Frage, zu denen die Verfahrenserzeugnisse mit den üblichen Trägerstoffen, wie Milchzucker, Stärke, Traganth und Magnesiumstearat, verarbeitet werden k¯nnen.
*) So führt beispielsweise die einmalSge Injektion von 10 y 1-Phenyl-2- [1', 1'-d'iphenyl-propyl- (1')-en- (3")]-amino-propan (Beispiel 3) zu einer starken Coronargefässerweiterung. Die Toxizität dieser Verbindung beträgt Dos. let. min. i. v. bei der Maus
15 mg/kg.
Beispiel 1
11, 4g1,1-Diphenyl-1-hydroxy-propyl-amin-(3) werden mit 6, 7 g Phenylaceton in Isopropanol in Gegenwart von Palladium-Mohr hydriert. Nach Aufnahme der berechneten Menge Wass'erstoff und Abtrennung des Katalysators durch Filtration wird die Lösung unter vermindertem Druck eingeengt. Man erhält 17 g 1-Phenyl-2- [1', 1'-diphenyl-1'-hydroxy-pro- pyl- (3')]-amino-propan als ¯ligen Rückstand, der nach Aufarbeitung in üblicher Weise das Hydrochlorid vom Schmelzpunkt 213-214¯C liefert.
Das als Ausgangsstoff dienende 1, 1-Diphenyl-
1-hydroxy-propylamin-(3) kann z. B. wie folgt hergestellt werden : ?-Hydroxy-?,?-diphenyl-propionitril (hergesteNt nach der in Liebigs Annalen der Chemie 603, S. 194, angegebenen Vorschrift) wird in 500 ml mit Ammoniak gesättigtem Methanol in Gegenwart von Raney-Nickel bei 80-85 C hydriert. Nach beendeter Reaktion wird filtriert, die Lösung eingeengt, der Rückstand'in verdünnter Salzsäure gelöst und unter Zusatz von Kohle filtriert. Beim Zusatz von 2n Natronlauge fällt das 1, 1-Diphenyl-l-hydroxy-pro- pylamin- (3) kristallin aus. Der Schmelzpunkt liegt bei 143-144 C.
Beispiel 2
12, 5 g ss (m-Methoxyphenyl)-?-phenyl-?-hydroxypropylamin werden mit 6, 7 g Phenylaceton nach der in Beispiel 1 beschriebenen Methode hydriert. Nach der Aufarbeitung erhält man 16 g
1-Phenyl-2-[1'-m-methoxyphenyl-1' hydroxy-propyl-(3')]-amino-propan-hydrochlorid vom Schmelzpunkt 175-176 C.
Das als Ausgangsstoff dienende ss-(m-Methoxy- phenyl)-?-phenyl-?-hydroxy-propylamin kann wie in Beispiel 1 beschrieben durch Hydrierung aus - (m-Methoxyphenyl)-jss-phenyl--hydroxy- propionitriit, das nach der in Liebigs Annalien der Chemie 603, Seite 194, angegebenen Vorschrift aus m-Methoxybenzophenon und Acetonitril hergestellt wird, gewonnen werden. Der Schmelzpunkt betrÏgt 61-62¯ C.
Beispiel 3 17, 9 g 3-Phenyl-3-hydroxy-1-amino-pentan und 13, 4 g Phenylaceton werden entsprechend der in Beispiet I angegebenen Vorschrift hydriert. Man erhält 16, 9 g 1-Phenyl-2- [3'-phenyl-3'-hydroxy-pentyl- (1')]-amino-propan vom Schmelzpunkt 93-95 C.
Das Hydrochlorid der Base schmilzt bei 161-163 C.
Das als Ausgangsstoff eingesetzte 3-Phenyl-3-hy droxy-l-amito-pentan kann z. B. durch Hydrierung aus ?-Phenyl-?-hydroxy-valeronitril vom Schmelz- punkt 59-60 C, das nach der in Liebigs Annalen der Chemie 603, Seite 194, beschriebenen Methode aus Propiophenon und Acetonitril hergestellt wurde, erhalten werden. Die Hydrierung wird wie in Beispiel 1 beschrieben durchgeführt.
Beispiel 4
11, 1 g 1, l-Diphenyl-l-hydroxy-3-amino-propan und 3, 9 g 1-Phenyl-2-chlor-propan werden drei Stunden im Ílbad auf eine Innentemperatur von 160 C erhitzt. Nach dem Abkuhlen wird das Reaktionsgemisch mit verdünnter Salzsäure behande?@. Man erhä$t 5, 8 g
1-Phenyl-2-[1',1'-diphenyl-1-hydroxy propyl- (3')]-amino-propan-hydrochlorid, das nach dem Umkristallisieren aus Athanol/Ather bei 211-212 C schmilzt.
Beispiel S
4, 4 g 1-Phenyl-2- [1', 1'-diphenyl-1'-hyd'roxy- propyl- (3')]-amino-propan werden etwa 15 Minuten lang in 30 m$ Toluol und 2 g p-Toluolsulfosäure erhitzt, bis sich kein Wasser mehr abscheidet. Nach dem Abkühlen wird das Reaktionsgemisch mit 2n Natronlauge alkalisch gemacht und ausgeäthert. Die ätherische Lösung wird getrocknet und das Lösungsmittel abdestilliert. Der Rückstand wird durch Zugabe von verdünnter Salzsäure in das
1-Phenyl-2-[1',1'-diphenyl-propyl-(1')-en (3')]-amino-propan-hydrochlorid übergeführt, das nach dem Umkristallisieren aus Atha- nol/Ather bei 202-203 C schmilzt.
Die Ausbeute beträgt 2, 5 g.

4.4 g of 1-phenyl-2- [1 ', 1'-diphenyl-1'-hydroxy-propyl- (3')] -amino-propane are in 30 m $ toluene and 2 g p-Toluenesulfonic acid heated until no more water separates out.
