Die Erfindung betrifft ein Verfahren zum Herstellen von neuen Chinuclidinderivaten der allgemeinen Formel

Der Ausdruck Alkyl bedeutet Gruppen wie Methyl, Äthyl, Propyl oder Butyl. Der Ausdruck Isoalkyl bedeutet Gruppen wie Isopropyl oder Isobutyl. Cycloalkyl bedeutet Gruppen wie Cyclopentyl oder Cyclohexyl. Typische Aryl- oder Heteroaryl-Gruppen sind Phenyl oder Thienyl.
Die Erfindung umfasst auch die Herstellung von solchen Salzen der obigen Verbindungen (I), welche zur pharmazeutischen oder veterinärmedizinischen Anwendung geeignet sind oder sich zur Reinigung und Kristallisation der neuen Verbindungen eignen. Unter derartigen Salzen können die Hydrochloride, Sulfate, Methansulfonate und Salze mit organischen Säuren genannt werden.
Unter den N-Alkyl-Derivaten können die N-Methyl-, N-Äthyl-, N-Benzyl-Verbindungen genannt werden, insbesondere in Form der Jodide, Bromide oder Chloride.

Die erfindungsgemäss erhältlichen Verbindungen sind auch wertvolle Weckmittel und können zusammen mit Barbituraten und andern Schläfrigkeit erzeugenden Verbindungen, z. B. einigen Antihistaminika und ähnlichen Drogen, verwendet werden. Erfindungsgemäss erhältliche Verbindungen sind ferner bei der Behandlung der Hyperkinese von Kindern, der Narcolepsie, geistigen Depressionszuständen organischen Ursprungs und der Fettleibigkeit nützlich.
Die Dosis beträgt im allgemeinen etwa 10 bis etwa 100 mg für einen Erwachsenen pro Tag.
Die erfindungsgemäss erhältliche Verbindungen enthaltenden Arzneimittel können auf dem Injektionswege in einem geeigneten Verdünnungsmittel oder Träger, per os, in Form von Suppositorien und nach jeglicher andern herkömmlichen Anwendungsart, z. B. durch Infusion, verabreicht werden.
Die erfindungsgemäss erhältlichen Verbindungen sind von besonderem Wert bei der Anwendung in Kombination mit Antihistaminika, die während des Tages angewendet werden sollen, da der resultierenden Zusammensetzung die Wirkung der Schläfrigkeit auf die so behandelte Person fehlt.
Bei der Anwendung für ophthalmologische Zwecke werden die erfindungsgemäss erhältlichen Verbindungen vorzugsweise in einem herkömmlichen Träger oder Puffer verwendet. Die Wirkstoffkonzentration kann je nach dem erwünschten Effekt variiert werden und beträgt im allgemeinen etwa 2 bis 10%-Masse.

Das erfindungsgemässe Verfahren wird im folgenden durch Ausführungsbeispiele näher erläutert.
Beispiel 1 : 1, 4-Äthano-6-oxo-7-phenyl-perhydrochinolin. Hydrochlorid.
Die Verbindung 1, 4-Äthano-6-oxo-7-phenyl-1, 2, 3, 4, 6, 7, 8, 8a-octahydrochinolin wurde gemäss einer Modifikation des Verfahrens von Oppenheimer und Bergmann, Synthesis 269 (1972), wie folgt hergestellt :
Eine aus 3 g Natriummetall und 100 mit absolutem Methanol hergestellte Lösung von Natriummethoxyd in Methanol wurde mit 45 g Methylbenzylketon versetzt, worauf die erhaltene Lösung während 30 min auf Rückflusstemperatur erhitzt und dann auf OoC gekühlt und schliesslich mit 39 g 2-Methylen-chinuclidin-3-on versetzt wurde. Die so erhaltene Lösung wurde 48 h bei Raumtemperatur gerührt, dann mit 3 ml Essigsäure neutralisiert und schliesslich vom Lösungsmittel befreit, wobei ein öliger Rückstand erhalten wurde, der mit Chloroform extrahiert wurde.
Durch Abdampfen des Lösungsmittels vom Extrakt wurden 52 g (Ausbeute 60%) der erwünschten Verbindung mit Fp. = 125, 7 C (aus Aceton) erhalten. Beim Behandeln dieser Verbindung mit Chlorwasserstoffsäure wurde das entsprechende Hydrochlorid mit Fp. > 300 C (Zers. ) erhalten.
Die Doppelbindung in der erhaltenen Verbindung wurde durch Auflösen von 15 g der freien Base in 150 ml Äthanol und Reduzieren mit Wasserstoff unter 0, 3 MPa Druck während 48 h in Gegenwart von 0, 5 g eines 10% Palladium auf Kohle enthaltenden Katalysators in einem Parr-Apparat selektiv hydriert, worauf das Reaktionsgemisch filtriert und das Filtrat eingedampft wurde. Der hiebei erhaltene Rückstand wurde aus Aceton umkristallisiert und lieferte 1, 4-Äthano-6-oxo-7-phenyl- perhydrochinolin mit Fp. = 122, 6 C, das beim Behandeln mit gasförmigem Chlorwasserstoff in Aceton das Hydrochlorid in fast quantitativer Ausbeute mit Fp. > 300 C (Zers.) ergab.
Beispiel 2 : 1, 4-Äthano-6-oxo-7-äthyl-1, 2, 3, 4, 6, 7, 8, 8a-octahydrochinolin. Hydrochlorid.
Eine aus 1 g Natrium und 100 ml Äthanol hergestellte Lösung Natriumäthoxyd in Äthanol wurde mit 6, 3 g Äthyl-Äthylacetoacetat versetzt, worauf die erhaltene Lösung während 30 min auf Rückflusstemperatur erhitzt, dann auf 0 C gekühlt und schliesslich mit 5, 5 g 2-Methylen-chinuclidin- - 3-on versetzt wurde. Das Reaktionsgemisch wurde über Nacht bei Raumtemperatur gerührt, dann mit Essigsäure neutralisiert und schliesslich durch Eindampfen vom Lösungsmittel befreit. Der hiebei erhaltene Rückstand wurde mit Toluol extrahiert, worauf der erhaltene Extrakt während 7 h mit 17 ml konzentrierter Salzsäure und 5 ml Wasser bei Rückflusstemperatur behandelt wurde.
Abdampfen des Lösungsmittels vom Reaktionsgemisch und Anreiben des Rückstandes mit Äthanol ergab das kristalline Hydrochlorid des I, 4-Äthano-6-oxo-7-äthyl-1, 2, 3, 4, 6, 7, 8, 8a-octahydrochinolins mit Fp.

45 g of methylbenzyl ketone were added to a solution of sodium methoxide in methanol prepared from 3 g of sodium metal and 100 with absolute methanol, whereupon the solution obtained was heated to reflux temperature for 30 min and then cooled to OoC and finally with 39 g of 2-methylene-quinuclidine-3 -one was moved.
